The scarcity and high cost of Pt-based electrocatalysts for the oxygen reduction reaction (ORR) hinder the practical application of proton exchange membrane fuel cells (PEMFCs). It is critical to replace platinum with non-noble metal electrocatalysts (NNMEs). Carbonized metalloporphyrins represent an important class of NNMEs, but most metalloporphyrins are costly and the corresponding NNMEs do not possess a high ORR activity. Herein, we report that the self-assembly of inexpensive hemin leads to porous nanomaterials in water under ambient conditions and subsequent heat-treatment of the unprecedented nanoporous hemin results in a magnetic NNME with a much enhanced ORR activity compared with directly carbonized hemin without self-assembly. The improvement of the ORR activity likely originates from the exposure of more ORR active sites, caused by the surface area increase of the nanoporous hemin after carbonization over that of micro-scale pristine hemin crystals. Moreover, the ORR activity of heat-treated nanoporous hemin is actually comparable to that of commercial Pt/C in alkaline solution. Additionally, the carbonized nanoporous hemin is much better than commercial Pt/C in terms of durability and tolerance to methanol. This study opens up a new avenue to the production of inexpensive metalloporphyrin-based NNMEs with a high ORR performance by using a self-assembly method in combination with traditional pyrolysis.

A simple, ultrasensitive peroxynitrite anion (ONOO − ) electrochemical sensing platform was developed by immobilizing hemin on a density controllable electrochemically reduced graphene oxide-Au nanoparticles (ERGO-AuNPs) nanohybrids . The ERGO-AuNPs in situ nanohybrids were produced onto a glass carbon electrode (GCE) by one-step electrodeposition, the density of which could be easily controlled by electrodeposited time. The morphology of ERGO-AuNPs nanohybrids was characterized by a scanning electron microscope (SEM). The ERGO-AuNPs nanohybrids showed a high electrocatalytic activity for immobilized-hemin, because the nanostructures hybrids could effectively promote electron transfer rate between hemin and the electrode. Due to nanohybrids-enhanced catalytic effect for hemin, they were firstly selected for use as a highly sensitive electrochemical platform for ONOO − detection. The resulted sensor showed a high electrocatalytic activity toward ONOO − oxidation , being free from the electroactive interferents, including nitrite, nitrate, dopamine and uric acid at an applied potential of 0.7 V. The sensor exhibited a high sensitivity of 123.1 nA μM − 1 and a lower detection limit of 0.1 μM, and a wide linear range of 2.4 × 10 − 6 to 5.5 × 10 − 5 M, which could be attributed to the synergy between ERGO and AuNPs in hybrids. The nanohybrids in situ preparation and ONOO − detection methods would be beneficial to developing other sensing interface and have promising applications in biological molecules analysis and clinical diagnostic.

Clenbuterol (CLB), a kind of β 2 -adrenergic agonists, could disrupt cardiovascular and central nervous system. In this work, a new electrochemical non-enzyme sensor for detecting Clenbuterol (CLB) was fabricated based on MoS 2 -Au-PEI-hemin layered nanocomposites . The two-dimensional (2-D) MoS 2 nanosheets were first in-situ assembled with Au nanoparticles , and polyethylenimine (PEI), then hemin molecules were immobilized onto the MoS 2 -Au-PEI film-modified glassy carbon electrode (GCE) via amide bond. Scanning electron microscopy (SEM) and Zeta potential measurements were employed to characterize the MoS 2 -based nanomaterials . Cyclic voltammetry (CV) was used to investigate electrochemical activity of the immobilized hemin on the modified electrode . Upon the optimum conditions, the proposed electrochemical sensor showed an excellent response for CLB including a wide linear ranging from 10 ng/mL to 2 μg/mL and a detection limit (LOD) of 1.92 ng/mL CLB (S/N=3) with favorable reproducibility and stability. Furthermore, this presented method could be feasible for determining CLB in the real pork samples.

