The glial scar forms a disadvantageous microenvironment for neurogenesis after stroke. Single treatment targeting glial scar suppression or neurogenesis promotion is insufficient for stroke repair. To improve the functional recovery post stroke, a synergistical therapy of reactive oxygen species (ROS)-scavenging polymers (hyperbranched poly(acrylate-capped thioketone-containing ethylene glycol, HBPAK) and chondroitin sulfate proteoglycans (CSPGs) receptor (protein tyrosine phosphatase σ, PTPσ) modulatory peptides (intracellular sigma peptide, ISP) was applied in a form of microgels for inflammation alleviation, glial scar inhibition and endogenous neuroblasts regulation. The microgels were prepared with a microfluidic device under UV irradiation. The microgels containing HBAPK displayed efficient elimination of typical ROS of 1-diphenyl-2-picryl hydrazyl radical (DPPH) and H 2 O 2 . The microgels containing HBAPK and ISP peptides decreased inflammation response of lipopolysaccharide (LPS) activated BV2 microglia. Promoted cell migration and proliferation of Schwann cells (SCs) and strengthened cell stemness and neuronal differentiation of neural stem cells (NSCs) were shown when these cells were incubated within the microgels containing HBAPK and ISP peptides. Injection of the microgels in vivo achieved positive therapeutic effects with respect to motor dysfunction recovery promotion, inflammation alleviation and cell apoptosis reduction at 7 d, and astrocytic scar inhibition, neuroblasts promotion and axonal neurofilaments increase at 28 d post stroke, suggesting the effectiveness of this synergistical therapy for stroke.

Reactive oxygen species (ROS) and subsequent inflammatory cascades hinder the healing of diabetic wounds, which should be tackled simultaneously when designing wound dressings. In this study, ROS-responsive di‑selenium-containing polyurethane nanofibers (PUF) loaded with manganese dioxide nanoparticles (MnO 2 NPs) and gelsevirine (GSV) with an average diameter of 0.6 ± 10 μm, were prepared to specifically target ROS and inflammation control, thereby enhancing healing in diabetic wounds. The resulting nanofibers exhibited a porous architecture and enhanced drug delivery capability. The MnO 2 /GSV/PUF dressing showed negligible cytotoxicity with cell viability over 80 %, effective ROS-scavenging ability of 87 %, and inhibition of pro-inflammatory cytokine expression. This dual mechanism of ROS-scavenging and inflammation modulation accelerated the healing of diabetic wounds up to 92 % in 14 d in a full-thickness diabetic wound model, as evidenced by reduced cytokine levels, enhanced epithelialization, and uniform collagen deposition with the highest percentage of 60 %.

A magnetic MXene aerogel (Fe 3 O 4 @MXene@PEI) was prepared by crosslinking amino modified MXene with polyethyleneimine using epichlorohydrin as a cross-linker. Adsorption properties of Fe 3 O 4 @MXene@PEI aerogel for phenolic acids were evaluated by adsorption kinetics and isotherms experiments, showing that the high adsorption affinity was governed by multilayer chemisorption process. An efficient MSPE/HPLC method was developed for the determination of phenolic acids with excellent selectivity, good linearity (0.025–5.0 μg mL −1 ), low LODs (0.007–0.017 μg mL −1 ), and satisfactory recoveries (80.0–120.0 %). Moreover, the antioxidant activity of the Fe 3 O 4 @MXene@PEI purified compounds was superior to that of the conventional method as demonstrated by the results of scavenging experiments on 2,2 -diphenyl-1-picrylhydrazyl radical scavenging assay. Finally, 65 organic acids were identified in the Fe 3 O 4 @MXene@PEI treated honeysuckle extracts by UHPLC-Q-Exactive Orbitrap MS/MS analysis. The proposed sorbent exhibits remarkable promise for the selective separation and purification of organic acids from herbal products.

Selective modulation of the types of reactive oxygen species (ROS) for the treatment of acute lung injury (ALI) has not been tackled, whereas the usually applied hydrophobic nanoparticles (NPs) are easily excreted and metabolized. Herein, a zwitterionic-segment containing polyurethane was synthesized to formulate into Pazopanib (Pazo)-loaded NPs (PUSB@Pazo NPs), which had the ability to selectively enhance the expression of H 2 O 2 but suppress other types of ROS, and achieved long retention in lung and thereby improved treatment efficacy. The zwitterionic group of PUSB effectively decreased the clearance rate of PUSB@Pazo NPs in vivo , and prolonged their resident time in lung post inhalation up to 48 h. The increase of local H 2 O 2 concentration decreased the permeability of lung epithelial cells, accompanied by the significantly reduced tissue edema and inflammatory cell recruitment. The system also reduced the expression of various inflammatory factors, revealing its effective and promising treatment for ALI.

