Exogenous electron mediators (EMs) can facilitate extracellular electron transfer (EET) via electron shuttling processes, but it is still unclear whether and how biofilm formation is affected by the presence of EMs. Here, the impacts of EMs on EET and biofilm formation were investigated in bioelectrochemical systems (BESs) with Shewanella oneidensis MR-1, and the results showed that the presence of five different EMs led to high density current production. All the EMs substantially promoted biofilm formation with 15–36 times higher total biofilm DNA with EMs than without EMs, and they also increased the production of extracellular polymeric substances, which was favorable for biofilm formation. The current decreased substantially after removing EMs from the medium or by replacing electrodes without biofilm, suggesting that both biofilm and EMs are required for high density current production. EET-related gene expression was upregulated with EMs, resulting in the high flux of cell electron output. A synergistic mechanism was proposed: EMs in suspension were quickly reduced by the cells and reoxidized rapidly by the electrode, resulting in a microenvironment with sufficient oxidized EMs for biofilm formation, and thus, besides the well-known electron shuttling process, the EM-induced high biofilm formation and high Mtr gene expression could jointly contribute to the EET and subsequently produce a high density current. This study provides a new insight into EM-enhanced current production via regulating the biofilm formation and EET-related gene expression.

Microbial fuel cells with mixed bacteria always show better power output capacity than those with single bacteria due to synergetic effects between species. Here, a rational-designed synthetic three-species alliance system based on Shewanella oneidensis MR-1 (the model exoelectrogen), Pseudomonas aeruginosa, and Lactobacillus plantarum was constructed upon process optimization, the three-species alliance system in MFC delivered a maximum power density of ∼ 207 mW/m 2 by using the glucose as the substrate, which was 2 ∼ 58 times higher than that of MFC inoculated with single species. In addition, the synthetic alliance system could generate electricity from broad substrate spectrum (lactose, saccharose, maltose, arabinose, cellobiose) with enhanced performance. Electrochemical and metabolism analysis indicated inter-species synergetic enhancement on charge transfer, substrate utilization, and biofilm formation was the underlying mechanism for performance improvement in MFC by this synthetic alliance system. In this system, L. plantarum uses glucose to produce lactic acid as the electrogenic substrate and electron donor of S. oneidensis MR-1, and P. aeruginosa uses glucose to produce another electron shuttle phenazine, which can also promote the growth of S. oneidensis MR-1 biofilm and help the alliance system to transfer electrons better. This work demonstrated a new strategy to enhance the MFC performance with a rational designed synthetic bacterial community, which would further renew the toolbox for MFC optimization.

The immobilized anthraquinone-2-sulfonate (iAQS) could significantly promote anaerobic biotransformation of the contaminants. During this process, the role of flavins secreted by bacteria remains unclear. In the present study, mutual interaction between extracellular flavins and AQS-modified polyurethane foam (AQS-PUF) during the reduction of azo dye Acid Red 18 and 3-nitrobenzenesulfonate (3-NBS) was investigated. Results showed that the amount of extracellular flavins secreted by Shewanella sp. RQs-106 was positively correlated with the concentration of iAQS ranging from 10 to 100 μM. The presence of iAQS resulted in the increased concentration of extracellular and intracellular flavins, implying that iAQS could induce the synthesis and secretion of flavins. The deletion of gene bfe encoding the flavin adenine dinucleotide exporter resulted in approximately 63.8% decrease in the amount of extracellular flavins. Further analysis showed that the decreased amount of extracellular flavins could contribute to around 50.8% reduction of iAQS. Moreover, around 23.2% and 34.0% decreases were observed in AQS-PUF-mediated removal rates of AR 18 and 3-NBS by mutant lacking bfe gene, respectively, compared with that by wild type strain RQs-106. These results indicated that the secreted flavins played an important role in the bio-reduction of AQS-PUF, resulting in their contribution to AQS-PUF-mediated removals of high-polarity aromatic compounds containing nitrogen.

