Combination of photodynamic therapy (PDT) and photothermal therapy (PTT) generally requires different components to build a composite irradiated with different excitation lights . One component photoactive agent for enhanced combination of PDT and PTT under the excitation of a single wavelength light source is more urgent in tumor phototherapy via adjusting spatial arrangement of photoactive units. Herein, porphyrin-based covalent organic framework nanoparticles (COF-366 NPs) were synthesized to control the orderly spatial arrangement of the photoactive building units and firstly used for antitumor therapy in vivo . COF-366 NPs provide the simultaneous therapy of PDT and PTT under a single wavelength light source with the monitoring of photoacoustic (PA) imaging, which makes the operation simpler and more convenient. COF-366 NPs had achieved good phototherapy effect even in the face of large tumors. The prepared multifunctional COF-366 NPs open up a new avenue to phototherapeutic materials and expand the application range of covalent organic framework.

Laminarin conjugate-based nano-scaled particles were in this study proposed as a delivery system for protoporphyrin IX (Pp IX) in photodynamic therapy (PDT) of human breast cancer cells (MCF-7). Hematin-Laminarin-Dithiodipropionic Acid-MGK, named as HLDM, was an amphiphilic carrier material with dual pH/redox sensitive that could be used to load hydrophobic drug to improve their solubility and enhance biocompatibility. Therefore, we combined photosensitizer (Pp IX) with HLDM to fabricate a novel nano-micelles, herein called Pp IX-loaded HLDM micelles. The Pp IX-loaded HLDM micelles were 149.3 ± 35 nm sized in neutral water. Phototoxicity, in vitro PDT effect, and dual sensibility to pH and redox microenvironment of Pp IX-loaded HLDM micelles were examined at different concentrations by using MCF-7 human breast cancer cells. The experiments on phototoxicity and reactive oxygen species (ROS) production proved that the micelles could produce PDT to kill the cancer cells with a certain wavelength light. The apoptosis experiment indicated that the micelles could cause nuclear damage. In vivo PDT effect of the micelles was studied by constructing the tumor-bearing nude mouse model of MCF-7 cells. In vivo studies showed that the Pp IX-loaded HLDM micelles could induce remarkable anti-tumor effect. A promising laminarin-based nanomedicine platform acts as a new drug delivery system to enhance the uptake, accumulation, and PDT efficacy of Pp IX in vitro and in vivo.

5-Aminolevulinic acid (ALA) based photodynamic therapy (PDT) is a modality for the treatment of cancers. However, due to its hydrophilicity and zwitterionic nature, the transdermal delivery of ALA is limited for the PDT of subcutaneous tumor. To address this problem, tip-loaded fast-dissolving microneedles made of sodium hyaluronate (HA) were fabricated by two casting method. 122 μg of ALA was loaded per microneedle patch and mainly distributed in the tips, which could improve the utilization of drug and avoid the waste of drug residue in the base of microneedle patch after use. The HA microneedles could pierce stratum corneum with insertion depth about 200 μm in isolated skin. After insertion, HA microneedles were rapidly dissolved to release the encapsulated drug to improve patients' convenience and compliance. Importantly, in a subcutaneous mouse tumor model established in BALB/c nude mice, the PDT efficacy of ALA-loaded HA microneedle group was much better than ALA injection group in spite of a relatively lower ALA dose with HA microneedles. The tumor inhibition rate of ALA-loaded HA microneedle group (containing 0.61 mg of ALA) was up to 97%, while the tumor inhibition rate of ALA injection group (containing 1.65 mg of ALA) was just 66%. In addition, microchannels created by microneedle patch were quickly recovered within 3 h after insertion. Overall, the tip-loaded fast-dissolving HA microneedle patch with ALA as drug was promising and might improve topical PDT efficacy of subcutaneous tumor in an efficient and safe manner.

