An electrophilicity-engineered magnetic sensor for MRI detection of dormant tumor cell clusters

Zeyu Liang,Bo Zhang,Xun Liu,Lin Xiao,Shangzhi Xie,Hui Du,Qiyue Wang,Fangyuan Li,Daishun Ling

In magnetic resonance imaging (MRI), direct dipole-dipole interactions between paramagnetic metal centers and water molecules govern the T 1 relaxation of contrast agents. Metal chelates featuring multiple unpaired electrons have long dominated MRI contrast agents. Despite theoretically offering more paramagnetic centers per probe, nanoparticle-based contrast agents have struggled because of the insufficient direct dipolar interactions with water, impeding their clinical adoption. Here, we present an electrophilicity-engineered magnetic sensor (EEMS), which leverages high-electronegativity metal atoms to enhance the electrophilicity of paramagnetic centers in nanosensors, enabling direct electrophilic catalytic dipolar interactions (ECD) with water for enhanced MRI. EEMS demonstrates robust T 1 contrast with a longitudinal relaxivity of 23.2 per millimolar per second at 9 tesla, visualizing tumor cell clusters as small as 68.5 micrometer in vivo. ECD-MRI allows detecting and precise resection of axillary lymph nodes containing dormant tumor cell clusters, achieving 100% survival in mice 100 days postsurgery. EEMS-enhanced ECD-MRI presents a transformative imaging principle for noninvasive visualization of previously undetectable biological entities.

Dynamic monitoring soft tissue healing via visualized Gd-crosslinked double network MRI microspheres

Chen Tongtong,Cai Zhengwei,Zhao Xinxin,Wei Gang,Wang Hanqi,Bo Tingting,Zhou Yan,Cui Wenguo,Lu Yong

By integrating magnetic resonance-visible components with scaffold materials, hydrogel microspheres (HMs) become visible under magnetic resonance imaging(MRI), allowing for non-invasive, continuous, and dynamic monitoring of the distribution, degradation, and relationship of the HMs with local tissues. However, when these visualization components are physically blended into the HMs, it reduces their relaxation rate and specificity under MRI, weakening the efficacy of real-time dynamic monitoring. To achieve MRI-guided in vivo monitoring of HMs with tissue repair functionality, we utilized airflow control and photo-crosslinking methods to prepare alginate-gelatin-based dual-network hydrogel microspheres (G-AlgMA HMs) using gadolinium ions (Gd (III)), a paramagnetic MRI contrast agent, as the crosslinker. When the network of G-AlgMA HMs degrades, the cleavage of covalent bonds causes the release of Gd (III), continuously altering the arrangement and movement characteristics of surrounding water molecules. This change in local transverse and longitudinal relaxation times results in variations in MRI signal values, thus enabling MRI-guided in vivo monitoring of the HMs. Additionally, in vivo data show that the degradation and release of polypeptide (K 2 (SL) 6 K 2 (KK)) from G-AlgMA HMs promote local vascular regeneration and soft tissue repair. Overall, G-AlgMA HMs enable non-invasive, dynamic in vivo monitoring of biomaterial degradation and tissue regeneration through MRI, which is significant for understanding material degradation mechanisms, evaluating biocompatibility, and optimizing material design.

ZIF-8 Film Coatings Render Polymeric Device Surfaces Addressable by 1H/129Xe MRI

Xinxin Yang,Qingbin Zeng,Jiawei Zhu,Xiuchao Zhao,Zhen Wang,Zhongyu Xiao,Yanling Feng,Yaping Yuan,Chenlu Yuan,Haidong Li,Sa Xiao,Louis-S. Bouchard,Xin Zhou,Qianni Guo

