The characteristic flavor of Coffea arabica from Yunnan is largely attributed to the primary processing treatments through affecting the VOCs accumulation. Therefore, a rapid and comprehensive detection technique is needed to accurately recognize VOCs in green coffee beans with different pretreatment methods. Hence, we conducted volatile profiles and identified nine markers of three different primary processed green coffee beans from the major production areas in Yunnan with the combined of HS-SPME-GC–MS and PTR-TOF-MS. The relationships between the chemical composition and the content of VOCs in green coffee beans were elucidated. Among the markers, palmitic acid ( F3 ), linoleic acid ( F6 ), α -ethylidene phenylacetaldehyde ( T4 ), and phytane ( T8 ) contributed to the antioxidant activity of sun-exposed green coffee beans. In conclusion, the analytical technology presented here provided a general tool for an overall and rapid understanding of a detailed volatile profiles of green coffee beans in Yunnan.

It is still difficult for a single antibacterial modality to realize satisfactory management of bacterial breeding in food preservation. To solve this problem, we developed a photothermal-derived dual-mode synergistic bactericidal konjac glucomannan (KGM)/polycaprolactone (PCL) bilayer film incorporated with quercetin-loaded melanin-like nanoparticles (Q@MNPs). The results showed that the mechanical properties (TS: 29.8 MPa, EAB: 43.1 %), UV shielding properties, and water resistance (WCA: 124.1°, WVP: 3.92 g mm/m 2 day kPa) of KGM-Q@MNPs/PCL bilayer films were significantly improved. More importantly, KGM-Q@MNPs/PCL bilayer film presented outstanding photothermal inversion and controlled release behavior of Q triggered by near infrared (NIR) radiation, thus contributing to excellent dual-mode synergistic antibacterial properties against E. coli and S. aureus . Meanwhile, the KGM-Q@MNPs/PCL bilayer film possessed good biocompatibility and low toxicity. As a proof-of-concept application, we further verified the significant value of film for the preservation of cherry tomatoes. Since KGM-Q@MNPs/PCL bilayer film showed excellent biodegradability, this work will aid the development of sustainable antibacterial food packaging materials.

Acute kidney injury (AKI) results from the rapid deterioration of renal function, which is mainly treated by transplantation and dialysis, and has a high mortality rate. Inflammation induced by excess reactive oxygen/nitrogen species (RONS) plays a crucial role in AKI. Although small molecule antioxidants have been utilized to alleviate AKI, low bioavailability and side-effect of these drugs tremendously limit their clinical use. Hence, we successfully construct ultra-small (2–4 nm) rhodium nanoparticles modified with l -serine (denoted as Rh-Ser). Our results show that Rh-Ser with multiple enzyme-mimicking activities, allows remove various RONS to protect damaged kidney cells. Additionally, the ultrasmall size of Rh-Ser is conducive to enrichment in the renal tubules, and the modification of l -serine enables Rh-Ser to bind to kidney injury molecule-1, which is highly expressed on the surface of damaged renal cells, thereby targeting the damaged kidney and increasing the retention time. Moreover, Rh-Ser allows the production of oxygen at the inflammatory site, thus further improving hypoxia and inhibiting pro-inflammatory macrophages to relieve inflammation, and increasing the survival rate of AKI mice from 0 to 80%, which exhibits a better therapeutic effect than that of small molecule drug. Photoacoustic and fluorescence imaging can effectively monitor and evaluate the enrichment and therapeutic effect of Rh-Ser. Our study provides a promising strategy for the targeted treatment of AKI via RONS scavenging and inflammatory regulation.

Atherosclerosis (AS) is a chronic inflammation vascular disease, with its ongoing progression can lead to the onset of cardiovascular diseases. Traditional drug therapy is limited by poor drug delivery, insufficient drug accumulation, and notable toxic side effects. In addition, the failure to receive an early AS diagnosis is primarily responsible for the delayed treatment, which subsequently contributes to the high frequency of life-threatening cardiovascular events. In this work, a macrophage membranes (MM)-camouflaged reactive oxygen species (ROS)-sensitive nanotheranostic platform (LC-MM) is constructed to improve AS target diagnosis and treatment efficacy. Thanks to the strong antioxidant properties of carbon dots (CDs), CDs as drug carriers for lovastatin aimed to synergistically treat AS by reducing ROS accumulation and suppressing inflammatory responses in vitro and in vivo are employed. The active functions of MM coupled with the ROS-responsiveness of LC nanoplatform are expected to enhance the efficacy of nanotherapy, particularly to reduce lipid deposition, macrophage infiltration, necrotic core size, collagen content, pro-inflammatory cytokines, and oxidative stress accumulation. Moreover, the near-infrared emission properties inherited from CDs facilitated precise fluorescence (FL) imaging for AS plaques. Thus, the biomimetic nanotheranostic agent LC-MM represented a powerful platform for safe and effective AS management.

