The development of intelligent nanoplatforms that integrate tumor targeting, stimuli-responsive drug release, and multimodal therapy remains a significant challenge for precise cancer treatment. Herein, a nanoplatform based on DNA nanoflowers and Mn:CuS nanoparticles with a unique architecture and multiple therapeutic functions was constructed. The DNA nanoflowers derived from rolling circle amplification (RCA) served as the scaffold, enabling the efficient intercalation of Mn:CuS nanoparticles and doxorubicin (DOX). Furthermore, the sgc-8 aptamer was incorporated into the DNA scaffold for specific recognition of HeLa cells. The intercalated Mn:CuS nanoparticles exhibited photothermal therapeutic activity, peroxidase (POD)/oxidase (OXD)-like properties and glutathione peroxidase (GPx)-like activity, thus achieving amplified therapeutic efficacy toward tumor cells. Moreover, the DOX release was demonstrated to be precisely controllable in acidic environments and under near-infrared (NIR) irradiation, which ensured tumor-specific drug release. Thus, synergistic chemotherapy, photothermal therapy and chemo-dynamic therapy (CDT) of cancers were achieved. Compared with the conventional single-function nanocarriers, this multifunctional hybrid system combines specific target recognition, responsive drug delivery, and multimodal therapeutic treatment, offering a promising avenue for precise and effective cancer therapy.

Highly sensitive nucleic acid detection is essential for biomedical diagnostics and bioanalytical applications, yet remains challenging due to the limited amplification efficiency and system complexity of existing sensing strategies. Herein, we report a simplified yet efficient cascade amplification strategy for nucleic acid detection by exploiting the multifunctionality of Exonuclease III (Exo III). In this design, a single Exo III enzyme simultaneously drives two mechanistically distinct amplification processes. In the first stage, target DNA triggers Exo III-assisted strand displacement, enabling efficient target recycling and generation of abundant trigger strands. In the second stage, the released strands activate an Exo III-powered DNA walker amplification process on gold nanoparticle surfaces, resulting in amplified fluorescence signal. Benefiting from the synergistic integration of solution-phase target recycling and interface-confined DNA walker-based amplification, the proposed system achieves high sensitivity with reduced enzymatic and operational complexity, and the limit of detection is 48.8 pM. This work provides a versatile and sequence-independent cascade amplification paradigm for advanced nucleic acid biosensing applications.

Capillary electrophoresis (CE) is widely used for DNA fragment analysis, but its separation performance depends strongly on the composition of the polymer sieving matrix, especially in multicapillary systems where matrix operability and reproducibility are critical. Herein, a laboratory-built multicapillary electrophoresis system was used to optimize the sieving matrix for high-throughput DNA fragment separation. The system integrated twelve fused-silica capillaries, high-voltage electrokinetic injection, fluorescence detection, and digital electropherogram acquisition. Using a 100 bp DNA ladder as the model sample, the effects of poly(ethylene oxide) (PEO), Tween 20, and glycerol on electropherogram quality were systematically investigated. PEO concentration was the dominant factor controlling the dynamic sieving network: 0.1% PEO provided insufficient separation, whereas 0.8–1.0% PEO produced clearly resolved DNA peaks. Tween 20 improved peak regularity and electropherogram quality under the tested conditions, with 0.05% providing sufficient improvement without prolonging migration time. Glycerol affected peak distribution by increasing apparent migration resistance in the polymer matrix; however, excessive glycerol slowed DNA migration and markedly extended the separation window. Considering separation quality, matrix operability, and analytical efficiency, 1.0% PEO, 0.05% Tween 20, 2.5% glycerol, 1× SYBR Gold, and 0.5× TBE were selected as the optimized sieving matrix. Under the optimized matrix, inter-capillary migration-time alignment improved the consistency of parallel capillary outputs, with corrected migration-time RSD values generally below 0.5%. The optimized formulation provides a practical basis for high-throughput CE-based DNA fragment analysis.

