Cell membrane damage induced by continuous stress of artemisinin sustained-release microspheres (ASMs) on Microcystis aeruginosa at different physiological stages

Ni Lixiao,Yue Feifei,Zhang Jianhua,Rong Shiyi,Liu Xuanyu,Wang Yifei,Wang Peifang,Li Danye,Wang Na,Wu Hanqi,Li Shiyin

Artemisinin sustained-release microspheres (ASMs) with long-term inhibition effects (> 40 days) on harmful freshwater bloom-forming cyanobacteria have been found in previous studies, but the inhibition mechanism is not completely clear. In the present study, we examined the growth effect of ASMs on Microcystis aeruginosa ( M. aeruginosa ) cells at different physiological stages. Growth experiments indicated that M. aeruginosa of different initial densities could be inhibited immediately and chlorophyll-a content both showed significant decreases following exposure of cyanobacteria to optimal dosage of ASMs for 20 days. The algicidal mechanism of ASMs was tested through a suite of physiological parameters (membrane permeability, antioxidant enzymes activity, and lipid peroxidation). The rise of cell membrane permeability indices (intracellular protein, nucleic acid contents, and conductivity) showed that the cellular membrane structure of M. aeruginosa was attacked by ASMs directly causing the leakage of cytoplasm. Antioxidant enzyme activity was a sensitive indicator of the impacts of ASMs which showed a significant downtrend after a few days. ASMs caused a great increase in •O 2 − and malondialdehyde (MDA) level of the algal cells which indicated the increase in lipid peroxidation of M. aeruginosa. Irreversible membrane damage induced by ASMs via the oxidation of ROS may be an important factor responsible for the algicidal mechanism of ASMs on M. aeruginosa cells . The application of ASMs might provide a new direction to control M. aeruginosa , especially before the exponential phase according to the optimal economy and inhibition effect. Graphical abstract

Microcystis aeruginosa co-exposure to bisphenol AF and perfluorooctanoic acid promotes cell division and increases microcystin and nitrous oxide emissions

Hao Ming,Jinhua Mou,Carol Sze Ki Lin,Liandong Zhu

The global rise in cyanobacterial blooms risk parallels the frequent detection of endocrine-disrupting compounds (EDCs) in freshwater, raising the unexplored possibility that EDCs may covertly trigger and sustain these events. This study investigated how bisphenol AF (BPAF) and perfluorooctanoic acid (PFOA) modulated the physiology of Microcystis aeruginosa under different exposure concentrations and sequences. Single-cell transcriptomic and proteomic analyses revealed that low-level co-exposure (0.5 μM) to these chemicals enhanced cell division by 28.99 % via FtsZ upregulation and increased microcystin release by 20.11 %, accompanied by elevated tryptophan- and tyrosine-like fluorescence peaks. High-concentration exposure (5 μM) induced oxidative stress, disrupted cell division, and triggered cell enlargement and secretion of dissolved organic matters. BPAF stimulated nitrous oxide (N₂O) production to 885.11 ng/(10 6 cells·h), whereas PFOA suppressed emissions, compared to 8.93 ng/(10 6 cells·h) in the control group. The nitrogen regulator NtcA was identified as a central node linking nitrogen metabolism, oxidative stress and photosynthetic redox reactions with N₂O synthesis. Molecular docking supported stable interactions between NtcA and both compounds. These findings uncovered distinct regulatory strategies in Microcystis aeruginosa under chemical stress, offering mechanistic insights into how endocrine-disrupting pollutants reshape microbial growth and greenhouse gas emissions in aquatic ecosystems.

