In Situ Encapsulation of Organophosphorus Hydrolase into a Reduction-Catalytic Cu-MOF for Chemoenzymatic Cascade Conversion of Methyl Parathion to 4-Aminophenol

Qianqian Li,Qinghong Shi,Xiaoyan Dong,Linling Yu,Yan Sun

The highly toxic organophosphate pesticide methyl parathion (MP) is widely used in agriculture and thus poses significant risks to the environment. Organophosphorus hydrolase (OPH) can effectively catalyze the hydrolysis of MP, but generate a toxic product of 4-nitrophenol (4-NP), so the following reduction of 4-NP to 4-aminophenol (4-AP) with low toxicity but high economic value has been designed for sustainable development. Therefore, the cascade conversion of highly hazardous MP to economically valuable 4-AP is an attractive strategy for both environmental remediation and industrial applications. In this study, we synthesized a reduction-catalytic cupric metal–organic framework (CuBDC) in solvent-free aqueous phase and under mild conditions and in situ encapsulated OPH within CuBDC via a cation polyelectrolyte poly(allylamine hydrochloride) (PAH)-assisted biomineralization, creating a chemoenzymatic cascade catalyst (OPH-PAH@CuBDC) for MP conversion to 4-NP. The optimized OPH-PAH@CuBDC exhibited 154% hydrolysis activity of free OPH, 130% reduction activities of pure CuBDC, and an average cascade conversion rate of 6.2 μmol·min–1·gcatalyst–1, affording a complete conversion of 50 μM MP to 4-AP in only 6 min at low reductant concentration (20 mM NaBH4), due to the Cu2+-activation effect on the encapsulated OPH and the PAH-induced exposure of more reduction-catalytic sites, respectively. Furthermore, OPH-PAH@CuBDC retained 95% activity in 10 day storage and about 50% activity in 10 recycles. The results proved that OPH-PAH@CuBDC is a promising chemoenzymatic catalyst with great potential in practical applications for MP detoxification and resourcilization.

Cu-MOF-Derived Cu/C Catalysts via Solvent Polarity Engineering with Renewable Ligand for Efficient Hydrogenation of Levulinic Acid to γ-Valerolactone

Weimeng Cai,Haojie Du,Erju Wang,Yuting Qiang,Kunhong Hu,Jianhua Guo

The synthesis of value-added chemicals from renewable biomass is becoming increasingly important in the transition to a sustainable and low-carbon energy system. Herein, we demonstrate a solvent-polarity modulation strategy to engineer biomass-derived Cu-MOF using 2,5-furandicarboxylic acid (FDCA) as a renewable ligand, where tailored β/α ratios via Kamlet-Taft parameters balance nucleation-growth kinetics to optimize textural properties. Pyrolyzed Cu-MOF at 500 °C under a nitrogen atmosphere yield the Cu/C catalysts, which were subsequently employed in the selective hydrogenation of levulinic acid (LA) to γ-valerolactone (GVL). Among the various solvents used, the Cu-MOF precursor synthesized in a mixed solvent of methanol and N, N-dimethylformamide (DMF) demonstrated the highest catalytic performance, outperforming those synthesized in other solvents. This highlights the significant impact of solvent selection on the structure of Cu-MOF, which in turn affects their hydrogenation activity. Advanced characterization confirming that the uniform Cu dispersion (confirmed by HRTEM mapping) and Cu⁰/Cu⁺ electronic synergy (via Cu Auger analysis) enhanced hydrogenation performance. Additionally, the biomass-derived catalysts exhibited excellent durability. Overall, this research provides a promising approach to develop highly efficient catalysts using renewable ligands and demonstrates the potential of regulating catalyst properties through the strategic selection of solvents, as informed by Kamlet-Taft parameters.

Flexible Sensor Array Based on Polythiophene/Cu-MOF-Derived CuO and Polyaniline/MXene Composites for Room-Temperature Detection of Hydrogen Sulfide and Ammonia

