Gelatin/Oxidized Konjac Glucomannan Composite Hydrogels with High Resistance to Large Deformation for Tissue Engineering Applications

Yongchao Jiang,Gaiying Li,Jie Liu,Mengya Li,Qian Li,Keyong Tang

In this study, gelatin hydrogels with remarkable compressive properties and recoverability were prepared via Shiff’s base chemical cross-linking between gelatin and oxidized konjac glucomannan (OKG). The process of OKG was first optimized by adjusting parameters, that is, oxidation temperature and time during processing. Various percentages of obtained OKG (1, 2, 3, 4, and 5 wt % to gelatin) were introduced to make composite hydrogels (G-OKGs). These G-OKGs were characterized by scanning electron microscopy (SEM), Fourier transform infrared (FTIR) spectroscopy, swelling ratio, and mechanical tests. In comparison with pure gelatin hydrogels, the G-OKGs exhibit significantly increased stress and modulus, and favorable recoverability after cyclic large deformation (up to 85% of the original height). Additionally, G-OKGs with uniform porous structures are biocompatible to support the proliferation of human umbilical vein endothelial cells. In conclusion, this study provides a reference for developing mechanically stable and recoverable hydrogels, and these kinds of hydrogels show potential capacity in tissue engineering and biomedical applications that may be undergoing extra forces and large deformation.

Changes in microstructure and rheological properties of konjac glucomannan/zein blend film-forming solution during drying

Chong Li,Fei Xiang,Kao Wu,Fatang Jiang,Xuewen Ni

During film formation at 60 °C, the microstructure and rheological properties of konjac glucomannan (KGM) film-forming solution and KGM/zein blend film-forming solution were investigated. The drying process of film-forming solutions was divided into two stages according to the drying curves. Scanning electron microscopy showed that KGM chains in the blend solution aggregated into thicker chains and formed a molecular network with larger pores. Zein particles grew larger but were homogeneously distributed during drying as observed by confocal laser scanning microscopy. The addition of zein improved the thermal stability of the film-forming solution. As the drying proceeded (up to 8 h), KGM solution exhibited a typical concentrated solution behavior due to molecular entanglement; whereas the blend solution gradually formed a weak gel after 2 h. Complex viscosity data for the film-forming solutions were well-fitted by the power-law model. The information obtained from the study is important for understanding the film-forming mechanism.

Konjac glucomannan/kappa carrageenan interpenetrating network hydrogels with enhanced mechanical strength and excellent self-healing capability

Yulong Chen,Cunzhen Song,Yukai Lv,Xin Qian

Konjac glucomannan (KGM) hydrogels have attracted considerable interest in many fields, such as drug delivery, tissue engineering, bio-medicine, and food processing, due to their good biocompatibility, biodegradability , gelation performance and security. However, KGM hydrogels prepared by traditional methods often exhibit poor mechanical properties , severely limiting their practical applications. To obtain hydrogels of excellent mechanical performance, a simple one-step method was developed. Through this method, KGM/kappa-carrageenan (KC) hydrogels of interpenetrating network (IPN) structure were prepared, where the chemical gel network was constructed via reaction between the KGM and borax and the physical network was built by introducing a KC component. Besides, the –OSO 3 − groups on the KC chains will also interact with the hydroxyl groups on the KGM chains to form hydrogen bonds , effectively binding the two networks into a whole. We find that the tensile strength of the obtained IPN hydrogels can reach to 100.2 kPa. Since the second physical network structure is temperature sensitive and recoverable, the hydrogels obtained by our method are of excellent self-healing performance. Our results showed that the mechanical strength of the self-healed hydrogels can recover by 96.7% after heat treating at 60 °C for 30 min.

