Fluorinated Poly(ε-caprolactone) (PCL) block copolymers PCL-PTFOA(2 h), PCL-PTFOA(4 h), PCL-PTFOA(6 h), PCL-PTFOA(8 h) were prepared via esterification by using poly(1H,1H,2H,2H-perfluorooctylacrylate) (PTFOA) with different molecular weights as reactants. It was found that PCL-PTFOA(6 h) and PCL-PTFOA(8 h) possessed the similar molecular weight via GPC analysis. PCL and fluorinated PCL block copolymers films were prepared by solution casting method. The structure and properties of film surface of the fluorinated PCL block copolymers were analyzed by energy dispersive X-ray spectrometer (EDS), scanning electron microscope (SEM), Differential scanning calorimetry (DSC), water contact angles (WCAs) and the surface free energy. The results showed that the fluorinated PCL films possessed improved hydrophobicity because of the microphase separation between fluoropolymer chain and PCL chain. The enzymatic degradation of PCL, PCL-PTFOA(2 h), PCL-PTFOA(4 h) and PCL-PTFOA(6 h) films were performed at 37 °C in a phosphate buffer solution (PBS, 0.1 M, with a pH of 7.2–7.4) with lipase and analyzed by weight loss, SEM and EDS . It was found that the introduction of fluorine-containing segment reduced the rate of enzymatic degradation of the copolymer film in the early period, while increased the finally weight loss rate with a prolonged time. In addition, the enzymatic degradation rate of PCL-PTFOA(2 h) film and PCL-PTFOA(4 h) film was faster than that of PCL-PTFOA(6 h) film.

Composite polymer devices have gained considerable interest in biomedical applications. In this work, a composite poly(lactic-co-glycolic acid)/poly(ε-caprolactone) (PLGA/PCL) microsphere with a layered architecture and an island–sea topography was developed. PCL was found to occupy the shell layer of the microsphere, while PLGA constituted surface islands and the internal core. This subtle structure was derived from the different external and internal rates of phase separation between PCL and PLGA. The existence of the PLGA islands enhanced the hydrophilicity of the PCL shell. Mouse mesenchymal stem cells (mMSCs) were cultured and they grew well on the PLGA/PCL microspheres. The surface of the microsphere was microscopically composed of PCL sea and PLGA islands, and the mMSCs showed a trend to attach to the more hydrophilic PLGA islands and bridged between adjacent microspheres. This paper provides a new alternative to design functional microspheres for potential application in tissue repair.

In this study, a composite drug delivery system was developed and evaluated for oral delivery of docetaxel: docetaxel-loaded micelles in pH-responsive hydrogel (DTX-micelle–hydrogel). Docetaxel was successfully loaded in micelles with small particle size of 20 nm and high drug loading of 7.76%, which contributed to the drug absorption in the intestinal tract. The experiments of cytotoxicity on 4T1 cells demonstrated the effective antitumor activity of DTX micelles. Meanwhile, a pH-responsive hydrogel was synthesized and optimized for incorporating the docetaxel micelles. The pH-responsiveness and reversibility of the hydrogel were investigated under the pH conditions of the gastrointestinal tract . Furthermore, the DTX-micelle–hydrogel system showed much quicker diffusion of micelles in simulated intestinal fluid than in simulated gastric fluid, which was mainly caused by the change of pH value. The docetaxel released from the micelle–hydrogel system quite slowly, so it had little influence on the absorption of DTX micelles in small intestine . More important, the pharmacokinetic study revealed that the DTX-micelle–hydrogel significantly improved the oral bioavailability of docetaxel (75.6%) about 10 times compared to DTX micelles, and this increase in bioavailability was probably due to the small intestine targeting release of the pH-responsive hydrogel. Consequently, the oral DTX-micelle–hydrogel system was effective in inhibiting tumor growth in subcutaneous 4T1 breast cancer model, and decreased systemic toxicity compared with intravenous treatment. The apoptosis cells in the immunofluorescent studies and the proliferation-positive cells in the immunohistochemical studies were also consistent with the results. Therefore, the DTX-micelle–hydrogel system might be a promising candidate oral drug for breast cancer therapy.

