Background: The design of novel nanoparticles with higher therapeutic efficacy and lower side effects, is still difficult but encouraging in cancer therapy. Specifically, for upconversion nanoparticles (UCNP)-based drug release, a high intensity of NIR light (1.4~5.0 W/cm2) above the maximum permissible exposure (0.33 W/cm2 for 980 nm) is commonly used and severely limits its practical application. Methods: The highly emissive UCNP is first synthesized and then coated with mesoporous silica (MS) shell (UCMS). Next, the surface of UCMS is modified with the thioether (-S-BP) linker, leading to UCMS-S-BP nanoparticles. Finally, after the drug doxorubicin (Dox) is loaded into the pore channels of UCMS, the pore openings are blocked by the β-cyclodextrin (β-CD) gatekeeper through the association with the -S-BP linker (UCMS(Dox)-S-BP@β-CD).Results: Upon 980 nm NIR light irradiation with an ultralow intensity of 0.30 W/cm2, it is found that the loaded Dox can be released through the cleavage of thioether linkers triggering dissociation of β-CD gatekeepers. The in vitro results exhibited significantly therapeutic efficacy with 85.2% of HeLa cells killed in this study. Conclusions: An ultralow-intensity NIR light triggered on-demand drug release system has been developed by employing highly emissive UCNP and photocleavable linker with low bond dissociation energy to avoid the potential photodamage on healthy neighbor cells.

Real-time tracking drug release behavior is fundamentally important for avoiding adverse effects or unsuccessful treatment in personalize medical treatment. However, the development of a non-invasive drug reporting platform still remains challenging. Herein the design of a novel synthetic magnetic resonance imaging (MRI) agent for drug release tracking (SMART) is reported, which integrates photothermal core and paramagnetic ion/drug loading shell with a thermal valve in a hybrid structure. Through near-infrared (NIR)-II photothermal effect originating from inner Au-Cu 9 S 5 nanohybrid core, burst release of drugs loaded in the mesoporous silica shell is achieved. The concomitant use of a phase change material not only prevents premature drug release, but also regulates heating effect, keeping local temperature below 45 °C, enabling synergistic chemotherapy and mild hyperthermia in vitro and in vivo. Furthermore, the drug release from SMART facilitates proton accessibility to the paramagnetic ions anchored inside mesopores channels, enhancing longitudinal T 1 relaxation rate and displaying positive signal correlation to the amount of released drug, thus allowing non-invasive real-time monitoring of drug release event. The current study highlights the potential of designed MRI nanophores such as SMART for real-time and in-situ monitoring of drug delivery for precision theranostic applications.

In this study, the potentiality of applying attenuated total reflectance near-infrared (ATR-NIR) and attenuated total reflectance mid-infrared (ATR-MIR) techniques combined with a partial least squares (PLS) regression technology to quantify the total polyphenols (TPs) in Dendrobium huoshanense (DHS) was investigated and compared. The real TP contents in the DHS samples were analysed using methods of reference. The capability of the two IR spectroscopic techniques to quantify the TPs in DHS was assessed by the root-mean-square error of calibration (RMSEC) and determination coefficients ( R 2 ). The results showed that both NIR and MIR might be used as a fast and simple tool to replace traditional chemical assays for the determination of the TP contents in DHS, and the best NIR model showed slightly better prediction performance [root-mean-square error of prediction (RMSEP): 0.307, R 2 : 0.9122, ratio performance deviation (RPD): 4.43] than the best MIR model (RMSEP: 0.440, R 2 : 0.9069, RPD: 3.09). Results from this study indicated that both the NIR and MIR models could be used to quantify the TP in DHS, and ATR-NIR appeared to be the more predominant and more robust technique for the quantification of the TP in DHS.

