Carrageenan-ferrocene-eicosapentaenoic acid composite hydrogel induce ferroptosis and apoptosis for anti-tumor recurrence and metastasis

Xiangyan Chen,Tianze Jiang,Yantao Li,Yifei Zhang,Jianqi Chen,Xia Zhao,Hai Yang

The immune-suppressive microenvironment of solid tumors is a key factor limiting the effectiveness of immunotherapy , which seriously threatens human life and health. Ferroptosis and apoptosis are key cell-death pathways implicated in cancers, which can synergistically activate tumor immune responses. Here, we developed a multifunctional composite hydrogel (CE-Fc-Gel) based on the self-assembly of poloxamer 407 , cystamine-linked ιota-carrageenan (CA)-eicosapentaenoic acid (EPA), and ferrocene (Fc). CE-Fc-Gel improved targeting in tumor microenvironment due to its disulfide bonds . Moreover, CE-Fc-Gel promoted lipid peroxidation, enhanced reactive oxygen species (ROS) production, and decreased glutathione peroxidase 4 (GPX4), inducing ferroptosis by the synergistic effect of Fc and EPA. CE-Fc-Gel induced apoptosis and immunogenic cell death (ICD), thereby promoting dendritic cells (DCs) maturation and T cell infiltration. As a result, CE-Fc-Gel significantly inhibited primary and metastatic tumors in vivo. Our findings provide a novel strategy for enhancing tumor immunotherapy by combining apoptosis, ferroptosis, and ICD.

Development of polyvinyl alcohol/carrageenan hydrogels and fibers via KOH treatment for Morse code information transmission

Shi Feng,Jing Guo,Qiang Yang,Fucheng Guan,Qiang Yao,Yonghe Wang,Fengyu Quan,Shijun Zeng

To solve the inherent problems of conductive hydrogels, such as relatively low mechanical properties and fatigue resistance, inability to work after water loss, and difficulty weaving. In this study, the borax-crosslinked polyvinyl alcohol/k-carrageenan (kC) conducting hydrogels (BPK KOH ) were prepared by a simple one-pot method, and KOH treatment was used instead of the cumbersome and time-consuming freeze-thaw cycle to improve the comprehensive properties. The KOH treatment increased the hydrogel hydrogen bonding content by 7.72 % and synergized with the induction of kC by K + to enhance the tensile and compressive strengths by 8.12 and 34.6 times, respectively. Meanwhile, the BPK KOH hydrogel's fatigue resistance and shape recovery after water loss were improved. Additionally, the BPK KOH hydrogels can be monitored for finger bending, showing clear and stable differences in electrical signals. BPK KOH hydrogels combined with Morse code realize applications in information transmission and encryption/decryption. Notably, introducing KOH ensures the molding and preparation of BPK KOH hydrogel fibers while having good signal responsiveness and monitoring ability. More importantly, it can be woven into fabrics that can be loaded with heavy weights, which has the potential to be directly applied in smart wearables. This work provides new ideas for applying flexible sensors and wearable smart textiles.

Highly conductive and stable double network carrageenan organohydrogels for advanced strain sensing and signal recognition arrays

Xinlong Liu,Longmeng Du,Yong Ma,Tingxi Li,Song Chen,Jia Yang,Zhenzhen Ran,Longbao Zhou,Qi Dong,Wenhui Zheng,Zaixing Jiang

Conductive hydrogels with excellent mechanical properties, a broad detection range, and stability in complex environments have remained a significant challenge for the development of flexible sensors. In this study, a straightforward freeze-thaw cycles strategy was developed to fabricate a polyvinyl alcohol (PVA)/carrageenan (CA)/calcium chloride (CaCl 2 )/MXene-based double network organohydrogel (PCCME) for highly flexible and responsive strain detection across a broad temperature spectrum. The PCCME organohydrogel features multiple interactive forces including hydrogen bonding, ionic interactions, and microphase crystallization, which contribute to the organohydrogel's exceptional mechanical and electrical performance. The PCCME organohydrogel exhibited excellent performance in a load-unload test repeated 100 times after being maintained at room temperature for 7 days, with a minimal mechanical decay of only 2.6%. Furthermore, the repaired PCCME organohydrogel retained its robust stability after storage at low temperatures followed by placement at room temperature. The organohydrogel sensor not only detects various movement amplitudes of the human body but also recognizes arrays of pressure signals and converts these into digital images, highlighting its significant potential for applications in rehabilitation monitoring, pressure sensing, and human-computer interaction.

