Healing wounds presents a significant challenge due to bacterial biofilm infections and the inherent drug resistance of these biofilms. This report introduces a multifunctional nanoplatform (NPs) designed to combat wound biofilm infections using NIR-II photothermal therapy. The NPs are self-assembled from amphiphilic polymers (AP) to encapsulate photothermal polymers (PT) through classic electrostatic interactions. Importantly, these NPs are electrically neutral, which enhances their ability to penetrate biofilms effectively. Once inside the biofilm, the NPs achieve complete thermal ablation of the biofilm under NIR-II laser irradiation. Additionally, when exposed to laser and the GSH microenvironment, the NPs exhibit strong photothermal effects and self-degradation capabilities. In vitro tests confirm that the NPs have excellent antibacterial and anti-biofilm properties against methicillin-resistant Staphylococcus aureus (MRSA). In vivo studies demonstrate that the NPs can efficiently clear wound biofilm infections and promote wound healing. Notably, the NPs show superior photothermal effects under NIR-II laser irradiation compared to NIR-I lasers. In summary, the developed NPs serve as an integrated diagnostic and therapeutic nano-antimicrobial agent, offering promising applications for biofilm wound infections and wound healing.

Photoacoustic (PA) imaging with functional nanoprobes in the second near-infrared region (NIR-II, 1000–1700 nm) has aroused much interest due to its deep tissue penetration and high maximum laser permissible exposure. However, most NIR-II PA imaging is performed using the two-dimensional (2D) imaging modality, which impedes the comprehension of the in vivo biodistribution, angiography and molecular-targeted performance of NIR-II nanoprobes (NPs). Herein, we report the systematic monitoring of biomineralized copper sulfide (CuS) NPs, typical NIR-II NPs, in mouse models by employing NIR-II three-dimensional (3D) PA imaging. The advanced imaging modality provides dynamic information about the 3D biodistribution and metabolic pathway of CuS NPs. We also achieved contrast-enhanced 3D PA imaging of abdominal and cerebral vessels at a high signal-to-background ratio. Moreover, the tumor-targeted CuS NPs conjugated with the bombesin peptide endowed NIR-II 3D PA with superior performance in imaging orthotopic tumors both deep in the prostate and in the brain beneath the intact scalp and skull. Our results highlight the potential of NIR-II 3D PA imaging for the evaluation of the in vivo behavior of NPs, thus providing a promising strategy for screening NPs in clinical translational studies.

Accurate detection and localization of micrometastases are extremely important for tumor diagnosis and comprehensive treatment. However, it is clinically difficult to finely localize micrometastases, especially focal lesions smaller than 1 mm. To that purpose, this work designed a tumor microenvironment factor (glutathione, GSH)-activated second near-infrared transparency window (NIR-II) fluorescent probe for visualization of micrometastases. It is constructed by complexing copper ions with 3-mercaptopropionic acid-modified silver sulfide (Ag 2 S-MPA) quantum dots . The experimental results showed that the probe has good and stable GSH activation properties and can achieve tumor-specific NIR-II fluorescence imaging. Further, in vivo imaging experiments demonstrated that the probe can precisely localize melanomas with a diameter of < 0.1 mm in lung micrometastases. This work provides a potential strategy for the development of novel tools for localization and visualization of micrometastases.

Lanthanide-doped near-infrared-II (NIR-II) luminescent nanoparticles have shown great potential in many fields. However, most previous studies were focused on the sensitizer-activator coupled systems in which the NIR-II luminescence intensity is seriously restricted by the low dopants concentration . Improving the NIR-II emission intensity in lanthanide-doped nanoparticles has remained a daunting challenge. Herein, we report a systematic research on the enhancement of the NIR-II luminescence of Er 3+ through lanthanide-mediated energy modulation. It is found that introducing proper content of Yb 3+ , Ce 3+ , Tm 3+ , and Ho 3+ in the erbium sublattice can indeed enhance the NIR-II emission under 980 and 808 nm excitations by mediating the energy transfer processes. Doping of other lanthanide ions (e.g., Pr 3+ , Tb 3+ , Dy 3+ , and Sm 3+ ) only led to a quick decline in luminescence intensity. The NIR-II emission was further enhanced by designing a core-shell-shell nanostructure which consists of an NaYbF 4 interlayer to maximize the absorption of 980 nm excitation. The NIR-II luminescence exhibits superior penetration depth in biological tissue, showing great promise in biological applications .

