Polarization-passivation synergistic antisolvent strategy for high-brightness CsPbBr3 QLEDs exceeding 100,000 cd/m2

Haoqi Li,Wenjun Yuan,Kun Zhang,Yifei Wang,Xinrui Chen,Yuqin Su,Xingle Shang,Hengyang Xiang,Haibo Zeng

The carrier injection limitation caused by the surface ligands of quantum dots (QDs) is a major challenge for achieving high-brightness emission in quantum dot light-emitting diodes (QLEDs). In this work, we enhanced the polarity of the antisolvent by incorporating inorganic bromides (ZnBr2) to improve ligand stripping capability, thereby achieving fewer insulating ligands to meet the demand for efficient carrier injection. Simultaneously, the inorganic bromide provided a Br-rich environment, suppressing the generation of Br vacancies under polar conditions, which frequently happened in traditional antisolvent strategies. Benefiting from this polarization-passivation synergistic strategy, ZnBr2-treated QDs exhibited a 17% reduction in ligand quantity and a 40% reduction in defect density, thus the photoluminescence quantum yield (PLQY) of ZnBr2-treated QDs increased from 47% to 86%. The fabricated QLEDs exhibited a 143% increase in current density and a maximum brightness reaching 104,126 cd/m2, which was 4.96 times that of samples treated with pure antisolvent. Correspondingly, the operational lifetime (T50@100 cd/m2) of the device extended from 20 h to 241 h. This simple inorganic bromide polarization-passivation synergistic strategy in QDs' purification engineering provides a new approach for realizing high-brightness QLEDs.

Scalable Preparation of Liquid Infused Coatings for Lubrication of 103 m2 Dry Ski Slopes

Yuehua Zhao,Bo Peng,Lijun Liu,Yanming Fu,Tianyu Zhao,Weichao Chi,Dong Li,Dong Ji,Xin Wang,Dapeng Wang

To facilitate effective training for freestyle skiers on artificial dry ski slopes, it is crucial to reduce the friction coefficient of the slopes and closely match it with that of snow. Traditional lubrication methods, such as water or soapy water, come with multiple disadvantages, including water waste, which leads to environmental pollution, short-lived effectiveness, and high costs. In this study, we have successfully developed a method for the scalable preparation of a liquid-infused coating (LIC) by tandem spraying inexpensive and environmentally friendly SiO2 particles and silicone oil lubricants. Experimental results showed that the resulting LIC is capable of imparting slippery properties to various surfaces, regardless of the surface chemistry. Moreover, the presence of LIC could reduce the friction coefficient significantly. By carefully regulating the surface composition, we achieved a friction coefficient of 0.059 between a snowboard and the LIC-functionalized ski slope, closely matching that between the snowboard and snow in a typical skiing competition venue (∼0.06). We successfully applied LIC onto 103 m2 dry ski slopes, providing a training ground for professional freestyle skiers.

M2 macrophage-mediated tigecycline nanoparticles for combating CRKP pneumonia via antibacterial and immunomodulatory therapy

Weijing Cheng,Ling Wu,Yu Liao,Tingrui Zhang,Xiujuan Han,Zongguang Tai,Quangang Zhu,Zhongjian Chen,Jing Tian,Nan Ding,Zhuo Wang

Pneumonia caused by carbapenem-resistant Klebsiella pneumoniae (CRKP) has a high mortality rate and exhibits multiple drug resistance, making current antibacterial treatments of limited efficacy. To address the challenges of ineffective drug accumulation at the infection site and excessive inflammation leading to tissue damage, this study developed a cell-based nano-bionic drug delivery system (denoted as PT@M2, PLGA-TIG nanoparticles loaded into M2 macrophages). This system encapsulates poly(lactic- co -glycolic acid) (PLGA) loaded with tigecycline within M2-type macrophages, thereby achieving stable circulation, rapid release in acidic microenvironments, and targeted delivery to the lungs. In vitro experiments showed that PT@M2 significantly reduced the minimum inhibitory concentration of CRKP, disrupted bacterial membranes, and induced reactive oxygen species accumulation. In a mouse pneumonia model, PT@M2 effectively reduced bacterial colonies, alleviated inflammatory responses, and improved pulmonary histopathology. Wefurther confirmed that PT@M2 could inhibit calcium ion influx by downregulating CACNG6, thereby indirectly modulating the MAPK pathway. In summary, PT@M2 enhances antibacterial efficacy through a dual mechanism of “bactericidal action + immune microenvironment remodeling,” providing a novel therapeutic strategy for the treatment of multidrug-resistant bacterial infections.

