Effect of pH-shifting combined with ultrasound treatment on the physicochemical, structural, and gelation properties of oxidized pea protein isolate

Qian Li,Siying Liu,Yi Cao,Yujie Cheng,Xudong Yan,Jun Chen,Taotao Dai,Peiyi Shen,David Julian McClements,Jian Xu,Ruiyun Chen

Oxidized pea protein isolate (OPPI) exhibits reduced solubility and impaired functional properties, limiting its application in food systems. This study aimed to modify the physicochemical and structural characteristics of OPPI using either pH-shifting alone or pH-shifting combined with sonication for enhancing its gelation properties. These treatments significantly promoted protein unfolding, reducing the aggregate size (from 1303 to 178 nm) and increasing protein solubility (from 5.8 to 23.2%), surface hydrophobicity (1.8-fold), and free sulfhydryl content (from 0.62 to 1.09 μmol/g). Only OPPI treated with both pH-shifting and sonication formed transglutaminase-induced gels. The gelation properties were pH-dependent, with higher pH values yielding stronger gels, as evidenced by increased shear modulus and hardness. This enhancement may result from greater exposure of active sites on protein surfaces at elevated pH, which promotes intermolecular interactions and enzymatic cross-linking. Thus, combining pH-shifting and ultrasonication methods is an effective strategy for enhancing the gelation properties of OPPI.

Fabrication of pea protein isolate-folic acid conjugates via free radical grafting: Formation mechanism, stability, and curcumin delivery potential evaluation

Zijun Wang,Huan Li,Shikai Zhang,Ke Ding,Ying Xie,Rongrong Wang,Liang Zhao,Wenjiang He,Yang Shan,Shenghua Ding

Plant proteins are increasingly used to encapsulate hydrophobic nutrients to formulate functional foods. A major challenge for plant protein-based nanocarriers is to improve the solubility and stability, thereby enhancing the absorption efficiency and bioavailability of hydrophobic nutrients within them. To address this issue, a free radical grafting strategy of folic acid (FA) modified pea protein isolate (PPI) was developed. The FA loading capacity of PPI-FA conjugates was 2.1-fold higher than that of PPI-FA mixtures. Conjugation of FA improved the solubility, thermal stability and storage stability of PPI. The photostability of FA was also enhanced (retention rate increased from 75.76 % to 94.92 %). Complexation with PPI-FA conjugate elevated the solubility, anti-cancer activity and cellular uptake of curcumin (Cur). The bioavailability of PPI-FA-Cur was 63.27 %, far exceeding those of free Cur (6.95 %) and PPI-Cur (24.47 %). These findings expand PPI's application and provide new reference for the development of advanced food delivery systems.

Synergistic effects of heating and ultrasonic co-treatment on pea protein functionality: Self-assembled protein carriers for enhanced resveratrol solubility

Lihua Tang,Qinhao Guan,Yuan Wang,Man Xu

A sequential heating and ultrasonic co-treatment was developed to structurally modify pea protein (PP) and enhance its performance as a self-assembled nanocarrier for resveratrol (RES). The impact of this co-treatment on PP's molecular structure and its subsequent RES encapsulation efficiency via pH-driven self-assembly was evaluated. Structural characterization confirmed that the treatment induced significant molecular reconfigurations, including covalent cross-linking through carbonyl formation, amino group oxidation, and sulfhydryl-to-disulfide bond conversion. These modifications directly improved the functional properties of PP. The optimal treatment (20 min heating followed by 30 min sonication, H 20 U 30 ) increased PP solubility by 194.77 % compared to the untreated control. Subsequent self-assembly with RES was most effective under alkaline conditions, a process governed predominantly by hydrophobic interactions and van der Waals forces. This treatment enhanced the resveratrol loading capacity of the pea protein by 115.19 % relative to its native counterpart. These findings demonstrate that sequential heating-ultrasound treatment is an effective strategy for engineering PP, markedly improving its solubility and capacity for encapsulating poorly soluble nutraceuticals. This pre-modification approach represents a valuable method for developing advanced delivery systems in functional foods.

Study on the mechanism of polyphenols regulating the stability of pea isolate protein formed Pickering emulsion based on interfacial effects

Lulu Ma,Xiaofan Yang,Jiaying Huo,Shugang Li

To improve the stability of pea isolate protein (PPI) Pickering emulsions, this study compared the stability effects of tannic acid (TA), epigallocatechin gallate, and gallic acid on PPI, and found PPI-TA the strongest binding and the best stability. When TA concentration increased from 0 to 0.5 mmol/L, the average particle size, zeta potential, and surface hydrophobicity of PPI-TA particles decreased by 23.1 %, 17.1 %, and 63.3 % respectively. The highest viscosity and elastic storage modulus G' which was also higher than and parallel to the loss modulus G", and the lowest Turbiscan stability index were observed in the emulsion with 0.5 mmol/L TA, indicating an elastic-based gel-like texture. The concentrations of conjugated diene and thiobarbituric acid reactive substances (TBARS) were also reduced by more than 58 %, showing improved oxidative stability. The study provides new insights into the interfacial behavior of PPI-polyphenols and technical support for their applications in food industry.

