A general strategy for controllable synthesis of Ba3(MO4)2:Mn5+ (M = V, P) nanoparticles

Xiaowen Zhang,Yang Li,Zhongliang Hu,Zhi Chen,Jianrong Qiu

The second near-infrared window (NIR-II, 1000–1400 nm) is ideal for fluorescence imaging owing to negligible tissue scattering and minimal autofluorescence. NIR-II luminescence nanophosphors are promising materials for fluorescence imaging. Mn5+ is emerging as a new luminescent indicator owing to its typical NIR-I (800 nm) to NIR-II (1190 nm) downconversion luminescence. Herein, we report a facile synthesis approach to prepare Mn5+ doped Ba3(MO4)2 (M = V, P) nanoparticles with a remarkably sharp emission peak around 1190 nm in the NIR-II region for the first time to our knowledge. The developed two-step approach based on efficient anion exchange reaction is able to not only stabilize the valence but also keep the controlled morphology and uniform size. It is highly possible that the general approach will provide a new perspective on synthesizing special valence ion doped nanosized materials.

Ag-Doped SnSe Nanosheets for Enhanced NIR-II Light-Induced Apoptosis

Dequan Xiao,Yan Gong,Jing Zhang,Hui Liu,Lile Dong

Triple-negative breast cancer (TNBC) is one of the most aggressive and challenging subtypes with limited treatment options and poor prognosis. To overcome these obstacles, Ag-doped SnSe (ASS) nanosheets were used as photonic hyperthermia agents for treating TNBC. ASS featured a high photothermal-conversion efficiency and photon hyperthermia effect. Additionally, biosafety assessments, including hemolysis assays and histopathological analysis, confirmed that ASS was biosafety. These findings suggest that ASS, in synergy with the second near-infrared biowindow (NIR-II) light irradiation, offers a noninvasive, effective, and safe therapeutic strategy for TNBC. This innovative approach exemplifies the evolving landscape of TNBC treatment, where technology and biology converge to offer more precise and personalized therapies.

Engineered manganese-BODIPY coordinated nanoadjuvants for enhanced NIR-II photo-metalloimmunotherapy

Yushi Zhang,Xuya Yu,Lingpeng Luo,Yigang Xu,Hanjie Zhang,Zhuo Mao,Yuqin Zhang,Cuihong Yang,Lu Wang,Pengcheng Zhang,Sitong Li,Meitong Ou,Ran Luo,Dunwan Zhu,Wen Li,Lin Mei

Immunotherapy, a pivotal and promising approach for tumor treatment, has demonstrated prominent clinical efficacy. However, its effectiveness is often impeded by insufficient antitumor immune responses attributed to the immunosuppressive tumor microenvironment (TME). The combination of immune activation through the stimulator of interferon genes (STING) pathway and phototherapy holds great potential for surmounting this challenge in advanced tumor immunotherapy. Herein, a novel manganese-boosted NIR-II photo-metalloimmunotherapy is proposed to synergistically enhance antitumour efficacy by fabricating Mn 2+ -BODIPY-based coordinated photo-immune nanoadjuvants (BMR), modified with tumor-targeted peptide cRGD. The obtained BMR could effectively deliver Mn 2+ to tumor sites, and immunogenic cell death (ICD) was evoked by localized photothermal ablation of tumors using NIR-II laser irradiation. Simultaneously, pH-responsive release of Mn 2+ would trigger the activation of STING pathway to promote the production of type I interferons (I-IFNs), significantly facilitating the maturation of dendritic cells (DCs) and polarization of macrophages to M1 phenotypes. Furthermore, by synergistically initiating systematic and robust antitumour immune responses, the BMR-mediated NIR-II photo-metalloimmunotherapy achieved remarkable therapeutic efficacy against both primary and lung metastasis of B16F10 tumors. Overall, in light of the versatile functionalities and synthetic flexibility of coordinated nanoadjuvants, formulated with photofunctional ligands and diverse metal ions, this work provides new insights into the design of metal coordination nanomedicine for effective antitumor photo-metalloimmunotherapy.

