Preparation of multi-functional magnetic–plasmonic nanocomposite for adsorption and detection of thiram using SERS

Ziwei Fu,Zhengdong Shen,Qinzhen Fan,Shaoxian Hao,Ying Wang,Xinquan Liu,Xiaoxue Tong,Xianming Kong,Zhanxu Yang

Magnetic materials have been widely used for constructing substrate in surface enhanced Raman scattering (SERS) sensing due to the magnetic responsibility. Here, we reported a facile and effective approach to construct multi-functional SERS substrate based on assembling Ag nanoparticles (NPs) on porous Fe microspheres. The porous Fe microspheres were prepared through hydrogen reduction of Fe 2 O 3 NPs with porous structure, in which the size and morphology of Fe could be well controlled. The surface of Fe was grafted with amino group, and then decorated with Ag NPs. The surface area and pore size of Fe microsphere were characterized by nitrogen adsorption and desorption. The Fe@Ag nanocomposite illustrated a good SERS activity. Furthermore, this substrate could be used for pesticide monitoring by portable Raman spectrometer. Especially, the porous Fe microsphere could adsorb analyte from target sample and the Fe@Ag could be concentrated by magnetic force to amplify the SERS signal for thiram detection.

Facilely Flexible Imprinted Hemispherical Cavity Array for Effective Plasmonic Coupling as SERS Substrate

Jihua Xu,Jinmeng Li,Guangxu Guo,Xiaofei Zhao,Zhen Li,Shicai Xu,Chonghui Li,Baoyuan Man,Jing Yu,Chao Zhang

The focusing field effect excited by the cavity mode has a positive coupling effect with the metal localized surface plasmon resonance (LSPR) effect, which can stimulate a stronger local electromagnetic field. Therefore, we combined the self-organizing process for component and array manufacturing with imprinting technology to construct a cheap and reproducible flexible polyvinyl alcohol (PVA) nanocavity array decorating with the silver nanoparticles (Ag NPs). The distribution of the local electromagnetic field was simulated theoretically, and the surface-enhanced Raman scattering (SERS) performance of the substrate was evaluated experimentally. The substrate shows excellent mechanical stability in bending experiments. It was proved theoretically and experimentally that the substrate still provides a stable signal when the excited light is incident from different angles. This flexible substrate can achieve low-cost, highly sensitive, uniform and conducive SERS detection, especially in situ detection, which shows a promising application prospect in food safety and biomedicine.Keywords:surface-enhanced Raman spectroscopy;localized surface plasmon resonance;flexible SERS substrate;cavity

Experimental and theoretical investigation for a hierarchical SERS activated platform with 3D dense hot spots

Chonghui Li,Chao Zhang,Shicai Xu,Yanyan Huo,Shouzhen Jiang,Cheng Yang,Zhen Li,Xiaofei Zhao,Suzhen Zhang,Baoyuan Man

Hot spots generated from the ultra-small nanogaps will tremendously contribute to the strong electromagnetic field. Therefore, the establishment and design of nanogaps are extremely significant. Here, a hierarchical structure, AgNPs/bilayer graphene/Au nanonet, was proposed to form the dense three-dimensional (3D) hot spots and demonstrated in experiment and theory. Numerous nanometer gaps in the complex Au nanonet structure (∼8.67 nm) and in AgNPs/Au nanonet isolated by bilayer graphene (0.64 nm) were achieved, which is much beneficial for the obtainment of the hot spots. Based on this surface enhanced Raman scattering (SERS) substrate, an excellent SERS activity (EF ∼ 9.1 × 10 9 ), high sensitivity (10 −13 and 10 −12  M respective for Rhodamine 6G (R6G) and crystal violet (CV)), and SERS signals homogeneity, outstanding reproducibility (maximum intensity deviation from substrate to substrate ∼10.43%) were obtained, which can be attributed to the abundant lateral and vertical hot spots introduced by the hierarchical structure. Moreover, the proposed SERS substrate was used to sensitively detect the harmful malachite green (MG), which exhibits a great prospect as a potential SERS sensor in food safety field and biochemical molecules.