Cajanstilbenoid A from Pigeon Pea: A Functional Food Component for Managing High-Fat Diet-Induced Hepatic Steatosis via PPARα Activation

Bailin Li,Fen Liu,Yaoguang Huang,Sui Kiat Chang,Chenqi Niu,Pu Liu,Xiancai Li,Liyuan Yao,Sheng-Xiang Qiu

Pigeon pea ( Cajanus cajan (L.) Millsp.), a plant with documented safety and dual use in food and traditional medicine, holds promise for developing hepatoprotective agents. This study aimed to evaluate its potential to mitigate high-fat diet (HFD)-induced hepatic steatosis. We investigated a stilbene-enriched extract (EX) from pigeon pea leaves and its major constituent, cajanstilbenoid A (CSA). In HFD-fed mice, both EX and CSA significantly reduced body weight, liver index, lipid accumulation and serum cholesterol/triglyceride levels. CSA also ameliorated oxidative stress and improved liver function markers. Mechanistic studies revealed that CSA acted as a selective agonist of peroxisome proliferator-activated receptor alpha (PPARα): Network pharmacology, transcriptomics, and molecular docking predicted PPARα as a target; this was confirmed by cellular thermal shift assay (CETSA), drug affinity responsive target stability assays (DARTS) and surface plasmon resonance (SPR), which demonstrated direct binding. Crucially, siRNA-mediated knockdown of PPARα in HepG2 cells abolished the lipid-lowering effects of CSA. Our findings demonstrated that CSA alleviated HFD-induced hepatic steatosis primarily by activating the PPARα pathway, reducing oxidative stress and promoting lipid metabolism. This study highlighted the therapeutic potential of CSA for non-alcoholic fatty liver disease and underscored pigeon pea as a valuable medicinal and edible resource.

Enhancing storage stability of pea peptides through encapsulation in maltodextrin and gum tragacanth via monitoring scavenge ability to free radicals

Kang-Ke Liu,Hao-Ran Liu,Li Wen,Zhou Xu,Li Ding,Yun-Hui Cheng,Mao-Long Chen

Pea peptides can lead to degradation through oxidation, deamidation, hydrolysis, or cyclization during production, processing, and storage, which in turn limit their broader application. To stabilize pea peptides, this study employed spray drying technology to create a pea peptide micro-encapsule using maltodextrin, gum tragacanth, and pea peptides. Four key factors, including polysaccharide ratio, glycopeptide ratio, solid-liquid ratio, and inlet temperature, were optimized to enhance the antioxidant properties of the pea peptide micro-encapsule. The results indicated that the utilization of maltodextrin and gum tragacanth significantly improves the storage stability and antioxidant activity of pea peptides. Moreover, optimal storage stability for pea peptides was achieved with a polysaccharide ratio of 9:1, a glycopeptide ratio of 10:1, a solid-liquid ratio of 4:40, and an inlet temperature of 180 °C. After 60 days of storage, the encapsulated pea peptides maintained 70.22 %, 25.19 %, and 40.32 % for scavenging abilities to hydroxyl radical, superoxide anion, and ABTS radical, respectively. In contrast, the unencapsulated pea peptides showed a decline to 47.02 %, 0 %, and 24.46 % in the same antioxidant activities after storage. These findings underscore the potential of spray drying technology to enhance the functional properties of pea peptides for various applications.

Green Pea (Pisum sativum L.) Hull Polyphenol Extracts Ameliorate DSS-Induced Colitis through Keap1/Nrf2 Pathway and Gut Microbiota Modulation

Fanghua Guo,Rong Tsao,Chuyao Li,Xiaoya Wang,Hua Zhang,Li Jiang,Yong Sun,Hua Xiong

