Zhikang Li,Gengyu Han,Boqing Jia,Jijian Lu,Changxu Zhang,Bin Wang,Jiaxiang Wang,Kang Zhao,Muhammad Afzal Khan Qureshi,Lan Yu,Min Li,Guoxi Luo,Tong Wang,Qijing Lin,Weishi Li,Libo Zhao,Yumeng Xue

Gelatin methacryloyl (GelMA) hydrogels are promising for wearable tactile sensors due to their biocompatibility, tissue‐mimic softness and conductivity, yet restricted by mechanical property, structure damage, dehydration, and freezing during usage. Herein, a mechanically robust, self‐healable, moisturizing, cryotolerant and adhesive GelMA hydrogel is developed by incorporating polyvinyl alcohol, acrylic acid, sodium tetraborate, and glycerol into GelMA matrix via a photoinitiated polymerization process. This unique approach overcomes the chemical‐mechanical conflictions among different molecules, and contributes to remarkable stretchability (≥220%, 5.5‐fold higher), elasticity (≈91.9% recovery), autonomous self‐healing within wide temperature range (from ‐40°C to 25°C), adhesion (≥23.5 kPa), moisture retention (≥81% after 10 d) and anti‐freezing (‐55.4°C) properties, significantly outperforming previous GelMA. Based on this, an omni‐healable ionotronic pressure sensor is constructed, featuring high sensitivity within broad pressure range (0–25 kPa), rapid recovery time, long‐term stability (≈120 h), excellent durability and performance self‐repairing (≈100% sensing range recovery) under subzero temperature. Sensitively and consistently monitoring of various physiological signals under harsh conditions, such as multiple structure damages, long period (≥5 d) and subzero temperatures (≤‐38.2°C) highlights its prominent durability, lifetime and environmental‐stability, demonstrating great promise toward practical application. A mechanically robust, self‐healable, moisturizing, cryotolerant and adhesive GelMA hydrogel is developed by incorporating polyvinyl alcohol, acrylic acid, sodium tetraborate, and glycerol into GelMA matrix via a photoinitiated polymerization process. The hydrogel‐derived omni‐healable ionotronic pressure sensor can sensitively and consistently monitor various physiological signals under harsh conditions, demonstrating great promise toward practical application.

Dual-layer conduit containing VEGF-A – Transfected Schwann cells promotes peripheral nerve regeneration via angiogenesis

Yuye Huang,Kai Ye,Andong He,Shaobo Wan,Miaoben Wu,Donghao Hu,Kailei Xu,Peng Wei,Jun Yin

Peripheral nerve injuries (PNIs) can cause neuropathies and significantly affect the patient's quality of life. Autograft transplantation is the gold standard for conventional treatment; however, its application is limited by nerve unavailability, size mismatch, and local tissue adhesion. Tissue engineering, such as nerve guidance conduits, is an alternative and promising strategy to guide nerve regeneration for peripheral nerve repair; however, only a few conduits could reach the high repair efficiency of autografts. The healing process of PNI is frequently accompanied by not only axonal and myelination regeneration but also angiogenesis, which initializes nerve regeneration through vascular endothelial growth factor A (VEGF-A). In this study, a composite nerve conduit with a poly (lactic-co-glycolic acid) (PLGA) hollow tube as the outer layer and gelatin methacryloyl (GelMA) encapsulated with VEGF-A transfected Schwann cells (SCs) as the inner layer was established to evaluate its promising ability for peripheral nerve repair. A rat model of peripheral nerve defect was used to examine the efficiency of PLGA/GelMA-SC (VA) conduits, whereas autograft, PLGA, PLGA/GelMA, and PLGA/GelMA-SC (NC) were used as controls. VEGF-A-transfected SCs can provide a stable source for VEGF-A secretion. Furthermore, encapsulation in GelMA cannot only promote proliferation and tube formation of human umbilical vein endothelial cells but also enhance dorsal root ganglia and neuronal cell extension. Previous animal studies have demonstrated that the regenerative effects of PLGA/GelMA-SC (VA) nerve conduit were similar to those of autografts and much better than those of other conduits. These findings indicate that combination of VEGF-A-overexpressing SCs and PLGA/GelMA conduit-guided peripheral nerve repair provides a promising method that enhances angiogenesis and regeneration during nerve repair. Statement of significance Nerve guidance conduits shows promise for peripheral nerve repair, while achieving the repair efficiency of autografts remains a challenge. In this study, a composite nerve conduit with a PLGA hollow tube as the outer layer and gelatin methacryloyl (GelMA) encapsulated with vascular endothelial growth factor A (VEGF-A)-transfected Schwann cells (SCs) as the inner layer was established to evaluate its potential ability for peripheral nerve repair. This approach preserves growth factor bioactivity and enhances material properties. GelMA insertion promotes Schwann cell proliferation and morphology extension. Moreover, transfected SCs serve as a stable VEGF-A source and fostering angiogenesis. This study offers a method preserving growth factor efficacy and safeguarding SCs, providing a comprehensive solution for enhanced angiogenesis and nerve regeneration.