C5aR1+ macrophages are involved in the progression of endometriosis

Chen Xi,Zuo Wei-Wen,Zhang Guo-Dong,Xu Cong-Jian

Objective: Immune dysfunction plays an important role in the pathogenesis of endometriosis. C5a, a by-product of complement activation, binds to its receptor (C5a receptor 1, C5aR1) in various cells, including macrophages. The combination of C5a and C5aR1 in macrophages induces M2 polarization. There is no consistent conclusion regarding macrophage polarization status and its participation in the pathogenesis of endometriosis. Methods: We analyzed the expression of C5aR1 and the immunophenotype of macrophages in samples from patients with endometriosis using immunohistochemistry and flow cytometry. We tested the equivalent item in a mouse model and the THP-1 cell line in vivo and in vitro . Single-cell RNA sequencing (scRNA-seq) from the Gene Expression Omnibus (GEO, GSE213216) was used to investigate the phenotype and function of C5aR1 + macrophages. Results: The infiltration of C5aR1 + macrophages was associated with the severity of endometriosis. C5aR1 + macrophages polarized more toward the M2 phenotype and transformed into an immunosuppressive state. Inhibiting C5aR1 using PMX-53 decreased the expression of C5aR1 + macrophages and reduced their polarization to the M2 phenotype, consequently shrinking lesion size in a mouse model. Conclusion: Our study emphasizes the importance of C5aR1 and the macrophage phenotype in the pathogenesis of endometriosis. We propose PMX-53 as a potential therapeutic agent for endometriosis.

C1QBP Drives M2 Macrophage Polarization Via TRAF2-CCL2 to Promote Oral Squamous Cell Carcinoma Progression

Xiyuan Qin,Kai Kang,Bowen Zhu,Yujie Shi,Shoushan Bu

Introduction and aims Oral squamous cell carcinoma (OSCC), frequently observed in the head and neck region, possesses mechanisms that have yet to be thoroughly clarified. Poor prognosis in OSCC is closely linked to the polarization of macrophages toward the M2 phenotype. Complement component 1q subcomponent-binding protein (C1QBP), a versatile protein commonly dysregulated in OSCC, has an unclear association with M2 macrophages. Methods Immunohistochemical (IHC) staining was conducted to determine the relationship between C1QBP expression and M2 macrophage infiltration. Flow cytometry (FC) and enzyme-linked immunosorbent assay (ELISA) were employed to explore the functional role of C1QBP in macrophages polarization. Furthermore, RNA sequencing combined with quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting (WB) were utilized to verify the increased expression of TNF receptor-associated factor 2 (TRAF2) and C-C motif chemokine 2 (CCL2). Additional validation of these findings was performed through in vivo experiments. Results A negative correlation was identified between C1QBP expression and CD206, a representative marker of M2 macrophages. Decreased levels of C1QBP in OSCC cells enhanced macrophage polarization toward the M2 phenotype and facilitated tumor progression by activating pivotal elements of the tumor necrosis factor (TNF) signaling pathway, notably TRAF2 and CCL2. Conclusion These findings suggest that C1QBP modulates the polarization of macrophages toward the M2 phenotype via the TRAF2-CCL2 signaling axis, thus contributing to OSCC malignancy progression. Consequently, targeting C1QBP to regulate M2 macrophage polarization may provide a promising therapeutic avenue for OSCC management.

