Passive daytime radiative cooling (PDRC), an energy-saving and environmentally friendly strategy, achieves cooling without any power consumption by reflecting sunlight and radiating energy to outer space. Herein, it is shown that a porous-structured poly(dimethylsiloxane)/silicon oxide (PDMS/SiO2) composite foam is fabricated through “pore in pore” engineering, in which a three-dimensional self-supporting silicon dioxide (SiO2) network is constructed in the pores of PDMS foam fabricated via freezing casting. The PDMS/SiO2 foams exhibit a hierarchical porous structure like “SiO2 assembling pores in PDMS micropores” and significantly benefit the reflectivity and infrared emissivity. An average solar reflectivity of ∼90.1% within a full spectral range of 0.3–2.5 μm and an average emissivity of ∼94.1% in the atmospheric window allow for the temperature inside the foam cavity to drop by ∼11.1 °C under direct sunlight with the solar radiation intensity of 871 W m–2. Notably, the hierarchical porous PDMS/SiO2 foams exhibit excellent thermal insulation, antifouling, and hydrophobic performances, weakening the heat flow from the external environment and ensuring long-term outdoor use. Such a powerful radiative cooling performance integrated with thermal insulating and antifouling capability by the foams constructed by “pore in pore” engineering offers a way to efficiently manage sunlight radiation energy to make devices, vehicles, buildings, and other urban objects cooler and helps to save energy in an outdoor sunlight environment.

Increasing antibiotic resistance becomes a serious threat to public health. Photothermal therapy (PTT) and antibacterial enzyme-based therapy are promising nonresistant strategies for efficiently killing drug-resistant bacteria. However, the poor thermostability of enzymes in PTT hinders their synergistic therapy. Herein, antibacterial glucose oxidase (GOx) is embedded in a Ag graphitic nanocapsule (Ag@G) arrayed silk film to fabricate a GOx-synergistic PTT system (named silk-GOx-Ag@G, SGA). The SGA system can stabilize GOx by a vitrification process through the restriction of hydrogen bond and rigid β -sheet, and keep the antibacterial activity in the hyperthermal PTT environment. Moreover, the arrayed Ag@G possesses excellent chemical stability due to the protection of graphitic shell, providing stable plasmonic effect for integrating PTT and surface enhanced Raman scattering (SERS) analysis even in the GOx-produced H 2 O 2 environment. With in situ SERS identification of bacterial intrinsic signals in the mouse wound model, such SGA realizes superior synergistic antibacterial effect on the infected Escherichia coli , Staphylococcus aureus , and methicillin-resistant Staphylococcus aureus in vivo, while without causing significant biotoxicity. This system provides a therapeutic method with low resistance and in situ diagnosis capability for efficiently eliminating bacteria.

Ensuring the long-term durability and safety of galvanized steel structures in industrial and environmental applications requires a deep understanding of their corrosion resistance, particularly at defects such as cut edges and scratches. These sites often initiate localized corrosion due to micro-environmental inhomogeneities. This study investigates the dynamic corrosion behavior of galvanized Zn/Q235 steel couples by integrating macro-electrochemical testing with in-situ high-resolution scanning electrochemical microscopy (SECM) to reveal the underlying interfacial physicochemical mechanisms. While macro-electrochemical measurements indicated significant passivation, with the corrosion current density decreasing by over an order of magnitude to 1.00 × 10⁻ 6 A·cm⁻ 2 after 24 h, micro-scale SECM mapping revealed a more complex mechanism. As corrosion progressed, oxygen reduction reactions became increasingly concentrated at the Zn/Q235 interface, where localized pH stabilized near 8. This remarkable pH buffering is attributed to the continuous precipitation of zinc corrosion products, an interfacial process that consumes both Zn 2+ and OH⁻ to form a passivating layer. At simulated scratch defects, an initially aggressive galvanic coupling with steep pH gradients was gradually suppressed as these precipitates formed, demonstrating a self-passivation effect. These findings elucidate a locally driven, interfacial chemical buffering mechanism, governed by precipitate formation, that controls passive film development and slows degradation. The results establish direct links between interfacial pH regulation, corrosion-product formation, and self-passivation at defects, advancing the interface science underpinning environmentally resilient galvanized coatings.

