Oligopeptide modified ICG/Cisplatin@CaCO3 for targeted osteosarcoma NIR-II imaging guided multimodal therapy

Li-chen Ji,Jia-qing Huang,Xu-gang Zhong,Qi-hong Sun,Yong Fan,Li Yin,Qiong Zhang,Yu Cai,Zeju He

Osteosarcoma, as the most prevalent primary malignant bone tumor, has consistently shown suboptimal clinical treatment outcomes. In this manuscript, we proposed an efficacious and novel phototherapy strategy for osteosarcoma based on a photoresponsive near-infrared (NIR) material, CaCO 3 -ICG-DDP-PEG-PT (CIDP-PT), which contained calcium carbonate (CaCO 3 ) and Cisplatin (DDP) via the gas dispersion method, and indocyanine green (ICG) was encapsulated through a nanoprecipitation reaction by employing DSPE-PEG 2000 -COOH to enhance the stability and hydrophilicity. The targeting oligopeptide (PT) for osteosarcoma cells was inserted into the outer membrane of nanoparticles, thereby further enhancing the tumor-targeting capability through receptor-mediated binding. The prepared CIDP-PT nanoparticles exhibited high targeting efficiency and multifunctionality. In vitro and in vivo experiments demonstrated that CIDP-PT significantly enhanced the therapeutic efficacy for osteosarcoma via the NIR-II photoresponsive and tumor-targeting approach. It achieves efficient tumor cell eradication through multiple modalities while maintaining an exceptional level of biocompatibility. Hence, the CIDP-PT nanocarriers possess considerable practical value for future clinical applications in adjunct osteosarcoma phototherapy.

NIR-II photo- and sono-responsive hyaluronic acid-capped nanozymes for glioblastoma-targeting theranostics

Huanran Qu,Yuanyuan Zhang,Minghao Chen,Shiyang Shao,Jianqiang Chen,Yundi Wu,Xilong Wu

Glioblastoma is the most aggressive and heterogeneous astrocytic tumor, with its intracranial location limiting the efficacy of conventional therapies. This study presents the synthesis of a multifunctional nanocomplex utilizing borophene nanosheets (B NSs) as a substrate. Gold nanoparticles (Au NPs) are modified onto the surface of B NSs to create Schottky heterojunctions (B Au). This is followed by co-precipitation solvothermal synthesis incorporating silver sulfide (Ag 2 S) and hyaluronic acid (HA) as a capping agent, yielding B-Au-Ag 2 S-HA (BAA-HA). The Schottky heterojunction reduces the bandgap and accelerates charge carrier separation, significantly enhancing the sonodynamic therapy (SDT) efficiency of B NSs. In comparison to B NSs, BAA-HA exhibits significantly improved photothermal conversion efficiency under 1064 nm laser irradiation, facilitating the cascade catalysis of glucose oxidase-like (GOx-like) and catalase-like (CAT-like) enzymes. This accelerates glucose and H 2 O 2 decomposition, increasing O 2 supply to amplify SDT efficacy and induce immunogenic cell death (ICD), inducing a robust anti-tumor immune response. Ultrasound-targeted microbubble destruction technology is employed to transiently open the blood-brain barrier, allowing for targeted delivery of BAA-HA to glioblastoma cells via HA-mediated recognition of the CD44 receptor. Additionally, the NIR-II fluorescence properties of Ag 2 S enable precise tumor imaging, guiding multimodal synergistic therapy. This platform provides a promising strategy for treating deep-seated tumors, integrating therapeutic and diagnostic functions to enhance efficacy and precision.