NIR-II responsive copper-nitrogen doped photocatalysts for pyroptosis and cuproptosis

Qinglin Wei,Xuejiao Song,Xiaorui Wang,Yawen Chen,Wenjun Wang,Ping Sun,Xiaochen Dong

Based on their distinctive manners of driving cell death, cuproptosis and pyroptosis exhibit significant potential in overcoming the apoptosis resistance of cancer cells. Nevertheless, limited copper concentrations and complex tumor microenvironment (TME) severely constrain the therapeutic efficacy. Herein, DSPE-PEG 2000 modified copper-nitrogen-doped carbon-based photocatalysts (Cu-NC@PEG) are developed as photo-activated reactive oxygen species (ROS) amplification systems to trigger pyroptosis and cuproptosis in a synergistic manner. Cu-NC@PEG enriches in tumor cells and generates a large amount of superoxide anions under the second near-infrared region (NIR-II) laser irradiation, followed by further induction of cell pyroptosis through caspase-mediated cleavage of Gasdermin D (GSDMD). The copper ions released from Cu-NC@PEG in the acidic TME can not only generate ·OH and consume the antioxidants to enhance ROS-induced pyroptosis, but also effectively activate cuproptosis through Cu + -mediated dihydrolipoamide S-acetyltransferase aggregation and Fe-S cluster protein loss. Simultaneously, the mitochondrial damage induced by pyroptosis leads to a considerable efflux of ATP and the restricted ATP availability could enhance the disruption of copper metabolic homeostasis, thereby potentiating cuproptosis. The increased tumor-inhibiting effects of the combined induction of cuproptosis and pyroptosis could be achieved. Statement of significance 1. NIR-II photo-activated copper-nitrogen-doped carbon-based photocatalysts (Cu-NC@PEG) were developed. 2. ROS amplification was achieved based on the photo catalytic properties and Fenton-like reaction of Cu-NC@PEG. 3. Therapeutic efficacy was enhanced via ROS-induced pyroptosis and Cu + -mediated cuproptosis.

NIR-II light activated photodynamic therapy with protein-capped gold nanoclusters

Chen Qian,Chen Jiawen,Yang Zhijuan,Zhang Lin,Dong Ziliang,Liu Zhuang

The use of near-infrared (NIR) light for photodynamic therapy (PDT) is a promising strategy to circumvent the limitations of current PDT, in which visible light with limited tissue penetration depth is usually used. In the present study, alkyl thiolated gold nanoclusters (AuNCs) were co-modified with human serum albumin (HSA) and catalase (CAT), and then employed as a multifunctional, optical, theranostic nano-agent. In the AuNC@HSA/CAT system, the AuNCs were able to produce singlet oxygen under excitation by a 1,064-nm laser, which locates in the second NIR window (NIR-II), and featured much lower tissue absorption and scattering, enabling NIR-II-triggered PDT. The HSA coating greatly improved the physiological stability of the nanoparticles, which showed efficient tumor retention after intravenous injection, as revealed by detecting the AuNC fluorescence. Moreover, the presence of CAT in the nanoparticles triggered decomposition of tumor endogenous H 2 O 2 to generate oxygen, thereby enhancing the efficacy of PDT by relieving tumor hypoxia. Compared with conventional PDT using visible light, NIR-II-triggered PDT exhibits remarkably increased tissue penetration. Thus, we developed a new type of photosensitizing nano-agent that simultaneously enables in vivo fluorescence imaging, tumor hypoxia relief, and NIR-II light-induced in vivo PDT in the treatment of cancer.

Resonant Strategy in Designing NIR-II SERS Nanotags: A Quantitative Study

Haoqi He,Yumin Zhang,Shuo Zhu,Jian Ye,Li Lin

Surface-enhanced Raman scattering (SERS) nanotags have garnered much attention for in vivo and intraoperative imaging due to their high sensitivity and intrinsic fingerprint spectrum. Recently, in vivo bioimaging at the second biological near-infrared (NIR-II) window has demonstrated significant advances with better spatial resolution and deeper tissue penetration. The development of SERS nanotags at the NIR-II window are therefore highly desired. The resonant strategy, i.e., the use of resonant substrates and resonant molecules, can be helpful in fabricating bright SERS nanotags at the NIR-II window; however, this strategy has not been demonstrated with the lack of quantitative assessments. Here we report a quantitative study on the Raman enhancements of the resonant strategy in preparing NIR-II SERS nanotags. By comparing the resonant and nonresonant plasmonic nanorod substrates, the resonant substrates have been shown to provide an enhancement factor of up to 4 orders of magnitude. When using the resonant Raman reporter molecules, SERS intensities can be further increased by 25–546 times. This work demonstrates the potential of the resonant strategy in fabricating ultrasensitive NIR-II SERS nanotags and provides insights into the design of related plasmonic devices.

