Numerous studies propose that epigallocatechin-3-gallate (EGCG), an abundant polyphenol in green tea, has anti-cancer properties. However, its mechanism of action in breast cancer remains unclear. This study investigated the capacity of EGCG to suppress breast cancer cell growth in vitro and in vivo, characterizing the underlying mechanisms, focusing on the effect of EGCG on glucose metabolism. EGCG reduced breast cancer 4T1 cell growth in a concentration- (10–320 μM) and time- (12–48 h) dependent manner. EGCG induced breast cancer apoptotic cell death at 24 h, as evidenced by annexin V/PI, caspase 3, caspase 8 and caspase 9 activation. Furthermore, EGCG affected the expression of 16 apoptosis-related genes, and promoted mitochondrial depolarization. EGCG induced autophagy concentration-dependently in 4T1 cells by modulating the levels of the autophagy-related proteins Beclin1, ATG5 and LC3B. Moreover, EGCG affected glucose, lactate and ATP levels. Mechanistically, EGCG significantly inhibited the activities and mRNA levels of the glycolytic enzymes hexokinase (HK), phosphofructokinase (PFK), and lactic dehydrogenase (LDH), and to a lesser extent the activity of pyruvate kinase (PK). In addition, EGCG decreased the expression of hypoxia-inducible factor 1α (HIF1α) and glucose transporter 1 (GLUT1), critical players in regulating glycolysis. In vivo, EGCG reduced breast tumor weight in a dose-dependent manner, reduced glucose and lactic acid levels and reduced the expression of the vascular endothelial growth factor (VEGF). In conclusion, EGCG exerts an anti-tumor effect through the inhibition of key enzymes that participate in the glycolytic pathway and the suppression of glucose metabolism.

Jellyfish stings are the most common type of marine life injuries. However, at present, the treatment measures against jellyfish stings are mostly empirical and supportive, with uncertain therapeutic outcomes, and there is a lack of specific antidotes based on the toxic mechanism of jellyfish venom in clinical practice. In our previous study, polyphenol epigallocatechin-3-gallate (EGCG) was found to neutralize the toxicity of jellyfishNemopilema nomuraivenom (NnV) in vivo and in vitro. Herein we further demonstrated that EGCG exerted its antagonistic effect against NnV through inhibiting the oxidative stress, pro-apoptotic proteins, and systemic inflammatory responses. Subsequently, we constructed a polyethylene glycol (PEG)-EGCG/tetracycline hydrochloride (HTC) co-loaded micellar nanocomplex in order to enhance the stability and bioavailability of EGCG in vivo, which successfully integrated the membrane-repair function of PEG, the enzyme inhibitory effect of HTC and the antioxidant properties of EGCG. Notably, this micellar nanocomplex demonstrated significant protective effects against both functional damage and pathological alterations in a non-lethal NnV-envenomed mouse model. When administered 1 h after NnV envenomation, EGCG (40 mg/kg), HTC and PEG-EGCG (containing 40 mg/kg EGCG) only partially improved abnormal blood biochemical indicators and moderately alleviated histopathologic damage, and PEG-EGCG/HTC containing merely 8 mg/kg EGCG completely mitigated the toxic reactions in envenomed mice. In the preventive regimen, the administration of EGCG, HTC or PEG-EGCG 30 min before exposure showed no significant improvement in abnormal blood biochemical indicators and histopathologic damage, while PEG-EGCG/HTC could still significantly improve the functional impairments and histopathologic damage of the heart and liver in NnV-envenomed mice. These findings suggest the clinical translational potential of PEG-EGCG/HTC against jellyfish envenomation.