The rapid degradation of magnesium-based implants in physiological environments in vivo not only will quickly deteriorate their mechanical strengths but will also lead to a severe change of the micro-environment around the implants, which may cause the final failure of magnesium-based implants. In this work, a polycaprolactone (PCL) layer was prepared to seal the plasma electrolytic oxidization coating (PEO) to form a PEO/PCL composite coating on a biodegradable AZ31 magnesium alloy, followed by further surface functionalization with polydopamine. The in vitro degradation behaviors of the bare AZ31 alloy and coated samples were evaluated in a simulated body fluid (SBF) using the potentiodynamic polarization curve test and the static immersion test. The bioactivity of the samples was investigated using the SBF soaking test. The cytocompatibility of all samples was evaluated using the cytotoxicity test and analysis of the adhesion and proliferation of osteoblast cells (MC3T3-E1) directly cultivated on the sample surface. The results showed that the PCL layer successfully sealed the pores of the PEO coating, and then the polydopamine layer formed on its surface. The in vitro degradation tests showed that the PEO/PCL composite coating improved the corrosion resistance of the AZ31 alloy in SBF with a more positive corrosion potential and a lower corrosion current density. Due to the protection of the PEO/PCL composite coating, the surrounding environment showed nearly no influence on the degradation of the coated sample, which led to no obvious local alkalization and hydrogen evolution. Moreover, compared with the AZ31 alloy and PEO coating, the PEO/PCL composite coating was more suitable for cell adhesion and proliferation. After further surface functionalization by polydopamine, the corrosion resistance of the composite coating was maintained, while its bioactivity was significantly enhanced with a large amount of hydroxyapatite (HA) formed on its surface after immersion in SBF. The initial cell adhesion and spread were also improved by the polydopamine. By further immobilizing polyhexamethylene biguanidine (PHMB) onto the coating surface via the assistance of polydopamine, good antibacterial ability was obtained. This feasible method for fabricating a cytocompatible and antibacterial composite coating on a biodegradable AZ31 alloy may be promising in implant applications due to the osteointegration and anti-infection properties of these materials post operation.

Electrospun biodegradable fiber mesh is a promising alternative scaffold for delivering progenitor cells for repairing damaged or diseased tissue, but its cripple mechanical stability has not met the requirement of tissue engineering yet. In this work, the well-defined poly(ε-caprolactone)-branched poly(methyl methacrylate- co -hydroxyethylmethacrylate) (PCL-PMH) has been successfully synthesized to toughen electrospun poly( l -lactide) (PLLA) fiber membrane. Characterization of the obtained nanofibrous meshes indicates that PCL-PMH and PLLA can be well blended to make smooth fibers, and fibrous diameter vary little with blending PCL-PMH. The aggregation state of two macromolecules is closely correlated with blend ratio, molecular structure, and molecular weight of PCL-PMH, and only when PCL-PMH and PLLA form good interfacial adhesion can PMH give full play to its potential for toughening the fiber membrane. The tensile strength and elongation at break of the blend are 6.20 MPa and 63.40% under the optimal conditions, respectively, and it also exhibits the representative feature of toughness materials. The blending fiber membrane is as no cytotoxic as original PLLA. This work will provide a new way for toughness of electrospun fiber membrane in practice.

In the study, a specific material system that contains poly-(ε-caprolactone) (PCL), polyvinyl acetate (PVAc) and hydroxyapatite (HA) was used to fabricate porous scaffolds employing a 3D printing technique for bone regeneration. Four groups of 3D printing scaffolds were fabricated: PCL, PCL/PVAc, PCL/HA and PCL/PVAc/HA for comparision. The morphologies, mechanical properties and biological characteristics of these scaffolds were analyzed using SEM, a material testing machine, in vitro cell culture and in vivo animal experiments. The results showed that these 3D printed scaffolds possessed porous channel structures with a hole size of 375–475 μm and porosity of 74.1–76.1%. The compressive moduli of the scaffolds increased with the addition of HA and decreased with the addition of PVAc. The PCL/PVAc/HA scaffold exhibited higher cell proliferation and bone formation rates than other groups (p < 0.001), which could be attributed to the synergistic effect of PAVc and HA components. Two types of new bone formation patterns in the scaffold were found in this study: one is the new bone formed directly on the grid matrix, and the other is the new bone initially formed in the center of the scaffold channel and then remolded to concentric circles. The osteogenesis pattern of the latter is analogous to the osteon structure of a cortical bone. The 3D printed scaffold based on PCL/PVAc/HA tri-component system is a promising prospect for future individualized bone repair applications.

