Cholesterol Targeted Catalytic Hydrogel Fueled by Tumor Debris can Enhance Microwave Ablation Therapy and Anti-Tumor Immune Response

Lin Shen,Zhijuan Yang,Yi Zhong,Yanran Bi,Junchao Yu,Qinwei Lu,Yanping Su,Xiaoxiao Chen,Zhongwei Zhao,Gaofeng Shu,Minjiang Chen,Liang Cheng,Liangzhu Feng,Chenying Lu,Zhuang Liu,Jiansong Ji

The immunosuppressive residual tumor microenvironment (IRTM) is a key factor in the high recurrence and metastasis rates of hepatocellular carcinoma (HCC) after microwave ablation (MWA). Cholesterol-rich tumor fragments significantly contribute to IRTM deterioration. This study developed a cholesterol-targeted catalytic hydrogel, DA-COD-OD-HCS, to enhance the synergy between MWA and immune checkpoint inhibitors (ICIs) for HCC treatment. Cholesterol oxidase (COD), modified with dimethyl maleic anhydride (DA) for release in acidic IRTM, is used to degrade cholesterol. Oxydextran (OD) and hemin-chitosan (HCS) formed a dual network gel, ensuring long-term fixation of COD and hemin in the IRTM post-MWA. In both in vitro and in vivo HCC models, DA-COD-OD-HCS effectively released COD, degraded cholesterol, and induced tumor cell ferroptosis, enhancing the antitumor immune response. Combined with anti-PD-L1 immunotherapy, this strategy inhibited primary tumor growth and distant metastases, without side effects on adjacent tissues. This work highlights that cholesterol-targeting catalytic hydrogels fueled by tumor debris can significantly improve the efficacy of MWA and ICIs, offering a novel therapeutic approach for HCC.

Surfactant-free porous nano-Mn3O4 as a recyclable Fenton-like reagent that can rapidly scavenge phenolics without H2O2

Zhenzhang Weng,Jie Li,Yali Weng,Miao Feng,Zanyong Zhuang,Yan Yu

Current methods that utilize Fenton-like reagents to remove persistent organic pollutants suffer from several limitations, i.e., the reagents may release metal ions into the sludge and cause secondary pollution, the reagents are easily inactivated after use, and the use of additives (e.g., 2KHSO5·KHSO4·K2SO4, Na2S2O8 and H2O2) can be costly but cannot be avoided. In this work, through a one-pot reaction, we prepared surfactant-free porous nano-Mn3O4 as an efficient Fenton-like reagent. Without the addition of external H2O2, it could remove 86.8% and 96.9% of the phenol from an aqueous solution within 1 and 5 min, respectively, and thus outperformed many other reported reagents. The underlying mechanism for the removal of organic pollutants by this Mn3O4 reagent was examined. It was found that the low-valence Mn in Mn3O4 reacted with the dissolved O2 to generate H2O2, and the H2O2 in turn created hydroxyl radicals (HO˙) to degrade organic pollutants. The Mn3O4 was also annealed to generate porous nano-Mn2O3, and a comparison of the performance of Mn3O4 and Mn2O3 in removing organic pollutants revealed that low-valence Mn in the oxides played a critical role in scavenging the pollutants. Note that this novel nano-Mn3O4 could remove pollutants without the addition of external H2O2, and it could be easily reactivated after use through reduction by NaBH4 and thus be recycled. The Mn3O4 can also be successfully applied to complex polluted water containing a mixture of phenolics. These findings pave the way for developing other efficient and environmentally friendly Fenton-like reagents for removing organic pollutants in wastewater.