Maillard reaction plays an important role in the flavour and color of some food. However, the excessive occurrence of Maillard reaction may affect the quality and safety of food. In this study, a simulated Maillard system composed of gluten and glucose was established to evaluate and analyze the regulatory role of tea polyphenols (TP) in it. The addition of tea polyphenols alleviated excessive browning of the Maillard reaction and significantly inhibited the accumulation of fructosamine and late glycosylation end products in the system. Based on the analysis of laser particle sizer and atomic force microscope, the large particle size distribution and aggregation structure of the Maillard reaction products in the tea polyphenol group disappeared. Further experiments showed that with the addition of tea polyphenols, the free radicals content in the system decreased, while the amount of reaction substrates increased. It was also shown that tea polyphenols effectively inhibited the isomerism of glucose to fructose and affected the content of free sulfhydryl and free amino groups in the system. The results indicated that tea polyphenols can regulate the Maillard reaction between gluten and glucose, which gives it the potential for application in the field of bakery and thermally processed foods.

Conventional radical grafting of proteins with catechins consumed the most antioxidant-active hydroxyls during grafting, thus failing to effectively retain antioxidant activity in conjugates. In this study, a novel strategy of selective protection of the most reactive hydroxyls before grafting was developed to preserve the most reactive hydroxyls and effectively retain antioxidant activity in conjugates. Selective protection of the most reactive hydroxyls of (−)-epigallocatechin-3-gallate (EGCG) was successfully realized in a yield of 87% applying trimethyl orthopropionate and catalytic calcium triflate at 40 °C. The novel ovalbumin (OVA)–EGCG conjugate with 93% grafting ratio was prepared by radical grafting with the selectively protected EGCG and subsequent deprotection. Substantially enhanced antioxidant performance of the novel OVA–EGCG conjugate in liposomes was unveiled with notably reduced curcumin degradation and leakage. The strategy and approaches developed in this study will be valuable to effectively improve the antioxidant activities of protein–catechin grafting conjugates.

In recent years, the application of biopolymer-based nanofibers prepared via microfluidic blow spinning (MBS) for food packaging has continuously increased due to their advantages of biocompatibility, biodegradability, and safety. However, the poor spinnability, undesirable water barrier capacity, and loss of antibacterial and antioxidant properties of biopolymer-based nanofibers strictly restrict their real-world applications. In this work, carvacrol (CV) incorporated konjac glucomannan (KGM)/polylactic acid (PLA) nanofibrous films (KP-CV) were produced by MBS. The FTIR spectra and XRD analysis revealed the hydrogen bonding interactions among CV, PLA, and KGM, thus significantly improving the TS of KP-CV nanofibrous films from 0.23 to 1.27 MPa with increased content of CV from 0 % to 5 %. Besides, KP-CV nanofibrous films showed improved thermal stability, excellent hydrophobicity (WCA: 128.19°, WVP: 1.02 g mm/m 2 h kPa), and sustained release of CV combined with good antioxidant activities (DPPH radical scavenging activity: 77.51 ± 1.57 %), and antibacterial properties against S. aureus (inhibition zone: 26.33 mm) and E. coli (inhibition zone: 22.67 mm). Therefore, as prepared KP-CV nanofibrous films can be potentially applied as packaging materials for the extended shelf life of cherry tomatoes.

Adsorbents with high adsorption capacity, selectivity, and antioxidant activity are crucial to enhance the efficacy and prognosis of hemoperfusion treatments for patients with lead poisoning. This study presents the development of polyphenol-analogous dextran hydrogels (PYG@Dex) with abundant chemically conjugated pyrogallol (PYG) groups on their networks, which were prepared by Hantzsch cross-linking reaction involving the formation of 1,4-dihydropyridine from acetoacetylated dextran, 3,4,5-trihydroxybenzaldehyde, and NH 4 OAc. The resulting PYG@Dex hydrogel exhibited superior biocompatibility and robust antioxidant capabilities, achieving a free radical scavenging rate of over 90 %. Notably, the hydrogel with high PYG content displayed an outstanding adsorption capacity of 102.35 mg·g −1 for Pb 2+ ions, due to the coordination interactions with PYG and β -ketoester groups. The adsorption affinity of PYG@Dex for Pb 2+ ions was confirmed in the presence of biologically-relevant metal ions (K + , Na + , Ca 2+ , and Mg 2+ ) in both aqueous solutions and simulated plasma. These findings underscore the potential of PYG-conjugated hydrogels to address the challenges of selective heavy metal removal from blood through hemoperfusion and to provide additional protection against heavy metal-induced oxidative stress responses.

