Natural Polyphenol Pentagalloyl Glucose as a Potent Xanthine Oxidase Inhibitor for Hyperuricemia Treatment

Jiana Du,Yan Liu,Lizi Li,Qin Yin,Dehong Yu,Pei He,Na Wang,Ruiying Yuan,Zhujun Yin,Yanbei Tu,Yanfang Li

Hyperuricemia (HUA) is a metabolic disorder characterized by increased serum uric acid (UA) levels. Here, we showed that pentagalloyl glucose (PGG), a well-known natural polyphenol, significantly decreased UA levels both in potassium oxonate/hypoxanthine-induced HUA mice and adenosine-induced hepatocytes. Mechanistically, the UA-lowering effect of PGG was attributed to the suppression of UA formation by inhibiting xanthine oxidase (XOD) activity and expression. PGG acted as a reversible mixed-type inhibitor of XOD (IC50 = 3.92 ± 0.06 μM), and its binding was driven primarily by hydrogen bonding and van der Waals forces. Structural analyses revealed PGG occupied the catalytic center of XOD, obstructing xanthine binding and inducing conformational changes. Integrated kinetics, isothermal titration calorimetry, multispectroscopic investigations, and computer simulations confirmed these interactions. These findings first comprehensively suggest the anti-HUA effect of natural XOD inhibitor PGG and highlight the promising potential of PGG as a functional food or drug candidate for treating HUA-related diseases.

Electrospun gelatin/tea polyphenol@pullulan nanofibers for fast-dissolving antibacterial and antioxidant applications

Jianfeng Zhou,Weiqiang Wang,Xingjian Yang,Deng-Guang Yu,Ping Liu

Bio-based active food packaging materials have a high market demand. We use coaxial electrospinning technology to prepare core–shell structured nanofibers with sustained antibacterial and antioxidant properties. The fiber core layer was composed of gelatin and tea polyphenols, whereas tea polyphenols provide antibacterial and antioxidant properties; the fiber sheath was composed of pullulan polysaccharides with antioxidant properties. By using a scanning electron microscope, it can be seen that the diameter distribution of the prepared nanofibers was uniform and the surface is smooth; using a transmission electron microscope, it can be clearly seen that the nanofibers have a core–shell structure; Fourier Transform Infrared and X-ray diffraction analysis indicate that the nanofibers have an amorphous structure; the 2,2-diphenyl-1-picrylhydrazyl free radical scavenging shows that nanofibers have higher antioxidant properties with the addition of tea polyphenols; antibacterial test showed that nanofibers had obvious inhibitory effect on the growth of Staphylococcus aureus and Escherichia coli ; and the nanofiber film dissolution test shows that nanofibers can be used as fast soluble active packaging. Finally, core–sheath-structured nanofibers can serve as active packaging for instant food, possessing both rapid water solubility and excellent antibacterial and antioxidant activity, making water-soluble nanofibers interesting applications in the field of food packaging.

Bioinspired Design of an Underwater Adhesive Based on Tea Polyphenol-Modified Silk Fibroin

Jialuo Chen,Zhipeng Li,Xinpeng Chen,Yurong Sun,Jin Cheng,Aijing Li,Shenzhou Lu,Tieling Xing

Adhesives have garnered significant interest recently due to their application in the field of biomedical applications. Nonetheless, developing adhesives that exhibit robust underwater adhesion and possess antimicrobial properties continues to pose a significant challenge. In this study, motivated by the adhesive mechanism observed in mussels in aquatic environments, dopamine (DA) was added to modify the silk fibroin (SF) solution. Subsequently, tea polyphenol (TP) was incorporated to form a sticky mixture, resulting in a biomimetic adhesive (TP-DA/SF). TP-DA/SF demonstrated rapid, robust, and indiscriminate adhesion to a wide array of substrates and even biological tissues (39 kPa). TP-DA/SF exhibits the ability to replicate the mussel adhesion mechanism of mussels underwater thanks to its biomimetic design. This characteristic provides the material with robust adhesion (40 kPa), notable reusability (at least 10 times), and long-lasting stability, especially in aquatic settings. It is worth noting that TP-DA/SF also demonstrated high adhesion in various water environments, even in solutions with a pH of 7.4 and buffered saline (PBS), which is one of the most widely used buffers in biochemistry research, offering salt-balancing and adjustable pH buffering capabilities. Meanwhile, TP-DA/SF exhibits excellent antibacterial and antioxidant properties due to its tea polyphenol content. After 15 days of wound closure in SD rats, the healing rate in the experimental group reached 93.4%, compared to 83.9% in the control group. Thus, the TP-DA/SF adhesive holds promising potential for biomedical applications, including sutureless wound closure and tissue adhesion.