Metal-organic framework (MOF) films synergize molecular-scale porosity with conformal surface chemistry on polymer substrates. We report a spectrally selective coating in which a ZIF-8 film, seeded by a Zn(II)-polydopamine (Zn-PDA) interlayer on polytetrafluoroethylene (PTFE), enables hyperpolarized 1 2 9 Xe MRI detection of the polymer surface, while conventional 1 H MRI delineates anatomy. The coating produces a film-bound 1 2 9 Xe resonance at 82.0 ppm in both water and fetal bovine serum. After 60 min, the resonance position remained unchanged, with signal-to-noise ratios decreasing by only 14.9% and 17.2%, respectively. Selective excitation yields coating-specific positive contrast on the 1 2 9 Xe channel; correspondingly, 1 H MRI reveals a distinct surface appearance likely due to altered local proton density and magnetic susceptibility. In phantoms, 1 2 9 Xe and 1 H images were co-registered with a center-point offset of 0.34 mm; in a flow simulator, the offset was 0.26 mm. A rat airway experiment confirmed in vivo spectral selectivity with a film-bound resonance near 92 ppm, co-localizing with 1 2 9 Xe ventilation and 1 H anatomical references. While demonstrated here using coated PTFE segments and direct xenon bubbling, these results provide a robust proof of concept for MOF-coated polymers as spectrally selective 1 2 9 Xe MRI labels, warranting future physiological translation.

Hydrophilicity engineering of amphiphilic macrocyclic MRI contrast agents for enhanced hepatic targeting efficiency

Yuting Jiang,Zhongyuan Cai,Haojie Gu,Shengxiang Fu,Yingzi Cao,Na Li,Li Liu,Xiaomin Fu,Chunchao Xia,Feijing Su,Huaiqiang Sun,Kui Luo,Su Lui,Bin Song,Qiyong Gong,Hua Ai

Effective hepatic enhancement and high kinetic inertness are pivotal driving forces for developing gadolinium-based hepatobiliary MRI contrast agents (CAs). Although lipophilic modifications have dominated efforts to promote hepatic uptake, the vital role of hydrophilicity has been overlooked. Herein, we investigated the structure-activity relationship by fine-tuning of an amphiphilic macrocyclic complex through hydrophilicity engineering. The water-soluble Gd-HE(BnOPh)-DO3A exhibited exceptional kinetic inertness with a dissociation half-life of 8.3 h at pH 1.2, nearly 5-fold longer than the 1.7 h observed for the non-hydroxyl Gd-BnOBn-DO3A. More importantly, Gd-HE(BnOPh)-DO3A achieved rapid and pronounced hepatic enhancement, with a 3.1-fold increase in signal intensity observed as early as 2 min post-injection, while the amphiphilic Gd-BnOBn-DO3A displayed delayed hepatic enhancement, peaking at 30 min. These markedly different pharmacokinetic profiles might arise from altered molecular assembly behavior. Hydroxyl incorporation in Gd-HE(BnOPh)-DO3A disrupted amphiphilic equilibrium and prevented nanoparticle self-assembly, enabling rapid hepatic enhancement in the form of small molecules. Conversely, Gd-BnOBn-DO3A dynamically formed nanoparticles with hydrodynamic size over 200 nm, leading to slower hepatic uptake. In summary, this work established strategic hydrophilicity engineering as an effective, previously underappreciated approach to optimizing hepatic targeting of CAs, offering unique insights into MRI agent design.

Enzyme-Activated MRI for In Vivo Glucose Imaging via a Biodegradable Chromium Nanoprobe

