In this paper, we propose a composite nano-microstructure comprising Ti3C2 and nanogold as a surface-enhanced Raman spectroscopy (SERS)-active substrate for the highly sensitive sensing of amine gases. The adsorption performance of Ti3C2 was tested by a quartz crystal microbalance (QCM) sensor. Excellent adsorption ability of Ti3C2 was observed for aniline, phenylenediamine, and diethylamine, with adsorption amounts exceeding 2210, 1740, and 970 ng cm−2, respectively. The synergistic effect of the electromagnetic properties of Ti3C2 and nano-Au in the Ti3C2/nano-Au composite nanostructures was verified by finite-difference time-domain simulation. Partial chemical enhancement between Ti3C2 and aniline was verified by photocurrent tests. To achieve assay specificity, mercaptoacetaldehyde (MA) was adopted as a recognition probe. The Schiff base reaction between aldehyde groups and amino groups produces the –CN– bond, with the Raman characteristic peak at 1635 cm−1, enabling precise amine identification with the SERS chip. To obtain the Ti3C2/nano-Au@MA SERS sensing chip, nano Au was deposited on an indium tin oxide substrate using the chronoamperometric method, followed by nano-Au aldolization modification through Au-SH interactions and the spin-coating of a Ti3C2 film layer. Due to the high affinity of Ti3C2 for amine molecules and the low SERS background signal of MA and Ti3C2, the SERS spectra could be used for ultra-sensitive detection of amines. Three amine gases, i.e. aniline, p-phenylenediamine, and diethylamine, were detected quantitatively using the intensity ratiometric method on the functionalized Ti3C2/nano-Au@MA SERS sensor chip. The detection limits were 1, 1, and 20 parts per billion, respectively. By identifying the characteristic fingerprint peaks and comparing the relative intensity of the –CN– characteristic peak, quantitative analysis of the components of aniline and p-phenylenediamine mixed gas was accomplished. These findings demonstrate that the Ti3C2/nano-Au@MA SERS sensor chip offers a promising approach for the detection of amine gas molecules and holds potential for applications in environmental monitoring, disease diagnosis, and chemical analysis.

Numerous chemical reactions and most life processes occur in aqueous solutions, where the physical diffusion of small molecules plays a vital role, including solvent water molecules, solute biomolecules, and ions. Conventional methods of measuring diffusion coefficients are often limited by technical complexity, large sample consumption, or significant time cost. Here, we present an optical imaging method to study molecular diffusion by combining stimulated Raman scattering (SRS) microscopy with microfluidics: a “Y”-shaped microfluidic channel forming two laminar flows with a stable concentration gradient across the interface. SRS imaging of a specific molecule allows us to obtain a high-resolution chemical profile of the diffusion region at varying inspection locations and flow rates, which enables the extraction of diffusion coefficients using the convection–diffusion model. As a proof of concept, we measured diffusion coefficients of molecules including water, protein, and multiple ions, with a sample volume of less than 1 mL and a time cost of less than 10 min. Moreover, we demonstrated a high-resolution three-dimensional (3D) reconstruction of the diffusion patterns in the microfluidic channel. The high-speed microfluidic SRS platform holds the potential for quantitative measurements of molecular diffusion, chemical reaction, and fluidic dynamics at the liquid–liquid interfaces.

Recurrent implantation failure is a critical bottleneck that limits the clinical success rates of assisted reproductive technologies. Existing in vitro implantation models often struggle to balance the conflicting needs of "fluid supply" and "embryo protection": static models lack necessary hydrodynamic stimulation, whereas traditional microfluidic perfusion frequently causes blastocyst damage due to excessive shear stress. Furthermore, most existing scaffold materials fail to mimic the critical dynamic mechanical remodeling of the endometrium during the implantation window. To overcome these limitations, this study constructed a biomimetic endometrium-on-a-chip based on methacryloyl gelatin (GelMA) hydrogels and micropillar arrays, aiming to reconstruct an implantation mechanical microenvironment that closely mimics in vivo conditions. In terms of engineering design, we optimized the micropillar array structure via COMSOL Multiphysics simulation to achieve spatial decoupling of shear stress. This design created a "mechanical sanctuary" for the embryo with shear stress lower than 0.3 dyn/cm2, effectively preventing fluid shear-induced damage while ensuring dynamic nutrient exchange. Regarding the material strategy, this study revealed the concentration-dependent degradation kinetics of GelMA hydrogels. Leveraging their "fast-then-slow" degradation behavior, we recapitulated the mechanical transition of the endometrium, aligning our matrix's softening profile with the physiological shift toward the receptive state characteristic of the implantation window. Biological experiments demonstrated that the GelMA-based hydrogel significantly enhanced the adhesion and zona hatching rates of mouse blastocysts compared to traditional static cultures. Furthermore, by integrating this biomimetic scaffold into a dynamically perfused microfluidic platform, we successfully established a robust in vitro model of mouse embryo implantation.

