K2SiF6:Mn4+ red-luminescent crystals with high external quantum efficiency (EQEmax of 78%) and high thermal quenching temperature (T1/2 > 500 K) enabling high brightness warm white LEDs

Wenrui Zhang,Yayun Zhou,Ce Shi,Jiajun Ren,Liying Zhang,Haipeng Ji

Mn4+-activated fluoride phosphors are some of the crucial red-emitting phosphors for white lighting systems driven by light-emitting diodes (LEDs). In this study, we successfully synthesized high-quality K2Si1−xF6:xMn4+ (nominal x = 0.03–0.70, measured x of 0.22%–33.3%) crystals, measuring approximately 1 mm in size, through a new cooling-induced crystallization method. These crystals exhibit narrow line-type photoluminescence emission bands and exceptional optical properties, including a high absorption efficiency of up to 92.9% and an external quantum efficiency of up to 78.5%, which are among the record values for Mn4+-doped fluoride phosphors. Furthermore, they exhibit a notably elevated luminescence thermal quenching temperature (T1/2) exceeding 500 K. A comprehensive investigation into the phase transformation, zero-phonon line tuning, and fluorescence decay behavior as a function of increasing Mn concentrations was also conducted. Practical applications were demonstrated through the fabrication of warm white LEDs, integrating a blue LED with a garnet yellow phosphor and the as-synthesized K2Si1−xF6:xMn4+ (measured x of 2.94%) crystals as the red component. The white LEDs exhibit an impressive luminous efficacy of 192.1 lm W−1, a color rendering index of Ra = 87.5 and R9 = 62 under a 20 mA driving current. Our study highlights the outstanding properties of Mn4+-doped fluoride crystals with bulk size rather than conventional powdery form and the potential of the cooling-induced crystallization method for cultivating the crystals.

Cong Tian,Zhe Tang,Yike Hou,Asim Mushtaq,Shafaq Naz,Zhangsen Yu,Jabeen Farheen,Muhammad Zubair Iqbal,Xiangdong Kong

Significant attention is paid to the design of magnetoplasmonic nanohybrids, which exploit synergistic properties for biomedical applications. Here, a facile method was employed to prepare plasmonic magnetic Au-MnO heterostructured hybrid nanoparticles for imaging-guided photothermal therapy of cancers in vitro, with the view to reducing the serious drawbacks of chemotherapy and gadolinium-based contrast agents. The biocompatibility of the prepared Au-MnO nanocomposites was further enhanced by Food and Drug Administration (FDA)-approved triblock copolymers Pluronic®F-127 and chitosan oligosaccharide (COS), with complementary support to enhance the absorption in the near-infrared (NIR) region. In addition, synthesized[email protected]nanocomposites exhibited promising contrast enhancement in T1MR imaging with a good r1relaxivity value (1.2 mM−1s−1), demonstrating a capable substitute to Gd-based toxic contrast agents. In addition, prepared[email protected]hybrid nanoparticles (HNPs) produced sufficient heat (62 °C at 200 μg/mL) to ablate cancerous cells upon 808 nm laser irradiation, inducing cell toxicity, and apoptosis. The promising diagnostic and photothermal therapeutic performance demonstrated the appropriateness of the[email protected]HNPs as a potential theranostic agent. Keywords:plasmonic nanoparticles;magnetic nanocomposites;T Magnetic contrast agents;photothermal therapy;cancer theranostics

Sialic acid-engineered mesoporous polydopamine nanoparticles loaded with SPIO and Fe3+ as a novel theranostic agent for T1/T2 dual-mode MRI-guided combined chemo-photothermal treatment of hepatic cancer

Gaofeng Shu,Minjiang Chen,Jingjing Song,Xiaoling Xu,Chenying Lu,Yuyin Du,Min Xu,Zhongwei Zhao,Minxia Zhu,Kai Fan,Xiaoxi Fan,Shiji Fang,Bufu Tang,Yiyang Dai,Yongzhong Du,Jiansong Ji

