Luteolin protects against myocardial ischemia/reperfusion injury by reducing oxidative stress and apoptosis through the p53 pathway

Pan Zhai,Xiao-hu Ouyang,Meng-ling Yang,Lan Lin,Jun-yi Li,Yi-ming Li,Xiang Cheng,Rui Zhu,De-sheng Hu

Objective Myocardial ischemia/reperfusion injury (MIRI) is an obstacle to the success of cardiac reperfusion therapy. This study explores whether luteolin can mitigate MIRI by regulating the p53 signaling pathway. Methods Model mice were subjected to a temporary surgical ligation of the left anterior descending coronary artery, and administered luteolin. The myocardial infarct size, myocardial enzyme levels, and cardiac function were measured. Latent targets and signaling pathways were screened using network pharmacology and molecular docking. Then, proteins related to the p53 signaling pathway, apoptosis and oxidative stress were measured. Hypoxia/reoxygenation (HR)-incubated HL1 cells were used to validate the effects of luteolin in vitro. In addition, a p53 agonist and an inhibitor were used to investigate the mechanism. Results Luteolin reduced the myocardial infarcted size and myocardial enzymes, and restored cardiac function in MIRI mice. Network pharmacology identified p53 as a hub target. The bioinformatic analyses showed that luteolin had anti-apoptotic and anti-oxidative properties. Additionally, luteolin halted the activation of p53, and prevented both apoptosis and oxidative stress in myocardial tissue in vivo. Furthermore, luteolin inhibited cell apoptosis, JC-1 monomer formation, and reactive oxygen species elevation in HR-incubated HL1 cells in vitro. Finally, the p53 agonist NSC319726 downregulated the protective attributes of luteolin in the MIRI mouse model, and both luteolin and the p53 inhibitor pifithrin‐α demonstrated a similar therapeutic effect in the MIRI mice. Conclusion Luteolin effectively treats MIRI and may ameliorate myocardial damage by regulating apoptosis and oxidative stress through its targeting of the p53 signaling pathway.

Overoxidized poly(3,4-ethylenedioxythiophene)-overoxidized polypyrrole composite films with enhanced electrocatalytic ability for rutin and luteolin determination

Meng Rongqian,Tang Jianke,Yang Hong,Guo Lijun,Song Yongbo,Li Qiaoling,Niu Yulan

In this study, a simple and effective method was proposed to improve the electrocatalytic ability of overoxidized poly(3,4-ethylenedioxythiophene)-overoxidized polypyrrole composite films modified on glassy carbon electrode for rutin and luteolin determination. The composite electrode was prepared by cyclic voltammetry copolymerization with LiClO 4 -water as the supporting electrolyte. The peak current of rutin and luteolin on the composite electrode gradually decreased or even disappeared with the increase in the positive potential limit. After incubation in NaOH-ethanol solution with a volume ratio of 1:1, the composite electrodes prepared at positive potential limit greater than 1.5 V exhibited enhanced differential pulse voltammetry peak currents, reduced charge transfer resistance, larger effective specific surface area and higher electron transfer rate constant. The composite electrode prepared in the potential range of 0–1.7 V showed optimal electrocatalytic performance. The X-ray photoelectron spectroscopy results indicated that the content of −SO 2 /−SO and −C=N− groups in the composite film increased significantly after incubation. Further, the Raman spectra and Fourier transform infrared spectra revealed that the thiophene ring structure changed from benzene-type to quinone-type, and the quinone-type pyrrole ring was formed. The electrocatalytic mechanism of the composite film was proposed based on the experimental results and further verified by Density Functional Theory calculation.