Citric Acid Loaded Hydrogel-Coated Stent for Dissolving Pancreatic Duct Calculi

Jing Li,Yanwei Lv,Zheng Chen,Jiulong Zhao,Shige Wang

In recent years, the incidence of chronic pancreatitis has increased significantly. Pancreatic calculi obstruct the pancreatic duct and induce abdominal pain in the patients. Pancreatic duct stenting is the major treatment option for chronic pancreatitis with calculi. In this study, a new kind of drug-eluting stent, a pancreatic stent coated by methacrylated gelatin (GelMA) hydrogel loaded with citric acid (CA), was designed for the interventional treatment of pancreatic duct calculi. The CA loading capacity reached up to 0.7 g CA/g hydrogel-coated stent. The GelMA hydrogel coating has higher mechanical strength and lower swelling performance after loading with CA. The in vitro experiments of stents exhibited good performance in CA sustained release and the calculi can be dissolved in almost 3 days. The stents also showed good blood compatibility and cell compatibility. This research has important clinical value in the treatment of chronic pancreatitis with pancreatic calculi.

Balancing Hydration and Mechanical Stability in One-Step Photocured GelMA/Poly(acrylic acid- co -hydroxyethyl acrylate) Composite Hydrogels

Jiru Jia,Fangqing Ge,Xiaotian Chen,Zhihe Zhang,Lvyi Yang,Jing Qu,Jun Peng,Qi Xiao,Ya Yang,Wei Ma,Jie Chen

Abstract To address the challenge of balancing high-water content with structural stability in hydrogels, gelatin methacryloyl (GelMA) was introduced into a poly(acrylic acid-co-hydroxyethyl acrylate) system. GelMA/PAH composite hydrogels (Gel-PAH) were then prepared through one-step ultraviolet (UV) photocuring, and their network formation and overall properties were tuned by varying the glycerol dimethacrylate (GDMA) content. The Gel-PAH hydrogels exhibited higher hydration capacity and greater surface wettability than the corresponding PAH hydrogels. As the GDMA content increased, the gel fraction and tensile strength of both the PAH and Gel-PAH series generally increased. The tensile strength of the Gel-PAH hydrogels ranged from approximately 1.6 to 2.5 MPa. Gel-PAH-0.15 exhibited an equilibrium water content of 82.55%, a water contact angle of approximately 39°, and a relative cellular metabolic activity of approximately 84% after 24 h. Overall, Gel-PAH-0.15 achieved a favorable balance among network stability, hydration capacity, mechanical performance, and preliminary direct contact cytocompatibility. These results show that a compositionally simple AA/HEA-GelMA system provides a straightforward route to tune hydration and mechanical performance through one-step UV photocuring and adjustment of GDMA content, without introducing additional reinforcing fillers or sequential cross-linking processes.