As a processing by-product, green pea hull (GPH) was found to be rich in phenolic components in our previous studies. In this study, UHPLC-LTQ-OrbiTrap-MS (Ultra performance liquid chromatography-linear ion trap orbitrap tandem mass spectrometry) technique was used to quantify polyphenols, and DSS (sodium dextran sulfate)-induced colitis mouse model was established to explore the effect of GPH extracts on colitis. The results showed that quercetin and its derivatives, kaempferol trihexanside and catechin and its derivatives were the main phenolic substances in the extract, reaching 2836.57, 1482.00 and 1339.91 µg quercetin/g GPH extract, respectively; GPH extracts can improved inflammatory status, repaired colonic function, regulated inflammatory factors, and restored oxidative balance in mice. Further, GPH extracts can activate Keap1-Nrf2-ARE signaling pathway, regulate downstream antioxidant protease and gut microbiota by increasing F/B value and promoting the growth ofLactobacillaceaeandLachnospiraceae, and improve the level of SCFAs (short-chain fatty acids) to relieve DSS-induced colitis in mice. Therefore, GPH may be a promising dietary resource for the treatment of ulcerative colitis.Keywords:polyphenols;colitis;UHPLC-LTQ-OrbiTrap-MS;GPH extracts;Keap1-Nrf2;gut microbiota

Pea-Resistant Starch with Different Multi-scale Structural Features Attenuates the Obesity-Related Physiological Changes in High-Fat Diet Mice

Danni Chang,Xinzhong Hu,Zhen Ma

The present study compared the modulatory effects of different resistant starches (RSs) isolated from native (NP-RS), acid-hydrolyzed (AHP-RS), and pullulanase debranched (PDP-RS) pea starches on the corresponding in vivo metabolic responses in high fat (HF)-diet-induced obese mice. The biochemical studies on serum lipid profile and antioxidant enzyme activities were supported by histological and gene expression analyses, which suggested a potential therapeutic role for RS in regulating obesity, possibly through the production of short-chain fatty acids and the proliferation of some beneficial colonic bacteria, including Allobaculum, Bifidobacterium, Odoribacter, Clostridium, and Prevotella. Particularly, a more pronounced effect of AHP-RS with a higher proportion of the crystalline region and a more ordered double-helical alignment on improving the hyperlipidemic symptoms in obese mice induced by a HF diet was observed. Our analysis revealed that the RS3 samples seemed to be more effective than RS2 in terms of attenuating obesity in mice that were fed a HF diet.

Self-assembly of amino acid-based random copolymers for antibacterial application and infection treatment as nanocarriers

Jie Zhu,Hua Han,Faxue Li,Xueli Wang,Jianyong Yu,Chih-Chang Chu,Dequn Wu

Bacterial infection is one of the most significant complications worldwide and has been one of the main factors of morbidity and mortality for the chronic wounds. Considering the negative charged feature of bacterial pathogens, a positive charged poly(ester amide) (PEA) micellar system based on lysine, arginine and phenylalanine is developed. In this study, a serials of PEA random copolymers can be obtained by altering the sorts of amino acids and feed ratio, and the self-assembled PEA micelles with an average diameter ranging from 150 to 200 nm exhibit the integrated properties of excellent biocompatibility and enzymatic biodegradation. More interesting, the degraded random block micelles can reassemble into smaller sized micelles with the diameter less than 20 nm which have promising applications in drug delivery. The PEA micellar nanocarriers display an intrinsic antibacterial property due to the pendant groups of lysine and arginine based moieties and this killing capacity can be enhanced by grafting levofloxacin without losing the original performance. The in vitro antibacterial evaluation proves all of the micelles display a concentration dependent efficiency of killing bacteria (up to 99.99%). The in vivo Staphylococcus aureus induced infection model demonstrates that the micelles are effective in killing the bacteria and infection treatment. The successful synthesis of the biocompatible and biodegradable amino acid based micellar nanocarriers may provide new insights into the development of biomedical materials for antibacterial applications and drug delivery.

Pore-structure-tunable carboxymethyl cellulose/polyvinyl alcohol aerogel functionalized with butterfly pea anthocyanin for real-time visual monitoring of pork freshness

Ke Zhang,Zhihua Li,Xiaowei Huang,Yaning Qin,Junjun Zhang,Sulafa B.H. Hashim,Haoyang Xu,Ruoying Zhang,Jiyong Shi,Xiaobo Zou

This study developed a pore-tunable colorimetric aerogel based on carboxymethyl cellulose (CMC), polyvinyl alcohol (PVA), and butterfly pea anthocyanin (BA) via sol-gel/freeze-drying, for real-time food freshness monitoring. The regulatory mechanisms of volume ratios on the aerogel's morphology, microstructure, physical properties, and release behavior were systematically investigated. The results demonstrated that CMC and PVA formed a stable 3D-H-bonded network, endowing the aerogel with outstanding water absorption capacity (2468.27 %), high hardness (7008.24 g), and excellent thermal stability. At a CMC/PVA ratio of 1:1, the uniform and dense pore structure modulated the BA diffusion pathways, achieving a continuous and controllable release behavior consistent with Fick diffusion kinetics. Furthermore, the BA-loaded aerogel exhibited high sensitivity to pH buffers and ammonia vapor (with a detection limit of 0.39 μM for ammonia), along with reversible color changes. During pork quality monitoring, the C1P1T (where 'C' and 'P' denote the mass of CMC and PVA, respectively, and 'T' indicates that the aerogel was loaded with BA extract) aerogel showed distinguishable color changes, clearly distinguishing between fresh, sub-fresh, and spoiled stages. This work provides a novel strategy for developing overly sensitive intelligent indicator systems for meat freshness monitoring.

