Dulcitol ameliorates LPS-induced acute lung injury (ALI) in mice by inhibiting TLR4/NF-κB activation

Rui Ge,Qikai Su,Jian Liang,Lu Zheng,Qing-shan Li,Li Tang

The identification of natural compounds with therapeutic efficacy for acute lung injury has consistently represented a critical area of research. Dulcitol, a natural product isolated from plants, is extensively used in the food and medical industries as a sweetener. Herein, we investigated whether dulcitol exerts an anti-inflammatory effect on LPS-induced acute lung injury (ALI) in mice. In our study, ICR mice were pretreated with dulcitol at dose of 6, 12 and 24 mg/kg for seven consecutive days, followed by intratracheally instilled with LPS (10 mg/kg) to induce acute lung injury. In vitro, RAW264.7 and A549 cells were incubated with LPS in the presence of dulcitol to investigate the anti-inflammatory activity and mechanism. The results demonstrated that dulcitol effectively inhibited the infiltration of inflammatory cells and reduced the excessive production of pro-inflammatory cytokines triggered by LPS to ameliorates acute lung injury in mice. Additionally, in vitro experiments indicated that dulcitol suppressed the release of NO, IL-6 and TNF-α while reducing mRNA expression levels of IL-1β, IL-6, COX-2, iNOS and TNF-α in RAW264.7 and A549 cells in a dose-dependent manner. Furthermore, dulcitol significantly decreased TLR4, MyD88, and TRIF levels and inhibited the translocation of NF-κB from the cytoplasm to the nucleus. Our finding suggested that dulcitol could serve as a protective agent against LPS-induced acute lung injury by inhibiting the TLR-4/NF-κB signaling pathway.

Charge-Engineered LPS-Targeting Magnetic Nano-adsorbents with Optimized Harvesting Strategy Advance Sepsis Blood Purification Nanotherapeutic

Xianda Liu,Shengjun Cheng,Xijing Yang,Yilin Wang,Shifan Chen,Ziyue Ling,Yujie Xiao,Weifeng Zhao,Changsheng Zhao

This study overcomes 2 critical barriers to the clinical translation of nano-adsorbents for sepsis blood purification: the weakening of adsorption function caused by nanoparticle biofouling and the limitations in clinical translation of recovery devices. We pioneer electrically neutral phosphocholine zwitterions as selective lipopolysaccharide (LPS) ligands. Their precise charge orientation resolves the core conflict between anti-fouling efficacy and LPS capture via differential dipole realignment upon LPS binding, enabling unprecedented selective LPS capture capacity with minimal protein adsorption. To address the persistent challenges of nano-adsorbent retrieval from blood, and clinical incompatibility of existing retrieval devices with blood purification systems, we developed a discretely assembled magnetic nanocomposite platform (PCAPAN-Fe) and an extracorporeal LPS-targeting magnetic array system (ELMAS), eliminating key risks inherent in monolithic designs while ensuring complete nanoparticle harvest. In septic rabbit models, the integrated platform exhibited 100% survival with early intervention, 84.7% LPS clearance (versus 20% survival and 45.6% LPS clearance for commercial adsorbents), and marked reduction of key proinflammatory cytokines. Crucially, the therapy achieved 82.2% LPS clearance efficacy in progressive sepsis, extending survival to 40% (versus 0% for commercial adsorbents). By ingeniously integrating molecular-level ligand design with a clinically viable device, this work pioneers a paradigm shifts in sepsis nanotherapeutic, resolving the performance–biosafety paradox in blood purification.