Oocyte cryopreservation represents a cornerstone technology in assisted reproductive medicine and biodiversity conservation. In this field, microfluidic chips have emerged as promising tools for minimizing osmotic shock and other damage through programmed and automated addition of cryoprotectants (CPAs). Nevertheless, existing microfluidic platforms still face significant challenges in inefficient cell loading/retrieval and operational complexity, which compromise the efficacy of oocyte cryopreservation, hindering their development and application. Here, we developed a novel microfluidic chip and combined it with magnetic hydrogel microencapsulation technology, to establish a comprehensive strategy for optimizing oocyte cryopreservation. Unlike previous microfluidic systems hindered by fluid shear stress or complex capture designs, our platform incorporates a quick-sealing cover for efficient retrieval and magnetic hydrogel immobilization, effectively preventing cell drift and loss without requiring intricate microchannel structures. This innovation not only simplifies operation but also enhances reproducibility. Furthermore, the magnetic hydrogel enables external magnetic control of oocytes in the microchannel, avoid the complex capture structure design of microchannels and provide physical shielding for oocytes during cryopreservation. Compared with conventional straw vitrification, this approach significantly improved oocyte survival rate (86.63% (n = 53) vs. 57.47% (n = 73)) and better preserved mitochondrial activity, cytoskeleton structure, and embryonic development potential. This work presents a combined cryopreservation method that marries high-efficiency protection with operational simplicity, offering a new scheme for oocyte cryopreservation with significant translational potential.

Emerging evidence underscores biophysical characteristics of cancer cells as key modulators of cancer progression and metastasis. Herein, we reported a cell-mechanophenotyping screening microfluidic chip (termed LesM) for the high-efficient capture of circulating tumor cells (CTCs) and evaluation of single-cell deformation to reveal the hematogenous metastatic potential of bacteria-infected breast cancer. LesM employs L-shaped traps to capture single cells, leveraging bacteria-infected CTCs with cytoskeletal reorganization traverse narrowed channels while rigid native cells are retained. The platform demonstrates an average single-cell capture efficiency of 95.42% and specificity of 85.34% in discriminating infected versus non-infected breast cancer cells, validated through parallel in vivo metastatic assays. LesM enables high-throughput sensing up to 10,240 cells of mechanical signatures and microbial cargo, correlating with metastatic risk and antibiotic response. By bridging biomechanics and intratumoral microbiota detection, LesM offers a transformative liquid biopsy tool for predicting distant metastasis and guiding antimicrobial therapies in bacteria-infected breast cancers.

In gout progression, uric acid (UA) accumulation triggers crystal deposition, while interleukin-6 (IL-6) drives the subsequent inflammatory flares. Therefore, monitoring both UA and IL-6 is important for gout management. In this study, for the first time a microfluidic electrochemical system (MES) has been developed to simultaneously detect UA and IL-6 levels in blood. The MES consists of a multilayer microfluidic chip that incorporates a plasma separation module and two screen-printed electrochemical (SPE) sensors, integrated with a wireless potentiostat for real-time data analysis. The working electrode (WE) of one SPE was modified with uricase and hydrophilic fumed nano-silica (FSiO₂) for UA sensing, and the WE of the other SPE was modified with gold nanoparticle-reduced graphene oxide (Au NPs/rGO) composites and antibodies for IL-6 sensing. The MES demonstrated excellent analytical performance for the simultaneous detection of UA and IL-6 in human blood, featuring high selectivity, broad linear ranges (10-1000 µM for UA and 0.001-100 ng/mL for IL-6), robust stability, and strong concordance with reference methods (R² > 0.95). This study provides a proof-of-concept platform for gout-related biomarker determination and offers a new strategy for the development of multi-biomarker electroanalytical devices.

