PLLA–gelatin composite fiber membranes incorporated with functionalized CeNPs as a sustainable wound dressing substitute promoting skin regeneration and scar remodeling

Yarong Lv,Yulong Xu,Xinyu Sang,Chenxi Li,Yong Liu,Quanyi Guo,Seeram Ramakrishna,Ce Wang,Ping Hu,Himansu Sekhar Nanda

The need for a wound dressing material that can accelerate wound healing is increasing and will last for a long time. In this study, cerium oxide nanoparticle (CeNP) incorporated poly-L-lactic acid (PLLA)–gelatin composite fiber membranes were fabricated using established electrospinning techniques for use as a low-cost sustainable wound dressing material. The obtained membranes were characterized for their morphology, and physical, mechanical and biological properties. The results showed that the membranes maintained an integrated morphology, and demonstrated water absorption and improved mechanical properties. An in vitro cell proliferation test confirmed that the cells presented better activities over the composite fiber membranes. In the rat scalding model, rapid wound recombination was observed. All these data suggested that electrospun CeNP incorporated PLLA–gelatin composite fiber membranes can be an ideal dressing substitute that can be used for wound healing applications. Furthermore, the use of biodegradable polymers and environmentally sustainable production technologies presented better sustainability for the commercial production of these composite membranes promoting tissue regeneration and scar remodeling.

siAkt2-Loaded Nanoparticles Reprogramming Macrophages to M2 Phenotype for Effective Bone Defect Repair

Shengjie Cui,Xiaowei Wu,Xiaotong Yu,Yiqian Huang,Daixing Zhang,Haotian Chen,Boon Chin Heng,Qing Cai,Yixiang Wang,Yan Gu,Xuliang Deng

Targeted siRNA delivery has been widely used to regulate cell function. However, the rapid degradation of siRNA within macrophages upon delivery severely limits its utility for regulating macrophage function. Here, a novel technique is developed to compact large amounts of siAkt2 strands into delivery nanoparticles, to achieve massive and sustained intracellular release of siAkt2 that overwhelms the degradation capacity of macrophages. Microspheres of RNA complexes are prepared by rolling circle transcription. For better deliverability, siAkt2 microspheres are compacted by cholesterol-modified DNA, and encapsulated by Poly-(L-lactic acid) (PLLA). These PLLA-encapsulated siAkt2-compacted nanoparticles (siAkt2 RNP@PLLA NPs), each containing multiple repeated siAkt2 sequences, displayed long-term stability and maintenance of integrity within body fluids and acidic environments. Extensive endocytosis by RAW 264.7 is followed by continuous massive siAkt2 release into the cytosol, which significantly increased Akt2 gene silencing, metabolic reprogramming, as well as enhanced macrophage alternative polarization to the pro-regenerative M2 phenotype. Local administration of siAkt2 RNP@PLLA NPs in bone defects caused by periodontitis markedly induced macrophage alternative M2 polarization and enhanced bone regeneration. Therefore, the study provides a novel and highly efficient siRNA delivery system for regulating macrophage function, which can be a promising therapeutic strategy for enhancing tissue regeneration.

Tumor microenvironment-responsive thermoelectric scaffold for on-demand antitumor therapy

Xiuwen Gao,Woding Deng,Jinhui Tan,Xiong Shuai,Jun Zan,Tailai Ye,Kaiwu Luo,Fangwei Qi,Yujun Wei,Cijun Shuai