Butylparaben-Loaded Aged Polystyrene Nanoplastics Amplify Its Toxicity in Microcystis aeruginosa via Quorum Sensing Suppression and Enhanced Microcystin-LR Release

Zhong-Hong Zhang,Yi-Fan Zhu,Yu-Cheng Yang,Yuan-Zhen Xiao,Shun-Ni Zhu,Wei-Dong Yang,Hong-Ye Li,Xiang Wang

Aged nanoplastics are emerging pollutants in aquatic environments, but the effects of their loaded pollutants on cyanobacteria are still poorly understood. This study evaluated the adsorption of butylparaben (BP) by pristine (PS) and aged polystyrene nanoplastics (APS) and prepared BP-loaded aged PS to analyze their effects on Microcystis aeruginosa. The results showed that APS had stronger BP adsorption and translocation capacity, with APS increasing BP adsorption by 4.34-fold and significantly enhancing its bioconcentration in M. aeruginosa. Compared with the PS–BP treatment, the 1.13 ng/g APS–BP treatment significantly inhibited algal growth, exacerbated cell membrane damage and oxidative stress, and promoted MC-LR synthesis and release. Transcriptome analyses and molecular docking showed that BP inhibited quorum sensing (QS) by binding to luxR, whereas APS further enhanced the binding affinity of BP and blocked the binding of N-acylhomoserine lactone, a natural QS signaling molecule, to its receptor, amplifying the QS inhibitory effect. QS inhibition downregulated genes related to photosynthesis, membrane synthesis, and oxidative stress and upregulated genes related to MC-LR synthesis. These findings revealed that APS is a pollutant carrier that exacerbates cyanobacterial toxicity and MC-LR release through pollutant adsorption, posing significant ecological risks to aquatic ecosystems.

Efficacy and mechanism of Microcystis aeruginosa removal by activated carbon fiber/nanoscale zero-valent iron/nickel foam cathodic electrochemical system

Guoming Zeng,Rui Zhang,Cenhui Liao,Dong Liang,Fei Wang,Qihui Wang,Da Sun,Xiaoling Lei

Cyanobacterial outbreaks seriously affect drinking water safety. Therefore, developing effective algal removal technologies is urgently needed. The study aims to investigate the effects of pH, electrode distance, current density, and initial algal concentration on electrochemical algae collection. Consequently, we loaded nanoscale zero-valent iron (nZVI) on activated carbon fiber/nickel foam (ACF/Ni) to form an ACF/nZVI/Ni composite cathode using the liquid phase reduction method and used titanium-based platinum plating (Pt/Ti) as the anode to construct a novel Pt/Ti- ACF/nZVI/Ni electrochemical system. The results showed that on pH of 6.0, the electrode distance was 10 mm, the current density was 75 mA/cm2, and the initial algal concentration was OD680 = 0.100; the algal cell removal rate was 94.20% after electrolysis for 30 min. This paper provides an efficient and environmentally friendly electrochemical algal removal technology for protecting the safety of drinking water and dealing with cyanobacterial outbreaks.

Repeated Exposure Enhanced Toxicity of Clarithromycin on Microcystis aeruginosa Versus Single Exposure through Photosynthesis, Oxidative Stress, and Energy Metabolism Shift

Yufeng Mao,Kailai Ye,Shengfa Yang,Muhammad Salam,Weiwei Yu,Qiang He,Ruixu He,Hong Li

Antibiotics are being increasingly detected in aquatic environments, and their potential ecological risk is of great concern. However, most antibiotic toxicity studies involve single-exposure experiments. Herein, we studied the effects and mechanisms of repeated versus single clarithromycin (CLA) exposure on Microcystis aeruginosa. The 96 h effective concentration of CLA was 13.37 μg/L upon single exposure but it reduced to 6.90 μg/L upon repeated exposure. Single-exposure CLA inhibited algal photosynthesis by disrupting energy absorption, dissipation and trapping, reaction center activation, and electron transport, thereby inducing oxidative stress and ultrastructural damage. In addition, CLA upregulated glycolysis, pyruvate metabolism, and the tricarboxylic acid cycle. Repeated exposure caused stronger inhibition of algal growth via altering photosynthetic pigments, reaction center subunits biosynthesis, and electron transport, thereby inducing more substantial oxidative damage. Furthermore, repeated exposure reduced carbohydrate utilization by blocking the pentose phosphate pathway, consequently altering the characteristics of extracellular polymeric substances and eventually impairing the defense mechanisms of M. aeruginosa. Risk quotients calculated from repeated exposure were higher than 1, indicating significant ecological risks. This study elucidated the strong influence of repeated antibiotic exposure on algae, providing new insight into antibiotic risk assessment.

