Stability assessments of coloration and functionalization of cationic cotton fabric using flavonoid-rich extracts from Zanthoxylum bungeanum pericarps

Huiyu Jiang,Wei Zeng,Hongwen Mei,Wen Zhang,Xing–Xiang Ji

This study investigates the use of flavonoid-rich extracts from Zanthoxylum bungeanum pericarps (ZBPFs) for sustainable dyeing and functionalization of cationized cotton fabrics. The extracts were obtained through ethanol/water extraction and purified using macroporous resin, with flavonoids such as rutin, astragalin, coumarin, baicalin, and quercetin making up over 72 % of the total composition. Stability tests revealed that ZBPFs were stable under neutral or mildly acidic conditions but were highly sensitive to reducing agents, prolonged light exposure, and metal ions. Cotton fabrics modified with 3-chloro-2-hydroxypropyl trimethylammonium chloride and dyed with ZBPFs reached a maximum color depth (K/S value) of 12.5 at 5 % owf. Fabrics dyed with 2 % owf ZBPFs demonstrated excellent UV protection (UPF of 115.54), antioxidant activity (98.1 % radical scavenging), and antibacterial efficacy (over 98 % against E . coli and S . aureus ). The adsorption of ZBPFs onto cationized cotton fitted both the pseudo-second-order and Sips models. This indicates that chemical adsorption is the predominant mechanism and suggests multiple interactions between the ZBPFs and the fabric. The dyed fabrics exhibited high stability in rubbing and washing fastness, although their light fastness was comparatively lower. Further, washing with potassium aluminum sulfate significantly diminished the antioxidant and antibacterial properties, while washing with other solutions, such as tap water and deionized water, showed minimal effects on performance. This research highlights the potential of ZBPFs as a stable, bioactive, and environmentally friendly dye, offering considerable functional and ecological benefits in textile applications.

Production of high antioxidant activity flavonoid monoglucosides from citrus flavanone with immobilised α-L-rhamnosidase in one step

Liu Aolu,Huang Baohua,Lei Lin,Lu Yu-Jing,Zhou Jin-Lin,Wong Wing-Leung

The rhamnosyl group of naringin dihydrochalcone, neohesperidin dihydrochalcone, naringin and hesperidin was selectively removed by enzymatic hydrolysis using an immobilised α-L-rhamnosidase. Monoglycosylated products, including trilobatin, hesperetin dihydrochalcone-7-O-glucoside, prunin and hesperetin-7-O-glucoside, were isolated and characterised by 1H and 13C NMR and ESI-MS. To optimise the enzymatic reaction conditions and its process costs, the hydrolysis of neohesperidin dihydrochalcone to produce trilobatin was selected as a model reaction. Using a ratio of neohesperidin dihydrochalcone: immobilised α-L-rhamnosidase equal to 1:0.6, the trilobatin yields was over 98%. The recycle of enzyme was also investigated, obtaining trilobatin with a yields of 80% even when the twentieth reaction cycle was conducted. Moreover, antiradical and antimicrobial activities of the obtained flavonoid monoglucosides were examined by DPPH, FRAP and ORAC methods, and compared with the efficacy of parental flavonoid glycosides and their aglycone. The results highlight that some of the obtained flavonoid monoglucosides show significant improvement in the antioxidant activity.

Metabolomics-guided valorization of microbial-enzyme co-fermented mulberry branches and leaves: Flavonoid biotransformation and improved antioxidant capacity

Jing Huang,Encun Du,Jiping Liu,Na Zhao,Wanzheng Guo,Fang Chen,Qiwen Fan,Wenjing Tao,Jiang Deng,Jintao Wei

Fermentation represents a sustainable bioprocess for upgrading the utilization value of underutilized agricultural by-products, such as mulberry branches and leaves (MBL). This study applied microbial-enzyme co-fermentation to MBL under simulated industrial conditions, using Lactiplantibacillus plantarum , Saccharomyces cerevisiae , Bacillus subtilis , and a composite enzyme cocktail, and analyzed dynamics in nutrients, bioactive compounds, antioxidant capacity, microbial communities, and flavonoid metabolites. Results showed that fermentation significantly improved nutritional profiles with CP content peaking at day 18 (17.93% relative to 14.30% unfermented MBL, 25.38% increase, p < 0.05) and fiber content reaching the lowest level at day 22 (NDF 49.99% relative to 57.07%, 12.41% reduction; ADF 28.90% relative to 30.45%, 5.09% reduction, p < 0.05). Meanwhile, antioxidant capacity was remarkably elevated, with the hydroxyl radical scavenging rate peaking at day 14 (138.94% increase, p < 0.05). Mechanistically, Lactiplantibacillus rapidly dominated the community and mediated the conversion of flavonoid glycosides to highly active aglycones via glycoside hydrolysis, which directly boosted antioxidant capacity; as fermentation proceeded, these aglycones were further modified to methoxylated flavonoids with improved structural stability. Overall, microbial-enzyme co-fermentation enhanced MBL’s nutritional and functional properties. Fermented MBL is a promising high-quality functional feed, alleviating food-feed competition and advancing sustainable circular agricultural bioeconomy.

