BACKGROUND In order to improve the stability of vitamin E and broaden its scope of application, an effective solution is to convert vitamin E into its derivatives. This work developed a new Candida rugosa lipase (CRL) nanogel based on modification of ionic liquid with vinyl functional groups. This novel CRL nanogel was used in the preparation process of vitamin E succinate based on the principle of non-aqueous enzymology. At the same time, various factors including enzyme concentration, substrate molar ratio, reaction temperature and reaction time, that affect the yield of vitamin E succinate were optimized and analyzed. RESULTS Different solvents with various hydrophobicity parameters (Log P values) from −1.3 to 3.5 were studied, it was found that dimethyl sulfoxide (DMSO) had the lowest Log P value among organic solvents but vitamin E succinate had the highest yield in DMSO. Furthermore, the effect of different operating conditions, such as molar ratios of substrate, enzyme concentration, reaction temperature and reaction time was studied. Under the optimal process conditions (enzyme concentration 6 mg mL −1 , substrate molar ratio 4:1, reaction temperature 55 °C and reaction time 15 h), the product yield was 62.58 ± 1.16%. CRL and CRL nanogel were characterized using Fourier-transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). It was shown that CRL nanogel influenced the catalytic behavior of lipase significantly by changing the structure and surface properties of CRL. CONCLUSION Novel lipase nanogel was an excellent biocatalyst for enzymatic synthesis of vitamin E succinate. © 2020 Society of Chemical Industry

NiCo layered double hydroxide (NiCo-LDH) was explored as electrocatalyst for nonenzymatic detection of H2O2, in which NiCo-LDH with different morphologies were synthesized through a hydrothermal method, and then acid-etched NiCo-LDH (E-NiCo-LDH) was obtained via an acid-base reaction. The morphology, composition and structure were characterized by transmission electron microscopy and X-ray diffraction. Electrochemical measurements reveal that E-NiCo-LDH exhibits an excellent electrocatalytic activity toward H2O2 oxidation and the sensor based on E-NiCo-LDH is able to detect H2O2 in a linear range of 2.0 μM to 1.1 mM with a high sensitivity of 728.6 μA mM−1 cm−2 and a low detection limit of 0.2 μM (S/N = 3). This indicated that E-NiCo-LDH is a promising material for electrochemical sensing of H2O2.

Magnetic particles can be combined with carbon materials to prepare core-shell microwave absorbers and the proper core/shell weight ratio yields the optimal absorption effects. However, few studies have focused on the influence of core surface modification on the microwave absorption performance. In this work, e-Fe 3 O 4 @C composite microspheres are synthesized by surface etching of core Fe 3 O 4 microspheres in HCl followed by polymerization/carbonization. Core surface modification improves the microwave absorption capability of the e-Fe 3 O 4 @C microsphere films. The minimal reflection loss (RL min ) is −54.38 dB at 12.4 GHz for a film thickness of 2.6 mm and the maximum effective bandwidth is extended to 4.1 GHz (3 mm). Etching of the Fe 3 O 4 core surface increases the interfacial contact area consequently enhancing interface polarization and defects created by surface etching produce additional dipole polarization to further increase the dielectric loss. The underlying mechanism is investigated and described. The results reveal a novel and effective strategy to enhance the properties of core-shell microwave absorption materials.

Hydrogels have attracted significant attention in various fields, such as smart sensing, human–machine interaction, and biomedicines, due to their excellent flexibility and versatility. However, current hydrogel electronic skins are still limited in stretchability, and their sensing functionality is often single-purpose, making it difficult to meet the requirements of complex environments and multitasking. In this study, we developed an MXene nanoplatelet and phytic acid-coreinforced poly(vinyl alcohol) (PVA) composite, denoted as MXene–PA–PVA. The strong hydrogen bonds formed by the interaction of the different components and the enhancement of chain entanglement result in a significant improvement in the mechanical properties of the PVA/PA/MXene composite hydrogel. This improvement is reflected in an increase of 271.43% in the maximum tensile strain and 35.29% in the maximum fracture stress. Moreover, the composite hydrogel exhibits excellent adhesion, water retention, heat resistance, and conductivity properties. The PVA/PA composite material combined with MXene demonstrates great potential for use as multifunctional sensors for strain and temperature detection with a strain-sensing sensitivity of 3.23 and a resistance temperature coefficient of 8.67. By leveraging the multifunctional characteristics of this composite hydrogel, electronic skin can accurately monitor human behavior and physiological reactions. This advancement opens up new possibilities for flexible electronic devices and human–machine interactions in the future.