The inhibition mechanism and death mode of Microcystis aeruginosa induced by the continuous pressure of artemisinin sustained-release microspheres (ASMs)

Cunhao Du,Lixiao Ni,Jakpa Wizi,Wenlu Sang,Shiyi Rong,Xianglan Li,Chu Xu,Yan Li,Xuqing Chen,Shiyin Li

Artemisinin , a natural allelochemical substance extracted from Artemisia annua , has been shown to have strong inhibitory effects on harmful cyanobacteria . However, only a few studies have systematically studied the mechanism of artemisinin in inhibiting algae and the process of algae cell death. In this study, the inhibition mechanism and death mode of M. aeruginosa under artemisinin sustained-release microspheres (ASMs) were investigated using a series of morphological, physiological and biochemical characteristics. The findings revealed that M. aeruginosa was significantly inhibited immediately after exposure to the 0.54 g·L −1 ASMs, with an inhibition rate (IR) exceeding 95 %, and the synthesis of phycobiliprotein and esterase activity were disturbed. In addition, ASMs exposure significantly increased caspase-3(-like) activities of M. aeruginosa . Under the continuous stress of ASMs, M. aeruginosa cells had three cell death modes: the first was necrosis-like cell death, which was characterized by cell wall rupture and leakage of cell contents; the second was apoptosis-like cell death, which was characterized by the disintegration of the cell structure, the integrity of the cell membrane , and the appearance of apoptotic bodies; and the last was autophagy-like cell death, which was characterized by cell swelling and appearance of autophagy vacuoles. These results indicated that ASMs could inhibit photosynthesis of M. aeruginosa cells, interfere with its antioxidant mechanisms and activate caspase-3(-like) activity, resulting in programmed cell death (PCD).

Impact of melatonin on the hydrogen peroxide treatment efficacy in Microcystis aeruginosa: Cell growth, oxidative stress response, and gene transcription

Giridhar Babu Anam,Dinneswara Reddy Guda,Young-Ho Ahn

A study was conducted to determine how melatonin (MLT), a growth regulator, affects Microcystis aeruginosa cell behaviour and how MLT exposed cells respond to hydrogen peroxide (H 2 O 2 ) treatment. MLT promotes the growth, chl- a content, Fv/Fm values, and microcystins (MCs) production of M. aeruginosa at low concentrations of 1–2.5 μmol/L but suppresses the growth at high concentrations (5–10 μmol/L). The cellular and genetic responses of MLT pre-treated cells to H 2 O 2 treatment were examined further. Further research found that the cells pre-treated with MLT were susceptible to a range of growth-promoting, inhibiting and lethal effects when exposed to higher levels of H 2 O 2 . A dose-dependent pattern was observed under conditions of 0.05–0.2 mmol/L H 2 O 2 with 0.5–2.5 μmol/L MLT concentrations to different degrees. High doses of H 2 O 2 (0.2 and 0.3 mmol/L) typically lead to cell lysis and release of MCs in 5.0 and 10 μmol/L MLT pre-treated cells. A decrease in SOD/CAT activities and an increase in MDA levels validated the growth reduction. Furthermore, higher cell lysis and release of intracellular MCs were observed when H 2 O 2 was increased for 5–10 μmol/L MLT pre-treated cells. This led to a higher accumulation of extracellular MCs. The results provide insight into how MLT influences H 2 O 2 damage and assist in identifying situations where H 2 O 2 treatment of cyanobacterial blooms is most appropriate.

Abundant Allelochemicals and the Inhibitory Mechanism of the Phenolic Acids in Water Dropwort for the Control of Microcystis aeruginosa Blooms

Jixiang Liu,Yajun Chang,Linhe Sun,Fengfeng Du,Jian Cui,Xiaojing Liu,Naiwei Li,Wei Wang,Jinfeng Li,Dongrui Yao