Specific spoilage organisms (SSOs) play a crucial role in causing the deterioration of aquatic products. This study investigated the antibacterial activity and mechanism of exogenous autoinducers through biochemical and metabolomics analysis against Shewanella putrefaciens . The potential application of exogenous autoinducers on large yellow croaker preservation was further investigated. C4-HSL, C10-HSL and C14-HSL with the strongest antibacterial activity on S. putrefaciens were selected, from the aspects of growth curve, biofilm formation, and motility ability, which damaged the integrity of cell membrane. Metabolomics analysis demonstrated C10-HSL significantly affected key metabolic pathways including alanine, aspartate and glutamate metabolism, glycine, serine and threonine metabolism, arginine biosynthesis, phenylalanine metabolism, tricarboxylic acid (TCA) cycle, pyrimidine and purine metabolism. Furthermore, C10-HSL exhibited the most effective preservation performance for large yellow croaker. These findings highlighted the antibacterial mechanism of exogenous acylated homoserine lactones (AHLs) and confirmed the potential of exogenous AHLs to prolong the shelf-life of aquatic products.

Large quantities of low-concentration uranium-contaminated water from the nuclear industry poses a critical environmental challenge due to the limitations of current methods on removal efficiency and adsorption selectivity. This study developed a novel biofilm cathode for enhanced uranium adsorption by leveraging the quorum sensing (QS) system of Shewanella oneidensis MR-1. Biofilm formation was induced using exogenous acyl-homoserine lactones (AHLs), with C4-HSL (10 μmol/L) identified as the most efficient signal molecule. It significantly improved biofilm properties, increasing thickness by 107.7 % versus the control without AHLs, boosting extracellular protein content, and increasing the proportion of living cells. Microstructure analysis (CLSM) revealed a “base-polysaccharide-gel” three-dimensional structure, where proteins, lipids, and β-polysaccharides form a functional gel layer that provides the primary functional matrix for uranium binding. The C4-HSL-induced biofilm exhibited a 43.4 % higher reduction peak current and a 33.3 % decrease in electron transfer resistance, confirming improved electron transfer efficiency. Furthermore, coupling CLSM, SEM-EDS, FT-IR and XPS analysis indicated uranium capture was primarily dominated by complexation/coordination with functional groups on extracellular polymeric substances (EPS), supplemented by electrochemical reduction of 25.91 % soluble U(VI) to insoluble U(IV). Accordingly, a possible mechanism model of QS-driven biofilm cathode is proposed for optimizing biofilm structure and enhancing uranium capture. Finally, verification tests in actual uranium-contaminated groundwater demonstrated the biofilm cathode exhibited exceptional performance with a 99.4 % uranium recovery rate and high selectivity (K d,U at 34.57 L/g). These findings highlight the significant potential of AHLs-triggered QS as a powerful strategy to optimize bioelectrochemical properties for highly-efficient uranium remediation and resource recovery.

The strong spoilage capability and cold adaptability of Shewanella putrefaciens present a serious threat to the quality and safety of fish. This study aimed to investigate the synergic antibacterial effects of six phenolic acids (Gallic acid, GA; Caffeic acid, CA; Protocatechuic acid; Ferulic acid; Chlorogenic acid; p-Coumaric acid) in pairs against S . putrefaciens , and to illustrate the antibacterial mechanisms. The fractional inhibitory concentration index (FICI) of 15 combinations of phenolic acids against S. putrefaciens ranged from 0.5-0.75, suggesting synergistic or partial synergistic antibacterial effects of these phenolic acids. Time-kill curves and morphologic observation demonstrated that GA and CA (GC) exhibited the strongest synergistic effect against S. putrefaciens . GC synergistically damaged the structure integrity of S. putrefaciens , leading to the leakage of intracellular substances and inhibiting the activity of Na + K + -ATPase and respiratory chain dehydrogenase. Both GA and CA could bind to DNA and cause changes in DNA content. Proteomics analysis revealed that 399 proteins were affected in the GC groups, a number significantly higher than that in the single GA and CA groups. GC inhibited dissimilatory nitrate and sulfate reduction, and enhanced fatty acid degradation and assimilatory sulfate reduction, which might cause the imbalance of energy supply and ROS accumulation. Meanwhile, the antioxidant-related enzymes, efflux systems and some two-components systems were upregulated, while chemotaxis were inhibited. GC synergistically inhibited S. putrefaciens in turbot, and retarded the spoilage process. The results revealed multi-targeted synergistic antibacterial mechanisms of GC against S. putrefaciens , suggesting GC might be a good strategy for controlling S. putrefaciens and fish spoilage.

