Emodin-8-O-β-D-glucopyranoside alleviates cholestasis by maintaining intestinal homeostasis and regulating lipids and bile acids metabolism in mice

Xiaolin Wu,Qiao Yu,Yuzhao Hou,Xuemei Zhang,Simon Sani Ocholi,Liming Wang,Ziping Yan,Jie Li,Lifeng Han

Cholestatic liver disease(CLD) is caused by impaired bile flow due to obstruction of the biliary tract, and long-term exposure to bile acids in the liver triggers inflammation, eventually leading to liver toxicity and liver fibrosis. Emodin-8- O-β -D-glucopyranoside(EG) is anthraquinone compound that is widely found in traditional Chinese medicine. It possessed antioxidative and anti-inflammatory activities. However, the effect of EG on cholestatic liver injury(CLI) has not been explored. In this study, Alpha-naphthyl isothiocyanate(ANIT)-induced CLI mice were used to investigate the anti-cholestasis and hepatoprotective effects of EG through serum biochemical index detection, non-targeted metabolomics, lipidomics, and intestinal flora 16S rRNA sequencing. The results suggested that EG restores homeostasis of the gut microbiome while regulating bile acids metabolism and lipid-related metabolic pathways to reduce liver damage in ANIT-induced cholestasis. This study provides a new perspective on the mechanism of EG, and help offer a more natural approach to managing liver damage.

Imprinted polymer/Fe3O4 micro-particles decorated multi-layer graphite paper: Electrochemical and colorimetric dual-modal sensing interface for aloe-emodin assay

Meng Wang,Xianwen Kan

A novel dual-modal sensing interface was constructed for the electrochemical/colorimetric dual-modal sensing of aloe-emodin (AE). Fe 3 O 4 micro-particles were directly synthesized on a multi-layer exfoliated graphite paper (EGP) by the in-situ hydrothermal approach, which was subsequently modified with a layer of molecularly imprinted polymer (MIP) using pyrrole and aloe emodin (AE) as monomer and template, respectively. Due to the specific recognition ability of MIP, the prepared sensing interface showed a good selectivity to AE. The advantages of the multi-layer EGP, such as the large surface area and the excellent conductivity, enabled the sensing interface to sensitively detect AE based on the electrochemical mode. Moreover, due to the peroxidase-like activity of the modified Fe 3 O 4 micro-particles, 3,3′, 5,5′-tetramethyl-benzidine can be oxidized to deep blue by the prepared sensor in the presence of H 2 O 2 . However, the rebound AE molecules in imprinted cavities blocked the activity of Fe 3 O 4 micro-particles and caused the light blue of color, which achieved the visual colorimetric assay of AE with the good sensitivity and selectivity. With the merits of easy preparation, good selectivity, and high sensitivity, the present electrochemical/colorimetric dual-modal strategy would greatly broaden the practical application of the molecular imprinting.

Silicon Carbide Nanoparticles Induce Pulmonary Fibrosis by Promoting Macrophage Apoptosis through Parkin-Mediated Mitophagy Disruption and the Intervention Effect of Emodin

Sufang Wu,Xianglong Wu,Nuo Xu,Ze Li,Yang Dong,Chenxing Zhang,Qian Liu,Huang Chen,Lele Liu,Yanxin Hu,Gancen Zhang,Yunru Liu,Jin Xu,Aihua Gu

