Comparative Study of Camellia oleifera Seed Oil on Chemical Constituents, Antioxidant Activities and Physicochemical Characteristics from Southern China

Qingqing Li,Yong Zhu,Likang Qin

Eleven kinds of Camellia oleifera seed oils (CSOs) were evaluated in terms of chemical constituents, antioxidant activities, acid value (AV) as well as peroxide value (POV). These CSOs contained abundant β-sitosterol, squalene, α-tocopherol and phenolics, in which the squalene was the distinct constituent with the content between 45.8±0.8 and 184.1±5.5 mg/kg. The β-sitosterol ranging from 143.7±4.8 to 1704.6±72.0 mg/kg contributed a considerable content to total accompaniments. Palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid were present in these CSOs, in which the dominant fatty acid was oleic acid with the content between 59.66±0.72 and 82.89±2.16 g/100 g. The AV ranged from 0.1±0.0 to 1.3±0.0 mg KOH/g, and the POV was between 0.1±0.0 and 1.0±0.0 g/100 g. These CSOs showed antioxidant activity based on DPPH and ABTS radical scavenging assay. Both α-tocopherol and β-sitosterol contents showed a positive correlation with DPPH and ABTS values, respectively, while the α-tocopherol content showed a negative correlation with AV. These results suggested that CSO can be categorized into high oleic acid vegetable oil with abundant active constituents, of which the quality presented variation among different origins. These accompaniments may contribute to the delay of its quality deterioration.

Characterization and functional properties of umami peptides from Douchi: taste mechanism and antioxidant activity

Shiyu Zhang,Feng Chen,Jianfei Li,Haohan Hu,Xiya Chen,Jingling Kang,Xiaodong Yu,Qiyi He

Douchi (DC) is a traditional Chinese condiment renowned for its rich umami flavor. This study aimed to comprehensively investigate the composition, characteristics, potential taste mechanisms, and antioxidant activity of umami peptides in DC. The aqueous extract of DC underwent ultrafiltration and gel column chromatography to isolate its high umami components. Three novel umami peptides (EEQEHT, FED, AEPE) were identified using liquid chromatography-tandem mass spectrometry (LC-MS/MS) coupled with bioinformatics analysis. Electronic tongue studies confirmed the high umami activity of these peptides, with scores ranging from 7.64 to 8.72 at a concentration of 0.2 mg/mL, with FED exhibiting the highest umami intensity, minimal bitterness and pronounced saltiness. Molecular docking studies revealed that all three umami peptides were embedded within the T1R1 subunit, facilitating their binding to the umami receptor via hydrogen bonding. Additionally, these peptides demonstrated significant in vitro antioxidant activities, scavenging 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl radicals. In vivo assessments on zebrafish embryos exposed to oxidative stress showed improved survival rates, normalized heart rates, and dose-dependent reductions in reactive oxygen species (ROS) levels. In conclusion, this study provides valuable insights into the umami peptides present in DC, highlighting their potential applications in the food and health supplement industries.

Seed Oils of Five Black Tartary Buckwheat Cultivars with Biochemical Characterization and Antioxidant Properties

Zhang Zhen,He Caian,Zhu Ruixue,Shen Jing,Yu Yueli,Peng Qiang,Gao Jinfeng,Li Yunlong,Wang Min