Curcumin-enhanced NIR-II-responsive gold nanobipyramids for targeted HSP 90 inhibition

Zhenying Diao,Youcheng Liang,Yong Liu,Dou Zhang,Long Qiu,Jianbo Sun,Qiaoyou Lu,Yanlei Liu,Daxiang Cui,Ting Yin

Blockade of heat shock protein 90 (HSP90) expression in multimodal synergistic therapy has a great prospect for cancer treatment. Nanomaterials combined with bioinformatic analysis provides accurate guidance for the design of anti-HSP90 nanomedicines. Herein, a NIR-II-responsive nanoplatform was developed under bioinformatics guided to effectly inhibit HSP90 for enhanced synergistic mild-photothermal chemotherapy without any notable tissue damage. The nanoplatforms were assembled from NIR-II-responsive gold nanobipyramids (GNBs) combined with curcumin (Cur) via hydrophobic-hydrophobic interactions and hydrogen bonds. On the basis of drug discovery and network pharmacology, we found that Cur has impressive anti-HSP90 capability and analyzed its therapeutic mechanism against NSCLC. Under the irradiation of NIR-II light, the obtained GNBs-Cur blocked the expression of HPS90 and inhibited related antiapoptotic pathways, thus enhancing the mild PTT of GNBs under 1064 nm laser irradiation. Meanwhile, Cur served as chemotherapeutic agents to induce apoptosis in tumor cells. In vivo photoacoustic imaging-guided, GNBs-Cur achieved effective tumor elimination through mild-photothermal chemotherapy without systemic toxicity. Overall, this work provides a new therapeutic modality paradigm for potential NSCLC treatment on the basis of synergistic therapies.

NIR-II responsive PEGylated nickel nanoclusters for photothermal enhanced chemodynamic synergistic oncotherapy

Yong Qian,Jiahui Zhang,Jinglu Zou,Xingyu Wang,Xiangfu Meng,Hongji Liu,Yefeng Lin,Qianwang Chen,Lei Sun,Wenchu Lin,Hui Wang

Rationale: All kinds of non-metal and metal-based nanozymes have been extensively explored as Fenton agents for Chemodynamic therapy (CDT). However, the low catalytic efficiency of non-metallic nanozymes and the susceptibility to oxidation and long-term toxicity of metallo-nanozymes limit their potential in CDT. Methods: In this study, we report a magneto-solvothermal method to tune the crystallinity and shape of polyethylene glycol (PEG)-ylated urchin-like nickel nanoclusters (named as 9T-PUNNC) at a high magnetic field with an intensity of 9 T for enhanced combined photothermal-chemodynamic therapy. Results: The needle-like protrusions on the surface of 9T-PUNNC can effectively increase the reception of NIR light in second NIR window (NIR-II) and transform it into local hyperthermia, achieving effective photothermal treatment. The light and heat generated by NIR-II further promotes the release of Ni 2+ and improves the ability of Ni 2+ -mediated chemodynamic therapy (CDT). In addition, the surface coating of PEG on the surface of 9T-PUNNC improves its stability and biocompatibility of nanocrystals. In vitro and in vivo results indicate that the 9T-PUNNC could efficiently kill tumor cells (nearly 12 times more than control group) and inhibit tumor growth (nearly 9 times smaller than control group) under NIR-II irradiation through the synergistic effect of combined treatments. Conclusions: we developed a novel synthetic strategy to tune crystallinity and shape of PUNNC for enhanced NIR-II responsive photothermal conversion efficiency and accelerated acid-induced dissolution for improved ·OH generation. Such 9T-PUNNC enable a combined chemodynamic-photothermal treatment to provide superior therapeutic efficacy due to their highly synergistic effect.

NIR-II photoresponsive antibacterial coatings on titanium implants using strontium titanate metastructures

Feiyang Hong,Honglian Wang,Xiaoyi Ma,Minggang Yang,Haitao Zhang,Yang Ma,Kai Yang,Xiaobing Zhao,Guocheng Wang

Implant-associated infections present a significant clinical challenge, exacerbated by the increasing prevalence of antibiotic resistance associated with conventional antibiotic therapies. Photoresponsive antibacterial strategies offer a promising alternative, but their effectiveness in treating deep tissue infections is often limited by the short wavelengths required for activation of most photosensitizers. This study reports the development of a strontium titanate (STO) metastructure coating on titanium alloy implants, engineered for long-wavelength photoresponsive antibacterial activity. We demonstrate that the size of the STO metastructure particles is precisely controlled by modulating the Sr(OH)₂ precursor concentration, enabling tunable optical responses within the 800 nm–1600 nm wavelength range. The resulting coating exhibits excellent antibacterial efficacy upon irradiation with second near-infrared window (NIR-II) light (1064 nm), while demonstrating good biocompatibility and minimal damage to healthy cells after NIR exposure.

