Icariin-loaded electrospun PCL/gelatin nanofiber membrane as potential artificial periosteum

Min Gong,Cheng Chi,Jingjing Ye,Meihong Liao,Wenqi Xie,Chengai Wu,Rui Shi,Liqun Zhang

Due to the significant role of the periosteum in bone regeneration, we hypothesised that using a specially engineered artificial periosteum could lead to an enhancement in osteogenesis in bone grafts. Herein, we describe our work aimed at fabricating an electrospun fibrous membrane as an artificial periosteum that exhibits flexibility, permeability and osteoinduction. This membrane was designed to cover the complex surface of bone grafts to facilitate and accelerate bone regeneration. The traditional Chinese medicine icariin (ICA) was subsequently introduced into poly (ε-caprolactone) (PCL) /gelatin nanofibers to fabricate an artificial periosteum for the first time. The effects of ICA content on morphology, physical properties, drug release profile, in vitro degradability, biocompatibility and osteogenic differentiation properties were investigated. The ICA-loaded electrospun membranes showed significant improvement in hydrophilicity, high mechanical strength, appropriate degradation rates and excellent biocompatibility. Furthermore, clear enhancement in alkaline phosphatase (ALP) activity, as well as an increase in osteocalcin (OCN) and type collagen I (COL I) expression in MC3T3-E1 cells were detected. Furthermore, we observed clear calcium deposition content in MC3T3-E1 cells cultured on ICA-loaded fibrous matrix. The membrane loaded with 0.05 wt.% ICA displayed properties contributing to cell attachment, proliferation and differentiation. These results indicate the huge potential of this ICA-loaded PCL/gelatin electrospun membrane as a biomimetic artificial periosteum to accelerate bone regeneration.

PCL NGCs integrated with urolithin-A-loaded hydrogels for nerve regeneration

Xue-Han Jin,Jia-Qi Fang,Jian-Guang Wang,Bo Xu,Xu Wang,Shu-Hao Liu,Feng Chen,Jun-Jian Liu

Inflammation and oxidative stress are among the leading causes of poor prognosis after peripheral nerve injury (PNI). Urolithin-A (UA), an intermediate product produced by the catabolism of ellagitannins in the gastrointestinal tract, has anti-inflammatory, antioxidant, and immunomodulatory properties for inflammation, oxidative damage, and aging-related diseases. Hence, we prepared UA-loaded hydrogels and embedded them in the lumen of PCL nerve guide conduits (NGCs). The hydrogels continuously released appropriate doses of UA into the microenvironment. Based on in vitro studies, UA facilitates cell proliferation and reduces oxidative damage. Besides, the experimental evaluation revealed good biocompatibility of the materials involved. We implanted NGCs into rat models to bridge the sciatic nerve defects in an in vivo study. The sciatic functional index of the PCL/collagen/UA group was comparable to that of the autograft group. Additionally, the consequences of electrophysiological, gastrocnemius muscle and nerve histology assessment of the PCL/collagen/UA group were better than those in the PCL and PCL/collagen groups and close to those in the autograft group. In this study, UA sustained release via the PCL/collagen/UA NGC was found to be an effective alternative treatment for PNI, validating our hypothesis that UA could promote regeneration of nerve tissue.

Modulating Cell–Scaffold Interaction via dECM-Decorated Melt Electrowriting PCL Scaffolds