Site-Specific Growth of Ag Islands on Concave Au Nanocubes for SERS and LSPR-Based Applications

Zhiwei Wang,Yefan Zhou,Junsheng Wang,Yun Wang,Jianwei Zhao,Wenjia Xu,Yuhua Feng

Currently, hybrid noble metal nanostructures have garnered significant interest due to their exceptional properties and diverse applications in fields such as catalysis, energy conversion, plasmonic sensing, and surface-enhanced Raman scattering (SERS). In this work, we report the highly site-specific growth of a single Ag island on both the concave face and the sharp tip of concave Au nanocube (c-AuNC) seeds, resulting in different c-AuNC-Ag Janus heterostructures. This site specificity was achieved by combining the surface curvature effect of the c-AuNCs with strong ligand-mediated interfacial energy control. Further integration of interfacial energy and growth kinetic control led to the formation of c-AuNC-(Ag)n core–satellite structures with varying numbers of Ag satellites. Benefiting from the continuous tunability of the interfacial energy and growth kinetics, the structural evolution of the c-AuNC-Ag hybrid structures continuously occurred, enabling precise control of the Au–Ag bimetallic structures. Notably, the resulting c-AuNC-Ag structures showed tunable intense localized surface plasmon resonance (LSPR) absorptions within the 400–1000 nm wavelength along with strong SERS signals. The broad spectral absorption and strong SERS made the c-AuNC-Ag structures promising candidates for SERS and LSPR-based applications, including sensing, imaging, light energy harvesting, and conversion. Moreover, the effective synthetic control demonstrated in this work could facilitate the development of advanced hybrid metal nanomaterials with valuable properties, further broadening their application potential across various fields.

Shape-controlled asymmetric bowl-like PDA@Au substrates for sensitive SERS detection of anabolic androgenic steroids

Yuanyuan Li,Jiayi Peng,Hong Chen,Weiling Yue,Yixuan Liu,Xiaojun Luo,Lu Yang

The widespread accumulation of androgenic steroid endocrine disruptors in water and food has garnered increasing attention due to their significant risks to ecosystems and human health. These steroids, which cannot be completely eliminated, highlight the urgent need for advanced detection technologies. In this study, we present a novel emulsion-induced interface-anisotropic assembly strategy to synthesize bowl-like mesoporous polydopamine (PDA) particles, which exhibit high sensitivity in surface-enhanced Raman scattering (SERS) detection. In-situ reduction of chloroauric acid leads to the formation of Au nanoparticles (NPs) on the PDA surface, where synergistic Au-N interactions enhance the SERS performance. The distinctive bowl-like structure generates abundant "hot spots" on both sides, resulting in exceptional sensitivity. The low relative standard deviation (RSD) values (<11.7 %) across different PDA@Au NPs, along with real sample analyses (1.9–4.0 %), confirm the high reproducibility and uniformity of the SERS substrates, all achieved without the use of additional reducing agents. This cost-effective and straightforward method eliminates the need for complex surface treatments, making it particularly suitable for real-time detection of anabolic steroids across various matrices. These findings underscore the potential of bowl-like PDA materials for broader applications in clinical diagnostics, environmental monitoring, and sports doping control.

A gold nanoflower-based traceable drug delivery system for intracellular SERS imaging-guided targeted chemo-phototherapy

Chunyuan Song,Yanxia Dou,Lihui Yuwen,Youzhi Sun,Chen Dong,Fang Li,Yanjun Yang,Lianhui Wang

Accurate and effective drug delivery in tumor cells significantly improves the curative effect with high drug delivery efficiency, low toxicity and side effects and has become an urgent demand for anticancer therapy. In this paper, a novel traceable and targeted drug delivery nanosystem (i.e. AuNF-nanocarriers) with high drug encapsulation and pH-controlled release was prepared based on gold nanoflowers (AuNFs) for efficient intracellular SERS imaging-guided chemo-phototherapy. SERS-active flower-like gold nanoparticles with large surface area were synthesized first and then modified with Raman and RGD molecules in sequence to prepare bright, traceable and targeted SERS tags of A549 human lung cancer cells. Furthermore, thiolated-PAA (PAA-SH) was synthesized and utilized for the first time to modify the SERS tags with a layer of negative charges for efficient pH-dependent loading and release of the anticancer drug doxorubicin. Based on the A549 human lung cancer cell model, the availability of the proposed AuNF-nanocarriers for efficient intracellular SERS imaging-guided chemo-phototherapy was studied and the results indicate that the AuNF-based drug delivery system exhibited attractive characteristics such as good stability, efficiency and pH-controlled drug loading and release, traceable and targeted delivery, as well as SERS imaging and chemo-phototherapy functions, and shows great potential for powerful SERS-imaging and as a theranostic candidate for precision nanomedicine that could achieve sensitive and accurate tumor detection and therapy.

