Pullulan nanofibrous films incorporated with W/O emulsions via microfluidic solution blow spinning technology

Liying Huang,Di Zhang,Nitong Bu,Yuanbo Zhong,Pingping Tan,Huanglong Lin,Jie Pang,Ruojun Mu

In this work, pullulan (PUL) nanofibrous films incorporated with water-in-oil emulsions (PE) were prepared by microfluidic blowing spinning (MBS). The microstructures of nanofibers were characterized by scanning electron microscopy (SEM), fourier transform infrared (FT-IR), and X-ray diffraction (XRD). With the addition of W/O emulsions, the thermal stability, mechanical, and water barrier properties of PUL nanofibers were improved. Increases in emulsion content significantly affected the antioxidant and antimicrobial properties of nanofibrous films. ABTS and DPPH free radical scavenging rates increased from 10.26 % and 8.57 % to 60.66 % and 57.54 %, respectively. The inhibition zone of PE nanofibers against E. coli and S. aureus increased from 11.00 to 20.00 and from 15.67 to 21.17 mm, respectively. In addition, we investigated the freshness effectiveness of PE nanofibrous films on fresh-cut apples. PE nanofibrous films significantly maintained the firmness, and reduced the weight loss and browning index of the fresh-cut apple, throughout the 4 days of storage. Thus, the PE nanofibrous films exhibited good potential to prolong the shelf life of fresh-cut fruit and promote the development of active food packaging.

Purification, characterization, and antioxidant ability of polysaccharides from Phascolosoma esculentas

Fengfang Zhou,Binxin Cai,Shaojiang Ruan,Qi Wei

The polysaccharide was extracted from Phascolosoma esculenta (PEP). Two purified polysaccharides (PEP-1 and PEP-2) were obtained by the column chromatography separation method. The molecular weights of PEP-1 and PEP-2 were 33.6 and 5.7 × 10 3 kDa, respectively. PEP-1 and PEP-2 had the same monosaccharides composition, but their molar ratios varied. The in vitro antioxidant activity of the PEP, PEP-1, and PEP-2 were investigated by scavenging free radicals like 3-ethylbenzoth-iazoline-6-sulfonic acid (ABTS), •OH, and 2,2-diphenyl-1-picrylhydrazyl (DPPH). Additionally, the in vivo antioxidant activity of PEP-1 was examined using the Caenorhabditis elegans ( C. elegans ) organism. Results showed that PEP-1 was much more effective than PEP and PEP-2 at scavenging DPPH, •OH, and ABTS radicals. Additionally, PEP-1 strengthened C. elegans' ability to endure oxidative stress. PEP-1 possessed the in vivo antioxidant capacity, including the reactive oxygen species (ROS) content reducing, and protective effect on antioxidant enzyme activities in C. elegans . In summary, PEP, PEP-1, and PEP-2 might have the potential to develop as functional foods and clinical medications.

Quality attributes of black tea-flavored Chardonnay wine processed by ultrasound

Liang Zijian,Zhang Pangzhen,Ma Wen,Zeng Xin-An,Fang Zhongxiang

An innovative Chardonnay wine flavored with black tea was prepared under optimal ultrasound conditions at 450.0 W for 22.6 min, and its physicochemical features, antioxidant power, total phenolic content, and volatile composition were systematically examined. A total of 165 phenolic and non-phenolic substances were characterized, including 27 newly formed phenolic substances and 10 non-phenolic substances that were not found in both black tea and Chardonnay wine. Catechin was found to be the phenolic compound with the largest amount detected, which was responsible for the potentiated antioxidant activities. The wine exhibited a profile of 44 volatile compounds, with 13 volatiles as odor-active compounds. Black tea infusion resulted in a decline in fruity, fermented and spicy aromas, but an increase in green and citrus odors, while ultrasound processing further intensified these odors. This study anticipated that ultrasound technology could hold promise for crafting flavored wines with heightened functionalities and appealing flavor profiles. Graphical Abstract Ultrasound-assisted infusion of black tea potentiated the antioxidant activity and enriched the volatile and phenolic profiles of Chardonnay wine.