Photodynamic therapy (PDT) received great attention in cancer therapy due to the advantages of negligible drug resistance, low side effects, and minimal invasiveness. Development of theranostic nanoprobes with specific imaging-guided PDT is of great significance in the field. Herein we report the fabrication of a novel theranostic nanoprobe porphyrin/G-quadruplex conjugated gold/persistent luminescence nanocomposites for imaging-guided PDT. The developed nanoprobe contains NIR-emitting persistent luminescent nanoparticles (PLNP) as the core for autofluorescence-free bioimaging and Au coating on PLNP for facile subsequent DNA conjugation. The DNA sequence is designed to contain G-rich AS1411 aptamer for recognizing the over-expressed cellular nucleolin of cancer cell and forming a G-quadruplex structure to combine with tetrakis (4-carboxyphenyl) porphyrin (TCPP) to realize PDT. The AS1411 aptamer-contained DNA conjugated Au-coated PLNP is rapidly prepared via a freezing method with high content of DNA and good aqueous stability. Meanwhile, TCPP is easily loaded into the G-quadruplex structure formed from G-rich AS1411 aptamer on the surface of Au/PLNP in presence of K + . The theranostic nanoprobe gives integrated merits of PLNP for autofluorescence-free bioimging, TCPP for PDT and AS1411 aptamer-contained DNA for specific binding to cancer cells. This work provides a new specially designed imaging-guided PDT nanoplatform for theranostics.

Considering traditional treatment methods, such as chemotherapy, with their potential side effects and lack of targeting, photodynamic therapy (PDT) and photothermal therapy (PTT), being innovative, with less side effects, and light-controlled strategies for antitumour applications, have received extensive attention, but the efficacy of PDT is seriously hindered by the hypoxia tumour microenvironment. Here, a multifunctional nanocomposite composed of an iron oxide (Fe3O4) core and two shells of manganese dioxide (MnO2) and polypyrrole (PPy) was successfully prepared and used to solve these issues. PPy, simultaneously as the photothermal agent and photosensitizer, was first combined with MnO2 to coat magnetic Fe3O4 for achieving an increase in the intracellular O2 concentration and to improve the generation ability of singlet oxygen upon laser irradiation, so that enhanced PDT/PTT could be obtained. After the efficient loading of doxorubicin (DOX) on the Fe3O4@MnO2@PPy nanocomposite, an acid controlled-release behaviour for DOX as well as magnetic-targeting-assisted synergistic effects of chemotherapy and improved PDT/PTT for tumour cells could be realized, which could also protect normal cells from damage. All these features may render our nanocomposite a promising platform to reverse hypoxia-triggered PDT resistance and chemotherapy-caused side effects in cancer therapy and other biomedical applications.

The accumulation of nanomedicines in tumor tissues determines their therapeutic efficacy. We herein exploit the tropism of macrophages to improve the accumulation and retention time of nanomedicine at tumors. Interestingly, macrophages are not merely as transporters, but killers activated by nanomedicine. The system( [email protected] /ICG) was established by decorating macrophages with hyaluronic acid-modified hollow mesoporous carbon (C) nanoparticles loading indocyanine green (ICG). Notably, C nanoparticles with superior photothermal conversion capability not merely guarantee the efficient delivery of ICG through high drug loading efficiency and inhibiting the premature leaky, but effectually activate the polarization of macrophages. The results exhibited that those activated macrophages could release pro-inflammatory cytokines (NO, TNF-α, IL-12), while [email protected] /ICG afforded about 2-fold higher tumor accumulation compared with pure nanoparticle C-HA/ICG and produced heat and singlet oxygen ( 1 O 2 ) under irradiation of an 808 nm laser , realizing the combination of photodynamic therapy (PDT), photothermal therapy (PTT) and cytokines-mediated immunotherapy . Specially, we also investigated the relationship of singlet oxygen ( 1 O 2 ) or temperature and tumor-killing activity for understanding the specific effectual procedure of PDT/PTT synergistic therapy. Overall, we firstly established an “all active” delivery system integrating the features of nanomedicine with biological functions of macrophages, providing a novel insight for cell-mediated delivery platform and tumor targeted multimodality anti-cancer therapy.