Yan Xu,Weitao Yang,Yanjing Yun,Hui Wang,Zhuoyao Wu,Youyi Yu,Bingbo Zhang

Non-invasive visualization of glucose metabolism in living organisms remains a major challenge, as existing techniques cannot specifically detect glucose molecules with deep-tissue penetration and without ionizing radiation. To overcome this, we developed an enzyme-activated magnetic resonance imaging strategy (eaMRI) using a biodegradable nanoprobe, CrGOx@Lip, that integrates endogenous Cr 3+ ions with glucose oxidase for glucose-responsive imaging. Our approach leverages GOx not only as a catalytic engine to specifically oxidize glucose but also as a structural template to guide the in-situ synthesis of a potent MRI reporter (paramagnetic chromium gluconate). This design enables direct, specific amplification of MRI signals in proportion to local glucose concentration, achieving an 8.28-fold relaxivity increase. We validated this method for sensitive glucose mapping in vivo, including delineating Warburg-effect-driven tumors and quantifying pathological glucose accumulation in metabolic dysfunction-associated fatty liver disease (ΔSNR% = 18.51 ± 3.72 vs. 1.14 ± 1.39 in controls; p <0.001), and further demonstrated its utility in monitoring therapeutic efficacy via glucose-responsive signal changes. By synergizing enzymatic precision with nanomaterial engineering, our CrGOx@Lip-mediated eaMRI platform provides a glucose-specific, radiation-free, and non-invasive strategy for sensitive metabolic diagnostics in precision medicine.

Multiparametric MRI for In Vivo Visualization of Renal Inflammation in Acute Kidney Injury

Chuang Liu,Shuai Wu,Mingyu Zhang,Xinyu Ren,Yuqiang Ma,Zhuo Ao,Wenyue Li,Yi Hou

Renal ischemia-reperfusion injury (IRI) is a major cause of acute kidney injury (AKI), yet early diagnosis remains challenging owing to the lack of sensitive and specific approaches for in vivo inflammation visualization, which severely limits accurate diagnosis and effective intervention. To address this limitation, this work reports an inflammation-targeting multiparametric MRI strategy based on a nanoprobe (NPs@PEG-PEP) for real-time visualization of AKI-associated inflammatory responses. NPs@PEG-PEP was modified with functional peptides targeting CD137, which is an activation-induced immune receptor serving as an upstream mediator of inflammatory cascades. This approach enables nanoprobes to selectively target and accumulate surrounding inflammatory cells in AKI-affected kidneys. In addition, constructed from NaGdF 4 nanoparticles, the nanoprobe simultaneously enhances dynamic contrast-enhanced MRI (DCE-MRI) and susceptibility-weighted imaging (SWI) to capture distinct vascular perfusion differences and inflammatory cell infiltration between injured and contralateral kidneys. The inflammatory imaging signal persists for up to 18 h and correlates well with histopathological findings, including H&E staining and CD11b immunofluorescence. Moreover, the biological safety of the nanoprobe is confirmed via cytotoxicity assays, hemolysis tests, and histopathological evaluations. Overall, this work establishes an inflammation-targeting multiparametric MRI platform that enables spatiotemporal mapping of IRI-induced AKI progression, offering a promising strategy for early diagnosis and therapeutic monitoring.

UV-Activated Zwitterionic Fluorine-Containing Hybrid Probe for Dual-Modal 19F/1H MRI

Deshuo Wang,Wenze Xiao,Susu Yan,Zipeng Wang,Junnan Ma,Shoujing Sun,Jianjiang He,Jingjiang Sun,Qingfu Wang,Yanan Li,Wei Zhao

Dual-modal 19F/1H MRI probes often suffer from hydrophobic aggregation caused by high fluorine content, limiting their solubility and imaging performance. Herein, a water-soluble dual-modal probe (PFCP-Gd) was developed by grafting fluorinated phosphocholine zwitterions and Gd3+ chelates onto a UV-responsive polymer via RAFT polymerization and ring-opening reactions. The design enables UV-triggered paramagnetic relaxation enhancement (PRE) modulation, allowing for activatable 19F/1H MRI contrast. Upon UV irradiation, PFCP-Gd exhibited a significantly enhanced 19F signal with a T2 relaxation time of 23 ms and a signal-to-noise ratio of 11.34 at 12 mM 19F. The probe also showed good biocompatibility and an efficient cellular uptake. This work provides a strategy for constructing water-soluble, activatable fluorinated MRI probes for dual-mode imaging.