The current strategy of co-delivering copper ions and disulfiram (DSF) to generate cytotoxic CuET faces limitations in achieving rapid and substantial CuET production, specifically in tumor lesions. To overcome this challenge, we introduce a novel burst-release cascade reactor composed of phase change materials (PCMs) encapsulating ultrasmall Cu 2-x Se nanoparticles (NPs) and DSF (DSF/Cu 2-x Se@PCM). Once triggered by second near-infrared (NIR-II) light irradiation, the reactor swiftly releases Cu 2-x Se NPs and DSF, enabling catalytic reactions that lead to the rapid and massive production of Cu 2-x Se-ET complexes, thereby achieving in situ chemotherapy. The mechanism of the burst reaction is due to the unique properties of ultrasmall Cu 2-x Se NPs, including their small size, multiple defects, and high surface activity. These characteristics allow DSF to be directly reduced and chelated on the surface defect sites of Cu 2-x Se, forming Cu 2-x Se-ET complexes without the need for copper ion release. Additionally, Cu 2-x Se-ET has demonstrated a similar (to CuET) anti-tumor activity through increased autophagy, but with even greater potency due to its unique two-dimensional-like structure. The light-triggered cascade of interlocking reactions, coupled with in situ explosive generation of tumor-suppressive substances mediated by the size and valence of Cu 2-x Se, presents a promising approach for the development of innovative nanoplatforms in the field of precise tumor chemotherapy.

Graphical Quinoid conjugated polymer PIS with broad absorption in the NIR-II region is developed based on multiple strategies. Benefiting from its remarkable photothermal conversion behavior under NIR-II laser (1064 nm), PIS nanoparticles show excellent antitumor performances in both in vitro and in vivo studies. This work presents a rational design strategy to explore NIR-II photothermal agents for hyperthermia therapy. Recently, extensive efforts have been devoted to the development of the second near-infrared bio-window (NIR-II, 1000–1700 nm) theranostic agents owing to the excellent tissue-penetration capability of NIR-II light. The exploration of organic NIR-II photothermal therapy materials, especially those with absorption peak over 1000 nm, is an appealing yet significantly challenging task. Herein, we have designed conjugated polymer nanoparticles (PIS NPs) with NIR-II absorption peak at 1026 nm through a combined strategy of introducing quinoid donor-acceptor (D–A) structures, constructing intramolecular “conformational locks” and extending the conjugation area to narrow the band gap. Irradiated at 1064 nm, PIS NPs showed remarkable photothermal conversion performance for efficient photothermal ablation of tumor cells in vitro and in vivo. This study provides useful insights into the rational design of organic NIR-II photothermal agents based on multiple strategies.

Conventional fluorescence imaging in the visible and first near-infrared (NIR-I, 700–900 nm) windows is limited by tissue scattering and autofluorescence. The second near-infrared (NIR-II, 1000–3000 nm) window offers deeper penetration depth and higher signal-to-noise ratio due to reduced photon scattering. Among the high-performance probes, atomically precise gold clusters emerge as a promising class of materials, as their defined structure can be engineered for enhanced NIR-II emission. Here, we report controllable Au 18 clusters for NIR-II bioimaging, which exhibit superior brightness, stability, and biocompatibility. We successfully dope a single Zn atom into the Au 18 cluster structure, thereby optimizing optical properties and improving photostability. Zn atom doping shifts the energy levels downward by approximately 1.2 eV and induces local charge redistribution, resulting in enhanced photoluminescence. The Au 17 Zn 1 clusters exhibit a 4.1-fold NIR-II fluorescence enhancement and high temporal stability with excellent biological safety. Furthermore, Au 17 Zn 1 shows potential for visualizing liver tissue in mice with hepatic ischemia-reperfusion injury (HIRI). In addition, three-dimensional (3D) imaging of HIRI mice using light-sheet microscopy (LSM) reveals vascular dilation from approximately 120 to 300 µm, clearly delineating the different stages of HIRI. Therefore, Au 17 Zn 1 shows potential as a tool for HIRI assessment.