Silicon carbide nanoparticles (SiC-NPs), an emerging class of nanomaterials, have garnered extensive applications in industrial and biomedical fields. The escalating use of SiC-NPs engenders substantial concerns regarding the potential biosafety implications. Previous studies indicate the association between silicon carbide exposure and elevated incidence of pulmonary fibrosis. However, the underlying mechanisms remain insufficiently elucidated. In this study, we investigated the role of SiC-NPs in pulmonary fibrosis development, employing THP-1-derived macrophages (THP-M) for in vitro experimentation and C57BL/6 mice for in vivo analyses. Our study demonstrated that exposure to SiC-NPs led to the production of reactive oxygen species (ROS) and mitochondrial damage in macrophages. This sequence of events impaired mitophagy and subsequently triggered macrophage apoptosis, ultimately contributing to the pathogenesis of pulmonary fibrosis. Mechanistically, we elucidated that SiC-NPs promoted the ubiquitin-mediated degradation of Parkin, resulting in dysregulated mitophagy. Furthermore, we revealed that emodin treatment mitigated SiC-NPs-induced pulmonary fibrosis by restoring Parkin expression. Our study provides novel insights into the detrimental effects and underlying mechanisms of SiC-NPs in pulmonary fibrosis, as well as highlighting the therapeutic potential of emodin.

Evaluating anticancer activity of emodin by enhancing antioxidant activities and affecting PKC/ADAMTS4 pathway in thioacetamide-induced hepatocellular carcinoma in rats

Hanan M. Hassan,Ahmed M. Hamdan,Abdullah Alattar,Reem Alshaman,Omar Bahattab,Mohammed M. H. Al-Gayyar

Emodin is a naturally occurring anthraquinone derivative with a wide range of pharmacological activities, including neuroprotective and anti-inflammatory activities. We aim to assess the anticancer activity of emodin against hepatocellular carcinoma (HCC) in rat models using the proliferation, invasion, and angiogenesis biomarkers. After induction of HCC, assessment of the liver impairment and the histopathology of liver sections were investigated. Hepatic expression of both mRNA and protein of the oxidative stress biomarkers, HO-1, Nrf2; the mitogenic activation biomarkers, ERK5, PKCδ; the tissue destruction biomarker, ADAMTS4; the tissue homeostasis biomarker, aggregan; the cellular fibrinolytic biomarker, MMP3; and of the cellular angiogenesis biomarker, VEGF were measured. Emodin increased the survival percentage and reduced the number of hepatic nodules compared to the HCC group. Besides, emodin reduced the elevated expression of both mRNA and proteins of all PKC, ERK5, ADAMTS4, MMP3, and VEGF compared with the HCC group. On the other hand, emodin increased the expression of mRNA and proteins of Nrf2, HO-1, and aggrecan compared with the HCC group. Therefore, emodin is a promising anticancer agent against HCC preventing the cancer prognosis and infiltration. It works through many mechanisms of action, such as blocking oxidative stress, proliferation, invasion, and angiogenesis.

A sustainable approach to enhance flame retardant, antibacterial and UV resistance of lyocell fabrics treated with luffa phosphate derivatives and emodin

Zhao Jieyun,Su Xiaowei,Jiang Lina,Ren Yuanlin,Liu Xiaohui,Cheng Chunzu

A multifunctional lyocell fabric with flame retardant, antibacterial and ultraviolet (UV)-resistant properties was fabricated. First, a novel and efficient eco-friendly intumescent flame retardant was synthesized by using natural luffa as raw materials, and was applied to lyocell fabric to improve its flame retardancy. Then, the flame retardant lyocell fabric was treated by emodin to simultaneously obtain the antibacterial and UV-resistant properties. The surface morphology and chemical structure of the treated fabric were analyzed by scanning electron microscopy, Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy, which confirmed the P–O–C covalent bond between the flame retardant and the fabric. Thermogravimetric, limiting oxygen index and cone calorimetry analysis demonstrated that the treated fabric possessed excellent char formation ability, thermal stability and flame retardancy. Thermogravimetric/infrared spectroscopy analyses of the treated fabric and the morphological study of the char residue after burning suggested that both gas-phase and condensed-phase mechanism contributed to the flame retardant improvement of lyocell fabric. In addition, after 20 laundering cycles, the treated fabric still maintained excellent flame retardant, antibacterial activity, and UV resistance properties. This study provided an eco-friendly and facile approach to produce lyocell fabric with multifunction of flame retardant, antibacterial and UV resistance. Graphical abstract

Construction of cinnamic acids derived β-cyclodextrins and their emodin-based inclusions with enhanced water solubility, excellent antioxidant and antibacterial activities