As a novel non-apoptotic cell death pathway, ferroptosis can effectively enhance the antitumor effects of photodynamic therapy (PDT) by disrupting intracellular redox homeostasis. However, the reported nanocomposites that combined the PDT and ferroptosis are cumbersome to prepare, and the unfavorable tumor microenvironment also severely interferes with their tumor suppressive effects. To address this inherent barrier, this study attempted to explore photosensitizers that could activate ferroptosis pathway and found that the photosensitizer aloe-emodin (AE) could induce cellular ferroptosis based on its specific inhibiting activity to Glutathione S-transferase P1(GSTP1), a key protein for ferroptosis. Herein, we prepared [email protected] /Fe nanocrystals (NCs) with synergistic PDT and ferroptosis therapeutic effects by one-step emulsification to obtain AE NCs cores and further modification of red blood cells (RBC) membranes and ferritin. Benefiting from the involvement of ferritin, the prepared [email protected] /Fe NCs provide not only sufficient oxygen for oxygen-dependent PDT, but also Fe 3+ for iron-dependent ferroptosis in tumor cells. Furthermore, the biomimetic surface functionalization facilitated the prolonged circulation and cancer targeting of [email protected] /Fe NCs in vivo . The in vitro and in vivo results demonstrate that [email protected] /Fe NCs exhibit significantly enhanced therapeutic effects for the combined two antitumor mechanisms and provide a promising prospect for achieving PDT/ferroptosis synergistic therapy.

Photodynamic therapy (PDT) is proved to be a promising modality for clinical cancer treatment. However, it also suffers from a key obstacle in association with its oxygen-dependent nature which greatly limits its effective application against hypoxic tumors. Herein, on the basis of the unique property of calcium peroxide (CaO 2 ), we propose an O 2 -self-supply strategy for the promotion of PDT by combining the in situ O 2 -generation characteristic of calcium peroxide with the photosensitive nature of porphyrin . A shell of ZIF-8 was synthesized surround the CaO 2 core to prevent the CaO 2 from premature decomposition and increased the loading of THPP efficiently. Depending on the in situ self-supply of O 2 , the photosensitizer was able to exhibit an enhanced PDT effect that significantly inhibit the growth of tumor. Moreover, the enrichment of free calcium ions derived from the decomposition of CaO 2 under acidic tumor microenvironment also shows the unique ion-interference effect and contributes to the obvious inhibition against tumor growth. This work presents a synergistic strategy for the construction of a photodynamic promotion/ion-interference combined nano-platform which can also serve as an inspiration for the future design of effective nanocomposites in tumor treatment.

The biofilms formed by bacteria at the wound site can effectively protect the bacteria, which greatly weakens the effect of antibiotics. Herein, a microneedle patch for wound treatment is designed, which can effectively penetrate the biofilms in a physical way because of the penetration ability of the microneedles and the motion behavior of the nanomotors, and deliver bacterial quorum sensing inhibitor luteolin (Le) and nanomotors with multiple antibacterial properties within biofilms. Firstly, the nanomotors-loaded microneedle patches are prepared and characterized. The results of in vitro and in vivo experiments show that the microneedle patches have good biosafety and antibacterial properties. Among them, Le can inhibit the growth of biofilms. Further, under near-infrared (NIR) irradiation, the nanomotors loaded with photosensitizer ICG and nitric oxide (NO) donor L -arginine (L-Arg) can move in the biofilms under the double driving effect of photothermal and NO, and can give full play to the multiple anti-biological infection effects of photothermal therapy (PTT), photodynamic therapy (PDT) and NO, and finally realize the effective removal of biofilms and promote wound healing. The intervention of nanomotor technology has brought about a new therapeutic strategy for bacterial biofilm-related infection of wound.