Can adsorbent of layered chalcogenide be regenerated? A case study of norfloxacin adsorbed by layered chalcogenide in water

Jian-Rong Li,Lei Xu,Ming-Lai Fu,Huachun Lan

The layered chalcogenide has been shown to efficiently absorb organic pollutant, as well as heavy metals and radioactive ions. Previous studied showed that the regeneration of spent layered chalcogenide was impossible because of the strong affinity between soft metal ions and sulfur ions. While can adsorbent of layered chalcogenide be regenerated for adsorption of organic pollutant? In this study a case study of norfloxacin (NOR) adsorbed by layered chalcogenide in water is illustrated. The typical layered chalcogenide, K 1.9 Mn 0.95 Sn 2.05 S 6 (KMS-1) was hydrothermally synthesized and then used to remove NOR in aqueous solution. The adsorption process was found strongly pH-dependent. The maximum adsorption capacities were obtained at pH of 4 at different temperatures. The shift of Bragg peaks indicated from XRD patterns at different pH accompanying SEM images and adsorption amount showed that cation exchange played an essential role to remove NOR. The intercalation of cationic NOR took titled orientations of di-molecular NOR in KMS-1 with the interlayer space of 1.38   nm and 0.97   nm, respectively. The KMS-1 intercalated with NOR can be successfully regenerated by cation exchange using high concentration (1.0   M) KCl solution, in which the decrease in adsorption capacity was found <10% after three adsorption – desorption cycles. The obtained results suggested layered chalcogenide could be a promising regenerated adsorbent to remove organic pollutant with potential application in the environmental remediation.

The post-processing temperature or humidity can importantly control the secondary structure and characteristics of silk fibroin films

Hai-Yan Wang,Su-Fang Zhou,Meng Zhang,Heng-Da Wang,Yu-Qing Zhang

Temperature and humidity (TH) are highly important factors that can control the secondary structure and characterization of silk fibroin (SF) biomaterials. In this study, the water stability, secondary structure, mechanical properties, surface morphology, and degradation of silk fibroin films (SFFs) with post-processing in different TH were investigated. Fourier transform infrared indicated that the SFF secondary structure did not change under low-relative humidity (RH) despite temperatures up to 180°C, while it transformed at 40°C with 100% RH in 10 min. A film with a higher tensile strength (42.1 ± 8.2 MPa) could be obtained after post-processing at 90°C/100% RH for 10 min. While a film with higher ductility (elongation at break: 198.8 ± 31.8%) was generated after post-processing at 40°C/100% RH for 10 min. Scanning electron microscope showed that the film presented a network structure of nanoparticles in series under certain TH post-treatment. Enzymatic hydrolysis proved that the SFFs containing a higher content of silk II structure degraded more slowly. Therefore, TH post-treatment is a relatively mild way to change the secondary structure and properties of SFFs, which can be widely used in loading drugs and maintaining the activity of drugs in SF biomaterials.

Addition of Lactic Acid Bacteria Can Promote the Quality and Feeding Value of Broussonetia papyrifera (Paper Mulberry) Silage

Wen-Tao Sun,Yuan Huang,Chang-Rong Wu,Chao Peng,Yu-Long Zheng,Chao Chen,Jun Hao

In this study, the influence of two lactic acid bacteria (LAB) strains [Lactobacillus rhamnosusBDy (LR-BDy) andLactobacillus buchneriTSy (LB-TSy)] selected from Southwest China on the fermentation characteristics and in vitro gas production ofBroussonetia papyrifera(paper mulberry) silage were experimentally explored. The experimental groups were a control group (C), an LB-TSy treatment (LB), an LR-BDy treatment (LR), and an LR-BDy + LB-TSy hybrid group (LR × LB). After the LAB were added, the pH value of paper mulberry silage significantly declined (p< 0.05), and the crude protein content was effectively preserved (p< 0.05). However, no significant changes were found in the levels of neutral detergent fiber, acid detergent fiber, and crude ash (p> 0.05). The lactic acid content in paper mulberry silage was evidently increased (p< 0.05). The in vitro gas production in the LR at 36, 48, and 72 h were markedly higher than that in the other treatments (p< 0.05). Owing to the addition of LAB, the microbial diversity in paper mulberry silage was reduced, while the relative bacterial abundance ofLactobacilluswas enhanced. Hence, the addition of LAB selected from the warm and humid region in Southwest China can improve the quality of paper mulberry silage and elevate its feeding value in this region. Keywords:paper mulberry;silage;in vitro gas production;microbial community;lactic acid bacteria