SEMA7A orchestrates the colorectal cancer tumor microenvironment via PlexinC1: coordinating angiogenesis and macrophage M2 polarization

Ling-Li Zhang,Ke-Yu Chen,Zhang-Xia Bai,Yan-Qi Huang

Colorectal cancer (CRC) is a leading cause of cancer-related deaths worldwide, creating an urgent need to identify new therapeutic targets. Semaphorin 7A (SEMA7A), a glycosylphosphatidylinositol (GPI)-anchored protein, has been implicated in tumor progression, but its role in CRC, tumor microenvironment (TME), and inflammation-related CRC remains unreported. In this study, we first confirmed that SEMA7A is highly expressed in CRC tissues, linked to lymphatic metastasis and advanced tumor node metastases (TNM) stage by bioinformatics and clinical cohort analysis. Furthermore, by silencing SEMA7A in CRC cell lines, there was a significant inhibition of tumor growth, endothelial cell migration, and angiogenesis. In the TME, culture medium derived from SEMA7A-knockdown CRC cells inhibited the infiltration of tumor-associated macrophage (TAM) and their polarization toward a pro-tumor M2 phenotype. Moreover, our study revealed that in the colitis-associated CRC model induced by azoxymethane/dextran sodium sulfate, down-regulation of SEMA7A significantly reduced the tumor burden, with marked suppression of macrophage recruitment, expression of M2-like TAM markers, and angiogenesis. Subsequently, we focused on PlexinC1, the high-affinity receptor for SEMA7A, through transcriptomic analysis and demonstrated that SEMA7A activates the RAP1/AKT signaling pathway through PlexinC1 receptor in TAM. The rescue experiments using recombinant SEMA7A (rhSEMA7A) further indicated that blocking PlexinC1 suppressed the conversion of macrophages into M2-like TAM in the CRC microenvironment. Collectively, our findings demonstrated that SEMA7A can act as a key regulator of CRC progression, coordinating tumor-intrinsic and TME processes, and highlight its potential as a therapeutic target.

Treatment of atherosclerosis by reinfusion of functional monocytes reprogrammed with M2 macrophage-derived exosomal nanovehicles

Xiaoyu Liang,Jianghui Zhou,Kaijing Liu,Xue Fu,Huiyang Li,Xuanling Li,Jing Yang

Atherosclerosis (AS) lacks the therapy of targeting and inhibiting foam cells. In this study, a biomimetic delivery system was designed utilizing monocytes as vehicles to transport reactive oxygen species (ROS)-responsive nanovehicles containing M2 macrophage secretion and ATV, thus generating reinfusion of functional monocytes (MAMS) after remodeling to treat atherosclerosis. MAMS utilized active transportation to specifically target and regulate immune responses at the focus. The findings demonstrated that MAMS effectively induced the polarization of M2 macrophages and maintained their optimal activity. MAMS exhibited significant scavenging capacity against ROS, effectively inhibited foam cell formation and promoted the efflux of lipids in vitro. Furthermore, MAMS promoted the conversion of monocytes into M2 macrophages and their migration towards foam cells. In murine models of AS, MAMS displayed significant lipid-lowering efficacy, regulated relevant enzymes, reduced foam cells and fibers, induced M2 macrophages, and regulated the proliferation of smooth muscle. The integration of this ROS-responsive material and the reinfusion of functional monocytes has resulted in a novel cell shipping administration method that exhibits a multi-faceted effect on AS treatment, including lipid reduction, inflammation resistance, antioxidant activity, targeted delivery, microenvironment modification, and immune repair. This innovative approach is poised to revolutionize the management of AS.