Ag NP-filter paper based SERS sensor coupled with multivariate analysis for rapid identification of bacteria

Rong Wang,Jiamin Luo

Rapid and accurate identification of bacteria is essential to ensure food safety and prevent pathogenic bacterial infection. In this study, a highly efficient method was established for accurately identifying bacterial species by applying Ag NP-filter paper based Surface enhanced Raman spectroscopy (SERS) analysis and Partial Least Squares-Discriminant Analysis (PLS-DA) statistical methods. The flexible Ag NP filter paper substrate with high sensitivity and uniformity was prepared by a facile and low-cost silver mirror reaction at room temperature, which exhibited desirable SERS activity in bacteria detection. Furthermore, PLS-DA was successfully employed to distinguish SERS spectra from S. aureus CMCC 26003, E. faecalis ATCC29212 and L. monocytogenes ATCC 19115 with a sensitivity of 93.3–100%, specificity of 96.7–97%, and overall predicting accuracy of 95.8%. This exploratory study demonstrates that a Ag NP-filter paper based SERS sensor coupled with PLS-DA has great potential for rapid and effective detection and identification of bacteria.

DNA-encoded plasmonic bubbles aggregating dual-microRNA SERS signals for cancer diagnosis

Yu Yang,Hao Lu,Dan Fang,Yuyuan Zhang,Yuteng Tang,Songsong Zhao,Jun Yan,Xiaojie Qin,Jianlei Shen,Fan Yang

Solid bubbles have expanded the SERS assay toolbox, but their detection performance in biofluids is still hampered by the irrational design of the plasmonic sensing interface. A plasmonic bubble aggregate-driven DNA-encoded SERS assay is reported here that enables simultaneous, ultrasensitive, and specific detection of multiple miRNAs in blood samples for accurate cancer diagnosis. In this assay, the buoyancy of plasmonic bubbles allows them to self-aggregate at a droplet apex for SERS reconfiguration, form single-layer bubble aggregates with plasmonic nanogaps, and prevent the coffee ring effect during evaporation assembly. Furthermore, DNA-encoded plasmonic bubbles seamlessly couple with dual-color catalytic hybridization assembly to amplify the specific miRNA-responsive Raman signal, and function as both an analyte concentrator and a Raman signal aggregator without external forces. Using these merits, this magnet-free, portable assay achieves femtomolar dual-miRNA quantitation with single-base resolution, simultaneous miRNA detection across four cell lines, and accurate cancer diagnosis (AUC = 1) via analyzing 40 blood samples with machine learning, thus providing a promising tool for clinical diagnosis.

GO/Au@Ag nanobones decorated membrane for simultaneous enrichment and on-site SERS detection of colorants in beverages

Lili Kong,Jie Chen,Meizhen Huang

In this paper, we have developed a graphene oxide/Au@Ag nanobones membrane (GO/Au@Ag NBs membrane) for simultaneous enrichment and SERS detection of colorants in beverages. Raman enhancement effect of Au@Ag nanobones (NBs) and enrichment capability of graphene oxide (GO) are contributed to the high sensitivity of GO/Au@Ag NBs membrane. Microporous membrane provides a support template for uniformly loading high-density GO/Au@Ag NBs. When beverage flow through the portable filtration unit with GO/Au@Ag NBs membrane, colorant molecules could be efficiently enriched. The prepared GO/Au@Ag NBs membrane exhibits an excellent sensitivity with a LOD of Rhodamine 6 G at 1.12 × 10 −9 mol/L, and has good performances of uniformity (with an RSD of 7.87 %), repeatability (with an RSD of 10.44 %), time stability (stored up to 60 days). Six types of colorants were sensitively detected combined with GO/Au@Ag NBs membranes, portable filtration unit and self-development portable Raman system. The lowest distinguished amounts of molecular accumulation are well below the maximum usage limits and the limits of visual recognition. Support vector machine model was established to identify the types of colorants, including colorants with coincident Raman peaks or similar visual colors, with an average accuracy of 94.8 %. In addition, GO/Au@Ag NBs membranes were used to enrich and detect colorant additives in commercial beverages, brilliant blue in energy beverage and carmine in bayberry wine were successfully detected. These results indicate GO/Au@Ag NBs membranes combined with a portable filtration unit can effectively enrich and rapidly detect colorants in beverages, and has great potential in the field of on-site and sensitive food safety detection.

