Diabetic cardiomyopathy (DCM) requires a systematic approach for effective management. Focusing on key signaling pathways in myocardial fibrosis could offer novel therapeutic approaches for DCM. The objective of this research is to investigate new biomarker and the underlying mechanisms by which baicalin intervenes in the progression of DCM. Swiss Target Prediction databases and weighted correlation network analysis (WGCNA) were employed to collect and analyze targets of baicalin and DCM. Intersection targets were obtained through venn online website, network visualization was conducted through STRING database and Cytoscape 3.7.2 software. Molecular docking validation was completed via AutoDock Vina software. Western blotting, RT-qPCR and histology were carried out to validate the direct targets of baicalin and explore the molecular mechanism during DCM progression. In mice induced to DCM, we observed that disruptor of telomeric silencing 1-like (DOT1L) was upregulated. Baicalin (BC) reduced myocardial fibrosis in mice as indicated by the reduced expression levels of fibrosis markers COL-1 and TGF-β. The inhibitory effect of BC was reversed by overexpressed DOT1L and further enhanced by silenced DOT1L. Our results indicate that cardiomyocyte DOT1L exerts a pathogenic role in DCM and BC specifically targets DOT1L/COL-1 signaling to reduce myocardial fibrosis in DCM.

Monocarboxylic acid transporter 1 (MCT1) maintains axonal function by transferring lactic acid from oligodendrocytes to axons. Subarachnoid hemorrhage (SAH) induces white matter injury, but the involvement of MCT1 is unclear. In this study, the SAH model of adult male Sprague-Dawley rats was used to explore the role of MCT1 in white matter injury after SAH. At 48 h after SAH, oligodendrocyte MCT1 was significantly reduced, and the exogenous overexpression of MCT1 significantly improved white matter integrity and long-term cognitive function. Motor training after SAH significantly increased the number of ITPR2 + SOX10 + oligodendrocytes and upregulated the level of MCT1, which was positively correlated with the behavioral ability of rats. In addition, miR-29b and miR-124 levels were significantly increased in SAH rats compared with non-SAH rats. Further intervention experiments showed that miR-29b and miR-124 could negatively regulate the level of MCT1. This study confirmed that the loss of MCT1 may be one of the mechanisms of white matter damage after SAH and may be caused by the negative regulation of miR-29b and miR-124. MCT1 may be involved in the neurological improvement of rehabilitation training after SAH.

This paper is dedicated to improve the conversion efficiency of carboxylation for polymers of intrinsic microporous (PIM-1). According to stable thermal properties of PIM-1, a hydrothermal method is used for the first time to achieve the carboxylation conversion of PIM-1 (C-PIM), achieving a conversion rate of more than 90% at a reaction time of 10 hours, which is nearly five times more efficient than the most efficient conversion process currently available, preserving the film-forming properties. The completion of the carboxylation conversion is verified using 1 H-NMR and 13 C-NMR, FT-IR, and organic elemental analysis. It is found that the change in molecular weight affected the film-forming properties of the membrane material, and the hydrophobicity of the C-PIM membrane did not decrease compared with C-PIM membrane reported in previous studies. The effect of C-PIM on the selectivity of CO 2 gas is explained from the microscopic to the macroscopic level by using physical characterization such as molecular structure analysis, XRD, cross-linkage calculation, pore size and volume distribution, and fractional free volume (FFV) calculation. It verifies that the efficiency of CO 2 adsorption and diffusion selectivity for PIM-1 membranes has been improved by carboxylation, preserving excellent separation, and capture capacity of CO 2 .

Background Beta-secretase 1 (BACE1) is a rate-limiting enzyme in the generation of amyloid beta peptides, which are associated with Alzheimer’s disease (AD). It has been reported that Schisandrin B could improve cognitive functions in animal models of AD, but the underlying mechanisms are not completely understood. Material/Methods In this research, in order to investigate the effects of Schisandrin B on amyloid-β (Aβ) metabolism and its mechanisms, amyloid precursor protein (APP) and its proteolytic products were determined by enzyme-linked immunosorbent assay (ELISA), western blotting, and RT-PCR after incubation of N2a/Swe cells with Schisandrin B. Results The results indicated that Schisandrin B can significantly reduce the level of secretion of Aβ 40 and Aβ 42 secreted in N2a/Swe cells. Additionally, there was nonsignificant change in APP level after Schisandrin B treatment. Treatment of Schisandrin B dramatically reduced the mRNA and protein expression levels of BACE1. Moreover, Schisandrin B treatment resulted in a reduction of protein level of sAPPβ, an APP fragment cleavage by BACE1. Conclusions These results suggest that Schisandrin B inhibits the transcription and translation of BACE1, suppresses the activity of BACE1, and ultimately attenuates Aβ generation, which provides a novel mechanism for the regulation of Aβ metabolism by Schisandrin B.

