Using Gd-Enhanced β-NaYF4:Yb,Er Fluorescent Nanorods Coupled to Reduced TiO2 for the NIR-Triggered Photocatalytic Inactivation of Escherichia coli

Huang Zhou,Fengjiao He

β-NaYF4:Yb,Er,Gd fluorescent nanorods were successfully coupled to a reduced TiO2(UCNPs@R-TiO2) nanocomposite and applied to visible-light catalytic sterilization under 980 nm near-infrared (NIR) light illumination. The UCNPs (β-NaYF4:Yb,Er,Gd) absorb the NIR light and emit red and green light. The visible light can be absorbed by the R-TiO2(Eg = 2.8 eV) for the photocatalytic reaction. About 98.1% ofEscherichia coliwere effectively killed upon 12 min of NIR light irradiation at a minimum inhibitory concentration (MIC) of 40 μg/mL UCNPs@R-TiO2nanocomposite. The bactericidal properties were further evaluated by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis. We found that the high bactericidal activity was due to the synergistic effect between the UCNPs and R-TiO2. Moreover, the UCNPs show excellent upconversion luminance properties, and the introduction of visible-light-absorbed R-TiO2nanoparticles (2.8 eV) was conducive to the efficient separation and utilization of photogenerated electron-hole pairs.

S-nitrosoglutathione functionalized polydopamine nanoparticles incorporated into chitosan/gelatin hydrogel films with NIR-controlled photothermal/NO-releasing therapy for enhanced wound healing

Wenyu Wang,Huan Sheng,Daihong Cao,Fenglian Zhang,Weifen Zhang,Fang Yan,Dejun Ding,Ni Cheng

Nitric oxide (NO) has aroused wide interest in the treating infected wounds due to its characteristic functionalities. However, its utilization is limited due to its volatile properties, high reactivity, direct potential toxicity, and byproducts of NO donors limited its application. Herein, endogenously NO donor S-nitrosoglutathione (GSNO) was connected covalently to polydopamine nanoparticles (PDA-GSNO NPs) to minimize the loss of NO in aqueous medium. Meanwhile, near-infrared (NIR)-controlled NO release and photothermal therapy (PTT) was obtained through the photothermal conversion by PDA. Then chitosan (CS)/gelatin (GE) biocomposite hydrogel films with preferable biocompatibility, surface hydrophilicity, hydroabsorptivity, and mechanical adhesive properties were constructed. By embedding PDA-GSNO NPs into the films, a multifunctional wound dressing was fabricated. Under NIR light irradiation, the combination of PTT, NO-releasing, and CS antibacterial agents can strengthen the in vitro antimicrobial efficacy and in vivo wound healing activities. Meanwhile, the obtained wound dressing presented good biocompatibility. This work outlines an approach for combating bacterial infections and demonstrating the possibility for synergistic NO-releasing wound healing.

NIR regulated upconversion nanoparticles@metal-organic framework composite hydrogel dressing with catalase-like performance and enhanced antibacterial efficacy for accelerating wound healing

Yu Chen,Danqi Li,Yaping Zhong,Zhentan Lu,Dong Wang

Developing a hydrogel dressing with excellent antibacterial efficacy for accelerating wound healing is high desirable in clinical applications. In this work, NIR regulated metal-organic framework composite hydrogel dressing was constructed for enhanced antibacterial efficacy and accelerated wound healing via the compounding between hydrogel and UCNPs@ZrMOF-Pt nanoparticles. The visible light emitted from upconvertion nanoparticles (UCNPs) activated porphyrin based metal-organic framework (MOF) in composite hydrogel to generate 1 O 2 for photodynamic antibacterial therapy under NIR laser irradiation. Moreover, the UCNPs@ZrMOF-Pt in composite hydrogel with catalase-like performance could effectively convert the high concentration H 2 O 2 in wound to abundant O 2 , which relieved the hypoxic in infected wound. Thus, the photodynamic antibacterial efficacy was remarkably enhanced, leading to accelerate the wound healing. This work presented a novel strategy for high efficient antibacterial therapy and accelerated wound healing.

