Odor profile characterization and variety identification of brown lactobacillus beverage based on untargeted metabolomics

Mingguang Yu,Baosong Wang,Ying Wang,Yuan Tang,Chen Liu,Huanlu Song,Baochao Hou,Baolei Li,Wen Zhao

Odor is an important attribute of fermented dairy products and is an important factor in determining consumer acceptance. However, the odor characteristics of brown lactobacillus beverage (BLB) and the differences in odor among different types of these products have rarely been studied. Here, a comprehensive analysis of different types of BLB were performed by SPME–GC–O–MS and SPME–GC×GC–O–MS techniques based on untargeted metabolomics. Compared with SPME–GC–O–MS, SPME–GC×GC–O–MS identified 26 new odor compounds in BLB, indicating that this method is more suitable for odor characterization of BLB. A total of 149 odor compounds were identified from 13 BLBs, of which 40 were odor-active compounds. Ketones , alcohols, acids, aldehydes, and esters accounted for about 80% of the total odor compounds and constituted the basic odor profile of the BLB. In addition to identifying indicator flavor compounds such as furfuryl alcohol , octanoic acid , and β -myrcene, which are responsible for the differences in odor profiles between different BLBs, the reference content ranges in BLBs were precisely quantified. These findings will provide new insights into the species traceability and odor characterization of BLB samples.

Peptides with Charged Amino Acids Mitigate nZnO-Induced Growth Inhibition of Lactobacillus rhamnosus LRa05

Yuezhen Ouyang,Shiying Nie,Xinquan Yang,Xin Xu,Miao Zhou,William Kwame Amakye,Erdong Yuan,Jiaoyan Ren

Growing concern is about the potential side effects of nanomaterials from food packaging, notably zinc oxide nanoparticles (nZnO). Previous research revealed that walnut-derived peptides could mitigate this inhibitory effect, but the mechanism involved is unclear. Here, we found that not all peptides have such an effect. Based on the growth inhibition model of Lactobacillus rhamnosus LRa05 induced by nZnO, we assessed the protective effects of various peptides. Notably, four peptides containing charged amino acids (PPKNW, WPPKN, ADIYTE, and WEREEQE) were found to effectively alleviate the growth inhibition phenomenon. We hypothesize that the peptide–nZnO interaction modifies this effect, as confirmed through infrared, Raman, and fluorescence spectroscopy. Our results highlight amide bonds, amino groups, carboxyl groups, and benzene rings as key peptide binding sites on nZnO, with static quenching primarily due to hydrogen bonds and van der Waals forces. This study elucidates peptide characteristics in nZnO interactions, facilitating a deeper exploration of food matrix–nanocomposite interactions.

Silk-fibroin-based nanofibrous scaffolds integrated with nano-engineered living bacteria enabling antibiotic-free accelerated infected wound healing

Ming Teng,Xiaomin Luo,Fen Ao,Liuying Li,Jianyan Feng,Peng Zhang,Xiaomeng Zhang,Xudan Liu,Chen Yang,Lufeng Ji,Xuechuan Wang,Haiyan Ju,Xinhua Liu

Bacteria‐infected skin wounds are an immense public health challenge for high mortality-rate and enormous financial burden worldwide. Emerging living bacterial therapy promises the desired therapeutic efficacy for wound healing, however, its indiscriminate negative effects on the bioactivity of beneficial bacteria remain enormous challenges for clinical far-ranging applications. Herein, silk fibroin-based nanofibrous scaffolds equipped with nano-engineered tannic acid (TA)-Fe(III) chelate-encapsulated Lactobacillus reuteri (denoted as Mbr/ L @FeTA) were proposed for antibiotic-free accelerated bacteria‐infected wound healing. Mbr/ L @FeTA exhibited excellent antimicrobial properties (99.03% and 98.60% bactericidal efficiency against Escherichia coli and Staphylococcus aureus , respectively), gratifying mechanical properties and biocompatibility. Furthermore, due to on-demand TA-Fe(III) chelate-encapsulated bacterial tactics, Mbr/ L @FeTA demonstrates exceptional environmental resistance and maintains satisfactory biological-activity even when administered with antibiotics. Additionally, Mbr/ L @FeTA enables the stable release of organic acids and other metabolites that significantly improve the wound microenvironment and modulate inflammatory reactions. In vivo and in vitro experiments confirmed that Mbr/ L @FeTA strongly promoted cell growth and proliferation, inhibited bacteria growth at the wound-site, and further accelerated the healing process in bacteria-infected wounds. These findings suggest that Mbr/ L @FeTA exhibits great potential for applications in antibiotic-free infected wound therapy and simultaneously represents a useful approach for clinical applications of living bacterial therapy.