A tea polyphenol-loaded cellulose/silk fibroin/polyacrylic acid hydrogel for wound healing

Liu Huijun,Chen Li,Peng Yan,Li Xia,Zhang Haiqiang,Chen Yanhao,Li Zhi,Dai Fangyin

Hydrogels can relieve wound pain and provide a moist environment for wound healing. Although hydrogels based on natural materials have proven to be biocompatible and biodegradable, their antibacterial and anti-inflammatory properties are not desirable. Natural extracts with antimicrobial and antioxidant properties are becoming increasingly attractive because of their environmentally friendly nature and good biocompatibility. Herein, composite cellulose/silk fibroin/polyacrylic acid hydrogels based on natural materials were fabricated by crosslinking between oxidized cellulose and silk fibroin together with radical polymerization of acrylic acid. The fabricated hydrogels displayed a uniform hierarchical topography with high porosity, as well as good mechanical properties, swelling properties, air permeability and biocompatibility. With the addition of tea polyphenols to natural extracts, the composite hydrogels exhibited significant antimicrobial and antioxidant properties such that they could prevent bacterial infection and relieve wound inflammation in the inflammation stage and could promote the synthesis of extracellular matrix (ECM), angiogenesis, collagenization and re-epithelization during wound healing. The tea polyphenols/cellulose/silk fibroin/polyacrylic acid hydrogels fabricated in this study can accelerate wound healing, demonstrating their great potential for application in wound dressings.

Tea polyphenol-engineered hybrid cellular nanovesicles for cancer immunotherapy and androgen deprivation therapy

Guo Yiming,Wu Jicheng,Chen Lefan,Liu Lujie,Bi Tianxiang,Pan Yuanwei,Meng Qian-Fang,Wang Chaoliang,Rao Lang,Li Qi

Androgen deprivation therapy (ADT) is a crucial and effective strategy for prostate cancer, while systemic administration may cause profound side effects on normal tissues. More importantly, the ADT can easily lead to resistance by involving the activation of NF-κB signaling pathway and high infiltration of M2 macrophages in tumor microenvironment (TME). Herein, we developed a biomimetic nanotherapeutic platform by deriving cell membrane nanovesicles from cancer cells and probiotics to yield the hybrid cellular nanovesicles (hNVs), loading flutamide (Flu) into the resulting hNVs, and finally modifying the hNVs@Flu with Epigallocatechin-3-gallate (EGCG). In this nanotherapeutic platform, the hNVs significantly improved the accumulation of hNVs@Flu-EGCG in tumor sites and reprogramed immunosuppressive M2 macrophages into antitumorigenic M1 macrophages, the Flu acted on androgen receptors and inhibited tumor proliferation, and the EGCG promoted apoptosis of prostate cancer cells by inhibiting the NF-κB pathway, thus synergistically stimulating the antitumor immunity and reducing the side effects and resistance of ADT. In a prostate cancer mouse model, the hNVs@Flu-EGCG significantly extended the lifespan of mice with tumors and led to an 81.78% reduction in tumor growth compared with the untreated group. Overall, the hNVs@Flu-EGCG are safe, modifiable, and effective, thus offering a promising platform for effective therapeutics of prostate cancer. Graphical Abstract

Tea polyphenol nano-crosslinked dynamical hyaluronic acid-based hydrogel for diabetic wound healing