Fertilization can modify the enantioselective persistence of penthiopyrad in relation to the co-influence on soil ecological health

Hui Liu,Mengqi Liu,Kuiyuan Chen,Mei Shan,Yongye Li

Penthiopyrad is a widely used chiral fungicide for controlling rust and Rhizoctonia diseases. Development of optically pure monomers is an important strategy to realize amount reduction and increment effects of penthiopyrad, wherein, fertilizers as the co-exiting nutrient supplement may alter the enantioselective residues of penthiopyrad in soil. In our study, influences of urea, phosphate, potash, NPK compound, organic granular, vermicompost and soya bean cake fertilizers on enantioselective persistence of penthiopyrad were fully evaluated. This study demonstrated that R -(−)-penthiopyrad dissipated faster than S -(+)-penthiopyrad during 120 days. High pH, available nitrogen, invertase activities and reduced available phosphorus, dehydrogenase , urease, catalase activities were situated to benefit removing the concentrations of penthiopyrad and weakening enantioselectivity in soil. With respect to the impact of different fertilizers on soil ecological indicators, vermicompost contributed to enhanced pH. Urea and compound fertilizer played an absolute advantage in promoting available nitrogen. All fertilizers didn't go against available phosphorus. Dehydrogenase responded negatively to phosphate, potash and organic fertilizers . Urea increased invertase, besides, it and compound fertilizer both diminished urease activity. The catalase activity was not activated by organic fertilizer. Based on all the findings, soil application of urea and phosphate fertilizers was recommended and considered as a better option to exhibit high efficiency for the dissipation of penthiopyrad. The combined environmental safety estimation can effectively guide the treatment of fertilization soils in line with the nutrition requirements and pollution regulation from penthiopyrad.

Antifungal mechanism of carvacrol and osthole can disrupt cell structure integrity and interfere with energy metabolism in Neopestalotiopsis ellipsospora

Huifang Liu,Jianmei Yao,Rongxiu Yin,Chiyu Ma,Lulu Li,Wen Yang,Zhiwei Lei

Introduction: Tea gray blight disease is a major leaf disease in tea plants, significantly reducing tea quality and yield. Plants are rich in bioactive compounds that are safe, non-toxic, and biodegradable. In this study, we evaluated the antifungal activities and mechanisms of 11 plant extracts against N. ellipsospora. Methods: This study evaluated the antifungal activity of 11 plant extracts against N. ellipsospora of tea gray blight disease and systematically examined the impacts of osthole and carvacrol on the mycelial morphology, mycelial weight, cell microstructure, membrane permeability, various biochemical substrate levels and related gene expression levels of N. ellipsospora. Results: The results showed that carvacrol and osthole exhibited significant antifungal effects among 11 plant extracts with EC50 values of 24.40 and 9.38 mg/L, respectively. Further research demonstrated that carvacrol and osthole significantly inhibited mycelial growth, reduced lesion areas on tea leaves, and markedly affected mycelial morphology and ultrastructure. Observations of mycelial morphology and ultrastructure revealed that carvacrol and osthole caused shrinkage and distortion of the mycelial surface, damage to cell wall and membrane, and disorganization of cellular organelles. Particularly carvacrol and osthole significantly increased chitinase activity, inhibited β-1,3-glucanase activity, and regulated the expression of genes encoding these enzymes.Discussion: The findings indicated that carvacrol and osthole could inhibit the growth of N. ellipsospora by disrupting the integrity of the cell wall and membrane and interfere with energy metabolism. This study would provide a theoretical basis for the development of novel plant-based fungicides.