Thiol-modified hyaluronic acid (HASH) improves zein nanoparticles (ZNPs) stability by disulfide bonding with zein. However, the effect of the degree of thiol substitution in HASH on the stability of the ZNPs remains unclear. In this study, HASH with 1%, 3%, and 5% degrees of thiol substitution was synthesized and used as stabilizers to prepare three composite NPs. The pH and salt ion stability of the composite NPs increased with increasing degree of thiol substitution, which improved the antioxidant and anti-inflammatory capacities of the composite NPs. Interestingly, when the degree of thiol substitution exceeded 3%, the physical stability of composite NPs did not improve further. Our results revealed the relationship between the degree of thiol substitution in HASH and the stability of ZNPs, and provided the optimal degree of thiol substitution. These findings provided important information for improving the physical stability of ZNPs and contributed to the development of a delivery system for natural actives.

Effects of red and blue light treatment on physiological quality, microbial loads, redox status and metabolomics profiles of fresh-cut sugarcane in vacuum and plastic packages were investigated during 15 d storage. The results showed that light synergistic vacuum treatment delayed the decrease of pH and the increase of respiration rate and microbial loads, enhanced antioxidant capacities and related enzymes activities. Light treatment was beneficial to 1 O 2 generation, but had opposite effects on O 2 − , H 2 O 2 and malondialdehyde. O 2− and H 2 O 2 was negatively associated with CAT, sucrose, fructose, glucose, 2-oxoglutaramate, liquiritigenin and dihydromyricetin, positively with PPO and malondialdehyde. Only phenylacetaldehyde exhibited a negative correlation with 1 O 2 . The biosynthesis of sugars, amino acids and flavonoids were the principal metabolite pathways corresponding to oxidative stress in fresh-cut sugarcane. It could be concluded that the concentration of ROS, especially O 2− and H 2 O 2 , should be appropriate to kill bacteria and retain the quality of fresh-cut sugarcane.

The intracellular delivery of protein drugs via nanocarriers offers significant potential for expanding their therapeutic applications. However, the unintended activation of innate immune responses and inflammation triggered by the carriers presents a major challenge, often compromising therapeutic efficacy. Here, we present oligoethylenimine-thioketal (OEI-TK), a reactive oxygen species-responsive cationic polymer with intrinsic anti-inflammatory properties, to overcome this challenge. OEI-TK self-assembles electrostatically with bovine serum albumin (BSA) to form stable nanoparticles (OTB NPs) with excellent encapsulation efficiency. In vitro studies confirmed that OTB NPs retained OEI-TK’s antioxidant and anti-inflammatory properties, enhanced biocompatibility, and efficiently delivered BSA into cells. Furthermore, OEI-TK facilitated the intracellular delivery of β-galactosidase while preserving its enzymatic activity, demonstrating its potential for functional protein transport. These findings highlight OEI-TK as a promising platform with dual benefits of inflammation modulation and intracellular protein delivery, holding potential for the synergistic treatment of inflammation-related diseases.

Primary sclerosing cholangitis (PSC) is a challenging cholestatic liver disease marked by progressive bile duct inflammation and fibrosis that has no FDA-approved therapy. Although obeticholic acid (OCA) has been sanctioned for PSC, its clinical utility in PSC is constrained by its potential hepatotoxicity. Here, we introduce a novel therapeutic construct consisting of OCA encapsulated within a reactive oxygen species (ROS)-responsive, biodegradable polymer, further cloaked with human placenta-derived mesenchymal stem cell (hP-MSC) membrane (MPPFTU@OCA). Using PSC patient-derived organoid models, we assessed its cellular uptake and cytotoxicity. Moreover, using a PSC mouse model induced by 3,5-diethoxycarbonyl-1,4-dihydro-collidine (DDC), we demonstrated that intravenous administration of MPPFTU@OCA not only improved cholestasis via the FXR-SHP pathway but also reduced macrophage infiltration and the accumulation of intracellular ROS, and alleviated mitochondria-induced apoptosis. Finally, we verified the ability of MPPFTU@OCA to inhibit mitochondrial ROS thereby alleviating apoptosis by measuring the mitochondrial adenosine triphosphate (ATP) concentration, ROS levels, and membrane potential (ΔΨm) using H 2 O 2 -stimulated PSC-derived organoids. These results illuminate the synergistic benefits of integrating an ROS-responsive biomimetic platform with OCA, offering a promising therapeutic avenue for PSC.