NIR-II Photoacoustic-Cavitation Boosted Metabolic Reprogramming Regulation Toward the Localized TNBC Growth Inhibition

Leilei Zhang,Haoheng Bai,Weiqi Sun,Luqi Wang,Minyan Duan,Qianying Liu,Chao Zhou,Xueyang Fang,Tao Zhang

Triple-negative breast cancer (TNBC) treatment with traditional phototherapeutics generally suffers from short light penetration depth and limitations of the hypoxic tumor microenvironment. Developing effective therapeutic modalities tailored to TNBC’s features remains highly desirable yet challenging. Herein, we propose a near-infrared dual windows (NIR-II) photoacoustic (PA) cavitation-based strategy to amplify oxidative stress and induce metabolic disruption in TNBC cells, achieving effective tumor suppression. With a fabricated nanoagent (DTG), we integrated the oxygen-independent therapeutic effects of photomechanical damage, cavitation-promoted oxidative damage and glucose oxidase (GOx)-mediated glucose depletion. Specifically, under NIR-II pulsed laser (PL) irradiation, PA cavitation generates collapsing bubbles and shock waves, causing mechanical injury to tumor cells. Meanwhile, GOx depletes intratumoral glucose, blocking glycolysis, reducing adenosine triphosphate (ATP) production, and generating hydrogen peroxide (H2O2) that would be further converted into more toxic hydroxyl radicals (•OH) by the photocavitation to suppress the tumors effectively. In addition, the strong NIR-II PA signal produced from the DTG facilitates high-resolution deep-tissue imaging to provide precise therapeutic guidance. Thus, these findings establish the multimodal strategy as a potent candidate for TNBC management.

Self-navigated NIR-II thermophoretic liposome nanomotors for directed tumor delivery and enhanced chemotherapy

Qing Hu,Yingfei Wang,Wei Chen,Nan Feng,Keju Tao,Jie Wu,Ying Liu,Fan Zhou,Qing Hao,Daoping Xiang,Junjie Chi,Hong Liu

Nanomotors are appealing drug carriers with deep tissue penetration to overcome biological barriers. Though high motion efficiency has been achieved with NIR-II light driven nanomotors, insufficient directional control and material biocompatibility limit their broad application. Herein, leveraging commercially available liposomal doxorubicin (DOX) as a platform, we develop a magnetic-steering NIR-II light thermophoretic liposomal nanomotor (LipNM) by asymmetrically incorporating magnetic photothermal Fe 3 O 4 @Cu 9 S 8 nanoparticles into the lipid bilayer, firstly realizing directional nanoscale self-thermophoresis. The embedded Fe 3 O 4 @Cu 9 S 8 nanoparticles respond to a brief magnet stimulation for spatiotemporal steering and NIR-II irradiation with local temperature increase for self-thermophoresis of LipNMs, which further facilitate tumor tissue targeting and deep tumor penetration, respectively. The direction-controllable thermophoretic LipNMs demonstrate good biocompatibility and increase the delivery efficiency of DOX by 186% compared to passive liposomal doxorubicin. Owing to the targeted accumulation and deep tissue penetration at the tumor site, the therapeutic efficiency of LipNMs for breast cancer in vivo is as high as 94.9%, providing a promising strategy for cancer therapy.

NIR-II Light-Activated Gold Nanocapsules for CO-Assisted Photothermal–Thermodynamic Synergistic Cancer Therapy

Mengcheng Tang,Lingfeng Pan,Hairui Deng,Xianan Li,Weili Yang,Jian Ge,Han Zhang,Yao Li,Ruibo Zhao,Shibo Wang,Xiangdong Kong

Photothermal therapy (PTT) based on a second near-infrared (NIR-II, 1000–1700 nm) light source holds great promise in the field of tumor treatment. However, hyperthermia-induced heat shock protein (HSP) expression and limited therapeutic efficacy remain the major challenges for PTT. Herein, a NIR-II laser-responsive nanocapsule PC/AC@GR@HA (abbreviated as P/A@G@H) was designed for carbon oxide (CO)-assisted tumor PTT combined with thermodynamic therapy (TDT). P/A@G@H was constructed by loading the CO storage polymer mPEG(CO) (PC) along with the free radical initiator 4,4′-azobis(4-cyanovaleric acid) (AC) into a gold hollow nanorod (GR), followed by the surface coating of hyaluronic acid (HA). After accumulating in tumor tissue through the active targeting of HA, P/A@G@H can quickly cause local hyperthermia upon 1064 nm laser irradiation and lead to cell damage by GR-mediated PTT. Moreover, the loaded PC can react with in situ H2O2 to produce CO, which inhibits the PTT-induced HSP90 expression. This can effectively weaken the heat resistance of tumor cells and enhance the PTT effect. Additionally, as a thermally decomposable radical initiator, AC can turn to toxic free radicals to ablate tumor cells via TDT under hyperthermia. Our research demonstrates that the synergistic strategy combining CO-assistance and TDT can effectively augment the ultimate therapeutic outcome of PTT, and the integrated nanocapsule P/A@G@H could be a powerful weapon for tumor suppression based on NIR-II light.