Luteolin Inhibits the Biofilm Formation and Cytotoxicity of Methicillin-Resistant Staphylococcus aureus via Decreasing Bacterial Toxin Synthesis

Yixuan Sun,Fengjun Sun,Wei Feng,Qian Wang,Fang Liu,Peiyuan Xia,Xuewen Qiu

Owing to the fact that luteolin has antibacterial activity against Staphylococcus aureus (S. aureus) and methicillin-resistant S. aureus (MRSA), its specific mechanism in MRSA is worthy of investigation, which is the focus of this study. Initially, the collected S. aureus strains were treated with luteolin. Then, the minimum inhibitory concentration (MIC) of luteolin against the S. aureus strains was measured by the broth microdilution. The growth curves, biofilm formation, and cytotoxicity of treated S. aureus were detected using a microplate reader. The live and dead bacteria were evaluated using confocal laser scanning microscopy, the bacterial morphology was observed using scanning electron microscopy, and the S. aureus colony-forming unit (CFU) numbers were assessed. The levels of alpha hemolysin (α-hemolysin), delta hemolysin (δ-hemolysin), and hlaA were detected via western blot and RT-PCR. The mortality of mouse model with S. aureus systemic infection was analyzed, and the levels of IL-6, IL-8, IL-10, and TNF-α were quantitated using ELISA. Concretely, the MIC of luteolin against MRSA N315 was 64 μg/mL. Luteolin at 16 μg/mL did not affect the growth of MRSA N315, but inhibited the biofilm formation and CFU, and promoted the morphological changes and death of MRSA N315. Luteolin decreased the cytotoxicity and the levels of α-hemolysin, δ-hemolysin, and hlaA in MRSA N315, elevated MRSA-reduced mice survival rate, and differentially modulated the inflammatory cytokine levels in MRSA-infected mice. Collectively, luteolin inhibits biofilm formation and cytotoxicity of MRSA via blocking the bacterial toxin synthesis.

Luteolin attenuates imiquimod–induced psoriasis-like skin lesions in BALB/c mice via suppression of inflammation response

Wei Zhou,Mengmeng Hu,Xiaohao Zang,Qifa Liu,Jiantang Du,Jingrong Hu,Lanyue Zhang,Zhiyun Du,Zhangmin Xiang

Psoriasis is considered as a common chronic immune-mediated skin disorder characterized by abnormal keratinocyte proliferation. Luteolin, an anti-inflammatory natural flavonoid with well-accepted inhibition effect against keratinocyte proliferation, was hypothesized to have a potential therapeutic effect for psoriasis. In this paper, we investigated the relieving effect of luteolin against imiquimod-induced psoriasis-like lesions on BALB/c mice and its possible anti-inflammatory mechanisms in lipopolysaccharide-stimulated macrophages (RAW264.7 cells). We found that luteolin ameliorated psoriasis-like skin lesions, suppressed the cutaneous infiltration of macrophages, T cells and neutrophils, and downregulated the expression of cytokines like IL-6, IL-1β, TNF-α, IL-17A and IL-23 in both skin lesions and eyeball blood of model mice. In vitro, we observed luteolin significantly suppressed the levels of psoriasis-related pro-inflammatory cytokines, such as IL-17A, IL-6, TNF-α and IL-23, and inflammatory mediators like nitric oxide NO, inducible NOS, COX-2 in RAW264.7 cells. The anti-inflammatory activity was accomplished by inhibiting NF-κB expression and activation. This study demonstrates luteolin is effective in alleviating psoriasis-like skin lesions and downregulating inflammatory response via NF-κB pathway, suggesting luteolin as a potential molecule for further therapeutic research of inflammation-related skin diseases like psoriasis.

Boronate-modified polyethyleneimine dendrimer as a solid-phase extraction adsorbent for the analysis of luteolin via HPLC

Baoyue Zhang,Yukui Tong,Jianghua He,Baodong Sun,Feng Zhang,Miaomiao Tian

Luteolin (LTL) is a flavonoid containing a cis-diol, which has significant anti-inflammatory, anti-allergic, anti-diabetic, anti-cancer and neuroprotective activities. In this work, a silver modified boric acid affinity polyvinyl imine (PEI) dendritic adsorbent (PPEI-Ag@CPBA) was prepared on polystyrene (PS) for the rapid recognition and selective separation of LTL. A thin layer of polydopamine (PDA) was formed on the surface of the substrate by self-polymerization, and a PDA-coated PS material (PS@PDA) was obtained. PEI with sufficient active amino groups was grafted onto PS@PDA to obtain a PEI-modified material (PS@PDA@PEI), then AgNO3 was reduced with NaBH4, and PS@PDA@PEI was embedded on Ag. Finally, PPEI-Ag@CPBA was obtained through the condensation reaction of PEI with 4-carboxyphenyl boric acid (CPBA). The adsorption conditions were optimized, the optimal pH and the optimum amount of adsorbent were determined, and the maximum adsorption capacity was found to be 2.49 mg g−1. This method has been successfully applied to the selective identification of LTL in peanut shell samples, and provides a practical platform for the detection of LTL in complex substrates.