Active drug-loaded nanocarriers have been widely employed as efficient drug delivery systems for tumor theranostics. Herein, we report folate-mediated “all-in-one” nanobubbles for tumor-targeted NIR/MR/US imaging and combined chemo-photothermal therapy. The surface-engineered nanobubbles are constructed from oleylamine-/IR-780-loaded hollow structures, folate and the Gd DTPA-BSA@5-FU complex via electrostatic adsorption and further filled with gas after freeze drying. DLS data show that the nanobubbles have a hydrodynamic diameter of 120.41 ± 18.30 nm. TEM observations show a hollow inner cavity and a shell thickness of approximately 10 nm. The relaxivity (r1) of the nanobubbles reaches 16.56 s −1 /mM, indicating suitable features for use as a T1-weighted MR contrast agent. Moreover, due to the gas core inside, the nanobubbles are suitable for ultrasound contrast imaging. Interestingly, ξ-potential data and cumulative release measurements demonstrate that the nanobubbles undergo charge-switchable behaviors and pH-/light-sensitive drug-release behaviors after surface engineering, which could facilitate deep tumor penetration and accelerate drug release for efficient killing of cancer cells. In vivo trimodal imaging and chemo-photothermal therapy for MGC-803 tumor-bearing mice reveal selective tumor accumulation, long tumor retention, and enhanced antitumor behaviors. Therefore, the all-in-one nanobubbles could be applied for active tumor-targeting theranostics.

Bilayer thermosensitive P(NIPAm-co-AAm) hydrogel discs were prepared by a facile UV light initiation process from N-isopropylacrylamide (NIPAm) and acrylamide (AAm) monomers’ cross-linking copolymerization. Poly(ethylene glycol) (PEG) as a pore-forming agent was added in order to form a porous structure and improve the water content in the hydrogel. Functional materials of NaYF4:Yb3+/Er3+ nanoparticles and multiwalled carbon nanotubes (MWCNTs) were incorporated into different layers of the P(NIPAm-co-AAm) hydrogel for the purpose of up-conversion luminescence labeling and the NIR light antenna effect, respectively. Significantly improved drug release from composite hydrogels was achieved in response to 980 nm NIR light irradiation by using lysozyme as a macromolecular drug. The multifunctional hydrogel reported here provides a platform for simultaneous NIR luminescence labeling and NIR-driven drug release.

Developing novel stimuli-responsive systems to optimize the co-delivery and concurrent controlled release of different anticancer drugs is highly pursued for reducing the probability of evolution of therapeutic resistance, which is the major cause of cancer mortality. In this work, we report a novel NIR light responsive drug co-delivery system based on hollow mesoporous Prussian blue nanoparticles (HMPBs) and a phase change material (PCM, such as 1-tetradecanol which has a melting point around 38 °C). The fabrication of this PCM@HMPB system simply involves filling the hollow interior and mesoporous shell of the HMPBs with PCM molecules, which acted as both a thermosensitive gatekeeper and a medium for loading both hydrophilic and hydrophobic anticancer drugs. The photothermal effect of the HMPBs under NIR light irradiation would induce the PCM to go through processes of melting and escaping from the HMPBs, concurrently releasing the encapsulated anticancer drugs into the surrounding medium. Notably, the as-prepared (PCM + drugs)@HMPB system could achieve near “zero release” of both hydrophilic doxorubicin hydrochloride (DOX) and hydrophobic camptothecin (CPT), as well as precise “on” or “off” drug delivery with various NIR light treatments in vitro to efficiently induce cell apoptosis. More importantly, according to the phenomenon that generated heat could improve drug absorption by increasing the cell metabolism and membrane fluidity, the (PCM + drugs)@HMPB system revealed significant photothermal-chemo synergistic effects against HeLa cancer cells. Therefore, the as-prepared (PCM + drugs)@HMPB system has good potential as a versatile NIR light responsive co-delivery platform of both hydrophobic and hydrophilic anticancer drugs for targeting adaptive therapeutic resistance as well as synergistic photothermal-chemo cancer treatment.