Thermoelectric material could realize the thermal-electricity-reactive oxygen species (ROS) conversion under temperature gradient, being considered a promising candidate for tumor therapy. Nevertheless, it required exogenous excitation such as light irradiation, causing difficultity in achieving on-demand therapy. Herein, a tumor microenvironment responsive Bi 2 Te 3 –CaO 2 nanohybrid was synthetized by in situ growing calcium peroxide (CaO 2 ) nanoparticles on thermoelectric bismuth telluride (Bi 2 Te 3 ) nanosheets, and then blended with poly- l -lactic acid (PLLA) to fabricate a PLLA/Bi 2 Te 3 –CaO 2 scaffold. Once the acidic microenvironment of the tumor was sensed, the CaO 2 would serve as an intelligent switch, releasing heat through rapid hydrolysis and triggering the thermoelectrocatalysis of Bi 2 Te 3 to generate ROS, thereby achieving on-demand therapy. Results demonstrated that the scaffold exhibited rapid temperature rise and significant ROS catalytic formation ability in an acidic solution (pH 5.5), while no significant changes occurred in a neutral environment (pH 7.4). Annexin V-FITC/PI staining and JC-1 assay demonstrated that the generated ROS could induce tumor apoptosis by reducing mitochondrial membrane potential. Besides, the fluorescent probe test revealed that CaO 2 could also produce hydrogen peroxide (H 2 O 2 ) and calcium ions (Ca 2+ ) to interfere with intracellular Ca 2+ , thereby further enhancing the antitumor effect. Overall, this work proposed a novel perspective for on-demand antitumor therapy.

Synthesis and characterization of bovine serum albumin-loaded microspheres based on star-shaped PLLA with a xylitol core and their drug release behaviors

Ding Amin,Teng Lijing,Zhou Yifeng,Chen Pengpeng,Nie Wangyan

The star-shaped poly( l -lactide) (s-PLLA) was synthesized via ring-opening polymerization of l -lactide, with xylitol as a multifunctional initiator and stannous octoate as a catalyst. The structures of s-PLLA were characterized by 1 H NMR spectroscopy, while the molecular weight ( M w ) and polydispersity index were determined by gel permeation chromatography. Bovine serum albumin (BSA) loaded s-PLLA microspheres were fabricated by a water–oil–water ( W 1 / O / W 2 ) double-emulsion solvent evaporation method. The morphology, drug encapsulation efficiency (EE), and in vitro release behavior of the prepared microspheres were studied in detail. Results indicated that the average diameters of s-PLLA microspheres could be controlled between 7 and 15 µm by varying the s-PLLA’s concentration or M w , and the drug encapsulation efficiency of BSA-loaded microspheres were 10–42%. The in vitro study shows that the BSA-loaded microspheres prepared by high molecular weight s-PLLA had a slow cumulative release behavior in phosphate buffer saline (PBS, pH 7.4) and the release process followed the Baker–Lonsdale equation. Present work supposes that the synthesized s-PLLA is a good candidate for drug delivery.

COF-reinforced tunable-degradation 5-FU@COF/PLLA-TPU coating with dual anti-sludge and anti- tumor hyperplasia for biliary stent restenosis prevention

Mingsheng Wang,Yunjie Chen,Yanghui Wen,Xiaomei Liu,Chang He,Yi Liu,Minghui Pan,Chao Pang,Hua Li

To overcome biliary stent failures caused by biofilm formation, bile sludge accumulation, and tumor-mediated restenosis, we designed a self-polishing, biodegradable coating by flame-spraying poly(L-lactic acid)/thermoplastic polyurethane (PLLA/TPU) composites incorporated with 5-fluorouracil-loaded covalent organic frameworks (5-FU@COF). This strategy integrates: 1) Programmable self-polishing via synergistic PLLA ester/urethane bond hydrolysis and pH-sensitive COF delamination, ensuring controlled erosion; 2) Mechanoadaptive reinforcement, where 5-FU@COF enhances coating-substrate adhesion by 111% (10FCLU: 3.19 MPa vs. LU:1.51 MPa) through COFs rigid filler anchoring and stress homogenization; and 3) Dual anti-biliary sludge accumulation/tumoricidal therapy enabled by 216.5 nm COF nanospheres achieving 43.1% 5-FU loading. The coatings resist biofouling via electrostatic repulsion and dynamic polishing, reducing bile salt coverage to 11.53% (vs. 47.44% on bare Ti). Biphasic 5-FU release from 10FCLU coating achieves 92.6% tumor Cholangiocarcinoma cell (RBE) inhibition for 3 days, validated by TEM ultrastructural apoptosis markers (nuclear fragmentation; mitochondrial collapse) and Korsmeyer-Peppas release kinetics. This multifunctional drug-loading coating enables next-generation biliary stents with sustained patency.