PVC as an important plastic material, has gained widespread application due to its excellent physical properties, good heat and chemical resistance, as well as low cost. By incorporating plasticizers and other plastic additives, rigid PVC can be extended to applications in the flexible PVC field. However, conventional low-molar-mass phthalate plasticizers tend to migrate from the PVC matrix to the external environment during long-term use. This migration not only deteriorates the physical properties of PVC but also poses risks to human health and the environment. Therefore, improving the stability of plasticizers in PVC materials is of great significance. In this study, two structurally different plasticizers were prepared by hydroxyl-initiated ring-opening polymerization of ε-caprolactone. The mechanical properties of PVC plasticized with Lignin-PCL, C 5 H 12 O-PCL, and the conventional plasticizer DOP were compared. The results show that PVC plasticized with Lignin-PCL exhibited improved mechanical properties compared to those plasticized with the conventional plasticizer. Interestingly, the hyperbranched plasticized PVC demonstrated higher modulus, strength, and toughness than its linear counterpart. The resulting plasticized PVC materials exhibited a high modulus of approximately 30 MPa while ensuring an elongation at break exceeding 300%. The migration rates of all Lignin-PCL-PVC samples were below 5%, which is only 0.026 times that of DOP-PVC. Moreover, as both lignin and PCL are degradable and environmentally friendly materials, the degradation performance of the materials was also investigated. The study found that the hyperbranched plasticizer Lignin-PCL could be degraded by more than 80%, which is crucial for the development of environmentally friendly flexible PVC materials.

This work aims to systematically elucidate the biodegradation and crystallization behaviors of the model hypergraft systems made of poly( ε -caprolactone) (PCL) and poly[tri(ethylene glycol) methyl ether acrylate] (PTEGMA) blocks. A series of hyperbranched PCL (HB-PCL n ) with varied subchain lengths were synthesized using AB 2 -type macromonomers with different degrees of polymerization ( DP s). The HB-PCLs were grafted with linear PTEGMA chains to prepare amphiphilic hypergraft copolymers (HB-PCL n - g -PTEGMA m ) to eventually get self-assembled structures through micellization. The DP of PTEGMA was also varied to get two sets of samples with varied sub-chain lengths of the hyperbranched core and the linear corona within each individual set to systematically study the crystallization and biodegradation behaviors of PCL. Differential scanning calorimetric (DSC) measurements of both hypergrafts and linear precursors were compared in order to get insights into the crystallization pattern of PCL. We found that the degree of crystallization in PCL can be controlled from ~0 (lowest limit) to ~52% (highest limit) by structural regulation. Biodegradation studies were performed in aqueous medium with Lipase from Pseudomonas using dynamic light scattering (DLS). A model was proposed to correlate the sub-chain length of core, the chain length of corona with the biodegradation rate of PCL, which reveals that an increase of PCL content in HB-PCL n - g -PTEGMA m increases the crystallinity while decreases the rate of biodegradation in a predictable manner.

Uncontrolled degradation of wound dressings may result in residues, causing several negative effects on wound healing, such as secondary damage, undesirable inflammation, and scar skin formation. Here, an available strategy associated with the synthesis of enzyme-loaded ( Burkholderia cepacia lipase, BCL) polycaprolactone (PCL) nanofiber scaffolds, aligning with wound healing effects is reported. These scaffolds are fabricated via fiber microfluidic electrospinning degradation-control technique. The obtained scaffolds exhibit tunable degradation rates, achieving complete degradation within 12–72-h cycles. The acidic degradation products are further elucidated and reveal the potential degradation mechanism. The acidic degradation products create an optimal microenvironment during the hemostasis and inflammation stages of wound healing. Notably, in vivo experiments demonstrate the enzyme-loaded scaffolds effectively promote angiogenesis, reduce inflammatory responses, mitigate collagen deposition, and regulate fibroblast differentiation. This promotes rapid wound healing with a remarkable scarless rate of over 99% by day 21. New guidelines for scar-free healing dressings are proposed, which carry out faster degradation without microplastics (MPs) and toxic byproducts before scar formation. These principles might provide valuable insights and promise for developing more effective wound dressings.