Wenchao Li,Xiang Gao,Peng Zhang

Aligned fibrous scaffolds are essential for directing soft-tissue regeneration, yet synthetic polymers lack native biochemical cues. To bridge this gap, bioactive and anisotropic scaffolds were developed by combining melt electrowriting (MEW) with decellularized extracellular matrix (dECM) decoration to enhance cell–scaffold interactions for soft tissue engineering. Porous polycaprolactone (PCL) scaffolds with aligned microfibers and tunable pore architectures (aspect ratios 1:1, 1:2, and 1:3) were fabricated via MEW and subsequently coated with porcine skeletal muscle dECM using a dip-gelation method. Comprehensive surface characterization confirmed the presence and robust adhesion of the dECM coating on the PCL scaffolds, which concurrently enhanced surface hydrophilicity. Furthermore, mechanical testing demonstrated that the resulting composite scaffold retained the structural integrity required to meet the mechanical demands of tissue regeneration. In vitro studies using L929 fibroblasts demonstrated that dECM decoration significantly improved cell adhesion, proliferation, and alignment along the fiber direction. Notably, scaffolds with 1:1 and 1:2 aspect ratios supported the highest cell density and guided morphological elongation most effectively. These findings highlight the synergistic potential of topographical cues and biochemical signaling in scaffold design for functional tissue regeneration.

Endothelium-Protective, Intimal Hyperplasia-Resistant PCL/KAT Scaffold for Vascular Implants

Dawei Jin,Pengfei Li,Yu Sun,Yanjun Pan,Jiang Yuan,Meng Yin

Small-diameter vascular grafts (SDVGs) have great potential in the treatment of cardiovascular diseases. However, thrombosis and restenosis of SDVGs, caused by incomplete endothelium and abnormal smooth muscle cell proliferation, limit their clinical applications. Hydrogen sulfide (H2S) is a crucial signaling molecule in the cardiovascular system, playing a vital role in physiological processes such as blood pressure regulation, angiogenesis, and the reduction of vascular hyperplasia, as well as exerting anti-inflammatory effects. In this study, a human hair keratin-based H2S donor was synthesized and coelectrospun with poly(ε-caprolactone) to develop an H2S-releasing vascular graft. The graft effectively promoted the growth and migration of HUVECs and suppressed the proliferation of HUASMCs by releasing H2S. Interestingly, the grafts accelerated endothelium formation under shear stress and protected them from oxidative stress. In vivo experiment also demonstrated that the endothelial layer regenerated without detectable thickening of the smooth muscle layer after 1 month of implantation, which was attributed to the H2S-mediated effect. Taken together, this study provided strategies for the tissue remolding of small-diameter vascular grafts.

PCL-based and Hirudin-containing Composite Nanofibers for Prolonged Anticoagulation Effect

Zheng Zhiwen,Dai Xin,Li Xueyang,Du Chang

M ore and more concerns about health bring the increasing demand for blood contact tissue engineering alternatives. In this paper, nanoparticles of poly(lactic-co-glycolic acid)/polyethyleneimine mixed with recombinant hirudin(rHNPs) were prepared by a double emulsion solvent volatilization method, which were then loaded onto the polycaprolactone(PCL) with polydopamine(PDA) coating to form the composite nanofibers of PCL/PDA/rHNPs. The hydrophilicity and mechanical properties of the composite nanofibers were improved significantly compared with pure PCL. The morphology kept almost unchanged after 30 d of degradation in phosphate buffer saline (PBS). The anticoagulant molecule of hirudin could be gradually released from the composite scaffolds through the degradation of rHNPs in vitro . When the concentration of rHNPs suspension was 5.0 mg/mL, the composite nanofibers could better promote the growth and proliferation of human umbilical vein endothelial cells(HUVECs). The anticoagulant ability of the composite nanofibers was also significantly improved in comparison with that of pure PCL. The design of controlled release anticoagulant materials would alleviate the sudden release of simple fixed hirudin, which could also provide a new idea for the development of novel blood contact materials.

Controllable microfluidic fabrication of monodisperse PCL microspheres for soft tissue augmentation

Xing-Long Zhou,Hao-Wen Zhu,Han Fu,Chang-Hai Zhou,Da-Wei Pan,Yu-Chao Deng,Wei Wang,Zhuang Liu,Rui Xie,Xiao-Jie Ju,Liang-Yin Chu

