Low doses of antibiotics are ubiquitous in the marine environment and may exert negative effects on non-target aquatic organisms. Using primary macrophages of common carp, we investigated the mechanisms of action following exposure to several common antibiotics; cefotaxime, enrofloxacin, tetracycline, sulfamonomethoxine, and their mixtures, and explored the immunomodulatory effects associated with the nuclear factor-κB (NF-κB) signaling pathway. A KEGG pathway analysis was conducted using the sixty-six differentially expressed genes found in all treatments, and showed that exposure to 100 μg/L of antibiotics could affect regulation of the NF-κB signaling pathway, suggesting that activation of NF-κB is a common response in all four classes of antibiotics. In addition, the four antibiotics induced nf-κb and NF-κB-associated cytokines expression, as verified by qPCR, however, these induction responses by four antibiotics were minor when compared to the same concentration of LPS treatment (100 μg/L). Antagonists of NF-κB blocked many of the immune effects of the antibiotics, providing evidence that NF-κB pathways mediate the actions of all four antibiotics. Moreover, exposure to environmentally relevant, low levels (0.01–100 μg/L) of antibiotics induced a NF-κB-mediated immune response, including endogenous generation of ROS, activity of antioxidant enzymes, as well as expression of cytokine and apoptosis. Moreover, exposure to mixtures of antibiotics presented greater effects on most tested immunological parameters than exposure to a single antibiotic, suggesting additive effects from multiple antibiotics in the environment. This study demonstrates that exposure of fish primary macrophages to low doses of antibiotics activates the NF-kB pathway.

Sardinella zunasi Bleeker, an edible and medicinal marine fish, is largely distributed in tropical oceans. However, the chemical composition and nutritional properties of this species have not yet been investigated. In the present study, proximate composition, fatty acids, amino acids, taurine, and minerals of S. zunasi Bleeker were characterized, and the immunostimulatory properties of its glycoprotein were evaluated. The results indicated the presence of crude protein (19.66%), crude lipid (6.29%) and carbohydrate (0.74%) in S. zunasi Bleeker; monounsaturated fatty acids and polyunsaturated fatty acids in the fatty acid composition of S. zunasi Bleeker were 25.00% and 31.01%, respectively; S. zunasi Bleeker was rich in taurine (219 mg/100 g) and essential amino acids (5.57 g/100 g). In addition, the glycoprotein of S. zunasi consisted of protein and sugars, with a total content of 34.25% and 16.27%, respectively. The glycoprotein showed significant effects on promoting NO, TNF-α and IL-6 in a dose-dependent manner in RAW264.7 macrophage cells. Thus, these findings provide a scientific basis for the further utilization of glycoprotein from S. zunasi Bleeker.

A rapid, specific and sensitive method based an immunochromatographic strip test has been developed for the determination of sodium nifurstyrenate (NFS) residues in fish. Sample preparation procedure includes ultrasonic bath extraction with acetonitrile, sample centrifugation at 8000 rpm for 10 min, filtration through the 0.22 µm membrane filter, and drying over nitrogen (60°C) and res-suspension. The antigen (NFS-ovalbumin) and a goat anti-mouse IgG antibody were drawn onto a nitrocellulose membrane as the test line and control line, respectively. Under optimized conditions, the NFS cut-off limits for test strips was determined as 5 ng/mL in 0.01 M phosphate buffered saline and 10 ng/mL in fish. Results were obtained within 5 min. Our data suggests this immunochromatographic assay can be used for the sensitive, rapid, and specific on-site screening for NFS in fish.

In this paper, a simple and effective fluorescent system was fabricated for sensitive detection of terramycin using ethylenediamine functionalized MoS 2 quantum dots (EDA-MoS 2 QDs) as probes. The monodisperse EDA-MoS 2 QDs were synthesized by microwave-assisted one-step bottom-up strategy. The obtained EDA-MoS 2 QDs have excellent water solubility and ultra-small sizes with an average diameter of 1.72 nm. In addition, they show strong fluorescence with high fluorescence quantum yield (22.62%). The existence of terramycin can effectively quench the fluorescence of EDA-MoS 2 QDs based on intermolecular charge transfer and aggregation-induced quenching mechanism. The fluorescence quenching efficiency is linearly dependent on the concentration of terramycin. The EDA-MoS 2 QDs exhibit high sensitivity for terramycin detection with a detection limit of 0.5 nM. Moreover, the EDA-MoS 2 QDs modified filter papers quickly determine the trace amounts of terramycin in fresh fish. The fluorescence quenching-induced color change can be observed by the naked eye under the UV light. The sensor system based on EDA-MoS 2 QDs has the good analytical performance, and can be applied to environmental water and food security monitoring.