Jing Zhang,Nini Li,Dongmei Zhang,Mengjin Dong,Changhao Wang,Yashao Chen

β -Cyclodextrin ( β -CD) is a natural macromolecule which has been regarded as a promising biomaterial and drug carrier. However, the poor water solubility and less functions of β -CD severely hinder its potential applications. Herein, we modified the β -CD with three cinnamic acid derivatives (ADs) of ferulic acid (FA), p -coumaric acid ( p- CA) and caffeic acid (CA) by sulfonylation , amination and amidation. The resulted β- CD-ADs are β- CD-FA, β- CD- p- CA and β- CD-CA, which were confirmed by proton nuclear magnetic resonance ( 1 H NMR) and high resolution mass spectrometry (HRMS). Compared to β -CD, the β- CD-ADs show enhanced water solubility (>600 mg/mL) and excellent antioxidant property in scavenging DPPH radicals. Furthermore, the inclusion complex of between emodin (ED) and β- CD-ADs were prepared and characterized by 1 H NMR, fluorescence spectroscopy , X-ray diffraction (XRD) and differential scanning calorimetry (DSC). The ED/ β- CD-ADs inclusions had antioxidant activity because of chemical modification of cinnamic acid derivatives, especially for ED/ β- CD-CA, which had the highest DPPH radical scavenging rate. The three ED/ β- CD-ADs inclusions showed excellent antibacterial activity against S. aureus with the bactericidal rates beyond 97 %. Additionally, the ED/ β- CD-ADs inclusions exhibited good blood compatibility by in-vitro hemolysis assay. This work provides an efficient way to modify β- CD with enhanced water solubility and more functional that could facilitate the application of β- CD in medicine and cosmetic.

Novel aloe emodin–hydroxyethyl piperazine hybrid dihydrochloride induces oral cancer CAL-27 cells apoptosis through ROS production, DNA damage and mitochondrial pathways

Long Xing,Yang Peilin,Chen Lin,Zhong Wen,Chen Shaofeng,Li Yanping,Lin Shijie,Tian Wei

Oral cancer severely affects the quality of life of patients, and the primary treatment is surgery, due to a lack of specific drugs. Aloe emodin, a natural product of anthraquinone, is considered a potential antitumor drug. However, it has poor water solubility and low biological activity. In this study, we designed and synthesized a novel aloe emodin–hydroxyethyl piperazine hybrid dihydrochloride ( AE-NPC ), which improved the water solubility and antitumor activity of aloe emodin in vitro. According to the results, the IC 50 value of AE-NPC on CAL-27 cancer cells was 14.4 ± 2.9 μM. The flow cytometry results showed that AE-NPC induced the production of reactive oxygen species (ROS) and apoptosis in CAL-27 cells. The spectroscopy and molecular docking results showed that AE-NPC can be well intercalated into DNA grooves. Further molecular mechanism studies revealed that AE-NPC not only regulates the expression levels of key proteins (Bax/Bcl-2, cytochrome C, cleaved caspase-9 and cleaved caspase-3) of the mitochondrial apoptotic pathway but also activates the expression levels of a key protein ( γ -H 2 AX) of the DNA damage pathway. Therefore, AE-NPC induces CAL-27 cell apoptosis via DNA damage and mitochondrial pathways. This study provides new insights into the further development of anthraquinone compounds as potential chemotherapeutic agents for oral cancer treatment. The novel aloe emodin derivative AE-NPC induces oral cancer CAL-27 cells apoptosis through ROS production, DNA damage and mitochondrial pathways. The anthraquinone compound AE-NPC may be a potential agent for the treatment of oral cancer.