In the course of extracorporeal circulation (ECC), real-time monitoring of blood electrolyte levels is crucial for ensuring patient safety and improving outcomes. A continuous detection chip of blood electrolytes potassium, sodium, and calcium is developed by integrating sensor array with a microchannel for real-time monitoring instead of traditional single-point detection. The primary issue to address for continuous blood monitoring is the nonspecific adhesion of plasma proteins and blood cells to the sensors and microchannel. To enhance the antifouling capability of the detection chip, chemical doping and surface modification are applied to both the sensors and the microchannel. These treatments significantly increase the surface hydrophilicity, thereby reducing the adhesion of proteins and platelets while preserving the potentiometric performance of the sensors. By further optimizing the storage conditions, the modified chips exhibit an extended operational lifespan and robust antifouling performance. This allows for reliable long-term detection in flowing blood without signal degradation due to biofouling. The proposed method thus provides a straightforward and effective route to construct continuous electrolyte detection chips compatible with ECC platforms.

Magnetic microrobots have emerged as transformative tools for biomedical applications, particularly in targeted cargo delivery within confined environments. Despite their potential, existing systems lack the capability for co-encapsulation of various incompatible components in isolated compartments. In this paper, we propose a magnetic compartmentalized microrobot (MCM) fabricated on-chip with hydrogel. Our approach leverages a microfluidic aqueous two-phase system to create hydrogel capsules with dual cores, where two core flows composed of dextran and shell flow containing sodium alginate and magnetic particles are sequentially sheared by oil phase, crosslinked by calcium ion, and magnetized via an electromagnet, to produce MCMs massively. The resulting architecture enables separate encapsulation of incompatible components within two cores isolated by a hydrogel shell. Precise control over flow rates allows tunable sizing of both core and capsule dimensions. Under rotating magnetic fields, MCMs demonstrate programmable individual and collective behaviors, including directional motion, dispersion, and aggregation. We further characterize the system’s drug-release kinetics and swelling properties in simulated gastrointestinal environments. These findings highlight MCMs’ significant potential for tissue engineering, combinatorial drug delivery, and transport of incompatible bioactive agents. Note to Practitioners—The motivation of this work is to develop a magnetic microrobot with multiple isolated compartments for co-encapsulation and target delivery of various incompatible components. We built a microfluidic aqueous two-phase system to create magnetic hydrogel capsules with dual cores. Physical isolation between cores enables co-encapsulation of incompatible therapeutic agents. Magnetic actuation permits controlled individual and collective behaviors. Fully biocompatible composition coupled with scalable on-chip high-throughput fabrication ensures clinical relevance. These unique characteristics position the MCMs as promising candidates for biomedical applications requiring contamination-free co-delivery of multiple therapeutic payloads.

Bioelectronic systems integrated with artificial intelligence (AI) are transforming neurochemical diagnostics, enabling intelligent, real-time decoding of brain chemistry. Here, we present an AI-driven biomimetic electrochemical chip with intestine-inspired wrinkled MoS2 electrodes, enabling dynamic in vivo dopamine monitoring and neuromodulation. This wrinkled structural design promotes efficient molecular capture and alleviates laminar flow limitations, achieving a 23-fold enhancement in dopamine sensitivity over planar electrodes, with detection limits as low as 37 nM in 4.1 μL of biofluid. Moreover, coupled with an AI-assisted decision module, the platform translates neurochemical fluctuations into adaptive modulation, enabling real-time closed-loop neuromodulation. In vivo studies in rats demonstrate autonomous regulation of endogenous dopamine transients, directly linking electrode architecture to functional neural outcomes. This work highlights how bioinspired electrode design, integrated with intelligent signal interpretation, establishes a scalable pathway toward next-generation neurodiagnostics and adaptive bioelectronic interfaces.