Novel luteolin@pro-phytomicelles: In vitro characterization and in vivo evaluation of protection against drug-induced hepatotoxicity

Kaichao Song,Liping Zhou,Cuicui Wang,Zhixin Yuan,Qilong Cao,Xianggen Wu,Mengshuang Li

A novel nanoformulation with the small molecule phytochemical dipotassium glycyrrhizinate as a nanomaterial was developed for the oral delivery of luteolin (Lut), a widely used phytochemical, but it suffered from poor water solubility and low oral bioavailability . This novel nanoformulation, named [email protected] , can be fabricated with a simple process. [email protected] can instantly dissolve into aqueous mediums and formulate through self-assembly a clear phytomicelle solution with a Lut encapsulation efficiency of 99.16 ± 0.90%, a small micelle size of 30.32 ± 0.12  nm , and a narrow polydispersity index of 0.138 ± 0.024. The optimized formulation demonstrated that Lut had solubility in up to 50 mg/ml of water as a result of its encapsulation within DG phytomicelles. [email protected] exhibited excellent characteristics, including good storage stability, a fast in vitro release profile, improvement in in vitro antioxidant activity , and high safety potential. In the oral bioavailability evaluation, a shorter Tmax, increased Cmax, and improved AUC0-t were obtained with [email protected] when compared to bare Lut. The distribution evaluation further showed that [email protected] could effectively increase the concentrations of Lut in all the tested organs and gastrointestinal segments. In the protection efficacy evaluation, 100 mg/kg [email protected] demonstrated strong effects against acetaminophen-induced hepatotoxicity. The mechanisms of inhibiting high-mobility group box 1 signaling and suppressing oxidative stress were involved in this strong treatment effect. These results showed that simple but novel Lut [email protected] provided a new, promising nano-delivery system for Lut with a significantly improved in vivo profile.

Electrochemical determination of luteolin using molecularly imprinted poly-carbazole on MoS2/graphene-carbon nanotubes nanocomposite modified electrode

Bingjie Xu,Bihong Zhang,Lite Yang,Faqiong Zhao,Baizhao Zeng

MoS 2 /graphene-carbon nanotubes (MoS 2 /GN-CNTs) nanocomposite was prepared by hydrothermal method . The nanocomposite combined the high catalysis of MoS 2 with the superior electronic conductivity of GN-CNTs, and exhibited high electrochemical sensitivity towards luteolin . A molecularly imprinted polymer (MIP) film deposited by electropolymerization , based on carbazole monomer and luteolin template, was used to improve the selectivity. Thus the resulting electrochemical sensor MIP/MoS 2 /GN-CNTs/GCE showed high performance. Its linear detection range and limit of detection were 0.040–2.0 μM and 9.0 nM (S/N = 3), respectively, while its sensitivity was 381.3 μA μM −1  cm −2 . In addition, 1000-fold K + , Na + , Ca 2+ , Mg 2+ , NO 3 − , Cl − , SO 4 2− and CO 3 2− 500-fold glucose and mannitol , 250-fold cystein, glycine, lysine, citric acid and oxalic acid , and 100-fold urea, ascorbic acid and dopamine did not interfere with the determination of luteolin. Moreover, 5-fold analogs rutin , catechin and naringenin , and 2-fold kaempferide, quercetin and morin also did not influence on its determination. The sensor was applied to the determination of luteolin in carrot and chrysanthemum tea samples and the results were in good agreement with those obtained through HPLC .