Multifunctional nanomicelles constructed via an aggregation and de-aggregation strategy for magnetic resonance/NIR II fluorescence imaging-guided type I photodynamic therapy

Lirong Wang,Ji Qi,Ke Zhang,Zeyan Zhuang,Keke Ding,Xu Chen,Hong Shan,Dan Ding,Anjun Qin,Ben Zhong Tang

Fluorescence and magnetic resonance imaging (FL/MRI) has received much attention due to their complementary characteristics. However, the simultaneous enhancement of fluorescence and MR signals plus the efficacy of the treatment is still a major challenge. To tackle this difficulty, we put forward a strategy of aggregation and de-aggregation based on luminogens with aggregation-induced emission (AIE) features. NIR II photosensitizer 4,4′-(6,7-diphenyl-[1,2,5]thiadiazolo[3,4-g]quinoxaline-4,9-diyl)bis(N,N-diphenylaniline) (TQ-TPA) with type I reactive oxygen species (ROS) generation ability and amphiphilic 2TPE-Gd were synthesized, and both of them showed the AIE feature. During the construction of nanomicelles, hydrophobic TQ-TPA spontaneously aggregated into the core of the nanomicelles formed by 1,2-distearoyl-sn-glycerol-3-phosphoethanolamine-N-methoxy(polyethylene glycol)-2000 (DSPE-PEG). Meanwhile, aggregates of 2TPE (tetraphenylethylene)-Gd in aqueous solution could be de-aggregated and inserted into the interface, and formed TGdTT nanomicelles (NMs). Based on this strategy and the AIE feature, the TGdTT NMs exhibited strong NIR II fluorescence and type I ROS generation ability, and enhanced T1 relaxivity (r1). Moreover, in vitro, in vivo, and pharmacokinetics results demonstrated that these nanomicelles have good biosafety and a long blood circulation time. Finally, they allowed successful realization of complementary MR/NIR II fluorescence dual-modal imaging-guided photodynamic therapy (PDT) to inhibit tumor growth. This work demonstrated that the aggregation and de-aggregation strategy of AIEgens in core–shell nanomicelles is very attractive for constructing multifunctional theranostic probes.

NIR-II Fluorescence and PA Imaging Guided Activation of STING Pathway in Photothermal Therapy for Boosting Cancer Immunotherapy by Theranostic Thermosensitive Liposomes

Qi Long,Yuliang Yang,Fangling Liao,Haoting Chen,Dongyue He,Shengliang Li,Li Pengcheng,Weisheng Guo,Yafang Xiao

Photothermal immunotherapy has shown great potential for efficient cancer treatment. However, the immunosuppressive tumor microenvironment forms a heavy barrier for photothermal-induced anti-tumor immunity by inhibiting dendritic cells (DCs) maturation and cytotoxic T cell response. Moreover, the lack of reliable spatiotemporal imaging modalities makes photothermal immunotherapy difficult to guide tumor ablation and monitor the therapeutic outcomes in real time. Herein, we designed a theranostic thermosensitive liposome (PLDD) as a versatile nanoplatform to boost adaptive anti-tumor immunity of photothermal immunotherapy and achieve multiple bioimaging modalities in a real-time manner. The PLDD contains two major functional components: a multifunctional photothermal agent (DTTB) and an immune potentiator STING pathway agonist (DMXAA). Upon irradiation, the heat generated by DTTB induced the immunogenic cell death (ICD) of the tumor and dissociated the structure of thermosensitive liposome to release DMXAA, which ultimately activated the STING pathway and promoted the ICD-induced immune response by increasing DC cells maturation and T cell recruitment. Meanwhile, the DTTB in PLDD displayed excellent second near-infrared (NIR-II) fluorescence and photoacoustic (PA) dual-modal imaging, which provided omnibearing information on the tumor and guided the subsequent therapeutic operation. Therefore, this versatile PLDD with light-triggered promotion of anti-tumor immunity and multiple spatiotemporal imaging profiles holds great potential for the future development of cancer immunotherapy.

