This study aimed to determine whether selection by antibiotics, chlorhexidine (CHX) and Rhizoma coptidis extract (RCE) would lead to cross-resistance or decreased susceptibility in Staphylococcus aureus . The S. aureus reference strain ATCC 25923 as well as 14 clinical isolates were exposed to antibiotics, CHX and RCE at sublethal doses for up to 14 days. Changes in susceptibility were determined by analysis of minimum inhibitory concentrations (MICs). All isolates were cross-resistant to more than one other antibiotic following tetracycline exposure, and increased resistance (≥4-fold MIC increase) to RCE and CHX was observed in six and three isolates, respectively. Following selection by CHX, most of the treated strains showed no significant change in sensitivity to CHX. However, all strains developed cross-resistance to at least one antibiotic, and decreased susceptibility (≥4-fold MIC increase) to RCE appeared in seven strains. Following exposure to RCE, 11 isolates showed cross-resistance to at least one antibiotic. In addition, three RCE-exposed strains showed reduced susceptibility to CHX (4- or 8-fold MIC increase). The results obtained in this study imply that antibiotics, biocides and antimicrobial Chinese herbs might employ some of the same mechanisms of action against bacteria, triggering mutual cross-resistance to further foster the development of bacterial resistance.

Aimed at decreasing the environmental pollution caused by petroleum-derived packing materials, many efforts have been directed towards fabricating biodegradable substitutes, especially of cellulose-based film with functional properties. In this paper, a totally plant-derived antibacterial film was successfully prepared. Regenerated cellulose films were used as substrate and poacic acid, a decarboxylated product from 8–5-diferulic acid, as fungicides. The physical properties and thermal stability of the prepared plant-derived antibacterial films were characterized, and the antibacterial activity was also assessed as the function poacic acid contents. The morphology showed that the cellulose/poacic acid films exhibited a homogeneous and smooth surface and poacic acid was successfully incorporated into cellulose films as revealed by Fourier-transform infrared (FT-IR) spectroscopy and fluorescent microscopy. It was noted that both the tensile strength and thermal stability of the antibacterial films decreased due to the incorporation of the poacic acid. Remarkably, the prepared cellulose/poacic acid films exhibited superior antibacterial activity against Staphylococcus aureus . The notable properties of the cellulose/poacic acid films are promising for applications in food and medical packaging to enhance their safety.

To develop a specific method for the detection of S. aureus , chicken anti-protein A IgY was adopted for specifically capturing S. aureus , depending on the specific recognition of staphylococcal protein A (SPA) by chicken anti-protein A IgY, which can eliminate the interference from protein G-producing Streptococcus . HRP labeled IgG , Fc region of which has a high affinity towards SPA, was paired with IgY for the colorimeter analysis of the system. By optimizing the system, a super-low detection limit of 11 CFU of S. aureus in 100 μL PBS without enrichment, with a linear range from 5.0 × 10 2 CFU mL −1 to 5.0 × 10 4 CFU mL −1 was obtained. The entire assay was accomplished in less than 90 min and no cross-reactivity with the other tested bacterial species was observed. Moreover, the developed assay has been applied for the detection of S. aureus in three different types of real samples (sodium chloride injection, apple juice and human urine) with satisfactory results. To the best of our knowledge, it is the first time to report using chicken anti-protein A IgY and any IgG to detect S. aureus based on the dual-recognition mode of SPA. The novel method opened up a way for monitoring S. aureus in food samples with high sensitivity, specificity and simple operation.

Staphylococcus aureus ( S . aureus ) is the leading cause of a broad range of human infections and associated with food poisoning worldwide. Nisin can be considered as a natural preservative and is widely used by the food industry. However, resistance of nisin has been reported in several species of bacteria. Identification of antibacterial agents that are synergistic with nisin is a method of minimising resistance to nisin. Cinnamaldehyde (CA), a well-studied essential oil isolated from cinnamon bark, has been reported to exhibit a broad spectrum of antimicrobial activity across a wide range of microorganisms. Previous studies have demonstrated that CA could be used in the food industry due to its active antibacterial activity and its safety. In this study, the synergistic antibacterial activity in vitro of nisin and CA against 13 food-borne isolates of S. aureus and S. aureus ATCC 29213 were evaluated by a checkerboard assay. The FICI values of nisin combined with CA ranged from 0.1875 to 0.375, which demonstrated a synergistic effect. The positive interactions were verified using the challenge tests in pasteurized milk and agar diffusion assays. The LIVE/DEAD BacLight experiment and scanning electron microscope (SEM) assay were also performed to distinguish the intact and damaged both of cell walls and membranes with the compounds, the combination group displayed stronger damage both of cell wall and cell membrane than when nisin or CA was used alone. These findings indicated that the combination of nisin and CA could not only be used as a promising naturally sourced food preservative but also to reduce the problem of bacterial resistance.