Probing of the single-cell level extracellular electron transfer highlights the maximum output current for microbial fuel cells (MFCs) at hundreds of femtoampere per cell, which is difficult to achieve by existing devices. Past studies focus on the external factors for boosting charge-extraction efficiency from bacteria. Here, we elucidate the intracellular factors that determine this output limit by monitoring the respiratory-driven shrinking kinetics of a single magnetite nanoprobe immobilized on a single Shewanella oneidensis MR-1 cell with plasmonic imaging. Quantified dissolving of nanoprobes unveils a previously undescribed bio-current fluctuation between 0 and 2.7 fA on a ∼40 min cycle. Simultaneously tracing of endogenous oscillations indicates that the bio-current waves are correlated with the periodic cellular electrokinesis. The unsynchronized electron transfer capability in the cell population results in the mean current of 0.24 fA per cell, significantly smaller than in single cells. It explains why the averaged output current of MFCs cannot reach the measured single-cell currents. This work offers a different perspective to improve the power output by extending the active episodes of the bio-current waves.

Hypoxia and lactate-overexpressed tumor microenvironment always lead to poor therapeutic effect of radiotherapy. Here, platinum nanoparticles-embellished hafnium metal-organic framework (Hf-MOF-Pt NPs) were elaborately integrated with Shewanella oneidensis MR-1 (SO) to construct an engineered biohybrid platform (SO@Hf-MOF-Pt) for enhancing radiotherapy. Benefiting from the tumor-targeting and metabolic respiration characteristics of SO, SO@Hf-MOF-Pt could enrich in tumor sites and continuously metabolize the overexpressed lactate, which specifically downregulated the expression of hypoxia-inducible factor (HIF-1α), thereby relieving the radiosuppressive tumor microenvironment to some extent. Moreover, SO@Hf-MOF-Pt would react with tumor-overexpressed hydrogen peroxide (H 2 O 2 ) to generate oxygen (O 2 ) and further inhibit the expression of HIF-1α, resulting in the downregulation of lactate dehydrogenase (LDHA) and subsequently reducing the lactate production. Under these multiple cascaded effects, the radiosuppressive tumor microenvironment was significantly reshaped, thus potentiating the radiosentization of SO@Hf-MOF-Pt and remarkably amplifying the therapeutic outcomes of radiotherapy. The designed biohybrid SO@Hf-MOF-Pt represented promising prospects in sensitizing radiotherapy via bacterium-based metabolic regulation.

Cell’s activities are highly dependent on signal molecules, of which reactive oxygen species of the superoxide anion (O2•–) and hydrogen peroxide (H2O2) are important ones that always work together to regulate biological processes such as apoptosis and oxidative stress. It is of significance to realize simultaneous qualification of O2•– and H2O2 but it still faces challenges particularly in live-cell assay with a complex environment. We report the design of a bifunctional sensing material by interfacially regulating ZIF-67 on bacteria Shewanella putrefaciens to generate cobalt nanoparticles/nitrogen-doped porous carbon nanorods (Co/N-doped CNRs) and its sensing chip for qualifying cell-released O2•– and H2O2. Co/N-doped CNRs exhibit unique properties including porous structure for significantly increased reaction surface area and coordinating Co nanoparticles for rich active sites. The bifunctional Co/N-doped CNRs is used to fabricate the electrochemical sensing chip, which achieves a fast response time (0.5 s for O2•–, 1.9 s for H2O2), a low detection limit (0.69 nM for O2•–, 2.25 μM for H2O2), and a remarkably high sensitivity (792.30 μA·μM–1·cm–2 for O2•–, 153.91 μA·mM–1·cm–2 for H2O2), among the best of reported bifunctional nanozymes.

