Preparation, Characterization of Amides from Piper wallichii (Miq.) Hand.-Mazz. and Key Compounds including Pellitorine Screened by Fingerprint-efficacy Correlation of Antioxidant Activity

Dezhi Shi,Jing Ji,Yue Wang,Lirong Lu,Shaohui Zhong,Zhifeng Tang,Yunyu Zhang,Wufeng Gao,Meng Cao,Yang Hu,Yunfeng Zheng,Shiwen Huang,Jianming Cheng

The industrial enrichment of fat-soluble compounds is a common challenge, which is also present in Piper wallichii (Miq.) Hand.-Mazz. (PWH). The stem of PWH is rich in amide alkaloid substances, although enrichment methods have been available for total amide alkaloids, their water-insoluble character has not yet been addressed by a green and efficient method. We have elaborated the adsorption characteristics of pellitorine and futoamide on macroporous resins. Synchronization of concentration and adsorption was used to solve the critical problem of enrichment on macroporous resins with lipid-soluble compounds that tend to clog the resin columns during the enrichment process. Fifteen batches of PSA (amide alkaloids of PWH) were prepared from PWH collected from multiple origins, and multidimensional fingerprints were established in combination with multivariate statistical analysis to evaluate the consistency and variability of the compounds in multiple batches. Then, the fingerprints were correlated with their corresponding antioxidant activities to identify the pharmacodynamic compounds, thus clarifying the key compounds. Finally, compound separation was performed to verify the accuracy of the antioxidant activity of the key compounds and fingerprint-efficacy correlation model. HP-20 macroporous adsorbent resin has optimal adsorption and desorption which was selected for the enrichment of PSA. The adsorption of PSA, including futoamide and pellitorine, on the HP-20 resin was consistent with the Pseudo-second order kinetic model and Langmuir isotherm model, and this adsorption was a spontaneous exothermic entropy increase mechanism. The on-column adsorption process was optimized to solve the phenomenon that the precipitation of fat-soluble amides were easy to block the glass column during the enrichment process of the macroporous adsorption resin, and the effective enrichment of PSA was achieved. The content of pellitorine and futoamide was 13.95 g/100g and 3.11 g/100g in prepared samples respectively. The percentage of PSA was more than 50 g/100g. Seventeen differentiated compounds were screened by multivariate statistical analysis, antioxidant activity of 4 individual compounds isolated by fingerprint-efficacy correlation such as pellitorine were validated. The present study provided a more effective method for the enrichment of PSA, and initially elucidated the active compounds of the antioxidant activity of PWH, which will establish the basis for the industrialization of PSA in the future.

Hydrolysis kinetics and structure characterization of ball milled bamboo impregnated with various diluted acids

Wei Yang,Cong Cong Wang,Lingjun Wang,Li Bao,Fan Yang,Shengji Wu,Lei Che

The hydrolysis mechanism of ball milled bamboo impregnated with deionized water (B-ball), H 2 SO 4 (B-H 2 SO 4 ), HCl (B-HCl) and CH 3 COOH (B-CH 3 COOH) in subcritical water was investigated in the present study. Various mechanochemical reactions were induced during acid-associated mechanical milling and bamboo with improved solubility, reduced thermal stability and solubility was obtained. Acid-associated mechanical treatment also significantly affected the hydrolysis behavior of bamboo in subcritical water. The hydrolysis kinetics of pretreated bamboo could be well expressed by the Coats-Redfern method. The acid strength dominated the hydrolysis of bamboo during ball milling and subcritical water treatment. H 2 SO 4 and HCl possessed the highest ability to dissociate the glycosidic bond and holocellulose-lignin chemical linkages. The above ability, together with their catalytic effects resulted in the lower activation energies required to initiate the hydrolysis of B-H 2 SO 4 (38.87 kJ mol −1 ) and B-HCl (38.23 kJ mol −1 ) compared with those of raw bamboo (74.34 kJ mol −1 ), B-ball (104.42 kJ mol −1 ) and B-CH 3 COOH (90.65 kJ mol −1 ) in subcritical water. The cleavage of holocellulose-lignin chemical linkages liberated the polycondensation of monomer and hydrolysable oligomers from holocellulose for glucose char and aromatic-linked polymer char production, leading to the increase of solid residue yield at elevated reaction temperature. It also promoted the glycosidic bond cleavage and ring scission reactions during hydrolysis in subcritical water, resulting in the improved yields of carbohydrate, 5-hydroxymethylfurfural and furfural.