Expression, biochemical characterization, and mutation of a water forming NADH: FMN oxidoreductase from Lactobacillus rhamnosus

Fei-Long Li,Wen Bin Su,Qing-Lan Tao,Liao-Yuan Zhang,Ye-Wang Zhang

Enzyme-catalyzed cofactor regeneration is a significant approach to avoid large quantities consumption of oxidized cofactor, which is vital in a variety of bioconversion reactions. NADH: FMN oxidoreductase is an ideal regenerating enzyme because innocuous molecular oxygen is required as an oxidant. But the by-product H 2 O 2 limits its further applications at the industrial scale. Here, novel NADH: FMN oxidoreductase (LrFOR) from Lactobacillus rhamnosus comprised of 1146 bp with a predicted molecular weight of 42 kDa was cloned and overexpressed in Escherichia coli BL21 (DE3). Enzyme assay shows that the purified recombinant LrFOR has both the NADPH and NADH oxidation activity. Biochemical characterizations suggested that LrFOR exhibits the specific activity of 39.8 U·mg −1 with the optimal pH and temperature of 5.6 and 35 °C and produces H 2 O instead of potentially harmful peroxide. To further study its catalytic function, a critical Thr29 residue and its six mutants were investigated. Mutants T29G, T29A, and T29D show notable enhancement in activities compared with the wild type. Molecular docking of NADH into wild type and its mutants reveal that a small size or electronegative of residue in position29 could shorten the distance of NADH and FMN, promoting the electrons transfer and resulting in the increased activity. This work reveals the pivotal role of position 29 in the catalytic function of LrFOR and provides effective catalysts in NAD + regeneration.

Effects of exopolysaccharide from Lactobacillus rhamnosus on human gut microbiota in in vitro fermentation model

Yuzhu Zhu,Jia-Min Zhou,Wei Liu,Xionge Pi,Qingqing Zhou,Ping Li,Tao Zhou,Qing Gu

Exopolysaccharides (EPS) produced by some bacteria have prebiotic potential and contribute to the regulation of the gut microbiota. Four types of EPS with different molecular composition from Lactobacillus rhamnosus ZFM231 were identified. In this work, we evaluated the potential of EPS on human gut microbiota by in vitro fermentation model. It was found that purified EPS fractions were degraded effectively by human gut microbiota. Gas chromatography (GC) assay revealed that EPS could be fermented to produce short chain fatty acids (SCFAs) by gut microbiota in human fecal samples from twelve healthy volunteers. 16S rDNA sequencing showed that EPS mainly increased the abundance of Ruminococcus , Dorea , Butyricicoccus , Blautia and others. The current research provides valuable new information on the mechanisms of absorption, utilization and functional activity of Lactobacillus rhamnosus EPS in the gastrointestinal tract of the human body.