Zhao Kuo,Shi Yunbo,Niu Haodong,Liu Jinzhou,Yang Xiaohui,Tang Bolun

Real-time detection of hydrogen sulfide (H 2 S) and ammonia (NH 3 ) is important for occupational safety in humid, mixed-gas environments, yet conventional chemiresistive sensors remain limited by cross-sensitivity, humidity dependence, and poor portability at room temperature. Here, we report a flexible dual-channel sensor array coupled with a gated recurrent unit-convolutional neural network (GRU-CNN). A polythiophene/Cu-MOF-derived CuO composite (PTh/CuO) targets H 2 S through sulfur-affinitive CuO surface sites, whereas a polyaniline/Ti 3 C 2 T x MXene composite (PANI/MXene) targets NH 3 through PANI deprotonation and interfacial charge modulation. At 25 ± 1 ° C, the optimized PTh/CuO and PANI/MXene channels exhibited responses of 87.5% to 100 ppm H 2 S and 189% to 100 ppm NH 3 , respectively, together with linear responses over 1−10 ppm ( R 2 = 0.997 and 0.998) and theoretical limits of detection of 73.3 and 40.4 ppb. Distinct responses were retained at 300 ppb H 2 S and 200 ppb NH 3 , with response/recovery times of 15/22 and 25/43 s, respectively. A GRU-CNN model trained using dynamic four-channel signals supported gas classification and concentration estimation across 0−10 ppm and 30%−80% relative humidity within the tested dataset; the maximum concentration-prediction mean squared errors were 0.020 for H 2 S and 0.015 for NH 3 . The channels retained 98.4% and 98.1% of their initial responses under 30 ° bending. Integration with a wireless wristband enabled remote signal acquisition and a wearable monitoring demonstration. This work combines chemically differentiated sensing channels, humidity-inclusive data analysis, and flexible wireless readout within a single room-temperature platform.

High-catalytic-activity conductive Cu-MOF coupled with carbon black Super-P for the rapid electrochemical detection of methylglyoxal in food and biological fluid

Yafeng Jin,Ruilong Zhang,Xinyue Song,Didi Tong,Haibo Shang,Fangming Han,Xiaobo Li,Guangri Xu

Elevated levels of methylglyoxal (MG) in the human body are linked to various metabolic disorders. Therefore, developing efficient methods for detecting MG in food and biological fluid is of significant importance. In this work, a high-catalytic-activity conductive copper-based metal–organic framework (Cu 3 (HHTP) 2 ) was synthesized via a hydrothermal method, and a sensitive electrochemical sensor based on a glassy carbon electrode (GCE), denoted as Super-P/Cu 3 (HHTP) 2 /GCE, for monitoring methylglyoxal (MG) was constructed using a composite of Cu 3 (HHTP) 2 and carbon black Super-P. Leveraging the catalytic activity of Cu 3 (HHTP) 2 and the enhanced conductivity provided by Super-P, the sensor demonstrated a wide linear detection range (1–500 µM) and a low detection limit (0.0275 µM). Moreover, it was successfully applied to the determination of MG in milk, beer, urine, and human serum samples, achieving recoveries ranging from 91.7 % to 115.7 %. This work opens up new avenues for the development of innovative electroanalytical tools for MG monitoring in both food and biological fluid.

A novel electro-spun Cu-MOF loaded PAN/cellulose membrane for efficient adsorption-assisted photocatalytic decontamination of bisphenol A: Kinetics, thermodynamics, and mechanistic study

Alamgir,Basem Al Alwan,Adeel Ahmed,Abdul Qadir,Muhammad Fayaz,Amira Alazmi,Muhammad Aadil,Mohamed R. El-Aassar,Atef El Jery,Muhammad Rafiq,Raza Ullah

Electrospun membranes have gained attention for wastewater treatment, but pristine nanofiber membranes like Polyacrylonitrile (PAN) often have limited adsorption and photocatalytic efficiency. A hybrid membrane was developed to address this, combining PAN, cellulose, and a metal–organic framework (MOF) to create a multifunctional photocatalytic adsorbent. The membrane was fabricated using an electrospinning process. PAN was infused with copper-based MOF (Cu-MOF), and cellulose was blended with polyethylene oxide (PEO), resulting in an integrated membrane structure. Structural characterization confirmed the uniform distribution and firm adhesion of Cu-MOF to the PAN fibers. The photocatalytic potential of the synthesized membrane was evaluated for the removal of bisphenol A (BPA) from water under visible light irradiation. The PAN/Cellulose nanofiber substrate loaded with 1.0 g of Cu-MOF (PAN/Cell/MOF-III) achieved 98 % removal efficiency. The PAN, PAN/Cell, pristine Cu-MOF, PAN/Cell/MOF-I, PAN/Cell/MOF-II, and PAN/Cell/MOF-III exhibited adsorption capacities of 4.0, 19.9, 69.1, 30.7, 47.4, and 87 mg/g, respectively. The effects of key factors, including membrane dosage, solution pH, and BPA concentration, on the adsorption and degradation efficiency of PAN/Cell/MOF-III were systematically explored. Trapping species and Electron Paramagnetic Resonance (EPR), studies identified holes (h + ) and superoxide radicals (O 2 − ) as the primary reactive species responsible for BPA degradation. Kinetic analysis revealed degradation followed a pseudo-first-order model, while adsorption followed a pseudo-second-order model. Thermodynamic parameters (ΔG ads , ΔH ads , ΔS ads ) indicated spontaneous, physical, and endothermic adsorption. The Langmuir and Dubinin-Radushkevich isotherm models accurately fit the adsorption data. The PAN/Cell/MOF-III membrane showed excellent reusability over five cycles, demonstrating its potential for practical wastewater treatment applications.