Preparation of Gelatin/Carrageenan Blend Film and Reinforced by Bagasse Hemicelluloses Extracted by Liquid Hot Water

Qin Chengrong,Wang Cheng,Liu Baojie,Huang Lingzhi,Liang Chen,Yao Shuangquan

The polymer-blend films (PBFs) have the potential to be utilized as packaging material for enhancing food shelf life. But the most important factor restricting the application is low mechanical strength. Recycling hemicellulose can be used as a new value-added product to increase economic efficiency. In this paper, bagasse hemicellulose was extracted by pH pre-controlled hot water extraction. Its utilization in the reinforcement of a gelatin/carrageenan PBFs was studied. The effect of reaction parameters on the tensile strength (TS) and elongation at break (EAB) of PBFs was studied. The most important reaction parameter was hemicellulose dosage. The optimum preparation process of PBFs was determined by the response surface analysis method. The optimum reaction conditions of TS was 34.84 °C, glycerin dosage 1.43%, hemicellulose dosage 0.92%, TS was 54.19 MPa. The optimum reaction conditions of EAB was 34.92 °C, glycerin dosage 1.45%, hemicellulose dosage 0.87%, EAB was 26.42%. The results provide theoretical support for the application of hemicellulose in food packaging.

Biocompatible κ-Carrageenan Gel Films with Clove Essential Oil Nanoemulsion: Optimization, Properties, and Strawberry Preservation Efficacy

Yanhong Liu,Zechen Liu,Yixin Wang,Yujie Ma,Lingling Zhang,Mengyao Liu,Shuo Wang

Emulsion-filled hydrogels have attracted considerable attention due to their potential for inhibiting foodborne pathogens. However, development of packaging films based on essential oil nanoemulsion-filled hydrogels and their efficacy in food preservation remains relatively limited. This study engineered a biocompatible κ-carrageenan (KC)-based gel film loaded with clove essential oil nanoemulsion (CEON), exhibiting excellent dual-function antibacterial and antioxidant properties for preserving strawberries. The results revealed that incorporating CEON dramatically boosted the film's ultraviolet (UV) barrier properties (reducing UV transmittance to 0.02%), antimicrobial activity (producing an inhibition zone diameter of 6.5 cm), and antioxidant activity (demonstrating DPPH/ABTS radical scavenging rates exceeding 90%). Concurrently, the film exhibited remarkable flexibility, with its elongation at break increasing from 4.8% to 35%. The thus-formed unique porous network, characterized by a “hydrophobic surface yet moisture-permeable matrix”, effectively modulated the internal microenvironment. The CEON-loaded KC gel film not only effectively inhibited the growth of spoilage microorganisms on strawberries, extending their refrigerated shelf life to 10 days, but also demonstrated excellent biocompatibility as confirmed by seed germination and cytotoxicity assays. This study provides novel insights for developing high-performance and environmentally friendly food preservation materials.

Effects of Xanthan Gum and Carrageenan on Texture and Structural Properties of Highland Barley Protein Gel

Mingze Fu,Yi Zhang,Xiaowei Peng,Jianquan Kan

Background Highland barley protein (HBP) is a plant-derived protein with a balanced amino acid composition. This study investigated the influence of xanthan gum (XG) and carrageenan (CGN) at varying concentrations (0%–0.5%, w/v) on the gelation properties of HBP. Results Both XG and CGN were found to enhance the water-holding capacity and gel strength of HBP gels. Thermogravimetric analysis demonstrated that their addition improved gel thermal stability, while Fourier-transform infrared (FTIR) spectroscopy confirmed that the secondary structure of HBP remained largely unchanged. Protein solubility tests and free thiol measurements revealed that XG and CGN reduced free thiol levels and promoted disulfide bond formation, with disulfide bonds playing a dominant role in the composite gel network. Microscopic observations showed that XG and CGN facilitated HBP aggregation, with CGN exhibiting a stronger aggregation effect. Conclusion This study offers critical insights into improving the gelation properties of HBP, establishing a theoretical basis for designing novel food systems incorporating barley protein and diverse colloids.