Impact of a Novel Nano-Protectant on the Viability of Probiotic Bacterium Lactobacillus casei K17

Jinsong Wang,Lanming Chen

Probiotics are considered as desirable alternatives to antibiotics because of their beneficial effects on the safety and economy of farm animals. The protectant can ensure the viability of probiotics, which is the prerequisite of the beneficial effects. The objective of this study was to evaluate the effects of a novel nano-protectant containing trehalose, skim milk powder, phytoglycogen nanoparticles, and nano-phytoglycogen Pickering emulsions on the viability ofLactobacillus caseiK17 under different conditions. The results indicated that the optimal concentration of the carbohydrate substrate was determined to be 10% skim milk powder (w/w) instead of trehalose. The combination of 10% skim milk powder (w/w), 1% phytoglycogen nanoparticles (w/w), and 10% Pickering emulsions (w/w) was selected as the optimal component of the protectant. Trilayer protectants with an optimal component had a more significant protective effect on the bacteria than that of the monolayer and bilayer protectants, or the control in feed storage, freeze-drying, and simulated gastrointestinal environment. A scanning electron microscope was used to monitor the morphological characteristics of the protectants for different layers onL. casei. In conclusion, the trilayer protectant exhibited a substantial effect onL. caseiduring storage and consumption, which could be used in the feed and functional food. Keywords:Lactobacillus casei;protectants;probiotics;phytoglycogen nanoparticle;pickering emulsions

Directed evolution of feruloyl esterase from Lactobacillus acidophilus and its application for ferulic acid production

Shujun Liu,Lubna Soomro,Xue Wei,Xufeng Yuan,Tianyi Gu,Zhen Li,Yulu Wang,Yuming Bao,Fengzhong Wang,Boting Wen,Fengjiao Xin

Producing ferulic acid (FA) from the natural substrate with feruloyl esterase is promising in industries, screening and engineering new enzymes with high efficiency to increase the FA yield is of great concern. Here, the feruloyl esterase of Lactobacillus acidophilus (FAELac) was heterologous expressed and the FAELac with different oligomerization states was separated. Interestingly, the activity of dimer was 37-fold higher than high-polymer. To further enhance the efficiency of FAELac, eight mutants were generated based on the simulated structure, of which Q198A, Q134T enhanced the catalytic efficiency by 5.4- and 4.3-fold in comparison with the wild type. Moreover, higher yields of FA (2.21, 6.60, and 1.67 mg/g substrate, respectively) were released by the mutants from de-starched wheat bran, insoluble wheat arabinoxylan, and steam-exploded corn stover. These results indicated that improving the purification process, engineering new FAELac and substrates bias studies hold great potential for increasing FA production yield.

Insights into quality attributes and digestive properties of Lactobacillus-assisted fermented Polygonatum cyrtonema rice cake

Jihui Gao,Tian Zheng,Yuyang Shi,Lei Jiang,Lei Huang,Bingsheng Qiu,Yongxin Li,Gang Chen,Jinping Si,Huqing Yang,Shangbin Bai

L. plantarum P5 improved the fermentation performance of PC-enriched FRCs. • L. plantarum P5 - assisted fermentation reduced off-flavors in PC-enriched FRCs. • The PCL group exhibited slower moisture migration, delaying starch retrogradation. • The PCL group predominantly formed V-type complexes, reducing starch digestibility. Although Polygonatum cyrtonema (PC) is rich in bioactive compounds, its direct incorporation may compromise the sensory quality of fermented products. This study systematically investigated the effects of PC on fermented rice cakes (FRCs), using Lactobacillus plantarum P5-assisted fermentation as a strategy to improve product quality. L. plantarum P5, as the optimal strain, significantly promoted yeast growth and increased total soluble solids in fermented glutinous rice, thereby improving the specific volume, height-to-diameter ratio, and textural characteristics of PC-enriched FRCs. SPME-GC–MS analysis showed that P5-assisted fermentation reduced off-flavor compounds, including hexanal and (E)-2-heptenal. Additionally, the group containing both PC and L. plantarum P5 (PCL) exhibited a slower staling rate and reduced moisture migration during storage. The resistant starch (RS) content in the PCL group significantly increased from 17.94% to 20.17%. X-ray diffraction analysis revealed that the PCL group exhibited predominantly V-type crystalline structures, indicating the development of inclusion complexes between bioactive compounds in PC and starch. Meanwhile, the PCL group also showed enhanced hydrogen bonding between PC polysaccharides (PCPs) and starch chains, promoting starch aggregation and the development of a more compact gel network. Overall, this study provides an approach for incorporating PC into fermented cereal products with enhanced product quality and reduced starch digestibility. Purification and structural characterization of atypical dextran-type capsular exopolysaccharide from Lactobacillus gasseri ATCC 33323