Background: Prostate cancer (PCa) is characterized by a unique metabolic dependency on the hyperactivated tricarboxylic acid (TCA) cycle, creating a distinct vulnerability to cuproptosis-a copper-dependent form of regulated cell death. However, metabolic plasticity allows tumor cells to evade single-pathway inhibition through compensatory glycolysis, limiting the efficacy of copper-based therapies. Therefore, there is an urgent need to develop integrated nanoplatforms capable of orchestrating a dual-pathway metabolic blockade to overcome this therapeutic resistance. Results: Herein, we engineered a self-assembling copper-epigallocatechin gallate (EGCG) nanoreactor (Cu-EQ NP) designed to execute a synchronized dual-metabolic assault. The Cu-EQ NPs leverage the tumor microenvironment to release copper ions that trigger TCA-dependent cuproptosis, while the EGCG component simultaneously inhibits key glycolytic enzymes to prevent metabolic escape. Mechanistically, this lethality is self-amplified by EGCG-quinone-mediated depletion of glutathione (GSH) and the downregulation of the ATPase copper-transporting beta (ATP7B) efflux pump, leading to irreversible intracellular copper accumulation. Furthermore, we demonstrate that this metabolic collapse is highly immunogenic. In a syngeneic RM-1 mouse model, Cu-EQ NP-induced immunogenic cell death (ICD) released damage-associated molecular patterns (DAMPs) that successfully transformed the immunologically "cold" tumor microenvironment. This remodeling was characterized by enhanced dendritic cell maturation, M2-to-M1 macrophage repolarization, and robust infiltration of cytotoxic CD8 + T cells, resulting in potent tumor regression. Conclusions: The Cu-EQ nanoreactor represents a precision nanomedicine strategy that converts the specific metabolic vulnerabilities of PCa into a fatal weakness. By integrating dual-metabolic disruption with robust immune activation, this platform offers a promising therapeutic paradigm for overcoming resistance in advanced prostate cancer.

EGCG was encapsulated within composite hydrogel beads reinforced with PPI. • An 81.91% encapsulation efficiency was achieved at a 5:5 PPI-to-SA mass ratio. • PPI incorporation delayed EGCG release kinetics. • PPI addition densified the hydrogel network microstructure. Epigallocatechin-3-gallate (EGCG) is a potent bioactive compound with notable physiological benefits. However, its poor stability and susceptibility to degradation in the gastric environment limit its bioavailability. In this study, composite hydrogel beads were developed as a controlled-release delivery system for EGCG. Composite hydrogel beads were fabricated via ionic gelation through the incorporation of peanut protein isolate (PPI) into a sodium alginate (SA) matrix, with varying SA: PPI mass ratios (8:2 to 4:6). As the content of PPI increased, the internal network structure of the hydrogel beads became more compact, and the encapsulation efficiency increased from 33.79% to 81.91%; During simulated in vitro digestion, the beads exhibited pH-responsive swelling behavior. Moreover, compared to the control group (i.e., pure SA beads), the EGCG release rate decreased from 43.27% to 20.38% after 2 h of simulated gastric digestion, demonstrating the composite hydrogel beads' efficacy in controlling EGCG release. Enhancing cancer immunotherapy using methods that induce immunogenic cell death (ICD) can significantly improve its effectiveness and profoundly influence its role as a highly efficient cancer treatment strategy. However, the limited penetration of cytotoxic T cells into tumors, owing to dense tumor fibrosis, remains a significant barrier to immunotherapy. A tumor microenvironment-sensitive intelligent dual-drug delivery system was developed to simultaneously deliver epigallocatechin-3-gallate (EGCG) and doxorubicin (DOX) to mitochondria. EGCG enhanced the mitochondria-targeted action of DOX and increased damage to the mitochondrial electron transport chain which facilitated capturing electrons in the mitochondrial matrix of DOX. Subsequently, DOX molecules form a semiquinone intermediate and electrons are transferred to oxygen to generate reactive oxygen species (ROS) that induce mitochondrial apoptosis. These results indicate that EGCG amplifies the combined effects of chemo/chemodynamic therapy of DOX, demonstrating a pronounced synergistic ICD effect that recruits CD8+ T cells to the tumor microenvironment (TME). In addition, EGCG promotes T-cell infiltration into tumor tissues by inhibiting the transforming growth factor-β signaling pathway, thereby significantly enhancing antitumor efficacy. This study advances the efficacy of immunotherapy through bidirectional synergy, which not only enhances intrinsic tumor immunogenicity but also overcomes the extrinsic physical barriers of tumors, providing a new direction for the development of broadly applicable immunotherapies.