Hepatic cancer is a serious disease with high morbidity and mortality. Theranostic agents with effective diagnostic and therapeutic capability are highly needed for the treatment of hepatic cancer. Herein, we aimed to develop a novel mesoporous polydopamine (MPDA)-based theranostic agent for T1/T2 dual magnetic resonance imaging (MRI)-guided cancer chemo-photothermal therapy. Superparamagnetic iron oxide (SPIO)-loaded MPDA NPs ( [email protected] ) was firstly prepared, followed by modifying with a targeted molecule of sialic acid (SA) and chelating with Fe 3+ ( [email protected] /Fe 3+ NPs). After that, doxorubicin (DOX)-loaded [email protected] /Fe 3+ NPs ( [email protected] /DOX/Fe 3+ ) was prepared for tumor theranostics. The prepared [email protected] /Fe 3+ NPs were water-dispersible and biocompatible as evidenced by MTT assay. In vitro photothermal and relaxivity property suggested that the novel theranostic agent possessed excellent photothermal conversion capability and photostability, with relaxivity of being r 1  = 4.29 mM −1 s −1 and r 2  = 105.53 mM −1 s −1 , respectively. [email protected] /Fe 3+ NPs could effectively encapsulate the DOX, showing dual pH- and thermal-triggered drug release behavior. In vitro and in vivo studies revealed that [email protected] /DOX/Fe 3+ NPs could effectively target to the hepatic tumor tissue, which was possibly due to the specific interaction between SA and the overexpressed E-selectin. This behavior also endowed [email protected] /DOX/Fe 3+ NPs with a more precise T1-T2 dual mode contrast imaging effect than the one without SA modification. In addition, [email protected] /DOX/Fe 3+ NPs displayed a superior therapeutic effect, which was due to its active targeting ability and combined effects of chemotherapy and photothermal therapy. These results demonstrated that [email protected] /DOX/Fe 3+ NPs is an effective targeted nanoplatform for tumor theranostics, having potential value in the effective treatment of hepatic cancer.

Dimeric PD-L1 specific affibody grafted Fe-based nanosheets for tumor-targeting dual-mode magnet resonance imaging and enhancement of Photothermal-Immunotherapy

Xinyu Zhang,Hui Liu,Chenyu Wang,Yuan Gao,Shiqi Zhao,Mengyao Mu,Ke Ren,Yufang Gong,Qing Fan,Xiao Sun

Biodegradable Two-dimensional (2D) nanomaterials have attracted tremendous interest in biomedical application. Herein, a biodegradable ferrous sulfide nanosheet with ultrathin structure and uniform size was developed. Following its development, the application of the self-enhanced photothermal nanoplatform (FMPN) in vivo by combining Mn-DOTA and dimeric PD-L1 affibody for targeted dual-mode magnetic resonance imaging (MRI) guided synergistic tumor therapy is explored. Dual-contrast enhanced subtraction imaging (DESI) was used to evaluate the excellent T1-T2 MRI ability of FMPN. FMPN displayed excellent photothermal conversion efficiency (η = 40.33 %) and biodegradability, thus had good tumor photothermal therapy effects with good biosafety. In addition, FMPN with pH-responsive Fe 2+ release can effectively induce tumor cell ferroptosis, further intensifying PTT and subsequent immunologic cell death. PD-L1 affibody could endow this nanoplatform with efficient tumor targeting, terminating T cells’ immune suppression by effectively blocking the interaction between PD-1 and PD-L1. Systemic delivery of FMPN significantly inhibited tumor progression in primary, metastatic and abscopal tumors, and showed obvious dual-mode MRI enhancement. Therefore, this nanoplatform exhibited promising potential for MRI-guided synergistic ferroptosis/photothermal/immunotherapy.

Tumor-targetable magnetoluminescent silica nanoparticles for bimodal time-gated luminescence/magnetic resonance imaging of cancer cells in vitro and in vivo

Bo Song,Qi Liu,Hua Ma,Zhixin Tang,Chaolong Liu,Jinhua Zou,Mingqian Tan,Jingli Yuan