Thermosensitive hydrogel with emodin-loaded triple-targeted nanoparticles for a rectal drug delivery system in the treatment of chronic non-bacterial prostatitis

Ye Yan,Zhong Wenzhen,Luo Ruifeng,Wen Hongzhi,Ma Ziyang,Qi Shanshan,Han Xiaoqin,Nie Wenbiao,Chang Degui,Xu Runchun,Ye Naijing,Gao Fei,Zhang Peihai

Background The complex etiology and pathogenesis underlying Chronic Non-Bacterial Prostatitis (CNP), coupled with the existence of a Blood Prostate Barrier (BPB), contribute to a lack of specificity and poor penetration of most drugs. Emodin (EMO), a potential natural compound for CNP treatment, exhibits commendable anti-inflammatory, anti-oxidant, and anti-fibrosis properties but suffers from the same problems as other drugs. Methods By exploiting the recognition properties of lactoferrin (LF) receptors that target intestinal epithelial cells (NCM-460) and prostate epithelial cells (RWPE-1), a pathway is established for the transrectal absorption of EMO to effectively reach the prostate. Additionally, hyaluronic acid (HA) is employed, recognizing CD44 receptors which target macrophages within the inflamed prostate. This interaction facilitates the intraprostatic delivery of EMO, leading to its pronounced anti-inflammatory effects. A thermosensitive hydrogel (CS-Gel) prepared from chitosan (CS) and β-glycerophosphate disodium salt (β-GP) was used for rectal drug delivery with strong adhesion to achieve effective drug retention and sustained slow release. Thus, we developed a triple-targeted nanoparticle (NPs)/thermosensitive hydrogel (Gel) rectal drug delivery system. In this process, LF, with its positive charge, was utilized to load EMO through dialysis, producing LF@EMO-NPs. Subsequently, HA was employed to encapsulate EMO-loaded LF nanoparticles via electrostatic adsorption, yielding HA/LF@EMO-NPs. Finally, HA/LF@EMO-NPs lyophilized powder was added to CS-Gel (HA/LF@EMO-NPs Gel). Results Cellular assays indicated that NCM-460 and RWPE-1 cells showed high uptake of both LF@EMO-NPs and HA/LF@EMO-NPs, while Raw 264.7 cells exhibited substantial uptake of HA/LF@EMO-NPs. For LPS-induced Raw 264.7 cells, HA/LF@EMO-NPs can reduce the inflammatory responses by modulating TLR4/NF-κB signaling pathways. Tissue imaging corroborated the capacity of HA/LF-modified formulations to breach the BPB, accumulating within the gland's lumen. Animal experiments showed that rectal administration of HA/LF@EMO-NPs Gel significantly reduced inflammatory cytokine expression, oxidative stress levels and fibrosis in the CNP rats, in addition to exerting anti-inflammatory effects by inhibiting the NF-κB signaling pathway without obvious toxicity. Conclusion This triple-targeted NPs/Gel rectal delivery system with slow-release anti-inflammatory, anti-oxidant, and anti-fibrosis properties shows great potential for the effective treatment of CNP.Graphical Abstract

Emodin Ameliorates Renal Damage and Podocyte Injury in a Rat Model of Diabetic Nephropathy via Regulating AMPK/mTOR-Mediated Autophagy Signaling Pathway

Hong Liu,Quan Wang,Ge Shi,Wenqiang Yang,Yanmin Zhang,Weidong Chen,Sheng Wan,Fei Xiong,Zengsi Wang