Electrochemical endotoxin aptasensor based on a metal-organic framework labeled analytical platform

Yangyang Duan,Nan Wang,Zixu Huang,Hongxiu Dai,Ling Xu,Shengjun Sun,Houyi Ma,Meng Lin

An electrochemical aptasensor for the lipopolysaccharide (LPS) detection was constructed by using copper-based metal-organic framework (Cu-MOF) as a label and the LPS aptamer of specific single-stranded DNA as a probe. The carboxyl-functionalized polypyrrole nanowires (PPy NWs) were synthesized by electrochemical polymerization method, and the amino-terminated aptamer covalently coupling with the carboxyl group of the PPy NWs was immobilized onto the modified electrode. The aptamer can specifically combine with the target LPS molecules, and Cu-MOF was labeled by adsorption based on specific interactions of LPS carbohydrate portions with anionic groups. The fabrication processes of the aptasensor were characterized by scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS) and electrochemical impedance spectroscopy (EIS). Differential pulse voltammetry (DPV) was used to measure electrochemical performance of the aptasensor, and the electrochemical signal can be directly measured by the electrochemical redox reaction of Cu(II)/Cu(I) existed in the Cu-MOF. The electrochemical aptasensor exhibited a high sensitivity toward LPS ranging from 1.0 pg/mL to 1.0 ng/mL with a detection limit of 0.29 pg/mL. Moreover, the developed sensor was found to have good selectivity, stability and regeneration properties, and the sensor also successfully detected LPS in real tap water samples.

Competitive electrochemical immunosensors by immobilization of hexahistidine-rich recombinant proteins on the signal labels

Yong Chang,Meiling Liu,Tong Wu,Ruting Lin,Lin Liu,Qijun Song

Most of proteins in the commercial ELISA kits are recombinant with hexahistidine (His 6 ) tags. Based on this fact, we proposed a simple procedure for the preparation of signal labels and the design of competitive immunosensors. The signal labels were fabricated through the high-affinity interaction between the His 6 tails on the surface of recombinant proteins and the coordinatively unsaturated copper ions on the surface of pristine Cu-based metal organic framework (Cu-MOF). The recombinant protein-modified MOF could be captured by the sensor electrode, thus producing a strong differential pulse voltammetry signal through the electrochemical reduction of Cu 2+ ions in MOF. However, the target protein in sample solution could compete with the His 6 -tagged protein on the surface of Cu-MOF to bind the antibody attached on the electrode, thus leading to the decrease in the electrochemical signal. The peak current showed an inversely relationship with the concentration of target protein. The competitive immunosensor exhibited a linear range of 1 pg/mL to 1 ng/mL with SARS-CoV-2 nucleocapsid protein ( N -protein) as the target analyte. We believe that the method can be used to detect other proteins by utilizing the characteristic of recombinant proteins in commercial kits.

Fast in situ construction of a Cu2O/Cu-MOF S-scheme heterojunction with synergetic interface matching and carrier separation ability for efficient photocatalytic nitrogen fixation

Yanning Qu,Wanquan Ma,Shihu Lei,Yuan Yu,Dongfeng Sun,Huayun Du,Madi Ren

Metal-organic frameworks (MOFs)-based heterojunctions, as one of porous materials, offer abundant unsaturated coordination sites, making them suitable for photocatalytic N 2 reduction. However, complicated reaction methods, mismatched interfaces and ineffective photo-induced carrier separation restricted their applications. In this study, a kind of Cu 2 O/Cu-MOF heterojunction was synthesized via a fast in situ method in acidic medium (CCM-a). It showed a typical core-shell structure with Cu-MOF coating surrounding the Cu 2 O. The photocatalytic NH 3 yield of CCM-a reached 83.25 μmol/g/h, which was 9.03 and 19.64 higher than that of pure Cu 2 O and CCM-b, respectively. The reaction pathway of N 2 reduction was investigated using density functional theory (DFT) and in-situ FT-IR spectra, revealing a preference for the alternating association pathway. The photocatalytic mechanisms were analyzed through the built-in electric field (IEF) theory, supporting its S-scheme charge-transfer route of CCM-a. The novel material achieved from in situ strategy not only ensured a good interface matching ability but also facilitated sufficient separation of photo-generated carriers via a S-scheme route for efficient photocatalytic N 2 fixation.

