Sustained-release nanofilms incorporated with oregano essential oil (OEO) were developed by emulsion electrospinning using polyvinyl alcohol (PVA) and quaternary-ammonium chitosan (QAC). The SEM, FTIR and XRD results showed that OEO was successfully encapsulated in the PVA/QAC nanofiber with a diameter range of 158.0–179.6 nm. The incorporation of OEO significantly enhanced the mechanical and barrier properties of PVA/QAC nanofilm, concomitantly mitigating its hydrophilic nature. The prepared PVA/QAC-OEO nanofilms demonstrated excellent antioxidant properties and antimicrobial activities against S. aureus and E. coli (the maximum inhibition zones achieving 23.13 ± 0.72 mm). The release kinetics results showed sustained release of OEO from the PVA/QAC nanofilms, and the release kinetics of OEO adhered to the Fickian + First order dynamics model (R 2 = 0.9570–0.9997), indicating that a combination of random diffusion and polymer structural relaxation influenced the release of OEO. Furthermore, preservation research showed that the PVA/QAC-OEO nanofilms significantly inhibited the rise in pH, total volatile base nitrogen (TVB-N), thiobarbituric acid reactive substances (TBARs), and total viable count (TVC) in chicken breasts stored at 4 °C, while also maintaining their color and texture properties. Particularly, chicken breasts wrapped in PVA/QAC-OEO 3 film exhibited an extended shelf life for 4 days. Therefore, the PVA/QAC-OEO nanofilms could be a prospective packaging material for meat preservation.

Hydrogels incorporating natural biopolymer and adhesive substances have extensively been used to develop bioactive drugs and to design cells encapsulating sturdy structure for biomedical applications. However, the conjugation of the adhesive in most hydrogels is insufficient to maintain long-lasting biocompatibility inadequate to accelerate internal organ tissue repair in the essential native cellular microenvironment. The current work elaborates the synthesis of charged choline-catechol ionic liquid (BIL) adhesive and a hydrogel with an electronegative atom rich polyphenol (PU)-laden gelatinmethacryloyl (GelMA) to improve the structural bioactivities for in vivo tracheal repair by inducing swift crosslinking along with durable mechanical and tissue adhesive properties. It was observed that Bioactive BIL and PU exhibited potent antioxidant (IC 50 % of 7.91 μg/mL and 24.55 μg/mL) and antibacterial activity against E. coli , P. aeruginosa and S. aureus . The novel integration of photocurable GelMA-BIL-PU revealed outstanding mechanical strength, biodegradability and sustained drug release. The in vitro study showed exceptional cell migration and proliferation in HBECs, while in vivo investigation of the GelMA-BIL-PU hydrogel on a rat's tracheal model revealed remarkable tracheal reconstruction, concurrently reducing tissue inflammation. Furthermore, the optimized GelMA-BIL-PU injectable adhesive bioink blend demonstrated superior MSCs migration and proliferation, which could be a strong candidate for developing stem cell-rich biomaterials to address multiple organ defects.

The ingrowth of endogenous cells into tissue scaffolds and modulation of Treg population take a pivotal function in restoring the structure and function of heart post-acute myocardial infarction (MI). However, the traditional hydrogels are failed to achieve such functions due to their critically small mesh size and inherent bioinertness. In this study, a Treg-regulated microgel scaffold (GTK-TK-drug) was prepared by inter-microgels assembly of microgels having cyclodextrin (CD) or adamantane (Ad) with a particle size of about 220 μm. The raw materials to obtain the microgels included gelatin methacryloyl (GelMA), reactive oxygen species (ROS)-responsive crosslinking agent poly(ethylene glycol)-thioketal-poly(ethylene glycol) (PEG-TK-PEG), ROS-responsive release prodrug aminooxyacetic acid (PEG-TK-AOA), PEG-CD or PEG-Ad, and photoinitiator lithium phenyl-2,4,6-trimethylbenzoylphosphinate (LAP). Quantitative real-time PCR (qPCR), cell counting kit-8, and immunofluorescence staining revealed the effectiveness of AOA for the differentiation of T cells into Treg and the ingrowth of cells in vitro and in vivo . During the short treatment of MI in vivo , the levels of plasma inflammatory factors, cardiomyocyte apoptosis and inflammatory cells were reduced significantly. In the long-term animal experiments, the recovery of cardiac function was significantly enhanced as characterized by echocardiography and the degree of myocardial fibrosis compared to the unassembled microgels control.