Longsheng Chen,Wei Xu,Rong Shao,Xianfeng Du

The profile and bioactivity of phytochemicals in Araiostegia yunnanensis ( Christ ) Cop were investigated. The total flavonoids content in A. yunnanensis is about 84.90   mg/g. By means of HPLC–DAD–ESI-MS, the main flavonoids in A. yunnanensis were tentatively identified as myricetin 3- O -rhamnosylglucoside, eriodictyol 7- O -rutinoside, quercetin 3- O -rutinoside, luteolin-7- O -apiosylglucoside, quercetin 3- O -rhamnosylgalactoside, and luteolin 7- O -glucoside. The extract (0.268   mg/ml total flavonoids) from A. yunnanensis showed very strong superoxide anion radical scavenging potential and reducing power, which are higher than those of rutin (0.25   mg/ml). The extract (0.268   mg/ml total flavonoids) from A. yunnanensis exhibited similar DPPH scavenging activity with rutin (0.25   mg/ml). However, rutin (0.25   mg/ml) showed a significantly higher ABTS radical scavenging effect than that of the extract (0.268   mg/ml total flavonoids) from A. yunnanensis. The methanol extract from A. yunnanensis showed obviously cytotoxic effects on A549 cells and it had no effect against acetylcholinesterase.

Yu Zhang,Fengrui Li,Honglei Yan,Yan Zhang,Shimiao Feng,Lei Deng,Lixia Zhao,Shuang Gao,Ying Fu,Fei Ye

The development of bio-based active food packaging is urgently needed to address food safety concerns. In this study, nerolidol/hydroxypropyl- β -cyclodextrin inclusion complex nanofibers (Nerolidol/HP β CD-IC-NF) were prepared by electrospinning to improve the thermal stability and water solubility of natural antimicrobial nerolidol. Structural characterization (FTIR, XRD, 1 H NMR) confirmed successful nerolidol encapsulation within HP β CD cavities. The resulting bead-free nanofibers exhibited high surface area and porosity. Nerolidol/HP β CD-IC-NF demonstrated a 3.3-fold increase in free radical scavenging capacity compared to free nerolidol, along with potent antimicrobial activity (88.1 % and 76.0 % inhibition against E. coli and S. aureus , respectively). In strawberry preservation trials, nanofiber membranes reduced B. cinerea lesion diameters by 5.56-fold after 5 days compared to untreated controls, significantly extending shelf life. These findings highlight Nerolidol/HP β CD-IC-NF as a promising solution for preventing microbial contamination and improving food preservation efficacy in active packaging applications.

Highly adhesive, stretch, antioxidative and antibacterial double-network hydrogel containing artemisinin for infected wound closure and healing

Xinxiang Chai,Ruifei Ge,Yanzhen Lou,Fozil Mamaraim Ugli Turaqulov,Khaydar E. Yunusov,Lei Nie,Muhammad Rafique Khan,Guohua Jiang

Infected wounds can trigger excessive oxidative stress and chronic inflammation, delaying wound healing, consequently placing a burden on human health and socioeconomics. In clinical applications, there is a lack of multifunctional wound dressings that combine antibacterial and antioxidant with good tension and adhesion properties. This study developed hydrogel dressings composed of polyvinyl alcohol (PVA)/mussel mucin (MUM)/ferric ion (PVA/MUM-Fe)/artemisinin (ART) (PVA/MUM-Fe/ART) by freeze-thawing method. The energy dissipation of PVA crystals and the double-network based on hydrogen and coordination bonds endow the hydrogel with high structural stability, strength and toughness. The hydrogel exhibited good tissue adhesion and ROS-scavenging properties due to the rich dopamine groups in the hydrogel. Furthermore, the hydrogel also exhibited good antibacterial, anti-inflammatory properties and high biocompatibility. In vivo results demonstrated that compared to commercial dressings, the resultant hydrogel dressing facilitated wound healing on an infected full-thickness skin defect by upregulating the growth factors such as VEGF-A and VEGF-C and downregulating that of inflammatory factor IL-6 and TNF-α. Transcriptomic results revealed that as-fabricated hydrogel dressing could promote the healing and regeneration of wound tissue by regulating inflammatory reaction, cell proliferation and migration, angiogenesis and matrix remodeling.