An additive- and pollution-free method for the preparation of biogenic silver and silver chloride nanoparticles ([email protected]NPs) was developed from the bacteriaShewanellasp. Arc9-LZ, which was isolated from the deep sea of the Arctic Ocean. The optimal synthesizing conditions were explored, including light, pH, Ag+concentration and time. The nanoparticles were studied by means of ultraviolet-visible (UV-Vis) spectrophotometry, energy dispersive spectrometry (EDS), X-ray diffraction (XRD) and inductively coupled plasma optical emission spectrometers (ICP-OES). The transmission electron microscope (TEM) showed that the nanoparticles were spherical and well dispersed, with particle sizes less than 20.00 nm. With[email protected]nanoparticles, the kinetic rate constants for congo red (CR) and rhodamine B (RhB) dye degradation were 2.74 × 10−1min−1and 7.78 × 10−1min−1, respectively. The maximum decolourization efficiencies of CR and RhB were 93.36% and 99.52%, respectively.[email protected]nanoparticles also showed high antibacterial activities against the Gram-positive and Gram-negative bacteria. The Fourier transform infrared spectroscopy (FTIR) spectrum indicated that the O-H, N-H and -COO- groups in the supernatant of Arc9-LZ might participate in the reduction, stabilization and capping of nanoparticles. We mapped the schematic diagram on possible mechanisms for synthesizing[email protected]NPs.Keywords:[email protected]nanoparticles;synthesis mechanisms;Shewanellasp.;dye reduction;antibacterial activity

The excessive lactate in the tumor microenvironment always leads to poor therapeutic outcomes of chemotherapy. In this study, a self-driven bioreactor (defined as SO@MDH, where SO is Shewanella oneidensis MR-1 and MDH is MIL-101 metal–organic framework nanoparticles/doxorubicin/hyaluronic acid) is rationally constructed via the integration of doxorubicin (DOX)-loaded metal–organic framework (MOF) MIL-101 nanoparticles with SO to sensitize chemotherapy. Owing to the intrinsic tumor tropism and electron-driven respiration of SO, the biohybrid SO@MDH could actively target and colonize hypoxic and eutrophic tumor regions and anaerobically metabolize lactate accompanied by the transfer of electrons to Fe3+, which is the key component of the MIL-101 nanoparticles. As a result, the intratumoral lactate would undergo continuous catabolism coupled with the reduction of Fe3+ to Fe2+ and the subsequent degradation of MIL-101 frameworks, leading to an expeditious drug release for effective chemotherapy. Meanwhile, the generated Fe2+ will be promptly oxidized by the abundant hydrogen peroxide in the tumor microenvironment to reproduce Fe3+, which is, in turn, beneficial to circularly catabolize lactate and boost chemotherapy. More importantly, the consumption of intratumoral lactic acid could significantly inhibit the expression of multidrug resistance-related ABCB1 protein (also named P-glycoprotein (P-gp)) for conquering drug-resistant tumors. SO@MDH demonstrated here holds high tumor specificity and promising chemotherapeutic efficacy for suppressing tumor growth and overcoming multidrug resistance, confirming its potential prospects in cancer therapy.

A comodified strategy was developed to connect inorganic catalysts with conjugated polymers and outer-membrane proteins, where an in situ growth method was used to stimulate synergistic interactions fully and thus enhance photocatalytic performance of the biocatalysts. The biocatalyst bCdS/PA@MR-1 decolored low-concentration dyestuff wastewater continuously, with decoloring completed within 15 min. Under visible-light irradiation, the photoelectrons from cadmium sulfide and bioelectrons in bCdS/PA@MR-1 formed a fast electron flow, and electron transport occurred in the comodified structure. Moreover, the separation of photoholes can effectively promote the properties of bioelectrons produced by the bacteria Shewanella oneidensis MR-1 that were used to clean the photoholes to prevent damage or corrosion, thereby ensuring efficient decolorization of anthraquinone and azo dyes. After decolorization in dyes, the activity of bCdS/PA@MR-1 was rejuvenated, with the OD600 value reaching 92.7% of that of the initial, illustrating advantages such as long shelf life and sustained renewable catalytic activity of the bCdS/PA@MR-1 biocatalyst.