AimsThe effects of a new glutaraldehyde–didecyldimethylammonium bromide combination disinfectant (GD) on the cell surface of Staphylococcus aureus, a representative Gram‐positive bacterium, were investigated in this study. Methods and ResultsResults of bacterial surface structural analysis showed that GD significantly changed the bacterial morphology. The membrane fluidity decreased and outer membrane permeabilization increased after contact with GD. Furthermore, the integrity of the cytoplasmic membrane was destroyed by over 99% after exposure to GD for a short time. Bacterial ATPase activity correlated negatively with the treatment of GD over time, and proteins were degraded. Assays of intracellular component leakage indicated that GD caused the rapid leakage of K+, Mg2+, ATP molecules, and proteins into the extracellular environment.ConclusionsThe effects of GD against S. aureus are probably attributable to the removal of the permeability barrier, changes in the S. aureus morphology, changes in the structures and functions of the cell membrane, leakage of intracellular substances and disturbance of the intracellular homeostasis. As a result, this irreversible damage accelerated the death of S. aureus.Significance and Impact of the StudyIn an earlier study, the bactericidal mechanism of GD against Escherichia coli was investigated. Hence, this study focused on the action of mechanism of GD against S. aureus. It is important to clarify the disinfectant bactericidal mechanisms of GD against bacterium, in general, and this study provides theoretical support to the prevention of bacterial resistance.

The treatment of infected pressure ulcers (IUPs) requires addressing diverse microenvironments. A pressing challenge is to effectively enhance the regenerative microenvironment at different stages of the healing process, tailoring interventions as needed. Here, a dual enzyme mimetic and bacterial responsive self-activating antimicrobial hydrogel designed to enhance IPUs healing is introduced. This hydrogel incorporates pH-responsive dual enzyme-active nanoplatforms (HNTs-Fe-Ag) encapsulated within a methacrylate-modified silk fibroin (SFMA) and dopamine methacrylamide (DMA) matrix. This composite hydrogel exhibits adaptive microenvironment regulation capabilities. Under the low pH microenvironment of bacterial infection, it has excellent antimicrobial activity by self-activating the •OH generation in conjunction with photothermal effects. Under the neutral and alkaline microenvironment of chronic inflammation, it catalyzes the decomposition of hydrogen peroxide (H 2 O 2 ) to produce oxygen (O 2 ), thereby alleviating hypoxia and scavenging reactive oxygen species (ROS), which in turn remodulates the phenotype of macrophages. The composite hydrogel demonstrates on-demand therapeutic effects in the microenvironment of infected wounds, significantly enhancing the regenerative microenvironment of IUPs by promoting wound closure, inflammation regulation, and collagen deposition through self-activated antimicrobial action during infection and adaptive hypoxia relief during recovery. This approach offers a novel strategy for developing smart wound dressings.

Developing antibiotics-independent antibacterial materials is of great importance for combating drug-resistant bacteria, such as methicillin-resistant Staphylococcus aureus (MRSA). MXene (transition metal carbides and nitrides), a class of novel 2D nanomaterials , has shown great potentials in biomedical areas. However, the effect of MXene size on its properties and bioactivity is still unknown. Herein, we report for the first time that the antibacterial photothermal therapy efficacy of Ti 3 C 2 T x MXene nanosheets is size-dependent. Three MXene suspensions with small size of 196 nm (MX-s), medium size of 347 nm (MX-m) and large size of 497 nm (MX-l) were prepared via ultrasonication. Upon NIR irradiation for 5 min, the temperature of MXene suspensions (10 μg/mL) increased to 64, 60 and 56 °C for MX-s, MX-m and MX-l, respectively. Accordingly, the viability loss of MRSA induced by MX-s, MX-m and MX-l under NIR was 93%, 69% and 56%, respectively. The in vivo study in the MRSA-infected mouse model showed that the photothermal therapy efficacy of MX-s was comparable to that of the positive control vancomycin . This is the first report on the size-dependent photothermal effect and photothermal antibacterial activity of MXene, which may guide the development of MXene-based therapeutics in the future. In addition, the drug-free antibacterial therapy has great implications for the treatment of antibiotics-resistant bacteria infections.

Staphylococcus aureus ( S. aureus ) is a common foodborne pathogen, which could cause serious bacteremia and infective endocarditis. Establishing a rapid and low-cost method to capture and detect S. aureus in food samples is necessary. In this study, we have developed a method on the basis of polyethyleneimine-functionalized magnetic nanoparticles (MNP-PEI) and recombinase aided amplification (RAA) for rapid enrichment and analysis of S. aureus in milk. The capture efficiency of MNP-PEI on multiple strains of bacteria and influencing factors were studied. The limits of detection (LOD) of the MNP-PEI-RAA strategy were 2.9 × 10 1 CFU·mL −1 in phosphate-buffered saline (PBS) and 2.9 × 10 2 CFU·mL −1 in milk. The entire enrichment and detection processes could be completed within 60 min. The detection strategy based on MNP-PEI and RAA had low cost, short detection time, and high sensitivity. In addition, the broad-spectrum of bacteria captured by MNP-PEI was important for simultaneous analysis of a variety of bacteria, which provided an effective tool for evaluating the microbiological safety of food samples.