Wound ulceration induces inflammation, delays tissue repair, and may trigger systemic infections, thereby endangering bodily health. Hydrogel films are capable of accelerating wound healing processes. This study fabricated polyvinylpyrrolidone/polycaprolactone (PVP/PCL) nanofiber-hydrogel membranes via juxtaposed electrospinning, overcoming the limitations of traditional electrospinning techniques. Hydrophilic PVP (15% w/v in ethanol with 0.03% DAS) and hydrophobic PCL (15% w/v in chloroform/ N , N -dimethylformamide (CF/DMF) 4:1) were employed, with PVP/PCL ratios precisely regulated. The optimal VC-8/2 (PVP/PCL = 8:2) exhibited excellent performance, featuring a viscosity of 176.5 cp, a smooth, bead-free morphology prior to swelling (diameter: 718.22 ± 142.96 nm), and a twisted structure post-swelling (diameter: 268.71 ± 94.21 nm)—all enabled by optimized solvents and juxtaposed electrospinning that eliminated phase separation. Additionally, the VC-8/2 demonstrated a tensile strength of 1.16 ± 0.10 MPa, an elongation at break of 94.35% ± 5.64%, and a 40% water absorption rate after 12 h. Attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) confirmed the successful cross-linking of PVP, while CCK-8 assays revealed high L929 cell viability (87.23%–91.44%), exceeding the 85% non-cytotoxic threshold. These biocompatible membranes integrate the exudate absorption capacity of PVP and the mechanical support of PCL, rendering them highly promising for advanced wound dressing applications. Graphical Wound ulceration endangers health by inducing inflammation, delaying tissue repair, and triggering systemic infections, while hydrogel films accelerate wound healing. This study fabricated polyvinylpyrrolidone/polycaprolactone (PVP/PCL) nanofiber-hydrogel membranes via juxtaposed electrospinning (overcoming traditional electrospinning limitations) using hydrophilic PVP (15% w/v in ethanol with 0.03% DAS) and hydrophobic PCL (15% w/v in CF/DMF 4:1). The optimal VC-8/2 (PVP/PCL = 8:2) showed a smooth pre-swelling morphology (718.22 ± 142.96 nm) and twisted post-swelling structure (268.71 ± 94.21 nm), with 1.16 ± 0.10 MPa tensile strength, 94.35% ± 5.64% elongation at break, 40% 12-h water absorption, successful PVP cross-linking (ATR-FTIR), and high L929 cell viability (87.23%–91.44%). Integrating PVP's exudate absorption and PCL's mechanical support, the membranes are promising for advanced wound dressings.

Mesoporous bioactive glass (MBG) is a potential biomedical material in bone defect repairment because of its bioactivity, biocompatibility, and osteoinduction properties. Here we report that Mg-doped MBG scaffold with 3:1 Ca/Mg ratio (MBG-Ca/Mg-3) is good for MC3T3-E1 osteoblast differentiation and mineralization. Mimicking bone extracellular matrix structure by electrospinning, we used MBG-Ca/Mg-3 adsorbed with Osteocalcin-Osteopontin-Biglycan (OOB), a new unique matrix fusion protein, to form [email protected] /Mg-3 scaffold, which has multifunctional ability in calvarial bone defect repairment in vivo . Intriguingly, we found that [email protected] /Mg-3 scaffold increases the expression of osteoblastic marker genes, including bone morphogenetic protein ( Bmp2 ), osteopontin ( Opn ), Osterix , Runx2 through activation of ERK1/2. We concluded that [email protected] /Mg-3 scaffold promotes osteoblast differentiation and mineralization through ERK1/2 pathway and it can also enhance bone formation in vivo , which provides a new biomaterial in bone tissue engineering.

Burns on human skin often leads to slow recovery because of continuous wound infections caused by the loss of skin protection. In this study, a seed-free soaking method is put forward to prepare PCL@ZIF-8/εPL nanofibers as a dressing for burn wounds. This ZIF-8-coated polycaprolactone (PCL) fiber exhibits increased specific surface area, significantly increasing the polypeptide loading of epsilon polylysine (εPL) from 12 to 20 wt.%. The antibacterial experiment found that ZIF-8 and εPL exhibited dual antibacterial properties, and after εPL was loaded to ZIF-8, the nanofiber demonstrated stronger antibacterial properties that can better kill the remaining tenacious bacteria. This high-efficiency bactericidal property, combined with the ability to protect wounds from common infections by bacteria in the air, would promote wound healing, shortening the healing time period from 24 to 17 days. Further tissue sections confirm that the shielding effect and dual antibacterial properties of the composite fiber membrane are the reasons behind the acceleration of the healing of burns and scalds. Shielding and antibacterial material are helpful for the treatment of burns and scalds, and it may also guide the treatment of other types of wounds.
