Enhanced burn wound healing by Controlled-Release 3D ADMSC-Derived exosome loaded hyaluronan hydrogel

Zhu Delong,Hu Ying,Kong Xiangkai,Luo Yuansen,Zhang Yi,Wu Yu,Tan Jiameng,Chen Jianwei,Xu Tao,Zhu Lei

Adipose mesenchymal stem cell (ADMSC)-derived exosomes (ADMSC-Exos) have shown great potential in regenerative medicine and been evidenced benefiting wound repair such as burns. However, the low yield, easy loss after direct coating, and no suitable loading system to improve their availability and efficacy hinder their clinical application for wound healing. And few studies focused on the comparison of biological functions between exosomes derived from different culture techniques, especially in exosome-releasing hydrogel system. Therefore, we designed a high-performance Exosome controllable releasing hydrogel system for burn wound healing, namely loading 3D printed microfiber culture derived exosomes in a highly biocompatible hyaluronic acid (HA). In this project, we compared the biological functions in vitro and in a burn model among exosomes derived from the conventional two-dimensional (2D) plate culture (2D-Exos), microcarrier culture (2.5D-Exos), and 3D printed microfiber culture (3D-Exos). Results showed that compared with 2D-Exos and 2.5D-Exos, 3D-Exos promoted HACATs and HUVECs cell proliferation and migration more significantly. Additionally, 3D-Exos had stronger angiogenesis-promoting effects in tube formation of (HUVECs) cells. Moreover, we found HA loaded 3D-Exos showed better burn wound healing promotion compared to 2D-Exos and 2.5D-Exos, including accelerated burn wound healing rate and better collagen remodeling. The study findings reveal that the hyaluronic acid (HA)-loaded, controllable-release 3D-Exos repair system distinctly augments therapeutic efficacy in terms of wound healing, while concurrently introducing a facile application approach. This system markedly bolsters the exosomal loading efficiency, provides a robust protective milieu, and potentiates the inherent biological functionalities of the exosomes. Our findings provide a rationale for more efficient utilization of high-quality and high-yield 3D exosomes in the future, and a novel strategy for healing severe burns.

A NIR light-activated PLGA microsphere for controlled release of mono- or dual-drug

Yiyu Wang,Wenlong Yu,Chunqing Niu,Guiting Yu,Xin Huang,Jian Shi,Da Ma,Xuezheng Lin,Kai Zhao

Microspheres produced from biodegradable poly (D, L-lactic-co-glycolic acid) (PLGA) have been studied widely as drug delivery carriers for therapeutics. This study aimed to develop a near-infrared (NIR) light-activated PLGA microsphere loaded with two kinds of drugs with different molecular weights respectively or simultaneously by the double emulsion method (bovine serum albumin (BSA) and tetracycline hydrochloride (TH) as the drug model agents, indocyanine green (ICG) as the NIR responsive component). To optimize the incorporation of drugs and particle size for PLGA microspheres loaded single drug, the effect of three processing parameters, including the amount of polymer, initial ICG content, and initial BSA or TH content on the drug entrapment efficiency (EE), drug loading and the particle size were investigated. The PLGA microspheres were spherical and ranged from 5 to 10 μm in diameter. The EE of ICG and BSA in PLGA@ICG-BSA was 78.41 ± 0.71% and 69.88 ± 5.14%, respectively. The EE of ICG and TH in PLGA@ICG-TH was 71.52 ± 5.07% and 28.39 ± 3.39%. The PLGA microspheres exhibited desirable biodegradability and low cytotoxicity. Based on the optimized processing of single drug-loaded microspheres, dual-drug-loaded microspheres containing both large and small molecule drugs were further prepared. The EE of ICG, BSA, and TH was 83.37 ± 0.59%, 52.65 ± 3.48%, and 12.88 ± 2.53%; the particle size was ∼7.21 μm. All the NIR-responsive PLGA microspheres exhibited an efficient photothermal property and enhanced photothermal stability under multiple NIR irradiation cycles. The drugs from the different PLGA microspheres were released continuously over 18 days, mainly determined by Fick's diffusion. Furthermore, the drugs can be controlled released through a NIR light trigger, resulting in a dosage increase at a specific time point.