Fermentation with lactic acid bacteria (LAB) enhances the nutritional value and flavor of food matrices. This study investigated the impact of Pediococcus acidilactici HNU168, Bifidobacterium animalis subsp. lactis HNU329, and their combined fermentation on Millennium cherry tomato juice (MFJ). Fermentation boosted organic acids, total flavonoid content (TFC), xanthine oxidase (XOD) inhibitory activity, and antioxidant activities. Mixed-strain fermentation significantly increased antioxidant activity and lycopene content beyond single-strain fermentation. Non-targeted metabolomics identified 745 differential metabolites, mainly enriched in purine metabolism and phenylpropanoid biosynthetic pathways, facilitating key metabolite biotransformation. Notably, lycoperodine l, 3-(4-hydroxyphenyl) lactate, indole-3-lactic acid, and dihydrocoumarin contents increased significantly. Molecular docking and XOD inhibitory activity confirmed strong interactions between key metabolites and XOD. Also, electronic tongue analysis showed that LAB-fermented MFJ had increased richness and reduced bitterness and astringency. These results illustrate that LAB fermentation improves MFJ's nutrition, bioactivities, and sensory properties, offering potential applications in functional or anti-hyperuricemia foods.

Plant polyphenols have gained increasing attention due to their abundant and powerful biological activities. However, polyphenols from roots of Ilex asprella (RIAPs) remain largely unexplored, with limited research on their composition and bioactivity, making it necessary to focus on RIAPs. In the present study, single - factor analysis and response surface optimization were performed for the extraction of RIAPs via an ultrasonic bath extraction method. The findings demonstrated that at 50 % ethanol concentration, 30 min of ultrasonic time, and 300 W of ultrasonic power, the maximum extraction content of RIAPs was 30.14 ± 0.41 mg/g. Compound and structural analysis via high-resolution liquid chromatography-mass spectrometry (LC-MS/MS) and a Fourier transform infrared (FT-IR) spectrometer confirmed that polyphenols were the majority of compounds comprising RIAPs, in addition to small amounts of alkaloids and trace amounts of other compounds such as amino acids. Scanning electron microscope (SEM) images revealed that RIAPs exhibited a densely reticulated and fragmented microstructure. In addition, bioactivity experiments demonstrated significant antioxidant, antibacterial, anti-inflammatory, and anti-hyperlipidemic activities of RIAPs, supporting their potential pharmaceutical applications. This study provides an experimental foundation for further pharmacological research and potential drug development of RIAPs.

Wound dressings equipped with disinfection and wound tissue regeneration capabilities demonstrate significant potential in chronic wound management. However, current multifunctional dressings can rarely accommodate multiple repair processes accurately due to the time-dependent variational wound microenvironment. Herein, a Janus wound dressing is developed based on electrospun nanofibrous scaffold featuring programmed drug release performance and asymmetrical wettability. Specifically, polyvinylidene fluoride (PVDF) nanofiber membrane as the outer layer to resist the external biofouling and provide essential mechanical support for the inner layer was firstly constructed. Subsequently, the inner layer composed of polyvinyl pyrrolidone (PVP) and polylactic acid (PLA) fibers which are separately loaded with bacterial-targeted nanoparticles (VHA NPs) and composite particle of zeolite imidazolate frameworks-8 containing deferoxamine (ZD NPs) is fabricated on the PVDF membrane. Experimental results demonstrated that the degradation rate of the inner layer could be precisely controlled to sequentially deliver the loaded drugs into the wound site. Taking advantages of the synergistic therapeutic effects including significant antibacterial activity, admired antioxidant property and remarkable angiogenesis performances, the fabricated Janus dressing (PF/PHPD) achieves superior performance for promoting infected wound healing in a full-thickness skin wound model, much better than that of conventional bandage and clinical antibiotic treated wound.