Antihyperuricemic Effect of Hawthorn Flavonoid Vitexin: LC–MS Analysis, Network Pharmacology, and In Vivo Verification of Molecular Mechanisms

Shu-Yu Wu,Kai-Wen Kang,Dan-Yu Huang,Shu-Ying Li,Hao Zheng,Ri-Hui Wu,Hui-Qing Lv,Wan-Bin Song,Jing Chen,Cheng-Ping Wen,Li-She Gan,Xiao Yuan

This study identified vitexin as one of the primary active flavonoids in hawthorn that ameliorate hyperuricemia by targeting the PPARγ/ABCG2 pathway. UPLC–MS/MS analysis indicated 17 flavonoid structures in total flavonoids of hawthorn (TFH), while SwissADME calculation then suggested vitexin as the prime candidate for further mechanistic and pharmacodynamic validation. Network pharmacology and molecular docking predicted PPARγ/ABCG2 as one of the key pathways for these flavonoids, which was confirmed by favorable binding with both PPARγ and ABCG2 and stable complex formation of vitexin with PPARγ in molecular dynamics studies. This hypothesis was validated in an HUA mouse model. Vitexin treatment effectively reduced serum uric acid by inhibiting hepatic XOD and ADA activities, and more notably, markedly upregulating renal and intestinal ABCG2 expression to near-normal levels, along with modulation of OAT1 and GLUT9, thereby significantly enhancing urate excretion. The compound also alleviated HUA-induced liver, kidney, and intestinal injuries, restored epithelial barrier integrity, and reduced oxidative stress and inflammation. These findings demonstrate that vitexin exerts multi-target protective effects against HUA by simultaneously suppressing urate production, enhancing its excretion, and mitigating tissue damage. This mechanism of action is consistent with the predicted pathways and supports its potential as a natural therapeutic candidate.

Genome-Wide Transcriptome Analysis Reveals GRF Transcription Factors Involved in Methyl Jasmonate-Induced Flavonoid Biosynthesis in Hedera helix

Feixiong Zheng,Zhangting Xu,Xiaoji Deng,Xiaoyuan Wang,Yiming Sun,Xiaoxia Shen,Zhenming Yu

Flavonoids are key bioactive compounds in plants that play important defense roles against abiotic stress and are involved in plant growth and development. Methyl jasmonate (MeJA) is a significant growth regulator that promotes the accumulation of flavonoids in a variety of plants, but the effect of MeJA inHedera helixremains poorly understood. In the present study, the flavonoid content was significantly increased after MeJA treatment and peaked at 6 h post-treatment. A total of 31,931 genes were identified using transcriptome, and 6484 DEGs were identified at 6 h post-treatment. Through GO and KEGG enrichment analysis, it was shown that DEGs were primarily enriched in phenylpropanoid biosynthesis pathways. Based on the putative transcription factors derived from DEGs, growth-regulating factor (GRF), a transcription factor potentially linking MeJA signaling to flavonoid accumulation and participating in plant growth and stress responses, was further identified. A total of 20 Hh-GRFd genes were identified on the whole genome level and clustered into five phylogenetic groups with conserved subfamily characteristics. Abundant MeJA-responsive cis-elements were presented in the promoter regions ofHhGRF1-HhGRF20. They exhibited a tissue-specific expression variation, andHhGRF10was dominantly expressed in leaves ofH. helix. Notably,HhGRF10exhibited MeJA-induced expression that correlated temporally with flavonoid accumulation, suggesting thatHhGRF10might play a potential role in promoting flavonoid biosynthesis, and overexpression and knockout assay substantiated this conclusion. The finding provides the first transcriptome-wide resource for flavonoid biosynthesis inH. helixand identifies the candidate GRF-mediated regulator for flavonoid accumulation.

