Sequential electrospinning of multilayer ethylcellulose/gelatin/ethylcellulose nanofibrous film for sustained release of curcumin

Peng Wang,Yang Li,Cen Zhang,Fengqin Feng,Hui Zhang

In this work, sequential electrospinning was utilized to fabricate a multilayer film with ethylcellulose nanofibers as the outer layer and curcumin-loaded gelatin nanofibers as the inner layer. Field-emission scanning electronic microscopy observations showed that the outer and inner layers had a smooth surface and clear boundary. The hydrophobic outer layers decreased the water vapor permeability and improved the water contact resistance of the hydrophilic inner layer, and the intimate interactions of hydrogen bonds between two adjacent layers enhanced the thermal stability . The multilayer film exhibited a sustained release manner of the encapsulated curcumin for 96 h, compared to the burst release within 30 min from the gelatin film. In addition, the antioxidant activities of the released curcumin from the multilayer film were well retained within 96 h. These results suggested that the multilayer nanofibrous film fabricated by sequential electrospinning has potential applications in bioactive encapsulation and controlled release.

Sustained release of cathepsin D/cathepsin K from injectable hydrogel microspheres in anti-photoaging

Wenyi Hou,Zhaoyi Lin,Xinling Chen,Yu Sun,Yao Lin,Mengran Peng,Feng Zhou,Huamin Jiang,Yan Li,Yue Zheng

There have been various interventions for photoaging skin, such as retinoic acid treatment and laser therapy. However, more precise, secure, and effective technologies and materials were still need to be explored. Cathepsin D (CTSD) repairs the epidermal barrier in chronic photodamaged skin through increasing TGase-1 expression and activity. Cathepsin K (CTSK) affects the metabolism of skin elastic fibers. Therefore, gelatin/alginate composite hydrogel microspheres loaded with CTSD or CTSK were fabricated. The microspheres with an average size of 79.8 ± 30.4 μm were stable in PBS (pH 7.4) for 5 days and then disintegrated in vitro . 0.1 mg/mL of microspheres continuously released 12-13 ng/mL CTSD or CTSK for the first three consecutive days. In vivo , the microspheres were almost completed degraded on day 7 after the subcutaneous injection. The biosafety and efficacy of CTSD- and CTSK- microspheres were confirmed in vitro and in vivo . They reduced ROS and inhibited the degradation of skin collagen and elastic fibers by upregulating Nrf2 and downregulating MMP-1 and MMP-3, which reversed photodamage of skin induced by UV. In conclusion, the gelatin/alginate composite microspheres loaded with CTSD or CTSK showed a great potential for chronic photodamaged skin.

Dual Antifreeze and Sustained Release Cellulose Nanocrystal-Humic Acid Nanocomposite Hydrogel for Soilless Cultivation

Pengxiao Liu,Kai Wei,Yong Liu,Liangjiu Bai,Hou Chen,Wenxiang Wang,Lixia Yang,Huawei Yang,Donglei Wei

Soilless cultivation relies on hydrogel matrices for water and nutrient management, but conventional hydrogels lose performance at low temperature and show unstable sustained release. Here this study develops an antifreeze sustained-release hydrogel (P-GTCH) that integrates l-proline (L-P) with a humic-acid-modified cellulose nanocrystal nanocomposite (CNCs-HA) for nanoenabled controlled release. L-P suppresses ice nucleation via hydrogen bonding, while CNCs-HA boosts HA loading and enables controlled transport within the cross-linked network. HA shows slow, continuous release governed by Fickian diffusion, achieving 96.7% cumulative release over 12 days at 0 °C and mitigating poor fertilizer release in cold environments. P-GTCH provides mechanical support for plant roots, and the synergistic effects of L-P and HA maintain lettuce germination above 86% at 0 °C while promoting root growth. This biobased, biodegradable platform is scalable for low-temperature soilless cultivation.

Wogonin-Loaded PLGA Sustained-Release Nanomicelle Inhibiting EGFR Pathway for Antitumor Effects in OSCC

Leina Gao,Dixin Cui,Xu Gao,Xiaoling Luan,Huali Gao

Oral squamous cell carcinoma (OSCC) is the most prevalent malignancy of the head and neck. Current treatment strategies, including surgery, radiotherapy, and chemotherapy, are limited by moderate efficacy and severe side effects, highlighting the need for novel therapeutic approaches. Wogonin (Wo), a natural flavonoid, has shown antiproliferative, pro-apoptotic, and antimetastatic activities in various cancer models; however, its effect on OSCC has not been investigated. In this study, Wo-loaded poly(lactic-co-glycolic acid) nanoparticles (PW NPs) were prepared to enhance water solubility and achieve sustained drug release. In vitro assays, including CCK-8, flow cytometry, live/dead staining, and wound healing assays, revealed that PW NPs significantly inhibited OSCC cell proliferation, induced apoptosis, and reduced cell migration. Network pharmacology analysis and qRT-PCR further suggested that Wo exerts its therapeutic effects through the EGFR signaling pathway. Hemolysis testing and histological examination of major organs confirmed the favorable biocompatibility of the nanoparticles. This study provides the first systematic evidence of the therapeutic potential of Wo in OSCC and lays a foundation for its further preclinical development.