Preparation, structural characterization, and functional attributes of zein-lysozyme-κ-carrageenan ternary nanocomposites for curcumin encapsulation

Jin Chen,Zhuangwei Zhang,Huihui Li,Mengchu Sun,Hongjin Tang

The low water solubility and inadequate bioavailability of curcumin significantly hinder its broad biological applications in the realms of food and medicine. There is limited information currently available regarding the particle characteristics and functional capabilities of zein-lysozyme-based nanomaterials . Thereby, the primary goal of the current work is to effectively develop innovative zein-lysozyme- κ -carrageenan complex nanocomposites (ZLKC) as a reliable carrier for curcumin encapsulation. As a result, ZLKC nanoparticles showed a smooth spherical nanostructure with improved encapsulation efficiency. Fourier-transform infrared, fluorescence spectroscopy, dissociation assay, and circular dichroism analysis revealed that hydrophobic and electrostatic interactions and hydrogen bonding were pivotal in the construction and durability of these composites. X-ray diffraction examination affirmed the lack of crystallinity in curcumin encapsulated within nanoparticles . The incorporation of κ -carrageenan significantly improved the physicochemical stability of ZLKC nanoparticles in diverse environmental settings. Additionally, ZLKC nanocomposites demonstrated enhanced antioxidant and antimicrobial properties, as well as sustained release characteristics. Therefore, these findings demonstrate the potential application of ZLKC nanocomposites as delivery materials for encapsulating bioactive substances.

κ-carrageenan mediated microwave 3D printing of Spirulina: Realizing immediate curing through temperature-dependent phase transition

Dingxuan Xu,Zilong Zhao,Hangwei Zhang,Xiaolin An,Nana Zhang,Bowen Yan,Yu Liu,Yang Zhang,Hongyuan Cheng,Daming Fan

Spirulina, a nutrient rich microalga, is predominantly utilized in powdered form, limiting its direct role in structuring food matrices. This study developed a Spirulina based printable system by incorporating κ-carrageenan and applying microwave 3D printing to achieve rapid solidification and high fidelity shaping. Rheological results showed that κ-carrageenan not only ensured the shear-thinning behavior of the slurry at high temperatures, but also imparted a modulus response that strengthened with decreasing temperature, thereby facilitating smooth extrusion and deposition during microwave 3D printing. Successful printing of complex structures was achieved at κ-carrageenan contents of 2% or higher and temperatures above 60 °C. The textural properties of the printed products, including gel strength, hardness and gumminess, increased with κ-carrageenan content and printing temperature. Meanwhile, the compressive stress increased markedly with both parameters, with distinct yielding behavior emerging under high-temperature conditions with ≥2% κ-carrageenan. Low-field NMR and scanning electron microscopy revealed that microwave-induced synergistic crosslinking further enhanced water binding and network crosslinking density. Finally, numerical simulations revealed the coupling of viscosity, temperature, and shear fields during extrusion, elucidating the synergistic mechanism of κ-carrageenan and microwave heating on the instant solidification and surface fidelity of the printed products.

Composite nanoparticle-filled oxidized hydroxypropyl starch/carrageenan films: Robust, water-resistant, antibacterial, antioxidant and biodegradable properties

Yinjuan Cao,Zhaoyang Song,Wenjing Ni,Yabin Ma,Keqi Xin,Qunli Yu,Li Zhang

In the present study, the structure of potato oxidized hydroxypropyl starch (OHS) was analyzed. Carrageenan (CA) was used as a reinforcing agent for the development of biodegradable food packaging materials . Broccoli leaf polyphenol/soybean protein isolate/hydroxypropyl methylcellulose composite nanoparticles (SPH NPs) were used to reinforce OHS/CA (OC) films, and their physicochemical, structural, antioxidant, antimicrobial, and biodegradable properties were characterized. The OHS had high transparency, low gelatinization temperature (GT), and easy film formation. Intermolecular hydrogen bonds were formed between the SPH NPs and the OC film matrix, but did not change the crystal structure of the matrix. Furthermore, the incorporation of SPH NPs improved the smoothness, densification, and thermal stability of the OC films. Since SPH NPs occupied the pores of the film matrix, the degree of swelling, light, and moisture-blocking properties were improved, causing the films to gradually turn red, yellow and dark. Notably, the tensile strength of the film was significantly increased from 11.06 MPa to 13.61 MPa at a concentration of 3% SPH NPs. The SPH NP 3 -loaded films showed scavenging activity up to 67.62% and 75.45% against DPPH and ABTS free radicals , with good inhibitory activity against S. aureus and E. coli . More importantly, the composite film was almost completely degraded after 45 d of burial in the soil, and it can regulate the soil microbial community . Consequently, OHS composites can be used as a potentially versatile and environmentally friendly film-forming material for food packaging.