High-Performance NIR-II Sn-Based Perovskite LEDs Enabled by the Functionalization of Sulfaguanidine

Xiuling Li,Yuechuan Bi,Xiang Guan,Jiaqi Wang,Jinli Liu,Yingjie Zhao,Chunli Zhao,Zhuojian Li,Shurui Chi,Junnan Wang,Renjing Chen,Zixun Zhao,Zemin Wang,Xin Tong,Ninggui Ma,Han Liu,Zhiming Wang,Zongwen Liu,Yang Ren,Zhanhua Wei,Kebin Lin

Eco-friendly CsSnI 3 -based perovskite light-emitting diodes (PeLEDs) with the emission peak extending to the near-infrared-II (NIR-II) region hold tremendous potential for applications in biological monitoring, night vision, and optical communications. However, the various defects caused by the instability of Sn 2+ within the CsSnI 3 film, and the non-equilibrium carrier injection rate in the devices are the two major factors that limit the device performance. Here, we report high-performance NIR-II CsSnI 3 -based PeLEDs achieved by employing functional sulfaguanidine (SG) molecules. Due to the strong S═O─Sn coordination bonds, NH 2 ···I − hydrogen bonds could be formed between CsSnI 3 and SG, which effectively eliminated various Sn(II)-related defects in the CsSnI 3 film. Moreover, the SG molecules significantly improved the morphological uniformity and regulated the carrier injection balance, resulting in more balanced carrier injection that enhanced the effective radiation recombination rate. Consequently, the optimized CsSnI 3 -SG PeLEDs achieved a breakthrough external quantum efficiency (EQE) of 8.1%, which is the highest efficiency reported for CsSnI 3 -based PeLEDs to date.

Erbium-based hydrothermal YOF with stark sublevels for in vivo NIR II luminescence nanothermometer

Zhenghao Liu,Ziyue Ju,Hanyu Liu,Zhan Wang,Ruichan Lv

Rare earth luminescent materials have attracted extensive attention in the biomedical field as noncontact temperature monitoring devices with microscopic resolution due to their properties in the visible and near-infrared regions. At the application level, it is required to have a certain temperature monitoring capability in the near-infrared region II window to enhance the tissue penetration depth. Here, two kinds of YOFs: Er 3 + , Yb 3 + were prepared by co-precipitation and hydrothermal method, and the luminescence was enhanced by ion doping. An Er 3 + -based ratiometric nanothermometer of 4 F 9 ∕ 2 → 4 I 1 5 ∕ 2 (672 nm, upconversion luminescence) to 4 I 1 3 ∕ 2 → 4 I 1 5 ∕ 2 (1580 nm/1566 nm, NIR II downshifting luminescence) were designed with the Stark energy level. When doped with 2% Zn 2 + , the relative temperature sensitivity of YOF prepared by co-precipitation method was improved from 0.30% ∘ C − 1 (30 ∘ C) to 0.59% ∘ C − 1 (30 ∘ C), expanding its use as a temperature monitoring device possibility. The temperature sensitivity of YOF prepared by hydrothermal method was 1.01% ∘ C − 1 (30 ∘ C). Finally, the NIR II luminescence of the prepared nanothermometer was used as a control for temperature monitoring of heating sites in mice. The results showed that it can distinguish heating site from control site and no significant cytotoxicity or damage to the tissues was revealed, indicating its broad prospects in the biomedical field and other temperature monitoring scenarios in the future.

