Nanotheranostics with integrated imaging functions can help monitor nanoparticle accumulation in tumors, thus achieving synergism and higher therapeutic accuracy in cancer therapy. However, it remains challenging to monitor the release of therapeutic drugs in real time from a nanoparticulate drug delivery system (nano-DDS) in the body. Herein, we developed a nano-DDS for fluorescence imaging in the second near-infrared window (NIR-II) region, which can be used for monitoring the responsive release of drugs and cancer-targeted combined photodynamic and chemotherapy. There is a linear correlation between the cumulative release of the drug and the NIR-II fluorescence intensity. Moreover, hyaluronidase/glutathione dual-response RGD-SS-DOX/Ce6@HA-IR-1061 (RSSDCHI) exhibited a higher tumor-to-normal-tissue ratio in NIR-II fluorescence imaging and enhanced antitumor efficacy in vivo. This makes it possible to visualize drug release at the cellular level by the nanocomposites and to predict the treatment effect according to the NIR-II fluorescence intensity in the tumor site, serving as a promising nanoplatform for precision nanomedicine.

Mitochondrial membrane potential (Δ Ψ m)-targeting molecular probes play an essential role in diagnosing and treating diseases. Recently, the second near-infrared window (NIR-II, 1000–1700 nm) fluorescent imaging has been actively studied as an attractive imaging modality. However, small molecule Δ Ψ m-targeting NIR-II probes are rarely reported, especially for highly efficient imaging and treatment of tumors. Herein, a small molecule probe named TQPTPP is designed and synthesized by conjugation of a novel D-A type dye TQT1009 with Δ Ψ m-targeting molecule alkyl triphenylphosphine (TPP) through a polyethylene glycol-8 (PEG 8 ) linker. The conventional ICG dye is also coupled with TPP through PEG 8 to produce ICGTPP as a comparison. TQPTPP showed a fluorescence quantum yield of 0.041% and excellent photothermal conversion efficiency (61.4%). It can be self-assembled into nanoparticles and still preserve Δ Ψ m-targeting capability. Tumor imaging is further performed, and results showed a long tumor retention time of TQPTPP (maximum tumor signal on day five and signal-to-noise ratio up to nine). As a comparison, ICGTPP remained a single molecule with Δ Ψ m-targeting capability. But it has shorter tumor retention and lower photostability. These results suggested the novel D-A small molecule ΔΨ m-targeting NIR-II probe TQPTPP provided a new tool for diagnosing and treating tumors.

The colon tumor microenvironment has a high concentration of H 2 S and glutathione, which is highly immunosuppressive and adverse to multiple therapeutic methodologies such as ferroptosis. Here, an engineered microbial nanohybrid based on Escherichia coli ( E. coli ) and Cu 2 O nanoparticles to specific colon tumor therapy and immunosuppression reversion is reported. The as-prepared E. coli @Cu 2 O hybrid can accumulate in tumor sites upon intravenous injection, and Cu 2 O nanoparticles convert to Cu x S by consuming the endogenous H 2 S, which exhibits strong photothermal conversion at near-infrared II (NIR II) biological window. Furthermore, E. coli @Cu 2 O is able to induce cellular ferroptosis and cuproptosis through inactivation of glutathione peroxidase 4 and aggregation of dihydrolipoamide S-acetyltransferase, respectively. Photothermal-enhanced ferroptosis/cuproptosis achieved by E. coli @Cu 2 O reverses the immunosuppression of colon tumors by triggering dendritic cell maturation (about 30%) and T cell activation (about 50% CD8 + T cells). Concerted with immune checkpoint blockade, the engineered microbial nanohybrid can inhibit the growth of abscopal tumors upon NIR illumination. Overall, the designed microbial nanohybrid can achieve tumor-specific photothermal-enhanced ferroptosis/cuproptosis and immunosuppression reversion, showing promise in precise tumor therapy in future clinical translation.