Tang Huazhi,Ye Zixin,Chen Mingsheng,Yang Xiaotong,Song Chengcheng,Tai Guihua,Zhou Yifa,Cui Sisi,Cui Liangnan

Background Probiotic strain Lactobacillus gasseri ( L. gasseri ) is widely used in the food industry due to its diverse bioactive activities, in which the exopolysaccharide (EPS) is a key functional component. While the secreted EPS from L. gasseri has been previously explored, its capsular form remains much less characterized, limiting the comprehensive understanding of EPS-derived postbiotics in this species. This study aims to isolate, purify, and structurally characterize the capsular exopolysaccharide from L. gasseri ATCC 33323. Results A capsular exopolysaccharide, designated C-EPS L.g -G, was isolated and purified from L. gasseri ATCC 33323. Structural characterization revealed that C-EPS L.g -G (Mw 36.1 kDa) is a highly branched, atypical dextran with a branching degree of 76.9%. It consists of an α-(1→6) backbone substituted at the α-(1→2) position by monomeric glucose side chains. These side chains have a defined arrangement with an unsubstituted/sparse central core that is symmetrically flanked by dense branches. Notably, 52.4% of the side chains carry 1-phosphoglycerol modifications at the O-6 position. Additionally, minor acetyl (8.3%) and 3-hydroxypropionyl (10.5%) modifications are present in C-EPS L.g -G at variable acylation sites. In 0.15 M NaCl solution, C-EPS L.g -G adopts a spherical conformation characterized by a small Z-average hydrodynamic diameter (7.64 ± 0.20 nm), moderate intrinsic viscosity (11.61 ± 0.21 mL/g), and slightly negative zeta potential (– 8.71 ± 0.32 mV). Comparative genomic analysis revealed that the biosynthetic gene locus for this atypical dextran is conserved across most L. gasseri and Lactobacillus johnsonii species. Conclusions This study reports the purification and comprehensive structural characterization of an atypical dextran-type capsular exopolysaccharide C-EPS L.g -G from L. gasseri ATCC 33323 for the first time. Resulting structural insights provide a foundation for future functional exploration and potential applications of this atypical dextran in the food industry. Milk extracellular vesicles attenuate pre-inflammatory bowel disease via Lactobacillus murinus-mediated TLR2 signaling modulation

Jiarun Xu,Guoyue Wei,Jianfeng Wang,Haoran Wang,Jian He,Jiaqi Xu,Wenguo Cao,Jialin Wang,Qi Shen,Nasi Ai,Yufang Su,Huaxi Yi,Lingjun Tong

Pre-inflammatory bowel disease (pre-IBD) represents a transitional state between irritable bowel syndrome and IBD, characterized by heightened susceptibility to mucosal inflammation driven by chronic high-fat diet intake and antibiotic exposure. However, effective intervention strategies remain poorly defined. Here, we identify milk-derived extracellular vesicles (mEVs) as a potent nanotherapeutic capable of attenuating pre-IBD pathology. Pre-IBD mice displayed marked gut microbiota dysbiosis and mucosal inflammation, both of which were substantially ameliorated by oral mEVs administration. Mechanistically, mEVs restored microbial homeostasis by enriching Lactobacillus murinus ( L. murinus ), which in turn reduced lysophosphatidylcholine (LPC) accumulation, two factors that exerted opposing and functionally instructive effects on intestinal inflammation. LPC activated the TLR2/MAPK axis to promote M1 polarization, whereas L. murinus engaged the TLR2/STAT3 pathway to drive M2 polarization. Collectively, these findings demonstrate that mEVs reprogram TLR2-dependent macrophage responses by modulating LPC and L. murinus , thereby mitigating mucosal inflammation and preventing the progression of pre-IBD.