Huan Liu,Ronger Ai,Bi-zhi Liu,Li He

Diabetic wound healing remains a significant clinical challenge for the complex wound microenvironment characterized by oxidative stress, inflammation, and bacterial infection. To address these challenges, we present a novel hydrogel incorporates tea polyphenol-stabilized silver nanoparticles (TP@Ag NPs) into a dynamic hyaluronic acid-phenylboronic acid network crosslinked via borate ester bonds. This design leverages the inherent biocompatibility and biodegradability of hyaluronic acid alongside the antioxidant, anti-inflammatory, and antibacterial properties of tea polyphenols and silver nanoparticles. The HP-TP@Ag hydrogel exhibited glucose-responsive degradation and TP@Ag NPs release, enabling targeted delivery within the diabetic wound microenvironment. In vitro assays demonstrated the hydrogel's potent antioxidant activity, effectively scavenging ROS and protecting both HaCaT and RAW264.7 cells from oxidative stress. Furthermore, the HP-TP@Ag hydrogel significantly suppressed the production of pro-inflammatory cytokines and exhibited robust antibacterial activity against both E. coli and S. aureus . In vivo studies using a diabetic mouse model revealed accelerated wound closure, reduced inflammation, enhanced collagen deposition, and promoted angiogenesis and tissue remodeling in HP-TP@Ag hydrogel-treated wounds. These findings highlight the promise of HP-TP@Ag hydrogel as an advanced wound dressing for effective diabetic wound management, offering a synergistic approach to overcome the multifaceted challenges associated with this complex condition.

Synergistic polyphenol–amino acid nanoparticles: a new strategy for reactive oxygen species management

Huizhou Ye,Jiayin Cai,Zhihao Shen,Qiuping Qian,Chunxia Zhang

Polyphenols exhibit strong antioxidant, and anti-inflammatory but are limited by chemical instability and low bioavailability. To address these challenges, we developed polyphenol–amino acid conjugates that self-assemble into stable nanospheres, enhancing their stability and bioavailability. These nanoparticles demonstrate significantly improved ROS scavenging efficiency and promote cell proliferation in vitro. The incorporation of amino acids enhances biocompatibility and facilitates effective ROS elimination. The polyphenol–amino acid nanoparticles offer a multifaceted therapeutic strategy to mitigate oxidative stress, overcoming traditional antioxidant limitations through advanced nanotechnology. This approach contributes to the development of next-generation wound care solutions with enhanced efficacy and safety profiles.

Protein-polyphenol conjugation through alkali treatment improves the structure and functionality of gluten

Liu Jianying,Ma Yan,Ukwatta Ruchika Hansanie,Xue Feng,Xiong Xiaohui,Li Chen

The conjugates of gluten and polyphenols (naringin, hesperetin or curcumin) were produced by alkaline method and the effects of conjugation on physicochemical properties of gluten were investigated in this study. The results show that the conjugates possess lower content of free amino and sulfhydryl groups, and higher molecular mass, which indicate the covalent binding occurred. As compared with corresponding mixtures, the conjugates show lower intensity of the amide A, surface hydrophobicity, intermolecular β-sheets, interfacial and surface tension, as well as higher fluorescence intensity, antiparallel β-sheets and particle size (in the range of 130–150 μm). Furthermore, the conjugates also exhibit better emulsifying and foaming properties, higher storage modulus and lower degradation temperature than native gluten. However, the changes in physicochemical properties of conjugates depend on the type of polyphenols and curcumin-gluten conjugates possess the improved emulsifying, foaming and gelation properties among conjugates, due to their more flexible structure.

Multiscale structure changes and mechanism of polyphenol-amylose complexes modulated by polyphenolic structures

Xianling Wei,Huan Xie,Ziqing Hu,Xiaofang Zeng,Hao Dong,Xiaoyan Liu,Weidong Bai

This study was designed to investigate the effect of polyphenolic structure on the interaction strength and process between polyphenols (gallic acid (GA), epigallocatechin gallate (EGCG) and tannic acid (TA)) and amylose (AM). The results of Fourier transform infrared spectroscopy, isothermal titration calorimetry, X-ray photoelectron spectroscopy and molecular dynamic simulation (MD) suggested that the interactions between the three polyphenols and AM were noncovalent, spontaneous, low-energy and driven by enthalpy, which would be enhanced with increasing amounts of pyrogallol groups in the polyphenols. The results of turbidity, particle size and appearance of the complex solution showed that the interaction process between polyphenols and AM could be divided into three steps and would be advanced by increasing the number of pyrogallol groups in the polyphenols. At the same time, MD was intuitively employed to exhibit the interaction process between amylose and polyphenols, and it revealed that the interaction induced the aggregation of amylose and that the agglomeration degree of amylose increased with increasing number of pyrogallol groups at polyphenols. Last, the SEM and TGA results showed that TA/AM complexes had the tightest structure and the highest thermal stability (TA/AM˃EGCG/AM˃GA/AM), which could be attributed to TA having five pyrogallol groups.