Bacillus cereus is a significant cause of foodborne illnesses, particularly through starchy foods like rice and pasta, leading to the vomiting symptom. The primary toxin involved, cereulide, is preformed within contaminated food and exhibits high resistance to heat, making it particularly difficult to control. Since cereulide persists even after standard food processing methods, its prevention and control remain critical for the food industry. Isoleucine (Ile), a branched-chain amino acid (BCAA), has been linked to the regulation of cereulide production, but the exact mechanisms remain unclear. Based on untargeted metabolomics analysis using liquid chromatography-mass spectrometry (LC-MS), this study demonstrated that Ile supplementation markedly downregulated the folate biosynthesis pathway in B. cereus . Supplementation with folic acid partially restored cereulide production that had been suppressed by Ile. In addition, RT-qPCR analysis revealed significant changes in the expression of key enzymes involved in folate metabolism and related pathways following Ile exposure, further supporting the notion that Ile inhibits cereulide synthesis by disruption of folate metabolism. Collectively, these findings suggest a novel mechanism by which Ile regulates cereulide production via folate metabolism, providing new insights for the effective prevention and control of cereulide production in B. cereus .

Sustained release microspheres loaded with goserelin are regarded as a promising candidate for treating prostate cancer and other sex hormone diseases. However, their widespread adoption has been hindered by issues such as wide particle size distribution and unstable release characteristics. To address these challenges, we employed a combination of the solid-in-oil-in-water microspheres preparation approach (S/O/W) and innovative premix membrane emulsification technology and deeply investigated the effects of four key parameters on the loaded performance of microspheres and the microscopic mechanisms behind them. With this approach, we successfully produced goserelin-loaded sustained release microspheres of narrow particle size distribution (Span 0.642), remarkable encapsulation efficiency (DL = 4.23 %, EE = 93.98 %), low initial burst release (about 0.50 % within 2 h), and compatibility with small injection needles (23-G, inner diameter 0.33 mm, outer diameter 0.64 mm, maximal force 59 N). In the animal model(administered dose, 2.4 mg·Kg −1 ), goserelin long-acting sustained release microspheres sustained release for over 32 days, maintaining effective concentrations above 2 ng·mL −1 , and effectively reduced serum testosterone concentrations to castration levels (<1.0 ng·mL −1 ) by day 4, maintaining this inhibition for up to 21 days, exhibiting comparable efficacy to the positive control group. In vivo release kinetics analysis revealed that goserelin-loaded sustained release microspheres exhibited a release pattern dominated by diffusion with corrosion assistance in vivo. In summary, the systematic and comprehensive evaluation of uniform-sized goserelin-loaded sustained release microspheres has highlighted their excellent translational potential, and the study herein may provide new strategies and ideas for the development of microsphere dosage forms.

Urine, an easily accessible body fluid, contains rich metabolic compounds reflecting the human health condition. Though urine analysis can be carried out by many conventional analytical methods, direct, rapid, and simultaneous identification of a variety of urinary metabolites by a handheld device remains challenging. Recent developments in nanopore small molecule sensing may suggest direct urine analysis performed by an engineered nanopore. Here, a hetero-octameric Mycobacterium smegmatis porin A (MspA) nanopore immobilized with a single nickel ion is confirmed to be suitable for the simultaneous identification of six urinary metabolites: creatinine, taurine, histidine, 1-methylhistidine, 3-methylhistidine, and hypoxanthine. With the assistance of machine learning, an overall accuracy of 98.5% was reported. This urine sensing capacity was further applied in real-time urinary metabolite monitoring after diet and physical exercise. These proofs of concept suggest a variety of nanopore-based body fluid analyses for early diagnosis, health management, pharmacokinetic monitoring, and doping tests.