Siraitia grosvenorii extract (Sweet Mogrosides) alleviates LPS-induced depressive-like behaviors through modulating neuroinflammation-pyroptosis axis

Yuan-yu Yang,Li-guo Li,Shang Li,Ya Su,Li-ming Liu,Quan Tang,Rui Qiu,Yong-Biao Li,Yong Cheng,Hui Zhou

This study investigates the antidepressant effects of Sweet Mogrosides (SMG) derived from Siraitia grosvenorii (SG), focusing on the outcomes of its oral administration and its novel dual-axis modulation of the neuroinflammation-pyroptosis pathway in an LPS-induced depression model. Behaviorally, oral SMG treatment significantly alleviated depressive-like behaviors in mice. Mechanistically, our results demonstrate that orally administered SMG not only rebalanced hippocampal pro- and anti-inflammatory cytokines (suppressing TNF-α while upregulating IL-10) but also significantly inhibited the NLRP3 inflammasome-mediated pyroptotic pathway, as evidenced by reduced IL-1β expression and decreased expression of GSDMD and microglial activation markers (CD68/CD86). Immunofluorescence analysis confirmed that oral SMG suppressed IBA1+ microglial activation and attenuated pro-inflammatory phenotypic shifts. By systematically elucidating that oral SMG can concurrently target both neuroinflammatory and pyroptosis cascades, this study highlights a novel mechanism of action and provides strong evidence for developing SMG as a dietary-based or orally active neuroprotective agent for inflammation-related depression.

Clostridium butyricum extracellular vesicles remodel the transcriptional network of pyroptosis-related genes in LPS-stimulated macrophages

Qingyu Zhang,Kailun He,Mohd Shafiq Aazmi,Mohd Fakharul Zaman Raja Yahya

Background: Extracellular vesicles (EVs) of probiotics are an important pathway for probiotics to exert probiotic effects. EVs of Clostridium butyricum (C. butyricum) has great potential in immunotherapy. The aim of this study is to investigate the regulatory effect of C. butyricum EVs on the pyroptosis-related gene network in LPS-stimulated macrophages and explore its potential molecular mechanisms. Methods: Differential centrifugation was used to isolate C. butyricum EVs, and EVs were characterized by transmission electron microscopy, NTA, and ZETA potential. PI staining was employed to confirm the stimulatory effect of LPS on RAW264.7 cells. Transcriptome sequencing and qPCR were performed to validate the regulatory effects of EVs on pyroptosis-related genes (PRGs) in LPS-stimulated RAW264.7 cells. Results: C. butyricum EVs exhibited a bilayer membrane structure with a median particle size of 144.4 ± 85.7 nm and a ZETA potential of -37.56 ± 1.08 mV. PI staining showed that LPS induced cell death in RAW264.7 macrophages, while EV treatment significantly affected the expression of pyroptosis-related genes IL-18 and IL-33 (q < 0.05). Transcriptome analysis revealed that C. butyricum EVs induce inflammation and cytokine secretion in RAW264.7 macrophages through the JAK-STAT signaling pathway and Cytokine receptor interaction. EVs pretreatment (10/20 μg/mL) regulated the expression of 14 PRGs in a concentration-related manner, including significant upregulation of IL-6 and P2rx7 (q < 0.01) and inhibition of ZBP1 and Tnf (q < 0.05). The gene interaction network showed a positive correlation (r=0.82) between IL-6 and P2rx7, and a negative correlation (r=-0.75) between ZBP1 and P2rx7.Conclusion: C. butyricum EVs remodel the pyroptosis-related regulatory network by modulating key genes such as IL-6, P2rx7, and ZBP1, suggesting their potential role in fine-tuning LPS-induced inflammatory responses in macrophages.

Anti-inflammatory effects of Glycine tabacina extract in LPS-stimulated macrophages and collagen-induced arthritis mice

