Adenosine triphosphate (ATP) is a central mediator of cancer cell metabolism and signaling, driving processes such as proliferation, invasion, metastasis, and therapeutic resistance. Current ATP imaging modalities-including fluorescence, chemiluminescence/bioluminescence, photoacoustic imaging, and magnetic resonance imaging (MRI)-face inherent trade-offs among sensitivity, spatial resolution, and tissue penetration capability. Here, a dual-mode probe is reported that synergistically couples the anatomical resolution of MRI with the high-sensitive, long-lasting signals of afterglow imaging for visualization of ATP in the tumor microenvironment. The probe is constructed via DNA hybridization between an afterglow unit and a T 1 -weighted MRI contrast agent, with an ATP-specific aptamer mediating probe disassembly upon ATP binding. This design generates simultaneous turn-on afterglow and MRI signals, enabling the imaging of intracellular and extracellular ATP fluctuations. Metabolic modulators-coenzyme Q10 (an ATP activator) and 2-deoxy-D-glucose (2-DG, a glycolysis inhibitor)-are applied to demonstrate dynamic, bidirectional tracking of ATP levels in vivo. Furthermore, by monitoring ATP depletion during synergistic metabolic therapy (cisplatin + 2-DG), a correlation between imaging readouts and therapeutic efficacy is established. The work provides anintegrated MRI/afterglow imaging platform for high-contrast, real-time ATP sensing in cancer, offering a powerful tool for elucidating tumor metabolism and evaluating metabolic interventions.

Increasing evidence has demonstrated that lactate and adenosine triphosphate (ATP) both play important roles in regulating abnormal metabolism in the tumor microenvironment. Herein, an O 2 self-supplying catalytic nanoagent, based on tannic acid (TA)–Fe(III) coordination complexes-coated perfluorooctyl bromide (PFOB) nanodroplets with lactate oxidases (LOX) loading (PFOB@TA–Fe(III)–LOX, PTFL), is designed for cascade metabolic-chemodynamic therapy (CDT) by dual-depletion of lactate and ATP with hydroxyl • OH radicals generation. Benefiting from the catalytic property of loaded LOX and O 2 self-supplying of PFOB nanodroplets, PTFL nanoparticles (NPs) efficiently deplete tumoral lactate for down-regulation of vascular endothelial growth factor expression and supplement the insufficient endogenous H 2 O 2 . Simultaneously, TA–Fe(III) complexes release Fe(III) ions and TA in response to intracellular up-regulated ATP in tumor cells followed by TA-mediated Fe(III)/Fe(II) conversion, leading to the depletion of energy source ATP and the generation of cytotoxic • OH radicals from H 2 O 2 . Moreover, TA–Fe(III) complexes provide photoacoustic contrast as imaging guidance to enhance therapeutic accuracy. As a result, PTFL NPs efficiently accumulate in tumors for suppression of tumor growth and show evidence of anti-angiogenesis and anti-metastasis effects. This multifunctional nanoagent may provide new insight for targeting abnormal tumor metabolism with the combination of CDT to achieve a synergistic therapeutic effect.

Energy levels in organisms are closely linked to metabolic diseases, with ATP—the universal energy currency—playing a central role in this relationship. This paper presents a novel electrochemical biosensor for ATP, integrating homogeneous bio-recognition reactions with terminal deoxynucleotidyl transferase (TdT)-mediated polymerization to enable precise ATP detection in metabolic research. Initially, a DNA oligonucleotide composed of a camouflaged aptamer and exposed hairpin DNA was designed to react homogeneously with ATP in the presence of exonuclease I (Exo I). This reaction generates blunt-ended hairpin DNA, which can activate the assembly of an ordered DNA-rich nanostructure layer on the gold electrode. Subsequently, the DNA strand is elongated through TdT-induced polymerization in the presence of deoxynucleotide triphosphates (dNTPs). A significant amount of Ru(NH 3 ) 6 3+ can be adsorbed onto the negatively charged DNA phosphate backbone via strong electrostatic interactions, effectively amplifying the detection signal and enhancing the sensor's performance. Under optimal conditions, the system exhibits excellent linearity over the target concentration range of 10 −2 to 10 3 nM, with a detection limit as low as 8.37 pM, enabling the detection of trace amounts of ATP. The biosensor demonstrates high stability and reliability, with recovery rates ranging from 97.8 % to 110 %. This method serves as a diagnostic tool to assess organismal metabolic status by monitoring ATP levels, enabling the tracking of metabolic disease progression. By integrating molecular monitoring with clinical diagnosis, it offers a potential approach for the preventing and intervention of metabolic diseases.