Encouraging progress in multifunctional nanotheranostic agents that combine photothermal therapy (PTT) and different imaging modalities has been made. However, rational designed and biocompatible multifunctional agents that suitfable for in vivo application is highly desired but still challenging. In this work, we rationally designed novel ultrasmall multifunctional nanodots (FS-GdNDs) by combining the bovine serum albumin (BSA)-based gadolinium oxide nanodots (GdNDs) obtained through a biomineralization process with a small-molecule NIR-II fluorophore (FS). The as-prepared FS-GdNDs with an ultrasmall hydrodynamic diameter of 9.3 nm exhibited prominent NIR-II fluorescence properties, high longitudinal relaxivity (10.11 mM −1 s −1 ), and outstanding photothermal conversion efficiency (43.99%) and photothermal stability. In vivo studies showed that the FS-GdNDs with enhanced multifunctional characteristics diaplayed satisfactory dual-modal MR/NIR-II imaging performance with a quite low dose. The imaging-guided PTT achieved successful ablation of tumors and effectively extended the survival of mice. Cytotoxicity studies and histological assay demonstrated excellent biocompatibility of the nanodots. Importantly, this novel FS-GdNDs can undergo efficient body clearance through both hepatobiliary and renal excretion pathways. The novel ultrasmall multifunctional FS-GdNDs with excellent features hold tremendous potential in biomedical and clinical applications.

ABSTRACT Radiotherapy plays a critical role in the clinical treatment of breast cancer. However, the efficacy of traditional X-ray radiotherapy is greatly limited by its low tumor specificity and treatment tolerance mediated by the tumor microenvironment. Herein, we proposed a novel nano-radiotherapy sensitization strategy to design and construct a cancer cell membrane-coated siRNA-decorated Au/MnO 2 nanosensitizer (R&F@Au/MnO 2 -CM) to synergistically enhance radio-immunotherapy for breast cancer. In the integrated nanosensitizer, the cancer cell membrane (CM) derived from 4T1 breast cancer cells is utilized for targeted functionality, while Au/MnO 2 is designed to improve X-ray absorption and alleviate tumor hypoxia. Additionally, PD-L1 siRNA (R) is used to downregulate PD-L1 expression in tumor cells. In an in situ mouse model of 4T1 breast cancer, R&F@Au/MnO 2 -CM demonstrated accurate tumor identification via CM-mediated homologous targeting after intravenous injection, which was monitored in real-time through NIR-II fluorescence imaging of NIR-935 (F). Subsequently, the radiotherapy sensitivity was achieved due to the strong radiation absorption properties and oxygen generation through catalysis of Au/MnO 2 upon X-ray irradiation. Furthermore, the immunosuppressive microenvironment of the tumor is improved by downregulating PD-L1, enhancing synergistic anti-tumor effect. Our findings demonstrate a promising approach for tumor treatment by combining targeted enhanced radiotherapy with immune activation.

Enlightened by the great success of drug repurposing strategy in pharmaceutic industries, in the current study, material repurposing was proposed where the performance of carbonyl iron powder (CIP), a nutritional intervention agent of iron supplement approved by the US FDA for iron deficiency anemia in clinical was explored in anti-cancer treatment. Except for the abnormal iron metabolic characteristics of tumors that serving as potential targets for CIP-based cancer therapy under repurposing paradigm, the efficacy of CIP as catalyst in Fenton reaction, activator for dihydroartemisinin (DHA) and thus increase the chemo-sensitivity of tumors, as well as the potent agent for NIR-II photothermal therapy (PTT) was fully evaluated by an injectable alginate hydrogel form. The CIP-ALG gel could cause rapid temperature rise in the tumor site under NIR-II laser irradiation, leading to complete ablation in the primary tumor. Further on, such photothermal-ablation would lead to ATP release significantly and in the bilateral tumor model, both primary tumor ablation and inhibition of secondary tumor were observed simultaneously under synergistic tumor treatment of nutritional-photothermal therapy (NT/PTT). Material repurposing could thus be confirmed by our pioneering trial and the CIP-ALG meditated NT/PTT/immunotherapy would provide new choice of strategy for safe and efficient tumor therapy.