Alkaline phosphatase mediated synthesis of carbon nanotube–hydroxyapatite nanocomposite and its application for electrochemical determination of luteolin

Feng Gao,Xiaoqian Chen,Hidekazu Tanaka,Ayaka Nishitani,Qingxiang Wang

A nanocomposite of hydroxyapatite (HAP)–carbon nanotube (CNT) was synthesized by in-situ transformation of CNT-calcium phenyl phosphate (CaPP) with the assist of alkaline phosphatase. The nanocomposite was characterized by powder X-ray diffraction (XRD), Fourier transform infrared (FTIR) and transmission electron microscope (TEM). Then the HAP–CNT was cast onto a glassy carbon electrode (GCE) and used as a sensing platform for the electrochemical determination of luteolin. The electrochemical experiments indicated that the HAP–CNT modified GCE (HAP–CNT/GCE) could greatly enhance the electrochemical response of luteolin. A series of experimental parameters including the type and pH value of supporting electrolyte, accumulation time and accumulation potential were optimized. The electrochemical parameters such as electron transfer rate constant and electron transfer coefficient of luteolin at the HAP–CNT/GCE were also investigated. Under the optimal conditions, the reduction peak currents presented a good linear relationship with the concentration of luteolin in the range of 4.0   ×   10 −7 –1.2   ×   10 −5   M. The detection limit was estimated to be 8.0   ×   10 −8   M.

Luteolin in Lonicera japonica inhibits the proliferation of white spot syndrome virus in the crayfish Procambarus clarkii

Hai-Feng Jiang,Cheng Chen,Xin-Yuan Jiang,Jing-Lei Shen,Fei Ling,Peng-Fei Li,Gao-Xue Wang

White spot disease caused by the white spot syndrome virus (WSSV) is a highly lethal infectious disease of crustacean aquaculture, with no effective means of control. Active compounds from herbal medicines are promising candidates for the development of antiviral agents for WSSV. We evaluated the efficacy of 12 herbal medicines in inhibiting WSSV proliferation in a WSSV infection model ( Procambarus clarkii ), Lonicera japonica extract had the strongest inhibitory effect on WSSV replication. Luteolin (LUT), the major bioactive compound of L. japonica extract, was further investigated and revealed to have a strong inhibitory effect on WSSV (96.04%, 50 mg/kg). LUT treatment decreased the WSSV load and the expression of important viral genes (immediate-early gene ie1 , DNA polymerase DNApol and envelope protein Vp28 ) in a dose-dependent manner and increased the survival of WSSV-infected crayfish by 60.00% (50 mg/kg). Co-incubation and time-of-addition assays revealed that LUT did not affect the infectivity of viral particles or viral absorption and entry. Expression of immune-related genes such as barrier-to-autointegration ( BAF ), heat shock protein 70 ( Hsp70 ) and signal transducer and activator of transcription ( STAT ), which may be beneficial for viral replication, were significantly decreased ( P  < 0.01) in LUT-treated crayfish. LUT also modified the expression of antioxidants ( SOD , CAT and GST ), pro-inflammatory factor ( COX-1 and COX-2 ), and apoptosis-related ( BAX and BI-1 ) genes ( P  < 0.01) to enhance antioxidant defenses, mitigate inflammation and induce apoptosis, respectively. Overall, LUT could be used as a prophylactic or therapeutic agent against WSSV infection in crustacean aquaculture.

Fabrication of rGO/MXene-Pd/rGO hierarchical framework as high-performance electrochemical sensing platform for luteolin detection