Disulfide bond critically govern gluten structure and its covalent interactions with polyphenols, ultimately determining gluten-polyphenol complex properties. This study investigated how disulfide bond cleavage modulates gluten structure, enhances epigallocatechin gallate (EGCG)-gluten covalent binding, and regulates the physicochemical/digestive properties of their complexes. Sodium metabisulfite treatment (0–25 mM treatment) induced disulfide bonds cleavage, which promoted the structural unfolding and exposed reactive groups for covalent binding with EGCG. With analysis of particle size, fluorescence spectroscopy, fourier-transform infrared spectroscopy, and thermogravimetric analysis, disulfide bond cleavage significantly reduced the particle size (from 124.5 to 62.26 μm) of gluten, altered the secondary (α-helix decreased from 23.4 % to 15.1 %, β-sheet from 32.8 % to 23.7 %) and tertiary structures of gluten, increased its surface hydrophobicity and enhanced its binding efficiency (from 51.98 % to 62.02 %) with EGCG. With increase of disulfide bond cleavage, EGCG-gluten covalent complexes exhibited enhanced thermal, pH, and oxidation stability, along with strong antioxidant activity. In addition, the disulfide bond cleavage of gluten inhibited the premature releasing and degradation of EGCG from covalent complexes during the digestion process, which was beneficial for improving the bioaccessibility of EGCG. All results suggest that the disulfide bond cleavage of gluten improve the physicochemical and digestive properties of EGCG-gluten covalent complexes, which provides a potential approach to regulate the properties of proteins-polyphenols covalent complexes as delivery systems of active compounds for functional foods and nutraceuticals.

Petroleum-based packaging materials raise serious environmental and food safety concerns, driving the development of sustainable smart packaging derived from natural polymers. In this study, natural pigment-zein-chondroitin sulfate-epigallocatechin gallate-ι-carrageenan (NZCEC) nanoparticles were fabricated and incorporated into κ-carrageenan-based films to enhance their functional properties. The effects of NZCEC addition on the films' physicochemical stability, intermolecular interactions, antioxidant activity, and responsiveness to ammonia and acetic acid vapors were systematically evaluated, along with their application in preserving mandarin fish ( Siniperca chuatsi ). Scanning electron microscopy revealed good compatibility between κ-carrageenan and moderately loaded NZCEC nanoparticles. Incorporation of NZCEC significantly improved the film's optical properties (opacity increased from 4.10 to 23.32 A mm −1 ) and reduced its water vapor permeability (from 8.23 × 10 −9 to 6.93 × 10 −9 g m −1 s −1 Pa −1 ). Moreover, the antioxidant activity was markedly enhanced (from 1.75 % to 77.65 %), along with notable responsiveness to ammonia and acetic acid. Storage trials with mandarin fish demonstrated that the smart film effectively delayed fish spoilage. Additionally, the film displayed distinct chromatic variations in response to freshness, allowing continuous assessment of food quality. These results establish NZCEC nanoparticle-reinforced κ-carrageenan films as a promising intelligent packaging material with improved stability, responsiveness, and food preservation performance.

The self-assembled peptide-based hydrogels have been widely utilized in foods . However, the self-assembled hydrogels still have some shortages such as low mechanical properties and hydrogel stabilities. Herein, we designed two co-assembled C 13 -WS hydrogels conjugated by Gallic Acid (CA) and Epigallocatechin Gallate (EGCG). The introduction of CA and EGCG into C 13 -WS hydrogels can significantly improve the mechanical properties and hydrogel stabilities. The storage modulus (G’) values of co-assembled C 13 -WS/CA and C 13 -WS/EGCG were 13.33 KPa and 16 KPa, respectively. From transmission electron microscope (TEM) results, CA and EGCG contributed to the rearrangement of C 13 -WS molecules during co-assembly. Moreover, the self-/co-assembled C 13 -WS hydrogels showed antimicrobial activity against E. coli , P. aeruginosa , and S. epidermidis . The introduction of CA and EGCG significantly enhanced the antibacterial activity of C 13 -WS hydrogels. Our findings provide a novel strategy for the development of stable supramolecular antibacterial short peptide hydrogels through co-assembly.

5 Hydroxymethylfurfural (5-HMF) is a heat-induced hazard that common in baked goods. Although bioactive compounds such as catechins and polysaccharides could inhibit 5-HMF, their inhibition efficiency was limited when used alone. Therefore, this study aimed to explore the synergistic inhibitory impact of catechins and polysaccharides on 5-HMF in biscuits. Epigallocatechin gallate (EGCG), chitosan, and sodium alginate were screened out due to their high maximum inhibition rates (55.90, 67.20 and 66.62%, respectively), and their synergistic inhibitory effects on 5-HMF were investigated by isobologram analysis. The interaction index (γ) varies inversely with the synergism. When γ < 1 or >1, it represented synergism or antagonism, respectively. The results showed the EGCG/chitosan ratios of 1:1 (γ = 0.221) had better synergism than that of 1:2 (γ = 0.671) and 1:3 (γ = 0.523). Moreover, EGCG and sodium alginate only showed a synergistic effect at ratio of 1:1 (γ = 0.358), while an antagonistic effect was observed at ratios of 1:2 and 1:3 (γ = 1.12 and 1.25, respectively). The optimal inhibitory effect (54.57%) was achieved when EGCG-sodium alginate (1:1) at 0.2 g/kg. Compared with those used alone, the co-addition of EGCG and chitosan could decrease the dosage of EGCG to 1/5 and that of chitosan to 1/7, indicating a significant improvement of inhibit efficiency. Finally, Inhibitors’ concentration correlated negatively with 5-HMF content and browning index, suggesting they might inhibit 5-HMF by hindering Maillard or caramelization reactions. This study provided an efficient and cost-effective solution for 5-HMF control in baking products.