Introduction. The advances in cerebral ischemia treatment have resulted in a larger proportion of patients get the benefits of rebuilding blood flow to the brain. Then, ischemia-reperfusion injury has emerged as a new essential problem. Dectin-1 plays an important role in cerebral ischemia-reperfusion injury by regulating the function of immune cells. Methods. C57BL/6 was blindly divided into four groups including the sham-operated group and the three different kinds of middle cerebral artery occlusion (MCAO) groups (after 6 hours, 12 hours, and 24 hours after plug removal). The protein expression levels of dectin-1, proapoptosis molecule, and antiapoptosis molecule were measured by using western blot analysis. The brain tissue was analyzed by flow cytometry to detect the M1 macrophage levels. Results. The correlation analysis of dectin-1 and infarct areas showed that there was an obviously positive correlation in between them (R = 0.9603). Dectin-1, cleaved caspase-3, and Bax increased, while antiapoptosis molecule, Bcl-2, decreased at three appropriate time points (after 6 hours, 12 hours, and 24 hours). The level of M1 macrophages in the experimental group increased after ischemia-reperfusion injury compared to the control group. Conclusions. The high expression level of dectin-1 may affect M1 macrophage polarization and brain cell apoptosis in cerebral ischemia-reperfusion injury.

P-Xylene is a prevalent indoor hazardous volatile organic compound(VOC). A long-term exposure limit of 100 ppm for p-xylene is generally recommended. Therefore, sensitive and selective detection of trace p-xylene is crucial. This work presents a method for detecting p-xylene gas by combining terahertz (THz) metasurface sensor with a metal-organic-framework (MOF) material, copper benzene-1,3,5-tricarboxylate (HKUST-1). The sensor with double split ring resonators is fabricated using a micromachining process and characterized by a terahertz time-domain-spectroscopy (TDS) system. The test results are consistent with the simulation. Subsequently, the sensor's surface is modified using the MOF material of HKUST-1 to enhance its sensitivity to the p-xylene gas. At room temperature, the sensitivity for p-xylene gas concentrations is between 1-100 ppm .The modified sensor has a maximum response frequency shift of 14 GHz for p-xylene gas at 100 ppm. Furthermore, the specificity, repeatability and stability of the sensor are characterized. The proposed method for highly sensitive and specific detection of p-xylene gas has important reference significance for the development of other types of gas sensors.

One-dimensional microporous zeolite, EU-1, has tighter restriction for bulky intermediates, exhibiting more favorable selectivity of propylene than ZSM-5 in catalytic cracking. However, the diffusion limitation accelerates coke generation, passivates acid sites and deactivates the zeolite. In this study, a strategy was proposed to etch amorphous species between EU-1 nanocrystals using NH 4 F solution to improve the diffusion property. It was found that the etching process occurred on the crystal surface and mesopores were introduced. Catalytic performance evaluation in n-hexane cracking demonstrated that the maximum propylene selectivity over parent EU-1 was 33.6 wt% ( vs . 5.7 wt% for ZSM-5). The n-hexane conversion was increased by 24 wt% over EU-1 etched in 20 wt% NH 4 F solution. Moreover, the introduction of mesopores provides accommodation space for coke. This study developed hierarchical EU-1 zeolite with favorable diffusion ability and reported a feasible strategy to improve the catalytic performance of EU-1 for propylene production.

Advanced oxidation processes (AOPs), particularly photocatalysis catalyzed by efficient and economical photocatalysts, is an emerging and sustainable approach. Band gap engineering, and spin polarization induction by anchoring spatially adjacent redox sites (metal cation vacancies) in the material is an excellent way for ensuring enhanced photocatalytic performance. Inspired by this, herein a novel and effective Sn rich Co co-doped Co 0.5 Sn 2 Mn 0.5 O 3 (C-SMO 3 ) oxide perovskite photocatalyst has been designed to accelerate charge separation and inhibit charge carrier recombination at redox intimate interfaces and defective Co, Mn 3 + /Mn 4+ dual sites for photocatalysis. The synthesized photocatalyst has been confirmed by various characterizations such as UV-Vis spectrophotometry, FTIR, XRD, SEM, TEM, HR-TEM, Raman, XPS, etc. The FTIR shows characteristics peaks of all constituent components. The crystalline morphology, d-spacing and shape of composite materials deduced from SEM, TEM, HR-TEM and XRD are all in agreement with each other hence the formation Co doped Sn rich SnMnO 3 has been confirmed. The experimental results demonstrate C-SMO 3 a potential dynamic dual site contender for wastewater decontamination. The spatially adjacent redox sites Co (1- x) - Mn 3+ /Mn 4+ inhibit photo-exciton recombination and accelerates charge carrier responsible for photocatalytic degradation of contaminants synchronized by light harvesting efficiency. The dual redox half reactions synergized photoinduced charge carriers dynamically enhanced the Sn 2 Mn 0.5 O 3 /Co (1- x) photodegradation property and performed 98 % breakdown of EB in 50 min, 96.7 % of CV dye in 90 min, and 90.9 % of CEF antibiotics under visible light irradiations optimized by RSM. The photocatalyst was recycled and reused due to its convenient recyclability, resulting in nearly stable degradation efficiency.