Thermally induced and physically cross-linked hydrogel doped with graphene oxide for controlled release

Peihong Li,Xueyan Dai,Yanlong Sui,Rui Li,Chunling Zhang

Graphene oxide (GO) is an ideal hydrogel material because of its water solubility, non-toxicity, and excellent mechanical properties. Here, we added GO to oligo(lysine)-modified F127 to prepare a series of FLGO composite hydrogels. The FLGO hydrogel was thermally induced, stable and injectable. And the content of GO would affect the sol–gel transition, rheological properties and glass transition temperature of the FLGO hydrogel. GO was connected to the matrix through electrostatic interactions and hydrogen bonds. The cross-linking effect of GO enhanced the FLGO hydrogel. We also studied the release properties of the FLGO hydrogel loaded with anticancer drug 5-fluorouracil. Compared with F127 hydrogel, the FLGO hydrogel showed a linear, slower and stable release pattern within one week. The release rate of FLGO hydrogel could be adjusted by the pH and it was faster under acidic conditions. Therefore, the FLGO hydrogel is expected to be used as a drug release system in the field of biomedicine.

Multilayer composite beads constructed via layer-by-layer self-assembly for lysozyme controlled release

Jiemin Zhao,Xiaoping Wang,Yanshen Kuang,Yufeng Zhang,Xiaowen Shi,Xingyun Liu,Hongbing Deng

Alginate (ALG)–lysozyme (LZ) beads were fabricated by a cross-linking process. Negatively charged ALG and positively charged LZ were alternately deposited on the positively charged ALG–LZ beads via layer-by-layer (LBL) self-assembly technique. The mechanical properties and the enzymatic activity of these samples were studied by regulating the number of deposition bilayers and the composition of the outermost layer. The scanning electron microscopy images indicated that the resultant samples exhibited good sphericity and porosity. The Fourier transform infrared spectra results implied the presence of electrostatic interactions between ALG and LZ. The pore size distribution results revealed that the samples mainly possessed mesopores with radius in the range of 2–7 nm. In vitro LZ release test performed at different time intervals showed that LZ could be released from ALG–LZ beads and LBL film-coated beads. The amount of released LZ increased with extended time intervals.

NGR-modified pH-sensitive liposomes for controlled release and tumor target delivery of docetaxel

Zili Gu,Minglu Chang,Yang Fan,Yanbin Shi,Guimei Lin

As current tumor chemotherapy faces many challenges, it is important to develop drug delivery systems with increased tumor-targeting ability, enhanced therapeutic effects and reduced side effects. In this study, a pH-sensitive liposome was constructed containing CHEMS-anchored PEG2000 for extended circulation and NGR peptide as the targeting moiety. The NGR-modified docetaxel-loaded pH-sensitive extended-circulation liposomes (DTX/NGR-PLL) prepared possess suitable physiochemical properties, including particle size of approximately 200   nm, drug encapsulation efficiency of approximately 70%, and pH-sensitive drug release properties. Experiments performed in vitro and in vivo on human fibrosarcoma cells (HT-1080) and human breast adenocarcinoma cells (MCF-7) verified the specific targeting ability and enhanced antitumor activity to HT-1080 cells. The results of intravenous administration demonstrated that NGR-modified liposomes can significantly and safely accumulate in tumor tissue in xenografted nude mice. In conclusion, the liposomes constructed hold promise as a safe and efficient drug delivery system for specific tumor treatment.

ATP-responsive near-infrared fluorescence MOF nanoprobe for the controlled release of anticancer drug

Chen Xi-Xi,Hou Mei-Jia,Mao Guo-Jiang,Wang Wen-Xin,Xu Fen,Li Yongfei,Li Chun-Yan

A near-infrared (NIR) fluorescence nanoprobe named RhI-DOX@ZIF-90 has been synthesized by wrapping the guest molecule (RhI and DOX) into ZIF-90 framework. The nanoprobe itself is non-fluorescent and the drug (DOX) is inactive. Upon the addition of ATP, the structure of RhI-DOX@ZIF-90 is degraded. The fluorescence of RhI is recovered and DOX is released. The nanoprobe can detect ATP with high sensitivity and selectivity. There is good linear relationship between the nanoprobe and ATP concentration from 0.25 to 10 mM and the detection limit is 0.10 mM. The nanoprobe has the ability to monitor the change of ATP level in living cells and DOX is released inducing apoptosis of cancer cells. RhI-DOX@ZIF-90 is capable of targeting mitochondria, which provides a basis for improving the efficiency of drug delivery by mitochondrial administration. In particular, the nanoprobe is preferentially accumulated in the tumor sites and detect ATP in tumor mice by fluorescence imaging using near-infrared fluorescence. At the same time, DOX can be released accurately in tumor sites and have good anti-tumor efficiency. So, this nanoprobe is a reliable tool to realize early diagnosis of cancer and improve effect of anticancer drug. Graphical abstract

A novel pH-responsive controlled release system based on mesoporous silica coated with hydroxyapatite