Synthesis of curcumin and indocyanine green co-loaded PLLA microparticles via solution-enhanced dispersion using supercritical CO2 for chemo-photothermal therapy of osteosarcoma

Yao Xiao,Zhongqing Wu,Zhiyuan Meng,Yongchao Wang,Zehao Li,Zheng Zhao

The therapy of tumor recurrence resulting from surgical resection of osteosarcoma is still a significant challenge for clinicians. In this study, we designed curcumin (CM) and indocyanine green (ICG) co-loaded poly(L-lactic acid) (PLLA) (PLLA-CM-ICG) composite microparticles , combining photothermal therapy with chemotherapy to enhance osteosarcoma therapy synergistically. PLLA-CM-ICG microparticles of sizes of (838.3 ± 5.4) nm were prepared by solution-enhanced dispersion by a supercritical CO 2 process. ICG in the PLLA-CM-ICG microparticles was more stable than free ICG. The PLLA-CM-ICG microparticles displayed pH- and near-infrared (NIR)-responsive CM release behavior. Moreover, NIR irradiation improved the anticancer activity of the PLLA-CM-ICG microparticles at a concentration of 250 μg/mL on MG-63 cells. Therefore, PLLA-CM-ICG microparticles could be employed as promising drug delivery systems for synergic photothermo-chemotherapy in osteosarcoma treatment.

Phospholipid-grafted PLLA electrospun micro/nanofibers immobilized with small extracellular vesicles from rat adipose mesenchymal stem cells promote wound healing in diabetic rats

Li Jing,Yan Shunshun,Han Weiju,Dong Zixuan,Li Junliang,Wu Qi,Fu Xiaoling

Small extracellular vesicles (sEVs) derived from mesenchymal stem cells (MSCs) can deliver a variety of bioactive factors to create a favorable local microenvironment, thereby holding huge potential in chronic wound repair. However, free sEVs administrated intravenously or locally are usually cleared rapidly, resulting in an insufficient duration of the efficacy. Thus, strategies that enable optimized retention and release profiles of sEVs at wound sites are desirable. Herein, we fabricated novel functional phosphoethanolamine phospholipid-grafted poly-l-lactic acid micro/nanofibers (DSPE-PLLA) to carry and retain sEVs from rat adipose MSCs, enabling the slow local release of sEVs. Our results showed that sEVs@DSPE-PLLA promoted the proliferation, migration and gene expression (Col I, Col III, TGF-β, α-SMA, HIF-1α) of fibroblasts. It also promoted keratinocyte proliferation. In addition, sEVs@DSPE-PLLA helped polarize macrophages toward the M2 phenotype by increasing the expression of anti-inflammatory genes (Arginase 1, CD 206, IL-10) and inhibiting the expression of pro-inflammatory genes (IL-1β, TNF-α). Further in vivo study in diabetic rat models showed that sEVs@DSPE-PLLA improved the wound-healing process by alleviating the inflammatory responses, stimulating cell proliferation, collagen deposition and angiogenesis. These results highlight the potential of using DSPE-grafted scaffolds for extracellular vesicle immobilization and suggest sEVs@DSPE-PLLA micro/nanofibers as promising functional wound dressings for diabetic wounds.