Probiotics are often subjected to adverse factors during processing, storage and digestion. To enhance the viability and function of probiotics, whey protein concentrate (WPC)-based microcapsules were systematically fabricated to co-encapsulate probiotic Lactiplantibacillus plantarum KLDS 1.0328 (LP KLDS 1.0328) and epigallocatechin gallate (EGCG) by electrospray. Scanning electron microscopy showed that spherical and smooth particles were developed. Attenuated total reflectance-Fourier transform infrared spectroscopy and X-ray diffraction suggested that probiotics and EGCG were successfully encapsulated in WPC-based microcapsules. Thermogravimetric analysis revealed that the addition of EGCG and probiotics improved the thermal stability of WPC-based microcapsules. Furthermore, the incorporation of polysaccharides and polyphenol significantly enhanced the antioxidant activity of the microcapsules ( P < 0.05). The viability of probiotics encapsulated in WPC/polysaccharides microcapsules was elevated compared with that of encapsulated in WPC microcapsules under simulated gastrointestinal digestion, thermal treatments and NaCl stresses. Notably, WPC/β-CD/EGCG/LP KLDS 1.0328 exhibited better protection against these environmental stresses. Moreover, the incorporation of polysaccharides could maintain viability over 7.50 lg CFU/g after 28 d of storage. Furthermore, WPC/β-CD/LP KLD 1.0328/EGCG microcapsules exhibited the highest scores by the results of principal component analysis. This study provides valuable insights for protecting probiotics with appropriate polysaccharide-protein composites and developing functional foods with probiotics.

The protein degradation, flavor and micromolecule metabolites changes of (−)-epigallocatechin gallate (EGCG)-gelatin biofilm treatment (EGT) on chilled tilapia fillets in 21 days were investigated. Morphology observations revealed EGT protected good connective myofibrillar protein. It maintained protein secondary structure by significantly increasing the proportion of α-helix (15.20%) and decreasing the ratio of random coils (22.02%) in the EGT group compared to the control (CON) group ( P < 0.05). Metabolomics with UHPLC-Q-TOF/MS analysis indicated a distinct separation between the CON and treatment groups at the end of storage. Small peptides analysis demonstrated that the EGT group increased the level of sweet peptides. Additionally, the EGT group significantly reduced the formation of amino acid derivatives and esters and off-flavor development. Overall, EGT effectively improved flavor, inhibited fish protein oxidation/degradation, and verified metabolomics results. This study unveiled the potential of metabolomics to analyze metabolites determined by tilapia and monitor the changes during storage.

Epigallocatechin gallate (EGCG) has shown antiviral potential against the hepatitis B virus (HBV) by restoring lysosomal acidification; however, its application is limited by poor stability and low intestinal permeability. Here, a dual-shell chitosan/ferritin nanosystem (CHE) was developed to enhance the EGCG delivery and functionality. CHE achieved an encapsulation efficiency of 13.61% with a uniform nanoscale size distribution. Compared with free EGCG and single-shell HE, CHE improved storage stability and delayed EGCG release under simulated gastrointestinal conditions. In a Caco-2/HepG2.2.15 or HepAD38 coculture model, CHE enhanced transepithelial transport and intracellular delivery while maintaining epithelial barrier integrity. Mechanistically, CHE promoted lysosomal acidification, increased cathepsin B maturation, and restored autophagic flux, accompanied by reductions in HBV DNA, pgRNA, HBsAg, and HBeAg levels. These findings demonstrate that a dual-shell nanocage integrates stability enhancement with functional intracellular modulation, providing a promising strategy to improve the antiviral efficacy of polyphenols.