Cui Liru,Lin Huiming,Guo Wei,Xiang Di,Zhou Dan,Zhang Ting,Qu Fengyu

With well bioactive and nontoxic, hydroxyapatite (HAp) was employed to seal the nanopores of mesoporous silica (MCM-41) to realize the pH-responsive controlled release. First, MCM-41 was modified with cationic polymer, poly-(diallyldimethylammoniumchloride) (PA). And after the addition of Ca 2+ /PO 4 3− , HAp precipitation can take place based on the cationic sites derived from PA. It is a simple and effective way to obtain HAp coating MCM-41 system (MHAs). The structure of the system was characterized by X-ray diffraction, scanning electron microscope, transmission electron microscope, N 2 adsorption–desorption and so on. Metformin hydrochloride was used as the model drug, and the drug release performance and the release kinetics of the system were investigated in detail. Because of the degradation of HAp under acid condition, the drug loading MHAs showed a well pH-sensitive controlled release behavior. From above investigation, MHAs is a promising platform to construct a pH-responsive controlled drug delivery system, especially for some low pH tissues, such as inflammatory and tumor.

Hydrogen-Bonding Reinforced Injectable Hydrogels: Application As a Thermo-Triggered Drug Controlled-Release System

Dong Wang,Yuzheng Xia,Dong Zhang,Xiaomeng Sun,Xiaonong Chen,Susan Oliver,Shuxian Shi,Lei Lei

Most injectable hydrogels are prone to irreversible deformation or damage under sustained shear forces and exhibit low mechanical properties, which limit their practical applications in biological engineering. In this work, the first hydrogen-bonding reinforced injectable thermoresponsive hydrogel system has been fabricated through an in situ hydrazide–aldehyde cross-linking reaction between hydrazide-functionalized poly(N-isopropylacrylamide) (PNIPAM) and dialdehyde dextrin. The mechanical strength and stability of the hydrogel were reinforced by the introduction of intermolecular hydrogen-bonding from poly(N-acryloyl glycinamide) (PNAGA), which possesses great biocompatibility and similarity to PNIPAM. The internal morphology, dynamic moduli, de-cross-linking properties, and temperature responsiveness of the hydrogels were systematically investigated. Upon hydrogen-bonding reinforcement, the hydrogel elasticity, reflected by the maximum storage modulus, dramatically increased from 90.2 to 517.0 kPa, which is about a 570% increase compared with unreinforced hydrogels. In addition, the mechanical properties of the hydrogel were reinforced as the de-cross-linking could be inhibited by PNAGA-based hydrogen-bonding. The temperature release behavior of the hydrogel was assessed with a model drug (propranolol hydrochloride, PHCl) and could be precisely controlled by adjusting the environmental temperature between 25 and 37 °C. Therefore, PNAGA reinforced thermoresponsive hydrogels are promising functional biological materials for use in tissue-repair engineering and controlled-release drug delivery.

Mechanically enhanced lipo-hydrogel with controlled release of multi-type drugs for bone regeneration

Ruoyu Cheng,Yufei Yan,Han Liu,Hao Chen,Guoqing Pan,Lianfu Deng,Wenguo Cui

Hydrogel is an excellent candidate in the field of drug delivery and tissue regeneration. However, hydrogel used as drug carriers is facing several problems including limited optional drug and burst drug release, besides hydrogel is still confronting some challenges in mechanical property involving decreased hydrogel's strength caused by interface separation between hydrogel and other solid composition. In this research, we designed a mechanical enhanced and release-controlling liposome modified hydrogel via hydrogen bone and hydrogel network micro-cross-linking double network as artificial scaffold for promoting the angiogenesis and osteogenesis in bone regeneration. Specifically, photo-cross linkable gelatin derivative (GelMA) was combined with drug loaded liposome by bonding effects, which lead to double improvement in mechanical property, such as compression, stretching, torsion and resiliency. Furthermore, the ability to control phased release was observed in the composite hydrogel including the early release of hydrophilic drug (deferoxamine), mid-term release of bioactive macromolecule (bovine serum albumin and bone morphogenetic protein 2) and long-term release of liposoluble medicine (paclitaxel). In vitro study revealed that, this composite hydrogel significantly promoted osteogenic and angiogenesis differentiation without significant influence on cell adhesion, proliferation. Finally, the composite hydrogel demonstrated a superior capacity in accelerating new and mature lamellar bone formation in distal femoral defect of Sprague-Dawley rats. Thus, the liposome modified hydrogel could supply a promising strategy for extending the application of hydrogel in drug delivery and tissue reconstruction.