Huang Hui,Xie Shuqian,Deng Lu,Yuan Jie,Yue Ruirui,Xu Jingkun

A  nanocomposite of rGO/MXene-Pd/rGO with hierarchical structure based on Ti 3 C 2 T x MXene, Pd nanoparticles, and reduced graphene oxide (rGO) was synthesized by a green approach. Ti 3 C 2 T x MXene decorated with Pd nanoparticles (MXene-Pd) was prepared first. Then, graphene oxide (GO), MXene-Pd, and GO were coated on the surface of the glassy carbon electrode (GCE) in sequence. After each coating of the GO layer, the GO nanosheets were reduced to rGO electrochemically. The fabricated rGO/MXene-Pd/rGO hierarchical framework performs a pie structure with MXene-Pd as the stuffing and rGO nanosheets as the crust, which will be beneficial to the enhancement of its electrochemical sensing performance. As compared with other electrodes, the rGO/MXene-Pd/rGO/GCE exhibited higher electrocatalytic activity and better sensing performance for luteolin detection, with a wide linear range of 6.0 × 10 −10 to 8 × 10 −7  M and 1.0 × 10 −6 to 1.0 × 10 −5  M (oxidation peak potential E pa  = 0.34 V vs . SCE), a low detection limit of 2.0 × 10 −10  M, and a high sensitivity of 112.72 µA µM −1  cm −2 . Moreover, the fabricated sensor also showed high selectivity, reproducibility, and repeatability toward the detection of luteolin. The real sample analysis for luteolin in honeysuckle was successfully carried out by rGO/MXene-Pd/rGO and verified with high-performance liquid chromatography (HPLC) analysis techniques with acceptable results. All the above tests indicate the promising application prospect of the rGO/MXene-Pd/rGO framework for luteolin detection in honeysuckle and other herbs containing luteolin. Graphical abstract

Synergistic chemo-/photothermal therapy based on supercritical technology-assisted chitosan–indocyanine green/luteolin nanocomposites for wound healing

Xu Pei-Yao,Kankala Ranjith Kumar,Li Yue-Wei,Wang Shi-Bin,Chen Ai-Zheng

Despite the success, it is highly challenging to battle against pathogenic biofilms-based chronic bacterial infections by conventional antibiotic therapy. Herein, we report a near-infrared (NIR)/acid-induced nanoplatform based on chitosan (CS)-coated indocyanine green (ICG, photosensitizer)/luteolin (LUT, a natural quorum sensing inhibitor) nanocomposites (ICG/LUT-CS) as antibacterial and antibiofilm agents for skin wound healing. Initially, the ICG/LUT nanoplatforms are prepared by the supercritical antisolvent technology and coated with the CS layer. The obtained ICG/LUT-CS with ultra-high encapsulation efficiency exhibited more favorable photothermal conversion effects and improved NIR laser/acid dual-induced drug release behavior than individual modalities, achieving exceptional bacteria-killing and biofilm elimination effects. Moreover, the ICG/LUT-CS realized the synergetic effects of chemotherapy and photothermal therapy outcomes for wound healing. Together, our findings provided an appealing strategy for the rapid preparation and future translational application of ICG/LUT-CS as an ideal agent for fighting against biofilm infections.

Luteolin-loaded biocomposite containing tantalum and polyimide with antibacterial effect for facilitating osteogenic differentiation and bone-bonding

Zhaoying Wu,Rames Kaewmanee,Qianwen Yang,Zimin Wang,EN Xie,Jie Wei,Chao Zhang

Polymer-based composites possess some outstanding advantages over ceramic/metallic/polymeric biomaterials, which is considered as a promising candidate for bone repair. Tantalum (Ta)/polyimide (PI) biocomposites (PT) containing Ta nanoparticles of 20 v% (PT20) and 40 v% (PT40) were fabricated, and luteolin (LU) was loaded on PT40 (LUPT40). Compared with PT20 and PI, PT40 with high Ta content displayed high surface behaviors (e.g., roughness, surface energy, and hydrophilicity). PT40 remarkably improved the cell adhesion and multiplication, and LUPT40 with LU displayed further enhancement in vitro. Moreover, LUPT40 obviously boosted osteoblastic differentiation while suppressed the osteoclastic differentiation. Furthermore, LUPT40 exhibited good antibacterial effects because of the slow release of LU. The in vivo results confirmed that PT40 markedly promoted bone-formation, and LUPT40 further enhanced bone-formation/bone-bonding. In short, incorporation of Ta particles improved surface behaviors of PT40, which stimulated cell-response/bone-formation. Moreover, the slow release of LU from LUPT40 not only promoted cell-response/bone-formation but also enhanced bone-bonding. The synergistic effects of Ta and LU release from LUPT40 enhanced bone-formation/bone-bonding. Therefore, LUPT40 would have great potential for repair of bear-loading bone.