Inflammatory bowel disease (IBD) compromises the intestinal barrier and disrupts gut microbiota, impacting liver function via the gut–liver axis, which in turn influences the intestinal microbiota through lipid metabolites exacerbating IBD. This study introduced a probiotic-based treatment using Lactobacillus acidophilus encapsulated in tungsten ion-loaded mesoporous polydopamine (LA@WMPDA) to ameliorate colitis and balance enterohepatic homeostasis. After oral administration, the encapsulation could protect Lactobacillus acidophilus, scavenge reactive oxygen/nitrogen species, and the released tungsten ions would inhibit abnormal Enterobacteriaceae growth during colitis, consequently restoring the intestinal barrier and regulating the gut microbiota. Nontargeted metabolomics and transcriptomics analyses showed increased short-chain fatty acids and indole derivatives, and decreased hepatic lipid metabolism. Pathways associated with immune response, cell migration and death, and response to bacterium showed significant down-regulation in the colon and liver transcriptome analysis. Thus, this study provided a pioneered paradigm for IBD treatment and highlighted the regulation of liver-related metabolic functions via the gut–liver axis.

Inflammatory bowel disease (IBD) is a recurrent chronic colitis disease with increasing incidence and prevalence year by year. The single efficacy and significant side effects of traditional IBD treatment drugs have promoted the flourishing development of new drugs. Inspired by many health benefits of carbon dots (CDs) based nanomedicine in biomedical applications, a metal-free carbon dots (CP-CDs) was synthesized from citric acid and polyethylene polyamine to treat colitis. Oxidative stress tests at the cellular and nematode levels demonstrated CP-CDs have good antioxidant effects, while the toxicity of CP-CDs to cells and nematodes is low. CP-CDs were further applied to dextran sodium sulfate (DSS)-induced colitis in mice models, and it was found that CP-CDs can reduce the disease activity index (DAI) score of colon tissue and restore the intestinal barrier. Further, the anti-colitis mechanisms of CP-CDs were explored, one of which is to regulate intestinal oxidative stress in inflammatory mice, further reducing the expression of inflammatory cytokines, and thus alleviating colitis. Notably, 16S rRNA sequence analysis showed that the abundance of beneficial bacteria ( Ligilactobacillus and Enterorhabdus ) in the intestinal tract increased, while that of harmful bacteria ( unclassified_Clostridia_UCG_014 ) decreased after CP-CDs treatment, indicating that CP-CDs rebalancing the gut microbiota destroyed by DSS is another important mechanism. In short, these non-toxic carbon dots not only have the potential for multi-factor combined relief of colitis but also offer an alternative therapy medicine for patients suffering from IBD.

In the development of drug delivery vehicles for full-thickness skin wounds, achieving a specific drug concentration at the active site remains a key challenge. In this work, we reported a novel protein-polysaccharide bio-based composite hydrogel (SG@CRG) with controlled continuous drug delivery for enhanced wound healing. This composite hydrogel was prepared by incorporating drug embedded spirulina protein isolate nanogels (SG) into carboxymethyl chitosan based hydrogel (CRG) network, which exhibited rapid gelation and excellent mechanical properties. Notably, the release rate of the embedded drug could be tuned by adjusting the proportion of SG within the composite hydrogels. In vitro experiments demonstrated that the composite hydrogel exhibited excellent antibacterial activity against both gram-positive and gram-negative bacteria (inhibition rates at 12 h for S. aureus and E. coli were 99.77 % and 98.64 %, respectively), and good biocompatibility. Additionally, the hydrogel could effectively alleviate excessive inflammation and oxidative stress damage. Lastly, a full-thickness mouse skin model was established to assess its wound healing performance in vivo, which found that the composite hydrogel markedly accelerated wound healing (the healing ratios achieved by day 7 and day 12 were 78.37 % and 99.13 %, respectively, compared to 56.99 % and 84.50 % in the control group) by diminishing inflammation, promoting granulation tissue regeneration, increasing collagen deposition, and accelerating angiogenesis. Altogether, this innovative protein-polysaccharide bio-based composite hydrogel provides new insights and technical support for the development of more efficient wound treatment method, showing its potential application as a wound healing drug delivery vehicle.

Background: The yellow-leafgl1mutant ofLagerstroemia indicaexhibits an altered phenylpropanoid metabolism pathway compared to wild-type (WT). However, details on the metabolites associated with leaf color variation, including color-specific metabolites with bioactive constituents, are not fully understood. Methods: Chemical and metabolomics approaches were used to compare metabolite composition and antioxidant capacity between thegl1mutant and WT leaves. Results: The mutant exhibited an irregular xylem structure with a significantly lower phenolic polymer lignin content and higher soluble phenolic compounds. Untargeted metabolomics analysis identified phenolic compounds, particularly lignans, as key differential metabolites betweengl1and WT, with a significant increase in the mutant. The neolignan derivative balanophonin-4-O-D-glu was identified as a characteristic metabolite in thegl1mutant. The soluble phenolic compounds of thegl1mutant exhibited higher FRAP, ABTS, DPPH, and hydroxyl radical scavenging activity than in WT. Correlation analysis showed a positive relationship between antioxidant capacity and phenolic compounds inL. indica. Conclusions: Metabolites associated with leaf color variation in theL. indicayellow-leafgl1mutant demonstrated high antioxidant capacity, particularly in scavenging hydroxyl radicals.