Lactobacillus salivarius li01 alleviates premature ovarian insufficiency via gut microbiota-mediated modulation of tryptophan metabolism

Lingnv Yao,Siyuan Xie,Shuobo Zhang,Furong Ba,Huijuan Jing,Bo Qiu,Lvwan Xu,Shengyi Han,Hang Xiao,Ming Zhao,Xiaowei Shi,Lanjuan Li,Mingfei Yao

Premature ovarian insufficiency (POI) threatens women’s reproductive health. Emerging evidence implicates the gut-ovary axis in POI pathogenesis. This study explored the probiotic Lactobacillus salivarius Li01 in a Tripterygium wilfordii glycosides-induced POI mouse model. Li01 effectively mitigated POI progression by restoring hormone levels, increasing functional follicle numbers, and improving ovarian antioxidant function. These beneficial effects were primarily mediated through modulation of the gut microbiota and host tryptophan metabolism. Notably, a tryptophan-enriched diet alone did not improve POI. When combined with Li01, it synergistically amplified the kynurenine pathway. This shift promoted ovarian NAD⁺ production by upregulating the levels of NAMPT, while attenuating NAD⁺ depletion through downregulation of CD38 expression. The elevated NAD⁺ levels subsequently activated SIRT1 expression, stimulated the synthesis of SOD and other antioxidants, and inhibited the Caspase-1/IL‑1β inflammasome axis. These findings highlight Li01 as a potential POI therapy through microbiota-metabolic regulation, offering new mechanistic insights into gut-ovarian interactions.

Effects and Mechanism of Lactobacillus gasseri CCFM1346 Combined with Rutin on Alleviating Hyperuricemia in Mice

Tang Xin,Zhang Shuling,Mao Bingyong,Zhang Qiuxiang,Zhao Jianxin,Chen Wei,Cui Shumao

Rutin shows potential for reducing uric acid (UA) but suffers from low bioavailability. Microbial biotransformation can convert rutin into metabolites with enhanced absorption, stability, and physiological activity. This study aims to investigate the biotransformation effects of probiotics containing α-L-rhamnose glycosidase and α-L-rhamnose glycosidase on rutin, as well as their therapeutic efficacy for hyperuricemia (HUA). In vitro, Lactobacillus gasseri CCFM1346 exhibited significantly higher conversion rates for transforming rutin into quercetin and isoquercetin compared to other strains. In HUA mice, both the CCFM1346-fermented rutin preparation (post-Rutin) and a synbiotic combination (Synbiotic) significantly lowered serum UA and creatinine levels ( p  < 0.05). Moreover, the Synbiotics group achieved superior serum UA reduction (74.7%) compared to the post-Rutin group (60.1%), despite more moderate xanthine oxidase inhibition. These treatments effectively decreased hepatic xanthine oxidase and enhanced urate oxidase activity, as well as downregulated mRNA levels of UA reabsorption transporters ( URAT1 and GLUT9 ) while upregulating the efflux transporter ABCG2 . Moreover, the combination of L. gasseri CCFM1346 with rutin significantly inhibited the transcription of renal inflammasome components ( NLRP3 , ASC , Caspase1 ) and inflammatory cytokines ( IL-6 , IL-1β , TNF-α ). Futuremore, correlation analysis indicates that these indicators exhibit a significant positive correlation ( p  < 0.001). In conclusion, L. gasseri CCFM1346-fermented rutin alleviates HUA through multi-mechanistic actions, with the synbiotic preparation demonstrating superior efficacy by concurrently targeting UA production, excretion, and inflammation. These findings provide a foundation for developing synbiotic functional foods for UA management.