Scandium (Sc), as a critical rare earth element (REE),
holds significant
application value in advanced manufacturing, energy materials, catalysis,
and other fields. However, due to the common coexistence of Sc with
light and heavy REEs, which exhibit high similarities in ionic radius,
valence state, and coordination chemistry, conventional spectrophotometric
methods suffer from poor selectivity, making accurate quantification
of Sc 3+ in complex matrices difficult. In this study, we
present a label-free ion sensing platform based on silicon nanowire-modified
glass micropipettes (SiNWs@GMP). By incorporating the Sc-1 DNA recognition
unit, the platform achieves highly sensitive and selective detection
of Sc 3+ as well as sequential discrimination between light
and heavy REEs. The three-dimensional porous SiNWs network inside
the GMP provides abundant immobilization sites for Sc-1 DNA. Under
an applied electric field, the specific binding between Sc 3+ and Sc-1 DNA modulates ion transport within the channel, generating
measurable changes in ionic current. The constructed sensor exhibits
a detection concentration range for Sc 3+ from 1 pM to 10 5 pM and a detection limit for Sc 3+ as low as 1
pM, which can effectively distinguish Sc 3+ from light REEs.
Meanwhile, the use of EDTA to selectively suppress interference from
coexisting heavy REEs further verifies the high selectivity of Sc-1
DNA. This platform demonstrates good stability, repeatability, and
reliability in aqueous environments, highlighting its potential for
applications in environmental monitoring and REEs analysis.

Effective detection of Enterocytozoon hepatopenaei (EHP) in aquaculture is currently hindered by the lack of field-deployable extraction methods capable of processing complex, inhibitor-rich hepatopancreatic tissue. This study presents a gravity-driven millifluidic platform for the rapid extraction of EHP genomic DNA using an optimized, surfactant-compatible magnetic solid-phase extraction (MSPE) chemistry. Utilizing 5% PEG 8000 and 2.0 M NaCl, the platform facilitates the selectively capture of DNA from inhibitor-rich crustacean lysates. The 3D-printed device employs a tilting rocking plate to generate passive, gravity-driven flow, maintaining homogeneous magnetic bead suspension and maximizing solid-phase capture efficiency without external pumps. The integrated platform achieved a DNA yield of 2804.33 ± 15.31 ng/μL, a 5.9-fold increase over manual magnetic bead extraction. TaqMan quantitative PCR (qPCR) validation targeting the EHP SSU rRNA gene was developed. Using a standard curve spanning 10 1 to 10 7 plasmid copies (Ct = −3.611 log 10 [copy] + 42.309, R 2 = 0.998, amplification efficiency 89.2%), the on-chip MSPE achieved a validated analytical limit of detection (LOD) of 1 spore per reaction (100% detection rate, n = 21), whereas a commercial CTAB-based DNA extraction kit failed to achieve a validated LOD even at 10 spores (85.7%, 18/21). Nested PCR targeting the SWP gene was employed for field evaluation. A pilot study across two cohorts (N = 40) demonstrated consistent detection of confirmed EPH-positive cases; however, the small sample size precludes definitive diagnostic accuracy claims. With a total processing time under 30 min, this platform provides a high-efficiency extraction module. Future work will couple the device with isothermal amplification (e.g., LAMP or RPA) to realize a sample-to-answer system for resource-limited aquaculture.