Successful surgical resection of solid tumours requires highly reliable real-time intraoperative tools to accurately delineate tumour boundaries, which remains challenging in routine clinical standards. Here, we identify endogenous substances with intense autofluorescence in the second near-infrared window (NIR-II, 1,000–1,700 nm) that are abundant in human liver tissues but negligible in cancerous tissues. Inspired by this discovery, we develop a label-free and wide-field imaging approach, named tissue autofluorescence NIR-II imaging (TANI) for visualizing human liver malignancies. TANI demonstrates exceptional contrast (7.69 ± 0.52), sensitivity (97.8%) and specificity (98.4%) in delineating various liver malignancies, including hepatocellular carcinoma, intrahepatic cholangiocarcinoma and liver metastasis from cirrhotic or non-cirrhotic livers, outperforming routine fluorescence-guided surgery and conventional autofluorescence imaging in the visible (400–650 nm) or first near-infrared (700–900 nm) window. The excellent performance of TANI remains unaffected by cancer grade/stage, benign lesions or blood/bile contamination. These findings represent a promising advance in intraoperative decision-making and suggest a strong correlation between near-infrared autofluorescence and diseases. We believe that clarifying the molecular insights underlying these autofluorescent substances may provide new diagnostic directions.

Achieving phosphor-converted light-emitting diodes (pc-LEDs) for portable NIR spectroscopy applications has evoked enormous demand for high-efficiency broadband NIR-II (1,000–1,700 nm) phosphors. Herein, a series of Li-alloyed (Li/Mg)O:Ni 2+ phosphors with tunable NIR-II emission is prepared. By alloying Li-ions into MgO:Ni 2+ , we break the uniform coordination environment of Ni 2+ emitters by substitutional disorder and induce many crystallographically equivalent Ni 2+ emitters. This leads to large spectral tuning in the NIR-II region with the peak position and full-width at half-maximum (FWHM) varying in the 1,330–1,460 and 204–350 nm regions, respectively. Consequently, ultra-broadband (FWHM = 350 nm, 10.1 mW at 350 mA) and high-efficiency (FWHM = 204 nm, 19.7 mW at 350 mA) NIR-II pc-LEDs have been fabricated simultaneously based on the explored phosphors, and its applications are also demonstrated. We anticipate that this strategy can be used to explore other NIR phosphor systems, thus opening up new avenues for next-generation NIR spectroscopy technology.

All-inorganic lead-free metal halide (MH) nanocrystals (NCs) have shown great promise for various optoelectronic applications; however, achieving efficient luminescence in the near-infrared-II (NIR-II) region remains a persistent challenge. Herein, we developed a new class of NIR-II-emitting materials based on Sb 3+ /Ln 3+ (Ln = Nd, Yb, Er) co-doped 0D Rb 3 InCl 6 NCs. By leveraging the spin-orbit allowed 1 S 0 → 3 P 1 transition of Sb 3+ as a sensitizer, we achieve intense multi-wavelength NIR-II emission arising from the 4f → 4f transitions of Nd 3+ , Yb 3+ , and Er 3+ under single-wavelength near-ultraviolet excitation, with a notable quantum yield of 7.1% for Er 3+ emission. The resulting NCs also demonstrate excellent ambient stability, enabling their use as visible/NIR dual emitters for applications such as bioimaging and night vision. This work provides a versatile co-doping strategy involving s- and f-electron ions to realize efficient NIR-II luminescence in lead-free MH NCs.