Menglu Li,Ruotong He,Ceyao Yang,Hao Chen,Chaoling Wu,Xiaomiao Zhang,Xin Fu,Chengbo Lu,Liu Yang,Shi Liu,Yi Zhang

Lilium brownii F.E.Brown var. viridulum Baker (Longya Baihe), a traditional medicinal and edible plant. In order to study the activity of its polysaccharide extracts, a three-factor three-level Box-Behnken response surface design was employed to optimize the extraction process. The optimal extraction conditions include: liquid-solid ratio of 32:1 mL/g, extraction temperature of 70 °C, and extraction time of 3.5 h. The purified Longya Baihe polysaccharide (LP), obtained via DEAE-52 cellulose chromatography, was structurally characterized by molecular weight distribution, monosaccharide composition and infrared spectrum. The results show that LP was a macromolecular polysaccharide composed of glucose and mannose (molar ratio of 1:1.56), with an average molecular mass (Mn) of 24.006 kDa. In vitro assays demonstrated its excellent antioxidant and anti-inflammatory activities. LP significantly inhibited lipopolysaccharide (LPS)-induced activation of the nuclear factor kappa-B (NF-κB) pathway in RAW264.7 cells, downregulated the overexpression of nitric oxide (NO), interleukin (IL)-6, inducible nitric oxide synthase (iNOS) and tumor necrosis factor (TNF)-α, and enhancing the levels of the anti-inflammatory factor interleukin (IL)-10. This study suggests LP holds promise as a natural antioxidant and anti-inflammatory agent for potential development and application.

Wang Hongji,Li Xiaoyu,Cui Liying,Guo LuLu,Wang Aijiao,Chu Guangru,He Siqi,Liu Shengkai,Fu Yujie,Liu Zhiguo

Vaccinium vitis-idaea L. leaves (VVLL), a renewable forest resource abundant in active compounds, have not been well utilized. This study employed microwave-assisted natural deep eutectic solvents (NADES) to efficiently extract the total flavonoids from VVLL. Among various combinations, the choline chloride-lactic acid solvent exhibited great selectivity for flavonoids, with the highest extraction rate of 269.99 ± 1.42 mg/g, ( n  = 3). Six flavonoids in the VVLL extract were further analyzed by UPLC-MS/MS and HPLC. They included two isomers, a triterpenic acid, and Asiatic acid which is firstly reported in VVLL. The total flavonoid content extracted by our method is 3.4 times higher than that of Vaccinium vitis-idaea L. fruit. Fourier-transform infrared spectroscopy results revealed NADES-flavonoid hydrogen bonding as the driving mechanism for enhanced extractability. The extracted flavonoids exhibited superior antioxidant capacity against DPPH and ABTS radicals in comparison to the traditional alcohol extraction methods. Specifically, the scavenging activities were also significantly improved. In conclusion, as a high-quality source of flavonoids, VVLL has substantial promise for the development of novel flavonoid products.

Preparation of Multifunctional Hydrogel with Curcumin-Controlled Release and Photothermal Synergistic Antibacterial Property for Wound Dressing