Rheumatoid arthritis (RA) is currently one of the most common systemic autoimmune disorders, with no clinically effective theranostics. Therapeutic manipulation of the gasotransmitter hydrogen sulfide (H 2 S) has exhibited promising potential as an innovative strategy in RA, whereas limited with non-targeting, low solubility, and systemic toxicity. In this work, based on the physicochemical and structural advantages of stimulus-responsive nanotechnology, we develop multifunctional glutathione (GSH)-responsive H 2 S nanogenerators (BDMA NGs), achieving activatable precise second near-infrared fluorescence (NIR-II FL)/photoacoustic (PA) bimodal imaging-guided gas/chemotherapy synergistic therapy. Contributed to the suitable size and excellent biocompatibility, the BDMA NGs accumulate passively in the RA region and then feature a responsive deformation reaction with GSH to release functional small molecules: H 2 S and methotrexate (MTX). The H 2 S undergoes a mineralization process with Ag + ions in situ, resulting in the rapid formation of Ag 2 S to “turn on” the NIR-II FL/PA signals. Remarkably, the effective generation of H 2 S downregulates hypoxia-inducible factor-1 α (HIF-1 α ) and promotes nuclear factor erythroid 2-related factor 2 (Nrf2) nuclear translocation, thereby restoring homeostasis in the RA microenvironment and facilitating the reprogramming of pro-inflammatory macrophages (M1) into anti-inflammatory phenotype (M2). Moreover, the controlled release of MTX, clinically approved drug for RA, synergistically promotes repolarization to further suppress local inflammation. Overall, the H 2 S-based nanogenerators BDMA NGs present a sophisticated and versatile strategy for implementing stimuli-responsive imaging and M1-to-M2 repolarization, demonstrating significant potential in the theranostics of RA and clinical transformation.

Elevating photothermal conversion efficiency (PCE) is the key to enhance curative efficacy during photothermal therapy. Nevertheless, current functionalized or synthetic methods for preparing photothermal agents with high PCE are too complicated, greatly hindering the development of photothermal therapy. In this study, we developed a tumor-targeting flower-like nanocomposite termed mesoporous platinum nanoparticles@Cu 2- x Se-AS1411 (MPNPs@Cu 2- x Se-AS1411, denoted as MPCSA) with remarkable PCE for high-resolution photoacoustic (PA) imaging guided photothermal/chemodynamic therapy for tumor ablation. We discovered that MPNPs served as carriers for delivering PA contrast agents. Cu 2- x Se nanosheets emerged as promising near-infrared-II (NIR-II) PA imaging and photothermal/chemodynamic therapy agents owing to their strong localized surface plasmon resonance property in the NIR-II region. The amine-modified AS1411 aptamer, which is connected to the surface of the flower-like MPNPs@Cu 2- x Se can target the nucleolar protein on the surface of 4T1 cells, achieving the accumulation of MPCSA at the tumor site in mice. Notably, these MPCSA nano-flowers exhibited an excellent PCE up to 82.33 % under 1064 nm laser irradiation, resulting in highly efficient cancer cell ablation and greatly hindering in vivo tumor growth. This study has paved the way for imaging-guided accurate cancer diagnosis and targeted cancer therapy.

Water-dispersed red upconversion nanoparticles (UCNPs) have great application prospects in the biomedical field. Thus, a series of Er 3+ -based upconversion nanoparticles (Er-UCNPs) with excellent red upconversion luminescence under three-wavelength NIR excitation (808/980/1550 nm) were prepared by a simple thermal decomposition method, and then further functionalized by PAA to achieve good water dispersibility. Compared with NaErF 4 bare core, the emission intensity of Er-UCNPs samples doping of 0.5%Tm 3+ and 0.5%Ho 3+ as energy capture centers was significantly enhanced, and further improved after coating with NaYF 4 inert shell. Meanwhile, among all Er-UCNPs@NaYF 4 samples, NaErF 4 @NaYF 4 exhibited the highest emission intensity and longest luminescence lifetime due to the efficient suppression of luminescence quenching effect, while NaErF 4 :0.5%Tm 3+ @NaYF 4 showed highest red luminescence purity owing to the energy transfer process between Tm 3+ ( 3 H 5 ) and Er 3+ ( 4 I 13/2 ). The hydrophilic Er-UCNPs@NaYF 4 @PAA samples still maintained good red luminescence under multi-wavelength excitation. The luminescence enhancement mechanism in Er-UCNPs@NaYF 4 was discussed in detail based on spectral characteristics. The as-prepared water-dispersed Er-UCNPs@NaYF 4 @PAA with bright red luminescence under multi-wavelength NIR excitation will have potential applications in targeted therapy and deep tissue imaging.