Background: Triple-negative breast cancer (TNBC) is a highly aggressive subtype of breast cancer characterized by significant treatment challenges and poor prognosis. Traditional therapies such as surgical resection, radiotherapy, and chemotherapy often suffer from limitations including insufficient therapeutic specificity, potential damage to normal tissues, and limited imaging capability. The integrated diag{Pareja, 2018 #13}nosis-treatment platform combines diagnostic and therapeutic functions, offering a novel approach for real-time tumor monitoring and precision therapy. Methods: Accordingly, this study designed and developed an integrated diagnostic and therapeutic platform called A2S-TT NPs, utilizing self-assembled nanoparticles incorporating organic conjugated molecules for precise photodynamic therapy (PDT) guided by dual modalities of Near-infrared II (NIR-II) fluorescence imaging and photoacoustic imaging (PA).Results: Under 808 nm laser excitation, A2S-TT NPs efficiently generated reactive oxygen species (ROS) at the tumor site, inducing mitochondrial membrane potential disruption and DNA damage to activate apoptosis. In vitro experiments demonstrated that A2S-TT NPs exhibited intense NIR-II fluorescence and photoacoustic signals, showcasing excellent dual-modal imaging potential. Moreover, it demonstrated minimal cytotoxicity in the absence of light exposure but significantly enhanced tumor cell killing under laser irradiation, demonstrating superior biosafety and photodynamic efficacy. In vivo studies further confirmed that A2S-TT NPs effectively accumulated at tumor sites, enabling visualization-based monitoring through its mediated NIR-II and PA dual-modal imaging. Additionally, A2S-TT NPs-mediated PDT exhibited remarkable tumor-suppressive effects while avoiding significant damage to normal tissues.Discussion: This study constructs a novel PA/NIR-II dual-modal imaging-guided PDT platform based on donor–acceptor conjugated polymer A2S-TT NPs for integrated TNBC theranostics. PA provides high-resolution deep-tissue imaging, while NIR-II fluorescence offers high sensitivity and deep penetration; their complementary strengths support precise tumor localization and real-time therapeutic monitoring. Under dual-modal guidance, A2S-TT NPs enrich in tumors and generate abundant ROS upon 808 nm laser irradiation to boost PDT efficacy. In vitro and in vivo biosafety tests confirm its favorable biocompatibility with minimal systemic toxicity at therapeutic doses. Conclusions: In conclusion, A2S-TT NPs achieved precise, efficient, safe, and controllable PDT through dual-mode imaging using NIR-II and PA, providing a novel strategy with clinical translation potential for the integrated diagnosis and treatment of TNBC.

The second near-infrared window (NIR-II, 1000–2000 nm) provides an ideal window for multichannel photoluminescence lifetime imaging (mPLI) because of its exceptional tissue penetration depth and minimal scattering. However, the inevitable spectral crosstalk that is caused by the broad and overlapping emissions of traditional luminescent materials (e.g., organic dyes and quantum dots) has restricted the scalability of mPLI channels. Herein, we developed a series of Ln 3+ -doped (Ln = Nd, Er, Tm, or Ho) stratified nanoparticles (Ln-SNPs) that enable highly efficient multiwavelength NIR-II luminescence. These nanoparticles can be further engineered into concentration-mediated Yb 3+ energy relay nanoparticles to enable the precise tuning of the luminescence lifetime across the 1060–2050 nm range, thereby expanding the NIR-II imaging channels in mPLI. Notably, the engineered Tm-SNPs exhibit optimized downshifting emission at ∼1850 nm (NIR-IIc subwindow) with distinguishable luminescence lifetimes that span three orders of magnitude at similar emission intensities. This advancement establishes a powerful toolkit for multidimensional information encryption and multiplexed bioimaging applications.