Mechanical and Sustained-Release Properties of Crosslinked Poly(vinyl alcohol)/Sodium Humate Composite Membranes

Shuai Kuang,Enwei Chen,Tian-en Shui,Piyue Gong,Feng Wang,Haiying Huang

Humic acid, as a natural macromolecular aggregate rich in functional groups, offers abundant modification sites and tunable chemical functionality, making it a promising building block for three-dimensional network construction. In this study, glutaraldehyde (GA) was employed as a crosslinking agent to incorporate sodium humate (NaHA, sodium salt of humic acid from alkaline treatment) into a polyvinyl alcohol (PVA) matrix, yielding composite membranes with enhanced structural stability and performance. The results demonstrate that NaHA effectively modulates the crosslinked network, and the physical and mechanical characteristics can be readily tailored by varying the PVA/NaHA/GA ratio. Compared with pristine PVA/GA hydrogel, the inclusion of NaHA significantly influences the mechanical response, with optimal comprehensive performance achieved at a NaHA content of 7.5 wt%, corresponding to a tensile strength of 60.4 MPa and an elongation at break of 74.2%. Furthermore, the cumulative release of NaHA after two days reached 64.4%, confirming that NaHA supramolecular aggregates were stably entrapped within the PVA/GA crosslinked matrix. The release kinetics were well described by the Korsmeyer–Peppas model. Overall, the covalent crosslinking of PVA with GA, together with hydrogen-bonding associations and physical entrapment mediated by NaHA, constructed a composite membrane network. This network exhibited tunable dry-state mechanical properties and sustained NaHA release, supporting its further evaluation as a prospective candidate for agricultural mulching films.

3D printing mesoporous bioactive glass/sodium alginate/gelatin sustained release scaffolds for bone repair

Jingwen Wu,Guohou Miao,Zhichao Zheng,Zhengmao Li,Wen Ren,Caijuan Wu,Yuanjing Li,Zhu Huang,Lan Yang,Lvhua Guo

Drug delivery and release are a major challenge fabricating bone tissue engineering. In this study, we fabricated new sustained release hydrogel scaffolds composited of mesoporous bioactive glass, sodium alginate and gelatin by a three-dimensional printing technique. Naringin and calcitonin gene-related peptide were used as drugs to prepare drug-loaded scaffolds by direct printing or surface absorption. The physicochemical properties of the scaffolds and the drug release profiles of the two drug-loading models were investigated. We also examined the biocompatibility of the scaffolds, as well as the effect of the released medium on the proliferation and osteogenic differentiation of human osteoblast-like MG-63 cell. The results showed that the scaffolds had a high porosity (approximately 80%) with an interconnected cubic pore structure, rough surface morphology, bioactivity and strong biocompatibility. Furthermore, the naringin or calcitonin gene-related peptide co-printed into the scaffold displayed a steady sustained release behaviour for up to 21 days without an initial burst release, while both naringin and calcitonin gene-related peptide absorbed onto the surface of the scaffold were completely released within two days. MG-63 cells cultured with the extraction containing released drugs displayed promoted cell proliferation and the expression of osteogenesis-related genes more effectively compared with the drug-free extractions. Therefore, these results demonstrate that the developed mesoporous bioactive glass/sodium alginate/gelatin sustained release scaffolds provide a potential application for bone tissue engineering.

Preparation of a colon-specific sustained-release capsule with curcumin-loaded SMEDDS alginate beads

Yuxia Zhang,Yan Bai,Huali Chen,Yuanrui Huang,Pei Yuan,Liangke Zhang

A colon-specific capsule with alginate beads containing a self-microemulsifying drug delivery system (SMEDDS) was developed and evaluated. The SMEDDS technique was used to improve the solubility of curcumin (Cur). After encapsulating the Cur-loaded SMEDDS, the alginate beads were placed inside an impermeable capsule body. A konjac glucomannan/lactose/hydroxypropyl methylcellulose (KG/Lac/HPMC) plug tablet was then prepared and placed in the mouth of the capsule. The capsule demonstrated a pulsatile drug-release profile with a specific lag time and subsequent sustained-release phase. The lag time was modified by changing the type of HPMC and the ratio of KG/Lac/HPMC. In addition, 0.5% β-mannase solution and 5% rat cecal solution were used to simulate the colon fluid, significantly decreasing the lag time of the capsule. The results show that the capsule has potential for use in colon-specific drug delivery and exhibits a sustained-release characteristic.

