The objective of this study was to compare the pharmacokinetics of vancomycin hydrochloride administered into rabbits through different routes and explore the feasibility of peptide drugs entering the systemic circulation through ocular administration.A convenient, accurate, and rapid liquid chromatography-trandem mass spectrometric (LC-MS/MS) method was established and used for the determination of vancomycin hydrochloride in rabbit plasma after intravenous administration (1.5 mg/kg), intragastric, and ocular administration (15 mg/kg). The pharmacokinetic parameters were analyzed using the DAS 2.0 software. We obtained a linear calibration curves vancomycin hydrochloride in plasma of rabbits over a concentration range of 0.05–10.0 μg/mL (R2 > 0.9995), the interassay accuracy was within 5%, precision of 1.66–3.38%, and recovery of >85%. No matrix effects were observed. The absolute bioavailability of vancomycin hydrochloride after intragastric and ocular administration was 1.0 and 7.3%, with the half-life values of 63.1 and 138.5 min, respectively. Therefore, the LC-MS/MS method established in this experiment was suitable for the determination of vancomycin hydrochloride. Vancomycin hydrochloride was rapidly absorbed into the blood circulation after ocular administration. Ocular administration was linked to higher bioavailability compared with intragastric administration, suggesting that the former will become a route for the delivery of peptide drugs.

A simple and sensitive method for the simultaneous determination of chito-oligosaccharides (COSs) with degrees of polymerization (DPs) from 2 to 7 was developed and used for COS quantification in rat plasma. Samples were separated on a Waters XBridge Amide column (3.5 μm, 2.1 × 150 mm) by isometric elution with 10 mM aqueous ammonium acetate (pH = 9) in acetonitrile and 10 mM aqueous ammonium acetate (pH = 9) (v/v, 50 : 50) employing multiple reaction monitoring (MRM) detection. Analytes and internal standards (IS) were extracted from rat plasma by protein precipitation with acetonitrile. The assay was linear over a concentration range of 20–10 000 ng mL−1 for COS2–7. The intra-day and inter-day precision of the investigated components exhibited an RSD within 15%, and the accuracy (RE%) ranged from −7.3% to 7.6%. The extraction recoveries of the six constituents were determined to be between 82.5% and 94.3%. No significant matrix effects for COS2–7 were observed in rat plasma. COS in plasma remained stable for 24 h at room temperature (short-term), after freeze–thaw cycles, and 30 days in a −40 °C freezer. In comparison to reported COS quantitation methods, this method is simple, sensitive and cost-effective and could be used for the simultaneous quantitation of COS2–7. This method meets the Food and Drug Administration guidelines and had been successfully applied to the analysis of pharmacokinetic samples collected from rats.

The spoiled dry-cured ham commonly shows unpleasant taste and odour. To deepen the understanding in the formation mechanism of unpleasant taste in spoiled ham, sensory attributes, texture parameters, protein degradation, metabolites were investigated between normal and spoiled hams; the relationship between the sensory quality and metabolites of dry-cured ham was further evaluated by partial least square discriminant analysis (PLS-DA). The scores of richness and overall acceptance were significantly lower in spoiled ham, and more than 12.5-fold values in adhesiveness were found in spoiled ham than normal ham. Myofibrillar proteins including actin, troponin-T and myosin light chain showed excessive degradation in spoiled ham. Forty-two kinds of metabolites mainly derived from protein degradation were identified by LC-MS/MS, and amino acid derivatives and oligopeptides were the key components to distinguish spoiled and normal hams demonstrated by PLS-DA. Purine metabolism, pyrimidine metabolism and protein degradation were the main metabolism pathways in spoiled ham.

