Fluorescence method for quickly detecting ochratoxin A in flour and beer using nitrogen doped carbon dots and silver nanoparticles

Chengke Wang,Rong Tan,Dan Chen

In this paper, a FRET (Forster resonance energy transfer) based fluorescence method was developed for the quickly detection of ochratoxin A (OTA) in agricultural products ( e.g. , flour and beer). A highly fluorescent nitrogen doped carbon dots (CD) were served as energy donor, the DNA and MCH (6-mercapto-1-hexanol) modified Ag nanoparticles were served as energy acceptor in the FRET system. OTA can be detected in a concentration range between 10 and 5000 nM, the limit of detection is 8.7 nM. This method has three advantages: (1) an enhanced fluorescent intensity can be acquired by utilizing the nitrogen doped CD synthesized by one-step approach without sophisticated modification of nanoparticles; (2) OTA detection was accomplished quickly (less than 30 min) by using MCH as assistant molecule; (3) an extended OTA detection linear range was acquired, which may facilitate the OTA detection in real agricultural samples, and is helpful for solving food safety problems.

Synthesis and transport characteristics of multi-color fluorescent Silica Nanoparticle Tracer in sandstone cores

Fang-Jun Jia,Hai-Tao Li,Zi-Min Liu,Xin Ma,Guo-Qiang Zhang,Qi-Hui Zhang,Jie Li

As an efficient monitoring and prediction tool, chemical tracers have been widely applied in reservoir characterization, production monitoring, water resources monitoring, and various other fields. Chemical tracer technology is characterized by high efficiency, high precision, relatively simple operational procedures, and low cost. Owing to the limitations of existing tracers, such as minimal options, limited transport efficiency, and complex detection methods, this study used fluorescein isothiocyanate and a ruthenium complex (Ru(phen 3 ) 2+ ) to synthesize 50 nm multi-color fluorescent silica nanoparticle tracers using an improved Stöber method based on fluorescence resonance energy transfer (FRET). Due to the FRET between the two compounds, the synthesized tracer exhibited the characteristics of multi-color fluorescence, and its fluorescent color varied with the mixing ratio of the two precursor solutions. The fluorescence intensity of the synthesized tracer was significantly higher than that of the monochromatic fluorescent nano-tracer. Fourier-transform infrared spectroscopy, ultraviolet spectrophotometry, and fluorescence spectrometry were used to characterize the structure, maximum absorption wavelength, and fluorescence characteristics of the synthesized tracer, respectively. The experimental results show that the synthesized tracer has a maximum absorption wavelength of 450 nm and an emission wavelength of 576 nm. Under field emission scanning electron microscopy, the tracer appears as uniformly spherical particles with a size of 50 nm (±5). It exhibited good dispersibility and fluorescence characteristics in reservoir environments that varied in temperature (25∼85 °C) and salinity (1000∼10000 mg/L). The effects of environmentally sensitive clay minerals, tracer particle size, injection concentration, fluid salinity, and flow rate on the transport characteristics (retention) of tracers in sandstone cores were studied using 12 sets of tracer breakthrough experiments. The experimental results showed that increased sensitivity to clay minerals, salinity, and tracer particle size were not conducive to tracer migration in the core. In contrast, increased tracer injection concentration and flow rate were beneficial to tracer migration in the core.

A FRET-based ratiometric fluorescent chemosensor for mitochondrial ONOO⁻ detection with application to Rheumatoid Arthritis

Jianhong Liu,Xinyue Zhao,Li Zhang,Haowen Zhou,Xudong Li,Guorui Zheng,Xibin Zhou,Yongjin Peng,Jie Zhang