Controlled release of hydrogen by implantation of magnesium induces P53-mediated tumor cells apoptosis

Rui Zan,Hao Wang,Weijie Cai,Jiahua Ni,Bérengère J.C. Luthringer-Feyerabend,Wenhui Wang,Hongzhou Peng,Weiping Ji,Jun Yan,Jiazeng Xia,Yang Song,Xiaonong Zhang

Hydrogen has been used to suppress tumor growth with considerable efficacy. Inhalation of hydrogen gas and oral ingestion of hydrogen-rich saline are two common systemic routes of hydrogen administration. We have developed a topical delivery method of hydrogen at targeted sites through the degradation of magnesium-based biomaterials. However, the underlying mechanism of hydrogen's role in cancer treatment remains ambiguous. Here, we investigate the mechanism of tumor cell apoptosis triggered by the hydrogen released from magnesium-based biomaterials. We find that the localized release of hydrogen increases the expression level of P53 tumor suppressor proteins, as demonstrated by the in vitro RNA sequencing and protein expression analysis. Then, the P53 proteins disrupt the membrane potential of mitochondria, activate autophagy, suppress the reactive oxygen species in cancer cells, and finally result in tumor suppression. The anti-tumor efficacy of magnesium-based biomaterials is further validated in vivo by inserting magnesium wire into the subcutaneous tumor in a mouse. We also discovered that the minimal hydrogen concentration from magnesium wires to trigger substantial tumor apoptosis is 91.2 μL/mm 3 per day, which is much lower than that required for hydrogen inhalation. Taken together, these findings reveal the release of H 2 from magnesium-based biomaterial exerts its anti-tumoral activity by activating the P53-mediated lysosome-mitochondria apoptosis signaling pathway, which strengthens the therapeutic potential of this biomaterial as localized anti-tumor treatment.

A Robust Gel Polymer Electrode with Controlled-Release Polymerization towards Practical Lithium-Sulfur Batteries

Li Jingkang,Wang Donghua,Gao Nanwu,Cai Wenlong,Zhang Junjie,Zou Zhijuan,Song Lixian,Song Yingze,Zhou Guangmin

While rational electrode engineering has promoted the kinetics of lithium-sulfur batteries, persistent lithium polysulfide diffusion continues to undermine the cyclability of conventional liquid electrolyte systems. We propose a quasi-solid gel electrode design that integrates the sulfur species-blocking attributes of a solid-state system within a liquid electrolyte framework. This is achieved through a well-designed free radical-controlled-release polymerization strategy, which precisely governs the formation of a processable poly(ethyl cyanoacrylate) network. This approach enables secondary in situ gelation triggered by electrolyte infiltration, forming gel-phase electrode architectures. Mechanistically, the resulting gel-based electrode provides dynamically adaptive stress dissipation to accommodate volume variations, establishes continuous ion-permeable pathways for deeper positive electrode discharge, and acts as a synergistic physico-chemical barrier for sulfur species confinement. As a result, the positive electrode delivers a high initial capacity of 812.8 mA h g –1 at 2 C and sustains stable cycling operation over 1000 cycles. A 1.7 Ah-level pouch cell demonstrates potential for a sustained power supply for an unmanned aerial vehicle.

ROS-responsive cinnamaldehyde-based delivery system for targeted delivery and controlled release of avermectin

Yuying Xie,Yangchun Ge,Zhuochao Shen,Yupeng Qiu,Haohua Zhang,Changmeng Sun,Jiangkuan Tang,Honglei Wang,Bingke Li,Junran Xu,Ping Liang,Xuqiang Peng,Lanlan Zhai,Zhongyu Li,Zhihui Li

ROS-triggered release of avermectin from cinnamaldehyde-based delivery system; • Enhanced insecticidal efficacy against Plutella xylostella larvae; • Improved leaf adhesion and favorable plant safety profile; • Stable carriers with high drug loading capacity and encapsulation efficiency. To achieve precise delivery and controlled release of pesticides, this study designed and developed a reactive oxygen species (ROS)-responsive nano-pesticide triggered by the excessively high ROS levels in pests, enabling precise and efficient control of target pests. Using the natural compound cinnamaldehyde as the core, an ROS-responsive amphiphilic small molecule was synthesized, and hydrophobic pesticide avermectin-loaded micelle suspension were successfully prepared through self-assembly. Characterization results showed that the avermectin-loaded micelle suspension had an average particle size of 136.0 nm, a drug loading capacity of 17.78 ± 0.13%, and an encapsulation efficiency of 77.05 ± 0.55%, significantly enhancing the adhesion of pesticides to leaf surfaces. In vitro release experiments confirmed that under micelle suspension, the cumulative release rate was minimal over 50 h, while under simulated high ROS conditions in pests, the cumulative release rate reached 86.02% ± 1.69% over 50 h. Indoor bioassays demonstrated that the micelle suspension exhibited superior efficacy against Plutella xylostella larvae compared to conventional pesticides. Plant safety experiments indicated that the micelle suspension possesses favorable safety profiles.