The evolution of antibiotic resistance poses a great challenge to the bacterial infectious disease therapy. Although antimicrobial peptides and photodynamic therapy have great potential in the treatment of bacteria, their clinical application is limited due to high toxicity and low therapeutic efficiency. Herein, an innovative nano antibacterial agent was designed based on cationic antimicrobial peptides (AMPs) modified with cholesterol succinate to load the hydrophobic photosensitizer TCPP (ACT) for selective killing of S. aureus. The modification of cholesterol succinate significantly increased the positive charge of AMPs while decreasing toxicity and hemolysis. The increased positive charge could capture bacteria through electrostatic interaction, which reduced the distance between the photosensitizer and bacteria to enhance the photodynamic sterilization effect. ACT selectively killed S. aureus, not E. coli, and showed excellent destruction and inhibition ability for S. aureus-formed biofilm. ACT accelerated wound healing by promoting collagen precipitation and angiogenesis after good bacterial clearance in mice with S. aureus-infected wounds. This strategy provides a paradigm for addressing toxicity and broadening the application of antimicrobial peptides and photosensitizers.

A novel antibiotic-affinity strategy was designed for fluorimetric detection of pathogenic bacteria based on the strong affinity of antibiotic agent to the cell wall of bacteria. In this proof-of-concept work, vancocin, a glycopeptide antibiotic for Gram-positive bacteria, was used as a molecular recognition agent to anchor Staphylococcus aureus (S. aureus) cell. To improve the specificity of this method for S. aureus detection, IgG was adopted as the second recognition agent utilizing the binding between Fc region of IgG and S. aureus protein A in the cell wall, to form a sandwich complex. By using fluorescein isothiocyanate as the signal probe, S. aureus whole cells could be directly assayed within a linear range of 1.0 × 103–1.0 × 109 CFU mL–1 with a detection limit of 2.9 × 102 CFU mL–1. The whole assay process could be completed within 130 min when a ready-for-use microplate was adopted. This proposed strategy for pathogenic bacteria detection possessed some attractive characteristics such as high sensitivity, wide linear range, simple manipulation, short assay time, and low cost. Furthermore, this sandwich mode also showed ideal specificity because vancocin and IgG bound with S. aureus at two distinct sites. It opened up a new pathway for high-throughput screening of pathogenic bacteria in medical diagnosis, food safety, bioterrorism defense, and drug discovery.

ABSTRACT In the present study, the antibacterial activity of monocaprylin in comparison with sodium benzoate and potassium sorbate against Staphylococcus aureus and Escherichia coli was assessed by measuring MIC, MBC, effect of pH on MIC, and incubation temperature on bactericidal efficacy. Results showed that monocaprylin exhibited an excellent antibacterial activity against both strains, with the lowest MIC and MBC of 1.28 mg/mL. A MIC of monocaprylin remained unchanged despite the pH values of culture medium, ranging from 5 to 9, unlike that of potassium sorbate or sodium benzoate. Furthermore, monocaprylin at MBC effectively reduced the population of E. coli and S. aureus by >5.5 log CFU/mL at 25°C within 6 h and decreased E. coli by approximately 5.0 log CFU/mL and S. aureus by 2.9 log CFU/mL at 12 h. The underlying mechanism of monocaprylin was then investigated by measuring β-galactosidase activity, membrane potential, release of cellular contents, scanning electron microscopy, and transmission electron microscopy observations. Results indicated that monocaprylin killed E. coli by the rapid change in permeability and integrity of cell membrane, leading to decline of membrane potential, leakage of nucleic acids and proteins, and ultimately cell membrane disintegration and lysis. On the other hand, monocaprylin might exert its antibacterial activity against S. aureus mainly by diffusing across the cell wall, collapsing the cell membrane, and disturbing the order of intracellular contents. These findings indicated that monocaprylin had better antibacterial ability compared with traditional synthetic preservatives and might be a potential antibacterial additive independent of pH.

Skin and soft tissue infections require effective and sustained topical administration. Platensimycin (PTM) is a natural drug lead that targets bacterial fatty acid synthases and has a great potential to treat infections caused by methicillin-resistant Staphylococcus aureus (MRSA). To facilitate the use of PTM against local MRSA infections, we prepared polyacrylamide hydrogels containing polyamidoamine (PAMAM)/PTM nanoparticles (NP-gel(PTM)) for the controlled release of PTM. NP-gel(PTM) can continuously inhibit the growth of MRSA and its biofilm formation in simulated drug flow models in vitro. In situ implantation of NP-gel(PTM) could treat MRSA-infected subcutaneous soft tissues without toxicity. For MRSA-infected skin wounds, NP-gel(PTM) not only showed strong anti-MRSA activity but also accelerated more wound healing than the widely used antibiotic mupirocin. Collectively, PTM is expected to be used in this safe and effective NP-gel delivery platform for the treatment of local infections, which might help to alleviate the current antibiotic resistance crisis.
