Integrated hydrogel of fucoidan and rhCol III for bioprosthetic heart valves to promote the antithrombosis, anti-inflammatory, and anti-calcification properties

Kaiyang Huang,Cheng Zheng,Xueyu Huang,Bangquan Wei,Lepeng Chen,Gaocan Li,Li Yang,Yunbing Wang

Glutaraldehyde cross-linked bioprosthetic heart valves (BHVs) have been widely used in clinical practice for its superior hemodynamic properties and significantly reduced requirement on anticoagulant therapy. However, the lifespan and biocompatibility of BHVs remain limited due to thrombus accumulation, poor endothelialization, inflammation, and progressive calcification. In this study, we introduced heparin-like fucoidan and recombinant humanized type III collagen (rhCol III) into glutaraldehyde cross-linked BHVs (GLUT) through photoinduced polymerization, thereby preparing an integrated hydrogel functionalized BHV (Fu-rhCol III). This multifunctional hydrogel formed a biocompatible barrier, providing robust protection for internal valve fibers and exhibiting strong resistance to platelet adhesion and thrombus formation in vitro. The Fu-rhCol III effectively reduced the cytotoxicity of GLUT, accelerating the endothelialization process. Fu-rhCol III maintained the structural stability and mechanical properties brought by glutaraldehyde cross-linking. This composite hydrogel demonstrated the significant suppression of acute inflammatory responses and satisfactory anti-calcification effect in subcutaneous implantation test. 60-day implantation results indicated that calcium deposition of Fu-rhCol III decreased by 96% compared with GLUT. In summary, this integrated hydrogel modification provides a promising strategy for future design of BHVs.

Improved bioavailability of polydatin and its protective effect against cisplatin induced nephrotoxicity through self-assembled fucoidan and carboxymethyl chitosan delivery system

Yinghan Wang,Pengyu Xue,Liang Gao,Xuefei Wang,Shilin Zhou,Xiaochen Wu,Chuanlong Guo

Cisplatin induced acute kidney injury (AKI) is clinically prevalent, with a complex pathogenesis and a lack of effective therapeutic drugs. Polydatin (Po) has excellent biological activity, but its low solubility and bioavailability limit its application. In this study, fucoidan (Fu) and carboxymethyl chitosan (Cs) self-assembled into nanoparticles through electrostatic interactions/hydrogen bonding and loaded Po (Fu/Cs Po NPs). In vitro studies found that Fu/Cs Po NPs protected human renal tubular epithelial (HK-2) cells from cisplatin induced damage and accumulation of reactive oxygen species (ROS). Mechanistic studies showed that Fu/Cs Po NPs inhibited cisplatin induced DNA damage and activation of cyclic guanosine monophosphate synthase (cGAS) and intron gene stimulator (STING) pathways. In vivo studies showed that Fu/Cs Po NPs treatment alleviated cisplatin induced AKI symptoms, including elevated blood urea nitrogen (BUN) and serum creatinine (SCr), as well as pathological damage to kidney tissues. In vivo mechanism studies also showed that Fu/Cs Po NPs treatment inhibited cisplatin induced DNA damage and activation of the cGAS-STING pathway. The pharmacokinetic and tissue distribution results demonstrated that the Fu/Cs delivery system enhanced the bioavailability and kidney accumulation of Po in vivo . In summary, our study provided potential drugs for the treatment of cisplatin induced AKI.

Inhibitory effects of fucoidan from Laminaria japonica against some pathogenic bacteria and SARS-CoV-2 depend on its large molecular weight

Xiaona Sun,Chunqing Ai,Chengrong Wen,Haoran Peng,Jingfeng Yang,Yuna Cui,Shuang Song