CAR-M2 immunotherapy resolves renal fibrosis via revascularization and apoptosis of profibrotic Cxcr2+ endothelial cells

Wenyan Zhao,Xin Zhou,Xingli Zhao,Hao Tian,Yang Su,Shanlan Zhao,Min Liu,Qiao Zhang,Lin Chen,Xiaochen Li,Di Liu,Junxuan Li,Lang Li,Yanhong Wang,Xingtong Li,Jin Yan,Wen Chen,Bing Liu,Chuhong Zhu,Wen Zeng

Renal fibrosis is a common outcome of chronic kidney disease (CKD), forming a fibrotic niche characterized by fibroblast activation and vascular rarefaction. Currently, there are no effective treatment strategies targeting fibrotic niche. Here, we show that chimeric antigen receptor-modified M2 macrophages (CAR-M2) targeting FAP and secreting interleukin (IL)-4 are delivered via an injectable HAMA-CS hydrogel beneath the renal subcapsule and attenuate renal fibrosis while promoting renal revascularization. The single-cell RNA sequencing reveals the heterogeneity and interaction of stroma and endothelial cells (ECs). A fibrosis-related Cxcr2 + EC subset is identified, and its specific depletion effectively mitigates renal fibrosis. Further results reveal that CAR-M2 can release matrix metalloproteinase 2 (MMP2) in close proximity to activate retinoid X receptor alpha (Rxra) in the Cxcr2 + ECs and further triggers its mitochondrial autophagy, leading to apoptosis. Our research provides innovative strategies and proof of principle for the immunotherapy of organ fibrosis.

Ultrasound molecular imaging of M2 macrophages for early detection of chronic rejection in heart transplantation

Xu Jia,Deng Cheng,Gao Tang,He Mengrong,Fu Wenpei,Zhang Xin,Bai Yin,Qiu Jiani,Wang Rui,Chen Yihan,Jin Qiaofeng,Zhang Li,Lv Qing,Xie Mingxing,Wu Wenqian

Chronic rejection (CR) remains the leading cause of morbidity and mortality in heart transplantation survivors. The primary pathological features of CR encompass cardiac allograft vasculopathy and myocardial fibrosis. Currently, its diagnosis heavily relies on invasive procedures, underscoring the pressing need for non-invasive evaluation methods. This study introduces a novel approach utilizing mannose-modified microbubbles (MBman) targeting CD206 (mannose receptor) positive M2 macrophages for early CR detection. In vitro experiments demonstrate substantial adhesion of MBman to M2 macrophages compared to common microbubbles (MBcon). In a CR rat model, MBman and MBcon are administered at three distinct time points (2 weeks, 4 weeks, and 6 weeks), followed by contrast-enhanced ultrasound images and quantitative analysis using the ultrasound destruction-supplementation method. Starting at 2 weeks and continuing through 6 weeks, MBman demonstrates significantly higher signal intensity than MBcon in allograft rats. However, this difference is not observed in isograft rats at any of the indicated time points. These findings suggest an increase in M2 macrophage infiltration in allografts compared to isografts. Furthermore, the signal intensity of MBman positively correlates with the percentage of CD206 in allograft rats. This study proposes a promising approach, simultaneous noninvasive ultrasound molecular imaging, for the early-stage evaluation of CR.

Hydrogen peroxide photosynthesis from water and air using a scaled-up 1-m2 flow reactor

Xiaoshan Zheng,Rito Yanagi,Zhenhua Pan,Chong Zhou,Tian Liu,Baoliang Chen,Kenji Katayama,Shu Hu,Chiheng Chu

Particulate photocatalysis (PC) has shown great potential in sustainable chemical synthesis. Until now, developing a scalable PC system remains a major challenge hurdling the practical application of hydrogen peroxide (H 2 O 2 ) photosynthesis. Here, we report a flexible hydrophobic photocatalyst sheet based on visible-light-responsive bismuth vanadate (BiVO 4 ) photocatalysts with (λ < 520 nm) to achieve flow-transport-dependent cascade photocatalytic H 2 O 2 production. Using dissolved oxygen from the air and deionized water or tap water, the flexible sheets showed solar-to-chemical conversion (STC) efficiency of 0.11%. These BiVO 4 photocatalyst sheets were arranged in a 4 × 4-panels array in a 1-m 2 flow-by reactor and achieved 1-month outdoor stability under diurnal solar cycles. We utilized this solar-produced H 2 O 2 solution for disinfection, achieving >99.9% inactivation of a coronavirus surrogate in 60 min. Techno-economic analysis (TEA) shows that at 2% STC efficiency, the cost becomes comparable to commercial approaches due to the elimination of transportation, storage, and deployment costs.