Bimodal photoluminescence-magnetic resonance (MR) imaging technique has attracted tremendous attention due to its great potential in biomedical researches and clinical practices. Herein a novel multifunctional magnetoluminescent nanocomposite, FA-Gd-Tb@SiO 2 , was found to serve as an effective probe for bimodal time-gated luminescence/MR imaging of cancer cells in vitro and in vivo . The nanoprobe was designed by integrating a luminescent Tb 3+ complex, a Gd(III)-based contrast agent and a tumor-targeting molecule, folic acid (FA), into a silica nanoparticle . The integration of these functional moieties allows the nanoprobe to be employed for specific imaging of cancer cells with background-free TGL and non-invasive MR imaging modes. In addition, the optical and magnetic properties were dramatically improved after implicating the newly synthesized nanoarchitecture. In vitro cellular TGL imaging demonstrated that the FA-Gd-Tb@SiO 2 nanoprobe could recognize and accumulate in cancer cells overexpressing FA receptor. Furthermore, in vivo study revealed that the as-prepared nanoprobe was able to effectively enhance T 1 -weighted MR contrast and TGL intensity in tumor tissue, which might contribute to the precise detection and tracing of cancer cells, as well as diagnosis and therapy of tumor in clinical.

Toxicological Risk Assessments of Iron Oxide Nanocluster- and Gadolinium-Based T1MRI Contrast Agents in Renal Failure Rats

Qinjie Weng,Xi Hu,Jiahuan Zheng,Fan Xia,Nan Wang,Hongwei Liao,Ying Liu,Dokyoon Kim,Jianan Liu,Fangyuan Li,Qiaojun He,Bo Yang,Chunying Chen,Taeghwan Hyeon,Daishun Ling

Gadolinium-based contrast agents (GBCAs) are widely used for T1-weighted magnetic resonance imaging (MRI) in clinic diagnosis. However, a major drawback of GBCAs is that they can increase the toxicological risk of nephrogenic systemic fibrosis (NSF) in patients with advanced renal dysfunction. Hence, safer alternatives to GBCAs are currently in demand, especially for patients with renal diseases. Here we investigated the potential of polyethylene glycol (PEG)-stabilized iron oxide nanoclusters (IONCs) as biocompatible T1MRI contrast agents and systematically evaluated their NSF-related risk in rats with renal failure. We profiled the distribution, excretion, histopathological alterations, and fibrotic gene expressions after administration of IONCs and GBCAs. Our results showed that, compared with GBCAs, IONCs exhibited dramatically improved biosafety and a much lower risk of causing NSF, suggesting the feasibility of substituting GBCAs with IONCs in clinical MRI diagnosis of patients with renal diseases.

Poly(acrylic acid) modified lanthanide-doped GdVO4 hollow spheres for up-conversion cell imaging, MRI and pH-dependent drug release

Xiaojiao Kang,Dongmei Yang,Yunlu Dai,Mengmeng Shang,Ziyong Cheng,Xiao Zhang,Hongzhou Lian,Ping'an Ma,Jun Lin

In this study, multifunctional poly(acrylic acid) modified lanthanide-doped GdVO4 nanocomposites [PAA@GdVO4: Ln3+ (Ln = Yb/Er, Yb/Ho, Yb/Tm)] were constructed by filling PAA hydrogel into GdVO4 hollow spheres via photoinduced polymerization. The up-conversion (UC) emission colors (green, red and blue) can be tuned by changing the codopant compositions in the matrices. The composites have potential applications as bio-probes for cell imaging. Meanwhile, the hybrid spheres can act as T1 contrast agents for magnetic resonance imaging (MRI) owing to the existence of Gd3+ ions on the surface of composites. Due to the nature of PAA, DOX-loaded PAA@GdVO4:Yb3+/Er3+ system exhibits pH-dependent drug releasing kinetics. A lower pH offers a faster drug release rate. Such character makes the loaded DOX easily released at cancer cells. The cell uptake process of drug-loaded composites was observed by using confocal laser scanning microscopy (CLSM). The results indicate the potential application of the multifunctional composites as theragnostics (effective bimodal imaging probes and pH-responsive drug carriers).