Purpose It is crucial for forensic analysis to differentiate clinical use from illegal abuse. Selegiline (SG) is mainly metabolized to desmethylselegiline (DM-SG), R -(−)-methamphetamine (R-MA) and R-(−)amphetamine (R-AM); while abused methamphetamine and amphetamine mainly contain the S -(+)-form. The aim of this study was to simultaneously determine SG, DM-SG, R / S -MA, and R / S -AM in human oral fluid (OF) and to differentiate clinical use from illegal use. We also aim to apply the present method to the OF samples from authentic cases in forensic toxicology. Methods Liquid–liquid extraction and liquid chromatography-tandem mass spectrometry operating in positive ion multiple-reaction monitoring mode were utilized. The chromatographic system consisted of a Chirobiotic TM V2 column (2.1 mm × 250 mm, 5 μm) and a mobile phase of methanol containing 0.1% (v/v) glacial acetic acid and 0.02% (v/v) ammonium hydroxide. Results and conclusions The calibration curves were linear from 1 to 100 ng/mL, and r > 0.995 for all analytes with imprecisions ≤ 15% and accuracy between 85 and 115%. Extraction recoveries ranged from 46.3 to 104.7% with coefficient of variation (CV) ≤ 10.3% and matrix effects (MEs) ranged from 47.4 to 114.8% with CV ≤ 12.9%. The lower limit of quantification was 0.2 ng/mL for SG and DM-SG and 1.0 ng/mL for S-MA, R-MA, S-AM and R-AM. The present method is simple, rapid (accomplished in 12 min), sensitive, and validated by authentic samples.

The mevalonate (MVA) pathway plays a crucial role in the occurrence and progression of various diseases, such as osteoporosis, breast cancer, and lung cancer, etc. However, determining all the MVA pathway intermediates is still challenging due to their high polarity, low concentration, chelation effect with metal compartments, and poor mass spectrometric response. In this study, we established a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method coupled with N 2 , N 2 , N 4 , N 4 -tetramethyl-6-(4-(piperazin-1-ylsulfonyl) phenyl)-1,3,5-triazine-2,4-diamine (Tmt-PP) labeling for the simultaneous analysis of all MVA intermediates in biospecimens. Chemical derivatization significantly improved the chromatographic retention, peak shape, and detection sensitivity of the analytes. Moreover, we employed a method named mass spectrum calculation to achieve the absolute quantification of the isomers, i.e., isopentenyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP). The established method was fully qualified and applied to explore the difference of these metabolites in cisplatin-resistant non-small cell lung cancer (NSCLC) cells. Additionally, several MVA intermediate analogs, including isopentenyl monophosphate or dimethylallyl monophosphate (IMP/DMAMP), geranyl monophosphate (GMP), 5-triphosphomevalonate (MTP), and isopentenyl triphosphate or dimethylallyl triphosphate (ITP/DMATP), were identified for the first time using a knowledge-driven prediction strategy. We further explored the tissue distribution of these novel metabolites. Overall, this work developed a sensitive quantification method for all MVA intermediates, which will enhance our understanding of the role of this pathway in various health and disease conditions. The novel metabolites we discovered warrant further investigations into their biosynthesis and biological functions.

Herba Lysimachiae inhibits synovial damage in osteoarthritis via regulating two bio labels (integrin alpha 2b/beta 3). However, the relevant active ingredients are still unknown. Here, the active ingredients of herbal medicines were analyzed based on the liquid chromatography–tandem mass spectrometry technology and public bioinformatics platforms. The liquid chromatography–tandem mass spectrometry technology was used for compound analysis, and public databases (PubChem BioAssay and STRING) were applied to establish the links between herbal compounds and both bio labels, and identify which herbal compounds may regulate these bio labels. Subsequently, the osteoarthritis model was used to confirm the results. Totally, ninety compounds in Herba Lysimachiae were identified based on the liquid chromatography–tandem mass spectrometry technology. Bioinformatics analysis showed that five compounds (myricetin, fisetin, esculetin, 7-hydroxycoumarin-4-acetic acid, and caffeic acid) may synergistically regulate bio labels through 11 targets, which may be the active ingredients of Herba Lysimachiae for osteoarthritis treatment. In the verification experiments, five compounds markedly suppressed the overexpression of bio labels in the synovium of the osteoarthritis model. In conclusion, the present study effectively and rapidly analyzed the active ingredients of Herba Lysimachiae for osteoarthritis treatment.