Cancer neuroscience has revealed the pivotal role of neural pathways in tumor progression and cancer-associated pain, particularly in bone metastases. To address both tumor growth and pain, we developed a novel nanomedicine strategy that targets mitochondrial dysfunction in cancer cells. The engineered H-MnCa/3MA-ALD nanoparticle system combines hollow mesoporous manganese-calcium (H-MnCa) nanoparticles functionalized with alendronate (ALD) for bone targeting, and loaded with 3-Methyladenine (3MA) to inhibit autophagy. This system disrupts mitochondrial ATP production, exploiting the inherent vulnerability of mitochondria to oxidative stress, and enhances reactive oxygen species (ROS) generation. The elevated ROS selectively induces cancer cell death while also reducing the secretion of nerve growth factor (NGF). This dual-action mechanism not only inhibits tumor growth and metastasis but also alleviates cancer-induced bone pain by downregulating neuro-pain mediators such as p-STAT-3 and NGF. This approach overcomes traditional chemodynamic therapy (CDT) limitations, ensuring targeted mitochondrial damage and apoptosis while preventing protective mitophagy. In vivo studies further confirm that H-MnCa/3MA-ALD provides effective tumor eradication and pain relief, offering a comprehensive solution for both cancer progression and pain management.

Exosomes are extracellular vesicles that inherit molecular markers from their parent cells for the transfer and delivery of membrane and cytosolic molecules between cells. Exosome detection and protein profiling provide noninvasive access to disease diagnosis and treatment. The nanozyme designed in this paper contains polydopamine-functionalization of graphene oxide (rGO@PDA) units that can adsorb ssDNA aptamers and CeO 2 metal oxide nanosheet units with peroxidase activity. As auxiliary regulator, adenosine triphosphate (ATP) enhances peroxidase activity of rGO@PDA@CeO 2 under a wide range of pH (4–7.4), which breaks through the limitation that traditional nanozymes can only exhibit peroxidase activity under acidic conditions. The ssDNA aptamers, which are recognition probes of exosome, can be adsorbed on the surface of rGO@PDA by aromatic stacking, competing with ATP molecules. Thus, the ssDNA aptamer acts as the switch for colorimetric sensor to flexibly regulate the peroxidase activity of the rGO@PDA@CeO 2 under physiological conditions , and realizes sensitive detection of exosomes. This assay achieves a detection linear range of 0.46 × 10 7 –10 8 particle/mL and a limit down to 3.81 × 10 5 particles/mL. In addition, this aptasensor sensitively profiles six exosomal protein across five cell types and clinical samples, suggesting their potential in clinic diagnosis.

The use of gaseous and air-captured CO 2 for technical biosynthesis is highly desired, but elusive so far due to several obstacles including high energy (ATP, NADPH) demand, low thermodynamic driving force and limited biosynthesis rate. Here, we present an ATP and NAD(P)H-free chemoenzymatic system for amino acid and pyruvate biosynthesis by coupling methanol with CO 2 . It relies on a re-engineered glycine cleavage system with the NAD(P)H-dependent L protein replaced by biocompatible chemical reduction of protein H with dithiothreitol. The latter provides a higher thermodynamic driving force, determines the reaction direction, and avoids protein polymerization of the rate-limiting enzyme carboxylase. Engineering of H protein to effectively release the lipoamide arm from a protected state further enhanced the system performance, achieving the synthesis of glycine, serine and pyruvate at g/L level from methanol and air-captured CO 2 . This work opens up the door for biosynthesis of amino acids and derived products from air.