A fluorescent GelMA hydrogel with covalently anchored nitrogen-doped carbon dots for efficient adsorption and sensitive detection of Cu(II)

Guanji Chen,Dan Qiu,Li Xu,Guangjie Liang,Peng Xie,Yuewen Huang,Shudong Lin,Bin Wang

Efficient removal and rapid in situ monitoring of Cu(II) in wastewater are of great importance for aquatic environmental remediation, yet conventional adsorbents generally fail to simultaneously achieve high adsorption capacity and quantitative detection. In this study, a N-doped carbon dot composite hydrogel strategy integrating adsorption and fluorescence sensing was proposed. A three-dimensional hydrogel network rich in N/O-containing functional groups was constructed using gelatin methacryloyl (GelMA), acrylic acid (AAc), acrylamide (AAm), and nitrogen-doped carbon dots (NCDs) were further introduced to fabricate the bifunctional fluorescent composite hydrogel NCDs-GA 1 A 2 gel. This structure not only provided abundant coordination and electrostatic adsorption sites for Cu(II), but also endowed the material with optical responsiveness for fluorescence detection. Batch adsorption experiments showed that the Cu(II) adsorption process followed the pseudo-second-order kinetic model and the Langmuir isotherm model, indicating monolayer chemisorption, with a maximum theoretical adsorption capacity of 203.1 mg·g −1 . After ten adsorption-desorption cycles, the hydrogel retained 86.7% of its initial adsorption capacity, demonstrating favorable regeneration stability. Meanwhile, NCDs-GA 1 A 2 gel could serve as a fluorescent probe for Cu(II), showing a good linear fluorescence response to Cu(II) concentration and a low detection limit of 0.65 μM. Coexisting ion interference experiments further confirmed its excellent selectivity and anti-interference capability. This work provides a facile strategy for developing multifunctional biomass-derived fluorescent hydrogel adsorbents and demonstrates promising application potential for Cu(II) removal and in situ detection in wastewater

Bioactive wound dressing based on decellularized tendon and GelMA with incorporation of PDA-loaded asiaticoside nanoparticles for scarless wound healing

Shuang Liu,Yingsong Zhao,Ming Li,Lei Nie,Qianqian Wei,Oseweuba Valentine Okoro,Hafez Jafari,Siyuan Wang,Jun Deng,Jianghai Chen,Amin Shavandi,Lihong Fan

In this study, bioactive composite hydrogels were created using the decellularized extracellular matrix (ECM), GelMA, and Polydopamine-loaded Asiaticoside (AC@PDA) nanoparticles for use as wound dressings that could promote healing. A decellularization method was used to obtain ECM from porcine Achilles tendon tissue. AC@PDA nanoparticles were then synthesized and found to have a uniform spherical structure with good cytocompatibility, particularly when compared to PDA nanoparticles alone. The mechanical properties of the bioactive composite hydrogels showed good elasticity and shape recovery after compression, with a slight decrease in compressive strength due to the addition of nanoparticles. The formation of interpenetrating networks through the use of EDC/NHS was also found to improve the mechanical properties and moisture retention of the hydrogels. The PDA/ECM-G and AC@PDA/ECM-G hydrogels showed higher water absorption capacity and similar moist retention capacity to the ECM-G hydrogel. The microstructure of the hydrogels was observed through SEM, with the ECM-G hydrogel showing a dense and compact structure, while the PDA/ECM-G and AC@PDA/ECM-G hydrogels displayed a more porous and interconnected structure due to the presence of nanoparticles. In vitro cytotoxicity tests on human skin fibroblasts showed good biocompatibility for all hydrogels. The in vivo wound healing performance of the hydrogels was also tested on a full-thickness excisional wound model in mice, with the AC@PDA/ECM-G hydrogel showing the fastest wound closure without scarring and the highest-formed hair follicles. The AC@PDA/ECM-G hydrogel had the best performance in promoting wound healing. These results suggest that the bioactive hydrogel has the potential for use as a wound dressing.