p-STAT4 directly binds to the Ak4 promoter to drive its transcription. • STAT4-AK4 signaling engages in the production of mitochondrial DNA (mtDNA). • TAMR inhibits Ak4 transcription and mtDNA synthesis in a STAT4-dependent manner. • TAMR ameliorates UC by suppressing the JAK2-STAT4 signaling pathway. • The JAK2-STAT4-AK4-mtDNA axis represents a novel therapeutic target for UC. Background Ulcerative colitis (UC), a chronic inflammatory bowel disease with increasing global incidence, has inadequate existing treatments, highlighting the urgent need for new therapeutic agents and strategies. The total alkaloids from Menispermi Rhizoma (TAMR) has been clinically used to treat inflammatory diseases in traditional Chinese medicine, but its specific therapeutic effects and mechanisms in UC remain poorly elucidated. Purpose This study aimed to systematically clarify the therapeutic efficacy of TAMR against UC and elucidate its potential molecular mechanisms. Methods A dextran sulfate sodium (DSS)-induced murine colitis model was used to evaluate the efficacy of TAMR, with RNA sequencing providing mechanistic insights. A variety of molecular biology experiments were conducted for subsequent pathway validation in RAW264.7 macrophage cells. STAT4-AK4 interactions were further explored via Stat4 knockdown and chromatin immunoprecipitation (ChIP) assays. Results TAMR treatment effectively ameliorated colonic pathological lesions and reestablished M1/M2 macrophage polarization in DSS-challenged mice. Integrated RNA-seq and Western blot profiling revealed that TAMR specifically suppressed JAK2-STAT4 signaling cascade and AK4 expression, whose inhibition curbed excessive mitochondrial DNA (mtDNA) synthesis in inflammatory macrophages. Notably, STAT4 overexpression entirely negated TAMR-driven AK4 and mtDNA suppression. ChIP experiments confirmed that p-STAT4 directly bound to the Ak4 promoter, and STAT4 silencing abrogated LPS-induced Ak4 transcription and mtDNA synthesis. Conclusions TAMR exerts its anti-colitis effects by inhibiting the JAK2-STAT4 signaling pathway, which in turn suppresses AK4-mediated mtDNA overproduction. This study provides a scientific basis for the clinical application of TAMR in UC treatment and identifies a novel JAK2-STAT4-AK4-mtDNA regulatory axis as a potential therapeutic target for UC. Investigation of rapid and sensitive method for detecting DNA damage is meaningful for screening the chemical compounds with potential genotoxicity and for earlier diagnosis of the related diseases. In this study, we reported a gold nanoparticle (AuNP) based exposure lateral flow strip (LFS) to determine oxidative DNA damage induced by two typical damaging reagents, including transition metal (Fenton reagents, Fe 2+ /H 2 O 2 ) and organic compounds (Rose Bengal under irradiation). The presence of human 8-oxoguanine DNA glycosylase (HOGG 1) and human apyrimidinic endonuclease (APE 1) converted the oxidized base, 8-oxo-7,8-dihydro-2′-deoxyguanosine (denoted as 8-oxodGuo, one of the major products of oxidative damage DNA), to strand break in a hairpin DNA probe. The opened loop acted as a linker to anchor the AuNP@DNA complexes on the test line (T-line). The visible red color appearing on the T-line was analyzed by employing Image J software for digital image acquisition, enabling quantification of DNA lesions with the measured mean grey value. Quantification of 8-oxodGuo was achieved with an estimated detection limit down to 0.003 pmol. About 67-fold more sensitive than the previously reported elimination LFS is mainly ascribed to the specific hairpin DNA structure to capture the signal probe complex and the exposure LFS signal readout. Furthermore, oxidatively generated DNA damage by Rose Bengal or Fe 2+ /H 2 O 2 were then assessed. The proposed exposure LFS detected as low as 2 nM of Rose Bengal or 0.06 nM Fe 2+ /0.24 nM H 2 O 2 produced DNA lesions, respectively. It shows the potential to serve as a screening tool for rapidly acquiring toxicological data on emerging chemicals.

Cervical cancer is one common cause of cancer relative death in women worldwide, while microRNA-21 (miR-21) can serve as one upregulated miRNA in cervical cancer for early diagnosis. Chemiluminescence is a powerful assaying method, but balancing the sensitivity and selectivity is often difficult at present. For this problem, a photocatalytic chemiluminescent biosensor was developed to homogeneously assay miR-21 with good performances. Based on a base stacking principle, cooperative hybridization is employed in double DNA probes for biosensor design. After DNA probe hybridization, photocatalytic DNA-dye complex can induce luminol’s chemiluminescence signals via several processes. Here the double-probe design results in the high sensitivity to assay miR-21 with a detection limit in pM level. Meanwhile, the short probe design and cooperative principle guarantee a good selectivity to discriminate other nucleotide variants. For comparison, both a single short and long probes were also studied to show the advantages of our biosensor in assay performances. So this photocatalytic chemiluminescent biosensor via cooperative hybridization is an ideal method to assay miR-21, showing a good potential for cancer early diagnosis.