Purpose The activation of autophagy has potential protective effect on diabetic nephropathy (DN) podocyte injury, and the AMPK/mTOR signaling pathway is an important regulatory pathway of autophagy. Emodin has been reported to effectively delay DN progression; however, the therapeutic mechanisms involved in vivo remain ambiguous. The present study aimed to elucidate the mechanism of emodin in improving renal tissue and podocyte injury in DN by regulating the AMPK/mTOR-autophagy signaling pathway. Methods All rats were divided into 4 groups: a Sham group, a Vehicle group, a low-dose emodin (LD-Emo) group (20 mg/kg/day) and a high-dose emodin (HD-Emo) group (40 mg/kg/day). The different doses of Emo and distilled water were daily administrated for 8 weeks after the induction of DN by the unilateral nephrectomy combined with intraperitoneal injections of streptozotocin (STZ). The rats' general status, blood glucose, biochemical parameters, urinary protein excretion, renal histological changes and cell apoptosis in renal tissue, as well as the key protein expressions in the AMPK/mTOR signaling pathway and apoptosis-related proteins were examined, respectively. Results Emodin ameliorated the general condition, kidney weight and urinary protein excretion of the rats, but has little influence on serum biochemical parameters and did not lower blood glucose; emodin attenuated renal fibrosis including the cell numbers, extracellular matrix rate and collagen area in glomerulus, simultaneously relieved podocyte foot process fusion, up-regulated the expression of nephrin protein and suppressed glomerular and tubular epithelial cell apoptosis. In addition, emodin can induce and enhance autophagy in podocytes including increased expression of LC3-II/I, Beclin-1, p-AMPK protein and decreased expression of p62, p-mTOR protein, as well as increased autophagosomes in podocytes. Conclusion We have demonstrated that emodin, as a natural regulator in vivo, reduced proteinuria and alleviated renal fibrosis without affecting hyperglycemia in DN rats. The potential mechanisms by which emodin exerts its renoprotective effects in vivo are through suppressing cell apoptosis and enhancing autophagy of podocytes via the AMPK/mTOR signaling pathway in the kidney.

β-1,3-d-glucan particles-based “nest” protected co-loaded Rhein and Emodin regulates microbiota and intestinal immunity for ulcerative colitis treatment

Yanli Wang,Siwei Zhong,Ke Yang,Ruifeng Luo,Linxin Dai,Wenzhen Zhong,Yan Ye,Chaomei Fu,Dasheng Lin,Nan Li,Jianping Chen,Chuan Zheng,Shu Fu,Fei Gao

Herein, a β-1,3- D -glucan based yeast cell wall loaded with co-loaded nanoparticles of Rhein (RH) and Emodin (EMO), was developed for the combined treatment of ulcerative colitis (UC) by modulating gut microbiota and the Th17/Treg cell balance. This was achieved through an oral “nano-in-micro” advanced drug delivery system. Specifically, RH was grafted onto the HA chain via disulfide bonds to synthesize a reduction-sensitive carrier material and then used to encapsulate EMO to form nanoparticles with a specific drug ratio (denoted as HA-RH/EMO NPs). As anticipated, HA-RH/EMO NPs were encased within the “nests”-yeast cell wall microparticles (YPs), efficiently reach the colon and then released gradually, this occurs mainly due to the degradation of β-1,3- D -glucan by β-glucanase. Additionally, HA-RH/EMO NPs demonstrated a significant reduction-sensitive effect in GSH stimulation evaluations and a remarkable ability to target macrophages in in vitro cell uptake studies. Notably, HA-RH/EMO NYPs reduced inflammatory responses by inhibiting the PI3K/Akt signaling pathway. Even more crucially, the oral delivery and drug combination methods significantly enhanced the regulatory effects of HA-RH/EMO NYPs on gut microbiota and the Th17/Treg balance. Overall, this research marks the first use of YPs to encapsulate two components, RH and EMO, presenting a promising therapeutic strategy for UC.

Rapid separation and enrichment of emodin and rhein in Sanhuang Xiexin Decoction by molecularly imprinted nanoparticles and evaluation of DPP-4 inhibitory activity

Chuangchuang Cao,Yue Li,Jingjing Qi,Dandan Liu,Xinyi Feng,Youqi Zong,Yufei Wang,Xiaoyu Wu,Guotai Wu,Wenda Duan