Exploring atherosclerosis imaging with contrast-enhanced MRI using PEGylated ultrasmall iron oxide nanoparticles

Ruru Zhang,Kuan Lu,Li Xiao,Xuelan Hu,Wu Cai,Linjiang Liu,Yan Liu,Weihua Li,Hui Zhou,Zhiyuan Qian,Sixia Wang,Can Chen,Jianfeng Zeng,Mingyuan Gao

Plaque rupture is a critical concern due to its potential for severe outcomes such as cerebral infarction and myocardial infarction, underscoring the urgency of noninvasive early diagnosis. Magnetic resonance imaging (MRI) has gained prominence in plaque imaging, leveraging its noninvasiveness, high spatial resolution, and lack of ionizing radiation. Ultrasmall iron oxides, when modified with polyethylene glycol, exhibit prolonged blood circulation and passive targeting toward plaque sites, rendering them conducive for MRI. In this study, we synthesized ultrasmall iron oxide nanoparticles of approximately 3 nm via high-temperature thermal decomposition. Subsequent surface modification facilitated the creation of a dual-modality magnetic resonance/fluorescence probe. Upon intravenous administration of the probes, MRI assessment of atherosclerotic plaques and diagnostic evaluation were conducted. The application of Flash-3D sequence imaging revealed vascular constriction at lesion sites, accompanied by a gradual signal amplification postprobe injection. T1-weighted imaging of the carotid artery unveiled a progressive signal ratio increase between plaques and controls within 72 h post-administration. Fluorescence imaging of isolated carotid arteries exhibited incremental lesion-to-control signal ratios. Additionally, T1 imaging of the aorta demonstrated an evolving signal enhancement over 48 h. Therefore, the ultrasmall iron oxide nanoparticles hold immense promise for early and noninvasive diagnosis of plaques, providing an avenue for dynamic evaluation over an extended time frame.

Role of Mn2+ Doping in the Preparation of Core-Shell Structured Fe3O4@upconversion Nanoparticles and Their Applications in T1/T2-Weighted Magnetic Resonance Imaging, Upconversion Luminescent Imaging and Near-Infrared Activated Photodynamic Therapy

Yang Luo,Wei Zhang,Zhengfang Liao,Shengnan Yang,Shengtao Yang,Xinhua Li,Fang Zuo,Jianbin Luo

Core-shell (C/S) structured upconversion coated Fe3O4nanoparticles (NPs) are of great interest due to their potential as magnetic resonance imaging (MRI) and upconversion luminescent (UCL) imaging agents, as well as near-infrared activated photodynamic therapy (PDT) platforms. When C/S structured Fe3O4@Mn2+-doped NaYF4:Yb/Er NPs were prepared previously, well-defined C/S-NPs could not be formed without the doping of Mn2+during synthesis. Here, the role of Mn2+doping on the synthesis of core-shell structured magnetic-upconversion nanoparticles (MUCNPs) is investigated in detail. Core-shell-shell nanoparticles (C/S/S-MUCNPs) with Fe3O4as the core, an inert layer of Mn2+-doped NaYF4and an outer shell consisting of Mn2+-doped NaYF4:Yb/Er were prepared. To further develop C/S/S-MUCNPs applications in the biological field, amphiphilic poly(maleic anhydride-alt-1-octadecene) (C18PMH) modified with amine functionalized methoxy poly(ethylene glycol) (C18PMH-mPEG) was used as a capping ligand to modify the surface of C/S/S-MUCNPs to improve biocompatibility. UCL imaging, T1-weighted MRI ascribed to the Mn2+ions and T2-weighted MRI ascribed to the Fe3O4core of C/S/S-MUCNPs were then evaluated. Finally, chlorine e6 (Ce6) was loaded on the C/S/S-MUCNPs and the PDT performance of these NPs was explored. Mn2+doping is an effective method to control the formation of core-shell structured MUCNPs, which would be potential candidate as multifunctional nanoprobes for future T1/T2-weighted MR/UCL imaging and PDT platforms.Keywords:Mn2+;Fe3O4;upconversion;core-shell-shell;MR/UCL imaging;PDT platforms

Redox-Sensitive Citronellol–Cabazitaxel Conjugate: Maintained in Vitro Cytotoxicity and Self-Assembled as Multifunctional Nanomedicine

Peng Xue,Dan Liu,Jing Wang,Na Zhang,Jiahua Zhou,Lin Li,Weiling Guo,Mengchi Sun,Xiangfei Han,Yongjun Wang