Yanbei Tu,Kai Wang,Jian-Bo Wan,Chengwei He

Glycine tabacina (Labill.) Benth, a renowned Chinese medicinal herb, has been used for treatment of rheumatoid arthritis (RA) and joint infection. This study aimed to investigate inhibitory effect of its ethanol extract (GTE) on RA-associated inflammation. Nine isoflavonoids of twelve ones which were found in G. tabacina roots were identified by UHPLC-DAD-MS/MS. GTE treatment suppressed lipopolysaccharide-stimulated overproduction of nitric oxide, TNF-α and IL-6, and iNOS expression in macrophages. GTE blocked the activation of NF-κB and MAPK pathways, as indicated by reduced levels of phosphorylation of IKKα/β, IκBα, p38, ERK and JNK, and the suppressed nuclear translocation of NF-κB p65. Additionally, GTE administration decreased swelling of arthritic paw and serum TNF-α and IL-1β levels in collagen-induced arthritis (CIA) mice. These results demonstrated that GTE exhibited strong anti-inflammatory and anti-arthritic activities through suppressing NF-κB and MAPK pathways activation in activated macrophages and CIA mice, suggesting the potential applications of this species in RA prevention and treatment as functional food or therapeutic agent.

Phosphatidylcholine Ameliorates LPS-Induced Systemic Inflammation and Cognitive Impairments via Mediating the Gut–Brain Axis Balance

Wen Tan,Qinjun Zhang,Zhijian Dong,Yubin Yan,Yukun Fu,Xuebo Liu,Beita Zhao,Xiang Duan

Systemic inflammation will cause an imbalance in the steady state of the gut–brain axis. Phosphatidylcholine (PC) is a phospholipid found in egg yolk that has anti-inflammatory and antioxidant properties. The present research proved that PC supplementation (60 mg/kg body weight) for 35 days prevented inflammatory responses and behavioral disturbances in lipopolysaccharide (LPS)-induced mice. PC could regulate the expression of neurotrophic factors and synaptic proteins, which effectively alleviated the nerve damage and synaptic dysfunction caused by LPS. In addition, PC supplementation ameliorated gut barrier damage, altered gut genes, and improved gut health by modulating the cell adhesion molecule (CAM) pathway. Furthermore, PC remodeled the gut microbiome structure in the mice of the LPS group by increasing the relative abundance of Rikenellaceae and Lachnospiraceae. PC also increased short-chain fatty acid (SCFA) production in LPS-induced mice, which in turn ameliorated brain inflammatory responses. In conclusion, PC supplementation may be a nutritional strategy for the prevention of systemic inflammation via the gut–brain axis.

Noncanonical Wnt5a/JNK Signaling Contributes to the Development of D-Gal/LPS-Induced Acute Liver Failure

Ji Xiang-Fen,Fan Yu-Chen,Sun Fei,Wang Jing-Wei,Wang Kai

Acute liver failure (ALF) is a deadly clinical disorder with few effective treatments and unclear pathogenesis. In our previous study, we demonstrated that aberrant Wnt5a expression was involved in acute-on-chronic liver failure. However, the role of Wnt5a in ALF is unknown. We investigated the expression of Wnt5a and its downstream c-Jun N-terminal kinase (JNK) signaling in a mouse model of ALF established by coinjection of D-galactosamine (D-Gal) and lipopolysaccharide (LPS) in C57BL/6 mice. We also investigated the role of Box5, a Wnt5a antagonist, in vivo. Moreover, the effect of Wnt5a/JNK signaling on downstream inflammatory cytokine expression, phagocytosis, and migration in THP-1 macrophages was studied in vitro. Aberrant Wnt5a expression and JNK activation were detected in D-Gal/LPS-induced ALF mice. Box5 pretreatment reversed JNK activation and eventually decreased the mortality rate of D-Gal/LPS-treated mice, with reduced hepatic necrosis and apoptosis, serum ALT and AST levels, and liver inflammatory cytokine expression, although the latter was not significant. We further demonstrated that recombinant Wnt5a (rWnt5a)-induced tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) mRNA expression and increased THP-1 macrophage phagocytosis in a JNK-dependent manner, which could be restored by Box5. In addition, rWnt5a-induced migration of THP-1 macrophages was also reversed by Box5. Our findings suggested that Wnt5a/JNK signaling plays an important role in the development of ALF and that Box5 could have particular hepatoprotective effects in ALF.