Fucoidan is a highly sulfated polysaccharide with a wide range of bioactivities, including anti-pathogenic activity. However, the relationship between structure and activity of fucoidan in inhibiting pathogen infections remains unclear. Here, different-molecular-weight fucoidans were prepared by photocatalytic degradation followed by membrane ultrafiltration, and their chemical structures and anti-pathogenic microbiota activity were compared. Results showed that photocatalytic degradation could effectively degrade fucoidan while its structure block and sulfate groups were not destroyed obviously. Fucoidan (90.8 kDa) of 5 mg/mL could inhibit the growth of S. aureus , S. typhimurium and E. coli , but its degradation products, Dfuc1 (19.2 kDa) and Dfuc2 (5.5 kDa), demonstrated lower inhibitory effect. In addition, compared to Dfuc1 and Dfuc2, fucoidan showed stronger capability to prevent the adhesion of S. aureus , L. monocytogenes , V. parahaemolyticus and S. typhimurium to HT-29 cells. Moreover, the inhibitory effect against SARS-CoV-2 and the binding activity to S protein were also positively correlated to molecular weight. These results indicate that natural fucoidan with higher molecular weight are more effective to inhibit these pathogenic bacteria and SARS-CoV-2, providing a better understanding of the relationship between structure and activity of fucoidan against pathogenic microbiota.

Fucoidan from Undaria pinnatifida prevents vascular dysfunction through PI3K/Akt/eNOS-dependent mechanisms in the L-NAME-induced hypertensive rat model

Xiaofei Li,Jian Li,Zhike Li,Ying Sang,Yunhui Niu,Qianying Zhang,Hong Ding,Shanye Yin

Despite major scientific advances in its prevention, treatment and care, hypertension remains a serious condition that might lead to long-term complications such as heart disease and stroke. The great majority of forms of hypertension eventually result from an increased vasomotor tone activity that is regulated by endothelial NOS (eNOS) in vascular endothelium. Here, we examined the effect of fucoidan on eNOS activation in human umbilical vein endothelial cells (HUVECs). We also examined the effects of functional components of Undaria pinnatifida fucoidan on blood pressure and vascular function in eNOS inhibition-induced hypertensive rats in vivo. Our results suggest that fucoidan increased nitric oxide production by activating eNOS and Akt phosphorylation, which could be impaired by Akt or eNOS inhibitors. In the hypertensive rat model, treatment of fucoidan resulted in potent and persistent reduction of high blood pressure (BP) even after drug withdrawal. Our results showed that the mechanisms might involve protection against vascular structure damage, enhanced endothelium-independent vascular function and inhibition of abnormal proliferation of smooth muscle cells, which are mediated by the Akt–eNOS signaling pathway. Moreover, fucoidan treatment reduced the vascular inflammation and oxidative stress control caused by iNOS expression. Together, these results support a putative role of fucoidan in hypertension prevention and treatment.

Low molecular fucoidan alleviated alcohol-induced liver injury in BALB/c mice by regulating the gut microbiota-bile acid-liver axis

Yiyun Sun,Qiuyue Men,Xiaomeng Ren,Chunhong Yan,Shuang Song,Chunqing Ai

Fucoidan has attracted significant attention owing to its remarkable bioactivities, but the effect of molecular weight (Mw) on its activities in the context of alcoholic liver diseases (ALD) is poorly understood. In this study, low Mw fucoidan (OSLF) was prepared, and its protective effect against alcohol-induced liver injury was assessed in a mouse model. OSLF increased weight gain and colon length, improved lipid disorders, and reduced oxidative stress in mice exposed to alcohol, alleviating liver injury. OSLF alleviated inflammation in the liver by inhibiting alcohol-activated NF-κB and MAPK pathways. The underlying mechanism can be attributed to the improvement of alcohol-induced dysbiosis of the gut microbiota, including a decrease in Proteobacteria and Bacteroidetes and an increase in microbiota diversity, as well as the abundances of Parabacteroides , Bacteroides , and Faecalibaculum . Metabolomics results showed that OSLF improved alcohol-induced abnormalities of microbiota metabolites, primarily involving amino acid metabolism and short chain fatty acids production. In addition, OSLF ameliorated bile acid metabolism in the gut and regulated the expression of bile acid-associated genes in the liver, affecting bile acid synthesis, regulation, and transport. It suggested that OSLF had the potential for the management of ALD by regulating the gut microbiota-bile acid-liver axis.