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by progressive joint destruction, where early diagnosis is crucial for impeding disease progression. Peroxynitrite (ONOO⁻), a key reactive nitrogen species, is critically involved in the pathogenesis of RA and represents a promising diagnostic target for early detection. Herein, we developed MRN, a mitochondria-targeting ratiometric fluorescent probe based on a Förster resonance energy transfer (FRET) mechanism for the dynamic monitoring of ONOO⁻ in RA inflammatory models. The probe integrates a rhodamine-naphthalimide FRET pair and a hydrazide group as the specific recognition unit, which facilitates a rapid, ONOO⁻-triggered ratiometric response within 30   seconds and achieves a detection limit of 7.5   nM. Furthermore, the conjugation of a triphenylphosphonium group ensures precise mitochondrial localization. Cellular assays demonstrated excellent biocompatibility and specific mitochondrial targeting. We successfully applied MRN for real-time ratiometric imaging of both endogenous and exogenous ONOO⁻ fluctuations in live BV-2 cells. Importantly, in a Complete Freund's Adjuvant (CFA)-induced rheumatoid arthritis mouse model, MRN enabled in situ ratiometric fluorescence imaging of ONOO⁻ at articular sites, revealing a strong positive correlation between the fluorescence signal intensity and the degree of inflammatory severity. This work establishes MRN as a novel and robust molecular tool for the early diagnosis of RA.

Silver nanoclusters based FRET aptasensor for sensitive and selective fluorescent detection of T-2 toxin

Imran Mahmood Khan,Sen Zhao,Sobia Niazi,Ali Mohsin,Muhammad Shoaib,Nuo Duan,Shijia Wu,Zhouping Wang

T-2 is a trichthecenes mycotoxin responsible for severe food poisoning and toxic influences in humans and animals. Therefore, a rapid and sensitive detection of T-2 is an urgent need to protect the public health mainly caused by the consumption of contaminated agricultural products. Herein, we have prepared one pot synthesis of aptamer functionalized silver nanoclusters (apt-AgNCs) for fluorescent detection of T-2 that relies on novel fluorescence resonance energy transfer (FRET). Apt-AgNCs, act as energy donor, were applied as fluorescent sensing probe whose fluorescence was quenched by MoS 2 employed as fluorescent acceptor. Introduction of target in proposed bioassay leads to desorption of Apt-AgNCs from MoS 2 , and recovery of fluorescence in a toxin concentration dependent-manner. A dynamic quantification range was 0.005–500 ng mL −1 while the LOD was 0.93 pg mL −1 . Spiking analysis reveals the efficiency and robustness of the developed assembly, with recovery percentages ranged from 89.46% to 102.08% and 90.41% to 107.75% in maize and wheat samples, respectively. The results were cross-verified using standard commercial T-2 EISA kit. In conclusion, developed bioassay may find good utility in testing for risk assessment of T-2 toxin.

In Situ Probing Intracellular Drug Release from Redox-Responsive Micelles by United FRET and AIE

Xuelin Wang,Juanjuan Li,Qi Yan,Yanrui Chen,Aiping Fan,Zheng Wang,Yanjun Zhao

Redox-responsive micelles are versatile nanoplatforms for on-demand drug delivery, but the in situ evaluation of drug release is challenging. Fluorescence resonance energy transfer (FRET) technique shows potential for addressing this, while the aggregation-caused quenching effect limits the assay sensitivity. The aim of the current work is to combine aggregation-induced emission (AIE) probe with FRET to realize drug release assessment from micelles. Tetraphenylethene (TPE) is selected as AIE dye and curcumin (Cur) is chosen as the model drug as well as FRET receptor. The drug is covalently linked to a block copolymer via the disulfide bond linker and TPE is also chemically linked to the polymer via an amide bond; the obtained amphiphilic polymer conjugate self-assembles into micelles with a hydrodynamic size of ≈125 nm. Upon the supplement of glutathione or tris(2-carboxyethyl)phosphine) trigger (10 × 10 −3 m ), the drug release induces the fluorescence increase of both TPE and Cur. Accompanied with the FRET decay, absorption enhancement and particle size increase are observed. The same phenomenon is observed in MCF-7 cells. The FRET–AIE approach can be a useful addition to the spectrum of available methods for monitoring drug release from stimuli-responsive nanomedicine.