Monodisperse PCL microspheres with precise size regulation are fabricated via droplet-based microfluidics. • PCL microspheres exhibit superior long-term stability and excellent biocompatibility. • Monodisperse 42 μm PCL microspheres exhibit the most superior ability to stimulate collagen neogenesis. Polycaprolactone (PCL) microspheres are emerging as versatile biomaterials for minimally invasive soft tissue augmentation due to their tunable biodegradability and favorable biocompatibility. Nevertheless, the extent to which monodispersity governs the functional performance of PCL microspheres, particularly in modulating host tissue responses and regenerative efficacy, remains poorly understood. Herein, we propose a method to controllably prepare monodisperse oil-in-water (O/W) droplet templates using coaxial flow-focusing microfluidics, and then obtaining monodisperse PCL microspheres with programmable sizes via solvent evaporation. The droplet dimensions are inversely regulated by the outer-to-inner phase flow rate ratio, while PCL concentration in the organic phase exerts minimal influence on initial droplet size but markedly reduces the volume shrinkage during solidification, yielding microspheres that better retain their geometries. Notably, the resultant PCL microspheres can maintain structural integrity and size uniformity over two-month period under physiologically mimetic conditions. Comprehensive biocompatibility assessments reveal that PCL microspheres exhibit negligible hemolytic activity and cytotoxicity, demonstrating excellent hemocompatibility and cytocompatibility. Notably, in a rabbit soft tissue implantation model, monodisperse PCL microspheres with an average diameter of 42  μm elicit attenuated foreign body reactions and potentiate endogenous collagen deposition relative to the polydisperse microsphere control group. These results provide useful guidance for the application of PCL microspheres in soft tissue augmentation. Download: Download high-res image (210KB) Download: Download full-size image

Solvent-Driven Interfacial Regulation of Lignin/PCL Composites Using γ-Valerolactone

Liang Minting,Xue Wenye,Tu Shiqiang,Ma Bole,Zheng Jiayi,Chen Liheng,Qiu Xueqing

The intrinsic incompatibility between lignin and biodegradable polyesters remains a key barrier to the development of high-performance biobased composite films. We propose a γ-valerolactone (GVL)-enabled strategy to regulate interfacial interactions between alkali lignin (AL) and polycaprolactone (PCL) through cross-linking network formation. GVL serves as a dissolution and purification medium, removing insoluble impurities and aggregates while promoting homogeneous lignin dispersion. A flexible cross-linked network is constructed via epoxy ring-opening reactions, enhancing interfacial adhesion and modulating chain mobility. The resulting films exhibit balanced mechanical strength (14.64 MPa) and ductility (1415% elongation), together with ultraviolet shielding, photothermal conversion (∼25 ° C), and tunable gas/water vapor transport (O 2 /CO 2 ≈ 1.2; WVTR < 100 g/(m 2 ·day)). This study provides fundamental insights and a scalable pathway for engineering biodegradable composite films.

Influence of electrohydrodynamic jetting parameters on the morphology of PCL scaffolds

Hang Liu,Sanjairaj Vijayavenkataraman,Dandan Wang,Linzhi Jing,Jie Sun,Kai He

One of the important constituents in tissue engineering is scaffold, which provides structural support and suitable microenvironment for the cell attachment, growth and proliferation. To fabricate micro/nano structures for soft tissue repair and three-dimensional (3D) cell culture, the key is to improve fibre-based scaffold fabrication. Electrohydrodynamic (EHD) jetting is capable of producing and orientating submicron fibres for 3D scaffold fabrication. In this work, an EHD-jetting system was developed to explore the relationship between vital processing parameters and fibre characteristics. In this study, polycaprolactone (PCL) solution prepared by dissolving PCL pellets in acetic acid was used to fabricate the scaffolds. The influence of voltage, motorized stage speed, solution feed rate, and solution concentration on fibre characteristics and scaffold pattern were studied. Morphology of the EHD-jetted PCL fibres and scaffolds were analysed using optical microscope images and scanning electron microscope (SEM) images. Multi-layer scaffolds with the varied coiled pattern were fabricated and analysed. Cell attachment and proliferation have to be investigated in the future by further cell culture studies on these multi-layer coiled scaffolds.