NH₂-MCOF enables ultra-sensitive detection of 17 PFASs down to 0.0346 ng/L. • Fast MSPE within 40 min and reusable for ≥6 cycles with stable performance. • Successfully detected PFASs in real urban waters, including 3.346 μg/L PFDS. Per - and polyfluoroalkyl substances (PFAS), as a class of persistent organic pollutants, are characterized by strong environmental persistence and high bioaccumulation potential, making them a major concern in global environmental and health fields. Currently, the trace detection of PFASs in urban environmental water samples, including surface water, source water, and domestic wastewater faces challenges such as insufficient adsorption selectivity of traditional materials and cumbersome pretreatment steps, highlighting the urgent need to develop efficient enrichment and highly sensitive analytical methods. To improve the enrichment efficiency of PFAS, this study designed and synthesized an amino-functionalized magnetic covalent organic framework material (NH₂-MCOF) as an adsorbent. Combined with magnetic solid-phase extraction (MSPE) and liquid chromatography-tandem mass spectrometry, a synchronous analytical method was established for the determination of 17 PFASs in real urban environmental water matrices. Through systematic optimization, the optimal pretreatment conditions were determined as follows: adsorbent dosage of 20 mg, solution pH of 3.0, ultrasonic-assisted adsorption time of 40 min, and a stepwise elution strategy using methanol, ammoniated methanol, and formic acid-modified methanol. Under the optimized conditions, the method demonstrated good linearity within the range of 0.5–200 μg/L, with detection limits of 0.0346–1.30 ng/L. The spiked recoveries ranged from 50% to 138%, and the intra -day and inter -day precision, expressed as relative standard deviations (RSD), were below 13.5% and 14.9%, respectively. Application to real water samples confirmed the method's effectiveness in detecting multiple PFASs, demonstrating its practicality and reliability. The analytical method established in this study integrates functionalized material design, efficient pretreatment, and sensitive detection, providing a reliable approach for the synchronous monitoring of multiple trace PFAS in real urban environmental water matrices and highlighting the potential application of functionalized magnetic covalent organic framework materials in environmental analysis. Mycobacterium tuberculosis culture filtrate proteins (MTB-CFPs) are critical for tuberculosis diagnosis and vaccine development. To explore their diagnostic potential, MTB was statically cultured in Sauton's medium for 15 days; MTB-CFPs were collected, fractionated using six biomimetic affinity chromatography (BiAC) resins, and analyzed by Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS). Four MTB-specific regions of difference (RDs) and the PstS1 protein were recombinantly expressed and purified, with their serological efficacy assessed by ELISA using sera from 157 pulmonary tuberculosis (pTB) patients and 71 healthy controls (HCs). LC-MS/MS identified 622 MTB proteins, 307 of which were validated with ≥2 peptides and P < 0.05. BiAC fractionation significantly increased the average peptide number (2.09-fold), peptide coverage (1.43-fold), and the exponentially modified Protein Abundance Index (emPAI) (1.70-fold) in 281 common identifications. Receiver Operating Characteristic (ROC) analysis revealed rEC (a fusion expression product of the Rv3875 and Rv3874 genes in the RD1 region, linked by a 15-nucleotide connector) as the optimal diagnostic marker Area Under the Curve (AUC = 0.8030), with sharply distinct IgG levels between pTB patients and HCs ( P < 0.0001). Notably, rEC combined with GmdA (encoded by the Rv1511 gene located in RD11) enhanced sensitivity while preserving high specificity, especially for Acid-Fast Bacilli (AFB) negative pTB cases demonstrating substantial clinical translation potential. Additionally, extended culture promoted MTB autolysis and cytoplasmic protein release, providing valuable insights into TB pathogenesis and biomarker discovery. This integrated approach of BiAC fractionation, proteomics, and serological validation advances TB precision diagnostics.