Encapsulation of Lactobacillus plantarum in W1/O/W2 double emulsions stabilized with the high-intensity ultrasound-treated pea protein and pectin

Chao Zhang,Yu Zhang,Bin Qiu,Zhenhua Liu,Xueyan Gao,Nan Zhang,Xia Liu,Shasha Qi,Lingfei Li,Wei Liu

This study focuses on developing a water-in-oil-in-water (W 1 /O/W 2 ) double emulsion system using high-intensity ultrasound (HIU)-treated pea protein isolate (HIU-PPI) and pectin to encapsulate Lactobacillus plantarum ( L. plantarum ). The effects of ultrasound treatment on pea protein isolate (PPI) characteristics such as solubility, particle size, emulsification, surface hydrophobicity, and surface free sulfhydryl group were examined, determining optimal HIU processing conditions was 400 W for 10 min. The developed W 1 /O/W 2 double emulsion system based on HIU-PPI demonstrated effective encapsulation and protection of L. plantarum , especially at the HIU-PPI concentration of 4 %, achieving an encapsulation efficiency of 52.65 %. Incorporating both HIU-PPI and pectin as emulsifiers increased the particle size and significantly enhanced the emulsion’s viscosity. The highest bacterial encapsulation efficiency of the emulsion, 59.94 %, was attained at a HIU to pectin concentration ratio of 3:1. These emulsions effectively encapsulate and protect L. plantarum , with the concentration of HIU-PPI being a critical factor in enhancing probiotic survival under simulated gastrointestinal digestion. However, the concurrent utilization of pectin and HIU-PPI as emulsifiers did not provide a notable advantage compared to the exclusive use of HIU-PPI in enhancing probiotic viability during in vitro simulated digestion. This research offers valuable perspectives for the food industry on harnessing environmentally friendly, plant-based proteins as emulsifiers in probiotic delivery systems. It underscores the potential of HIU-modified pea protein and pectin in developing functional food products that promote the health benefits of probiotics.

Ca2+-mediated pea protein isolate and low-methoxyl pectin systems for structuring peach kernel oil into reduced-oil semi-solid emulsions

Wanheng Gao,Yuxiao Wang,Shuoran Huang,Xin Sun,Bing Peng,Qingyong Li,Rentang Zhang

Interfacial-matrix coordination sustained reduced-oil semi-solid emulsions. • Moderate Ca 2+ reinforced pectin networks and optimized emulsion stability. • A PPI:LM ratio of 5:5 with 25 mM Ca 2+ supported cohesive reduced-oil emulsions. Structuring unsaturated liquid oils into reduced-oil semi-solid emulsions requires simultaneous droplet stabilization and continuous-phase matrix formation. This study examined how pea protein isolate (PPI), low-methoxyl pectin (LM), and Ca 2+ regulate these structural contributions, using peach kernel oil at a water-to-oil ratio of 5:1. Water distribution, water-holding capacity, storage stability, molecular environments, rheology, thermal degradation, and microstructure were evaluated. PPI-rich systems lacked sufficient matrix support, whereas LM-rich systems improved water retention and continuous-phase structuring but showed less uniform droplet distribution. PPI-LM blending balanced droplet stabilization with bulk matrix formation. Moderate Ca 2+ promoted semi-solid structuring. Spectroscopic analysis of corresponding model systems revealed changes in carboxylate-associated and Trp environments, supporting Ca 2+ -dependent biopolymer association. The 5:5 PPI:LM emulsion containing 25 mM Ca 2+ combined restricted water mobility, refrigerated storage stability, solid-like viscoelasticity, shear-thinning behavior, and relatively uniform droplet distribution. Its dominant T2 decreased from 135.1 ± 0.0 to 54.9 ± 3.8 ms after Ca 2+ addition. Increasing Ca 2+ to 50 mM produced only a small further decrease in dominant T2, while rheological properties showed no consistent improvement. A model involving pectin junctions, protein-associated Ca 2+ interactions, and possible protein-pectin bridging was proposed to explain the composition-dependent structuring. These findings link biopolymer composition and ionic regulation to the storage and flow properties of reduced-oil semi-solid emulsions, informing the development of spreadable food bases.