To achieve efficient separation of emodin and rhein, a double-template Fe₃O₄-imprinted polymer (DT-MIP@Fe₃O₄) was successfully synthesized. The characterization (FT-IR, XRD, TGA, SEM, TEM, VSM) demonstrated that the synthesized DT-MIP@Fe₃O₄ exhibited a core-shell architecture, exhibiting thermal stability up to 270 °C and superparamagnetic characteristics, facilitating separation in 8 s. This material exhibited significant adsorption capacities (31.7823 mg/g and 15.8942 mg/g) and quick adsorption kinetics (equilibrium achieved within 35 min), coupled with the benefits of facile separation, high selectivity, and exceptional reusability. DT-MIP@Fe₃O₄ was utilized as an adsorbent for magnetic solid-phase extraction (MSPE) in conjunction with high-performance liquid chromatography (HPLC) for the simultaneous separation and detection of emodin and rhein in the Chinese herbal medicine SXD. The optimization of parameters influencing MSPE-HPLC was conducted, such as adsorbent dosage, type and volume of desorbent, desorption time, and salt effect. The established method demonstrated exceptional linearity (R² > 0.9999) across the 0.1–100 mg/L, exhibiting high sensitivity and stability. The limits of detection (LOD) and the limits of quantification (LOQ) were established at 0.0092–0.02 mg/L and 0.0307–0.0668 mg/L, respectively. Moreover, the separated fractions demonstrated remarkable hypoglycemic efficacy by DPP-4 inhibition. These findings confirmed the effectiveness of the strategy and indicated that it has considerable prospects in the rapid separation of rhein and emodin from complex Chinese herbal compound, providing a method for the search of natural DPP-4 inhibitors.

Emodin alleviates severe acute pancreatitis-associated acute lung injury by decreasing pre-B-cell colony-enhancing factor expression and promoting polymorphonuclear neutrophil apoptosis

Hongzhang Cui,Shu Li,Caiming Xu,Jingwen Zhang,Zhongwei Sun,Hailong Chen

The present study aimed to evaluate the protective effects of emodin on severe acute pancreatitis (SAP)‑associated acute lung injury (ALI), and investigated the possible mechanism involved. SAP was induced in Sprague‑Dawley rats by retrograde infusion of 5% sodium taurocholate (1 ml/kg), after which, rats were divided into various groups and were administered emodin, FK866 [a competitive inhibitor of pre‑B‑cell colony‑enhancing factor (PBEF)] or dexamethasone (DEX). DEX was used as a positive control. Subsequently, PBEF expression was detected in polymorphonuclear neutrophils (PMNs) isolated from rat peripheral blood by reverse transcription‑quantitative polymerase chain reaction and western blotting. In addition, histological alterations, apoptosis in lung/pancreatic tissues, apoptosis of peripheral blood PMNs and alterations in the expression of apoptosis‑associated proteins were examined by hematoxylin and eosin staining, terminal deoxynucleotidyl‑transferase‑mediated dUTP nick end labeling assay, Annexin V/propidium iodide (PI) assay and western blotting, respectively. Serum amylase activity and wet/dry (W/D) weight ratios were also measured. An in vitro study was also conducted, in which PMNs were obtained from normal Sprague‑Dawley rats and were incubated with emodin, FK866 or DEX in the presence of lipopolysaccharide (LPS). Apoptosis of PMNs and the expression levels of apoptosis‑associated proteins were examined in cultured PMNs in vitro by Annexin V/PI assay and western blotting, respectively. The results demonstrated that emodin, FK866 and DEX significantly downregulated PBEF expression in peripheral blood PMNs. In addition, emodin, FK866 and DEX reduced serum amylase activity, decreased lung and pancreas W/D weight ratios, alleviated lung and pancreatic injuries, and promoted PMN apoptosis by regulating the expression of apoptosis‑associated proteins: Fas, Fas ligand, B‑cell lymphoma (Bcl)‑2‑associated X protein, cleaved caspase‑3 and Bcl‑extra‑large. In addition, the in vitro study demonstrated that emodin, FK866 and DEX significantly reversed the LPS‑induced decrease of apoptosis in PMNs by regulating the expression of apoptosis‑associated proteins. In conclusion, the present study demonstrated that emodin may protect against SAP‑associated ALI by decreasing PBEF expression, and promoting PMN apoptosis via the mitochondrial and death receptor apoptotic pathways.