Carboxymethyl-β-cyclodextrin grafted chitosan embedded oyster (Crassostrea gigas) ferritin can alleviate lead-induced liver injury by oral administration

Han Li,Shuzhen Cheng,Kai Zhou,Jiachen Zang,Zhenyu Wang,Ming Du

Ferritin is expected to alleviate lead-induced oxidative damage. Despite being a relatively stable macromolecular protein that can withstand high temperatures and extreme pH conditions without its structure and activity being compromised, oral administration of ferritin may still result in its degradation within the gastrointestinal tract, reducing its bioavailability. In recent years, microcapsules have emerged as an effective approach in oral protein delivery to enhance bioavailability. In this study, carboxymethyl-β-cyclodextrin grafted chitosan embedded oyster ( Crassostrea gigas ) ferritin (CMCD-CS@GF1) was fabricated with favorable encapsulation efficiency (57.73 ± 4.84 %) and loading capacity (5.78 ± 0.48 %), as well as a certain degree of resistance to digestion in the gastric environment. In the simulated gastric fluid with an acidic pH, the cumulative release of GF1 over 4 h was 24.18 %. In the simulated intestinal fluid with a neutral pH, the cumulative release of GF1 over 8 h was 70.56 %. Animal experiments showed that oyster ferritin could reduce the accumulation of lead in the body, enhance the activity of antioxidant enzymes, activate the Nrf2 signaling pathway, and down-regulate the NLRP3 signaling pathway to reduce oxidative stress and inflammation levels. The findings demonstrate that oyster ferritin may mitigate the toxic effects of lead in the body, providing a significant theoretical foundation for its application in functional foods.

Ca2+ crosslinked gelatin-sodium alginate film loaded with berberine hydrochloride that can effectively promote wound healing of MRSA infection

Peiyuan Li,Huan Liao,Yingbin Lv,Siyan Liang,Jingwen Zhong,Wei Su

Wound infections are a primary cause of delayed wound healing, and film dressings have garnered significant attention in wound management. However, the development of biologically sourced and green-synthesized film dressings with rapid antimicrobial functionality remains an urgent technological breakthrough. In this study, we successfully developed gelatin sodium alginate/sea sheath molecularly modified berberine hydrochloride film (GS/CB). The incorporation of sea sheath powder significantly enhances the mechanical properties of the gelatin‑sodium alginate film. The calcium ions (Ca 2+ ) provided by the sea sheath create a cross-linking effect within the sodium alginate-gelatin network, resulting in tensile properties of the GS/CB films that are 2.6 times greater than those of the control films. This enhancement allows the films to meet the requirements for wound applications. In in vitro antimicrobial experiments, the films incorporated with sodium oleate-modified berberine hydrochloride showed good antimicrobial effects, with an inhibition rate of 97.79 % against Staphylococcus aureus ( S. aureus ) and 98.57 % against methicillin-resistant Staphylococcus aureus ( MRSA ). In in vivo wound healing experiments, the film effectively eliminated MRSA from the wound, achieving a bacterial clearance rate of 98.5 %. Compared to transparent film dressings, the GS/CB film demonstrated superior wound care efficacy and significantly accelerated wound healing. Furthermore, both in vivo and in vitro tests indicated that the film possesses excellent biocompatibility. Therefore, the GS/CB film holds great potential and application value as a new wound dressing for the treatment of infectious wounds.

Biodegradable piezoelectric PHB-BT nanofiber scaffolds combined with ultrasound stimulation to accelerate bone regeneration by regulating Ca2+/CaN/NFAT

Yangyang Qu,Yifeng Shang,Shixing Luo,Xiaomin Pei,Yuanming Xiao,Jinmin Zhao,Li Zheng,Chuanan Liao,Ruiming Liang