Fucoidan-based dual-targeting mesoporous polydopamine for enhanced MRI-guided chemo-photothermal therapy of HCC via P-selectin-mediated drug delivery

Gaofeng Shu,Lin Shen,Jiayi Ding,Junchao Yu,Xiaoxiao Chen,Xiaoju Guo,Enqi Qiao,Yaning Chen,Chenying Lu,Zhongwei Zhao,Yongzhong Du,Minjiang Chen,Jiansong Ji

The development of novel theranostic agents with outstanding diagnostic and therapeutic performances is still strongly desired in the treatment of hepatocellular carcinoma (HCC). Here, a fucoidan-modified mesoporous polydopamine nanoparticle dual-loaded with gadolinium iron and doxorubicin (FMPDA/Gd 3+ /DOX) was prepared as an effective theranostic agent for magnetic resonance imaging (MRI)-guided chemo-photothermal therapy of HCC. It was found that FMPDA/Gd 3+ /DOX had a high photothermal conversion efficiency of 33.4% and excellent T 1 −MRI performance with a longitudinal relaxivity (r 1 ) value of 14.966 mM −1 · s   −   1 . Moreover, the results suggested that FMPDA/Gd 3+ /DOX could effectively accumulate into the tumor foci by dual-targeting the tumor-infiltrated platelets and HCC cells, which resulted from the specific interaction between fucoidan and overexpressed p-selectin receptors. The excellent tumor-homing ability and MRI-guided chemo-photothermal therapy therefore endowed FMPDA/Gd 3+ /DOX with a strongest ability to inhibit tumor growth than the respective single treatment modality. Overall, our study demonstrated that FMPDA/Gd 3+ /DOX could be applied as a potential nanoplatform for safe and effective cancer theranostics

Targeting 4-1BB with Fucoidan from Fucus vesiculosus: A Strategy to Activate Macrophages and Remodel the Immunosuppressive Microenvironment for Pancreatic Cancer Therapy

Yi Wang,Yabin Ding,Xiaomei Wei,Wenxiu Tian,Wenxiang Zheng,Yongliang Liu,Chuanqin Shi,Xinxin Xiang

Pancreatic ductal adenocarcinoma (PDAC) presents a significant clinical challenge due to its high mortality and resistance to chemotherapy. Over the past decade, fucoidan has been explored as a potential treatment for cancers of the lung, breast, bladder, colon, and pancreas. However, investigations into the efficacy of fucoidan within the tumor microenvironment (TME) of pancreatic cancer are scarce, and its governing mechanisms are not fully understood. Herein, fucoidan (Mfu) from Fucus vesiculosus significantly induced the mRNA expression of inflammatory cytokines (interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-ɑ) by real-time fluorescence quantitative PCR (qPCR) in macrophages. RNA spatial transcriptome sequencing and western blotting showed that Mfu specifically targeted the tumor necrosis factor receptor superfamily member 9 (4-1BB), thereby triggering the TNF signaling pathway in RAW 264.7 cells. Mfu-activated macrophages indirectly elicited the apoptosis and G1-phase arrest of PAN02 cells, as shown by apoptotic protein detection and flow cytometry, suggesting a regulatory effect on pancreatic cancer cell death. In a PDAC allograft model in mice, Mfu (100–400 mg/kg/day, oral gavage) exhibited robust anti-tumor efficacy. Besides, immunohistochemistry (IHC) revealed that Mfu targeted 4-1BB specifically to activate macrophages within the TME. By engaging with the 4-1BB receptor and activating macrophages, Mfu offers a promising strategy to advance the development of pancreatic cancer immunotherapy.