Black tartary buckwheat oils (BTBOs) were extracted from five major industrial tartary buckwheat cultivars grown under similar agronomical activities and environmental conditions. These oils were characterized for the bioactive compounds containing fatty acids, β-carotene, lutein, α-, β-, δ- and γ-tocopherol, and for their antioxidant properties. The total tocopherol contents that were obtained ranged from 704.66 to 1156.19 mg/kg, with γ-tocopherol (588.98–977.91 mg/kg) as the main component. The concentration of lutein ranged from 253.14 to 429.63 mg/kg, which was almost ten times higher than that of β-carotenoid (46.71–69.2 mg/kg), indicating that black tartary buckwheat seed oils were a good source of lutein. The predominant fatty acids were unsaturated oleic acid (C18:1) (35.27–40.61 %) and linoleic acid (C18:2) (38.25–42.90 %). Excellent values of 2,2-diphenyl-1-picrylhydrazyl radical (DPPH), diammonium salt (ABTS) radical scavenging activities were obtained and the highest oxygen radical absorbance capacity (ORAC) value of 13.89 mmol Trolox equiv/g oil was detected in the variety of Chuanqiao No. 1, which was clearly separated by principal component analysis (PCA) on the basis of the highest content of tocopherols and carotenoids. Moreover, the correlation analysis showed that tocopherols and carotenoids were the major contributors to the antioxidant activities of the BTBOs. This study demonstrates that lipophilic extraction in the tartary buckwheat seed contains many interesting bioactive compounds, which are beneficial for human health.

Jianping Chen,Xuehua Chen,Jiarui Li,Baozhen Luo,Tugui Fan,Rui Li,Xiaofei Liu,Bingbing Song,Xuejing Jia,Saiyi Zhong

In the present study, a selenium-chondroitin sulfate (SeCS) was synthesized by the sodium selenite (Na2SeO3) and ascorbic acid (Vc) redox reaction using chondroitin sulfate derived from shark cartilage as a template, and characterized by SEM, SEM-EDS, FTIR and XRD. Meanwhile, its stability was investigated at different conditions of pH and temperatures. Besides, its antioxidant activity was further determined by the DPPH and ABTS assays. The results showed the SeCS with the smallest particle size of 131.3 ± 4.4 nm and selenium content of 33.18% was obtained under the optimal condition (CS concentration of 0.1 mg/mL, mass ratio of Na2SeO3to Vc of 1:8, the reaction time of 3 h, and the reaction temperature of 25 °C). SEM image showed the SeCS was an individual and spherical nanostructure and its structure was evidenced by FTIR and XRD. Meanwhile, SeCS remained stable at an alkaline pH and possessed good storage stability at 4 °C for 28 days. The results on scavenging free radical levels showed that SeCS exhibited significantly higher antioxidant activity than SeNPs and CS, indicating that SeCS had a potential antioxidant effect. Keywords:nanoselenium;chondroitin sulfate derived from shark cartilage;structure characterization;antioxidant activity

Synthesis, characterization and effect of alkyl chain unsaturation on the antioxidant activities of chlorogenic acid derivatives

Yan Xu,Jing Zhang,Teng Pan,Fazheng Ren,Hailing Luo,Hao Zhang

Chlorogenic acid (CGA), a hydrophilic polyphenolic compound, is difficult to use in lipophilic systems. Accordingly, to improve its lipophilicity and explore the effect of alkyl chain unsaturation on antioxidant activity, CGA was acylated with fatty acids of the same chain length, but an increasing degree of unsaturation, namely, stearic acid [C18:0], oleic acid [C18:1], linoleic acid [C18:2], and linolenic acid [C18:3]. The major products were monoesters, as confirmed by ESI-MS, 1 H NMR, 13 C NMR, and Fourier transform infrared spectra (FT-IR) analysis. The lipophilicity of CGA derivatives was enhanced by acylation, and decreased with an increasing degree of fatty acid unsaturation. All CGA derivatives showed a high level of antioxidant activity. In detail, CGA-C18:0, CGA-C18:1, and CGA-C18:2 showed similar antioxidant activities, while CGA-C18:3 had the lowest antioxidant activity. The DPPH radical and ABTS radical cation scavenging activities of CGA-C18:3 were equivalent to 66.9% and 62.0% of those of CGA, respectively. The results of peroxide value (PV) indicated that the oxidative stability of CGA derivatives was enhanced. These results indicated that CGA derivatives could serve as potential sources of antioxidants for lipophilic food systems.