Yufei Ling,Yuxuan Wang,Yuanpeng Cai,Shizhen Zhao,Hui Liu

Considering the dynamic changes in the wound microenvironment and the risk of bacterial infection during healing, the synergistic design of antibacterial hydrogel dressings has become an effective treatment strategy. A multifunctional hydrogel Cur-F127/CMCs/PDA was prepared through the cross-linking of carboxymethyl chitosan (CMCs) with genipin (GP), followed by the loading of curcumin-F127 (Cur-F127) micelles and polydopamine (PDA) nanoparticles. Successful synthesis of the hydrogel was confirmed by Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), and X-ray photoelectron spectroscopy (XPS). Results showed that the introduction of PDA endowed the hydrogel with excellent photothermal capability, achieving a temperature increase to 54.3 °C within 10 min under near-infrared (NIR) irradiation. Meanwhile, rapid Cur release was observed under NIR irradiation, exhibiting distinct “on/off” switching characteristics. The drug-controlled releasing behavior of the hydrogel was evaluated in physiological environments, and relatively high cumulative Cur release was obtained under simulated wound microenvironmental conditions (acid region and high temperature) due to the shrinking of polymer chains and micelle structure. The synergistic effect of Cur release and PDA photothermal therapy enabled the hydrogel to exhibit excellent antibacterial effects, and the survival rates of E. coli and S. aureus were reduced to 6.25% and 1.31%, respectively. Furthermore, in vitro biological studies demonstrated that the hydrogel possessed a good antioxidant property, hemostatic performance, blood compatibility, cell compatibility, and cell migration ability. In summary, the multifunctional wound dressing Cur-F127/CMCs/PDA provided a synergistic antibacterial strategy combining natural active compound release with photothermal effects, which accelerated the wound-healing process and holds significant clinical translational potential.

Probiotics Behavior-Inspired Biomimetic Platform for Ulcerative Colitis Prevention and Therapy

Mengjun Wang,Linyu Nian,Shuo Zhang,Yan Zeng,Wei Man,Xiaoyang Sun,Suilou Wang,Chongjiang Cao

Despite significant advancements in probiotics therapy for ulcerative colitis, its unsatisfactory efficacy and safety concerns impede further clinical applications. Herein, inspired by the natural secretion of outer membrane vesicles (OMVs) and colon colonization behavior of probiotics, a biomimetic probiotics platform (OSM) using bicontinuous single primitive metal–phenolic networks (SP-MPNs) loaded with OMVs derived from E. coli Nissle 1917 (EcN) is developed. Notably, the ordered mesopores, high specific surface area, and excellent antioxidant properties of SP-MPNs demonstrate a high loading capacity (≈30%, w / w ) and protective effects for OMVs. Impressively, OSM not only responds to transferrin (Tf) for the slow release of OMVs but also strongly interacts with mucin in the mucus layers, prolonging the retention time of OMVs in the colon (>48 h). Furthermore, OSM exhibits robust antioxidant, anti-inflammatory, and immunoregulatory activities. Significantly, in comparison with the probiotic EcN, OSM demonstrates superior adhesion properties, higher OMVs release amount, and eliminates the risk of inducing bacteremia. To protect OSM from gastric acid degradation, OSM is encapsulated into ascorbyl palmitate (AP) to form OMVs@SP-MPNs@AP (OSMA) hydrogel, which demonstrates preventive and therapeutic efficacy in colitis mouse models. In summary, the biomimetic platform provides a promising approach for the prevention and treatment of gastrointestinal disorders.

Zhongmei Tan,Mengguo Wu,Bailiang Li,Zhanmei Jiang,Jiage Ma,Juncai Hou

To achieve the positive effects of probiotics, it is crucial to enhance their viability and stability throughout gastrointestinal tract. In the study, polyvinyl alcohol-fucoidan@ethyl cellulose (PVOH-FUC@EC) core-shell electrospun nanofibers with flow ratios of 1:1, 1:1.5 and 1:2 by coaxial electrospinning were developed to co-encapsulate Lactiplantibacillus plantarum 69-2 (LP69-2) and dihydromyricetin. The average diameter of PVOH-FUC@EC nanofibers decreased, while the shell thickness of PVOH-FUC@EC nanofibers increased as the shell solution flow ratios increased. Furthermore, the successful formation of core-shell structure of PVOH-FUC@EC nanofibers was verified by confocal laser scanning microscopy and transmission electron microscope. PVOH-FUC@EC nanofibers with higher shell flow rate presented superior thermal stability. Compared with uniaxial nanofibers, the antioxidant capacity of PVOH-FUC@EC core-shell nanofibers was significantly enhanced when core-shell flow ratio was 1:1 ( P  < 0.05). Moreover, the survival rate of LP69-2 embedded in PVOH-FUC@EC core-shell nanofibers was significantly elevated after simulated gastrointestinal digestion ( P  < 0.05). Overall, this study further revealed that EC-coated core-shell nanofibers was an effective and novel approach for co -delivering probiotics and polyphenols by coaxial electrospinning, thereby improving their efficacy in functional foods.