Egg yolk lecithin delivery system constructed by ovalbumin-fucoidan (OVA-FUC) binary complex: Storage stability, release yield and in vitro digestion characteristics

Xiefei Li,Yingmei Wu,Fei Liu,Wenshan Duan,Lei Chen,Lujie Cheng,Yujie Huang,Xin Li,Chao Ai,Qun Huang

Unfavorable environmental factors limit the utilization of egg yolk lecithin in functional foods. Therefore, it is necessary to enhance its stability and effectiveness to cater to the diverse food needs of individuals. This study aimed to explore the feasibility of protecting and delivering egg yolk lecithin through O/W emulsions, which were stabilized using ovalbumin-fucoidan (OVA-FUC) binary complexes as emulsifiers. This study investigated the protective effect of OVA-FUC emulsion on egg yolk lecithin and its impact on delivery by examining the storage stability, in vitro release yield, and stability during in vitro digestion of egg yolk lecithin in the emulsion. The results indicate that egg yolk lecithin, when encapsulated in the OVA-FUC emulsion, exhibits a relatively stable behavior during simulated in vitro digestion. Additionally, encapsulation enhanced both the delivery efficiency and drug release of egg yolk lecithin. Furthermore, the thermal and light stability of egg yolk lecithin embedded in OVA-FUC emulsion was improved, significantly improving its storage performance. The emulsion, prepared using the OVA-FUC binary complex, effectively safeguarded and enhanced the delivery efficiency of bioactive substances as an encapsulating material, thus exhibiting immense potential for application in the active protection and delivery of such substances.

Synthesis and characterization of innovative poly(lactide-co-glycolide)-(poly-L-ornithine/fucoidan) core–shell nanocarriers by layer-by-layer self-assembly

Jingqian Fan,Yuangang Liu,Shibin Wang,Yulu Liu,Siming Li,Ruimin Long,Ran Zhang,Ranjith Kumar Kankala

Layer-by-Layer (LbL) self-assembly of nanocarriers has garnered the interest of researchers for a wide variety of biomedical applications. In this study, we demonstrated the preparation of poly(lactide-co-glycolide) (PLGA)-(poly-L-ornithine (PLO)/fucoidan)4 core–shell nanoparticles (LbL NPs) by a LbL-based self-assembly process, which possessed a mean size of 170 nm. In LbL NPs, a drug carrying PLGA nano-core is coated with alternating PLO and sulfated polysaccharide fucoidan composite films as a shell on the surface. The anti-tumor drug doxorubicin (DOX) loaded into the PLGA core, resulted in better encapsulation efficiency and its in vitro release from LbL NPs demonstrates that this core–shell strategy takes an advantage of its ability to hold the drug cargo and exhibit controlled release. Further, in vitro cell uptake studies by confocal laser scanning microscopy (CLSM) examination in breast tumor cells (MCF-7 cell line) have confirmed that the nanocarriers are successfully internalized and outlined their presence in the cytoplasm after 4 h of incubation. These intracellularly delivered DOX-loaded LbL NPs exhibited significant anti-tumor activity against breast tumor cells. This innovative chemotherapeutic design taking above advantages of successful internalization along with controlled release property signifies as a promising interventional therapeutic delivery system.

Protection and delivery of probiotics by degraded fucoidan-chitosan nanogel-based W1/O/W2 double emulsion incorporated in self-assembled hydrogel particles

Biyang Zhu,Siyu Guo,Peter Chi-Keung Cheung,Kseniya Hileuskaya,Lijun You

Probiotics as pharmaceutical and nutraceutical products are essential to human health, whereas maintaining their viability in adverse environments remains an obstacle. This study presents a well-designed W 1 /O/W 2 emulsion system incorporated in calcium-alginate hydrogel particles for the targeted delivery and sustained release of Lactobacillus bulgaricus in the gastrointestinal tract. Firstly, the degraded fucoidan (DF) with the highest carboxyl content (2.99%), prepared using UV/H 2 O 2 treatment, was more favorable for forming DF-CS nanogel through ionic cross-linking with chitosan (CS). The W 1 /O/W 2 double emulsion fabricated by 0.4 mg/mL DF-CS nanogel possessed the highest interface stability, as concluded from visual appearance, confocal laser scanning microscope (CLSM), droplet size distribution, and ζ-potential results. The double emulsion loaded in 2% alginate hydrogel showed desirable textural properties and swelling behavior, thereby offering a high encapsulation efficiency of Lactobacillus bulgaricus (74.5%). Additionally, the hydrogel delivery system not only achieved sustained release of Lactobacillus bulgaricus in the simulated gastrointestinal condition (>10 7  CFU/g) but also maintained at least 71.3% and 55.1% survival of probiotics at −20 °C and 4 °C, respectively, after 28 d of long-term storage. Overall, these W 1 /O/W 2 double emulsion-filled multilayer self-assembled hydrogel particles can be utilized for better protection and delivery of probiotics in the food and pharmaceutical industries.