Physicochemical characterization and antimicrobial properties of konjac glucomannan/κ-carrageenan composite films filled with curcumin nanoemulsion

Xu Wei,Li Lala,Xin Yingying,Kang Mengyao,Li Penglin,Luo Denglin,Zhang Lihua

This study develops novel antimicrobial camellia oil composite films based on konjac glucomannan (KGM) and κ-carrageenan (KC) incorporated with a curcumin (CUR)-loaded camellia oil nanoemulsion to address food packaging safety and sustainability. The composite solutions and films were systematically characterized. The results demonstrate that the integration of KGM significantly enhanced the rheological characteristics of the composite solution, while also improving the hydrophilicity of the resulting composite film. Thermogravimetric analysis (TGA) revealed that 0.4% KGM elevated the maximum thermal degradation temperature to 250.23 °C, significantly enhancing thermal stability. Fourier transform infrared (FTIR) and X-ray diffraction (XRD) analyses further demonstrated that KGM and KC established a stabilizing network structure through hydrogen bonding interactions. Confocal laser scanning microscopy (CLSM) and scanning electron microscopy (SEM) indicated that KGM concentrations between 0.1% and 0.3% promoted uniform emulsion droplet dispersion and a smooth film surface. It is noteworthy that when the concentration of KGM was 0.1%, the composite film showed a zone of inhibition of 4.64 mm against Escherichia coli and 4.67 mm for Staphylococcus aureus , which were significantly larger than those of other formulations. In summary, this research presents a theoretical framework designed to enhance the development of high-performance, multifunctional antimicrobial packaging materials, with the objective of fostering sustainable food practices.

Effect of Potassium-Ion-Triggered Double Helix Aggregation on Shakedown Behavior of κ-Carrageenan/Polyacrylamide Hydrogel

Xueqi Zhao,Yudong Pan,Zhanrong Zhou,Yang Gao,Aijun Li,Binkai Shi,Jian Hu,Liuying Wang

This study investigates the effect of potassium ion (K+) concentration on double helix aggregation inκ-carrageenan-based hydrogels, which significantly influences their shakedown behavior. The shakedown behavior ofκ-carrageenan/polyacrylamide (PAAm) hydrogels was characterized by the evolution of maximum stress and energy dissipation during cyclic load. The experimental results indicate that higher K+concentrations significantly improve the maximum stress in the steady state, but barely influence the energy dissipation in the steady state. The improved maximum stress can be explained by the higher density of double helix aggregation. The steady energy dissipation elucidates that the K+concentration does not affect the breaking–recovering balance of the sacrificial network in cyclic loading. These results provide mechanistic insights into how ion-triggered double helix aggregation influences the shakedown behavior ofκ-carrageenan-based hydrogels.

Effect of moisture content and Konjac glucomannan on the mechanical properties of κ-carrageenan-derived hydrogels

Zhang Lihua,Zhang Lanlan,Yue Mengge,Zang Jiaxiang,Kang Mengyao,Li Lala,Luo Bo,Xu Wei

κ-Carrageenan (CRG)/konjac glucomannan (KGM) composite gels have promising applications in food texture modification and drug delivery, but are limited by poor mechanical strength. This study investigated the effects of KGM concentration and moisture content on the mechanical properties of these gels. The results show that increasing KGM concentration combined with moisture regulation substantially enhances the mechanical strength of the gels. At a KGM concentration of 0.6% and 60% water loss, the CRG/KGM gels exhibited maximum shear force and tensile strength, increasing 20-fold and 33-fold, respectively, compared to pure CRG gels. Furthermore, the resistance to deformation in the composite gels demonstrated a positive correlation with both KGM concentration and moisture content. Microstructural analyses showed that while KGM did not alter the crystalline structure type within the gels, it promoted cross-linking between CRG and KGM, resulting in a denser gel network with lower water mobility. Scanning electron microscopy further illustrated that increased KGM concentration enhanced the density and cross-linking strength of the gel network, whereas reduced water content contributed to a more compact gel structure. The interplay of these factors improved the mechanical properties of the composite gels. This study provides valuable insights for the rational design of polysaccharide gels.