Luteolin Stabilized in Nanosheet and Cubic γ-Cyclodextrin-Based Metal Organic Framework for Enhanced Bioavailability and Anti-inflammatory Therapy

Mubarak G. Bello,Shiyu Huang,Zifan Qiao,Zhenhua Chen,Lihua Chen

Luteolin (LT), a biologically active flavonoid with potent anti-inflammatory properties, faces significant limitations due to its instability, poor absorption, and low bioavailability. To overcome these challenges, this study explores the encapsulation of LT within nanosheet and cubic γ-cyclodextrin-based metal-organic frameworks (N-MOF and CD-MOF). This approach achieved a two-fold improvement in LT loading efficiency compared to previously reported methods. A critical finding was the identification of pH as a key factor in optimizing the loading process, which significantly enhanced encapsulation efficiency. The resulting LT/N-MOF and LT/CD-MOF formulations demonstrated improved photo-stability and thermal stability, alongside significantly enhanced cellular permeation. In vitro studies revealed remarkable anti-inflammatory properties of these formulations. Moreover, the bioavailability of LT was increased by 6-fold compared to pure luteolin. In a rat paw edema model, the optimized LT/CD-MOF and LT/N-MOF formulations exhibited 4.86- and 5.4-fold improvements in anti-inflammatory activity, respectively, over pure luteolin. These results underscore the potential of CD-MOFs as effective delivery systems for enhancing the therapeutic efficacy of luteolin.

Preparation, Characterization of Amides from Piper wallichii (Miq.) Hand.-Mazz. and Key Compounds including Pellitorine Screened by Fingerprint-efficacy Correlation of Antioxidant Activity

Dezhi Shi,Jing Ji,Yue Wang,Lirong Lu,Shaohui Zhong,Zhifeng Tang,Yunyu Zhang,Wufeng Gao,Meng Cao,Yang Hu,Yunfeng Zheng,Shiwen Huang,Jianming Cheng

The industrial enrichment of fat-soluble compounds is a common challenge, which is also present in Piper wallichii (Miq.) Hand.-Mazz. (PWH). The stem of PWH is rich in amide alkaloid substances, although enrichment methods have been available for total amide alkaloids, their water-insoluble character has not yet been addressed by a green and efficient method. We have elaborated the adsorption characteristics of pellitorine and futoamide on macroporous resins. Synchronization of concentration and adsorption was used to solve the critical problem of enrichment on macroporous resins with lipid-soluble compounds that tend to clog the resin columns during the enrichment process. Fifteen batches of PSA (amide alkaloids of PWH) were prepared from PWH collected from multiple origins, and multidimensional fingerprints were established in combination with multivariate statistical analysis to evaluate the consistency and variability of the compounds in multiple batches. Then, the fingerprints were correlated with their corresponding antioxidant activities to identify the pharmacodynamic compounds, thus clarifying the key compounds. Finally, compound separation was performed to verify the accuracy of the antioxidant activity of the key compounds and fingerprint-efficacy correlation model. HP-20 macroporous adsorbent resin has optimal adsorption and desorption which was selected for the enrichment of PSA. The adsorption of PSA, including futoamide and pellitorine, on the HP-20 resin was consistent with the Pseudo-second order kinetic model and Langmuir isotherm model, and this adsorption was a spontaneous exothermic entropy increase mechanism. The on-column adsorption process was optimized to solve the phenomenon that the precipitation of fat-soluble amides were easy to block the glass column during the enrichment process of the macroporous adsorption resin, and the effective enrichment of PSA was achieved. The content of pellitorine and futoamide was 13.95 g/100g and 3.11 g/100g in prepared samples respectively. The percentage of PSA was more than 50 g/100g. Seventeen differentiated compounds were screened by multivariate statistical analysis, antioxidant activity of 4 individual compounds isolated by fingerprint-efficacy correlation such as pellitorine were validated. The present study provided a more effective method for the enrichment of PSA, and initially elucidated the active compounds of the antioxidant activity of PWH, which will establish the basis for the industrialization of PSA in the future.