This experiment was conducted to explore the effects of betaine on the DNA methylation level, expression level, and fat synthesis ofVNN1in goose hepatocytes by isolating the primary hepatocytes of goose at the cellular level and constructing a fatty degeneration model of goose hepatocytes. In the study, 23-day-old Taizhou goose embryos were used as the research object, and free fatty acid (PA:OA = 2:1) was used to induce steatosis of goose primary hepatocytes. The experiment was randomly divided into seven groups: control group, steatosis model group, and betaine (2, 10, 25, 50, 100mM) group. After 24 h of cell culture, cell viability, oil red O staining, and lipid metabolism-related indicators in cell supernatant were measured, and cells were collected to determineVNN1,FAS,ACC,SCD,SREBPQgene expression andVNN1promoter region DNA methylation level. (1) The addition of 0.5 mM fatty acids successfully determined the degeneration of goose hepatocytes. The levels of TG and LDL were significantly increased (p< 0.05), and the level of HDL was significantly decreased. (2) The addition of 100 mM betaine significantly reduced TG levels, and 10, 25, 50, and 100 mM betaine significantly reduced LDL levels. The addition of betaine had no significant effect on HDL level compared to the FFA group (p> 0.05), although a significant overall model effect was observed. Oil red O staining showed that the area of lipid droplets in cells with 50 mM betaine decreased most significantly. (3) The expression levels ofVNN1,FAS,ACC,SCD, andSREBPQgenes in the fat model group were significantly higher compared to the control group (p< 0.05), and the DNA methylation level in the promoter region of theVNN1gene decreased. (4) The addition of 2, 10, 25, 50, and 100 mM betaine significantly reduced the expression ofVNN1. The expression ofFASin 2, 10, and 25 mM betaine groups significantly decreased. Betaine at 10 and 100 mM significantly reduced the expression ofSREBPQ, but it showed no significant effect onACCexpression. Addition of 2, 50, and 100 mM betaine led to an increased DNA methylation status at theVNN1gene promoter region. In summary, the addition of betaine can reduce the expression of fatty acid synthesizing genes such asFAS,SCD, andSREBPQ, down-regulate the expression level of theVNN1gene, increase the DNA methylation level of the VNN1 promoter region, and reduce lipid deposition in goose liver steatosis cells.

Surface-Enhanced Raman Spectroscopy (SERS) is a powerful vibrational spectroscopy technique capable of single-molecule sensitivity, yet its performance is critically dependent on the precise engineering of plasmonic hotspots. Conventional dimer-based substrates offer limited enhancement due to their restricted hotspot volume and intensity. Here, we overcome this limitation by leveraging the programmable addressability of DNA origami to construct a plasmonic nanofork antenna (DONA) that directs the assembly of 80 nm gold nanoparticles into well-defined multimers (trimers, tetramers, and pentamers). By systematically increasing the number of thiolated staple strands and refining the post-assembly conjugation workflow, we achieve reproducible sub-2 nm interparticle gaps, enabling dense and intense electromagnetic hotspots. This multimeric platform enables label-free SERS detection of Rhodamine 6G (R6G) down to an ultralow concentration of 10−14 M, with characteristic Raman peaks at 613 cm−1 and 1512 cm−1 clearly resolved. Furthermore, we demonstrate the versatility of this platform by detecting proteins (streptavidin, thioredoxin) at sub-picomolar levels. Our work establishes DNA-origami-engineered multimers as a robust and generalizable strategy for ultrasensitive, label-free biochemical sensing.

The widespread misuse of ciprofloxacin (CIP) poses a serious threat to environmental safety and human health, necessitating its trace detection in food matrices. In this study, a novel electrochemical aptasensor was developed for the highly sensitive detection of CIP. Au@ZnCoN-C nanocomposites with superior conductivity were synthesized and employed as the electrode modification material. To improve assay sensitivity, a target-triggered cyclic amplification method relying on RecJf exonuclease was adopted. Notably, a cross-linked DNA framework (CLDF) was utilized for the first time in CIP detection. Its interconnected architecture provides abundant active sites for high-density loading of methylene blue (MB), thereby significantly amplifying the electrochemical response. Under optimized conditions, the aptasensor exhibited a wide linear range from 0.005 to 1000 ng/mL, with a low limit of detection (LOD) of 3.2 pg/mL. Furthermore, the sensor was successfully applied to the analysis of CIP in milk, egg, and pork samples with satisfactory results. Leveraging its high specificity, long-term stability, and high sensitivity, this method provides a robust strategy for the precise monitoring of CIP in food safety applications.