The gut bacteria not only play a crucial role in maintaining human health but also exhibit close associations with the occurrence of numerous diseases. Understanding the physiological and pathological functions of gut bacteria and enabling early diagnosis of gut diseases heavily relies on accurate knowledge about their in vivo distribution. Consequently, there is a significant demand for noninvasive imaging techniques capable of providing real-time localization information regarding gut bacteria. In this work, we developed a second near-infrared (NIR-II) fluorescent nanoprobe labeling-based visualization strategy for real-time tracking of the biodistribution of Gram-negative bacteria in the gastrointestinal tract of mice. By utilizing positively charged silver sulfide quantum dots (Ag2S QDs) as NIR-II nanoprobes and exploiting electrostatic interactions to efficiently label the Gram-negative probiotic Escherichia coli Nissle 1917 with negative surface charges, we have achieved rapid and effective labeling. Leveraging the exceptional NIR-II fluorescent performance of Ag2S QDs, our approach enables high spatiotemporal resolution visualization via NIR-II imaging in mouse gastrointestinal areas where Ag2S QD-labeled probiotics are present, facilitating real-time in vivo tracking capabilities for these labeled probiotics. This work not only establishes a powerful intestinal bacterial imaging strategy but also introduces novel concepts for constructing nanomaterial-bacteria hybrid systems.

Second near-infrared (NIR-II) window responsive phototheranostic agents have a precise spatiotemporal potential for the diagnosis and treatment of cancer. In this study, a full-spectrum responsive ZrO2-based phototheranostic agent was found to achieve NIR-II photoacoustic (PA) imaging-guided tumour-targeting phototherapy. Initially, the ZrO2-based phototheranostic agent was fabricated through NaBH4 reduction to form boron-doped oxygen-deficient zirconia (ZrO2−x-B), an amino-functionalised SiO2 shell and a further covalent connection with hyaluronic acid (HA), namely, ZrO2−x-B@SiO2-HA. In the ZrO2−x-B@SiO2-HA system, the oxygen vacancy and boron doping resulted in full-spectrum absorption, enabling a high NIR-II photothermal conversion, high-resolution PA imaging ability and a remarkable production of reactive oxygen species (ROS). The surface modification of HA provided ZrO2−x-B@SiO2-HA with water dispersibility and a targeting capability for CD44 overexpressed cancer cells. Furthermore, in vitro and in vivo experiments showed that NIR-II activated ZrO2−x-B@SiO2-HA had a targeted photothermal/photodynamic effect for cancer elimination under the real-time guidance of NIR-II PAs. Hence, ZrO2−x-B@SiO2-HA displays a precise NIR-II radiation-activated phototheranostic potential for PA imaging-guided cancer-targeting photothermal/photodynamic therapy.

Rational design of tumor-microenvironment (TME)-activated nanoformulation for precisely targeted cancer treatment has recently attracted an enormous attention. However, the all-in-one TME-activated theranostic nanosystems with a simple preparation and high biocompatibility are still rarely reported. Herein, catalase nanocrystals (CatCry) are first introduced as a tumor microenvironment activatable nanoplatform for selective theranostics of colon cancer. They are engaged as (i) a “nanoreactor” for silver nanoparticles (AgNP) synthesis, (ii) a nanovehicle for tumor delivery of anticancer drug doxorubicin (DOX), and (iii) an in situ O2 generator to relief tumor hypoxia. When CatCry-AgNP-DOX nanoformulation is within a tumor, the intratumoral H2S turns AgNP into Ag2S nanoparticles, inducing a photothermal effect and NIR-II emission under 808 nm laser irradiation and also triggering DOX release. Simultaneously, CatCry catalyzes intratumoral H2O2 into O2, relieving hypoxia and enhancing chemotherapy. In contrast, when delivered to healthy tissue without increased concentration of H2S, the developed nanoformulation remains in the “off” state and no theranostic action takes place. Studies with colon cancer cells in vitro and a murine colon cancer model in vivo demonstrated that CatCry-AgNP-DOX delivered a synergistic combination of PTT and enhanced chemotherapy, enabling complete eradication of tumor with minimal side effects. This work not only introduces nanoplatform for theranostics of H2S-rich tumors but also suggests a general strategy for protein-crystal-based nanomedicine.