Ultrasound assisted hot water extraction (UAHWE) was applied to extraction of polysaccharides from Taraxacum mongolicum with hot water as extract solvent. Experimental factors in UAHWE process were optimized by response surface methodology. The optimal extraction parameters to achieve the highest Taraxacum mongolicum polysaccharides (TMPs) yield (12.08 ± 0.14)% by UAHWE were obtained under the ultrasound power of 200 W, extraction temperature of 62°C, solid-to-liquid ratio of 1:20 g/mL, and extraction time of 40 min, and then the crude TMPs were further purified by DEAE-52 and Sephadex G-100 chromatography to obtain a homogenous polysaccharide fraction (TMPs-1-SG). Subsequently, the structure of TMPs-1-SG was characterized by UV-vis, Fourier transform infrared spectroscopy (FT–IR), high performance gel permeation chromatography (HPGPC), high performance liquid chromatography (HPLC), scanning electron microscope (SEM), transmission electron microscopy (TEM), and Congo red test. The results display that TMPs-1-SG with an average molecular weight of 5.49 × 10 4 Da was comprised of mannose (Man), galactose (Gal), xylose (Xyl), and arabinose (Ara) with a molar ratio of 39.85:52.61:27.14:6.30. Moreover, TMPs-1-SG did not contain a triple helix structure. Furthermore, TMPs-1-SG and TEM presented a sheet-like, rod-shaped, and irregular structure. Finally, the antioxidant activity of TMPs-1-SG was evaluated by in vitro experiment. The IC 50 values of scavenging DPPH and OH radicals for TMPs-1-SG achieved 0.71 mg/mL and 0.75 mg/mL, respectively. The findings can provide an effective method for extracting polysaccharides from natural resources.

Ultrasound assisted aqueous two-phase extraction (UATPE) is used to extract polysaccharides from Ophiopogon japonicus (OJPs). Experimental factors in UATPE process are optimized by response surface methodology. The optimal extraction conditions are obtained as follows: the ethanol concentration of 34%, ammonium sulfate concentration of 23%, solid-to-liquid ratio of 1:17 g/mL, and ultrasound power of 480 W, and the yield of OJPs is 9.41% ± 0.12%, and then the crude OJPs are further purified by different column chromatography to obtain a homogenous fraction (OJPs-2-SG), and identified its structure via different methods. OJPs-2-SG with a molecular weight of 1.25 × 105 Da is comprised of mannose (Man), galactose (Gal), glucose (Glc), and rhamnose (Rha) with a molar ratio of 5.15: 26.39: 68.72:8.41. Moreover, OJPs-2-SG contained α and β glycosidic bonds, and didn't showed a triple helix structure. Furthermore, OJPs-2-SG described irregular structures, and cylindrical and conical block like features. The IC50 values of scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl (OH), and superoxide (O2-) radicals for OJPs-2-SG achieved 3.72 mg/mL, 3.89 mg/mL, and 8.71 mg/mL, respectively. These results provide important references for the extraction and application of polysaccharides.

In the current study, a novel crude polysaccharide (cNCEP) was extracted from N. commune Vaucher utilizing ultrasonic-assisted extraction (UAE) with 60 % ethanol, employing response surface methodology. The optimal yield of cNCEP was determined to be 8.07 ± 0.08 mg/g, achieved through ultrasonic-assisted extraction under the conditions of a material-to-liquid ratio of 1:22, temperature of 56 °C, power of 570 W, and duration of 147 min. Subsequent purification of NCEP via Sephadex G75 resulted in a novel polysaccharide with a molecular weight of 20.466 kDa. NCEP exhibited significant scavenging activites against DPPH and hydroxyl radicals, as well as notable in vitro immunomodulatory properties. Furthermore, the mechanisms underlying the immunomodulatory effects of NCEP, involving enhancement of immunity, were investigated, revealing potential regulation of MAPK and TLR4-IRF7-NF-κB signaling pathways through RNA-Seq and Western blot analyses. These findings highlight the promising potential of NCEP as an organic immunomodulatory agent and functional food ingredient.