In this study, pectin (PE) composite films containing tea polyphenol (TP) and graphene oxide (GO) were developed. The effects of TP and GO on the appearance, structure, mechanical properties, barrier properties, hydrophobicity, and antioxidant properties of pure PE films were investigated. The results demonstrated that the addition of 1 % w /w TP and 1 % w/w GO increased the tensile strength (TS) and elongation at break (EB) of the composite films to 20.22 MPa and 34.18 %, respectively. Additionally, the water vapor permeability (WVP) was reduced to 0.67 ± 0.17 (×10 −10 g −1 s −1 Pa −1 ), and both the moisture content and water contact angle were significantly decreased. Furthermore, the incorporation of TP enhanced the antioxidant properties of the composite films and demonstrated the slow-release capability of TP. Blueberry fruits packaged in PE-TP-GO film maintained an optimal appearance after eight days of storage at 25 °C, with a reduction in shrinkage index of approximately 29.4 % compared to those packaged in PE film.

Litsea cubeba essential oil (LCEO) has garnered widespread attention due to its robust biological activity. However, challenges such as high volatility, limited water solubility, and low bioavailability impede its application. Nano-emulsion encapsulation technology offers an effective solution to these issues. In this study, we prepared litsea cubeba essential oil nano-emulsion (LCEO-NE) for the first time using whey protein (WP) as the emulsifier through an ultrasonic-assisted method, achieving high efficiency with minimal energy consumption. Transmission electron microscopy and dynamic light scattering analyses revealed that the nanoparticles were uniformly spherical, with a particle size of 183.5 ± 1.19 nm and a zeta potential of −35.5 ± 0.95 mV. Stability studies revealed that LCEO-NE exhibited excellent thermal and salt stability, maintaining its integrity for up to four weeks when stored at 4 °C and 25 °C. In vitro digestion assays confirmed the digestibility of LCEO-NE. Furthermore, evaluation of the DPPH, ABTS, and antimicrobial activities revealed that LCEO-NE displayed superior bacteriostatic and antioxidant properties compared to LCEO. Scanning electron microscopy elucidated that its bacteriostatic effect involved the disruption of bacterial microstructure. Hemocompatibility and cytotoxicity assays demonstrated the safety of LCEO-NE within the effective concentration range. This research supports the utilization of nanoparticles for encapsulating LCEO, thereby enhancing its stability and bioactivity, and consequently expanding its applications in the food and pharmaceutical industries.

Scallops are renowned for their delicious taste and rich nutritional profile, including high contents of eicosapentaenoic acid and docosahexaenoic acid. However, they are highly susceptible to spoilage caused by lipid oxidation and microorganism proliferation. Therefore, in this study, trilayer films based on gelatin/gelatin-dextran/gelatin with different concentrations (0%, 0.3%, 0.6%, 0.9%, and 1.2%, w/w) of active fillers (citronellal and α-tocopherol) were developed to preserve scallop adductor muscle during cold storage. Scanning electron microscopy images showed that each layer was well-bonded with no gaps, indicating the successful preparation of the trilayer films. The addition of citronellal and α-tocopherol resulted in a porous cross-section with a slightly rough surface and increased hydrophobicity of the trilayer films. With increase in concentrations of citronellal and α-tocopherol, the thickness, opacity, antioxidant capability, and antibacterial activity of the trilayer films increased, while water vapor permeability and mechanical properties decreased. Both citronellal and α-tocopherol showed controlled release properties in four food simulants, with the longest release times in 95% (v/v) ethanol solution. Furthermore, the incorporation of 1.2% (w/w) of active fillers into the trilayer film significantly enhanced the chemical and microbiological stabilities of the scallop adductor muscles, thereby prolonging their shelf life by six days during cold storage. The developed trilayer films showed excellent controlled release, antibacterial properties, and antioxidant capabilities, indicating their potential for effective preservation of aquatic food products.

Vitamin C (VC) possesses antioxidant and whitening effects. However, its effectiveness is hindered by challenges such as instability, impaired solubility, and limited bioavailability hinder. In this study, VC was encapsulated in nanoliposomes by primary emulsification and high-pressure homogenization. The VC nanoliposomes were comprehensively characterized for their microscopic morphology, particle size, polydispersity index (PDI), and encapsulation efficiency (EE). Orthogonal experiments were designed to optimize the optimal preparation process, and the antioxidant activity, whitening efficacy, transdermal absorption, and stability of VC nanoliposomes were evaluated based on this optimized process. The findings demonstrated the high reproducibility of the optimal process, with particle size, PDI, and EE values of 113.502 ± 4.360 nm, 0.104 ± 0.010, and 56.09 ± 1.01 %, respectively. Differential scanning calorimetry analysis showed effective encapsulation of VC nanoliposomes with better thermal stability than aqueous VC solution. Besides, the VC nanoliposomes demonstrated excellent antioxidant and whitening effects in efficacy experiments, stronger skin permeability in transdermal experiments and fluorescence tracking. Furthermore, storage stability tests indicated that the VC in nanoliposomes remained relatively stable after 60 days of storage. These findings highlighted the potential use of VC nanoliposomes in a wide range of applications for the cosmetic market, especially in the development of ingredients for skin care products.