Multifunctional Polyphenolic Nanoparticle-Cross-Linked Aminoglycoside Hydrogels

Zhan Li,Jianhua Zhang,Hengjie Zhang,Tianyou Wang,Yuxian Song,Pengyu Liu,Yiwen Li,Hong Liu,Wancai Guo,Zhipeng Gu

Multifunctional hydrogels with integrated antioxidant and antibacterial activities are vital for modulating the wound microenvironment, mitigating oxidative stress, and preventing infection. However, efficient construction of such hydrogels with programmable responsiveness and structural integrity that can be tailored to therapeutic demands remains challenging. Herein, we developed a modular, functionally programmable hydrogel platform constructed via an in situ Schiff base reaction between aldehyde-functionalized polyphenolic nanoparticles and aminoglycoside antibiotics. The resulting nanocomposite hydrogels exhibited excellent mechanical properties, pH-responsiveness, biodegradability, and biocompatibility, attributed to the synergistic interactions between the functional nanoscale building blocks and cross-linkers. In vitro and in vivo evaluations confirmed the hydrogel’s potent antibacterial and antioxidant capabilities, enabling effective infection control and attenuation of oxidative stress in wound environments. This strategy offers a versatile route for engineering adaptive, multifunctional hydrogels for advanced wound management.

Co-fermentation of Xuxiang kiwifruit juice by Lacticaseibacillus paracasei with other lactic acid bacteria: impacts on quality properties, nutrients, volatiles, and in vitro antioxidant activities

Shufan Xie,Rong Wang,Xiaoli Xiao,Wen Zhang,Zhiming Pan,Chang Shu,Jiaqi Chen,Xiao Yang,Yunlu Wei

This study addressed kiwifruit's postharvest perishability and nutrient loss during juicing by co-fermenting Xuxiang kiwifruit juice with Lacticaseibacillus paracasei and five other LAB strains. We evaluated fermentation performance, nutrition, volatiles, sensory attributes, and antioxidant changes during digestion to develop a high-nutrition functional beverage. Co-fermentation significantly increased total phenolics, flavonoids, amino acids, and organic acids while boosting antioxidant activities (DPPH/ABTS, SOD). Volatile profiles strongly correlated with sensory attributes, and specific co-fermentation combinations produced complex aromas and a smooth mouthfeel preferred by consumers. Strain-specific metabolic activities created distinct flavors, such as creamy, or apple-citrus notes. L. paracasei : L. fermentum produced 3-methyl-1-butanol (13.70 ± 0.11 %), enhancing fruity notes. Fermented samples maintained high SOD activity and ABTS scavenging after in vitro digestion, the L. paracasei : L. acidophilus combination showed the highest ABTS radical scavenging activity (51.88 ± 0.06 %) and SOD activity (135,300 U/mL). LAB co-fermentation is a sustainable strategy to simultaneously improve kiwifruit juice's nutritional and sensory quality.

A natural oleanolic acid self-assembly system for curcumin: Structural insights, enhanced physicochemical properties, and synergistic antioxidant activity

Kangkang Zhi,Yi Shi,Weibing Lin,Huilan Yang,Yawen Zhu,Jiajia Cui,Wen Zhang,Junlong Wang,Ji Zhang