Inhibiting HNF4A suppresses inflammation whilst promoting trophoblast invasion and migration: A promising target for the treatment of preeclampsia

Xuan Zhang,Lei Sun

Preeclampsia (PE) is a complex disease of pregnancy, and an important cause of this disease is insufficient trophoblast invasion and migration. However, the underlying mechanism of PE remains largely unknown. Here, transcriptome sequencing analysis found the high expression of hepatocyte nuclear factor 4 alpha (HNF4A) in PE placentas. Meanwhile, we found that HNF4A expression was up-regulated in the placentas of PE patients. Thus, we assumed that HNF4A might be involved in PE progression. To validate our hypothesis, l -arginine methyl ester ( l -NAME) or lipopolysaccharide (LPS)-treated rats were used to mimic the pathological status of PE in vivo . Consistently, HTR8/SVneo cells were treated with hypoxia/reoxygenation (H/R) or LPS to simulate PE progression in vitro . The results observed an increase in elevated urine protein levels, systolic blood pressure (SBP), diastolic blood pressure (DBP), and mean arterial pressure (MAP), which indicated that the PE-like rat model was successfully established. Meanwhile, the expression of pro-inflammatory cytokines interleukin (IL)-6 and IL-1β was increased in PE placentas. HTR8/SVneo cells were used to further explore the underlying mechanism of PE in vitro . H/R conditions up-regulated the acetylation level of HNF4A. Further analysis showed that HNF4A overexpression inhibited trophoblast invasion and migration, while HNF4A knockdown promoted the progression. Additionally, inhibiting HNF4A was found to reduce the levels of IL-6 and IL-1β secretion in HTR8/SVneo cells following H/R or LPS exposure. Conclusively, these findings suggest that inhibiting HNF4A suppresses inflammation whilst promoting trophoblast invasion and migration in PE, providing a promising target for the treatment of PE.

GKLF, a transcriptional activator of Txnip, drives microglia activation in kainic acid-induced murine models of epileptic seizures

Zhihong Zhuo,Yao Wang,Huimin Kong,Tianjiao Fu

Neuroinflammation is a major component of epilepsy. Gut-enriched Kruppel-like factor (GKLF), a transcription factor of Kruppel-like factor family, has been reported to promote microglia activation and mediate neuroinflammation. However, the role of GKLF in epilepsy remains poorly characterized. This study focused on the function of GKLF in neuron loss and neuroinflammation in epilepsy and the molecular mechanism underlying microglia activation induced by GKLF upon lipopolysaccharides (LPS) treatment. An experimental epileptic model was induced by an intraperitoneal injection of 25 mg/kg kainic acid (KA). Lentivirus vectors (Lv) carrying Gklf CDS or short hairpin RNA targeting Gklf (shGKLF) was injected into the hippocampus, resulting in Gklf overexpression or knockdown in the hippocampus. BV-2 cells were co-infected with Lv-shGKLF or/and Lv carrying thioredoxin interacting protein ( Txnip ) CDS for 48 h and treated with 1 μg/mL LPS for 24 h. Results showed that GKLF enhanced KA-induced neuronal loss, pro-inflammatory cytokine secretion, activation of NOD-like receptor protein-3 (NLRP3) inflammasomes and microglia, and TXNIP expression in the hippocampus. GKLF inhibition showed negative effects on LPS-induced microglia activation, as evidenced by reduced pro-inflammatory cytokine secretion and activation of NLRP3 inflammasomes. GKLF bound to Txnip promoter and increased TXNIP expression in LPS-activated microglia. Interestingly, Txnip overexpression reversed the inhibitory effect of Gklf knockdown on microglia activation. These findings indicated that GKLF was involved in microglia activation via TXNIP. This study demonstrates the underlying mechanism of GKLF in the pathogenesis of epilepsy and uncovers that GKLF inhibition may be a therapeutic strategy for epilepsy treatment.