The large-scale synthesis of ternary nanocomposite hemin–graphene sheets/poly(3,4-ethylenedioxythiophene) (H–GNs/PEDOT) was achieved via a microwave-assisted method, in which PEDOT was polymerized with graphene oxide and induced with hemin on the H–GNs nanosheets without additional oxidants; meanwhile, graphene oxide was partially reduced simultaneously. The morphology and structure of the as-prepared nanocomposites were characterized by transmission electron microscopy, Fourier-transform infrared spectroscopy, and ultraviolet-visible absorption spectroscopy. The H–GNs/PEDOT nanocomposite combines the main excellent properties of the three components, maintaining the peroxidase-like activity of hemin, and displaying a high-speed electron transfer ascribed to the presence of PEDOT and the graphene sheets. The H–GNs/PEDOT exhibited remarkable electrocatalysis towards the reduction of H2O2 due to the direct electron-transfer between the hemin and the modified electrode. The excellent synergic effect enables an enhanced electrochemical performance of the H–GNs/PEDOT modified electrode with good biocompatibility and fast redox property. The novel biomimetic H2O2 biosensor based on the well-designed ternary hybrid H–GNs/PEDOT has a low detection limit (0.08 μM) with a high dynamic response range (10−7 to 10−5 M) and a high sensitivity (235 μA mM−1 cm−2). The sensor displays good selectivity, repeatability, reproducibility and stability.

Hemin-functionalized MoS2 nanosheets (hemin/MoS2-NSs) are first obtained via van der Waals interactions between few-layered MoS2 nanosheets (MoS2-NSs) and hemin molecules. It is demonstrated that a portion of MoS2-NSs undergoes a phase transition from semiconducting to metallic phase under the influence of hemin, which shows the coexistence of semiconducting and metallic phases in the crystal structure of hemin/MoS2-NSs. MoS2-NSs prepared from sonication-induced exfoliation of bulk MoS2 crystals in aqueous surfactant solution exhibit intrinsic peroxidase-like activity for the oxidation of 3,3,5,5-tetramethylbenzidine in the presence of H2O2, which is further improved by the functionalization of hemin. Significantly, MoS2-NSs are presented as a new support of hemin, and when compared to MoS2-NSs, hemin/MoS2-NSs exhibit better dispersity in aqueous solution, which is used in the development of H2O2 sensor based on the enhanced peroxidase-like activity.

Oxidative stress mediated secondary injury contributes to neurological deterioration after intracerebral hemorrhage (ICH). Astrocytes, the most dominant cells in the central nervous system (CNS), play key roles in maintaining redox homeostasis by providing oxidative stress defense. Hemoglobin (Hb), the primary component released by hemolysis, is an effective activator of astrocytes. Hemin, the product of Hb degradation, is highly toxic due to the induction of reactive oxygen species (ROS). We speculate that Hb-activated astrocytes are resistant to hemin-induced toxicity. To verify our speculation, Hb-pretreated astrocytes were exposed to hemin, intracellular ROS accumulation and cell apoptosis were evaluated. Heme oxygenase 1 (HO-1) and nuclear transcription factor-erythroid 2 related factor (Nrf2) expression were observed to explore the potential mechanism. The results demonstrated that Hb induced upregulation and nuclear translocation of Nrf2 in astrocytes, resulted in HO-1 upregulation, which contributed to reduced ROS accumulation and apoptosis rate . Knocking down Nrf2 expression by siRNA suppressed Hb-induced upregulation of HO-1 expression and increased the susceptibility of Hb-pretreated astrocytes to hemin-induced toxicity. Taken together, Hb-activated astrocytes acquired resistance to hemin-induced toxicity via Nrf2/HO-1 pathway. This phenomenon can be considered as the adaptive self-defense in the pathological process of ICH. Hb pre-warned astrocytes and enhanced their capability of handling the coming hemin “flood”. Nrf2/HO-1 may be employed as a target for neuroprotection after ICH.