Pterostilbene (Pte) is a dietary phenolic compound derived from plants that offers numerous physiological advantages. Nonetheless, its low water solubility and limited oral bioavailability restrict its bioactivity. To address these limitations, nanoparticles were prepared using anti-solvent precipitation method. Optimal nanoparticles were achieved at a 10:1 mass ratio of whey protein isolate (WPI) to Pte, resulting in a particle size of 104.54 nm, a polydispersity index of 0.42, and an impressive encapsulation efficiency of Pte at 85.02%. The water solubility of Pte in the nanoparticles was found to be 53.14 times higher than that of free Pte and remained stable under various environmental conditions. Furthermore, the DPPH and ABTS + free radical scavenging activities of the nanoparticles were maximized at 55.58% and 92.97%, respectively. Pte release from WPI-Pte nanoparticles was well-fitted to Korsmeyer-Peppas model and followed a non-Fickian mechanism controlled by both diffusion and erosion. This environmentally friendly, straightforward, and efficient approach shows promise for large-scale production of Pte products with amplified health benefits.

Rosa roxburghii Tratt fruits (RRT) exhibit extremely high nutritional and medicinal properties due to its unique phytochemical composition. Probiotic fermentation is a common method for processing fruits. Variations in the non-volatile metabolites and bioactivities of RRT juice caused by different lactobacilli are not well understood. Therefore, we aimed to profile the non-volatile components and investigate the impacts of L. plantarum fermentation (LP) and L. paracasei fermentation (LC) on RRT juice (the control, CG). There were both similarities and differences in the effects of LP and LC on RRT juice. Both of the two strains significantly increased the content of total phenolic, total flavonoid, and some bioactive compounds such as 2-hydroxyisocaproic acid, hydroxytyrosol and indole-3-lactic acid in RRT juice. Interestingly, compared with L. paracasei , L. plantarum showed better ability to increase the content of total phenolic and these valuable compounds, as well as certain bioactivities. The antioxidant capacity and α-glucosidase inhibitory activity of RRT juice were notably enhanced after the fermentations, whereas its cholesterol esterase inhibitory activity was reduced significantly. Moreover, a total of 1466 metabolites were identified in the unfermented and fermented RRT juices. There were 278, 251 and 134 differential metabolites in LP vs CG, LC vs CG, LC vs LP, respectively, most of which were upregulated. The key differential metabolites were classified into amino acids and their derivatives, organic acids, nucleotides and their analogues, phenolic acids and alkaloids, which can serve as potential markers for authentication and discrimination between the unfermented and lactobacilli fermented RRT juice samples. The KEGG enrichment analysis uncovered that metabolic pathways, purine metabolism, nucleotide metabolism and ABC transporters contributed mainly to the formation of unique composition of fermented RRT juice. These results provide good coverage of the metabolome of RRT juice in both unfermented and fermented forms and also provide a reference for future research on the processing of RRT or other fruits.

In this study, the structural design and physicochemical property enhancement of undenatured type II collagen (UC-II) nanofibrils with sodium alginate (SA) coating induced by calcium ions (Ca 2+ ) were investigated. The research aimed to elucidate the impact of Ca 2+ concentration on the morphology, thermal stability , and digestive resistance, as well as to assess the potential of UC-II/SA nanofibrils as a delivery system for curcumin (Cur). A series of Ca 2+ concentrations (1–9 mM) were methodically applied to optimize the condition that maintains the triple-helical structure of UC-II, thereby enhancing its functional properties. It was found that the Ca 2+ level up to 5 mM effectively preserved the structural integrity and improved thermal stability of UC-II, with the added benefit of ensuring the substantial delivery of active fragment to small intestine (70.7 %), which was 3.43 times greater than that of uncoated UC-II. Moreover, incorporating Cur into the UC-II/SA nanofibrils resulted in a 300 times increase in Cur solubility and showcased the superior dispersion stability, antioxidant activity , and sustained release profile during simulated digestion. These findings underscored the dual functionality of the UC-II/SA system as both a stabilizing agent for UC-II nanofibrils and an efficient carrier for Cur delivery.