Synovial hyperplasia and inflammation are the main pathological features of rheumatoid arthritis (RA). Fibroblast‑like synoviocytes (FLSs) are the major effector cells contributing to chronic inflation and joint injury in the synovial microenvironment. Neutrophil extracellular traps (NETs) play an important role in the pathogenesis of RA, but their downstream regulatory mechanisms remain unclear. In order to improve the therapeutic prospects for RA, it is crucial to identify the targets of NETs and the molecular mechanisms of synovial dysfunction. ATP-citrate lyase (ACLY) directs glucose metabolism to de novo lipogenesis (DNL) by generating acetyl-CoA, which is also an acetyl donator to protein acetylation. ACLY has gained attention in studies on tumors, fatty liver, inflammation, and metabolic diseases. However, its involvement in RA progression is still uncertain. The present study revealed increased expression of NETs in the RA synovial microenvironment, including synovial fluid and synovial tissue, and a positive correlation to disease activity. In vitro experiments demonstrated that NETs activate ACLY, which not only triggers DNL, and enhances procreation, migration and invasiveness of RA-FLSs, but also stimulates the nuclear factor kappa-B (NF-κB) signaling pathway. This enhances the expression and nuclear translocation of acetyl-NF-kappaB p65 (Ac-p65), intensifying the transcription of inflammatory mediators and exacerbating synovial inflammation. Additionally, a high level of NETs expression in synovial tissues of arthritis animal models and the therapeutic effect of inhibiting ACLY on joint inflammation, bone erosion and bone destruction were also confirmed in vivo. In conclusion, our results elucidate the molecular mechanisms involved in the activation of RA-FLSs by NETs, and ACLY may be a candidate target for regulating metabolic reprogramming and inflammation to mitigate RA joint injuries.

Adenosine triphosphate (ATP) is crucial for cellular activity. The need for ATP detection in the field of biomedicine is on the rapid rise. Several biosensor-based approaches have been developed as a result of the growing demand for ATP detection. An electrolyte-insulator-semiconductor (EIS) sensor is a type of field-effect device that has the ability to detect surface-potential changes with a specific level of sensitivity. In this study, a label-free ATP detection biosensor that is based on poly(amidoamine)-modified EIS sensors was developed, in which ATP-sensitive aptamer (Apt) was utilized as sensitive element and EIS sensor was used as transducer. It is possible to monitor the binding of charged molecules, such as aptamers using EIS sensors in a label-free manner with a straightforward setup. To improve the coupling efficiency of Apt with EIS sensor, positively charged polyelectrolyte, i.e. poly (amidoamine) (PAMAM) dendrimers, was utilized to modify the surface of the EIS sensors to attach the negatively charged Apt based on electrostatic attraction. The adsorptive binding of Apt and ATP results in a change in capacitance of EIS sensor. During the process of surface modification, the electrochemical measurements. of capacitance-voltage (C-V) curves and constant-capacitance (ConCap) characteristics were utilized as indicators for the corresponding processes of EIS sensor surface modification. The measurements results indicated this biosensor was able to detect ATP with high sensitivity and good specificity. The detection range of ATP was 0.1. nM to 100. nM and the detection limit was as low as 0.12 nM. This biosensor has the potential to be utilized in the detection of ATP in the surrounding microenvironment of cells and tissues, which have promising prospects to be applied in the field of biomedicine such as energy and metabolism monitoring of cells and tissues.

Enlightened by the great success of drug repurposing strategy in pharmaceutic industries, in the current study, material repurposing was proposed where the performance of carbonyl iron powder (CIP), a nutritional intervention agent of iron supplement approved by the US FDA for iron deficiency anemia in clinical was explored in anti-cancer treatment. Except for the abnormal iron metabolic characteristics of tumors that serving as potential targets for CIP-based cancer therapy under repurposing paradigm, the efficacy of CIP as catalyst in Fenton reaction, activator for dihydroartemisinin (DHA) and thus increase the chemo-sensitivity of tumors, as well as the potent agent for NIR-II photothermal therapy (PTT) was fully evaluated by an injectable alginate hydrogel form. The CIP-ALG gel could cause rapid temperature rise in the tumor site under NIR-II laser irradiation, leading to complete ablation in the primary tumor. Further on, such photothermal-ablation would lead to ATP release significantly and in the bilateral tumor model, both primary tumor ablation and inhibition of secondary tumor were observed simultaneously under synergistic tumor treatment of nutritional-photothermal therapy (NT/PTT). Material repurposing could thus be confirmed by our pioneering trial and the CIP-ALG meditated NT/PTT/immunotherapy would provide new choice of strategy for safe and efficient tumor therapy.