Citronellol-cabazitaxel (CIT-ss-CTX) conjugate self-assembled nanoparticles (CSNPs) were designed and prepared by conjugating cabazitaxel with citronellol via the disulfide bond that is redox-sensitive to the high concentration of glutathione within tumor cells. Notably, the CSNPs maintained in the cell cytotoxicity. Moreover, the AUC0-t of CSNPs was 6.5-fold higher than that of cabazitaxel solutions and the t1/2 was prolonged 2.3 times. Furthermore, we found that CSNPs could be employed as an efficient carrier for other hydrophobic drugs or imaging agents. Thus, the in vivo targeting study was implemented via using 1,1-dioctadecyl-3,3,3,3-tetramethylindotricarbocyanine iodide (DiR)-loaded CSNPs as imaging agent, which showed CSNPs could effectively accumulate at the tumor site. Curcumin, a hydrophobic anticancer drug, was successfully loaded in CSNPs which exhibits good stability and synergistic antitumor effects. The citronellol–cabazitaxel conjugate therefore has a promising perspective as a multifunctional nanomedicine for combination therapy and theranostics attributed to its long-circulation property, redox-sensitive mechanism, and high drug coloading capability.

Theranostic Vesicles Based on Bovine Serum Albumin and Poly(ethylene glycol)-block-poly(l-lactic-co-glycolic acid) for Magnetic Resonance Imaging and Anticancer Drug Delivery

Qiuming Liu,Hongshi Zhu,Jingya Qin,Haiqing Dong,Jianzhong Du

Presented in this article is the preparation of a new theranostic vesicle which exhibits excellent in vitro and in vivo T1 magnetic resonance (MR) imaging contrast effect and good anticancer drug delivery ability. The theranostic vesicle has been easily prepared based on an amphiphilic biocompatible and biodegradable dibock copolymer, poly(ethylene glycol)-block-poly(l-lactic-co-glycolic acid) (PEG-b-PLGA) and bovine serum albumin-gadolinium (BSA-Gd) complexes. Dynamic light scattering (DLS), transmission electron microscopy (TEM), UV–vis spectroscopy, and inductively coupled plasma atomic emission spectroscopy (ICP-AES) measurements confirmed the formation and physiological stability of BSA-Gd@PEG-b-PLGA vesicles. Furthermore, the in vitro and in vivo MR imaging experiments revealed their excellent T1-weighted MR imaging function. Red blood cell hemolysis and cytotoxicity experiments confirmed their good blood compatibility and low cytotoxicity. Doxorubicin (DOX) loading and release experiments indicated a more retarded release rate of DOX in those theranostic vesicles than sole PEG-b-PLGA nanoparticles without BSA. Overall, this new biocompatible and biodegradable vesicle shows promising potential in theranostic applications.

Enhancing catalase-like activity of Prussian blue nanozyme by gadolinium-doping for imaging-guided antitumor amplification via photodynamic therapy and chemotherapy

H. Wang,Y. Shen,L. Chen,K. Li,Y. Shi,Z. Xu,D. Li,H. Chen,W. Wang,L. Gao

Deprivation of O 2 in most solid tumors greatly limits the effectiveness of photodynamic therapy. Prussian blue nanozyme (PBzyme) are reported to possess catalase-like (CAT) activity to alleviate tumor hypoxia . However, the low catalytic activity is far from satisfactory. Metal doping may improve the catalytic activity of PBzyme, but there are few reports in this regard. Herein, by a screening of metal dopants , Gd-doped PB with enhanced CAT activity is constructed on the basis of small-sized hollow mesoporous silica nanoparticles . Theoretical calculation demonstrates that Gd regulates the state density population of Fe in PBzyme and reduces the Gibbs free energy of CAT catalytic cycle, thus improving its CAT activity (approximately 4 times higher than that of pure PBzyme). After conjugated with photosensitizer Ce6 and tumor-targeted molecule folic acid and loaded with chemotherapy drug doxorubicin , the nanoplatform displays dual-enhanced T 1 and T 2 imaging capacity simultaneously. Upon high accumulation in tumors, the nanoplatform demonstrates augmented antitumor effects by combining enhanced photodynamic therapy and doxorubicin-mediated chemotherapy both in vitro and in vivo . This work demonstrates that the integration of Gd into PBzyme is an effective strategy to improve the catalytic activity of PBzyme and further potentiate the effectiveness of tumor theranostics.
