Whether the copper ion (Cu 2+ ) exposed to us functions as a necessary part of metabolism or toxicant for organs depends on subtle concentration difference of trace-level. Although miniature sensors have combined various recognition probes and signal transducer moieties to form novel sensing scheme, haunting limits on practicality or analytical specifications like sensitivity and selectivity remain thwarting high Cu 2+ detection performance with excellent cost-effectiveness. In this work, we report a Cu 2+ -specific biosensor integrating Deoxyribozyme (DNAzyme) and single-particle plasmon. The basic principle here is that severity of cleavage corresponding to various Cu 2+ concentration determines these elements’ aggregation capacity. The evaluation criteria is set up on the shift of dark-field Localized Surface Plasmon Resonance (LSPR) wavelengths caused by the binding with free AuNPs. Not only excellent analytical specifications like limit of detection (LOD) of 0.082 nM, broad dynamic range from 0.1 nM–5 μM, ultra-specificity are demonstrated but recovery rate with real sample and term of validity is also investigated to be 101.25 ± 9.15 % at 3 nM and longer than 9 days corroborating its potential for practical scenario. Therefore, the sensor is introduced as an attract alternative to remove hindrances for high sensing performance and to generalize on-chip cleavage-inducing spectral shift to heavy metal ions concentration measurement.

A highly sensitive and selective impedimetric immunosensor with triple signal amplification was designed for ultrasensitive detection of prostate-specific antigen (PSA) by using anti-PSA antibody and DNAzyme-functionalized gold–palladium hybrid nanotags (Ab 2 –AuPd–DNA). The signal was amplified based on the Ab 2 –AuPd–DNA toward the catalytic precipitation of 4-choloro-1-naphthol (4-CN). DNAzyme (as a kind of peroxidase mimic) could catalyze the oxidation of 4-CN, whilst AuPd hybrid nanostructures could not only provide a large surface coverage for immobilization of biomolecules but also promote 4-CN oxidation to some extent. The produced insoluble benzo-4-chlorohexadienone via 4-CN was coated on the electrode surface , and hindered the electron transfer between the solution and the electrode, thereby increasing the Faradaic impedance of the base electrode . Three labeling strategies including Ab 2 –AuNP, Ab 2 –AuPd and Ab 2 –AuPd–DNA were investigated for determination of PSA, and improved analytical features were obtained with the Ab 2 –AuPd–DNA strategy. Under optimal conditions, the dynamic concentration range of the impedimetric immunosensor spanned from 1.0 pg mL −1 to 50 ng mL −1 PSA with a detection limit of 0.73 pg mL −1 . Intra- and inter-assay coefficients of variation were below 8.5% and 9.5%, respectively. Importantly, no significant differences at the 0.05 significance level were encountered in the analysis of 6 clinical serum specimens and 6 diluted standards between the impedimetric immunosensor and the commercialized electrochemiluminescent method for PSA detection.

Accurate tumor subtyping through multiplex miRNA profiling is critical for precision medicine but remains challenging due to limitations in current methods, including cross-reactivity, cost, and stability. Herein, we present a DNAzyme-powered DNA fiber barcode platform that combines G-quadruplex (G4)/hemin catalysis, miRNA-responsive displacement, and polydopamine (PDA)-mediated fluorescence modulation for sensitive and specific miRNA detection. The system operates via a target-dependent “switch”: in the absence of target miRNAs, G4/hemin DNAzymes catalyze dopamine (DA) oxidation into PDA that efficiently quench fluorescence through Förster resonance energy transfer (FRET); when target miRNAs are present, they competitively binding to the capture strands on DNA fibers with higher affinity, displacing the G4 structures, inhibiting DNAzyme formation and preserving the fluorescence signal, with fluorescence intensity showing a positive correlation with target concentration. This mechanism enables dual qualitative and quantitative analysis with a broad linear range (50 pM-100 nM) and low detection limits (5.50 pM). Key advantages include stable, tunable G4/hemin transduction, enhanced kinetics and signal-to-noise through synergistic displacement-quenching, and multicolor barcoding for one-pot multiplex miRNA detection. Validated against qPCR with high concordance, this platform overcomes existing technical barriers to enable robust miRNA-based cell typing classification and precision diagnostics.