With the aggravation of pollutants damage to the environment, it is crucial yet rather challenging to design powerful matrixes for nanomaterials with high ions permeability and migration, as well as multiple exposed active sites. However, most of the current nanomaterial matrixes just act as carrier supporters with introduced invalid mass. Here, a gradient natural polycation electrolyte (chitosan) hydrogel was engineered as nanomaterials matrix to boost anionic heavy metals removal such as antimonate (Sb(V)) and arsenate (As(V)) for the first time, based on the generated interfacial and build-in electric potentials due to the diffusion of counterions. The resulting dual electric potentials allow for rapid diffusion and removal efficiency of heavy metal ions. Exemplified by Sb(V) removal, the chitosan gradient hydrogel matrixes with embedded UiO-66 MOFs exhibited a 3-fold higher removal velocity than the homogeneous hydrogel matrix, and their removal efficiencies were also promoted by gathering on the high charge density side of hydrogels. In contrast to the other reported matrix materials without driving force, the gradient polyelectrolyte hydrogel can greatly boost the removal efficiency and shorten the treatment times. This study offers inspirations on eliminating heavy metal contaminations based on self-driving forces stemming from the heterogeneous structure of materials.

Small chiral metabolic molecules are increasingly recognized as pivotal biomarkers for disease monitoring and treatment. Here, a wearable microfluidic patch is presented that integrates chiral 3D plasmonic nanostructures with surface-enhanced Raman spectroscopy (SERS) sensing activity for the in situ and real-time metabolic profiling of chiral molecules in sweat. The microfluidic patch is designed for the direct, in situ capture and storage of microliter volumes of sweat. By exploiting the 3D chiral plasmonic coupling interactions within the nanostructures, the integrated SERS sensor on the patch allows for highly sensitive and quantitative enantiomer detection through their unique fingerprint SERS spectra. In a proof-of-concept demonstration, the first in situ, real-time quantitative detection of chiral drug metabolite and pH in human sweat using this device is successfully conducted. This capability enables the capturing of an individual's dynamic metabolic profile. Leveraging this solution, pharmacokinetic correlations are established, showcasing the potential application of the device in assessing human health.

The emulsion binder system has been widely used in the inkjet printing technology of energetic materials because of its remarkable advantages. A water-in-oil (W/O) binder system was designed using an aqueous polyvinyl alcohol solution as the aqueous phase and fluorocarbon resin ethyl acetate solution as the oil phase. The physicochemical properties of a typical oil-in-water (O/W) and the proposed W/O binder systems were characterized. The W/O binder system demonstrated prominent advantages in its water resistance, moisture absorption, and curing rate. To verify the advantages of the W/O binder system in the field of energetic inks, the curing rates of two kinds of inks containing a high melting explosive (HMX) were compared. The results showed that the W/O binder system rapidly solidified and the corresponding ink exhibited a higher curing rate. Thus, the energetic material HMX was suspended in the W/O binder system and inkjet-printed. The molded sample of HMX-based W/O suspension ink exhibited excellent surface morphology, internal structure, and mechanical properties compared with the O/W molded sample. An innovative research path toward optimizing the curing rate of emulsion binder systems is presented, thereby improving the mechanical properties and microscale detonation characteristics of inkjet printing samples.