Konjac glucomannan/pectin/Ca-Mg hydrogel with self-releasing alkalinity to recover phosphate in aqueous solution

Xingyu Wu,Xinyu Zhang,Yumei Xian,Yan Liu,Ling Luo,Lilin Wang,Chengyi Huang,Chao Chen,Jinsong He,Yanzong Zhang

The combination of polysaccharides can obtain stable, degradable, and environmentally friendly hydrogels, which have broad application prospects in adsorbents assembly. With Ca 2+ and Mg 2+ as crosslinkers, a new pectin/Konjac glucomannan/Ca-Mg composite hydrogel was prepared for phosphate adsorption by the alkali-thermal co-reaction method. Since Mg(OH) 2 can create a suitable pH condition for phosphate adsorption by Ca, Ca and Mg synergistically promoted phosphate adsorption and remained stable in the pH range of 4 to 10. FTIR, SEM-EDS, XRD, XPS , and zero potential analysis corroborated that the hydrogel used Ca and Mg as active sites to trap pollutants by electrostatic adsorption and fix phosphate through complexation to form Mg 3 (PO 4 ) 2 ·8H 2 O and CaPO 3 (OH) 2 ·H 2 O. Furthermore, it is unnecessary to separate the recovered phosphate from the hydrogel, and it can be used directly as a fertilizer. By being reused in the soil, it promoted seed germination and seedling growth. This adsorbent has the potential for recovery as a phosphorus-containing organic fertilizer after phosphorus adsorption.

Drug-Loaded Konjac Glucomannan/Metal–Organic Framework Composite Hydrogels as Antibacterial and Anti-Inflammatory Cell Scaffolds

Chuanyi Gu,Chunru Wang,Wenjie Ma,Yunli Gao,Junyao Li,Qing Jin,Xiaochen Wu

Bacterial infections severely threaten human health; therefore, it is important to endow the matrix for tissue engineering with antibacterial efficiency. The loading of antibacterial drugs on nanomaterials provides an efficient strategy to realize synergistic antibacterial efficiency. By depositing various metal–organic frameworks, such as UIO-66, onto konjac glucomannan (KGM), composite hydrogels (KGM/UIO-66) were created. These hydrogels were used as drug carriers, enabling the development of antibacterial hydrogels with high drug loading capacities (e.g., the maximum loading amount of pterostilbene on KGM/UIO-66 reached 0.157 mg/mg) and sustained drug release. The resulting KGM/UIO-66/pterostilbene hydrogel exhibited a three-dimensional porous structure, excellent biocompatibility, antibacterial efficiency, and anti-inflammatory activity. It effectively protected cells from bacterial attacks while ensuring cell adhesion and proliferation, demonstrating great potential as a three-dimensional substrate for biomedical applications, including tissue engineering and regenerative medicine.

Yeast protein-based meat analogues: Konjac glucomannan induces the fibrous structure formation by modifying protein structure

Songgang Xia,Jian Song,Ku Li,Tingting Hao,Chengxin Ma,Shuo Shen,Xiaoming Jiang,Changhu Xue,Yong Xue

Carbohydrate polymers can induce the fibrous structure formation of meat analogues during high-moisture extrusion (HME). In this study, meat analogues were prepared using HME (60% moisture) from yeast protein (YP) and konjac glucomannan (KGM) blends. The impact of KGM (0, 1, 2, 4%, wet basis) addition on the textural, rheological, thermal, and structural properties of high-moisture meat analogues (HMMA) was investigated. The results indicated that KGM strengthened the hardness and chewiness and adjusted the formation and orientation of fibrous structures in HMMA. When the content of KGM reached 1%, the texture of HMMA was optimal. The rheological and thermal results showed that an appropriate KGM content (<4%) enhanced the structure of HMMA, which is consistent with the textural results. In addition, the structural results (ultraviolet–visible spectroscopy, fluorescence spectroscopy, Fourier transform infrared spectroscopy , surface hydrophobicity, disulfide bond content, and X-ray diffraction) demonstrated that KGM increased the protein aggregation, β-sheet structure, S–S bond content, and ordered degree of HMMA, which were closely related to its texture and fibrous structure. The formation mechanism of KGM-induced fibrous structures in HMMA was preliminary clarified by the protein structure results. Moreover, the study suggested that YP in combination with KGM can be used to produce HMMA with a rich fibrous structure.