Doping Ag2S quantum dots with Pb yields significantly enhanced in vivo fluorescence imaging in the NIR-II window and comparable toxic effects

Qingyuan Cheng,Liman Li,Mingxia Yu

Silver chalcogenide (Ag2X; X = S, Se, Te) quantum dots (QDs) have facilitated the biomedical applications of QDs due to their low toxicity and tunable emission reaching the second near-infrared window (NIR-II window, 1000–1700 nm). The photoluminescence intensity of Ag2S QDs and Ag2Se QDs is considerably enhanced via Pb doping. However, the developed Pb-doped Ag2S QDs lacked in vivo verification, and the toxicity of the Pb-doped Ag2X QDs was only preliminarily assessed. Although QDs containing Pb have been widely introduced into the field of biomedicine, there are still concerns regarding toxicity associated with Pb. In the present study, we prepared polyethylene glycol (PEG)-modified Pb-doped Ag2S QDs with better biocompatibility and tested their in vivo bioimaging and toxicity in comparison with Ag2S QDs modified with the same PEG under identical concentrations. Results indicated that the PEG-modified Pb-doped Ag2S QDs significantly enhanced the brightness of in vivo bioimaging and spatial resolution for vessels inside the tumor without increasing toxicity, assessed based on cell viability post incubation for 96 h, intracellular ROS level, DNA damage, early apoptosis and mitochondrial injury. Our study demonstrated that the PEG-modified Pb-doped Ag2S QDs could serve as an advanced fluorescent probe for various biomedical applications.

Can magnetic β-NaYF4/g-C3N4 heterojunction with highly and stable NIR enhanced photocatalytic activity be synthesized by one-step solvothermal process

Chen Zhangxu,Zhu Danchen,Zhang Lidan

To overcome the intrinsic disadvantages of pristine g-C 3 N 4 and make it reusable, improve the activity of g-C 3 N 4 by making use of the near-infrared region, the magnetic Fe 3 O 4 /β-NaYF 4 /g-C 3 N 4 (FO/UC/CN) heterogeneous materials with high and stable NIR enhanced photocatalytic activity were prepared via a new facile one-step solvothermal synthesis method. It can be revealed that FO/UC/CN can activate UV–visible light under excitation of 980 nm. Compared with pure Fe 3 O 4 , g-C 3 N 4 and Fe 3 O 4 /g-C 3 N 4 , the obtained FO/UC/CN15 heterojunction exhibits good photodegradation rates of 65.56, 21.35, 59.22 and 20.49% for MB, MO, RhB and phenol, respectively. In addition, after four cycles of the photocatalytic degradation test, the photodegradation rates remain 62.14, 20.54, 57.64 and 19.36%, respectively. The results illustrate magnetic FO/UC/CN heterojunction exhibits highly NIR-enhanced photocatalytic activity and excellent stability. This study indicates a promising system to synthesize g-C 3 N 4 -based magnetic photocatalysis for effectively utilizing the NIR and treating industrial wastewater, which can be expected to alleviate the energy crisis and enhance the application of solar energy for dealing with environmental remediation in the future.

A H2O2-activatable nanoprobe for diagnosing interstitial cystitis and liver ischemia-reperfusion injury via multispectral optoacoustic tomography and NIR-II fluorescent imaging

Chen Junjie,Chen Longqi,Wu Yinglong,Fang Yichang,Zeng Fang,Wu Shuizhu,Zhao Yanli

Developing high-quality NIR-II fluorophores (emission in 1000–1700 nm) for in vivo imaging is of great significance. Benzothiadiazole-core fluorophores are an important class of NIR-II dyes, yet ongoing limitations such as aggregation-caused quenching in aqueous milieu and non-activatable response are still major obstacles for their biological applications. Here, we devise an activatable nanoprobe to address these limitations. A molecular probe named BTPE-NO 2 is synthesized by linking a benzothiadiazole core with two tetraphenylene groups serving as hydrophobic molecular rotors, followed by incorporating two nitrophenyloxoacetamide units at both ends of the core as recognition moieties and fluorescence quenchers. An FDA-approved amphiphilic polymer Pluronic F127 is then employed to encapsulate the molecular BTPE-NO 2 to render the nanoprobe BTPE-NO 2 @F127. The pathological levels of H 2 O 2 in the disease sites cleave the nitrophenyloxoacetamide groups and activate the probe, thereby generating strong fluorescent emission (950~1200 nm) and ultrasound signal for multi-mode imaging of inflammatory diseases. The nanoprobe can therefore function as a robust tool for detecting and imaging the disease sites with NIR-II fluorescent and multispectral optoacoustic tomography (MSOT) imaging. Moreover, the three-dimensional MSOT images can be obtained for visualizing and locating the disease foci.