Graphdiyne oxide (GDYO) is a novel type of two-dimensional carbon allotrope nanomaterial consisting of a large conjugated system and excellent chemical stability. To date, application of GDYO as a nanozyme in biosensing has been rarely reported. In this study, a novel ultrasensitive colorimetric bioassay was constructed using a hemin/GDYO nanocomposite (H/GDYO) as a new nanozyme with superior peroxidase-like activity for the detection of H2O2 and glucose. It was discovered that H/GDYO exhibited 6-fold higher peroxidase-like activity than pure hemin. Catalytic kinetic analysis showed that H/GDYO had a much higher affinity for H2O2 and glucose than that of hemin. The designed colorimetric bioassay displayed excellent sensitivity for H2O2 and glucose detection with a wide linear range of 0.015–0.5 mM and 0.1–10 mM, respectively, while the limit of detection (LOD) was as low as 4.39 μM and 38 μM, respectively. Moreover, it was successfully applied for the analysis of H2O2 in milk and glucose in real human serum samples with acceptable recoveries. Importantly, the developed colorimetric bioassay shows good agreement with the results obtained from a commercial blood glucose meter. We believe that the proposed method could provide a promising prospect for medical diagnosis and biotechnology.

Combined photo-induced cancer therapy (photothermal therapy (PTT) and photodynamic therapy (PDT)) is a promising anti-cancer strategy for its non-invasiveness property. However, severe hypoxia in tumor microenvironment (TME) would weaken the PDT effect and limit the clinical efficiency of PTT/PDT treatment. To solve this problem, denatured BSA-hemin-IR780 (dBHI) was designed and synthesized for anti-cancer treatment in this work. Denatured bovine serum albumin (dBSA) was used as the binding matrix to bind hemin and IR780 via covalent reaction and hydrophobic self-assembly, respectively. The obtained dBHI nanoparticles possess both PTT and PDT effect due to the presence of IR780 in the composites. Moreover, because of the existence of hemin, dBHI also exhibited photo-enhanced Fenton-like catalytic performance, which could decompose H 2 O 2 to form oxygen for improving the PDT effect and generate hydroxyl radical (·OH) for chemodynamic therapy (CDT). Therefore, by using dBHI, synergistic effect of PTT/PDT/CDT combined treatment has been achieved for cancer therapy.

Zeolitic imidazolate frameworks (ZIFs), a famous subfamily of metal-organic frameworks (MOFs), are considered promising electrocatalysts. Herein, ZIF-67 was selected as an electrocatalyst for designing electrochemical sensors due to having the best electrocatalytic activity in ZIFs. To overcome the insufficient electrocatalytic activity of ZIFs, ZIF-67 derivatives (QZIF-67-X, where X represents calcination time) were obtained by calcining at 250 °C for a certain time. The porous structure of the precursor in QZIF-67-X is maintained, exposing more active centers. QZIF-67-X could accelerate electron transfer and lead to improve the electrocatalytic performance. Moreover, QZIF-67-2 was chosen as an Au nanoparticle-supported nanocarrier to further bind G-quadruplex/hemin DNAzymes with strong catalytic activity due to the best supporting activity of QZIF-67-2 among QZIF-67-X. The synergistic catalysis of QZIF-67-2 and G-quadruplex/hemin DNAzymes effectively amplified the reduction current signal of H 2 O 2 . The linear range of the prepared electrochemical sensor was 2 μM-65 mM, and the detection limit was 1.2 μM. Moreover, the real-time detection of H 2 O 2 from HepG2 cells was achieved by the sensor, providing a novel technique for efficient anticancer drug evaluation. These results suggested that QZIF-67 can be utilized as an efficient electrocatalyst for improving the sensitivity of sensors.