Cobalt-based metal–organic framework as a dual cooperative controllable release system for accelerating diabetic wound healing

Li Jiankai,Lv Fang,Li Jinxiu,Li Yuxin,Gao Jingduo,Luo Jian,Xue Feng,Ke Qinfei,Xu He

Insufficient angiogenesis in the chronic wound of the diabetic is one of the most important causes that making the wound unable to heal itself. In this work, a cobalt-based metal–organic framework (ZIF-67) was introduced as a carrier for loading a pro-angiogenic small molecular drug (dimethyloxalylglycine, DMOG). To achieve a long-term angiogenic therapy on the diabetic wound beds, a dual cooperative controllable release system has been designed by incorporating the drug-loaded ZIF-67 nanoparticles into the micro-patterned PLLA/Gelatin nanofibrous scaffolds. The results showed that DMOG was incorporated into ZIF-67 with a high loading ratio (359.12 mg/g), and the drug-loaded ZIF-67 nanoparticles were well embedded in the circular patterned scaffold. Notably, the DMOG as well as Co ions could continuously release from the scaffold for more than 15 days. The in vitro studies showed that the released Co ions and DMOG from the micropatterned nanofibrous scaffolds could synergistically promote the proliferation, migration and tube formation of the human umbilical vein endothelial cells (HUVECs) by inducing a hypoxia response and upregulating the expression of angiogenesis-related genes such as HIF-1α, VEGF and e-NOS. Furthermore, the in vivo results demonstrated that the composite scaffolds could significantly enhance angiogenesis, collagen deposition and eliminate inflammation in the diabetes wounds. These results indicate that the cobalt-based metal–organic framework as a dual cooperative controllable release system provides a new strategy for enhancing angiogenesis and promoting diabetic wound healing.

Micro- and nano-environment dual-modulated anti-tendon adhesion barrier membranes

Qiang Zhang,Kui Ma,Chun-Hei Lam,Ho-Pan Bei,Yu Liu,Xing Yang,Xin Zhao

Despite promise in preventing peritendinous adhesion, electrospun membranes face many challenges related to their complex fabrication process, untargeted/uncontrolled drug delivery and consequently low therapeutical effect. Here, a micro-and nano-environment dual-modulated barrier membrane (MNBM) with on-demand gene delivery capability is presented. Our MNBM is developed by first preparing extracellular signal-regulated kinase-2 (ERK2) siRNA-loaded gelatin methacryloyl (GelMA) nanogels via facile nano-emulsification technique, then incorporating these nanogels into poly-L-lactic acid (PLLA) fibers via simple blending electrospinning. The GelMA nanogels offer a nano-niche for ERK2-siRNA encapsulation and allow for a nano-environment controlled siRNA release by readily tuning the GelMA concentrations during nano-emulsification, while the resultant MNBM can mediate a micro-environment controlled siRNA delivery in response to the matrix metalloproteinase-2 (MMP-2) enriched micro-environment at the tendon repair site. Such MNBM can not only biologically orchestrate fibroblast behaviors by silencing the target gene expression, but also physically shield the tendon from extrinsic cell/tissue invasion. This study provides a proof-of-concept of anti-adhesion barrier membrane as an intelligent gene delivery system to offer a spatiotemporal and biophysical dual control over tendon recovery according to disease state and ensure long-term therapeutic efficacy. We envision such MNBM represents a promising therapeutic platform with great efficacy to achieve adhesion-free tendon repair.

An electrical microenvironment constructed based on electromagnetic induction stimulates neural differentiation

Fangwei Qi,Ruobing Liao,Ping Wu,Huixing Li,Jun Zan,Shuping Peng,Cijun Shuai

Electrical stimulation is considered an effective way to accelerate peripheral nerve cell growth. However, reliance on external wires and a power supply restricts its further applications. In this work, a wireless magnet-powered electrical stimulation system was constructed based on the electromagnetic induction effect. Specifically, amino-functionalized conductive MXene nanosheets were first wrapped on the PLLA particles’ surface by electrostatic assembly. Then the conductive NH2-MXene/PLLA conduit was fabricated by laser additive manufacturing. Triggered by a rotating magnetic field, the conduit could act as a coil to cut magnetic induction lines, converting magnetic energy into electrical energy and thereby achieving wireless electrical stimulation. The results indicate that the conduit presented an excellent conductivity of 8.44 S m−1, benefiting from its formed conductive network structure. Under excitation of the rotating magnetic field, the conduit generated an electric current of 10 μA, which is in the appropriate range for nerve cell growth. An in vitro cell test confirmed that the generated current effectively enhanced the PC12 cell proliferation, neurite growth and differentiation-related mRNA (Nestin, MAP2 and Tuj1) expression. Moreover, it also promoted PC12 cells to differentiate into mature neurons, evidenced by calcium sparks produced in PC12-derived neurons. Collectively, this wireless electrical stimulation system offers a new perspective on peripheral nerve repair.