Nanocodelivery of an NIR photothermal agent and an acid-responsive TLR7 agonist prodrug to enhance cancer photothermal immunotherapy and the abscopal effect

Bo Chen,Ruijie Huang,Wei Zeng,Wei Wang,Yuanzeng Min

The immunosuppressive tumor microenvironment (TME) greatly limits the actual outcome of immunotherapy. Therefore, it is urgent to develop appropriate strategies to reshape the TME and ultimately induce a strong immune response. Here, we developed a dual-functional liposome loaded with the photothermal agent IR808 near the infrared region (NIR) and Toll-like-receptor-7 (TLR7) agonist loxoribine prodrug (Lipo@IR808@Loxo) to achieve NIR light-triggered photothermal therapy (PTT) and the targeted delivery of immune adjuvants. Under NIR irradiation, Lipo@IR808@Loxo could greatly improve the efficiency of PTT to directly kill tumor cells and release tumor-associated antigens, which could work together with loaded loxoribine to relieve the immunosuppressive TME, effectively promoting the activation of antigen-presenting cells and subsequent antigen presentation . In this way, Lipo@IR808@Loxo could act as an in situ therapeutic cancer vaccine, eventually inducing a potent antitumor T-cell response. When further combined with immune checkpoint blockade, Lipo@IR808@Loxo-mediated photothermal immunotherapy could not only eliminate the primary tumors but also inhibit the growth of distant tumors, thus enhancing the abscopal effect.

Preparation of NIR-responsive, ROS-generating and antibacterial black phosphorus quantum dots for promoting the MRSA-infected wound healing in diabetic rats

Shuocheng Huang,Shibo Xu,Yanan Hu,Xingjun Zhao,Linna Chang,Zhenhua Chen,Xifan Mei

Multidrug-resistant (MDR) bacteria-induced infection is becoming a huge challenge for clinical treatment, especially for non-healing diabetic wound infections, which increase patient mortality. MRSA infections and delayed wound healing (methicillin-resistant Staphylococcus aureus) accounted for a higher proportion. Although surgical debridement and continuous use of antibiotics are still the main clinical treatments, new multifunctional therapeutic nanoplatform are attractive for MIDW. Thus, in the present study, black phosphorus quantum dots (BPQDs) encapsulated in hydrogel ( [email protected] ) were utilized as nanoplatforms for MIDW treatment to achieve the multifunctional properties of NIR (near infrared) responsiveness, ROS (reactive oxygen species) generation and antibacterial activity . Upon NIR irradiation, the temperature of the [email protected] MIDW area rapidly increased up to 55 °C for sterilization. In vitro experiments showed that [email protected] exerted a synergistic effect on inhibiting MRSA by producing of ROS, lipid peroxidation , glutathione , adenosine triphosphate accumulation and bacterial membrane destruction upon NIR irradiation. The resulting [email protected] achieved an effective sterilization rate of approximately 90% for MRSA. Furthermore, animal experiments revealed that [email protected] achieved an effective closure rate of 95% for MIDW after 12 days by reducing the inflammatory response and regulating the expression of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF). Meanwhile, intravenous circulation experiments showed good biocompatibility of BPQDs, and no obvious damage to rat major organs was observed. The obtained results indicated that [email protected] achieved the synergistic functions of NIR-responsiveness, ROS generation, and antibacterial activity and promoted wound healing, suggesting that they are promising multifunctional nanoplatforms for MIDW healing. Statement of significance 1. NIR-triggered ROS-generating and antibacterial nanoplatforms are attractive in the wound healing field. 2. In this work, black phosphorus quantum dots encapsulated in a hydrogel were used as a nanoplatform for treating MRSA infected wounds. 3. The obtained materials have achieved an effective sterilization rate for MRSA and effective wound closure rate.