Homo-FRET mediated antenna effect on the DNA tetrahedron for hepatitis C virus DNA detection

Yu Zou,Xu Zhang,Chen Yue Lin,Shu Jun Zhen,Lei Zhan,Cheng Zhi Huang

Home-Förster resonance energy transfer (Homo-FRET) has been widely utilized in artificial light-harvesting systems to construct antenna structures with enhanced energy-capture efficiency. Although these systems typically exhibit remarkable antenna effects and signal amplification capabilities, their application in detection systems remains limited. Herein, we reported a rationally designed homo-FRET mediated three-donor/one-acceptor (3D-A) antenna nanostructure assembled on a rigid DNA tetrahedron scaffold for the quantitative detection of Hepatitis C virus (HCV) DNA. Robust homo-FRET interactions among the three donors significantly amplified the hetero-FRET efficiency to the single acceptor, generating an enhanced antenna effect. Upon target recognition, the system provided a ratiometric readout, where the fluorescence intensity ratio between Cy5 and Cy3 increased proportionally with HCV DNA concentration. The proposed 3D-A nanoprobe exhibited a linear response range from 1 to 100 nM with a detection limit of 65 pM. Furthermore, by simply altering the target-binding sequence, this system offered a high versatile platform for universal DNA detection detection.

Virus Mimetic Shell-Sheddable Chitosan Micelles for siVEGF Delivery and FRET-Traceable Acid-Triggered Release

Shengyu Zhang,Ye Gan,Lanlan Shao,Tianqing Liu,Danyi Wei,Yanyan Yu,Hongwei Guo,Hongyan Zhu

Targeting vascular endothelial growth factor (VEGF) using small interfering RNA (siVEGF) has shown great potential in inhibiting the growth, proliferation, and migration of tumors by reducing the proliferation of blood vessels. On the basis of bionic principles, a novel pH-responsive and virus mimetic shell-sheddable chitosan (CS) micelles (CMs) as siRNA delivery system was introduced in this study. The cyclo(Arg-Gly-Asp-d-Phe-Lys) (cRGD) modified poly(enthylene glycol) (PEG) was conjugated to the HA2 modified chitosan via a hydrazone linkage (cRGD-PEG-Hz-CS-HA2). The cRGD-PEG-Hz-CS-HA2 conjugate could form micelles by interacting with the complex of octanal, Boc-l-lysine, and 9-d-arginine (9R) (octyl-Lys-9R) as a hydrophodic core forming agent, termed as cRGD-PEG-Hz-CS-HA2/octyl-Lys-9R (abbreviated as cRGD/HA2/Hz-CMs).The CMs modified with cRGD can accurately target glioma cells (U87MG cells) with high expression of αvβ3. The payloads of siVEGF were packed into the core of cRGD/HA2/Hz-CMs via electrostatic interaction and hydrophobic interaction. The intracellular cargo release was achieved by the pH-responsive lysis of the hydrazone bond in acidic environment of endosome. Moreover, the exposed HA2, as a pH-sensitive membrane-disruptive peptide, assists the escape of the carriers from endosome into cytosol. In addition, cRGD/HA2/Hz-CMs can effectively deliver siVEGF and silence VEGF gene expression in U87MG cells, leading to the significant tumor growth inhibition. This study demonstrates that cRGD/HA2/Hz-CMs can deliver and release siVEGF in a controlled manner, which was traced by the fluorescence resonance energy transfer (FRET) system in order to achieve RNAi-based anti-angiogenic treatment of cancer in vivo.

A new FRET-based ratiometric probe for fluorescence and colorimetric analyses of adenosine 5′-triphosphate

Nianqi An,Qiang Zhang,Jing Wang,Cui Liu,Luqing Shi,Lihua Liu,Ludi Deng,Yan Lu

This paper reports a new ratiometric probe for the detection of adenosine 5′-triphosphate (ATP) that is based on the binding-induced modulation of fluorescence resonance energy transfer (FRET) coupled with the aggregation-caused quenching (ACQ) sensing mechanism. The introduction of coumarin fluorophores as FRET donors into the side-chain of polythiophene (PT) as the FRET acceptor provides the ratiometric probe. Upon addition of ATP, the formation of a polymer/ATP complex leads to the conformational changes of the PT backbone from a random coil to the planar conformation and aggregation. As such, the ratiometric ATP detection can be achieved by taking advantage of spectral overlap changes to modulate the FRET efficiency from coumarin fluorophores to the PT backbone before and after the addition of ATP, coupled with ACQ. The probe exhibited a clear seesaw-type dual-emission signal change upon binding with strong affinity (Kapp = 3.12 × 104 M−1) to ATP in aqueous solution, whereas no obvious emission changes were observed with ADP, AMP or various other anions. The probe can also be utilized for naked-eye detection of ATP and real-time monitoring of the ATP hydrolysis reaction by apyrase under physiological pH conditions.