The present study developed a natural nano-assembled delivery system with curcumin (Cur)-synergistic bioactivity using natural oleanolic acid (OA). The OA self-assembled nanoparticles (OA NPs) were systematically prepared via region-limited self-assembly, with their size (20–600 nm) regulated by varying key preparation parameters. The XPS, FTIR, CAT, and MD revealed that the 3-hydroxy oxygen and 17-carboxyl oxygen of OA act as hydrogen bond donor and acceptor, respectively, to form hydrogen bonds. Together with van der Waals forces, these interactions drive the formation of hydrophilic surface-exposed nanoparticles. The OA NPs exhibited good biocompatibility, demonstrated by favorable blood safety (hemolysis rate <5 %) and low cytotoxicity (cell viability >90 %). The Cur was efficiently encapsulated into OA NPs, achieving a LC of 22.81 % and EE of 68.43 %. The encapsulated Cur showed significantly improved physicochemical properties, with a 29.02-fold increase in water solubility, a 3.64-fold enhancement in photostability, and a 2.77-fold improvement in thermal stability compared to free Cur. In vivo , OA NPs exhibited intestinal retention time three times longer than gastric retention, and release studies confirmed their intestinal-selective and sustained-release behavior, which followed the Korsmeyer–Peppas model. The Cur-OA NPs demonstrated optimal antioxidant activity, showing 2.45-fold higher potency (based on EC 50 values) than free Cur, attributable to the synergistic effect between Cur and OA NPs. This study highlights the potential of Cur-OA NPs as an oral antioxidant formulation and offers a promising strategy for developing natural nanocarriers with curcumin-synergistic bioactivity.

Rational design of caffeic acid amides as novel antioxidants in lipid systems: Experimental and theoretical studies

Zhiheng Yuan,Qicheng Sun,Yaoyao Zhang,Sichang Ti,Meimei Shan,Yang Xiao,Shangde Sun

Caffeic acid (CA), known as a natural phenolic compound, is widely recognized for its antioxidant properties and potential applications in food preservation. However, its practical antioxidant efficacy and potential applications have been limited due to its poor lipophilicity. To overcome this issue, based on the hydrophobic residue structure and antioxidant activity of amino acids, three amino acids (methionine, tyrosine, and tryptophan) were screened for amidation with CA to both improve the lipophilicity of CA and enhance its antioxidant performance. The antioxidant efficacy of CA amides was evaluated by experimental and theoretical approach. The experimental results showed that CA amides with tryptophan exhibited the excellent antioxidant efficacy, which increased by 49% and 47% compared to CA in DPPH and FRAP assays, respectively. The induction period of lard with CA amides in the Rancimat test was increased by 45-55% compared to CA. Density Functional Theory (DFT) analysis demonstrated that tryptophan conjugation improved the electron-donating ability (E HOMO from -7.306 to -6.863 eV) and reduced the O-H bond dissociation energy (from 79 to78 kcal/mol) of CA amide. Moreover, the 4-OH site was identified as the primary active site for radical scavenging. These results clarified how specific amino acid residues modulate the antioxidant behavior at the molecular level and indicated that CA amides with tryptophan were more effective in scavenging free radicals. These findings offer valuable insights into the structure-antioxidant activity relationship of CA amides, highlighting their potential as antioxidants and application in inhibiting lipid oxidation.

A dual stimuli-responsive cellulose-based double network hydrogel crosslinked with fluorescent carbon dots for controlled drug release

Jingwei Gong,Yern Chee Ching,Shuangwu Huang,Q. Jason Niu,Anyu Li,Ching Kuan Yong,Thennakoon M. Sampath Udeni Gunathilake,Nguyen Dai Hai,Chuah Cheng Hock

Cellulose-based carbon dots (C-CDs) with fluorescence and non-toxicity properties have attracted wide attention in the field of biomedicine. However, constructing dual-stimuli hydrogels incorporated with C-CDs using biomass-based raw materials with biodegradable and antimicrobial characteristics for drug sustained-release remains a challenge. Herein, an interpenetrated-network method was proposed in which the N-isopropyl acrylamide (NIPAM) monomer, regarded as the second network, was polymerized via photo-initiation. Additionally, C-CDs derived from lychee waste were used as cross-linking agent, which was introduced into β-cyclodextrin (β-CD)/carboxymethyl cellulose (CMC)/polyvinyl alcohol (PVA) first-network system, to fabricate a dual-network fluorescence hydrogels called β-CD/CMC-(PVA-NIPAM). The design of dual-network hydrogel can effectively enhance swelling and water retention abilities within a certain time. To evaluate the biomedical application of the obtained hydrogel, ibuprofen, as a poorly water-soluble model drug, was loaded into the hydrogel utilizing advantages of possessing hydrophobic inner cavity of β-CD and the highly dense structure of double-network hydrogels, to complete release behavior under different pH and temperature, with the maximum release rate reaching up to 96 %. Furthermore, it is noted that the cellulose-based interpenetrated-network hydrogel has access to cells but exhibits no cytotoxicity in vitro. Herein, this dual-responsive fluorescent hydrogel is expected to play a potential role in drug delivery and drug tracking.