Impact of polyphenol-loaded edible starch nanomaterials on antioxidant capacity and gut microbiota

Wenzhi Lei,Maodong Qi,Pei Tan,Siqi Yang,Lingjia Fan,Hongcai Li,Zhenpeng Gao

Starch nanoparticles (SNPs) have the capability to adsorb polyphenol components from apple pomace efficiently, forming bound polyphenols (P-SNPs). These bound polyphenols may have potential bioactivities to affect human health positively. Therefore, in-depth in vivo observation of the antioxidant activity and evaluation of its gut microbiota regulatory function are essential. The results revealed that P-SNPs indicated significant scavenging abilities against DPPH, ABTS, and hydroxyl radicals. Furthermore, the nanomaterials exhibited non-toxic properties, devoid of hepatorenal and intestinal damage, while concurrently stimulating the production of short-chain fatty acids (SCFAs) within the gastrointestinal tract. Notably, P-SNPs significantly enhanced antioxidant capacity in serum, liver, and kidney tissues, fostering the proliferation of beneficial bacteria ( Lactobacillus , Bacillus , norank_f__Muribaculaceae ) while suppressing pathogenic bacterial growth ( Helicobacter , Odoribacter ). This study proposes a novel research concept for the scientific use of polyphenols in promoting gut health.

Natural Polyphenol Delivered Methylprednisolone Achieve Targeted Enrichment for Acute Spinal Cord Injury Therapy

Taoyang Yuan,Weijie Liu,Tianyou Wang,Feng Ye,Jianhua Zhang,Zhipeng Gu,Jianguo Xu,Yiwen Li

The strong anti-inflammatory effect of methylprednisolone (MP) is a necessary treatment for various severe cases including acute spinal cord injury (SCI). However, concerns have been raised regarding adverse effects from MP, which also severely limits its clinical application. Natural polyphenols, due to their rich phenolic hydroxyl chemical properties, can form dynamic structures without additional modification, achieving targeted enrichment and drug release at the disease lesion, making them a highly promising carrier. Considering the clinical application challenges of MP, a natural polyphenolic platform is employed for targeted and efficient delivery of MP, reducing its systemic side effects. Both in vitro and SCI models demonstrated polyphenols have multiple advantages as carriers for delivering MP: (1) Achieved maximum enrichment at the injured site in 2 h post-administration, which met the desires of early treatment for diseases; (2) Traceless release of MP; (3) Reducing its side effects; (4) Endowed treatment system with new antioxidative properties, which is also an aspect that needs to be addressed for diseases treatment. This study highlighted a promising prospect of the robust delivery system based on natural polyphenols can successfully overcome the barrier of MP treatment, providing the possibility for its widespread clinical application.

Polyphenol mediated zinc-oxygen synergistic hydrogel remodels senescent microenvironment for periodontal tissue regeneration

Chengxinyue Ye,Jin Liu,Jinhui Ran,Shoushan Hu,Lu Yang,Jialiang Zhao,Xinyi Fang,Zhenzhen Zhang,Xin Xiong,Xueman Zhou,Yating Yi,Xiaolong Zeng,Xiong Lu,Jun Wang,Chaoming Xie

Senescent mesenchymal stem cells residing in an inflammatory, dysbiotic, and hypoxic microenvironment pose a barrier to periodontal regeneration. Here we introduce a hydrocaffeic acid (HCA)-mediated silk fibroin hydrogel incorporating Mn/HCA-modified calcium peroxide (Mn-hCaO₂) and HCA-modified zeolitic imidazolate framework-8 (hZIF8) to rejuvenate this environment. The strategy imbues the hydrogel with enhanced adhesion and adaptability in periodontal pockets. The Mn-HCA complex acts catalytically to reduce oxidative stress and correct hypoxia. Simultaneously, Zn²⁺ and HCA restore microbial balance and mitigate inflammation. This multifunctional hydrogel targets the senescent periodontal niche by normalizing microbiota homeostasis and promoting a regenerative immune profile. Concurrently, it directly alleviates telomere shortening, DNA damage and oxidative stress in stem cells, thereby rejuvenating cellular function. By specifically addressing hypoxia and zinc deficiency, this polyphenol-mediated synergistic strategy offers a pathway to rejuvenate the senescent periodontal microenvironment, thereby overcoming tissue regeneration barriers and offering a translatable pathway for treating periodontitis and other aging-associated inflammatory diseases. Periodontitis treatment is challenged by senescent stem cells in the periodontal niche. Here, authors develop a multifunctional hydrogel that alleviates hypoxia and stem cell aging, and restores microbial balance to accelerate regeneration.