A graphene oxide-based FRET sensor for rapid and specific detection of unfolded collagen fragments

Xiuxia Sun,Jun Fan,Yuping Zhang,Hongli Chen,Yongqing Zhao,Jianxi Xiao

The unstructured collagen species plays a critical role in a variety of important biological processes as well as pathological conditions. In order to develop novel diagnosis and therapies for collagen-related diseases, it is essential to construct simple and efficient methods to detect unfolded collagen fragments. We therefore have designed a FITC-labeled collagen mimic triple helical peptide, whose adsorption on the surface of GO effectively quenches its fluorescence. The newly constructed GO/FITC–GPO complex specifically detects unstructured collagen fragments, but not fully folded triple helix species. The detection shows a clear preference for the collagen targets with complementary GPO-rich sequences. The conformation-sensitive, sequence-specific GO-based approach can be applied as an efficient biosensor for rapid detection of unfolded collagen fragments at nM level, and may have great potential in drug screening for inhibitors of unfolded collagen. It may provide a prototype to develop GO-based assays to detect other important unstructured proteins involved in diseases.

Surface enhanced FRET for sensitive and selective detection of doxycycline using organosilicon nanodots as donors

Shuyi Huang,Long Yu,Pengchen Su,Tao Wen,Mingtai Sun,Dejian Huang,Xiangke Wang,Suhua Wang

In order to achieve on-site fluorescent detection of antibiotic doxycycline (DOC) residue, a new kind of fluorescent organosilicon nanodots (OSiNDs) was successfully constructed by a one-pot strategy and used as a novel ratiometric fluorescent probe . This method could enhance the fluorescence of DOC by conformational fixation on the surface of OSiNDs, and subsequently induced fluorescence resonance energy transfer (FRET). Control experimental results showed that phenylamine functional groups on the as-prepared OSiNDs possessed favorable interactions with DOC by hydrogen bond/π–π stacking, leading to both conformational change of DOC molecules and fluorescence resonance energy transfer process from OSiNDs to DOC. This method was then adapted and optimized for visual sensitive and selective detection of DOC based on ratiometric fluorescence signal, displaying a good linear range for DOC in the range of 1–35 μM with an excellent limit detection of ∼80 nM. Other tetracycline derivatives (TCs) and possible interferences have little influence on the determination of DOC using the method. The spike and recovery test of DOC in real water samples demonstrated the feasibility of the OSiNDs for DOC assay in practical samples and a test paper was fabricated with OSiNDs loading, achieving easy and visual detection of DOC by naked eyes.

Color-Tunable Carbon Dots with Aggregation-Induced Emission Constructed by FRET between Surface Luminescence Centers

Yijie Wang,Shengju Zhou,Shengmei Pan,Xiaofeng Sun,Jin Zhou,Hongguang Li

As a class of new fluorescent materials, carbon dots (CDs) have drawn widespread attention by virtue of their tunable photoluminescence (PL), excellent biocompatibility, and novel physicochemical properties. Unfortunately, in solid state, most CDs experience serious aggregation-caused quenching (ACQ). In this work, color-tunable CDs with aggregation-induced emission (AIE) are prepared by simply changing the filling ratio of the autoclave. The obtained CDs show dual emission (green/red) in solid state, and the fluorescent color can be tuned from green to red. These CDs all exhibit blue fluorescence in dilute solution, and aggregation-induced redshift behaviors are observed when the CDs form aggregates. Structural and optical characterizations prove that blue fluorescence is contributed by the carbon cores, whereas the green/red emission is attributed to the surface groups. Multicolor fluorescence is caused by increased fluorescence resonance energy transfer (FRET) efficiency among the dual emission (green/red) peaks from g-CDs to r-CDs. Surprisingly, delayed fluorescence (DF) of CDs are brightened by inserting the CDs into poly(vinyl alcohol) (PVA) films, which can be easily recognized by the naked eyes with a lifetime up to ms. Finally, owing to the unique AIE and DF properties, the four CDs can be applied to multimodal advanced anti-counterfeiting and information encryption.