An ultra-small organic dye nanocluster for enhancing NIR-II imaging-guided surgery outcomes

Wang Yajun,Zhou Ding,Ma Huilong,Liu Dahai,Liang Yongye,Zhu Shoujun

Purpose The accuracy of surgery for patients with solid tumors can be greatly improved through fluorescence-guided surgery (FGS). However, existing FGS technologies have limitations due to their low penetration depth and sensitivity/selectivity, which are particularly prevalent in the relatively short imaging window (< 900 nm). A solution to these issues is near-infrared-II (NIR-II) FGS, which benefits from low autofluorescence and scattering under the long imaging window (> 900 nm). However, the inherent self-assembly of organic dyes has led to high accumulation in main organs, resulting in significant background signals and potential long-term toxicity. Methods We rationalize the donor structure of donor–acceptor-donor-based dyes to control the self-assembly process to form an ultra-small dye nanocluster, thus facilitating renal excretion and minimizing background signals. Results Our dye nanocluster can not only show clear vessel imaging, tumor and tumor sentinel lymph nodes definition, but also achieve high-performance NIR-II imaging-guided surgery of tumor-positive sentinel lymph nodes. Conclusion In summary, our study demonstrates that the dye nanocluster-based NIR-II FGS has substantially improved outcomes for radical lymphadenectomy.

Novel ultrasmall multifunctional nanodots for dual-modal MR/NIR-II imaging-guided photothermal therapy

Fenfen Li,Tuanwei Li,Debo Zhi,Pengping Xu,Wenshen Wang,Yi Hu,Yuanyuan Zhang,Shaozhen Wang,J. Matula Thomas,J. Beauchamp Norman,Weiping Ding,Lifeng Yan,Bensheng Qiu

Encouraging progress in multifunctional nanotheranostic agents that combine photothermal therapy (PTT) and different imaging modalities has been made. However, rational designed and biocompatible multifunctional agents that suitfable for in vivo application is highly desired but still challenging. In this work, we rationally designed novel ultrasmall multifunctional nanodots (FS-GdNDs) by combining the bovine serum albumin (BSA)-based gadolinium oxide nanodots (GdNDs) obtained through a biomineralization process with a small-molecule NIR-II fluorophore (FS). The as-prepared FS-GdNDs with an ultrasmall hydrodynamic diameter of 9.3 nm exhibited prominent NIR-II fluorescence properties, high longitudinal relaxivity (10.11 mM −1  s −1 ), and outstanding photothermal conversion efficiency (43.99%) and photothermal stability. In vivo studies showed that the FS-GdNDs with enhanced multifunctional characteristics diaplayed satisfactory dual-modal MR/NIR-II imaging performance with a quite low dose. The imaging-guided PTT achieved successful ablation of tumors and effectively extended the survival of mice. Cytotoxicity studies and histological assay demonstrated excellent biocompatibility of the nanodots. Importantly, this novel FS-GdNDs can undergo efficient body clearance through both hepatobiliary and renal excretion pathways. The novel ultrasmall multifunctional FS-GdNDs with excellent features hold tremendous potential in biomedical and clinical applications.

Engineered Microbial Nanohybrids for Tumor-Mediated NIR II Photothermal Enhanced Ferroptosis/Cuproptosis and Immunotherapy

Yihang Ruan,Huilan Zhuang,Xuemei Zeng,Lili Lin,Xuechun Wang,Panpan Xue,Shan Xu,Qi Chen,Shuangqian Yan,Wei Huang