The highly specific detection and mechanism of Cu-MOF-74 fluorescent probe to amino trimethylene phosphonic acid: Experimental study and theoretical calculation of quantum chemistry

Hua Qiang,Mingzhu Xia,Fengyun Wang,Wu Lei,Xin Lu,Yue Ni

Due to the lack of rapid and efficient detection methods for the determination of trace organic phosphonate concentrations in the natural aqueous system, little is known about their impact on the ecosystem. Amino trimethylene phosphonic acid (ATMP) is an organic phosphonate that has been widely used in many fields. In the present work, the copper-based organic framework material (Cu-MOF-74) for the specific detection of amino trimethylene phosphonic acid (ATMP) in aqueous solution was prepared by hydrothermal synthesis using 2,5-dihydroxyterephthalic acid (DHTP) as raw material and copper nitrate trihydrate as metal source. The synthesized material can be used as a “turn-on” fluorescent sensor for the specific sensing of ATMP, the fluorescence intensity of Cu-MOF-74 can be enhanced 24.2 times by ATMP (90 μΜ), the emission wavelength of the fluorescence emission spectrum of the system was blue-shifted, and the detection limit (LOD) of ATMP was 0.844 μΜ. Moreover, the mechanism of ATMP greatly increasing the fluorescence intensity of Cu-MOF-74 was investigated by experimental and theoretical calculations of quantum chemistry. The experimental results showed that ATMP was involved in the coordination of open metal sites in Cu-MOF-74. Multiwfn wavefunction analysis results demonstrated that the charge transfer from the benzene ring and hydroxyl groups of the ligand to the metal (LMCT) occurred during the electron excitation of Cu-MOF-74, but after adding ATMP, the coordination of ATMP with copper led to the interruption and reduction of the energy transfer and electron transfer from ligand DHTP to Cu 2+ , and the fluorescence intensity of Cu-MOF-74 was improved. The Cu-MOF-74 fluorescent probe exhibited excellent anti-interference ability, high selectivity, good fluorescence intensity stability in the short term, so it can be used as a reliable fluorescent probe for the highly specific detection of low concentration ATMP, the revealed microscopic mechanism of Cu-MOF-74 fluorescent probe to ATMP from the atomic level can provide a theoretical basis for the research of new fluorescence detectors.

A photocatalytic and superhydrophobic self-cleaning nanocellulose-based membrane based on Cu-MOF for highly efficient oil/water separation, the removal of dyes and anti-biofouling towards sewage remediation

Zuozhu Yin,Tonghan Zhou,Zihao Li,Yuanting Deng,Mingshan Xue,Yuhua Chen,Junfei Ou,Yu Xie,Yidan Luo,Chan Xie,Zhen Hong

Multifunctional superhydrophobic nanocellulose-based membranes for self-cleaning, oil/water separation, photocatalytic degradation, and anti-biofouling are in great demand for sewage treatment. This demand led to the in situ deposition of Cu-MOF micro/nanoparticles on the surface of CB membranes, followed by stearic acid (STA) modification to provide excellent superhydrophobicity, which enables effective oil/water separation, photocatalytic degradation, and anti-biofouling. The CB/Cu-MOF/STA membranes created in this manner demonstrated good superhydrophobicity with a water contact angle of 160 ± 2° and superoleophobicity with an oil contact angle of 0°. The superhydrophobic CB/Cu-MOF/STA membrane also demonstrates remarkable stability under extreme conditions, including high temperatures, acid and alkali solutions, etc. Furthermore, it has a high separation efficiency of 98 % and a large separation flux of 127.71 L·m −2 ·h −1 for the n-Hexane/water separation. After 8 cycles of separation, the n-Hexane/water mixture's separation efficiency was maintained at about 95 %. Additionally, the superhydrophobic CB/Cu-MOF/STA membrane shows good photocatalytic activity because of its high porosity and large specific surface area. Chlorella's capacity to cling to the superhydrophobic CB/Cu-MOF/STA membrane was significantly diminished, and the membrane demonstrated strong anti-biofouling capability. The superhydrophobic CB/Cu-MOF/STA membrane is thus demonstrated to have a significant impact on promoting the widespread development of superhydrophobic materials with a variety of applications.