BACKGROUND Roasted fish enjoys great popularity in Asia, but how roasting and subsequent digestion influence the oxidation and proteolysis of fish meat is unknown. This study aimed to investigate the effect of roasting time on lipid and protein oxidation and their evolution and consequence on proteolysis during simulated digestion of fish fillets. RESULTS Several oxidation markers (thiobarbituric acid-reactive substances (TBARS), free thiols, total carbonyls and Schiff bases) were employed to assess the oxidation of fish. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and the 2,4,6-trinitrobenzenesulfonic acid (TNBS) assay for free amino groups were used to study the proteolysis during gastrointestinal digestion. The results showed that significant lipid and protein oxidative changes occurring in roasted fish fillets were reinforced after gastric digestion and were much more intense after intestinal digestion. Throughout roasting and digestion, a close interconnection between lipid and protein was also manifested as the levels of total carbonyls and Schiff bases rose while TBARS fell. Furthermore, free amino groups decreased with prolonged roasting time, signifying that protein oxidation before digestion resulted in impaired proteolysis during digestion. CONCLUSION This study indicated that the lipid and protein oxidation of fish fillets could be dependent on time of roasting, and the oxidation continued to develop and have an impact on proteolysis during in vitro digestion. © 2018 Society of Chemical Industry

A photonic crystal (PC) sensor was prepared through molecular imprinting by using dispersed SiO 2 microspheres as PC, sulfaguanidine (SG) as template, methacrylic acid as monomer, and ethylene glycol dimethacrylate as crosslinker. The preparation conditions were optimized as follows: molar ratio of template, monomer, and crosslinker was 1:10:4; pH was 6.0 in phosphate buffer; and volume fraction of methanol was 15%. The relationship between wavelength shift of reflection peak and SG concentration was found as: Δλ = 7.8887 lg(C) + 79.9664. The response time was only 5 min. The limit of detection was 2.8 × 10 −10 mol L −1 . The sensor exhibited higher specificity for SG than its sulfonamide analogs. The sensor maintained original sensing performance after five cycles of usage. The recoveries of SG ranged from 93.8% to 111.2% in lake water and from 88.5% to 115.2% in fish samples. This finding suggested that the sensor can be used in food samples with complicated matrix.

Seafood consumption is widely considered as the primary route for human exposure to the neurotoxin methylmercury (MeHg) that is produced by certain anaerobic microorganisms and can bioaccumulate to high concentration levels in natural aquatic food webs. In this study, a novel methylmercury-binding peptide with seven amino acids was displayed on the cell surfaces of Escherichia coli strain W-1, which was isolated from fish feces and fused with ice nucleation protein. These cells exhibited high affinity and selectivity toward methylmercury. They efficiently removed more than 96% of 12 μM methylmercury, and accumulation of methylmercury in the engineered strain was four times higher than that in the wild type. Transmission electron microscopy confirmed methylmercury accumulation on cell membranes. Carassius auratus was fed by engineered bacteria, which showed a decrease in methylmercury concentration in muscles of about 36.3 ± 0.7%; whereas an increase in methylmercury concentration was observed in the feces (36.7 ± 0.8%) in comparison to the control group. The engineered strain in the gut captured methylmercury and prevented it’s absorption by muscles, while some bacteria with methylmercury were excreted in the feces. The surface-engineered E. coli effectively protected fish from methylmercury contamination.

A facile and practical ratiometric fluorescence probe based on two CdTe quantum dots (QDs) coated with molecularly imprinted polymers (MIPs) was prepared for the detection of trace malachite green (MG) in fish. Two CdTe QDs coated with MIPs were fabricated by a one-pot method using MG, (3-aminopropyl) triethoxysilane (APTES) and tetraethyl orthosilicate (TEOS) as template, functional monomer, and cross-linker, respectively. CdTe QDs with λ em 530 nm (gQDs) and 630 nm (rQDs) were used as the referential fluorophore and target sensitive fluorophore, respectively. The fluorescence intensity of gQDs remained unchanged in the presence of MG, while the fluorescence of rQDs could be quantitatively quenched by MG based on the strategy of fluorescence resonance energy transfer. The ratiometric fluorescence probe (MIPs@gQDs&rQDs) was characterized by transmission electron microscopy and Fourier transform infrared spectroscopy. The linear range of MG detection was 0.1–32 μmol L −1 with a detection limit of 8.8 μg kg −1 . The constructed probe has been successfully applied to the detection of MG in fish with the recoveries of 92.3–109.1%, which were validated by the method of HPLC. The result indicated that the probe possessed rapid response, wide linear range, high sensitivity, and relatively high selectivity, and was low-cost and easy in operation in the detection of MG in fish samples.