Aberrant copper (II) homeostasis is intimately linked to numerous neurodegenerative pathologies, yet ultrasensitive detection of Cu 2+ in complex biological matrices remains a challenge due to its trace-level concentrations and pervasive interference. Here, we introduce a novel ratiometric electrochemical sensor that couples Cu 2+ activated DNAzyme with a metal-organic framework (2D MOF) as an intrinsic internal reference and methylene blue loaded in MIL-101(MB@MIL-101) as an external reporting element. Upon Cu 2+ binding, an interfacial DNAzyme cleaves its substrate strand, triggering the release of MB@MIL-101 from the electrode while concomitantly exposing additional electroactive sites on the MOFs. This results in an antiparallel dual-signal response: the MB current ( I MB ) decreases, and the 2D MOFs current ( I MOFs ) increases, enabling Cu 2+ quantification by the ratio I MB / I MOFs . The developed ratiometric platform demonstrates a wide linear range from 10 -12 to 10 -4 M and a remarkable detection limit of 2.37 × 10 -13 M. Furthermore, it demonstrates reliable performance in complex samples such as sweat and serum. Beyond Cu 2+ sensing, this work establishes electroactive MOFs as intrinsic ratiometric calibrants and provides new avenues for DNAzyme-based dual-signal electrochemical biosensing.

A simple and sensitive method was developed for the electrochemiluminescence (ECL) determination of DNA. • Au-luminol NPs addressed the issues of weak solubility and ECL signals of luminol in neutral conditions. • The target DNA induced the formation of heme and G-quadruplexes as biomimetic enzymes, thereby enhancing the electrochemical luminescence intensity of luminol. A novel signal amplification approach was developed for DNA detection, leveraging the synergistic effect of G-quadruplex/hemin DNAzyme and gold nanoparticles (Au NPs) to enhance the electrochemiluminescence (ECL) of luminol. The optimized system exhibited excellent linear response toward target DNA with a linear detection range from 10 fM-50 pM and achieved fM-level DNA detection. The spiked recoveries of serum samples ranged from 95.1% to 107.6%, demonstrating its reliable performance for practical sample analysis. Compared with recently reported analogous ECL sensors for DNA detection, this system featured a wider linear range, lower limit of detection and stronger anti-matrix interference capability, offering a new strategy for rapid and ultrasensitive detection of target analytes in complex matrices. The 10-23 DNAzyme is a potent catalytic nucleic acid for biosensing; however, traditional single-blocker designs suffer from high background signal leakage due to spontaneous thermal “breathing.” To overcome this, we engineered a triangular DNA probe-DNAzyme (TDPzyme) that spatially separates the enzyme components onto a rigid 3D scaffold. By splitting the binding arms and catalytic center, the TDPzyme functions as a fault-tolerant logic “AND” gate, strictly requiring the simultaneous displacement of multiple blocker strands by the target miR-21. This architectural constraint physically shields the probe from nuclease degradation and virtually eliminates false-positive activation, yielding a near-zero background state. Functionalized with a Cy3-BHQ2 pair and an AS1411 aptamer for targeted, carrier-free cellular delivery, the TDPzyme achieved an approximately 18-fold enhancement in sensitivity over conventional closed-arm designs, establishing a linear range of 0.1 nM to 40.0 nM and a limit of detection of 50.3 pM (3σ). Furthermore, time-lapse dynamic imaging demonstrated that the TDPzyme enables highly reliable, high-contrast discrimination of miR-21 expression between tumor and normal cells. Ultimately, the TDPzyme design offers a robust, structurally controllable platform for developing highly sensitive, low-background biosensors applicable to demanding live-cell imaging and precision molecular diagnostics.

The contamination of marine environments with antibiotics, such as kanamycin, poses a significant ecological threat due to its potential harmful effects on marine organisms. Conventional methods for detecting kanamycin, including high-performance liquid chromatography (HPLC) and enzyme-linked immunosorbent assays (ELISA), face challenges in terms of cost, complexity, and long detection times. This research aims to address these limitations by developing a novel, highly sensitive biosensing platform for kanamycin detection based on a DNAzyme-driven cascade reaction coupled with nanoparticle technology. The method utilizes aptamers for specific kanamycin recognition, which activates DNAzyme-1, triggering a downstream DNA walker system. This cascade reaction amplifies fluorescence signals, significantly enhancing sensitivity. The optimized platform demonstrated a wide detection range (0.05–3 nmol/L) and a low detection limit of 0.0327 nmol/L. This system showed excellent specificity for kanamycin, even in the presence of other antibiotics, and was successfully applied to detecting kanamycin in seawater samples with high recovery rates (94%–102.9%). This DNAzyme-based biosensor offers a promising approach for rapid, cost-effective, and high-sensitivity monitoring of antibiotic contamination in aquatic environments, with potential applications in environmental monitoring and ecological protection.