Accurate identification of volatile organic compounds (VOCs) in gas mixtures remains a significant challenge, especially for isomers with similar structures. We propose a metal-organic framework (MOF)-functionalized micro-ring resonator (MRR) array for butanol isomer recognition and mixture analysis. By growing HKUST-1 thin films on MRRs with diverse waveguide geometries using spin-assisted liquid-phase epitaxial layer-by-layer (SA-LPE-LbL) methods, the optimized racetrack-type MRR achieves 17.7 pm/ppm sensitivity and a calculated 0.41 ppm detection limit for 1-butanol. Additionally, the MRR array demonstrated reliable environmental tolerance under near-realistic humidity conditions (30–50% RH). While sensitivity slightly decreased, the ability to distinguish isomers remains effective. Molecular dynamics (MD) simulations reveal that steric hindrance governs the adsorption differences. Specifically, linear isomer access Cu 2 + sites more effectively in HKUST-1, leading to stronger binding and faster diffusion. Deep learning models analyze the multi-channel spectral data to achieve a 94.4% classification accuracy for the three isomers and effectively decompose gas mixture spectra with a per-wavelength-point (PWP) reconstruction error below 0.04. This platform significantly enhances sensitivity and selectivity for isomer differentiation and mixture analysis, offering a scalable solution for optical gas sensing.

Exosomes have gained increasing attention as robust, biocompatible carriers for targeted therapy. However, current techniques for exosome drug loading suffer from low drug loading efficiency, substantial exosome loss during repeated purification and quantification processes. Here, we present an integrated microfluidic chip (IMC) that streamlines drug loading, separation, and electrochemical detection of exosomes from plasma in a single device. In this design, the three-dimensional (3D) macroporous scaffold and the magnetoresponsive electrode are successfully assembled into the modeling microchip, playing the functions of “3D chaotic flow mixer”, “magnetic separator” and “electrochemical detector”. When plasma, doxorubicin (DOX), boron clusters and immunomagnetic nanoprobes (IMP) are simultaneously injected into the IMC, the exosomes are loaded with DOX-boron cluster (EDB) complexes and synchronously recognized by IMP in the “3D chaotic flow mixer”. Our strategy exhibits high DOX loading efficiency owing to the superchaotropic effect of boron cluster and enhanced immunolabeling efficiency by the thorough mixing of 3D scaffold. Meanwhile, the novel magnetoresponsive electrode enables magnetic separation and real-time, enzyme-linked immunoelectrochemical quantification of exosomes, thereby simplifying the workflow from drug loading to quantification. The resulting EDB in combination with magnetic hyperthermia achieves up to 90% cell-killing efficiency against DOX-resistant breast cancer cells. Overall, our system could simultaneously realize the enhanced DOX loading into exosomes, efficient magnetic immunoseparation of exosomes, and sensitive electrochemical quantification of exosomes, offering a promising approach for autologous exosome-based drug delivery for cancer treatment.

Circulating tumor cells (CTCs) captured from blood fluid represent recurrent cancers and metastatic lesions to monitor the situation of cancers. We develop surface-enhanced Raman scattering (SERS)-coding microsphere suspension chip as a new strategy for fast and efficient capture, recovery, and detection of targeting cancer cells. Using HeLa cells as model CTCs, we first utilize folate as a recognition molecule to be immobilized in magnetic composite microspheres for capturing HeLa cells and attaining high capturing efficacy (up to 95%). After capturing cells, the composite microsphere, which utilizes a disulfide bond as crosslinker in the polymer shell and as a spacer for linking folate, can recycle 90% cells within 20 min eluted by glutathion solution. Taking advantage of the SERS with fingerprint features, we characterize captured/recovered cells with the unique signal of report-molecule 4-aminothiophenol through introducing the SERS-coding microsphere suspension chip to CTCs. Finally, the exploratory experiment of sieving cells shows that the magnetic composite microspheres can selectively capture the HeLa cells from samples of mixed cells, indicating that these magnetic composite microspheres have potential in real blood samples for capturing CTCs.