Rationale: Bone defects pose a persistent challenge in orthopedic medicine due to their limited self-repair capacity. Although guided bone regeneration scaffolds have shown therapeutic potential, their clinical efficacy remains constrained by their suboptimal osteoinductive capability. Methods: Herein, we developed biodegradable piezoelectric polyhydroxybutyrate-barium titanate (PHB-BT) nanofiber scaffolds capable of generating synergistic piezoelectric stimulation for bone repair when integrated with low-intensity pulsed ultrasound (LIPUS). Results: Compared with conventional PHB scaffolds, PHB-BT nanofiber scaffolds showed enhanced piezoelectric properties and excellent biocompatibility, thereby facilitating sustained osteogenic activity. In vitro studies revealed that these scaffolds significantly promoted the osteogenic differentiation of bone marrow mesenchymal stem cells under LIPUS stimulation. Notably, in vivo evaluations demonstrated that these scaffolds substantially accelerated bone defect repair, with complete scaffold degradation observed after eight weeks. Mechanistically, PHB-BT nanofibers improved osteogenesis via activating the Ca 2+ /calcineurin/nuclear factor of activated T-cells signaling pathway in response to ultrasound stimulation. Conclusions: These findings have significant implications for the design of next-generation, implantable electrical stimulators capable of providing sustained electromechanical cues for personalized bone tissue engineering applications.

Hyodeoxycholic acid in Arisaema Cum Bile can alleviate pentylenetetrazol-induced epilepsy by activating p53/Nrf2/GPX4 to inhibit ferroptosis

XiaoYang Qu,LiJuan Chen,JiangHua He,YuFei Fang,Jing Sun,ZhiYing Yu,Zhen Yang,ZiJing Zhang,JianBo Chen,Ling Dong

Background Epilepsy is a prevalent neurological disorder characterized by recurrent seizures and has limited effectiveness of available clinical treatments. Ferroptosis of neurons is one of the main mechanisms of epilepsy. Arisaema Cum Bile (ACB) is a traditional Chinese medicine that is prepared by fermenting animals bile with Rhizoma Arisaematis rhizomes, has demonstrated therapeutic effects on epilepsy and other nervous system diseases. Furthermore, the active components responsible for the anti-epileptic effects of ACB and the intrinsic mechanisms are still unclear. Purpose This study aims to clarify the principal bioactive constituents of ACB and their mechanisms in the treatment of epilepsy. It also seeks to provide data support for its clinical application in antiepileptic combination therapy. Method Pentylenetetrazol injections were used to induce epilepsy in rats. ACB and brain metabolites were identified via UPLC-Q-Exactive Orbitrap MS/MS, and active components were predicted through bioinformatics analysis. In vivo , ACB and hyodeoxycholic acid (HDCA) were administered by gavage, and the effects on oxidative stress, inflammation, and cognitive impairment were evaluated. Additionally, ferroptosis-related protein and gene expression were analyzed using western blot, PCR, immunofluorescence, and flow cytometry. In vitro , using primary neurons and HT22 cells, with electrophysiology employed to assess neuronal activity and glutamate used to induce ferroptosis in HT22 cells, in order to explore how HDCA alleviates epilepsy by inhibiting neuronal ferroptosis. Result A total of 37 chemical components were identified from the ACB, of which 10 active constituents can cross the blood-brain barrier, including steroids and terpenoids. Bioinformatics analysis suggests that HDCA may be one of the key active components responsible for ACB anti-epileptic effects and ferroptosis may represent a crucial underlying therapeutic mechanism. Studies in animals have demonstrated that treatment with ACB and HDCA reduced the frequency and severity of seizures in epileptic rats, mitigated hippocampal neuronal damage, improved post-epileptic cognitive learning capacity, alleviated neuroinflammatory injury and decreased markers of ferroptosis. This neuroprotective effect likely occurs through activation of the p53/Nrf2/GPX4 signaling pathway. Further in vitro cell experiments demonstrated that HDCA can inhibit the generation of action potentials in primary hippocampal neurons, protects HT22 neurons from glutamate-induced cell death, an effect that is reduced by Nrf2 inhibitors. These findings corroborate earlier bioinformatics analyses, confirming that the neuroprotective effects of HDCA are closely linked to Nrf2-mediated ferroptosis pathways. Conclusion HDCA is a key component of ACB anti-epileptic effects, potentially inhibiting ferroptosis through the p53/Nrf2/GPX4 pathway to reduce epilepsy.