Adenosine triphosphate (ATP) biomolecules play critial roles in the biomineralization process during the formation of amorphous calcium phosphate composites (ACPC), and ACPC is an important drug carrier due to its significant advantages of biocompatibility and biodegradability. Hence, studying the behavior of ACPC nanodrug carriers is crucial to investigate the structural regulation of biomimetic minerals and calcium phosphate (CaP)-based drug delivery systems. However, it is difficult to probe these interactions using traditional characterization methods. In this paper, XANES analysis together with STXM successfully provided a method to reveal the interaction of ATP and drug molecules with individual mesoporous ACPC. We found that the adenosine and phosphate groups of ATP biomolecules coordinated with Ca2+ and played critical roles in the formation of ACPC; drug molecules with the –COOH groups were linked to Ca2+via carboxylic acid groups primarily by electrostatic interactions, and the N-containing ring structures within the drug molecules also coordinated with Ca2+.

Herein, a highly sensitive and selective ratiometric fluorescence platform was constructed for probing adenosine triphosphate (ATP) for the first time by using magnetic Fe 3 O 4 nanozymes that were modified by ATP-specific aptamer labeled with 7-Amino-4-methyl-3-coumarinylacetic acid (Apt-AMCA) to yield Fe 3 O 4 @Apt-AMCA. It was discovered that the incorporation of Apt-AMCA greatly enhance the peroxidase-like activity of Fe 3 O 4 nanozymes, which might catalyze more o -phenylenediamine (OPD) substrates into fluorescent oxidized OPD (DAP) with a emission of 560 nm. More importantly, upon ATP addition, Apt-AMCA could potentially be released from Fe 3 O 4 @Apt-AMCA because of Apt-target binding and the competition between ATP and Apt. The rational increase in the levels of free Apt-AMCA could thereby be attained in the reaction solutions so as to induce an enhancement of fluorescence at 450 nm. Meanwhile, the desorption of Apt-AMCA from the Fe 3 O 4 @Apt-AMCA surface would decrease the peroxidase-like activity of Fe 3 O 4 @Apt-AMCA, thereby reducing its ability to catalyze OPD into fluorescent DAP and thus decreasing fluorescence at 560 nm. A ratiometric fluorescence method was accordingly formulated to permit the sensitive and selective identification of ATP, featuring a linear dynamic range that ranges from 0.10 μM to 80 μM. Accurate measurements of ATP in practical blood samples were also confirmed, promising a new detection method for ATP analysis that can widely applied in clinical laboratories.

Specific delivery of NCEH1 plasmid is a promising approach to boost the cholesterol removal from lipid-laden macrophages for antiatherosclerosis. Polyethylenimine (PEI) is one of the most efficient gene carriers among nonviral vectors. However, the high transfection activity of PEI is always accompanied by profound cytotoxicity. To tackle the paradox between transfection efficiency and safety, we constructed a novel ATP-responsive multifunctional supramolecular polymer by cross-linking functionalized low-molecular-weight PEI via a boronic ester bond for NCEH1 plasmid delivery. The supramolecular polymer could condense NCEH1 plasmids to form stable nanosized polyplexes when the w/w ratios of the polymer and gene were higher than 2. ATP-triggered degradation of the polymer and pDNA release were characterized by a series of studies, including 1H NMR, 31P NMR, XPS, agarose gel electrophoresis, and ethidium bromide exclusion tests. In addition, the changes in particle size and morphology were observed in the presence of ATP. Interestingly, the supramolecular polymer showed broad spectrum antioxidant activities by measuring the elimination rates of different reactive oxygen species. In addition, the supramolecular polymer displayed a high buffering capability and good cytocompatibility as demonstrated by the results of the buffering capacity, a hemolysis assay, and a cytotoxicity test. Importantly, it was revealed that the supramolecular polymer/NCEH1 plasmid polyplex formulated at a w/w ratio of 20 was most effective in enhancing cholesterol removal from lipid-laden macrophages and reducing the accumulation of lipid droplets as evidenced by transfection study, cholesterol efflux assay, and oil red O staining studies. Collectively, the ATP-responsive multifunctional supramolecular polymer holds great potential for safe and efficient gene delivery for antiatherosclerosis.