Meiqi Jin,Ningning Sun,Wenxian Weng,Zhentao Sang,Taotao Liu,Wei Xia,Shuze Wang,Xiaoting Sun,Tianlin Wang,Heran Li,Huazhe Yang

Bone tissue is a natural composite, exhibiting complicated structures and unique mechanical/biological properties. With an attempt of mimicking the bone tissue, a novel inorganic-organic composite scaffolds (ZrO 2 -GM/SA) was designed and prepared via the vacuum infiltration method and the single/double cross-linking strategy by blending GelMA/alginate (GelMA/SA) interpenetrating polymeric network (IPN) into the porous zirconia (ZrO 2 ) scaffold. The structure, morphology, compressive strength, surface/interface properties, and biocompatibility of the ZrO 2 -GM/SA composite scaffolds were characterized to evaluate the performance of the composite scaffolds. Results showed that compared to ZrO 2 bare scaffolds with well-defined open pores, the composite scaffolds prepared by double cross-linking of GelMA hydrogel and sodium alginate (SA) presented a continuous, tunable and honeycomb-like microstructure. Meanwhile, GelMA/SA showed favorable and controllable water-uptake capacity, swelling property and degradability. After the introduction of IPN components, the mechanical strength of composite scaffolds was further improved. The compressive modulus of composite scaffolds was significantly higher than the bare ZrO 2 scaffolds. In addition, ZrO 2 -GM/SA composite scaffolds had highly biocompatibility and displayed a potent proliferation and osteogenesis of MC3T3-E1 pre-osteoblasts compared to bare ZrO 2 scaffolds and ZrO 2 -GelMA composite scaffolds. At the same time, ZrO 2 -10GM/1SA composite scaffold regenerated significantly greater bone than other groups in vivo . This study demonstrated that the proposed ZrO 2 -GM/SA composite scaffolds had great research and application potential in bone tissue engineering.

Production and Characterization of an Integrated Multi-Layer 3D Printed PLGA/GelMA Scaffold Aimed for Bile Duct Restoration and Detection

Xiang Yang,Wang Weijia,Gao Yuanhui,Zhang Jianquan,Zhang Jing,Bai Zhiming,Zhang Shufang,Yang Yijun

We successfully fabricated artificial bile duct via 3D printing technique which was composed of poly (lactic-co-glycolic acid) (PLGA) and gelatin methacrylate (GelMA). The PLGA-inner layer provided sufficient strength to support the bile duct contraction, the GelMA-outer layer possessed good biocompatibility to provide a good living environment for the cells. Moreover, IKVAV laminin peptide (Ile-Lys-Val-Ala-Val) and ultrasmall superparamagnetic iron oxide (USPIO) were used to regulate scaffold cell adhesion and magnetic resonance imaging (MRI) detection, respectively. After BMSCs co-culture with IKVAV at a certain concentration, the survival rate and adhesion of BMSCs was increased obviously. Meanwhile, the fabricated scaffold exhibited the tensile modulus in the range of 17.19 – 29.05 MPa and the compressive modulus in the range of 0.042 – 0.066 MPa, which could meet the needs of human implantation. In an animal experiment in vivo pig bile duct regeneration, PLGA/GelMA/IKVAV/USPIO duct conduits could promote bile duct regeneration and enhance cytokeratin 19 (CK19) expression. In summary, the composite bile duct scaffold with excellent MRI imaging function and biocompatibility could be used to develop bioactive artificial bile ducts.

3D bioprinting of high-performance hydrogel with in-situ birth of stem cell spheroids

Shunyao Zhu,Xueyuan Liao,Yue Xu,Nazi Zhou,Yingzi Pan,Jinlin Song,Taijing Zheng,Lin Zhang,Liyun Bai,Yu Wang,Xia Zhou,Maling Gou,Jie Tao,Rui Liu