Preparation of M2 macrophage membrane hybrid lipid nanoparticles based on microfluidic device for atherosclerosis treatment

Qi Liu,Li Qiao,Xia-Feng Peng,Shi-Qi Chang,Xiao Fang,Ya-Jun Li,Ning-Ping Huang

With the continuous advancement of bionanomaterial technology, the design and fabrication strategies of drug delivery systems have undergone significant strategic transformations and innovations. Herein, we report a microfluidic chip that enables one-step production of cell membrane hybrid lipid nanoparticles by fusing synthetic phospholipids with M2-macrophage membrane fragments and simultaneously encapsulating simvastatin and rapamycin. This greatly simplifies the preparation process, improves efficiency and produces M2-macrophage-membrane hybrid lipid nanoparticles loaded with simvastatin and rapamycin (M2LNPs@SIM&RAPA) with small particle size, homogeneous distribution and enhanced cumulative release in vitro. Western blotting and SDS-PAGE confirm the successful embedding of M2-membrane proteins. The formulation merges the long-circulating stability of synthetic phospholipids with the inflammation-targeting capacity and biocompatibility of M2 macrophage membranes. Stability tests and cell uptake experiments both proved its excellent storage stability and inflammation targeting. Cell cytotoxicity tests optimized the safe dosages of simvastatin and rapamycin. In the subsequent experiments, M2LNPs@SIM&RAPA could significantly reduce the expression of TNF-α in inflammatory cells and inhibit the formation of lipid droplets in foam cells, demonstrating its excellent anti-inflammatory activity and enhanced cholesterol excretion ability. In summary, the M2LNPs@SIM&RAPA prepared based on microfluidics can precisely regulate lipid metabolism and inflammatory responses through the synergistic effect of low-dose drugs, and is expected to provide a theoretical basis for the future treatment of atherosclerosis.

Ionizable cationic lipid nanoparticles loaded with miRNA-125b/BLZ945 for pancreatic cancer treatment

Jiajie Zhang,Ming Qu,Zhanhao Mo,He Sui,Lin Liu,Deliang Fu

In prior research, both miRNA-125b and BLZ945 have shown potential in effectively inhibiting M2 macrophage polarization and producing antitumor effects. Nevertheless, their physicochemical characteristics present significant challenges for efficient in vivo delivery. Ionizable cationic lipid nanoparticles (LNPs), recognized for their superior biocompatibility and drug-loading capacity, serve as a novel carrier for nucleic acid-based therapeutics. In our study, we successfully encapsulated both agents within LNPs and conducted a thorough characterization. Subsequently, we investigated their potential to repolarize M2 macrophages in vitro and evaluated their in vivo distribution, biosafety, and antitumor efficacy. The findings revealed that the LNPs maintained excellent drug-loading efficiency, consistent particle size, and stable zeta potential. All formulations effectively inhibited M2 macrophage polarization in vitro. Upon administration in vivo, the LNPs not only demonstrated favorable biosafety profiles but also accumulated efficiently in tumor tissues, substantially reducing tumor burden, particularly notable in co-loaded LNPs. Our results affirm that LNPs are an effective carrier for miRNA-125b and BLZ945, highlighting this encapsulation approach as promising for the treatment of solid tumors and meriting further investigation. Practitioner points : (i) Ionizable cationic nanoparticles provide high and stable encapsulation rates to efficiently load nucleic acid polymers into the LNP, avoiding the rapid accumulation of circulating macrophages, which can lead to reduced penetration of the LNP into target tissues. Therefore, it can be used as a novel drug delivery method to benefit clinical patients. (ii) miRNA-125b LNP/BLZ945 LNP attenuated the depleting effect of BLZ945 on macrophages and significantly inhibited macrophage M2 polarization. It could be effectively distributed in tumors and showed good biosafety while exerting antitumor effects, bringing hope to clinical pancreatic tumor patients.