Electrochemical Determination of Aflatoxin B1 (AFB1) Using a Copper-Based Metal-Organic Framework (Cu-MOF) and Gold Nanoparticles (AuNPs) with Exonuclease III (Exo III) Assisted Recycling by Differential Pulse Voltammetry (DPV)

Chunyan Wang,Hui Zhang,Xiaoqing Jiang,Bo Zhou

A sensitive electrochemical biosensor was designed for determination of aflatoxin B1 (AFB1) using a copper-based metal-organic framework (Cu-MOF), which has strong electrochemical activity and exonuclease III (Exo III)-assisted recycling for dual signal amplification. Hairpin DNA (S1) was immobilized on the electrode. The AFB1 was recognized by aptamer DNA (S2) and complementary DNA (S3) was released. The S3 hybridized with the hairpin S1 to form the Exo III hydrolyzed double-stranded DNA, leaving a partial sequence of hairpin DNA (S1′) on the electrode and releasing S3 for the next cycle of the opening and digestion of hairpin S1. The amplified S1′ then was able to combine with more signal probes. Cu-MOF bond gold nanoparticles (AuNPs) by -NH2 were immobilized to capture DNA (S4) to obtain Cu-MOF/AuNPs/S4. This signal probe Cu-MOF/AuNPs/S4 was able to hybridize with the electrode and generate an amplified electrochemical signal. Under the optimized conditions, this electrochemical biosensor for AFB1 exhibited a low detection limit of 6.7 × 10−7 ng/mL at a signal-to-noise equal to 3 and a wide linear range from 10−6 to 1 ng/mL. The biosensor was also used to analyze AFB1-spiked beer sample with recovery values between 96% and 103%. This method has the potential to become a valuable technology for detecting various toxins by the selection of the appropriate aptamer DNA.

Hypoxia-Adapted Sono-chemodynamic Treatment of Orthotopic Pancreatic Carcinoma Using Copper Metal–Organic Frameworks Loaded with an Ultrasound-Induced Free Radical Initiator

Yu Sun,Jing Cao,Xue Wang,Cong Zhang,Jiali Luo,Yiqing Zeng,Chao Zhang,Qunying Li,Ying Zhang,Wen Xu,Tao Zhang,Pintong Huang

The efficacy of sonodynamic therapy (SDT) is largely dependent upon oxygen availability to generate deleterious reactive oxygen species, and as such, hypoxic microenvironments greatly constrain the efficacy of SDT. Development of free radical generators that are not dependent on oxygen and related combination treatment strategies thus have the potential to enhance the antitumor potential of SDT. Combined treatment strategies are expected to improve the efficacy of sonodynamic antitumor therapy. As metal–organic framework (MOF) platforms are highly amenable to integration with other therapeutic approaches, we herein report the development of tumor microenvironment (TME)-responsive nanoparticles constructed by embedding the azo initiator 2,2′-azobis[2-(2-imidazolin-2-yl)propane]dihydrochloride (AIPH) into hypoxia-triggered copper metal–organic framework (Cu-MOF) nanovectors to achieve synergistic sono-chemodynamic therapy in an orthotopic murine pancreatic carcinoma model system. When exposed to hypoxic conditions within the TME, this Cu-MOF structure underwent degradation, leading to the release of Cu2+ and AIPH. Cu2+ was then able to deplete local glutathione stores, resulting in the reduction of Cu2+ to Cu+, which then reacts with endogenous H2O2 in a Fenton-like reaction to yield cytotoxic hydroxyl radicals (•OH) for chemodynamic therapy. When exposed to ultrasound irradiation, AIPH further degraded in an oxygen-independent manner to yield nitrogen bubbles and alkyl radicals, the former of which enhanced the ability of these nanoparticles to penetrate deeply into the tumor. The resultant radicals induced substantial DNA damage and apoptotic cell death within target tumors under different levels of oxygen availability. As such, this hypoxic TME-responsive synergistic sono-chemodynamic approach offers an ideal means of achieving oxygen-independent free radical generation and enhanced treatment efficacy.