Digital light processing (DLP)-based bioprinting technology holds immense promise for the advancement of hydrogel constructs in biomedical applications. However, creating high-performance hydrogel constructs with this method is still a challenge, as it requires balancing the physicochemical properties of the matrix while also retaining the cellular activity of the encapsulated cells. Herein, we propose a facile and practical strategy for the 3D bioprinting of high-performance hydrogel constructs through the in-situ birth of stem cell spheroids. The strategy is achieved by loading the cell/dextran microdroplets within gelatin methacryloyl (GelMA) emulsion, where dextran functions as a decoy to capture and aggregate the cells for bioprinting while GelMA enables the mechanical support without losing the structural complexity and fidelity. Post-bioprinting, the leaching of dextran results in a smooth curved surface that promotes in-situ birth of spheroids within hydrogel constructs. This process significant enhances differentiation potential of encapsulated stem cells. As a proof-of-concept, we encapsulate dental pulp stem cells (DPSCs) within hydrogel constructs, showcasing their regenerative capabilities in dentin and neovascular-like structures in vivo . The strategy in our study enables high-performance hydrogel tissue construct fabrication with DLP-based bioprinting, which is anticipated to pave a promising way for diverse biomedical applications.

pH-responsive resveratrol delivery using chitosan-based nanoparticles embedded in GelMA: in vitro evaluation of acute brain injury-relevant cellular responses

Chen Zhongxin,Cao Wei,Zhao Minghua,Wang Qing,Li Yujiang,Fu Rong,Gao Xinyu,Ke Zunqian,Xu Xinqing,Li Shengcun

Background Secondary injury after acute brain injury (ABI) involves interconnected oxidative, inflammatory, and apoptosis-associated cellular responses. Resveratrol (RSV) has antioxidant and anti-inflammatory activities, but its limited aqueous stability and rapid clearance restrict sustained biological activity. This study developed a pH-responsive RSV delivery platform consisting of chitosan-based nanoparticles embedded in gelatin methacryloyl (GelMA) and evaluated its physicochemical properties, release behavior, and effects on ABI-relevant cellular responses in vitro. Results RSV-loaded chitosan-based nanoparticles (RSV@PCCC) were prepared through electrostatic self-assembly and subsequently incorporated into GelMA hydrogel. The optimized nanoparticles maintained relatively stable hydrodynamic characteristics at pH 7.2 and exhibited increased RSV release under acidic conditions. After 72 h, cumulative RSV release from RSV@PCCC reached ~ 74% at pH 6.4 and ~ 50% at pH 7.2. Incorporation into GelMA (RSV@PCCC-Hydrogel) prolonged RSV release, resulting in cumulative release of ~ 58% and ~ 36% at pH 6.4 and pH 7.2, respectively. In LPS-stimulated neural and microglial cells, the RSV-containing formulations reduced intracellular reactive oxygen species. RSV@PCCC reduced IL-1β and TNF-α protein expression by ~ 60% and ~ 50% respectively. RSV@PCCC and RSV@PCCC-Hydrogel reduced the Bax to Bcl-2 ratio by ~ 50% and ~ 40% respectively. These results indicate attenuation of oxidative stress, inflammatory activation, and apoptosis-associated changes under the tested in vitro conditions. Conclusions The chitosan-based nanoparticle/GelMA platform enabled pH-responsive and sustained RSV release and attenuated LPS-induced oxidative, inflammatory, and apoptosis-associated changes in neural and microglial cells. These findings provide an in vitro proof-of-concept for modulating cellular processes associated with secondary injury after ABI and support further evaluation of this platform in physiologically relevant ABI models.

The use of machine learning to predict the effects of cryoprotective agents on the GelMA-based bioinks used in extrusion cryobioprinting

Qiao Qian,Zhang Xiang,Yan Zhenhao,Hou Chuanyu,Zhang Juanli,He Yong,Zhao Na,Yan Shujie,Gong Youping,Li Qian