Optimization of Ultra-High-Pressure Extraction and Comparative Evaluation of Antioxidant Activity and Flavonoid Composition in Guanxiang Leaves

Jinyan Zhang,Jiyao Kang,Haoqian Yan,Yang Zhang,Zhenhu Guo,Lihui Liu,Guifeng Zhang,Yu Zhang

This paper aimed to optimize the ultra-high-pressure extraction process of flavonoids from guanxiang leaves, evaluate their antioxidant capacity, and identify the flavonoids. Methods: Guanxiang leaves were used as the raw material. The extraction process was optimized through single-factor experiments and a response surface methodology. Ascorbic acid (Vc) was employed as a positive control, and the in vitro antioxidant activity of flavonoids was assessed by determining the DPPH radical-scavenging rate, ABTS radical-scavenging rate, and FRAP ferric-reducing ability. Results: The optimal extraction conditions were determined as follows: ethanol concentration of 50%, solid-to-liquid ratio of 1:60 (g/mL), extraction pressure of 300 MPa, and pressure-holding time of 3 min. Under these conditions, the yield of guanxiang leaf flavonoids was found to be 2.21%, better than the solvent extraction method (1.94%). The antioxidant test results indicated that the IC50values of the extract from the ultra-high-pressure treatment of guanxiang leaves were 11.8 and 50.72 μg·mL−1on DPPH and ABTS scavenging, which were lower than those of the solvent’s extract (13.40 and 54.29 μg·mL−1). Moreover, the antioxidant ability could also be confirmed by Fe3+ion reduction. Mass spectrometry results indicate that ultra-high-pressure extraction can yield components such as spiraeoside, scutellarin, luteolin, and nepetin, which are not present in solvent extraction methods. Conclusions: Therefore, the ultra-high-pressure extraction method can help to improve the flavonoid yield and antioxidant activity as well as to obtain special product types, including spiraeoside, scutellarin, luteolin, and nepetin, than the solvent extraction method.

A Novel Triphenylamine-Based Flavonoid Fluorescent Probe with High Selectivity for Uranyl in Acid and High Water Systems

Bing Liu,Wenbin Cui,Jianliang Zhou,Hongqing Wang

Developing a fluorescent probe for UO22+, which is resistant to interference from other ions such as Cu2+and can be applied in acidic and high-water systems, has been a major challenge. In this study, a “turn-off” fluorescent probe for triamine-modified flavonoid derivatives, 2-triphenylamine-3-hydroxy-4H-chromen-4-one (abbreviated to HTPAF), was synthesized. In the solvent system of dimethyl sulfoxide:H2O (abbreviated to DMSO:H2O) (v/v= 5:95 pH = 4.5), the HTPAF solution was excited with 364 nm light and showed a strong fluorescence emission peak at 474 nm with a Stokes shift of 110 nm. After the addition of UO22+, the fluorescence at 474 nm was quenched. More importantly, there was no interference in the presence of metal ions (Pb2+, Cd2+, Cr3+, Fe3+, Co2+, Th4+, La3+, etc.), especially Cu2+and Al3+. It is worth noting that the theoretical model for the binding of UO22+to HTPAF was derived by more detailed density functional theory (DFT) calculations in this study, while the coordination mode was further verified using HRMS, FT-IR and1HNMR, demonstrating a coordination ratio of 1:2. In addition, the corresponding photo-induced electron transfer (PET) fluorescence quenching mechanism was also proposed.

The Contents of Terpene Trilactone and Flavonoid in Leaves of Seedlings from Ancient Female Ginkgo Trees in China

Yanhui ZHANG,Peiying LIU,Qianqian KONG,Shiyan XING,Xiaojing LIU,Limin SUN

Flavonoids and terpene trilactones, especially, ginkgo flavonglycosides, ginkgolides and bilobalides in leaves of ginkgo trees, need to be studied for effective application of these active components with high medical and health-care values. This study was aimed to provide scientific bases for genealogies selection and harvest season confirmation for Ginkgo biloba . A high-performance liquid chromatographic method (HPLC-ELSD) was developed to determine the contents of terpene trilactone and flavonoid of 36 ancient G. biloba genealogies from 19 provinces in China. The study indicated that the content gradually increased from April to August, and thereafter declined. Analysis of variance indicated that the contents of terpene trilactone, flavonoids, and their respective components had significant difference among 36 genealogies. The cluster analysis showed that No. 72 (Xing'an, Guangxi), No. 58 (Youyang, Chongqing), No. 82 (Rugao, Jiangsu), No. 123 (Huixian, Gansu), No. 99 (Dujun, Guizhou), No. 10 (Tai'an, Shandong) and No. 133 (Mentougou, Beijing) genealogies have higher content of terpene trilactone and flavonoid. These results can help us to select superior variety containing high content of terpene trilactone and flavonoid.