Improving the anti-washout property of calcium phosphate cement by introducing konjac glucomannan/κ-carrageenan blend

Guowen Qian,Xingmei Li,Fupo He,Jiandong Ye

Anti-washout calcium phosphate cement (CPC) was prepared by dissolving water-soluble konjac glucomannan (KGM) and κ-carrageenan (KC) blend in the cement liquid. The anti-washout property, setting time, compressive strength and in vitro cytocompatibility of the CPC modified with KGM/KC blend were evaluated. The results indicated that the CPC pastes modified with KGM/KC blend exhibited excellent anti-washout property. The addition of KGM/KC blend shortened the setting time and increased the injectability of CPC. Although the introduction of KGM/KC blend reduced the compressive strength of CPC, the compressive strength still surpassed that of human cancellous bone. The optimal KGM/KC mass ratio was 2:8, with which the modified cement exhibited the most efficient washout resistance and the highest compressive strength. The introduction of KGM/KC blend obviously promoted the proliferation of mouse bone marrow mesenchymal stem cells. This anti-washout CPC modified by KGM/KC blend with excellent in vitro cytocompatibility will have good prospects for application in bone defect repair.

Fabrication of chitosan-coated konjac glucomannan/sodium alginate/graphene oxide microspheres with enhanced colon-targeted delivery

Yi Yuan,Xiaowei Xu,Jingni Gong,Ruojun Mu,Yuanzhao Li,Chunhua Wu,Jie Pang

Microspheres play an increasingly important role in the food and medicine industries. In this study, konjac glucomannan (KGM)/sodium alginate (SA)/graphene oxide (GO) solution was injected into CaCl 2 solution under high-voltage static electricity assistance to fabricate microspheres. Then, chitosan (CS) was coated on the surface of the microspheres to enhance their stability. SEM images confirmed that increasing voltage decreased the particle size of microspheres obviously. Furthermore, GO was beneficial in maintaining the full structure of freeze-dried microspheres, and the CS membrane improved the surface of the microspheres with no relatively obvious gully. Results indicated that KGM interacted with SA by hydrogen bond, and GO improved this interaction in microspheres. Furthermore, swelling tests showed that the microspheres exhibited different swelling properties in different media, and the CS membrane could improve the stability of microspheres in simulated intestinal fluid and simulated colon fluids. Moreover, GO could greatly improve the ciprofloxacin (CPFX) loading efficiency of microspheres, and achieving a sustained release effect of CPFX. Thus, CS-coated KGM/SA/GO microspheres showed great potential application in drug and/or nutrition factor colon-targeted delivery.

Mussel-inspired fabrication of konjac glucomannan/microcrystalline cellulose intelligent hydrogel with pH-responsive sustained release behavior

Lin Wang,Yu Du,Yi Yuan,Ruo-Jun Mu,Jingni Gong,Yongsheng Ni,Jie Pang,Chunhua Wu

Intelligent hydrogels are attractive biomaterials for various applications, however, fabricating a hydrogel with both adequate self-healing ability and mechanical properties remains a challenge. Herein, a series of novel intelligent konjac glucomannan (KGM)/microcrystalline cellulose (MCC) hydrogels were prepared vis the mussel-inspired chemistry. MCC was firstly functionalized by the oxidative polymerization of dopamine, and the intelligent hydrogels were obtained by mixing aqueous solutions of KGM and functionalized MCC (PDMCC). By introducing PDMCC, a more compact interconnected porous structure formed for the resulting hydrogels. The self-healing ability and mechanical properties of intelligent hydrogels were dependence on the PDMCC content. Compared with KGM hydrogels, KGM/PDMCC hydrogels exhibited a more distinct pH sensitivity and a lower initial burst release, which was attributed to the compact structure and strong intermolecular hydrogen bond interaction between PDMCC and KGM. These results suggest that the KGM/PDMCC intelligent hydrogels may be promising carriers for controlled drug delivery.