Dual sustained-release PTMC/PCL porous microspheres for lipid-soluble drugs

Liang Qiao,Fuhua Deng,Xulin Hu,Tianfang Huang,Junlin An,Dong Pan,Junqiang Yan,Gaofeng Liang,Jian He

It is still a considerable research challenge to design biodegradable drug delivery systems that efficiently transport lipid-soluble drugs with controlled release. In the present study, a microsphere delivery system, γ-cyclodextrin (γ-CD)/glutathione (GSH)-atorvastatin (AT) fibrous nanoparticles (γ-CD/GSH-AT FNs) loaded polytrimethylene carbonate (PTMC)/polycaprolactone (PCL) microspheres (PTMC/PCL/γ-CD/GSH-AT MPs), was constructed through an exquisite method. The results show that a large number of γ-CD/GSH-AT FNs can be encapsulated during the formation of microspheres and released in a controllable manner. Notably, the high dispersibility and drug loading of FNs can load with lipid-soluble drugs and protect the drugs from the solvent during microsphere preparation. FNs and MPs can achieve dual release. First, with the degradation of the microspheres, the encapsulated FNs were gradually released into the local microenvironment. Subsequently, the FNs began to dissolve and release the loaded AT over a period of more than 3 days. Therefore, the dual sustained-release drug delivery system can maintain the blood drug concentration for a longer time and reduce the number of administrations and the side effects of the drug. This study provides new ideas for the on-demand manufacture of controllable degradable drug delivery platforms to treat diseases.

Construction of ZnO/PCL Antibacterial Coating Potentially for Dental Unit Waterlines

Min Xing,Haifeng Zhang,Ling Zhang,Wenhao Qian

The formation of bacterial biofilms and the contamination of treatment water within dental unit waterlines can lead to a risk of secondary bacterial infections in immunocompromised patients. Although chemical disinfectants can reduce the contamination of treatment water, they can also cause corrosion damage to dental unit waterlines. Considering the antibacterial effect of ZnO, a ZnO-containing coating was prepared on the surface of polyurethane waterlines using polycaprolactone (PCL) with a good film-forming capacity. The ZnO-containing PCL coating improved the hydrophobicity of polyurethane waterlines, thus inhibiting the adhesion of bacteria. Moreover, the continuous slow release of Zn ions endowed polyurethane waterlines with antibacterial activity, thus effectively preventing the formation of bacterial biofilms. Meanwhile, the ZnO-containing PCL coating had good biocompatibility. The present study suggests that ZnO-containing PCL coating can realize a long-term antibacterial effect on the polyurethane waterlines by itself, providing a novel strategy for the manufacture of autonomous antibacterial dental unit waterlines. Keywords:polyurethane;dental unit waterlines;ZnO;antibacterial

Characterization, biocompatibility, and optimization of electrospun SF/PCL composite nanofiber films

Yanping Feng,Zhang Jupei,Zhihong Dong,Lu Tang

In this study, composite nanofiber films for the wound dressing application were prepared with silk fibroin (SF) and polycaprolactone (PCL) by electrospinning techniques, and the SF/PCL composite nanofiber films were characterized by the combined techniques of scanning electron microscopy (SEM), the equilibrium water content, Fourier transform infrared spectrometer test, X-ray diffraction (XRD) and cell viability test. The results indicated several parameters, including the rotating roller speed, solution concentration, and SF/PCL ratio, affected SF/PCL composite nanofibers’ diameter size, distribution, and wettability. The SF/PCL composite nanofiber manifested a smaller fiber diameter and more uniform nanofibers than pure PCL nanofibers. The contact angle changed from 121 ± 2° of the neat pure PCL to full wetting of 40% SF/PCL composite nanofiber films at 2,000 rpm, indicating good hydrophilicity. Meanwhile, cells exhibit adhesion and proliferation on the composite nanofiber films. These results testified that SF/PCL composite nanofiber films may provide good wettability for cell adhesion and proliferation. It was suggested that optimized SF/PCL composite nanofiber films could be used as a potential biological dressing for skin wound healing.