Degradable and Excretable Ultrasmall Transition Metal Selenide Nanodots for High-Performance Computed Tomography Bioimaging-Guided Photonic Tumor Nanomedicine in NIR-II Biowindow

Lile Dong,Lining Sun,Wenjuan Li,Yunping Jiang,Yingying Zhan,Luodan Yu,Yu Chen,Guobin Hong

Transition metal selenide nanodots (NDs) represent distinctive antitumor agents for cancer treatment, but their non-biodegradability may bring serious adverse effects and potential long-term toxicity to internal tissues/organs, which substantially hinder their further clinical translations. In this work, the construction of a multifunctional theranostic nanosystem based on degradable and excretable ultrasmall Rh 3 Se 8 NDs by a general bovine serum albumin-templated strategy is reported. The constructed Rh 3 Se 8 NDs exhibit distinctively high photothermal-conversion efficiency (57.5%) in the second near-infrared biowindow, making them highly applicable for photoacoustic imaging and photonic hyperthermia at the desired wavelength. Rh 3 Se 8 NDs exhibit a high tumor growth inhibition rate (98.1%) on 4T1 breast tumor-bearing mice due to the desirable photonic hyperthermia performances. Especially, the fabricated Rh 3 Se 8 NDs feature the large X-ray attenuation coefficients of the Rh component for contrast-enhanced X-ray computed tomography imaging. Importantly, the prominent biodegradability of Rh 3 Se 8 NDs enables their quick excretion out of the body for potentially avoiding inflammation and mitigating long-term toxicity. Therefore, this work highlights the construction of proof-of-concept biodegradable and excretable ultrasmall inorganic theranostic nanosystems for multiple bioimaging-guided cancer nanotherapeutics, guaranteeing the further clinical translations of inorganic nanoparticles in biomedicine.

Fabrication of dopamine enveloped WO3−x quantum dots as single-NIR laser activated photonic nanodrug for synergistic photothermal/photodynamic therapy against cancer

Jianping Sheng,Ling Zhang,Liu Deng,Yajing Han,Liqiang Wang,Haichuan He,You-Nian Liu

Tungsten oxide is a promising nanodrug in phototherapy via transforming light energy to reactive oxygen species and heat. Nevertheless, the phototherapeutic activity of the tungsten oxide nanomaterials is generally activated under 980 nm laser, which is close to the absorption of normal tissue, resulting in unfavorable heating effect on normal tissues. Therefore, the tungsten oxide nanomaterials which can be excited under 808 nm are highly required to avoid overheating and obtain deep tissue penetration of near-infrared (NIR) laser. Here, we successfully synthesized a novel dopamine enveloped tungsten oxide nanodots (WO 3−x /Dpa-Mel NPs) via facile one-step solvothermal route, using dopamine as the template and reductant in triethylene glycol solvent. Under the 808 nm laser irradiation, the as-obtained WO 3−x /Dpa-Mel NPs exhibit excellent photodynamic activity and stability. Meanwhile, the WO 3−x /Dpa-Mel NPs also possess high photothermal conversion property. Thus, the photothermal therapy (PTT) and photodynamic therapy (PDT) can be triggered simultaneously under single 808 nm laser irradiation. Additionally, the inherited good biocompatibility and dispersity in aqueous solution from dopamine hydrochloride (Dpa-Mel) also make the WO 3−x /Dpa-Mel NPs more suitable for in vivo application. The as-obtained WO 3−x /Dpa-Mel NPs exhibit an excellent synergistic phototherapy effect on solid tumor ablation in vivo without damaging healthy tissues under single 808 nm NIR light irradiation, indicating WO 3−x /Dpa-Mel NPs can serve as a multifunctional therapeutic nanoplatform to realize the synergistic cancer therapy.