Steroid hormones play crucial regulatory roles in human growth, development, reproduction, and the maintenance of internal environmental homeostasis. Because the concentrations of most hormones in the body are extremely low and their chemical properties vary widely, developing an accurate and sensitive quantitative method has become a primary objective in clinical diagnostics. In this study, an isotope dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) method was developed and validated for the simultaneous quantification of 22 steroid hormones in human serum. To enhance extraction and purification efficiency, a protein precipitation–solid-phase extraction (PP-SPE) approach was employed, forming a “dual purification” strategy. The limits of quantification for progesterone, cortisol, dihydrotestosterone, and dehydroepiandrosterone sulfate were 0.05, 0.50, 0.05, and 1.00 ng/mL, respectively, representing an improvement over previously reported values obtained using similar sample pretreatment methods. Method validation results demonstrated strong linearity for all 22 steroid hormones ( R 2 > 0.98). Accuracy ranged from 86.9% to 119.5%, intra-day precision was below 9.7%, inter-day precision was below 12.0%, and the matrix effect was controlled within 80.3%–118.1%. These findings indicate that the method is accurate, reliable, and suitable for analyzing multiple components in complex serum matrices. Twenty real serum samples were analyzed to evaluate the clinical applicability of the method. The results demonstrated that this method can be used clinically to monitor steroid hormone levels, assist in diagnosing and treating endocrine disorders, and provide a diagnostic basis for adrenal diseases. It holds significant reference value for clinical laboratory testing. Graphical

The quantitative analysis of 10 neurotransmitters, including Gamma-aminobutyric acid (γ-GABA), L-Glutamate (Glu), L-Glutamine (Gln), L-Tryptophan (Trp), Serotonin (5-HT), 5-Hydroxyindoleacetic acid (5-HIAA), L-Phenylalanine (Phe), L-Tyrosine (L-Tyr), Dopamine (DA), Levodopa (L-DOPA) was achieved within a chromatographic separation time of 4.5 min by the application of ThermoFisher Scientific™ Vanquish™ HPLC coupled to ThermoFisher ScientificTM TSQ QuantisTM mass spectrometer. Optimized chromatographic separation as well as high sensitivity allow the simultaneous analysis and precise quantification of ten neurotransmitters in mouse hippocampus. The limits of detection in sample ranged from 0.3pg (Phe) to 0.1 μg (γ-GABA). Recoveries were between 88.0 to 113.8 %, the mean intra - day and inter - day coefficient of variation of 5.56% and 4.72%, respectively. The matrix effect was from 85.1 to 114.9%. A series of significant changes in five neuroinflammatory mouse models can be detected. γ-GABA, Glu, Gln, DA, Phe, L-Tyr, 5-HIAA were decreased in depression model. Gln and Glu were decreased in PTSD model. 5-HIAA was elevated in anxiety. 5-HIAA and L-Trp were decreased in CFA and SNI model, respectively. In conclusion, a LC-MS/MS method for simultaneous measurement of 10 neurotransmitters in mouse hippocampus was developed and successfully applied to investigation of the changes in five neuroinflammatory mouse models. The method would be expected to provide applicability to the study of the mechanisms of neurological diseases associated with these neurotransmitters.

MicroRNAs (miRNAs) play a vital role in regulating gene expression and are associated with a variety of cancers, including breast cancer. Their distorted and unique expression is a potential marker in clinical diagnoses and prognoses. Thus, accurate determination of miRNA expression levels is a prerequisite for their applications. However, the assays currently available for miRNA detection typically require pre-enrichment, amplification and labeling steps, and most of the assays are only semi-quantitative. Therefore, we developed a quasi-direct liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based targeted proteomics approach to quantify target miRNA by innovatively converting the miRNA signal into the mass response of a reporter peptide via a covalently immobilized DNA-peptide probe. Specifically, the probe containing the targeted proteomics-selected substrate/reporter peptide, GDR AVQLGVDPFR/AVQLGVDPFR, and the DNA sequence complementary to the target miRNA (i.e., miR-21) was first immobilized on APMTS modified silica nanoparticles using PDITC. After the immobilized probe was recognized and hybridized with the target miRNA, the excess probe was degraded using MBN and followed by a trypsin digestion of the hybrids. The reporter peptide was released and quantified using LC-MS/MS. The obtained LOQ was 5 pM. Finally, the developed assay was used for the quantitative analysis of miR-21 in breast cells and tissue samples.