The colon tumor microenvironment has a high concentration of H 2 S and glutathione, which is highly immunosuppressive and adverse to multiple therapeutic methodologies such as ferroptosis. Here, an engineered microbial nanohybrid based on Escherichia coli ( E. coli ) and Cu 2 O nanoparticles to specific colon tumor therapy and immunosuppression reversion is reported. The as-prepared E. coli @Cu 2 O hybrid can accumulate in tumor sites upon intravenous injection, and Cu 2 O nanoparticles convert to Cu x S by consuming the endogenous H 2 S, which exhibits strong photothermal conversion at near-infrared II (NIR II) biological window. Furthermore, E. coli @Cu 2 O is able to induce cellular ferroptosis and cuproptosis through inactivation of glutathione peroxidase 4 and aggregation of dihydrolipoamide S-acetyltransferase, respectively. Photothermal-enhanced ferroptosis/cuproptosis achieved by E. coli @Cu 2 O reverses the immunosuppression of colon tumors by triggering dendritic cell maturation (about 30%) and T cell activation (about 50% CD8 + T cells). Concerted with immune checkpoint blockade, the engineered microbial nanohybrid can inhibit the growth of abscopal tumors upon NIR illumination. Overall, the designed microbial nanohybrid can achieve tumor-specific photothermal-enhanced ferroptosis/cuproptosis and immunosuppression reversion, showing promise in precise tumor therapy in future clinical translation.

In Situ Polymerization of Aniline Derivative in Vivo for NIR-II Phototheranostics of Tumor

Aihui Wang,Hongyan Li,Hao Feng,Huimin Qiu,Rimei Huang,Yiqin Wang,Shichen Ji,Hong Liang,Xing-Can Shen,Bang-Ping Jiang

Natural biopolymers can be controllably in situ synthesized in organisms and play important roles in biological activities. Inspired by this, the manipulation of in situ biosynthesis of functional polymers in vivo will be an important way to obtain materials for meeting biological requirements. Herein, in situ biosynthesis of functional conjugated polymer at the tumor site was achieved via the utilization of specific tumor microenvironment (TME) characteristics for the first time. Specially, a water-soluble aniline dimer derivative (N-(3-sulfopropyl) p-aminodiphenylamine, SPA) was artfully in situ polymerized into polySPA (PSPA) nanoparticles at the tumor site, which was activated via the catalysis of hydrogen peroxide (H2O2) overexpressed in TME to produce hydroxyl radical (•OH) by coinjected horseradish peroxidase (HRP). Benefiting from outstanding near-infrared (NIR)-II absorption of PSPA, the in situ polymerization process can be validly monitored by photoacoustic (PA) signal at the NIR-II region. Meanwhile, in situ polymerization would induce the size of polymeric materials from small to large, improving the distribution and retention of PSPA at the tumor site. On the combination of NIR-II absorption of PSPA and the size variation induced by polymerization, such polymerization can be applied for tumor-specific NIR-II light mediated PA image and photothermal inhibition of tumors, enhancing the precision and efficacy of tumor phototheranostics. Therefore, the present work opens the way to manipulate TME-activated in situ biosynthesis of functional conjugated polymer at the tumor site for overcoming formidable challenges in tumor theranostics.

NIR-II organic small molecule probe for labeling lymph nodes and guiding tumor imaging

Lin Yuan,Yingbin Su,Runfeng Zhang,Jie Gao,Bing Yu,Hailin Cong,Youqing Shen

Organic small molecule fluorescent groups have injected new material support into the field of medical imaging due to their unique luminescence mechanism and easy tuning of structure. The great potential of NIR-II window imaging forces us to continuously optimize the structure of organic fluorophores to design better fluorescent molecules for fluorescence imaging-guided surgery. An ideal organic small molecule fluorescent group: it can penetrate into the inside of the organism, clearly present the internal structure and the edge contour of different tissues, so as to perfectly achieve internal imaging and accurately guide external surgery. In vivo, fluorescent groups do not damage normal tissues and organs. However, problems such as low quantum yield and poor biocompatibility greatly limit the clinical transformation of NIR-II fluorescent small molecules. To avoid the shortcomings of NIR-II fluorescent probes as much as possible and better realize image-guided surgery, in this experiment, the biplane donor unit was incorporated into the twisted D-π-A-π-D structure to expand the conjugated structure of the fluorescent group, which not only realized NIR-II emission, but also had high quantum yield and biosafety.