In recent years, with the frequent global occurrence of harmful algal blooms, the use of plant allelopathy to control algal blooms has attracted special and wide attention. This study validates the possibility of turning water dropwort into a biological resource to inhibit the growth of harmfulMicrocystis aeruginosablooms via allelopathy. The results revealed that there were 33 types of allelopathic compounds in the water dropwort culture water, of which 15 were phenolic acids. Regarding water dropwort itself, 18 phenolic acids were discovered in all the organs of water dropwort via a targeted metabolomics analysis; they were found to be mainly synthesized in the leaves and then transported to the roots and then ultimately released into culture water where they inhibitedM. aeruginosagrowth. Next, three types of phenolic acids synthesized in water dropwort, i.e., benzoic, salicylic, and ferulic acids, were selected to clarify their inhibitory effects on the growth ofM. aeruginosaand their mechanism(s) of action. It was found that the inhibitory effect of phenolic acids on the growth ofM. aeruginosaincreased with the increase of the exposure concentration, although the algae cells were more sensitive to benzoic acid than to salicylic and ferulic acids. Further study indicated that the inhibitory effects of the three phenolic acids on the growth ofM. aeruginosawere largely due to the simultaneous action of reducing the number of cells, damaging the integrity of the cell membrane, inhibiting chlorophyll a (Chl-a) synthesis, decreasing the values ofF0andFv/Fm, and increasing the activity of the antioxidant enzymes (SOD, POD, and CAT) ofM. aeruginosa. Thus, the results of this study indicate that both culture water including the rich allelochemicals in water dropwort and biological algae inhibitors made from water dropwort could be used to control the growth of noxious algae in the future. Keywords:Microcystis aeruginosa;biological control;water dropwort;allelopathic metabolite;phenolic acids;cellular structure;photosynthesis;antioxidant enzyme

Effects of Dihydroartemisinin and Artemether on the Growth, Chlorophyll Fluorescence, and Extracellular Alkaline Phosphatase Activity of the Cyanobacterium Microcystis aeruginosa

Shoubing Wang,Ziran Xu

Increased eutrophication in the recent years has resulted in considerable research focus on identification of methods for preventing cyanobacterial blooms that are rapid and efficient. The objectives of this study were to investigate the effects of dihydroartemisinin and artemether on the growth of Microcystis aeruginosa and to elucidate its mode of action. Variations in cell density, chlorophyll a, soluble protein, malondialdehyde, extracellular alkaline phosphatase activity (APA), and chlorophyll fluorescence parameters (Fv/Fm, ΦPSII, ETR, rapid light curves, fast chlorophyll fluorescence curves on fluorescence intensity, and relative variable fluorescence) were evaluated by lab-cultured experiments. Our results demonstrated that both dihydroartemisinin and artemether inhibited the growth of M . aeruginosa by impairing the photosynthetic center in photosystem II and reducing extracellular APA, with a higher sensitivity exhibited toward artemether. The inhibitory effects of dihydroartemisinin on M . aeruginosa increased with concentration, and the maximum growth inhibitory rate was 42.17% at 24 mg·L -1 after 120h exposure, whereas it was 55.72% at 6 mg·L -1 artemetherafter 120h exposure. Moreover, the chlorophyll fluorescence was significantly inhibited (p<0.05) after 120h exposure to 12 and 24 mg·L -1 dihydroartemisinin. Furthermore, after 120h exposure to 6 mg·L -1 artemether, Fv/Fm, ΦPSII, ETR and rETR max showed a significant decrease (p<0.01) from initial values of 0.490, 0.516, 17.333, and 104.800, respectively, to 0. One-way analysis of variance showed that 6 mg·L -1 artemether and 24 mg·L -1 dihydroartemisinin had significant inhibitory effects on extracellular APA (p<0.01). The results of this study would be useful to further studies to validate the feasibility of dihydroartemisinin and artemether treatment to inhibit overall cyanobacterial growth in water bodies, before this can be put into practice.

Cell density-dependent regulation of microcystin synthetase genes (mcy) expression and microcystin-LR production in Microcystis aeruginosa that mimics quorum sensing