NIR-II Light-Activated, ROS-Amplifying Janus Nanozymes for Multimodal Synergistic Therapy

Lin Fan,Shao Yong,Zhang Yuanqing

Nanozyme-catalyzed reactive oxygen species (ROS) generation has emerged as a promising strategy for advanced ROS-based therapeutics. However, achieving efficient ROS production with favorable catalytic kinetics in a confined microenvironment remains challenging. Herein, a cancer-targeting heterogeneous Janus nanozyme (JN, Cu 2– x Se–Au-TLS11a) was rationally fabricated via a facile modular synthesis and assembly. Upon near-infrared II (NIR-II) light irradiation, the well-designed JN realized multimodal synergistic anticancer effects via three coordinated mechanisms: (i) efficient photothermal conversion with a high efficiency of 61.9% for photothermal therapy (PTT); (ii) NIR-II-triggered hot-electron transfer from the Au domain to Cu 2– x Se promoted the generation of Cu + , which accelerated Fenton-like reactions to produce hydroxyl radicals (•OH); and (iii) NIR-II photoexcitation induced the formation of electron–hole (e – -h + ) pairs on the Janus nanostructure, enabling additional photocatalytic •OH generation. Under NIR-II light activation, the JNs, integrating photothermal, photoenhanced chemodynamic, and photocatalytic effects, demonstrated highly specific and potent in vitro anticancer efficacy, achieving an IC 50 value of approximately 12.5 μg mL –1 against HepG2 cells and reducing cell viability to 13.8%, thereby providing an innovative paradigm for designing spatiotemporally controlled, ROS-amplifying nanoplatforms toward precision cancer therapy.

NIR-II Light-Activated Nanoplatform for Synergistic Photothermal and H2S-Mediated Thrombolysis

Lei Chen,Jianwen Song,Zhenya Shen,Ji Qi

Occlusive thrombi within blood vessels are a primary cause of life-threatening cardiovascular diseases, which continue to pose significant global health challenges. Here, we report a polymer nanoplatform that integrates near-infrared II (NIR-II) light-triggered photothermal thrombolysis with intrinsic antithrombotic functionality. A novel semiconducting polymer, featuring intense NIR-II absorption and exceptionally high photothermal conversion, is rationally engineered to enable rapid and localized light-to-heat thrombolysis. A thermosensitive hydrogen sulfide (H 2 S) donor is also incorporated, generating multifunctional “nanosweepers” that combine synergistic photothermal and gas-mediated thrombolytic activity. The formulation is further functionalized with bis-serotonin groups, which confer myeloperoxidase- and hydrogen peroxide-responsive thrombus-targeting capability, ensuring selective accumulation and site-specific therapy. This agent possesses robust NIR-II absorption, high photothermal conversion efficiency (77.2%), and pronounced thrombus-specific enrichment. Importantly, the combination of localized photothermal heating and on-demand H 2 S release enable rapid, near-complete thrombolysis in both carotid and lower-limb arterial thrombosis models, achieving almost full restoration of blood flow. This work establishes a versatile and highly efficient strategy that integrates precise NIR-II photothermal conversion with H 2 S-mediated redox regulation, providing a promising platform for site-specific multimodal synergistic treatment of life-threatening thrombotic disorders. The findings offer new insights into the design of multifunctional, precision therapeutics for vascular disease management.

Xinyao Dong,Na Zhou,Minfeng Tian,Yanan Ji,Jingxuan Wang,Jianing Fan,Heyang Li,Wen Xu,Ping Duan,Xiumei Yin

Rare-earth doped oxysulfides have gained considerable attention as a luminescence host material owing to its good chemical stability and high efficiency. In this study, Er3+-sensitized La2O2S nanoparticles are prepared by the high-temperature co-precipitation method. La2O2S:2%Er3+ is selected to construct La2O2S:Er3+@La2O2S core/shell structures through epitaxial growth, and the luminescence properties are studied. The surface defects of La2O2S:Er3+ are repaired by the inert protective shell, and the luminescence intensity of the core/shell nanocrystals is increased by 9.6 times. Finally, MAPbI3 is spin-coated over the La2O2S:Er3+@La2O2S film to construct narrow band near-infrared photodetectors. Under 1550 nm excitation, the response rate is 23 mA W−1, detection rate is 2.57 × 108 Jones, and response time is 220 ms.