Background: Obesity is associated with hormonal imbalance, increased oxidative stress, and inflammation in the testis. These conditions adversely affect sperm quality, leading to impaired male fertility. Therefore, therapeutic interventions to counteract the adverse effects of obesity are crucial. This study explored the therapeutic effects of 4-hydroxyisoleucine (4-HIL) on fertility in male mice with diet-induced obesity. Methods: C57BL6 male mice (n=45) were randomly divided into normal diet (ND, n=15) and high-fat diet (HFD, n=30) groups for 10 weeks. The HFD group was then randomized into untreated (HFD, n=15) and 4-HIL-treated (200 mg/kg/day, intraperitoneal injection, HFD + 4-HIL group, n=15) for 6 weeks. ND and HFD controls received saline (0.3 mL/30 g body weight) throughout the intervention period. Comprehensive evaluations included (1) metabolic assessments (body weight, glucose, insulin and pyruvate tolerance tests, and blood serum lipids), (2) sperm analysis (count, concentration, and morphology), (3) fertility testing (mating trials and in vitro fertilization), (4) testicular histopathology (fat deposition and apoptosis), (5) biochemical assays (reproductive hormones, oxidative stress markers, and inflammatory cytokines), and (6) molecular analyses (mRNA sequencing and qPCR validation of differentially expressed genes). Results: 4-HIL treatment improved metabolic parameters, including reduced weight gain, enhanced glucose tolerance, and optimized blood serum lipids, compared to HFD controls. Treated mice exhibited superior sperm quality with increased count and concentration, reduced histomorphological abnormalities in the testis, and attenuated oxidative stress and inflammation. Furthermore, the key spermatogenic gene expressions, including spem1 and spata24, were significantly optimized in the testes of mice treated with 4-HIL compared to those of untreated mice (HFD group). Conclusion: This study demonstrates that 4-HIL therapy ameliorates obesity-induced testicular dysfunction and improves fertility markers in mice. The beneficial effects of this compound on metabolic parameters, sperm quality, and spermatogenic gene expression suggest its potential as a therapeutic agent for obesity-related male infertility. Further research is warranted to elucidate the underlying mechanisms and assess the clinical translatability of these findings.

l -threonine aldolase (LTA) catalyzes the synthesis of β -hydroxy- α -amino acids, which are important chiral intermediates widely used in the fields of pharmaceuticals and pesticides. However, the limited thermostability of LTA hinders its industrial application. Furthermore, the trade-off between thermostability and activity presents a challenge in the thermostability engineering of this enzyme. This study proposes a strategy to regulate the rigidity of LTA's V-shaped subunit by modifying its opening and hinge regions, distant from the active center, aiming to mitigate the trade-off. With LTA from Bacillus nealsonii as targeted enzyme, a total of 25 residues in these two regions were investigated by directed evolution. Finally, mutant G85A/M207L/A12C was obtained, showing significantly enhanced thermostability with a 20 °C increase in T 50 60 to 66 °C, and specific activity elevated by 34 % at the optimum temperature. Molecular dynamics simulations showed that the newly formed hydrophobicity and hydrogen bonds improved the thermostability and boosted proton transfer efficiency. This work enhances the thermostability of LTA while preventing the loss of activity. It opens new avenues for the thermostability engineering of other industrially relevant enzymes with active center located at the interface of subunits or domains.

Researches have proposed that obesity might contribute to development of oligoasthenospermia. This study was performed to confirm whether obesity contributes to oligoasthenospermia as well as the underlying mechanisms in mice fed with a high fat diet (HFD). Meanwhile, the actions of metformin, a drug of well-known weight-lowering effect, on sperm quality in obese mice were investigated. Our results showed that HFD feeding reduced sperm quality and steroid hormone levels in mice, associated with disruptions in testicular histomorphology and spermatogenesis. Moreover, obesity increased sperm apoptosis. These effects could be prevented by metformin treatment in HFD-fed mice. Mechanistically, an increasement in lipid contents associated with decreased hormone-sensitive lipase (HSL) protein expression in testes in HFD-fed mice was observed, which could be improved by metformin treatment. Then, the model of TM4 mouse Sertoli cells stimulated with palmitic acid (PA) was used to investigate the potential effect of lipid retention on testicular apoptosis and sperm quality reduction. In consistent, PA exposure elevated lipid contents as well as apoptosis in TM4 cells, which could also be improved by metformin treatment. Of note, the protein expression of HSL was reduced stimulated by PA in TM4 cells, also rescued by metformin. Then, anti-apoptosis effect of metformin would be lost with the deficiency of HSL . In summary, our study propose that obesity contributes to oligoasthenospermia by increasing sperm apoptosis induced by impaired lipid hydrolysis due to HSL down-regulation, which could be prevented with metformin treatment via regulating the expression of HSL in testis in mice.