Biosensors working in the second near-infrared (NIR-II) spectral region have recently gained great attention owning to their deep tissue bioimaging and high spatiotemporal resolution . However, there is still a challenge to develop efficient nanothermometers based on NIR-II luminescence. Herein, we present a pioneering advancement in the field of nanothermometry by synthesizing a novel nanothermometer, a composite of fluoride nanocrystals and quantum dots . This NaYF 4 :1Nd-Ag 2 S QDs@SiO 2 nanocomposite demonstrates exceptional functionality, being excited by a single light irradiation at 808 nm for NIR-II luminescence imaging. Notably, the composite emits at two distinct wavelengths-1064 nm and 1125 nm, attributable to Nd 3+ and Ag 2 S QDs, respectively. This unique emission profile enables deeper tissue penetration, mitigating overheating risks compared to conventional excitation sources at 980 nm. Furthermore, the nanomaterials exhibit remarkable temperature-sensing capabilities, with maximum sensitivities of 0.81%K −1 (S R ) at 333 K. These superior sensitivities outperform many existing temperature sensors relying on thermally coupled Stark sublevels. The SiO 2 matrix not only encapsulates and protects the fluoride nanocrystals and quantum dots but also shields them from aqueous environments, ensuring outstanding luminescence stability. Our findings highlight the potential of this nanothermometer as a highly efficient temperature sensor with promising applications in cutting-edge bioapplications . This development introduces a new frontier in temperature sensing and bioimaging, showcasing its potential for transformative contributions to the field.

Colon cancer is one kind of malignant digestive tract tumor with high morbidity and mortality worldwide, treatments for which still face great challenges. Recently emerged intervention strategies such as phototherapy and gas therapy have displayed promising effects in the treatment of colon cancer, but their application are still hindered due to insufficient tumor targeting and deeper tissue penetrating capacity. Herein, in the present study, we developed one theranostic nanoplatform Cet-CDs-SNO (CCS) to realize multimodal imaging-guided synergistic colon cancer therapy. Among the CCS, Cetuximab (Cet), one first-line clinical drugs for colorectal cancer, endowed CCS with tumor-targeting capacity and enhanced drug accumulation in tumor cells; CDs doped by Ni2+ and Mn2+ served as NIR-II photothermal therapy (PTT), chemodynamic therapy (CDT), and photothermal/magnetic resonance/fluorescence imaging (PTI/MRI/FLI) agents; SNO, a nitric oxide (NO) donor, exerted gas therapeutic (GT) effects under thermal stimulation derived from PTT. In vitro and in vivo experiments proved that CCS had excellent colon cancer-targeting ability. Proliferation of colon cancer cells and tumor growth were significantly inhibited by the administration of CCS without detectable cytotoxicity. This study presented one strategy for developing a multifunctional nanoplatform to be applied in imaging-guided precise tumor therapy.

Near-infrared (NIR) luminescence capable of covering NIR-I and NIR-II is highly promising for biomedical imaging and non-invasive inspection. However, achieving both broadband and long-wavelength emission remains challenging due to the activator ion site-occupation constraints. In this work, we report La 3 TiGa 5 O 14 :Cr 3+ (LTG:Cr 3+ ) phosphor that exhibits an ultrawideband NIR emission spanning the spectral range of 650–1400 nm. The underlying mechanism involves Ti and Ga site exchange, which triggers local polyhedral reorganization. This structural transformation creates a weak crystal field, large Stokes shift, lower energy barrier of activator ion site occupancy, and more sites favorable for Cr 3+ incorporation ─ collectively relaxing Cr 3+ site-occupation constraints. The optimized LTG:0.03Cr 3+ phosphor exhibits NIR emission at 1017 nm with an exceptionally broad full width at half maximum (FWHM) of 476 nm. Moreover, introducing Ta 5+ as a charge-compensating dopant boosts NIR emission intensity by nearly 3.9-fold. A prototype NIR LED device fabricated with this phosphor demonstrates effective multi-channel signal detection. This work advances site-occupancy engineering from extrinsic doping to intrinsic polyhedral reorganization, providing a molecular design framework for high-performance NIR phosphor.