Therapeutic Effects of Lifei Decoction in a Murine Model of COPD Induced by LPS and Cigarette Smoke

Liguo Lu,Chengdong Zhu,Jian Xu,Yulan Hu,Juxiang Dai,Sheng Wang,Tao Wei

Introduction The Lifei Decoction (LD) is a commonly utilized Chinese medicine for the treatment of sepsis and bronchial inflammation. However, its therapeutic potential in chronic obstructive pulmonary disease (COPD) remains unknown. Therefore, the objective of this study was to investigate the therapeutic efficacy and underlying mechanism of LD in a mouse model of COPD induced by cigarette smoke (CS) combined with lipopolysaccharide (LPS).Methods Hematoxylin-eosin (H&E) staining was employed to observe the pathological alterations in lung tissue, while ELISA was utilized for the detection of levels of inflammatory factors in both lung tissue and bronchoalveolar lavage fluid (BALF). Additionally, Western blot analysis was conducted to assess the expression of p-NF-κB, GDF11, ZO-1, and Occludin-1 proteins. The changes in intestinal flora were evaluated using the viable bacteria count method. Results The administration of LD demonstrates significant efficacy in mitigating pulmonary tissue damage in a murine model, while concurrently inhibiting the activation of the inflammatory pathway NF-κB to attenuate the levels of pro-inflammatory factors. Moreover, LD exhibits the capacity to enhance the expression of intestinal functional proteins ZO-1 and Occludin-1, thereby rectifying dysbiosis within the gut microbiota. Conclusion The LD shows great promise as a potential treatment for COPD.

High salt condition alters LPS synthesis and induces the emergence of drug resistance mutations in Helicobacter pylori

Hongming Huang,Huang Yang,Shunhang Feng,Xiaoyan Zhang,Chu Chen,Hongyu Yan,Rui Li,Mengxin Liu,Juan Lin,Yancheng Wen,Feifei She

The burgeoning emergence of drug-resistant Helicobacter pylori strains poses a significant challenge to the clinical success of eradication therapies and is primarily attributed to mutations within drug-targeting genes that lead to antibiotic resistance. This study investigated the effect of high salt conditions on the occurrence of drug-resistance mutations in H. pylori. We found that high salt condition significantly amplifies the frequency of drug resistance mutations in H. pylori. This can be chiefly attributed to our discovery indicating that high salt concentration results in elevated reactive oxygen species (ROS) levels, initiating DNA damage within H. pylori. Mechanistically, high salt condition suppresses lipopolysaccharide (LPS) synthesis gene expression, inducing alterations in the LPS structure and escalating outer membrane permeability. This disruption of LPS synthesis attenuates the expression and activity of SodB, facilitates increased ROS levels, and consequently increases the drug resistance mutation frequency. Impairing LPS synthesis engenders a reduction in intracellular iron levels, leading to diminished holo-Fur activity and increased apo-Fur activity, which represses the expression of SodB directly. Our findings suggest a correlation between high salt intake and the emergence of drug resistance in the human pathogen H. pylori, implying that dietary choices affect the risk of emergence of antimicrobial resistance.

κ/β-Carrageenan oligosaccharides promoting polarization of LPS-activated macrophage and their potential in diabetes wound healing

Rijian Song,Xuan Mao,Shunqing Tang

Here we prepared a low-degree-sulfated κ/β-carrageenan oligosaccharide (L-DS-KOS) by DMSO-methanol desulfation method, and fabricated a sponge dressing for the wound healing of diabetic rats. The immunomodulatory effects of L-DS-KOS on M1-like macrophages were evaluated. Results showed that L-DS-KOS could effectively promote the secretion of anti-inflammatory factors and accelerate polarization of LPS-activated macrophages from M1 to M2 type. The gross examination result showed that the sponge dressing with the mass ratio of L-DS-KOS: collagen = 3: 7 could effectively accelerate the repair process of the full-thickness excisional wound in diabetic rats; and H&E and Masson staining results disclosed that the L-DS-KOS/collagen dressing could better shorten the inflammation period of the wound site, and improve the process of collagen deposition and epithelial formation, thereby promote the repair of skin wounds in diabetic rats. These results demonstrate that L-DS-KOS has potential to be used as a new type of immunomodulatory biomaterial for diabetic wound dressings.