Spray-dried magnetic chitosan/Fe3O4/halloysite nanotubes/ofloxacin microspheres for sustained release of ofloxacin

Qin Wang,Junping Zhang,Aiqin Wang

A series of magnetic chitosan/Fe3O4/halloysite nanotubes/ofloxacin (CTS/Fe3O4/HNTs/OFL) microspheres was prepared by spray-drying for the sustained release of OFL. CTS was in situ crosslinked by glutaraldehyde in the spray-drying process. The magnetic microspheres were characterized by FT-IR, scanning electron microscopy and thermogravimetric analysis. The magnetic properties, encapsulation efficiency, drug loading and in vitro sustained release properties of the magnetic microspheres for OFL were also evaluated. The CTS/Fe3O4/HNTs/OFL microspheres show superparamagnetic properties and fast magnetic response. The Fe3O4 nanoparticles have a remarkable effect on the surface morphology of the microspheres compared with HNTs. The encapsulation efficiency and the cumulative release are closely related to HNTs and Fe3O4 contents, crosslinking density and CTS concentration. OFL is released from the magnetic CTS/Fe3O4/HNTs/OFL microspheres via anomalous transport.

Long-Acting Sustained-Release Hydrogel for Soil-Borne Pathogen Control in Chinese Herbal Medicine

Wang Xiaoyan,Xiang Shunyu,Wang Jing,Luo Xingyi,Du Changxue,Sun Xianchao

The Chinese herbal medicine industry plays a crucial role globally, providing traditional remedies and significantly contributing to healthcare. Disease prevention and control in Chinese herbal medicine face unique challenges distinct from conventional plant protection methods. The distinctive requirements for disease control add complexity to disease management, necessitating specialized approaches. In this study, we developed a bio-based hydrogel (CSA-gel) using a straightforward procedure designed for the sustained release of azoxystrobin for soil disinfection with the aim of preventing fungal diseases in Chinese herbal medicine. The synthesized hydrogel was characterized using FTIR, zeta potential measurement, cryo-TEM, SEM, and elemental analysis. CSA-gel demonstrated a release duration of azoxystrobin for over 14 days, maintaining high antifungal activity throughout the entire release period. Additionally, CSA-gel exhibited the ability to release a significant quantity of calcium ions during pesticide release, fostering plant growth and enhancing plant disease resistance. The use of CSA-gel is anticipated to decrease both how often and how much pesticide is needed, highlighting the significance of our research in improving sustainable disease control in traditional Chinese herbal medicine farming.

Development and characterization of chitosan/bacterial cellulose/pullulan bilayer film with sustained release curcumin

Miaoyu Gan,Caoyu Guo,Wenying Liao,Xiaoli Liu,Qi Wang

A natural biopolymer bilayer film based on chitosan and bacterial cellulose with a protective layer of pullulan was developed by a two-step solution casting method. Curcumin was incorporated as an active antioxidant and antibacterial agent into the inner layer. The films with different curcumin concentrations were systematically characterized. Fourier transform infrared spectroscopy and X-ray diffraction analyses showed high compatibility between curcumin and the polysaccharide matrix through intermolecular interactions, which was verified by enhanced mechanical and barrier properties. The curcumin incorporation improved the thermal stability by >35.4 %, along with lower visible and ultraviolet light transmittance (< 8.6 %) and water solubility (< 25.1 %). The film had both antibacterial and antioxidant properties, and the sustained release of curcumin was largest (> 58.8 %) in the fatty food simulant lasting for over 155 h. The results suggested that the film containing 0.2 % curcumin had ideal physical and functional properties, suggesting its potential as a novel packaging material for the preservation of high-fat food.

Gastrointestinal Digestion and Microbial Hydrolysis of Alkyl Gallates: Potential Sustained Release of Gallic Acid

Xinmiao Wang,Qian Wang,Yuanyuan Hu,Fawen Yin,Xiaoyang Liu,Dayong Zhou

Phenolipids such as alkyl gallates (A-GAs) have been approved by the food industry as non-toxic antioxidant additives, which are also regarded as an emerging source of functional food ingredients. However, comprehensive understanding of their digestive absorption is needed. Thus, the models of live mice and anaerobic fermentation were used to clarify the distribution and microbial hydrolysis characteristics of A-GAs in the gastrointestinal tract. HPLC-UV results demonstrated that A-GAs could be hydrolyzed by intestinal lipases and gut microorganisms includingLactobacillusto produce free gallic acid (GA). Through regulating the chain length of the lipid part in A-GAs, the sustained and controllable release of the GA can be easily achieved. Furthermore, A-GAs were also able to reach the colon and the cecum, which would lead to potential gastrointestinal protective effects. Therefore, A-GAs may be applied as possible ingredient for functional foods. Keywords:alkyl gallates;distribution;hydrolysis;gastrointestinal tract;oral administration