Numerous studies reported that betulinic acid (BA), a natural product extracted from birch bark, exhibited various beneficial effects in vitro. However, its pharmacological activities in aging are rarely understood. In this study, Caenorhabditis elegans was deployed as a whole animal model to investigate the impacts of BA on lifespan and stress resistance. Wild-type C. elegans were fed in the presence or absence of BA and tested for a series of phenotypes, including longevity, mobility, reproductive capacity, pharyngeal pumping, heat stress, and oxidative stress. BA at the optimal dose (50 μg/mL) extended the lifespan, improved the healthspan, and significantly evoked the increased oxidative stress resistance in C. elegans. Incorporating the genetic analysis with different types of longevity mutants, DAF-16, the downstream effector of the Insulin/IGF-1 receptor signaling, was revealed to mediate the protective effects of BA on lifespan and antioxidant activity. Together, these data showcased the potential of BA in promoting healthy aging, which shall facilitate its further development in the food and pharmaceutical industries.

Microvascular obstruction (MVO) is recognized as an independent risk factor for adverse prognosis in patients with myocardial infarction (MI), and its development is mechanistically linked to inflammation and microthrombus. Maresin 1 (MaR1), a specialized pro-resolving mediator, has demonstrated therapeutic potential in counteracting these processes, but its translation has been hindered by poor stability and rapid degradation. To overcome these limitations, we report the development of a cardiac-targeted liposomal nanotherapeutic (nano-MaR1) designed to stabilize and selectively deliver MaR1 to the injured myocardium. Nano-MaR1 was generated through a scalable nanoprecipitation method in a herringbone microfluidic mixer, with MaR1 encapsulated by a freeze–thaw approach to minimize degradation, and further modified with a cardiac-homing peptide for organ-specific targeting. In a murine MI model, intravenous administration of nano-MaR1 led to rapid myocardial enrichment within 12 h, with 20 μM identified as the optimal therapeutic dose. Treatment markedly reduced infarct size, cardiomyocyte apoptosis, inflammatory infiltration, and MVO formation, while promoting reparative macrophage polarization toward the M2 phenotype. Mechanistic studies revealed that these effects were mediated by downregulation of Itgam (CD11b) within the coagulation cascade and inhibition of monocyte–platelet aggregate formation, which were abolished upon CD11b blockade. Collectively, this work establishes cardiac-targeted liposomal encapsulation as a powerful strategy to enhance the stability and bioactivity of MaR1, and highlights nano-MaR1 as a promising therapeutic candidate for mitigating MVO and improving post-MI repair.

(1) Background: Ulcerative colitis (UC) is a persistent inflammatory condition of the intestine, characterized by dysregulated T cell-mediated immune responses. Curcumin (CUR), a common food additive and health supplement, is noted for possessing anti-inflammatory and immunomodulatory properties. Nevertheless, the molecular mechanisms underlying its therapeutic effects remain incompletely elucidated. This research aims to investigate the therapeutic mechanisms of CUR in UC, focusing on its role in restoring T cell homeostasis by modulating TIGIT and Neuropilin-1 (NRP1). (2) Methods: We employed a DSS-induced murine colitis model, combined with network pharmacology, molecular docking, protein–protein interaction docking, molecular dynamics simulations, andinvitroassays with Jurkat T cells. (3) Results: CUR markedly ameliorated clinical manifestations and histopathology in DSS-treated mice, restoring the balance of T cell and memory T cell subsets. Computational predictions and experimental validation showed that CUR downregulated TIGIT and NRP1 expression in inflamed colonic tissue and directly inhibited their expression in activated T cellsinvitro. (4) Conclusions: This study reveals a novel immunoregulatory mechanism of this natural compound. These findings suggest CUR modulates TIGIT/NRP1 to inhibit excessive T cell activation and restore immune homeostasis in UC.