Superhydrophilic Fluorinated Polymer Probe for Zero-Background 19F MRI with Adaptable Targeting Ability

Chang Guo,Xiaoyao Xiong,Xinxing Zhao,Yumin Li,Sijia Li,Suying Xu,Tony D. James,Leyu Wang

19F magnetic resonance imaging (19F MRI), with zero background, high tissue penetration depth, excellent spatial resolution, and nonradioactive features, has attracted considerable attention but faces tough challenges due to the shortage of sensitive and selective targetable probes. Herein, we report a biocompatible and highly sensitive 19F MRI probe with an adaptable tumor-targeting ability. The fluorine-grafted polymer (PIBMA-FSON) probes were rich with sulfoxide and carboxy groups, containing a high fluorine content (∼17 wt %). The probes exhibit superhydrophilicity, strong 19F MRI signals (enhancement of ∼95-fold), long transverse relaxation time (T2, 422 ms), and excellent 19F MRI capability. Conjugation using a targeting peptide (Arg-Gly-Asp, RGD) afforded ultrasmall soft polymer probes (PIBMA-FSON-RGD) with superhydrophilicity and tumor-targeting ability suitable for the 19F MRI of orthotopic bladder cancer. Amidification of 5% of the carboxylate units with oleylamine resulted in PIBMAOAm-FSON nanoprobes (NPs) via self-assembly, displaying different targeting toward subcutaneous tumors. Further grafting with near-infrared (NIR) dyes renders the probe suitable for NIR-fluorescence and 19F MRI dual-modality imaging. This study provides a suitable approach for designing highly sensitive and zero-background 19F MRI probes with a tunable tumor-targeting ability.

Mesothelin-targeted MRI for assessing migration, invasion, and prognosis in malignant pleural mesothelioma

Huang Yilong,Shen Shasha,Xiao Jie,Luo Cici,Ma Jiyao,Huang Xin,Qi Tianfu,Gao Chao,Li Guiyun,Li Fan,He Bo,Chen Bingdi,Han Dan

Background Mesothelin (MSLN) has been implicated in cancer migration, invasion, and prognosis, making it a potential tumor marker. However, the precise role of MSLN in the migration and invasion of malignant pleural mesothelioma (MPM) remains elusive, and effective noninvasive methods for assessing MSLN status are currently lacking. In this study, we focused on MSLN expression and elucidated the underlying mechanisms by which MSLN regulates migration and invasion in MPM. Building upon this knowledge, we developed an MRI nanoprobe that targets MSLN to assess its status in vitro and in vivo by comparing T2 signal intensity and T2 values on magnetic resonance imaging examinations. This nanoprobe combines the anatomical information obtained from MRI with biological information obtained from MSLN for comprehensive evaluation of MPM. Results Notably, we observed that MSLN expression in the epithelial type of MPM was higher and increased continuously with tumor growth than that in other types. In addition, MSLN upregulation promoted N-cadherin, matrix metalloproteinase-7, and MMP9 expression and resulted in higher migration/invasion ability and shorter survival. We synthesized MSLN-targeted nanoprobes (Fe 3 O 4 @SiO 2 -PEG-MSLN, FSPM) to assess MSLN expression by comparing the T2 signal intensity and T2 value of different cell lines and mice after 14, 28, and 42 days of modeling. Remarkably, MSLN-targeted nanoprobes demonstrated excellent targeting capabilities. In vitro studies revealed a pronounced reduction in T2 signal intensity and T2 values of the epithelial type as the probe concentration increased. In addition, in vivo experiments demonstrated a gradual decline in these parameters over time, particularly in the epithelial type as compared to the biphasic type, corresponding to the dynamic expression patterns of MSLN during different growth stages. Conclusion Our comprehensive research succeeded in confirming the regulatory mechanisms by which MSLN influences migration and invasion. Moreover, we introduced a promising method for monitoring MSLN expression that may help in facilitating the early detection, histological subtype identification, and assessment of migration, invasion, and prognosis in MPM.