Shanlin Wang,Ping Ding,Siyu Lu,Pian Wu,Xiaoqian Wei,Ruixue Huang,Tianhan Kai

As the secondary metabolites of cyanobacterial harmful algal blooms (Cyano-HABs), microcystins (MCs) were generated under various environmental and cellular conditions. The understanding of the causes of MCs generation is of great interest in the field of water treatment and environmental science. In this work, we studied how Microcystis aeruginosa (FACHB-905) cell densities affect the MCs synthetase genes ( mcy ) expression, microcystin-LR (MC-LR) and quorum sensing molecules (Acyl-homoserine lactones (AHLs)) production. An electrochemical sensor was developed here for sensitive and quantitative detection of MC-LR that cultured at different cell densities. The results showed that mcy expression and MC-LR concentration started to increase when the cell density reached ca. 22 × 10 6 cells/mL, and was significantly increased with increasing cell densities. Moreover, the up-regulation of AHLs with increasing cell densities revealed that MC-LR is quorum sensing-mediated. Our results undoubtedly confirmed that MC-LR was produced in a cell density-dependent way that mimics quorum sensing, and the minimum cell density ( ca. 22 × 10 6 cells/mL) that was required to produce MC-LR was provided and offered a reference standard for the prevention and control of MCs pollution in the actual water environment.

Activation of peroxymonosulfate by La2CuO4 perovskite for synergistic removal of Microcystis aeruginosa and microcystin-LR in harmful algal bloom impacted water

Panpan Gao,Yunyi He,Shihuan Lu,Mengfan He,Zhiqun Liu,Yang Deng,Zhiquan Liu,Ting Xu,Hangjun Zhang

The cyanobacterial blooms induced water safety problems have attracted tremendous concerns due to the adverse influences on aqueous ecosystem and human health. This study developed a novel perovskite-type catalyst for heterogeneous peroxymonosulfate (PMS) activation as an emergency measure to remove harmful algae. The results indicated that significant removal efficiency of Microcystis aeruginosa cells (97%) and chlorophyll-a (100%) was achieved within 20 min in La 2 CuO 4 /PMS system without significant release of dissolved organic carbon and metals in water. • OH, SO 4 •- , 1 O 2 and • O 2 - generated were all responsible for algae inactivation and subsequent algal organic matters removal. Moreover, the heterogeneous catalytic mechanism was proposed that La portion in La 2 CuO 4 favored the cells destabilization by increasing zeta potential of algae suspension, while variable Cu portion facilitated the electron transfer to improve the yields of • OH and SO 4 •- . This study demonstrates that the new catalytic PMS process promises viable and effective control of harmful algal bloom impacts in water treatment.

Toxicity of the disinfectant benzalkonium chloride (C14) towards cyanobacterium Microcystis results from its impact on the photosynthetic apparatus and cell metabolism

Yunlu Jia,Yi Huang,Jin Ma,Shangwei Zhang,Jin Liu,Tianli Li,Lirong Song

Quaternary ammonium compounds (QACs) are commonly used in a variety of consumer and commercial products, typically as a component of disinfectants. During the COVID-19 pandemic, QACs became one of the primary agents utilized to inactivate the SARS-CoV-2 virus on surfaces. However, the ecotoxicological effects of QACs upon aquatic organisms have not been fully assessed. In this study, we examined the effects of a widely used QAC (benzalkonium chloride-C 14 , BAC-14) on two toxigenic Microcystis strains and one non-toxigenic freshwater Microcystis strain and carried out an analysis focused on primary, adaptive and compensatory stress responses at apical (growth and photosynthesis) and metabolic levels. This analysis revealed that the two toxic Microcystis strains were more tolerant than the non-toxic strain, with 96 hr-EC 50 values of 0.70, 0.76, and 0.38 mg/L BAC-14 for toxigenic M. aeruginosa FACHB-905, toxigenic M. aeruginosa FACHB-469, and non-toxigenic M. wesenbergii FACHB-908, respectively. The photosynthetic activities of the Microcystis , assessed via F v /F m values, were significantly suppressed under 0.4 mg/L BAC-14. Furthermore, this analysis revealed that BAC-14 altered 14, 12, and 8 metabolic pathways in M. aeruginosa FACHB-905, M. aeruginosa FACHB-469, and M. wesenbergii FACHB-908, respectively. It is noteworthy that BAC-14 enhanced the level of extracellular microcystin production in the toxigenic Microcystis strains , although cell growth was not significantly affected. Collectively, these data show that BAC-14 disrupted the physiological and metabolic status of Microcystis cells and stimulated the production and release of microcystin, which could result in damage to aquatic systems.