Carbon-Dot-Zn2+ Assembly Hybrid Multifunctional Hydrogels for Synergistically Monitoring Local Glucose and Repairing Diabetic Wound

Xu Yang,Xinmei Duan,Li Zhu,Xiong Zhao,Shirong Zhang,Sheng Ni,Shuai Wu,Zhiqiang Han,Wuquan Deng,Da Sun,Qiao Chen,Wei Wu

The prolonged healing period and increased rates of amputation and mortality are significant challenges for diabetic patients’ wound care in clinical practice. Therefore, there is an urgent need to develop timely and efficient wound management strategies to monitor the local glucose while promoting rapid wound healing. In this study, a multifunctional hydrogel (PF127@Zn/C-G) was constructed by coloading the monitoring and therapeutic carbon dot (CD)-based agents (Zn/C) and glucose oxidase (GOx). For applications, the ″off–on″ characteristic in fluorescence (FL) signal of the Zn/C assemblies enabled the real-time monitoring of the local glucose levels in diabetic wounds. Moreover, the synergistic effect of Zn2+ and CDs was developed to promote wound healing. In vitro and in vivo results showed that the PF127@Zn/C-G hydrogel exhibited a good linear relationship between FL intensity and glucose concentration, and the FL signal intensity in the wound environment in vivo correlated well with the blood glucose, suggesting its function for timely reflection of the blood glucose content. In addition, the PF127@Zn/C-G hydrogel could accurately fit the wound contour to form a protective film, while effectively promoting cell proliferation and migration, and scavenging the accumulated ROS at the local wound, exhibiting an excellent angiogenic capacity and collagen deposition for significantly accelerating the wound healing during tissue remodeling. Therefore, the multifunctional hydrogel lPF127@Zn/C-G provides an effective and feasible candidate for improving the diabetic wound management.

Synthesis and Characterization of Argan Oil-Loaded Polyacrylamide Hydrogels: A Multifunctional Approach

Ahmad Ali,Jiaqi Yu,Qasim Qasim,Faizan Ikhlaq,Yangxufei Huang,Shun Yao

This study presents the pioneering development and comprehensive characterization of argan oil (AO)-loaded polyacrylamide (PAm) hydrogel (AO-PAm), establishing its multifunctional potential for biomedical applications. Systematic evaluation revealed that the AO-PAm hydrogel combines excellent mechanical properties (90% transparency, > 25 cm stretchability) with remarkable biological activity, demonstrating: (1) potent antibacterial effects against E. coli (1.2 ± 0.2 mm inhibition zone), P. aeruginosa (0.7 ± 0.2 mm), and S. aureus (0.6 ± 0.2 mm); (2) exceptional antioxidant capacity (97.6% DPPH scavenging at 75 μg/mL); and (3) pH-responsive swelling behavior. GC–MS analysis identified key bioactive compounds (β-pinene, camphene, undecanal, β-bisabolene) in AO that contribute to these properties, while microscopic characterization confirmed successful AO encapsulation within the PAm matrix. The hydrogel exhibited controlled drug release following Higuchi kinetics ( R 2 = 0.9547), with enhanced performance at physiological pH (6.5–6.8). These findings position AO-PAm hydrogel as a promising platform for advanced wound care, transdermal drug delivery, and daily healthcare applications, combining the therapeutic benefits of AO with the structural advantages of a synthetic hydrogel matrix.