Glyphosate is the most widely used herbicide in the world. There is still no complete clarity about the degree of its genotoxicity and mutagenicity . In addition, its effect on bacterial biofilms, the main life form of soil microbial communities , has not been adequately studied. Toxicity and mutagenicity, as well as changes in the bacterial biofilm biomass, physiological activity, and the number of living cells in its composition in the presence of glyphosate were assessed using the Escherichia coli model. To assess damage to cellular components under the action of this pesticide, luminescent whole-cell bacterial lux-biosensors were used. Changes in the level of mutagenesis were studied by the method of rifampicin mutants. High integral toxicity of glyphosate, the average level of increased oxidative stress and protein damage were shown with the help of bacterial biosensors . All the studied concentrations of the pesticide completely or partially suppress the matrix and structure of the E. coli CDC F-50 biofilm formation, as well as the bacterial cells metabolic activity in the biofilm. At the concentrations of 6.7 and 0.67 g/L, glyphosate suppresses mutagenesis, probably due to general suppression of metabolism, and at the concentration of 0.0067 g/L, it enhances mutagenesis by six times compared with the spontaneous level. Suppression of bacterial biofilms formation, toxic effects on microorganisms , and mutagenesis enhancement by glyphosate can lead to negative consequences for natural microbiomes.

Electronic skins (e-skins) have shown great potential in “bionic-machine” technologies. The development of bio-compatible e-skins that mimic the functions of the human skin, which is sensitive to multiple external stimuli, remains a challenge and a highly pursued goal. In this work, we report the synthesis, characterization, and performance-optimization of a thermo-sensitive and conductive hydrogel that displays multimodal sensing capacity. The optimal composition of a hydrogel with a volume phase transition temperature (VPTT) of 38 °C and a transition width of 15 °C was developed. This gel has an elastic modulus similar to that of the human skin, a large stretchability of 1900%, and a high gauge factor of 9.8. The gel is also puncture-resistant, self-healable, and self-adhesive. The as-fabricated hydrogel can sense multimodal stimuli, including force and temperature, with high sensitivity. We demonstrate that the gel possesses important attributes as a candidate for e-skin to monitor physiological signals including temperature, bending, and pressure in real time.

Wet-spinning technology plays an essential role in the development of wearable electronics. Poly(3,4-ethylene dioxythiophene): poly(styrene sulfonate) (PEDOT: PSS) fibers have been widely investigated in this field due to their advantages of being environment-friendly and low-cost. They are mainly wet-spun from a coagulation bath containing ethanol and water with additional metal salts. However, the conductivity of PEDOT: PSS fibers spun by this method is low. Here, we propose a novel coagulation bath made of ionic liquid (IL), ethanol, and water, and the resultant PEDOT: PSS fibers could achieve a conductivity of 118.3 S·cm −1 . Furthermore, the fiber-shaped supercapacitor prepared by such PEDOT: PSS/IL fibers reveals an optimal capacitance retention rate of 97 % even after 10,000 cycles, and the capacitance loss of the supercapacitor is only 7 % at −20 ℃. The sensor derived from the PEDOT: PSS/IL fibers has superb sensing capability, indicating their promising applications for wearable electronics.

Accurate and rapid detection of bacteria in complex environmental samples using simple and portable devices is still a major challenge. This research presents a simple nanofiber-based platform for highly sensitive colorimetric/fluorometric detection of Escherichia coli ( E. coli ). Nanofiber membranes (NFM) were loaded with target molecules (fluorescent and chromogenic substrate) via chemical modification to prepare functional NFM (NFM-MUG and NFM-XG). The β -glucuronidase secreted by E. coli during the metabolic process triggered the functionalized NFM to produce biological signals and color change. The intensity of bio-signals and color was shown to enable quantitative and qualitative detection of E. coli . The nanofiber-based platforms exhibited high stability and a wide detection range (10 2 -10 7 CFU/mL). The limit of detection (LOD) of NFM-MUG and NFM-XG sensors for E. coli were 26 and 69 CFU mL −1 , respectively. The sensing time required for NFM-MUG and NFM-XG was 15 mins and 30 mins respectively. The nanofiber-based platforms also exhibited high specificity and low interference from ionic compounds and pH changes. Notably, this assay was easily combined with a smartphone app as a portable device for on-site detection of E. coli , showing a great promise in the field of environmental and food safety testing.