In situ targeted MRI detection of Helicobacter pylori with stable magnetic graphitic nanocapsules

Li Yunjie,Hu Xiaoxiao,Ding Ding,Zou Yuxiu,Xu Yiting,Wang Xuewei,Zhang Yin,Chen Long,Chen Zhuo,Tan Weihong

Helicobacter pylori infection is implicated in the aetiology of many diseases. Despite numerous studies, a painless, fast and direct method for the in situ detection of H. pylori remains a challenge, mainly due to the strong acidic/enzymatic environment of the gastric mucosa. Herein, we report the use of stable magnetic graphitic nanocapsules (MGNs), for in situ targeted magnetic resonance imaging (MRI) detection of H. pylori . Several layers of graphene as the shell effectively protect the magnetic core from corrosion while retaining the superior contrast effect for MRI in the gastric environment. Boronic-polyethylene glycol molecules were synthesized and modified on the MGN surface for targeted MRI detection. In a mouse model of H. pylori -induced infection, H. pylori was specifically detected through both T 2 -weighted MR imaging and Raman gastric mucosa imaging using functionalized MGNs. These results indicated that enhancement of MRI using MGNs may be a promising diagnostic and bioimaging platform for very harsh conditions.

Fe3O4@M nanoparticles for MRI-targeted detection in the early lesions of atherosclerosis

Xin Huang,Chenyu Lin,Cici Luo,Yuhan Guo,Jun Li,Yiping Wang,Jiahong Xu,Yuwen Zhang,He Wang,Zhongmin Liu,Bingdi Chen

Atherosclerosis can lead to most cardiovascular diseases. Although some biomimetic nanomaterials coated by macrophage membranes have been reported in previous studies of atherosclerosis, to our knowledge, no studies regarding the detection of early lesions of atherosclerosis (foam cells) using such a strategy have yet been reported. In the present study, Fe 3 O 4 biomimetic nanoparticles coated with a macrophage membrane (Fe 3 O 4 @M) were prepared to investigate the imaging effect on the early lesions of atherosclerosis (foam cells). The results showed that the Fe 3 O 4 @M particles are spheres with average diameters of approximately 300  nm . T1 and T2 relaxation values showed that the ratio of r2 to r1 was 26.09. The protein content accounted for approximately 27% of the total weight in Fe 3 O 4 @M, and Fe 3 O 4 @M nanoparticles exhibited high biosafety. Further testing showed that Fe 3 O 4 @M effectively targets early atherosclerotic lesions by the specific recognition of integrin α4β1 to VCAM-1. Taken together, Fe 3 O 4 @M is a promising contrast agent for the diagnosis of early stage atherosclerosis.

Biodegradable organosilica magnetic micelles for magnetically targeted MRI and GSH-triggered tumor chemotherapy

Tiangang Yang,Dechao Niu,Jianzhuang Chen,Jianping He,Shaobo Yang,Xiaobo Jia,Jina Hao,Wenru Zhao,Yongsheng Li

Recently, block copolymer micelles have attracted widespread attention due to their controlled biodegradability and excellent loading capability. Unfortunately, the poor in vivo stability and low delivery efficiency of drug-loaded micelles greatly hampered their biomedical applications. Herein, we develop a new kind of biodegradable magnetite/doxorubicin (Fe3O4/DOX) co-loaded PEGylated organosilica micelles (designated as FDPOMs) with both high circulating stability and smart GSH-triggered biodegradability for magnetically targeted magnetic resonance imaging (MRI) and tumor chemotherapy. The FDPOMs are prepared by the self-assembly of biodegradable polycaprolactone-block-poly(glutamic acid) (PCL-b-PGA), a chemotherapeutic DOX drug and Fe3O4 nanoparticles in an oil/water system, subsequent organosilica cross-linking with 3-mercaptopropyltrimethoxysilane (MPTMS) molecules and surface PEGylation. The resultant FDPOMs exhibit excellent dispersity and stability in biological media, remarkable T2-weighted MR imaging capability, unique GSH-responsive release behavior and selective toxicity to tumor cells. The in vivo experiments show that the FDPOMs not only have improved MR tumor imaging capability, but also exhibit high anti-tumor efficacy due to the strong magnetic targeting ability under an external magnetic field. Consequently, the FDPOMs are promising candidates for magnetically targeted MR imaging and imaging-guided tumor chemotherapy.