In emerging strategies for localized tumor therapy, FDA-approved indocyanine green (ICG) is a promising candidate for phototherapy. Despite its potential, ICG's application is hindered by aqueous instability, intracellular antioxidant scavenging, and rapid clearance, necessitating nanomaterial-based formulations. Herein, a multifunctional nanoplatform integrating ICG into a glutathione (GSH)-responsive metal–organic framework (MOF) functionalized with AS1411 aptamers (AS1411@ICG@MOF, MIA) for stimuli-responsive delivery and intracellular redox regulation is developed. Constructed with a GSH-degradable MOF core for ICG encapsulation and an AS1411 aptamer shell for nucleolin targeting, MIA enables active tumor cell accumulation. Under laser irradiation, ICG acts as both a photosensitizer and photothermal agent, generating reactive oxygen species (ROS) and heat for synergistic photodynamic therapy/photothermal therapy (PDT/PTT). Thereout, GSH-triggered MOF disintegration facilitates intracellular ICG release, overcoming bioavailability challenges and GSH-mediated ROS consumption. Moreover, mild PTT minimizes off-target thermal injury while enhancing PDT via GSH depletion. Taken together, through aptamer-targeted delivery and GSH-responsive degradation, MIA achieves spatiotemporal control of ROS amplification and mild PTT, offering a promising strategy for synergistic cancer therapy.

Candida albicans ( C. albicans ), a ubiquitous polymorphic fungus in humans, causes different types of candidiasis, including oral candidiasis (OC) and vulvovaginal candidiasis (VVC), which are physically and mentally concerning and financially costly. Thus, developing alternative antifungals that prevent drug resistance and induce immunity to eliminate Candida biofilms is crucial. Herein, a novel membrane-targeted aggregation-induced emission (AIE) photosensitizer (PS), TBTCP-QY, is developed for highly efficient photodynamic therapy (PDT) of candidiasis. TBTCP-QY has a high molar absorption coefficient and an excellent ability to generate 1 O 2 and •OH, entering the interior of biofilms due to its high permeability. Furthermore, TBTCP-QY can efficiently inhibit biofilm formation by suppressing the expression of genes related to the adhesion ( ALS3 , EAP1 , and HWP1 ), invasion ( SAP1 and SAP2 ), and drug resistance ( MDR1 ) of C. albicans , which is also advantageous for eliminating potential fungal resistance to treat clinical infectious diseases. TBTCP-QY-mediated PDT efficiently targets OC and VVC in vivo in a mouse model, induces immune response, relieves inflammation, and accelerates the healing of mucosal defects to combat infections caused by clinically isolated fluconazole-resistant strains. Moreover, TBTCP-QY demonstrates excellent biocompatibility, suggesting its potential applications in the clinical treatment of OC and VVC.

In order to effectively avoid the side effects induced by multiple components and tedious synthesis process, a simple therapy system based on one material to simultaneously realize both photothermal therapy (PTT) and photodynamic therapy (PDT) under single laser irradiation will promote the overall phototherapeutic efficiency and make the PTT/PDT system easier to operate. Here, by using transferrin (Tf) as protein template, ultrasmall [email protected] nanodots ( [email protected] NDs) were successfully synthesized through a facile one-pot protein-based biomineralization method. The obtained [email protected] NDs exhibited not only excellent photothermal conversion ability (34.4%) but also high photoactivated formation of reactive oxygen species (ROS) upon 980 nm near-infrared (NIR) irradiation. By loading the drug doxorubicin (DOX) to [email protected] NDs, a synergistic therapy system with multiple therapeutic effects combined PTT, PDT, chemotherapy (CT) and tumor targeting properties could be perfectly implemented together by [email protected] NDs without any complicated post-modification process. Results from the in vitro cell experiments confirmed that these [email protected] NDs could produce excellent effect on cancer cells with 88.5% cell inhibition rate. In comparison with the complicated systems based on “multiple-components-in-one” strategy, this therapy system based on one single material but possess multifunctional purpose is easy to operate and more suitable for clinical applications.