A highly selective colorimetric detection method for putrescine on the basis of an optimized derivatization reaction was established. With the aids of o -phthalaldehyde (OPA) and thioglycolic acid (TGA), putrescine was derived to become red derivatives (PUT-RD), the form that can be detected qualitatively and quantitatively by visual inspection and UV-vis spectrophotometer at λ max of 490 nm, respectively. Key parameters affecting the experiment were investigated by one-factor-at-a-time and response surface analysis. At the optimal condition, this colorimetric method achieved good linearity at putrescine concentrations ranging from 0.8 to 200 μM with detection limit of 0.44 µM. The method also has good selectivity when common amino acids and inorganic ions, as well as ethylenediamine, 1,3-propanediamine, 1,5-pentanediamine, and 1,6-hexanediamine were used as interferences. The established colorimetric method was successfully employed for the detection of putrescine in 10 commercial fish products.

In this study the waste fish scales (FS) were dissolved in 1-butyl-3-methylimidazolium acetate ionic liquid to obtain a valuable product of hydroxyapatite (HAp). The HAp was obtained in the yield of 32 ± 2%. The obtained HAp was characterized using Fourier Transform Infrared Spectroscopy (FTIR), Powder X-rays Diffraction (PXRD), Thermal Gravimetric Analysis (TGA), Field Emission Scanning Microscopy (FE-SEM), Energy Dispersive X-rays spectroscopy (EDX), and Brunauer–Emmett–Teller (BET). The results of FTIR and XRD showed the characteristic peaks of the HAp. The thermal degradation temperature of the extracted HAp was relatively high. Furthermore, low weight loss was measured which confirmed the removal of organic part of FS during ionic liquid treatment. The FE-SEM result showed the particles with different morphologies and EDX analysis showed a Ca/P ratio of 1.60 for the extracted HAp. The biocompatibility of the extracted HAp was assessed through MTT cell viability assay using known Human Embryonic Kidney 293 cells (HEK cells) and epidermoid carcinoma cells (A431 cells).

This work made the first attempt to construct a florescence assay based on CDs@FMs (carbon dots-based fluorescence microspheres) for ultrasensitively probing malachite green (MG). Through static quenching and inner filter effect (IFE), the 581-nm emission peak of CDs@FMs could be significantly suppressed by MG. Our results demonstrated two good linear fluorescence responses for MG in the concentration scopes of 0.01–0.1 μM and 0.1–10.0 μM, respectively. Strikingly, the detection limits (LODs) achieved were down to picometer (56.7 pM) and nanometer levels (0.57 nM). Furthermore, this method exhibited good consistency with the HPLC results when applied for quantitatively screening MG from fish samples. Our current research highlights a promising application of CDs@FMs for enhanced MG probing in aquatic products with the merits of rapidity, simplicity, high accuracy and stable signal readout.

Unlike predictability in mammals, the responses of PPARα activation treatment in fish varies between different fish species. Existing results indicated that the varied effects may result from fish feeding habits difference within their varied dietary protein content, while the exact mechanism remains obscure. In this study, we conducted an in vivo 8-week trial of low protein 30 % (LP) or high protein 38 % (HP) diet fed tilapia (8.83 ± 0.02 g) supplemented with or without fenofibrate and in vitro 24-h of BCAA (leucine, valine and isoleucine) treated primary cell trial, aiming to reveal the unknown mechanism. The in vivo results showed that, in LP group, fenofibrate significantly increased serum levels of TG, glucose, AST, ALT, MDA and liver TG ( P < 0.05), while decreased liver, muscle and serum BCAA, liver mitochondria quantity, glucose tolerance ( P < 0.05). Moreover, fenofibrate notably lifted the hepatic bckdha , bckdhb , mff , fis , fas and pparγ expression ( P < 0.05), while decreased genes expression of pgc1α , tfam , parkin , pink , foxo3a, sod2, cat , ir and irs ( P < 0.05). No great impact was found in aco , cyp4a3 and cytb ( P > 0.05). In HP group, fenofibrate improved lipid and glucose metabolism along with unaltered BCAA, increased expression of aco , cytb , cpt1a , pparβ , foxo3a , sod2 , nrf1 , mfn1 and fis ( P < 0.05). In vitro findings revealed that the PGC1α and PPARα could be activated by BCAA, particular for leucine ( P < 0.05). Taken together, we conclude that: 1) increased lipid deposition, lipid peroxidation, liver damage and glucose intolerance in fenofibrate-LP group could be ascribed to decreased BCAA and done through PGC1α/FOXO3, PGC1α/MQC regulation; 2) improved lipid-lowering and glucose tolerance in fenofibrate-HP group would be attributed to the enhanced PGC1α/MQC, organelles proliferation and fatty acid β-oxidation.