Aims Osteoarthritis (OA) is a chronic inflammatory disease, characterized by degradation of extracellular matrix, apoptosis of chondrocytes and inflammation in articular cartilage. Zinc finger E-box binding homebox 2 (ZEB2), a transcription repressor, has been demonstrated with anti-inflammatory role in some cells. The analysis from GEO data reveals that ZEB2 expression is upregulated in articular cartilage of OA patients and experimental OA rodents. This study aims to confirm the function of ZEB2 in OA process.Material and methods The experimental OA was induced by anterior cruciate ligament transaction (ACLT) in rats, and the adenovirus loaded with ZEB2 coding sequence was intra-articularly injected into rats (1 × 10 PFU). The primary articular chondrocytes were stimulated by interleukin-1β (IL-1β) (10 ng/ml) to mimic the osteoarthritic injury, and transfected with the adenovirus carrying ZEB2 coding or silencing sequence. The apoptosis, content of extracellular matrix, inflammation, and the activity of NFκB signaling in chondrocytes and cartilage were determined. Results ZEB2 was highly expressed in osteoarthritic cartilage tissues and IL-1β-treated chondrocytes. The overexpression of ZEB2 restrained ACLT- or IL-1β administration-induced apoptosis, matrix degradation and inflammation in vivo or in vitro, evidenced by changed levels of cleaved caspase-3/PARP, collagen-II, aggrecan, matrix metalloproteinase 3/13, tumor necrosis factor-α and interleukin-6. Additionally, the phosphorylation of NFκB p65, IκBα and IKKα/β, and the nuclear translocation of p65 was blocked by ZEB2, implying inactivation of this signaling. Conclusions ZEB2 mitigated osteoarthritic symptoms in rats and chondrocytes, and NFκB signaling might be involved. These findings may provide novel insights for clinical treatment of OA.

Polymyxin E (colistin) has been considered as an important peptide antibiotic for clinical treatment of Gram-negative pathogens by selectively targeting lipopolysaccharide on the outer membrane. Recent reports showed that colistin can also kill Gram-positive bacteria, but the mechanism of colistin against Gram-positive bacteria remains unclear. In this study, we found that colistin can kill its producer Paenibacillus polymyxa , a Gram-positive bacterium, by both disrupting its cell integrity and inducing oxidative stress. Enhancement of the cell permeability is the major action of colistin against P. polymyxa . In relative, colistin-induced oxidative stress plays an associated role in killing of P. polymyxa . Notably, we, for the first time, found that exogenous catalase is able to intensify the colistin-induced killing by inhibiting the intracellular catalase activity and thus increasing oxidative stress. Our findings would shed light on a not yet clearly described bactericidal mechanism of colistin against Gram-positive bacteria. More importantly, our findings pave the road for the future to lower clinical use of colistin for avoiding bacterial resistance.

A new multifunctional emulsifier was synthesized by coupling maltodextrin with a dextrose equivalent of 19 to vitamin E succinate. Two emulsifiers with varying degrees of vitamin E succinate substitution were prepared based on different mass ratios of vitamin E succinate to maltodextrin. The molecular structure and purity of these emulsifiers were analyzed. Nanoemulsions were prepared using octenyl succinic anhydride modified starch as a control to investigate the physical stability, antioxidant capacity, oxidative stability, and in vitro simulated digestive properties of the nanoemulsions. The emulsifying and antioxidant activity of the maltodextrin-vitamin E succinate conjugate was significantly superior to that of octenyl succinic anhydride modified starch, demonstrating good physical and oxidative stability. Additionally, they were rapidly digested under simulated small intestinal conditions. This new emulsifier shows broad application potential for the encapsulation, protection, and delivery of hydrophobic bioactive substances in the fields of medicine, food, and healthcare products.

Bacteria-based metabolic therapy has been acknowledged as a promising strategy for tumor treatment. However, the insufficient efficiency of wild-type bacteria severely restricts their therapeutic efficacy. Here, we elaborately develop an ʟ-cyst(e)ine-addicted bacteria-nanodrug biohybrid for metabolic therapy through a dual-selection directed evolution strategy. Our evolved strain exhibits a 36-fold increase in ʟ-cystine uptake and a 23-fold improvement in total activity of cysteine desulfhydrases compared with the wild-type strain. By conjugating with DMXAA-loaded liposomes, the engineered bacteria-nanodrug biohybrid not only prevents the influx of nutrients into the tumor by blocking neovasculature but also achieves efficient and durable CySS catabolism locally. The unavailable of Cys species disrupts redox homeostasis and strikingly increases intracellular ROS level, achieving favorable therapeutic outcomes in multiple tumor models. Our study not only highlights the promise of directed evolution strategy in enhancing the stability and efficiency of bacteria-based living biocatalyst but also provides new opportunities for antitumor metabolic therapy.