Extraction and identification of new flavonoid compounds in dandelion Taraxacum mongolicum Hand.-Mazz. with evaluation of antioxidant activities

Wang Rong,Li Weihua,Fang Cao,Zheng Xinxin,Liu Chao,Huang Qing

Due to the interest in the potential pharmacological application of dandelion, the chemical constituents and activities of Taraxacum mongolicum Hand.-Mazz were studied. Box–Behnken response surface methodology was employed to optimize the protocol for extraction of flavonoid from dandelion. The molecular structures of different flavonoid compounds were acquired and analyzed by liquid chromatography-mass spectrometry (LC–MS) and nuclear magnetic resonance (NMR) spectroscopy. Several major flavonoid compounds were isolated and purified, namely, hesperetin-5′-O-β-rhamnoglucoside, hesperetin-7-glucuronide, kaempferol-3-glucoside, baicalein, hyperseroside, which were extracted for the first time from dandelion. Hesperetin-5′-O-β-rhamnoglucoside was identified as a new type of flavonoid that had never reported in the literature. This new flavonoid has outstanding antioxidant activity, as shown by its IC 50 value (8.72 mg/L) for scavenging DPPH free radicals. The determination of the structure-related antioxidant activities could be interpreted based on DFT calculations. As such, we have not only illustrated the rich flavonoid contents in Taraxacum mongolicum Hand.-Mazz , but also revealed new types of flavonoid compounds in dandelion in terms of structure and antioxidant properties.

Henghui Zhang,Xiaoli Wang,Dongliang He,Dongliang Zou,Runzhu Zhao,Huifang Wang,Shuying Li,Yongping Xu,Buhailiqiemu Abudureheman

In the present work, the extraction process of total flavonoids (TFs) fromX. sorbifoliaflowers by ultrasound-assisted extraction was optimized under the response surface methodology (RSM) on the basis of single-factor experiments. The optimal extraction conditions were as follows: ethanol concentration of 80%, solid–liquid ratio of 1:37 (g/mL), temperature of 84 °C, and extraction time of 1 h. Under the optimized conditions, the extraction yield of the TFs was 3.956 ± 0.04%. The radical scavenging capacities of TFs against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) were much greater than that of rutin. The results of antibacterial experiments indicated that the TFs displayed strong inhibitory activities onE. coli,S. aureusandBacillus subtilis. Therefore,X. sorbifoliaflowers can be used as a novel source of natural flavonoids, and the TFs have potential applications as natural antioxidants or antibacterial agents in the food and pharmaceutical industries. Keywords:Xanthoceras sorbifoliaBunge;total flavonoids;ultrasound-assisted extraction;response surface methodology;antioxidant activity;antibacterial activity

Effect of crosslink structure on mechanical properties, thermal stability and flame retardancy of natural flavonoid based epoxy resins

Yonggang Du,Ge Zhao,Gonghui Shi,Yiming Wang,Wei Li,Shitong Ren

Three novel bio-based epoxy resins were synthesized based on natural flavonoids (daidzain, naringenin and luteolin). Maleic anhydride was used as curing agent. The effect of crosslink structure on mechanical properties, thermal stability and flame retardancy of bio-based epoxy resins was investigated. The results showed that the mechanical properties of bio-based epoxy resins were significantly better than that of di-glycidyl ether of bisphenol A, but the flame retardancy of bio-based epoxy resins was not significantly improved compared with di-glycidyl ether of bisphenol A. The cycloaddition reaction of benzopyrone ring is conducive to the formation of high crosslink density . The mechanical properties and thermal stability of bio-based epoxy resins increased with increasing crosslink density. The impact strength and initial thermal degradation temperature of cured daidzein epoxy resin which with the highest crosslink density were 0.86 kJ/m 2 and 22 °C higher than that of di-glycidyl ether of bisphenol A, respectively. The glass transition temperature of cured daidzein epoxy resin was 184.4 °C.

A PLGA/Silk Fibroin Nanofibre Membrane Loaded with Natural Flavonoid Compounds Extracted from Green Cocoons for Wound Healing

Xiang Chen,Jiaqi Liu,Yaru Lu,Huijun Liu,Lan Cheng,Zhi Li,Fangyin Dai

The use of biodegradable materials combined with natural metabolites in wound dressings has received much attention. Flavonoids (FLs) from green cocoons, as metabolites, have antibacterial, antioxidant, anti-inflammatory, and other pharmacological effects. In this study, composite membranes of FL-loaded polylactic glycolic acid (PLGA)/silk fibroin (SF) were prepared by an electrospinning method. The prepared membranes, including SF, exhibited a good slow-release effect and cytocompatibility. An in vitro evaluation of the FL-loaded PLGA/SF membranes demonstrated good antioxidant, antibacterial, and anti-inflammatory properties. Animal experiments showed that the wound healing rate of PLGA/SF-2.5FL membranes within 15 days was 97.3%, and that of the control group was 72.5%. The PLGA/SF-2.5FL membranes shortened the inflammatory period of a full-layer wound model and promoted skin regeneration and wound healing by downregulating expression of the pro-inflammatory cytokines IL-1β and TNF-α and promoting expression of the growth factors VEGF, TGF-β, and EGF. In summary, the PLGA/SF-2.5FL composite nanofibre membrane with anti-inflammatory properties is an ideal wound dressing to promote acute wound healing.