Chitosan/γ-polyglutamic acid-based Pickering emulsion gel: A promising transdermal carrier for crocetin

Yicheng Yang,Qinan Huang,Lipei Miao,Qihong Zhang,Chongyang Wu,Weike Su,Jingbo Yu

Crocetin (CT), a natural carotenoid dicarboxylic acid known for its high medicinal value, faces challenges such as poor water solubility, low bioavailability and instability, limiting its therapeutic applications. To overcome these challenges, this study introduces a novel Pickering emulsion gel stabilized by naturally sourced chitosan (CS) and γ -polyglutamic acid ( γ -PGA) nanoparticles (CS/ γ -PGA NPs) as a potential transdermal carrier for CT. The CS/ γ -PGA NPs were synthesized using an electrostatic crosslinking method pretreated by mechanical ball-milling, producing uniform and stable nanoparticles. These NPs stabilized a CT-loaded Pickering emulsion gel (CT-PE) with cornmint oil as the oil phase, achieving up to 98.53 % encapsulation efficiency. The CT-PE exhibited enhanced rheological properties, storage stability, sustained and targeted in vitro transdermal release, potent antioxidant activity, and effective antimicrobial effects with no cytotoxicity. Microstructural analysis revealed a dense network of aggregated particles forming a moderately strong gel structure, wherein encapsulated CT enhances emulsion stability through interaction with the gel network. The synergy between CS and γ -PGA facilitated a biocompatible and efficient carrier system, markedly boosting CT's therapeutic effectiveness. These results highlight CT-PE's potential as an effective transdermal carrier for CT, promising for future dermatological treatments and drug delivery systems.

Effect and mechanism of polysaccharide from Crataegus pinnatifida on nonalcoholic fatty liver disease

Hanju Fan,Meng Yan,Yongshuai Jing,Ziyan Li,Yaqian He,Zhiwei Li,Junna Song,Lanfang Wu

Crataegus pinnatifida is the dried fruit of Crataegus pinnatifida . Crataegus pinnatifida polysaccharides (CPPs) eluted with water as the mobile phase (CPPs-H-1) is one of the important active components. CPPs-H-1 has anti-oxidation and blood lipid regulation activities in vitro. Therefore, we investigated the effects of CPPs-H-1 on nonalcoholic fatty liver disease (NAFLD) in C57/BL6J mice. As a result, CPPs-H-1 inhibited the rapid growth of body weight and alleviated the liver injury caused by high-fat diets, significantly reducing the liver index and blood lipid levels in the model group, inhibiting the production of inflammation and cytokines, and inhibiting oxidative stress. CPPs-H-1 also enhanced intestinal barrier function. At the phylum level, CPPs-H-1 reduced the ratio of Firmicutes to Bacteroides (F/B) and regulated the content of short-chain fatty acids (SCFAs). The CPPs-H-1 may regulate intestinal flora composition, and improve intestinal barrier function, ultimately delaying the progression of NAFLD in mice. Also, CPPs-H-1 improved NAFLD progression by enhancing the Amp-activated protein kinase (AMPK)/peroxisome proliferator-activated receptor α (PPARα) signaling pathway to enhance fatty acid oxidation. All these results confirm that CPPs-H-1 plays a multi-pathway and multi-target role in the improvement of NAFLD.