A dual signal and label-free electrochemical aptasensor for mucin 1 was constructed based on hemin/ [email protected] nanocomposite ( [email protected] ). Hemin attached on the graphene surface not only improves the solubility of graphene and acts as an in-situ electrochemical probe but also exhibits excellent peroxidase-like properties to electrocatalyze the reduction of H 2 O 2 . PdPtNPs also show outstanding catalytic capacity to the reduction of H 2 O 2 and provide numerous binding sites for loading dDNA (mucin 1 aptamer and cDNA) to form the sensing interface. In the presence of mucin 1, due to the specific affinity between aptamer and mucin 1, double helix would be induced dissociation and the aptamer would be pulled off from the electrode. As a result, the electrochemical signals of hemin and H 2 O 2 were recovered. Based on these properties, the label-free and sensitive dual signal electrochemical biosensor for mucin 1 detection has been developed. The one is differential pulse voltammetry (DPV) signal of hemin and the other is chronoamperometry signal arisen from the catalytic reduction of H 2 O 2 . The linear ranges for mucin 1 were 8.0 pg mL −1 to 80 ng mL −1 and 0.8 pg mL −1 to 80 ng mL −1 with the limit of detection 2.5 pg mL −1 and 0.25 pg mL −1 by DPV and chronoamperometry, respectively. The recovery of mucin 1 in human blood serum samples was from 95.0% to 104.2%. The detection platform does not need signal labeling which greatly reduced the sophisticated and expensive procedures. The aptasensor provide a promising strategy for the determination of mucin 1 in clinical diagnostics.

Reconstruction of enzymatic active site in an artificial system is key to achieving high catalytic efficiency. Herein, we report the self-assembly of the lysine-containing peptides with guanine-rich DNA and hemin to form peroxidase-mimicking active sites and catalytic nanoparticles . The DNA strand self-folds into a G-quadruplex structure that provides a supramolecular scaffold and a potential axial ligand for hemin. The β-sheet forming capability of the lysine-containing peptides is found to affect the catalytic synergy between the G-quadruplex DNA and the peptide. It is hypothesized that the β-sheet formation of the peptides results in the enrichment of the lysine residues , which distribute on the distal side of hemin to promote the formation of Compound I, like distal arginine residue in natural heme pocket. Incorporation of the histidine residues into the lysine-containing peptides further enhanced the hemin activities, indicating the cooperation between the lysine and histidine. Furthermore, the peptide/DNA/hemin complexes can be switched between active and inactive state by reversible formation and deformation of the DNA G-quadruplex, which was attributed to the peptides-promoted conformational changes of the DNA components. This work opens an avenue to mimic the catalytic residues and their spatial distribution in the natural enzymes, and shed light on the design of the smart biocatalysts that can respond to the environmental stimuli.

As a disease of high mortality rate, acute lung injury (ALI) has been getting increased attention. Heme oxygenase-1 (HO-1), which plays an essential role in alleviating the pathological symptoms of ALI. There have few studies noted that the improvement effect of HO-1 on ALI may associated to NOD-like receptor protein 3 (NLRP3) inflammasome. However, the roles of HO-1 in the process of NLRP3 inflammasome activation is remains unknown. This study investigated the mechanism that HO-1 blocking NLRP3 inflammasome in ALI. The sepsis model induced by LPS was used in C57BL6 mice, LPS triggered obviously pulmonary damage and edema formation, increased the expression of NLRP3 inflammasome component proteins and IL-1β, but those negative effects were reversed by the pretreatment of hemin. NOD-like receptor signaling pathway was down-regulated after hemin treatment through RNA-Seq. Meanwhile, hemin pretreatment exerted significantly inhibit effect on the activation of NLRP3 inflammasome, and the component proteins of NLRP3 inflammasome were not influenced in immortalization of bone marrow-derived macrophages (iBMDMs). Moreover, the immunoprecipitation and immunofluorescence experiments were indicated that HO-1 could bind to NLRP3, silencing HO-1 prevented the block effect of hemin on NLRP3 inflammasome activation. Our study suggested that hemin-inhibited NLRP3 inflammasome activation involved HO-1 bind to NLRP3 and alleviated ALI mice induced by LPS.