Deep Eutectic Solvents as New Extraction Media for Flavonoids in Mung Bean

Jingyu Gao,Longli Xie,Yu Peng,Mo Li,Jingming Li,Yuanying Ni,Xin Wen

Mung beans contain abundant flavonoids like vitexin and isovitexin, which contribute to their strong bioactivities, such as antioxidant effects, so efforts should focus on extracting bioactive flavonoids as well as aligning with the goal of green extraction for specific applications. Deep eutectic solvent coupled with ultrasound-assisted extraction (DES-UAE) was applied to extract flavonoids from mung beans, and eight different DESs were compared on the extraction yield. In addition, the traditional extraction method with 30% ethanol was performed as the reference. The results showed that ethylene glycol-glycolic acid achieved the highest yield among all the DESs, 1.6 times that of the reference values. Furthermore, the DES-UAE parameters were optimized as a 60 mL/g liquid–solid ratio, 30% water content in DES, 200 W ultrasonic power, 67 °C ultrasonic temperature, and 10 min extraction time, leading to the DES extract with the maximum extraction yield of 2339.45 ± 42.98 μg/g, and the significantly stronger DPPH and ABTS radical scavenging ability than the traditional extract. Therefore, employing DES and ultrasonic extraction together offers a green method for extracting flavonoids from mung beans, advancing the development and utilization of plant-derived effective components in a sustainable manner.

Phenolic compounds and the antioxidant activities in litchi pericarp: Difference among cultivars

Hui-Cong Wang,Zhi-Qun Hu,Yan Wang,Hou-Bin Chen,Xu-Ming Huang

The pericarps of 10 litchi cultivars from two production seasons were studied for their phenolic concentrations and composition and activities of anti-oxidation. The total phenolic concentrations in dried litchi pericarp ranged from 51 to 102   g   k   g −1 . Different cultivars varied considerably in phenolic concentration and composition. Cultivars ‘Nuomici’ exhibited significantly higher concentrations of proanthocyanidins, flavonoids, phenolic acids and therefore total soluble phenolics than the other cultivars, while ‘Lanzhu’ and ‘Guiwei’ had minor values in phenolics. The extracts of litchi pericarp exhibited considerably high ferric reducing antioxidant power (FRAP) and strong activities of 1,1-diphenyl-2-picryhydrazyl (DPPH) scavenging, lipid peroxidation inhibiting, and oxidative DNA damage protection. The crude ethanol extracts of pericarp displayed significantly higher FRAP and stronger DPPH scavenging capacity than butylated hydroxylytoluene (BHT) and catechin. Among the tested cultivars, concentrations of major phenolics had significant linear correlations with FRAP and DPPH scavenging activities. These results indicated that litchi pericarp is rich in natural antioxidant phenolic compounds and different litchi cultivars differed considerably in phenolic concentration and antioxidant activities.

(−)-Epigallocatechin gallate (EGCG) possesses potent antioxidant properties and can enhance the encapsulation efficiency and stability of oil microcapsule with sodium caseinate (NaCas). Hence, the present study aimed to investigate the effects of different concentrations of EGCG on the physicochemical properties of walnut oil microcapsules. The impact of EGCG on the morphology, structure, physical attributes, and functional traits of walnut oil microcapsules was characterized through scanning electron microscopy, X-ray diffraction, Fourier transform infrared spectroscopy, molecular docking, thermogravimetric analysis, etc. Results demonstrated that EGCG interacted with NaCas via hydrogen bonding and hydrophobic interactions to form walnut oil microcapsules, significantly enhancing the encapsulation efficiency up to 86.42%. With increasing concentration of EGCG, the NaCas-EGCG complex displayed heightened DPPH free radical scavenging capacity (up to 68.27 μg Trolox/mL). Furthermore, in vitro digestion indicated that the microcapsules could effectively regulate the release rate of free fatty acids. In conclusion, the present study successfully prepared walnut oil microcapsules with elevated oxidative stability by employing EGCG and NaCas as the wall materials, providing new insight for preparing walnut oil microcapsules with improved stability and release properties.