An aptamer-based colorimetric/SERS dual-mode sensing strategy for the detection of sulfadimethoxine residues in animal-derived foods

Nan Zhang,Haiyang Lv,Jihong Wang,Zhanye Yang,Yanru Ding,Bing Zhao,Yuan Tian

Residues of sulfadimethoxine (SDM) in animal-derived foods have attracted widespread public concern. Herein, we propose an aptamer-based colorimetric/SERS dual-mode sensing strategy for the determination of SDM based on hexadecyl trimethyl ammonium bromide (CTAB) induced aggregation of nanoparticles. In the absence of SDM, the SDM aptamer formed a supramolecular composite with CTAB, and the 4-mercaptopyrimidine functionalized gold nanoparticles (AuNPs@4-MPY) remained dispersed due to the lack of CTAB. Upon the addition of SDM, the SDM aptamer preferentially combined with SDM, resulting in the release of CTAB and subsequent aggregation of AuNPs@4-MPY, and the solution color changed from red to blue and presented a dynamic UV-absorbance curve based on different aggregation states. On the other hand, when, gold–silver core–shell nanoparticles (Au@AgNPs) were added additionally, the released CTAB narrowed the nanogap between AuNPs@4-MPY and Au@AgNPs, thus exhibiting enhanced SERS intensity of 4-MPY. This strategy achieved colorimetric detection of SDM with a linear range of 4.00–200.00 ng mL−1 and a detection limit of 2.41 ng mL−1, while SERS had a detection range of 1.20–120.00 ng mL−1 with a detection limit of 0.89 ng mL−1. This strategy is simple and cost-effective for the rapid detection of SDM within 20 minutes. It was further applied for the detection of SDM in spiked milk and honey samples with satisfactory recoveries. Therefore, it exhibits great potential for fast and on-site SDM detection.

Tailored SiO2-TiO2 Aerogel/Ag Flexible Films as Stable SERS Substrates for Hazardous Materials Detection

Wei Liu,Xue Wang,Zhiyang Zhao,Xin Zhou,Shiliang Huang,Longjin Huang,Zihan Wang,Jingru Li,Junjie Fu,Yu Liu,Sheng Cui,Xuan He

Flexible surface-enhanced Raman spectroscopy (SERS) substrates have received more and more attention for their non-invasive sample collection ability. However, majority of them have poor stability and reproducibility because noble metal is easily oxidized and led to lose SERS activity. Herein, SiO 2 -TiO 2 -Ag hybrids aerogel (STAHA) material is first applied as a flexible SERS substrate for explosives and dyes long-term sensitive detection. The sensitive non-invasive detection of 3-nitro-1, 2, 4-triazol-5-one (NTO) residues can be achieved with a limit of 1 × 10 −6 m by wiping sample collection method. Meanwhile, similar SERS sensitivity from long-term (50 days) kept substrates is observed compared with fresh-made substrates. This long-term sensitivity is contributed to electronic transfer compensation from TiO 2 aerogel. This proposed flexible STAHA enrich the type of SERS substrates, and their long-term sensitivity can provide a prospective application for trace analyses of explosives and other hazardous residues in criminal forensic, military, and security check.

A balsam pear-shaped CuO SERS substrate with highly chemical enhancement for pesticide residue detection

Liang Pei,Cao Yu,Dong Qianmin,Wang Dan,Zhang De,Jin Shangzhong,Yu Zhi,Ye Jiaming,Zou Mingqiang

Simple and traditional hydrothermal fabrication of a novel balsam pear-shaped CuO with high SERS enhancement is presented. XRD (X-ray diffraction), SEM (scanning electronic microscopy), TEM (transmission electron microscope), HRTEM (high-resolution transmission electron microscope), UV-Vis, and Raman are adopted to ensure that this balsam pear-shaped CuO with dense nanoparticle protuberance is successfully prepared. The LOD of this CuO SERS substrate is 4.79 μg L −1 with R6G as molecular probe. By using DFT (density functional theory) calculation and FDTD (finite difference time domainmethod) simulation, both EM (electromagnetic enhancement) and CM (chemical enhancement) mechanisms are investigated, and the results show that these two-enhancement mechanisms can coexist in this balsam pear-shaped CuO. Finally, the prepared substrate has been applied for the determination of trace levels of paraquat in solution , and results show that its LOD for paraquat is 275 μg L −1 (optimum Raman band: 1646 cm −1 Raman shift), which is better than the government standard in China. A dexterous and facile way for fabrication of CuO SERS-active substrates with low cost and high performance, quite promising in detection of chemically hazardous substances and pesticide residue is provided.