A vascular graft with hydrogen-bonded network structure for leak-free puncture resistance, spatiotemporal anticoagulation, and rapid endothelialization via fucoidan-heparin functionalization

Meihong Xu,Enxiang Jiao,Hua Xie,Ziru Sun,Xuehai Zhang,Jinglan He,Peizhao Shi,Chengchen Deng,Hui Tang,Duanqiang Xiao,Huimin Wang,Yuechuan Li,Yanwei Zhao,Guiying Ren,Guiying Li,Taoxia Wang,Haijun Zhang

Small-diameter vascular grafts for vascular access applications face three major challenges: thrombosis, difficulty in endothelialization, and post-puncture blood leakage. Existing vascular grafts struggle to meet these three critical requirements simultaneously. To address these challenges, we designed and fabricated a dual-network vascular graft composed of thermoplastic polyurethane (TPU)/polyvinyl alcohol (PVA) &fucoidan-heparin (TPFH). Using a dual-nozzle electrospinning technique combined with covalent modification, we constructed a functionalized graft with strong hydrogen bonding and microphase separation characteristics, which exhibited self-healing capability after puncture and effectively prevents blood leakage. After undergoing 24 puncture cycles (equivalent to 18 months of clinical use), the mechanical properties of the graft remained superior to those of human coronary arteries, meeting the long-term usage requirements for arteriovenous grafts. In terms of anticoagulant function, fucoidan acted as a fast-response molecule that migrated to the damaged site after puncture to achieve immediate anticoagulation, while the covalently grafted heparin provides stable long-term antithrombotic protection for the whole lumen. Furthermore, the TPFH dual-network structure not only mimics the extracellular matrix but also significantly improves material hydrophilicity, effectively promoting endothelial cell adhesion and proliferation. Subcutaneous implantation in rat models confirmed its good biocompatibility. This study provides a feasible strategy for developing novel vascular grafts with integrated leakage resistance, spatiotemporal anticoagulation, and pro-endothelialization functions, showing promising potential for hemodialysis vascular access.

Cold plasma induced amyloid β-lactoglobulin fibrils-fucoidan self-assembled complexes for lycopene delivery: Structural characterization, emulsion stability, and in vitro release behaviour

Gongshuai Song,Taijiao Xiang,Haina Hou,Danli Wang,Tinglan Yuan,Ling Li,Jianchang Jin,Ziyuan Wang,Fuping Zheng,Mengna Zhang,Jinyan Gong

In this study, a novel delivery system based on self-assembled complexes of amyloid beta-lactoglobulin fibrils (β-LGFs) and fucoidan (FD) was developed to encapsulate and protect lycopene in emulsions. β-LGFs were generated using a cold plasma (CP)-assisted acid-heat induction process under β-LG concentration of 10 mg/mL, CP treatment time of 80 s, and CP power of 60 W. Structural characterization of β-LGFs confirmed the formation of well-defined fibrils and their subsequent interaction with FD at pH 3.0 with a mass ratio of 3:1. The emulsions stabilized by the β-LGF-FD complex exhibited the superior thermal and ultraviolet stability of encapsulated lycopene. The highest retention rate (70.4 %) was obtained in the emulsion with 1.5 wt% β-LGF-FD complex and 50 % oil mass fraction after thermal treatment at 75 °C for 4 h. The increasing the concentration and oil mass fraction could promote the formation of a denser interface layer and more compact gel network structure.