Curcumin-loaded core-shell biopolymer nanoparticles produced by the pH-driven method: Physicochemical and release properties

Zhenpeng Li,Quanquan Lin,David Julian McClements,Yuying Fu,Hujun Xie,Teng Li,Guowen Chen

In this study, core-shell biopolymer nanoparticles were fabricated for the encapsulation and delivery of curcumin using a pH-driven method. The influences of the coating composition on the physicochemical properties and curcumin release characteristics of the core-shell nanoparticles were studied. Fourier transform infrared spectroscopy and X-ray diffraction analyses indicated that curcumin was encapsulated in an amorphous state inside the nanoparticles. Particle size and ζ-potential measurements indicated that the biopolymer nanoparticles were relatively stable under different environmental conditions: long term storage, heating, pH changes and salt. The DPPH radical scavenging activity of the curcumin was increased after encapsulation within the nanoparticles, whereas the gastrointestinal release of curcumin was prolonged. These results were attributed to the ability of alginate and NaCas to form a thick layer around the nanoparticles, which increased the steric and electrostatic repulsion between them, as well as inhibiting the release of curcumin.

Zwitterionic polymer modified xanthan gum with collagen II-binding capability for lubrication improvement and ROS scavenging

Kai Ren,Xiang Ke,Zhu Chen,Yao Zhao,Lu He,Peng Yu,Jiaqi Xing,Jun Luo,Jing Xie,Jianshu Li

High friction of damaged cartilage requires long-acting lubricated additive, which can also effectively scavenge reactive oxidative species (ROS) produced by mechanically stimulated chondrocytes . In this study, xanthan gum (XG) was grafted by poly (sulfobetaine methacrylate) (PSBMA) (the [XG]/[SBMA] molar ratio is 1:5 or 1:10), forming nanoparticles and then conjugated with collagen II-binding peptide, finally obtaining CBPXGSB1/5 or CBPXGSB1/10. Therein, the CBPXGSB1/5 was chosen as optimal lubricated additive. The results show that hydrated effect of PSBMA side chains endows CBPXGSB1/5 with favorable lubrication property (COF is 0.063). Furthermore, the CBPXGSB1/5 combining lubrication property and specific binding capability together may achieve the long-acting lubrication for injured cartilage in medical field. The CBPXGSB1/5 also possesses antioxidation verified by DPPH assay and exhibits synergistically enhanced ROS ( OH, O 2 − and H 2 O 2 ) scavenging. Besides, cytotoxicity experiment demonstrates that CBPXGSB1/5 has good biocompatibility. Therefore, multifunctional CBPXGSB1/5 developed here may have promising application potential in osteoarthritis treatment.

Evaluation of physicochemical characteristics, bioactivity, flavor profile and key metabolites in the fermentation of goji juice by Lacticaseibacillus rhamnosus

Xin An,Tongtong Li,Jiaxue Hu,Yaoran Li,Huiyan Liu,Haitian Fang,Xiaobo Wei

This study aimed to investigate the changes in physicochemical properties, bioactivities and metabolites of fermented goji juice (FGJ) by Lacticaseibacillus rhamnosus at different fermentation stages. The results showed that Lacticaseibacillus rhamnosus fermentation significantly decreased the content of soluble protein, total phenolic, total flavonoid and total sugar. Meanwhile, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability and the inhibition rate of xanthine oxidase (XOD) activity were remarkably enhanced by Lacticaseibacillus rhamnosus fermentation. Flavor profiles analysis indicated that FGJ produced novel volatile compounds such as 4-methylpentanol and 2-butanol, which provide its distinct aroma. The non-targeted metabolomics analysis showed that the differential metabolites in the FGJ28 vs. FGJ0 group were mainly included 1,7-bis (3,4-dihydroxyphenyl) heptan-3-yl acetate, isoplumbagin, triacetylresveratrol, sulochrin, indole-3-acetaldehyde, etc. , which might have an effect on the promotion of the bioactivity of goji juice. These findings will contribute to understanding the biotransformation effect of Lacticaseibacillus rhamnosus fermentation on goji juice.