Mussel-Inspired Fabrication of Konjac Glucomannan/Poly (Lactic Acid) Cryogels with Enhanced Thermal and Mechanical Properties

Lin Wang,Yi Yuan,Ruo-Jun Mu,Jingni Gong,Yongsheng Ni,Xin Hong,Jie Pang,Chunhua Wu

Three-dimensional nanofibers cryogels (NFCs) with both thermally-tolerant and mechanically-robust properties have potential for wide application in biomedical or food areas; however, creating such NFCs has proven to be extremely challenging. In this study, konjac glucomannan (KGM)/poly (lactic acid) (PLA)-based novel NFCs were prepared by the incorporation of the mussel-inspired protein polydopamine (PDA) via a facile and environmentally-friendly electrospinning and freeze-shaping technique. The obtained KGM/PLA/PDA (KPP) NFCs were characterized by field emission scanning electron microscopy (FE-SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) and compressive and tensile test. The results showed that the hierarchical cellular structure and physicochemical properties of KPP NFCs were dependent on the incorporation of PDA content. Moreover, the strong intermolecular hydrogen bond interactions among KGM, PLA and PDA also gave KPP NFCs high thermostability and mechanically-robust properties. Thus, this study developed a simple approach to fabricate multifunctional NFCs with significant potential for biomedical or food application. Keywords:nanofibers cryogels;konjac glucomannan;polydopamine;poly (lactic acid);mechanical properties

Building covalent crosslinks of carboxymethyl konjac glucomannan with boronic ester bonds for fabricating multimodal hydrogel sensor

Xuexue Ding,Lunbo Zhang,Chenguang Jiang,Siyuan Liu,Huajun Li,Juqun Xi,Defeng Wu

As the derivative of konjac glucomannan (KGM), carboxymethyl konjac glucomannan (CMK) has attracted increasing attention in the polysaccharide hydrogel fields with the aim of improving the performance related to drug delivery and release. In this study, we prepared a CMK-based hydrogel with dual characteristic crosslinks, and unlocked new applications of this type of hydrogel in soft sensor fields. CMK and poly (vinyl alcohol) were used as substrates, and physical crosslinks were constructed via the freeze-thawing treatments and covalent crosslinks were built via the boronic ester bonding. As-prepared hydrogel possessed significantly improved mechanical performance because the boronic ester bonding, on the one hand, well associated the two kinds of polymer chains, and on the other hand, played the role of ‘sacrificial crosslinks’. Furthermore, the occurrence of dynamic boronic ester bonding gave the hydrogel strain- and temperature-sensitive ionic conductivity, and therefore, the hydrogels could be used to identify human motions and as-resulted environmental temperature alterations, and worked well in various scenarios. This work activates new application of CMK in the multimodal sensing field, and also proposes an intriguing way of building multiple crosslinks in the KGM derivative-based hydrogels.

High-absorbent antibacterial konjac glucomannan/xanthan gum/bamboo fiber aerogel with carvacrol for chilled pork preservation

Chao Li,Fengxian Wang,Pengfei Xiang,Fatang Jiang,Man Xiao

To address exudate accumulation and enhance the preservation of chilled pork, a biodegradable aerogel composed of konjac glucomannan (KGM), xanthan gum (XG), bamboo fiber (BF), and varying concentrations of carvacrol (CV) was developed. The aerogels were characterized by their microstructure and properties, which exhibited rapid water absorption and good mechanical performance in both dry and wet states. The incorporation of CV endowed the aerogels with antibacterial activity against E. coli and S. aureus and antioxidant properties, as demonstrated by DPPH radical scavenging activity. Additionally, the aerogels exhibited soil biodegradability. When applied as absorbent pads for chilled pork preservation, the aerogels significantly inhibited the increase in total volatile basic nitrogen (TVB-N), thiobarbituric acid reactive substances (TBA), and total viable counts (TVC) after 24 h of storage. Moreover, the aerogels extended the pork's shelf life to 9 days at 4 °C, surpassing the 5-day shelf life achieved with commercial pads. These results underscore the aerogels' potential as absorbent pads for preserving chilled pork, offering effective solutions for both short- and long-term storage applications.