Enhancing bone scaffold interfacial reinforcement through in situ growth of metal–organic frameworks (MOFs) on strontium carbonate: Achieving high strength and osteoimmunomodulation

Guowen Qian,Yuqian Mao,Yang Shuai,Zhikui Zeng,Shuping Peng,Cijun Shuai

Bioceramics have been extensively used to improve osteogenesis of polymers because of their excellent bone-forming capabilities. However, the inadequate interfacial bonding between ceramics and polymers compromises their mechanical properties . In this study, zeolitic imidazolate framework-8 (ZIF-8) was grown in situ on strontium carbonate (SrCO 3 ) to construct a core–shell SrCO 3 @ZIF-8, which was then added to poly- l -lactic acid (PLLA) to print a SrCO 3 @ZIF-8/PLLA composite scaffold using selective sintering technology. First, ZIF-8 characterized by its multiple organic ligands, forms a robust interface with PLLA. Second, SrCO 3 characterized by its negative zeta potential in solution, exhibits the ability to adsorb positively charged zinc ions . This, in turn, promotes the in situ growth of ZIF-8 on SrCO 3 , eventually achieving perfect bonding between the second phase and the PLLA matrix. Our findings indicated that the composite scaffold exhibited the highest compressive strength (21.93 MPa) and significantly promoted the osteogenic differentiation of mouse mesenchymal stem cells. Moreover, the in vivo results established that the SrCO 3 @ZIF-8/PLLA scaffold significantly accelerated bone regeneration efficiency in rat femur defects. The prepared scaffold, with its favorable mechanical properties and osteogenic activity, shows considerable promise for applications in bone repair.

Fabrication, Crystalline Behavior, Mechanical Property and In-Vivo Degradation of Poly(l–lactide) (PLLA)–Magnesium Oxide Whiskers (MgO) Nano Composites Prepared by In-Situ Polymerization

Hui Liang,Yun Zhao,Jinjun Yang,Xiao Li,Xiaoxian Yang,Yesudass Sasikumar,Zhiyu Zhou,Minfang Chen

The present work focuses on the preparation of poly(l–lactide)–magnesium oxide whiskers (PLLA–MgO) composites by the in-situ polymerization method for bone repair and implant. PLLA–MgO composites were evaluated using Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), differential scanning calorimetry (DSC), scanning electron microscopy (SEM) and solid-state13C and1H nuclear magnetic resonance spectroscopy (NMR). It was found that the whiskers were uniformly dispersed in the PLLA matrix through the interfacial interaction bonding between PLLA and MgO; thereby, the MgO whisker was found to be well-distributed in the PLLA matrix, and biocomposites with excellent interface bonding were produced. Notably, the MgO whisker has an effect on the crystallization behavior and mechanical properties; moreover, the in vivo degradation of PLLA–MgO composites could also be adjusted by MgO. These results show that the whisker content of 0.5 wt % and 1.0 wt % exhibited a prominent nucleation effect for the PLLA matrix, and specifically 1.0 wt % MgO was found to benefit the enhanced mechanical properties greatly. In addition, the improvement of the degrading process of the composite illustrated that the MgO whisker can effectively regulate the degradation of the PLLA matrix as well as raise its bioactivity. Hence, these results demonstrated the promising application of PLLA–MgO composite to serve as a biomedical material for bone-related repair. Keywords:PLLA–MgO whiskers composites;in-situ polymerization;bone repair and fixation;mechanical properties and crystallization behavior;regulating in-vivo degradation