A series of new visible and near-infrared (NIR) luminescent lanthanide-hydroxybenz[de]anthracen derivatives (Ln(HBAN) 3 (H 2 O) 2 (NO 3 ) 3 , Ln(HBAN) 3 phen(NO 3 ) 3 ) were synthesized (HBAN-6-hydroxybenz[de]anthracen-7-one, Ln = Eu, Yb, Nd). Further, chemically bonded lanthanide hybrids of functionalized mesoporous silica (SBA-15) are prepared using HBAN-functionalized alkoxysilane (HBAN-Si) as linker, which is named as Ln(HBAN-SBA-15) 3 (NO 3 ) 3 , Ln(HBAN-SBA-15) 3 phen(NO 3 ) 3 (Ln = Eu, Yb, Nd), respectively. The physical characterization and especially the photoluminescence properties for these hybrids are performed. SAXRD and BET of them revealed that all of these hybrid materials have uniformity in the mesostructure. Worth noting here is that the excitation spectra of these hybrid materials are extended to visible light region (439–535 nm). Upon ligand-mediated excitation with the ultraviolet and visible light, the visible luminescence for europium hybrids and NIR luminescence for Yb 3+ and Nd 3+ hybrids are obtained.

Gastrodia elata Blume (Tianma) is widely cultivated and consumed as a food and herbal material in China. With its increasing use as a functional food, Tianma is gaining popularity in the market. However, during the cultivation process, the harvest time affects its maturity and storage time, while the total content of gastrodin (GAS) and p -hydroxybenzyl alcohol (HA) affects its nutritional value , making it difficult to control the quality of Tianma. In this study, a combination of visible-near-infrared (Vis-NIR) spectroscopy and chemometrics was used to identify the harvest time and determine the quality indicator content. A Vis-NIRRamanNet modeling method based on the architecture of RamanNet was proposed and compared with harvest time discrimination models constructed using partial least squares discriminant analysis (PLS-DA), k-nearest neighbor (KNN), support vector machine (SVM), and one-dimensional convolutional neural network (1D-CNN). The results demonstrated that Vis-NIRRamanNet exhibited superior performance, achieving accuracy rates of 0.995 and 0.973 on the training and testing sets, respectively. Subsequently, Vis-NIR spectroscopy combined with support vector regression (SVR) was employed for quantitative analysis of quality indicator content. Wavelength selection methods such as uninformative variable elimination (UVE), genetic algorithm (GA), and competitive adaptive reweighted sampling (CARS) were utilized to optimize the model. The results indicated that the CARS-SVR model provided the best prediction of quality indicator content, with root mean square error of prediction (RMSEP) and coefficient of determination ( R p 2 ) values of 0.257 and 0.901, respectively. Therefore, the combination of Vis-NIR spectroscopy and chemometrics shows significant potential in assisting the selection of high-quality Tianma.

Fluorescence/luminescence bioimaging, utilizing rare-earth doped nanocrystals (RENCs) within the NIR-IIb (1500–1700 nm) sub-window, has drawn interest for its deep penetration depth and superior resolution. Herein, Er 3+ /Yb 3+ couple is rationally designed in topologically arranged RENCs that facilitate a boosted 1530 nm emission of Er 3+ via premeditated Yb 3+ -mediated energy transfer processes under 808 nm excitation. To maximize this modulation effect for boosting 1530 nm emission, a regional active ions concentration and spatial regulation strategy is explored in the optimized concentration and core–shell–shell structure (NaErF 4 :20%Yb@NaYbF 4 :20%Er@NaYF 4 with a quantum yield of 16.46% in NIR-IIb region). Leveraging the superior emission and multi-wavelength excitation characteristics of the as-obtained nanocrystals, a NIR-IIb-based ratiometric fluorescence imaging measurement is achieved under 808 and 980 nm excitation, which cannot only show the 2D distribution of probes but also provide spatial depth information (range from 0 to 2.8 mm with a tissue depth resolution of 66 µm). The findings hold significant potential for advancing both vascular science and clinical medicine.