Decapod Iridescent Virus 1 (DIV1) leads to high mortality rates in shrimp farming, significantly impacting the aquaculture industry economically. Currently, there are no effective treatments for controlling DIV1 infections, and management primarily relies on preventative measures. Rapid and precise diagnosis of DIV1 is crucial. In this study, we identified two DNA enzymes, MCP-1 and MCP-8, that can specifically recognize DIV1 via in vitro selection. The detection limit of MCP-1 was 6.89 copies/μL of DIV1. We developed a DNAzyme-based “turn off” biosensor for quick detection of DIV1, which is both highly specific and sensitive, by integrating SYBR Green I to enhance the signal. The detection limit of biosensor could reach 2.01 copies/μL of DIV1. Compared to conventional detection methods, this sensor delivers results in 5 min without the need for complex procedures, greatly streamlining the detection process.

Hemin/G-quadruplex (hG4) complexes are frequently used as artificial peroxidase-like enzymatic systems (termed G4 DNAzymes) in many biosensing applications, in spite of a rather low efficiency, notably in terms of detection limits. To tackle this issue, we report herein a strategy in which hemin is chemically modified with the amino acids found in the active site of parent horseradish peroxidase (HRP), with the aim of recreating an environment conducive to high catalytic activity. When hemin is conjugated with a single arginine, it associates with G4 to create an arginine–hemin/G4 (R-hG4) DNAzyme that exhibits improved catalytic performances, characterized by kinetic analysis and DFT calculations. The practical relevance of this system was demonstrated with the implementation of biosensing assays enabling the chemiluminescent detection of G4-containing DNA and colorimetry detection of the flap endonuclease 1 (FEN1) enzyme with a high efficiency and sensitivity. Our results thus provide a guide for future enzyme engineering campaigns to create ever more efficient peroxidase-mimicking DNA-based systems.

Gold nanoparticles (Au NPs) have been widely utilized in developing DNAzyme-functionalized nanosensors, most of which were engineered by attaching the thiolated DNAzymes to Au NPs via Au-S bonding. However, the Au NP-DNAzyme nanosensors always suffer from signal distortion when applied in complex environment with abundant thiols, which poses challenge for practical applications. Here, we focus on addressing the root cause of the issue and propose to decorate the Au NPs with a thin layer of platinum, thus facilitating the conjugation of DNAzymes through Pt-S bonding, a thiol-resistant cross-linking. The Pt-S bond stabilized DNAzyme nanosensor effectively minimized false positive signals when detecting L-histidine in infant formulas, as compared to the Au-S stabilized counterpart. This innovative strategy holds promise for high-fidelity biosensing, improving the practical applicability of Au NP-based DNAzyme nanosensor.

Enrichment of rare cancer cells from various cell mixtures for subsequent analysis or culture is essential for understanding cancer formation and progression. In particular, maintaining the viability of captured cancer cells and gently releasing them for relevant applications remain challenging for many reported methods. Here, a physically cross-linked deoxyribozyme (DNAzyme)-based hydrogel strategy was developed for the specific envelopment and release of targeted cancer cells, allowing the aptamer-guided capture, 3D envelopment, and Zn2+-dependent release of viable cancer cells. The DNAzyme hydrogel is constructed through the intertwinement and hybridization of two complementary DNAzyme strands located on two rolling circle amplification-synthesized ultralong DNA chains. The enveloping and separation of target cells were achieved during the formation of the DNAzyme hydrogel (sol–gel transition). Triggered by Zn2+, the encapsulated cells can be gently released from the dissociated DNAzyme hydrogel with high viability (gel–sol transition). Successful isolations of target cells from cancer cell mixtures and peripheral blood mononuclear cells (PBMC) were demonstrated. This method offers an attractive approach for the separation of target cancer cells for various downstream applications that require viable cells.

In this work, after optimizing the original aptamer sequence by truncation and site-directed mutagenesis, a simple and sensitive colorimetric aptasensor was established for detecting the widespread food-borne pathogen Vibrio parahemolyticus (V. parahemolyticus). The detection strategy was based on the competition for an V. parahemolyticus specific aptamer between its complementary DNA (cDNA) and V. parahemolyticus. The aptamer-conjugated magnetic nanoparticles (MNPs) were used as capture probes, and the G-quadruplex (G4) DNAzyme was employed as the signal amplifying element. Under optimal conditions, a wide linear detection range (from 102 to 107 cfu/mL) was available, and the detection limit could be as low as 10 cfu/mL. This method was also used to detect V. parahemolyticus in contaminated salmon samples, and the results showed good consistency with those obtained from standard plate counting method. Therefore, this novel aptasensor could be a good candidate for sensitive and selective detection of V. parahemolyticus without complicated operations.