Cryobioprinting has tremendous potential to solve problems to do with lack of shelf availability in traditional bioprinting by combining extrusion bioprinting and cryopreservation. In order to ensure the viability of cells in the frozen state and avoid the possible toxicity of dimethyl sulfoxide (DMSO), DMSO-free bioink design is critical for achieving successful cryobioprinting. A nontoxic gelatin methacryloyl-based bioink used in cryobioprinting is composed of cryoprotective agents (CPAs) and a buffer solution. The selection and ratio of CPAs in the bioink directly affect the survival of cells in the frozen state. However, the development of universal and efficient cryoprotective bioinks requires extensive experimentation. We first compared two commonly used CPA formulations via experiments in this study. Results show that the effect of using ethylene glycol as the permeable CPA was 6.07% better than that of glycerol. Two datasets were obtained and four machine-learning models were established to predict experimental outcomes. The predictive powers of multiple linear regression (MLR), decision tree (DT), random forest (RF), and artificial neural network (ANN) approaches were compared, suggesting an order of ANN>RF>DT>MLR. The final selected ANN model was then applied to another dataset. Results reveal that this machine-learning method can accurately predict the effects of cryoprotective bioinks composed of different CPAs. Outcomes also suggest that the formulations presented here have universality. Our findings are likely to greatly accelerate research and development on the use of bioinks for cryobioprinting. A GelMA hydrogel co-loaded with ursolic acid, ferulic acid and naringin facilitates dentin regeneration by providing a multi-component odontogenic microenvironment

Jun Che,Ai Yang,Feng Mo,Yaqin Gao,Yunshu Li,Yuting Cui,Feihong Zhuo,Yun Hong,Yiheng Zhang,Tianyi Chen,Fei Qin,Gaohong LYU,Zhipeng Chen,Liu Xu

GelMA hydrogelco-loaded anti-caries/odontogenic components (Gel/UFN) was developed. • Gel/UFN was designed to achieve prolonged release over 55 days. • Gel/UFN upregulates odontogenesis-related genes and regenerative mineralization. • Gel/UFN promotes angiogenesis and enables the regeneration of dentin in vivo . Dental caries is a prevalent bacterial infectious disease, yet current clinical treatments remain limited because conventional restorative materials provide only mechanical filling and fail to induce dental tissue regeneration. Herein, we developed a composite gelatin methacryloyl (GelMA) hydrogel system (Gel/UFN) co-loaded with three active components from traditional Chinese medicine, including ursolic acid (UA), ferulic acid (FA) and naringin (Nar), to remodel the carious microenvironment and promote dentin remineralization and vascularized pulp–dentin-like tissue formation. Gel/UFN showed excellent biocompatibility and favorable physicochemical and mechanical properties for dental applications, supporting adhesion and proliferation of human dental pulp stem cells (hDPSCs). Functionally, Gel/UFN significantly inhibited Streptococcus mutans ( S. mutans ) growth and biofilm formation, downregulated inflammatory cytokine expression and promoted the migration and angiogenesis of human umbilical vein endothelial cells (HUVECs). More importantly, Gel/UFN upregulated odontogenesis-related genes in hDPSCs, thereby enhancing odontogenic differentiation, alkaline phosphatase activity, collagen maturation and regenerative mineralization. In vivo , Gel/UFN exhibited slow degradation and sustained drug release for over 55 days. After implantation of Gel/UFN-filled dentin discs into nude mice, H&E staining demonstrated the formation of vascular structures, dentin-like mineralized tissue and cell-rich pulp-like tissue. Western blot analysis further revealed increased expression of the odontogenesis-related proteins RUNX2, DSPP, DMP1, and COL1A1 in the Gel/UFN group, providing additional protein-level evidence of its pro-odontogenic effects in vivo . These newly formed tissues were observed to cover exposed dentinal tubules on the surface of dentin disc using scanning electron microscopy (SEM). Micro-CT further confirmed that dental tissue induced by Gel/UFN exhibited a higher remineralization density at the defect site, supporting enhanced dentin remineralization. Overall, Gel/UFN may serve as a promising bioactive hydrogel platform for dentin repair and vascularized pulp–dentin-like tissue formation through antibacterial, anti-inflammatory, pro-angiogenic and pro-differentiation effects.
